背景技术
复杂介质中蛋白质的超灵敏检测对于医学诊断、生物标志物发现和药物研发等至关重要。例如,在神经源性疾病中,如帕金森病(PD)和阿尔茨海默病(AD),蛋白的生物标志物由于血脑屏障的存在而以极低的水平存在于外周血中。使用如酶联免疫吸附测定(ELISA)或化学发光等传统分析方法定量检测这些生物标志物是具有挑战性的。因此,目前PD和AD在临床诊断上仍依赖于医学成像和病理分析,而不是分子图谱。近年来,超灵敏的免疫传感取得了快速发展,有代表性的如单分子免疫传感技术和超灵敏电化学发光等。其中,数字免疫传感在超高灵敏度(如亚飞摩尔检测)方面引起了越来越多的关注。原则上,数字免疫传感依赖于将目标混合物随机划分为大量微孔或微滴后对阳性信号进行统计分析(如泊松分布)。目前,数字免疫传感器已应用于癌症的早期筛查、低丰度蛋白生物标记物的检测和发现以及其他医学诊断等。

一种超灵敏数字化免疫传感方法,磁颗粒与初级抗体偶联;次级抗体利用单链信号DNA修饰;所述的初级抗体、次级抗体分别识别目标蛋白形成三明治结构的磁颗粒免疫复合物;
分离除去磁颗粒,得到游离的三明治结构的免疫复合物;通过液滴发生器将游离的三明治结构的免疫复合物加载到油包水(W/O)微滴中;
扩增单链信号DNA,产生荧光微滴;计数荧光微滴从而得到目标蛋白的含量。
所述的磁颗粒为金包磁纳米颗粒(Au@MNPs)。
所述的磁颗粒与初级抗体偶联使用两端分别带不同基团的双链链接DNA,磁颗粒与双链链接DNA之间形成金硫共价键,初级抗体和双链链接DNA之间形成酰胺键。

3)使用限制性核酸内切酶特异性酶切双链链接DNA,释放三明治结构的免疫复合物;
4)使用微滴发生器将游离的免疫复合物加载入微滴中,使得每个微滴中有且仅有一个三明治结构的免疫复合物;
5)对微滴进行数字PCR扩增产生荧光信号;
6)对荧光微滴和总微滴进行计数,从而得到目标蛋白的浓度或含量。
本发明的有益效果:
本发明将免疫PCR应用于微滴发生装置,实现了aM蛋白分子水平,甚至单分子水平的蛋白免疫检测。
所述的免疫传感方法减少了因Au@MNPs等固相载体引起的非特异性吸附;只有在连接和信号两部分同时存在时,数字化免疫传感才可以正常进行,进一步提高了该免疫传的特异性。
所述的免疫传感方法对双链核酸上的特异性位点酶切使得金包磁纳米颗粒和免疫复合物分离,这部分的双链核酸可以使用例如可控光化学响应等材料替换,更快更高效的释放免疫复合物。
所述的免疫传感方法可使用聚合酶链式反应的方法生成检测信号,核酸自身的多样性为蛋白的联检提供了更多的可能。
该免疫传感方法具有操作简单、成本低、适用性广、灵敏度高和特异性强等优势,可应用于多个领域,多个物种,尤其是对于体外诊断、筛查、生物标志物的发现等应用具有较高的价值。

具体实施方式
为了使本发明的目的、技术方案及优点更加清楚明白,以下结合附图对本发明做进一步阐述。
如图1所示,一种超灵敏数字化免疫传感方法,步骤包括:
(1)免疫复合物的形成
磁颗粒与初级抗体偶联;
次级抗体利用单链信号DNA修饰;
所述的初级抗体、次级抗体识别目标蛋白形成三明治结构的磁颗粒免疫复合物;
分离除去磁颗粒,得到游离的三明治结构的免疫复合物;
(2)微滴生成
通过液滴发生器将游离的三明治结构的免疫复合物加载到油包水(W/O)微滴中;
扩增单链信号DNA,产生荧光微滴;
(3)单分子计数
计数荧光微滴从而得到目标蛋白的含量。
优选的,所述的磁颗粒为金包磁纳米颗粒(Au@MNPs)。
优选的,所述的磁颗粒与初级抗体偶联使用两端分别带不同基团的双链链接DNA,磁颗粒与双链链接DNA之间形成金硫共价键,初级抗体和双链链接DNA之间形成酰胺键。
优选的,所述的单链信号DNA修饰的次级抗体使用生物素和链霉亲和素之间的相互共价结合。
优选的,所述的双链链接DNA使用限制性核酸内切酶特异性酶切的方法处理,释放三明治结构的免疫复合物。
优选的,所述的方法,通过液滴发生器将游离免疫复合物加载到油包水(W/O)微滴中,所述的液滴发生器类型包括离心式(如CN216688147U等)、微流控芯片式(如CN113275048A等)或振荡式(如CN108654706A等)。
优选的,所述的单链信号DNA扩增的方法包括实时荧光定量PCR、环介导的等温扩增。
优选的,所述的扩增单链信号DNA步骤中 ,DNA扩增染料包括双链DNA结合染料(如SYBR Green、ROX)或分子探针(如Tapman)。
所述的目标蛋白的来源包括未经处理或者预处理后的人的体液,所述的体液包括血清、尿液、脑脊液,或唾液。
实施例1
磁核颗粒的制备:将360 mg FeCl3·6H2O(1.33 mM)和131.94 mg MnCl2添加到20mL二甘醇(DEG)和乙二醇(EG)的混合物(1:1)中。搅拌20分钟直到溶液透明。加入2 g聚乙烯吡咯烷酮(PVP)粉末,在120 ℃下加热混合物直至PVP粉末完全溶解。随后,加入1.5 g乙酸钠,搅拌30分钟。将溶液转入反应釜中在205 ℃下反应10小时。反应完成后,用乙醇将合成的MnFe2O4磁性纳米颗粒洗涤5次,并在真空下干燥24小时,得到棕色粉末。
金种子纳米颗粒(~15 nm)的制备:对装有200 mL去离子水的锥形瓶加热,加入11.11 mL柠檬酸钠溶液(5 mM),1200 rpm将溶液加热至沸腾状态。加入2.22 mL氯金酸溶液(25 mM)搅拌5分钟。随后加入5 mL新鲜制备的NaBH4(100 mM),25 ℃下连续搅拌4小时。得到的产物在4℃下储存以备之后使用。
金包磁纳米颗粒(Au@MNPs)的形成:将金种子颗粒吸附在磁核的表面,通过金种子生长的方法使磁的表面附着生长金壳。将100 mg的MnFe2O4磁性纳米颗粒与200 mL的聚醚酰亚胺(PEI)溶液(5 mg/mL)相混合中,超声处理20分钟。向混合溶液中加入200 mL Au种子溶液,超声处理30分钟。随后将纳米颗粒磁力分离,用水洗涤五次。将产物分散到100 mL水中以获得1 mg/mL Auseeds-PEI@MnFe2O4。将250 mg PVP加入到5 mL(0.25 mg/mL)Auseeds-PEI@MnFe2O4溶液中使其完全溶解,再加入200 mL盐酸羟胺溶液,并将混合物超声处理3分钟。然后加入适量(~500 μL)的氯金酸溶液(25 mM)反应5分钟,生成Au@MnFe2O4。用磁分离和去离子水洗涤3次,并在4℃下储存。
通过扫描电子显微镜(SEM,Carl Zeiss AG,GeminiSEM 300)和动态光散射(DLS,Malvern Panalytical,Zetasizer Nano ZS)表征合成的纳米颗粒的形态、尺寸和Zeta电位。通过UV-Vis光谱仪(岛津株式会社,UV-2550)测量纳米颗粒的吸收峰情况。
纳米颗粒的表征:如图2的(1)-(2)部分所示,合成的金包磁纳米颗粒呈球形,在Au涂覆前测得的直径为~130 nm,金壳形成后直径增加至~160 nm。动态光散射测量结果表明Au@MNPs平均尺寸约为700 nm(图2的(3)部分),这可能是由于纳米颗粒的聚集导致。纳米颗粒的紫外-可见吸收光谱如图2的(4)部分所示。在金壳形成后,Au@MNPs在~540nm处显示出一个新的吸收峰,这在Au涂覆前的纳米颗粒中未观察到。从SEM结果可以看出,不光滑的金壳可能导致相邻Au纳米颗粒的耦合和吸收峰的红移。
DNA双链的形成:使用DNA退火缓冲液(5×)对单链DNA-SH和DNA-COOH进行退火反应以产生双链DNA(SH-DNA-COOH)。用退火缓冲液对两条DNA寡核苷酸单链进行稀释以达到相同的摩尔浓度(5 μM),反应条件为:在95℃下变性2分钟;每90秒降低1℃,然后慢慢将温度降至25℃(整个冷却过程至少持续45分钟)。将获得的双链DNA(SH-DNA-COOH)在4℃下短暂储存,用于之后的实验。
其中,本实施例中使用的DNA 序列如下表1所示。

用终浓度为10mM的三(2-羧乙基)膦(TCEP)对双链DNA一端的巯基基团进行活化,室温下避光孵育20分钟。使用CENTRI·SPIN-10纯化柱对活化后的双链DNA进行纯化以去除多余的TCEP。纯化过程应至少进行三次。
免疫传感样本蛋白的捕获:使用上述制备的SH-DNA-COOH双链对Au@MNPs和初级抗体偶联:核酸双链一端使用活化的巯基基团连接Au@MNPs,室温孵育4h,磁选分离并用去离子水清洗多次除去未结合的核酸双链;另一端的羧基使用终浓度为10 mM的1-乙基-(3-二甲基氨基丙基)碳酰二亚胺(EDC)和N-羟基丁二酰亚胺(NHS)进行活化30min。磁选分离并用MEST(0.1M,pH 5.0)缓冲液清洗多次除去多余的EDC和NHS。随后与初级抗体的胺基连接,室温孵育2h。初级抗体偶联的Au@MNPs复合物经磁选分离洗涤。这里使用的双链链接DNA可以使用可控光化学响应等材料替换,链接金包磁纳米颗粒和初级抗体。使用磁珠封闭液对初级抗体偶联的Au@MNPs复合物封闭处理45 min。随后加入不同浓度梯度的α-突触核蛋白样本,室温孵育1h。用磁选分离、PBST洗涤以除去未结合的α-突触核蛋白。加入信号DNA标记的次级抗体去连接被捕获到的α-突触核蛋白,室温孵育20分钟。用磁选分离、1×PBS缓冲液(pH 7.2)洗涤除去未结合的次级抗体。次级抗体上修饰的信号DNA自身序列的多样性为多重蛋白的联检提供了更多的可能。由于磁纳米颗粒在离心力的作用下会产生不可逆的聚集,因此我们使用酶切的方法去除磁性纳米颗粒。将上述结合的Au@MNPs免疫复合物重悬到EcoR Ⅰ 酶切反应体系中:10×EcoR Ⅰ Buffer(5μL),EcoR Ⅰ 105U/mL(1μL),H2O(44μL),37℃ 1h进行水浴酶切反应。使用磁选分离除去Au@MNPs得到包含游离免疫复合物的上清液。游离的免疫复合物短暂保存于4℃,用于后续实验。
免疫PCR:将得到的免疫复合物(5μL)加入到PCR反应体系中:2×SYBR Green qPCRMaster Mix(10μL),10μM Primer 1(0.4μL),10μM Primer 2 (10μM)(0.4μL),免疫复合物(5μL),H2O(4.2μL),混合均匀。随后将10μL反应混合物加载到微滴发生装置中,6000g转速离心5分钟生成液滴,将离心管底部的液滴转移至PCR仪(Bio-Rad CFX96)中进行扩增,扩增程序:25℃ 10 min;94 ℃ 30 s;94 ℃ 5 s,60 ℃ 45 s,40×。本发明中使用的油相为7%Abil EM 180+棕榈酸异丙酯,在高温下,油包水微滴可以保持稳定状态。
荧光液滴计数:将微滴转移到玻璃板上,然后使用配备有激发滤光片(BP460~495)和发射滤光片(BA510~550)的IX73倒置荧光显微镜进行观察。使用image J对所有微滴以及荧光微滴进行计数。以α-突触核蛋白在缓冲液或血清中的浓度为横坐标,以相对应的荧光微滴数量的占比为纵坐标建立标准曲线和定量线。使用H2O作为空白对照组。每次至少进行三次测定。
免疫传感α-Syn检测:如图3所示,通过对扩增后的荧光微滴计数,在0.1-1000 fM动态范围内观察到α-突触核蛋白在缓冲液中的浓度与荧光微滴的比例呈现线性相关,线性相关系数>0.99。其中,检测限为43.36 aM,定量限为99.59 aM。如图4所示,液滴数字化免疫传感在血清样本中的性能与在缓冲液中相当。当α-突触核蛋白浓度在0.5-1000 fM时观察到线性关系,线性相关系数为0.9904。在血清环境中的检测限LOD为173.02 aM,定量检测限LOQ为660.91 aM。
实施例2
本发明的超灵敏数字化免疫传感方法的特异性检测。
本发明使用α-突触核蛋白的异构体β-突触核蛋白、γ-突触核蛋白和其他血液中存在的蛋白:白介素-6(IL-6)、白介素-10(IL-10)、肿瘤坏死因子α(TNF-α)、人类白细胞抗原(HLA-DR)和人脂多糖蛋白(LBP)评估液滴数字化免疫传感的特异性,所有的步骤与实施例1的步骤一致。如图5所示,液滴数字化免疫传感对α-突触核蛋白表现出高的特异性。
实施例3
本发明提出了离心式数字化免疫传感技术对磷酸化α-突触核蛋白ser129进行检测。
本发明通过对α-突触核蛋白磷酸化Ser129位点进行检测,使用重组抗α-突触核蛋白 (磷酸化 S129)抗体作为初级抗体进行液滴数字化免疫传感,所有的步骤与上述实施例1的步骤基本保持一致。如图6所示,磷酸化的α-突触核蛋白ser129检测的检测限LOD为147.23 aM,定量检测限LOQ为1.567 fM。
上述描述中的实施方案可以进一步组合或者替换,且实施方案仅仅是对本发明的优选实施例进行描述,并非对本发明的构思和范围进行限定,在不脱离本发明设计思想的前提下,本领域普通技术人员对本发明的技术方案做出的各种变化和改进,均属于本发明的保护范围。本发明的保护范围由所附权利要求及其任何等同技术方案给出。
Claims (10)
1.一种超灵敏数字化免疫传感方法,其特征在于:
磁颗粒与初级抗体偶联;
次级抗体利用单链信号DNA修饰;
所述的初级抗体、次级抗体分别识别目标蛋白形成三明治结构的磁颗粒免疫复合物;
分离除去磁颗粒,得到游离的三明治结构的免疫复合物;
通过液滴发生器将游离的三明治结构的免疫复合物加载到油包水(W/O)微滴中;
扩增单链信号DNA,产生荧光微滴;计数荧光微滴从而得到目标蛋白的含量。
2.根据权利要求1所述的方法,其特征在于:所述的磁颗粒为金包磁纳米颗粒(Au@MNPs)。
3.根据权利要求1所述的方法,其特征在于:所述的磁颗粒与初级抗体偶联使用两端分别带不同基团的双链链接DNA,磁颗粒与双链链接DNA之间形成金硫共价键,初级抗体和双链链接DNA之间形成酰胺键。
4.根据权利要求1所述的方法,其特征在于:所述的单链信号DNA修饰的次级抗体使用生物素和链霉亲和素之间的相互共价结合。
5.根据权利要求3所述的方法,其特征在于:所述的双链链接DNA使用限制性核酸内切酶特异性酶切的方法处理,释放三明治结构的免疫复合物。
6.根据权利要求1所述的方法,其特征在于:通过液滴发生器将游离免疫复合物加载到油包水(W/O)微滴中,所述的液滴发生器类型包括离心式、微流控芯片式或振荡式。
7.根据权利要求1所述的方法,其特征在于:所述的单链信号DNA扩增包括实时荧光定量PCR、环介导的等温扩增;DNA扩增染料包括双链DNA结合染料或分子探针。
8.根据权利要求1所述的方法,其特征在于:所述的目标蛋白的来源包括动物、植物、
微生物,或者人工产物。
