Description

Compound vitamin pharmaceutical preparation for broiler chickens and preparation method and application thereof
Technical Field
The invention relates to the technical field of biological medicines, in particular to a compound vitamin pharmaceutical preparation for broiler chickens and a preparation method and application thereof.
Background
Calcium and phosphorus are the two mineral elements with the greatest demand for poultry organisms, and the ratio of calcium to phosphorus is approximately seven of the ratio of mineral microelements of the poultry organisms. In modern broiler farming, feed is the main source of calcium and phosphorus, which are mainly absorbed by the intestinal tract, re-absorbed in the kidneys and retained in the bones.
Vitamin D3 is used for regulating the absorption of calcium and phosphorus in the intestinal tracts of broilers, and vitamin D3 needs to be activated in two steps in the body of broilers: firstly, converting into 25-hydroxy vitamin D3, then converting into 1, 25-dihydroxyvitamin D3, and finally, regulating the absorption of calcium and phosphorus in the intestinal tracts of broilers in the active form of 1, 25-dihydroxyvitamin D3.
However, in the prior art, the biological activity of vitamin D3 used in the feed industry is very low, so that the content of the finally converted 1, 25-dihydroxyvitamin D3 is low, and the calcium and phosphorus absorbed in the intestinal tracts of the broiler chickens are insufficient, thereby influencing the growth and development of the broiler chickens, and in order to ensure the growth performance of the broiler chickens, the calcium and phosphorus absorption in the intestinal tracts of the broiler chickens is generally promoted by increasing the addition amount of the vitamin D3. However, excessive addition of vitamin D3 can result in high pressure of 25-hydroxylation of the liver of broiler chickens, damage to the liver, and decrease the activation efficiency of vitamin D3 after liver injury, and the problems caused by such circulation are as follows:
1. the usage amount of vitamin D3 is large, the cost is high, the vitamin D3 can not be fully absorbed by livestock organisms, and finally the vitamin D3 is discharged out of the body, so that the raw materials are wasted.
2. The vitamin D3 has low activation efficiency, reduces the absorption of calcium and phosphorus in the intestinal tracts of the broiler chickens, and influences the bone growth and development of the broiler chickens.
3. Vitamin D3 is contained in the liver of the broiler chickens in a large amount, so that the liver is burdened, and the liver function of the broiler chickens is damaged.
From the above, it is clear that the regulation of calcium and phosphorus by vitamin D3 is ultimately achieved in the active form of 1, 25-dihydroxyvitamin D3, paper: compared with 1 alpha-hydroxy vitamin D3 and 1, 25-dihydroxyvitamin D3 in daily ration of broiler chickens, the biological cost performance ratio (feed additive, 2015,36 (22)) of the 1 alpha-hydroxy vitamin D3 is disclosed, and the 1, 25-dihydroxyvitamin D3 can promote the growth and mineralization of bones of low-calcium and low-phosphorus broiler chickens. The preparation of glaucophyllum plant extracts with high content of 1, 25-dihydroxyvitamin D3 glycoside and quercetin and their use are disclosed in the publication No. CN 102014945B, wherein 1, 25-dihydroxyvitamin D3 is disclosed as useful for the prevention and treatment of diseases associated with reduced bone mass, in particular for the treatment of tibial cartilage dysplasia in broilers, as well as for the treatment of milk fever, and in addition as a food supplement for humans or animals.
However, the above prior art does not disclose how 1, 25-dihydroxyvitamin D3 promotes the growth of low-calcium, low-phosphorus broiler bones. In addition, since the commercial 1, 25-dihydroxyvitamin D3 is produced in very low yields and is very expensive, it is impossible to use it as a conventional feed additive for a long period of time.
Disclosure of Invention
In view of the above, the invention provides a compound vitamin pharmaceutical preparation for broiler chickens, and a preparation method and application thereof. The method directly uses the 1, 25-dihydroxyvitamin D3, does not need to carry out twice conversion in vivo, has good activity of the 1, 25-dihydroxyvitamin D3, can fully play a role, does not need to add a large amount of vitamin D3, can ensure calcium and phosphorus absorption of the body of the broiler chickens, and can avoid liver injury to a certain extent.
The technical scheme of the invention is realized as follows:
in a first aspect, the invention provides a compound vitamin pharmaceutical preparation for broiler chickens, which comprises the following components in parts by weight:
20-30 parts of vitamin A, 0.02-0.08 part of 1, 25-dihydroxyvitamin D, 3-8 parts of vitamin K, 120-150 parts of vitamin E, 10-30 parts of vitamin B, 50-100 parts of vitamin B2, 32-38 parts of vitamin B6, 0.06-0.15 part of vitamin B12, 330-370 parts of nicotinic acid, 90-120 parts of pantothenic acid, 5-6 parts of folic acid, 1.5-2 parts of biotin and 500-700 parts of corncob powder.
In the vitamin D complex pharmaceutical preparation, 1, 25-dihydroxyl vitamin D3 is directly used, and the 1, 25-dihydroxyl vitamin D3 is mainly stored in the liver of broiler chickens, and takes part in the regulation of blood calcium and phosphorus, maintains bone steady state, regulates cell proliferation and differentiation, enhances immunity, and has the effects of anti-inflammatory, anti-fibrosis and the like. Compared with vitamin D3, the method does not need two steps of activation, is not influenced by the high and low conversion rate, and the 1, 25-dihydroxyvitamin D3 can fully play the role of promoting calcium and phosphorus absorption.
Vitamin A is mainly stored in the liver of broiler chickens, and has the effects of maintaining normal visual function, improving immunity, maintaining normal growth and development of bones, promoting growth and reproduction, and the like.
Although normal amount of 1, 25-dihydroxyvitamin D3 has a protective effect on chicken liver, and promotes intestinal calcium and phosphorus absorption. However, excessive vitamin D3 also impairs liver function, resulting in a decrease in vitamin a storage capacity and a decrease in vitamin a utilization. When the vitamin A is excessively added (15000 IU/kg), the liver of the chicken is damaged, anabolism of vitamin D in the liver is blocked, the concentration of serum vitamin D is reduced, the absorption of calcium and phosphorus in intestinal tracts is reduced, the discharge of calcium and phosphorus is increased, the calcium and phosphorus content in legs is reduced, the bone strength is weakened, and fracture is easily caused.
Thus, vitamin a and vitamin D are two mutually antagonistic and mutually constraining vitamins. According to the invention, the 1, 25-dihydroxyl vitamin D3 and the vitamin A respectively exert the promotion functions by controlling the mixing proportion of the 1, 25-dihydroxyl vitamin D3 and the vitamin A in the compound vitamin pharmaceutical preparation, and the compound vitamin pharmaceutical preparation can restrict each other, so that the damage to the body of broilers is avoided.
Vitamin E is mainly stored in the liver and plays an important role in resisting oxidation, maintaining cell membrane stability and protecting cells from oxidative damage. The vitamin A and the vitamin E have synergistic effect, and the vitamin A and the vitamin E can jointly offset the side effect of the 1, 25-dihydroxyvitamin D3 by controlling the mixing proportion of the vitamin A and the vitamin E, so that the liver health is ensured, and the normal vitamin D effect and metabolic activity of broilers are maintained.
Vitamin K belongs to an accelerator for bone formation, and is added into a compound vitamin pharmaceutical preparation to have the effect of resisting osteoporosis; vitamin B1 has the function of maintaining normal sugar metabolism and nerve conduction; vitamin B2 participates in the growth metabolism of cells, and is a necessary nutrient for metabolism and repair of body tissues, such as strengthening liver function and regulating secretion of epinephrine; vitamin B6 is involved in a variety of metabolic reactions, particularly in the close relationship with amino acid metabolism; vitamin B12 can increase folic acid utilization rate and promote metabolism of carbohydrate, fat and protein; the nicotinic acid can improve the utilization rate of feed protein; pantothenic acid is a substance necessary for fatty acid synthesis of steroids; folic acid can promote the growth of animals and improve the immunity of organisms; biotin is an essential substance for normal metabolism of fats and proteins.
Corncob meal is used as a diluent for 1, 25-dihydroxyvitamin D3.
In a second aspect, the invention also provides a preparation method of the compound vitamin pharmaceutical preparation for broiler chickens, which comprises the following steps:
weighing all raw materials according to the weight ratio;
step two, taking 1, 25-dihydroxyvitamin D3 crystals, dissolving the 1, 25-dihydroxyvitamin D3 crystals by using absolute ethyl alcohol, and then adding 1, 2-propylene glycol for dilution to obtain a 1, 25-dihydroxyvitamin D3 solution;
adding the 1, 25-dihydroxyvitamin D3 solution into the corn cob powder serving as a diluent, and pre-dispersing and dissolving the 1, 25-dihydroxyvitamin D3 solution to obtain mixed powder of the 1, 25-dihydroxyvitamin D3 solution and the corn cob powder.
And step four, adding the mixed powder obtained in the step three into the rest raw materials, and uniformly mixing.
In the invention, the volume concentration of the absolute ethyl alcohol is 75%; the 1, 2-propylene glycol is analytically pure with mass concentration more than 99%. The mixing operation of the remaining raw materials in the fourth step is a conventional operation performed by those skilled in the art in the process of producing and processing feeds, for example, stirring in a stirring kettle, so that the stirring operation is a prior art, and the present invention will not be repeated.
In the invention, the addition volume ratio of the absolute ethyl alcohol to the 1, 2-propylene glycol is 1:17-20. The volume mass ratio of the 1, 25-dihydroxyvitamin D3 solution to the corncob powder is 1:850-1200mL/g. The mass concentration of the 1, 25-dihydroxyvitamin D3 solution is 8-14 mug/mL.
Because the 1, 25-dihydroxyl vitamin D3 is a fat-soluble substance, alcohol substances are needed to be dissolved, and the absolute ethyl alcohol has good solubility, but can affect the activity of other vitamins, and a large amount of absolute ethyl alcohol cannot be added into the broiler chicken pharmaceutical preparation, so that the absolute ethyl alcohol cannot be used in a large amount as a diluent; 1, 2-propanediol can dissolve 1, 25-dihydroxyvitamin D3, but is oily and viscous, has poor dissolution effect on 1, 25-dihydroxyvitamin D3, can only be used as a diluent, and 1, 2-propanediol is a food emulsifier and can be used as a food additive, and is harmless to animals and human bodies, so that 1, 2-propanediol is used as a diluent.
In the invention, after 1, 25-dihydroxyvitamin D3 crystals are fully dissolved and diluted to a certain concentration, 1, 25-dihydroxyvitamin D3 can be fully mixed in other raw materials, and if 1, 2-propanediol is not used for dissolution and dilution, the 1, 25-dihydroxyvitamin D3 is unevenly mixed in other raw materials, so that antagonism is generated between different vitamins, and the drug effect is reduced.
Although the amount of 1, 25-dihydroxyvitamin D3 added in the multivitamin is very small, if the 1, 25-dihydroxyvitamin D3 solution is directly added into other multivitamins, antagonism is still generated between different vitamins, so that the pharmaceutical activity of the multivitamin is reduced, and therefore, the 1, 25-dihydroxyvitamin D3 solution needs to be pre-dispersed and dissolved in corncob powder which does not have an effect on the activity. Because of the small amount of 1, 25-dihydroxyvitamin D3 solution added to the corncob meal, the diluent corncob meal remains slightly moist in powder form after pre-dispersion dissolution.
In a third aspect, the invention also provides an application method of the compound vitamin pharmaceutical preparation for broiler chickens, the compound vitamin pharmaceutical preparation is added into broiler chickens feed of 1-3 weeks old, and the addition amount of the compound vitamin pharmaceutical preparation in the broiler chickens feed is 160-230mg/kg.
As 1-3 weeks old is in the growth period, which is the stage of the fastest growth of the bones of the broiler chickens, a large amount of 1, 25-dihydroxyvitamin D3 is needed to promote the absorption of calcium and phosphorus so as to ensure the normal bone development and growth, and therefore, the compound vitamin pharmaceutical preparation is mainly applied to the feed of the broiler chickens of 1-3 weeks old. The broiler chickens after 3 weeks old are not in the stage of fastest bone growth, so the demand for calcium and phosphorus is reduced, the vitamin D3 with lower price can be replaced, and a small amount of vitamin D3 can be added in the stage, so that the demand of the broiler chickens for calcium and phosphorus is maintained, and the liver of the broiler chickens is not damaged.
The dosage form of the compound vitamin pharmaceutical preparation can be powder or granules.
The compound vitamin pharmaceutical preparation for broiler chickens, and the preparation method and the application thereof have the following beneficial effects compared with the prior art:
1. the 1, 25-dihydroxyvitamin D3 is selected in the compound vitamin pharmaceutical preparation, and because the absorption of the broiler chickens to calcium and phosphorus mainly depends on the action of the vitamin D3, the vitamin D3 can really play a role only after being converted into the 1, 25-dihydroxyvitamin D3 in vivo, the 1, 25-dihydroxyvitamin D3 is directly used without being converted in vivo twice, the 1, 25-dihydroxyvitamin D3 has better activity, can play a role fully, does not need a large amount of addition, can ensure the calcium and phosphorus absorption of the broiler chickens, and can avoid liver injury to a certain extent.
2. Although the addition amount of 1, 25-dihydroxyvitamin D3 is less than that of vitamin D3, liver injury can be avoided to a certain extent, the addition amount of 1, 25-dihydroxyvitamin D3 needs to be enough, which is unavoidable and slightly burdens the liver, so that the damage to the body of broilers is avoided by the aid of the mutual antagonism of vitamin A and 1, 25-dihydroxyvitamin D3 by adding vitamin A into the multi-vitamin pharmaceutical preparation.
3. On the basis of vitamin A, in order to further eliminate the side effect of 1, 25-dihydroxyvitamin D3, vitamin E is added into the compound vitamin pharmaceutical preparation, and the vitamin E and the vitamin A have synergistic effect, so that the side effect of 1, 25-dihydroxyvitamin D3 is better eliminated.
4. In the preparation process of the compound vitamin pharmaceutical preparation, the 1, 25-dihydroxyvitamin D3 crystal can be dissolved by the cooperation of absolute ethyl alcohol and 1, 2-propylene glycol in a specific proportion, so that the uniform mixing degree of the 1, 25-dihydroxyvitamin D3 and other vitamins is ensured; meanwhile, the 1, 25-dihydroxyvitamin D3 solution and the corncob powder are required to be pre-dispersed and dissolved, so that the mixing uniformity degree of the 1, 25-dihydroxyvitamin D3 and other vitamins can be further improved, and the mutual antagonism among different vitamins caused by uneven mixing is avoided.
5. Because the price of the 1, 25-dihydroxyvitamin D3 crystal is relatively high, the compound vitamin medicinal preparation is added into the 1-3 week old broiler feed for reducing the raw material cost of broiler chicken breeding, and the compound vitamin medicinal preparation is not required to be used in the whole broiler chicken breeding process, so that the skeletal growth and development of broiler chickens can be ensured, and the cost can be saved.

TABLE 2 growth performance index
Table 3 index of tibial mineralization of broiler chickens
From the data in the above 4 examples, the indexes in the examples 1 and 2 have better effect, so the preferable proportion of the compound vitamin pharmaceutical preparation for broiler chickens is as follows: 25 parts of vitamin A, 0.06 part of 1, 25-dihydroxyvitamin D3, 5 parts of vitamin K, 130 parts of vitamin E, 1 part of vitamin B1, 2 parts of vitamin B2, 6 parts of vitamin B35, 0.1 part of vitamin B12, 350 parts of nicotinic acid, 100 parts of pantothenic acid, 5.5 parts of folic acid, 1.8 parts of biotin and 690 parts of corncob powder. It can also be: 27 parts of vitamin A, 0.05 part of 1, 25-dihydroxyvitamin D, 4.6 parts of vitamin K, 135 parts of vitamin E, 1 part of vitamin B1, 60 parts of vitamin B2, 36 parts of vitamin B6, 0.12 part of vitamin B12, 365 parts of nicotinic acid, 115 parts of pantothenic acid, 5.3 parts of folic acid, 1.6 parts of biotin and 660 parts of corncob powder. The addition volume ratio of the absolute ethyl alcohol to the 1, 2-propylene glycol is preferably 1:18 or 1:19. The volume mass ratio of the 1, 25-dihydroxyvitamin D3 solution to the corncob meal is preferably 1:960 or 1:1100mL/g. The mass concentration of the 1, 25-dihydroxyvitamin D3 solution is preferably 10 or 12. Mu.g/mL. The addition amount of the compound vitamin medicinal preparation in the broiler feed is preferably 200 or 220mg/kg.
As can be seen from the data in comparative example 1 and example 2, the feed directly added with vitamin D3 has low activity of vitamin D3, and can not be completely converted into 1, 25-dihydroxyvitamin D3, and the calcium and phosphorus absorption of intestinal tracts is insufficient, so that various indexes are relatively poor.
As can be seen from the data in comparative examples 2-4 and example 2, antagonism between vitamin a and 1, 25-dihydroxyvitamin D3 helps to eliminate the side effects of 1, 25-dihydroxyvitamin D3, improve the calcium and phosphorus absorption effect of broilers, and promote growth performance; meanwhile, the addition amount of the vitamin A needs to be controlled within the range of 20-30 parts to exert good antagonism.
The data of comparative examples 5-6 show that the addition amount of 1, 25-dihydroxyvitamin D3 needs to be controlled to be 0.02-0.08 part, so that the compound vitamin medicinal preparation for broiler chickens can exert the optimal effect and ensure that the broiler chickens have the best calcium and phosphorus absorption and growth performance.
As can be seen from the data of comparative examples 7 to 8, the addition amount of vitamin E is controlled to be 120 to 150 parts, so that the vitamin E and vitamin A can play a synergistic effect together, and the side effects of 1, 25-dihydroxyvitamin D3 can be eliminated together.
As can be seen from the data of comparative examples 9 to 10, the crystals of 1, 25-dihydroxyvitamin D3 are diluted by the dissolution of absolute ethyl alcohol and 1, 2-propanediol, so that 1, 25-dihydroxyvitamin D3 and other components can be uniformly mixed, antagonism between different vitamins is avoided, and the drug effect is reduced; meanwhile, the volume ratio of the absolute ethyl alcohol to the 1, 2-propylene glycol needs to be controlled within a reasonable range to better exert the effect.
As can be seen from the data of comparative examples 11-12, the pre-dispersion dissolution of 1, 25-dihydroxyvitamin D3 is required by using corncob powder, so that 1, 25-dihydroxyvitamin D3 can be uniformly mixed with other components, antagonism between different vitamins is avoided, and the drug effect is reduced; meanwhile, the volume and mass ratio of the 1, 25-dihydroxyvitamin D3 solution to the corncob powder needs to be controlled within a reasonable range to better exert the effect.
As can be seen from the data of comparative examples 13 to 14, the effect of the compound vitamin pharmaceutical preparation is reduced when the addition amount of the compound vitamin pharmaceutical preparation in the broiler feed is less than 160mg/kg, and the effect is not improved when the addition amount of the compound vitamin pharmaceutical preparation is more than 230mg/kg, so that the addition amount can be controlled to be not more than 230mg/kg in order to reduce the cost.
The foregoing description of the preferred embodiments of the invention is not intended to be limiting, but rather is intended to cover all modifications, equivalents, alternatives, and improvements that fall within the spirit and scope of the invention.
