背景技术
抗生素的发现在过去75年中挽救了无数的生命。然而,抗生素时代的黄金时代正在逐渐消逝,我们现在正在进入后抗生素的黑暗时代。每年,仅在美国,由多重耐药性ESKAPE病原体(屎肠球菌(Enterococcusfaecium)、金黄色葡萄球菌(Staphylococcusaureus)、肺炎克雷伯氏菌(Klebsiella pneumoniae)、鲍氏不动杆菌(Acinetobacterbaumannii)、铜绿假单胞菌(Pseudomonas aeruginosa)和肠杆菌(Enterobacter)属物种)引起的医院获得性感染超过200万例,造成了估计200亿美元的过度医疗保健成本。这些ESKPE病原体已经进化为产生碳青霉烯酶(carbapenemase,KPC)的肺炎克雷伯氏菌、铜绿假单胞菌、耐甲氧西林金黄色葡萄球菌(MRSA)和耐万古霉素肠球菌(VRE),对这些病原体没有治疗选择。尽管目前存在抗菌剂耐药性危机,但因为开发新的抗菌剂的平均成本约为8千亿美元,并且开发所需的时间为10年或更长,所以主要制药公司都不愿开发新的抗菌剂。而且,病原体很快会对新的抗菌剂产生抗性。鉴于这些问题,本发明内容不是开发新的抗菌剂,而是涉及一种有用的选择,即通过提高它们的功效和扩展它们的活性谱来强化目前存在的抗菌剂。
发明内容
本发明提供了药物缀合物、药物组合物,和通过将抗生素与氟化的磷酸己糖进行缀合并通过磷酸己糖转运蛋白(UhpT)的摄取(uptake)将这些缀合的抗生素递送到细菌来改善药物如抗生素的一种或多种药代动力学性质的方法,以及通过将抗菌剂如抗生素与氟化的磷酸己糖进行缀合而使用磷酸己糖转运蛋白来递送药物如抗菌剂的方法。
在一个实施方案中,本发明提供了细菌UhpT表达受磷酸己糖调节的机制。
在另一个实施方案中,本发明使用氟化的磷酸己糖作为载体分子以通过UhpT转运抗菌剂来改善抗菌剂如抗生素的一种或多种药代动力学性质。
在另一个实施方案中,本发明提供了一种合成不可代谢的氟化的磷酸己糖的方法,该不可代谢的氟化的磷酸己糖稳定地诱导UhpT的高水平表达。
在一个具体实施方案中,本发明提供了缀合药物,其包括与磷酸己糖或氟化的磷酸己糖缀合的药物。
本发明的药物组分可以是抗菌剂如抗生素的缀合物(conjugate)。抗生素可以优选地选自利奈唑胺(linezolid)和磷霉素(fosfomycin)。

图1说明了磷酸己糖(HP)调控UhpT表达的机制。HptA膜蛋白感知细胞外的磷酸己糖,磷酸己糖诱导HptS的自磷酸化(autophosphorylation)。随后,HptS使HptR磷酸化,这引起HptR的二聚化。然后二聚化的HptR与uhpT基因的启动子区(promoter region)结合并诱导UhpT的表达以促进UhpT对磷酸酯(phosphates)的摄取。

图3示出了合成氟化的磷酸己糖以防止合成的磷酸己糖被磷酸己糖脱氢酶代谢的化学-酶促方法(chemo-enzymatic method)的实例。
图4示出了3-氟-葡萄糖-6-磷酸(3FG6P)不被原代犬膀胱上皮细胞代谢。将3FG6P(500μM)重悬于RPMI1640细胞培养基中,并在存在和不存在原代犬膀胱细胞的情况下孵育24小时。将培养基的甲醇提取物在氮气下蒸发至干,然后在100μL的1:1(v/v)乙腈/25mM甲酸铵水溶液中重构(reconstituted)。将每个样品以2μL注射到与Waters UPLC和ThermoQuantum三重四极杆质谱仪(Thermo Quantum triple-quadrupolemassspectrometer)(电喷雾电离(electrospray ionization))连接的2.1mm×100mmHILIC柱上。在整个色谱运行中监测3FG6P(m/z 261.0>79.3)的质量转变(mass transition)(参见图)。没有观察到膀胱细胞代谢3FG6P的证据。
图5说明了用于通过测量生物发光的光信号以及3FG6P对UhpT表达的强而稳定的诱导来监测UhpT的表达的LuxABCDE报告系统(LuxABCDE reporter system)。在补充有500μM葡萄糖(Glc)、葡萄糖-6-磷酸(G6P)和3-氟-葡萄糖-6-磷酸(3FG6P)的脑心浸液肉汤(brain heartinfusion broth)中培养含有(harboring)LuxABCDE报告系统的金黄色葡萄球菌(S.aureus)LAC菌株,并使用Cytation5监测生物发光信号。虽然G6P暂时诱导生物发光信号,但3FG6P诱导组成型生物发光信号(constitutivebioluminescent signal)。葡萄糖不诱导生物发光信号。这些结果表明不可代谢的3FG6P稳定激活HptARS系统并诱导UhpT的表达。

具体实施方式
对抗菌剂的耐药性由于两种主要机制而产生。一种机制涉及通过基因突变修饰抗生素作用的靶标。另一种机制通过表达抗生素外排泵(effluxpumps)、降低膜的渗透性和/或破坏抗生素来阻止抗生素以足够高的浓度达到其靶标。前一种机制只能通过开发可以作用于新靶标的新抗生素来解决。后一种机制可以通过改善本发明中抗生素的一种或多种药代动力学性质来解决。
如本文所用,术语“磷酸己糖(hexose phosphate)”可以具体指磷酸己糖或一般指磷酸己糖和氟化的磷酸己糖。
如本文所用,“药代动力学性质(pharmacokinetic properties)”是指药物递送、药物吸收、药物分布、药物代谢和药物排泄中的一种或多种。
最近,对细菌基因调控系统(HptARS)进行了表征。HptARS控制来自金黄色葡萄球菌的摄取磷酸己糖的转运蛋白(Uptake of Hexose PhosphateTransporter,UhpT)的表达(图1)。HptA是感知细胞外磷酸己糖(HP)的膜蛋白。HptA对HP的识别会诱导HptS随后HptR的顺序磷酸化(sequentialphosphorylation)。HptR是转录调控因子(transcriptionalregulator),其与UhpT基因的启动子区结合并诱导UhpT的表达以促进微生物如细菌将磷酸化的己糖(phosphorylated hexose)分子摄取到其中。
重要的是,细菌基因组序列分析示出UhpT系统在包括ESKAPE病原体在内的许多革兰氏阳性和革兰氏阴性病原体中是高度保守的。由于某些抗生素对此类病原体的疗效低下和/或窄谱活性,已经不鼓励使用它们,而这些发现使我们开发了一种概念,即利用HptARS和UhpT系统来转运上述抗生素。更具体地,这些抗生素与磷酸己糖(HP)或氟化的磷酸己糖缀合以提供磷酸己糖缀合抗生素(AB-HP)。磷酸己糖缀合抗生素激活HptARS系统并诱导UhpT的表达。这有利于通过UhpT摄取AB-HP。缺乏磷酸己糖部分的非缀合的抗生素(AB)由于缺乏合适的转运系统而无法通过UhpT或以其他方式转运到细菌中。与磷酸己糖或氟化的磷酸己糖的这种缀合增加了此类抗生素的功效并扩展了它们的抗菌活性谱。与磷酸己糖或氟化的磷酸己糖的这种缀合适用于许多抗生素,这些抗生素由于它们目前的疗效低下和/或窄谱的抗菌活性而被放弃或不鼓励使用。
本发明涉及抗菌剂如抗生素通过接头(linker)与磷酸己糖部分的缀合。可以选择相对容易与药物缀合的接头,以确保与多种药物如抗生素分子的通用用途。接头可以是可切割接头(cleavable linker)或不可切割接头(non-cleavable linker)。优选地,接头是可切割接头,因为在许多情况下,抗生素分子一旦进入细菌体内就需要释放以提供所需效果。然而,根据抗生素分子的SAR,可以使用不可切割的接头以提供提高的稳定性。
下表1示出了本发明的示例性合适抗生素和可切割接头的列表。

缀合药物的第三个要素是葡萄糖-6-磷酸(G6P)单元,其优选被稳定化以防止被代谢。3FG6P是具有足够活性以及对代谢具有抗性的合适部分的一个实例。也可以使用4FG6P和其他FG6P。合适的氟化的磷酸己糖还包括3-脱氧-3,3-二氟葡萄糖(A)、2-脱氧-2-氟葡萄糖(B1)、2-脱氧-2-氟葡萄糖的2,2-二氟化衍生物(B2)、2,3-双脱氧-2,3-二氟葡萄糖(C1)和2,3-双脱氧-2,2,3,3-四氟化类似物(C2)。在1-位处的脱氧将阻止糖酵解和戊糖磷酸途径,因此1DG6P(D1)和对应的3-氟化类似物(D2)也可能是合适的,如同4-脱氧-4-氟葡萄糖(E)一样。
用于合成多氟化化合物的合适方法可在文献中获得。己糖激酶可以使单氟化葡萄糖磷酸化。对于不能被己糖激酶转化的底物,可以使用化学磷酸化。
第二类合适的类似物包括6-磷酸基团(6-phosphate group)(F)。糖环中的氟化(Fluorination)不影响代谢的第三途径,即磷酸基团(phosphate group)的水解。这可以通过用亚甲基(F1)或氟化亚甲基(F2)取代磷酸(phosphate)的桥接氧来合成不可水解的磷酸酯类似物(phosphate analogs)来解决。
当取代基在下式中表示为“F/OH”时,这表示取代基可以是-F或-OH,条件是这些式子的每个化合物都必须具有至少一个为-F的取代基。

在其他方面,本发明提供了通过将抗生素与氟化的磷酸己糖进行缀合并通过摄取磷酸己糖转运蛋白(UhpT)将这些缀合抗生素递送至细菌来改善药物如抗生素的一种或多种药代动力学性质的方法,以及通过将抗菌剂如抗生素与磷酸己糖或氟化的磷酸己糖进行缀合而使用磷酸己糖转运蛋白递送药物如抗菌剂的方法。上文描述了通过将利奈唑胺或磷霉素与3-氟-葡萄糖-6-磷酸或4-氟-葡萄糖-6-磷酸进行缀合来改善利奈唑胺和磷霉素的一种或多种药代动力学性质的方法的实例。这些方法可用于提高利奈唑胺和磷霉素的抗菌活性。
本发明还提供了细菌UhpT的表达受磷酸己糖调控的机制。

实施例
实施例1:3-氟-葡萄糖-6-磷酸的合成
在第一步中,合成3-氟-葡萄糖-6-磷酸,由此葡萄糖环第三个碳上的羟基被氟原子取代。这种合成的第一步是制备3-氟-葡萄糖。在该步骤中,1,2:5,6-二-O-异亚丙基-α-D-呋喃葡萄糖与(二乙基氨基)三氟化硫(DAST)在二氯甲烷中反应,在3’-位置产生立体特异性氟化(stereospecificfluorination),得到3-氟-1,2:5,6-二-O-异亚丙基-α-D-呋喃葡萄糖。然后将3-氟-1,2:5,6-二-O-异亚丙基-α-D-呋喃葡萄糖与三氟乙酸(TFA)反应以除去异亚丙基保护基,从而产生3-氟-葡萄糖。通过快速色谱法纯化3-氟-葡萄糖,并进行酶促磷酸化以形成3-氟-葡萄糖-6-磷酸。在这个反应中,己糖激酶将磷酸基团从ATP转移到3-氟-葡萄糖的6’-OH基团(图3)。然后通过配备有阴离子交换柱的HPLC纯化产物3-氟-葡萄糖-6-磷酸,并冻干直至使用。
为了测试3-氟-葡萄糖-6-磷酸(3FG6P)是否被宿主细胞代谢,将3FG6P(500μM)重悬于RPMI1640细胞培养基中,并在存在和不存在原代犬膀胱细胞的情况下培养24小时。将含有x的培养基的甲醇提取物在氮气下蒸发至干,然后在100μL的1:1(v/v)乙腈/25 mM甲酸铵水溶液中重构。将每个样品以2μL注射到与Waters UPLC和Thermo Quantum三重四极杆质谱仪(电喷雾电离)连接的2.1mm×100mm HILIC柱上。在整个色谱运行中监测3FG6P的质量转变(m/z 261.0>79.3)。在存在犬膀胱上皮细胞的情况下,细胞培养基中3FG6P峰值的强度与不存在犬膀胱上皮细胞的同一细胞培养基的峰值没有差异,这表明3FG6P不被膀胱上皮细胞代谢(图4)。
实施例2:3-氟-葡萄糖-6-磷酸对UhpT的影响
为了测试3FG6P是否能诱导UhpT的表达,我们生产了一种生物发光报告质粒(reporter plasmid),该质粒中uhpT的启动子区与无启动子的LuxABCDE操纵子融合(图5)。参见Francis KP,Joh D,Bellinger-Kawahara C,Hawkinson MJ,Purchio TF,ContagPR.Monitoring bioluminescent Staphylococcus aureus infections in living miceusing a novel luxABCDE construct.Infection and immunity.2000;68(6):3594-600.Epub 2000/05/19.PubMed PMID:10816517;PMCID:PMC97648和Karsi A,LawrenceML.Broad host range fluorescence and bioluminescence expression vectors forGram-negative bacteria.Plasmid.2007;57(3):286-95.Epub 2007/01/09.doi:10.1016/j.plasmid.2006.11.002.PubMed PMID:17207855,用于通过测量生物发光信号来监测UhpT的表达。葡萄糖-6-磷酸暂时诱导生物发光信号持续8小时,而3FG6P诱导显著更高的生物发光信号持续18小时以上(图5)。正如预期的那样,葡萄糖没有诱导任何生物发光信号。这些结果表明,3FG6P稳定而强烈地激活HptARS系统并诱导了UhpT的表达。
要使用3FG6P作为载体分子将抗生素转运到细菌中,即使在与抗生素缀合后,3FG6P也必须被UhpT识别。为了验证这一点,我们产生了缺乏uhpT基因的金黄色葡萄球菌LAC(ΔUhpT),其结果是不能转运磷酸己糖。含有3FG6P(500μM)的PBS在金黄色葡萄球菌LAC野生型(WT)菌株和金黄色葡萄球菌LACΔUhpT菌株存在下培养6小时,并在不存在金黄色葡萄球菌(PBS对照)的情况下培养6小时。3FG6P的浓度使用如上所述的与Waters UPLC和Thermo Quantum三重四极杆质谱仪连接的HILIC柱来测定。当与金黄色葡萄球菌LAC野生型(WT)菌株孵育2小时时,3FG6P浓度迅速降至PBS对照的约40%,并在6小时内从PBS中完全消失(图6)。相比之下,当与金黄色葡萄球菌LACΔUhpT菌株一起培养时,100%的3FG6P保留2小时,然后3FG6P的浓度在6小时内逐渐降低到PBS对照的约60%。这些结果表明,3FG6P通过UhpT有效地转运到金黄色葡萄球菌WT菌株中。
总之,这些结果表明,氟化的磷酸己糖(3FG6P)能够诱导UhpT的稳定和强表达,并且尽管进行了氟修饰,氟化的磷酸己糖(3FG6P)仍然被UhpT有效识别并转运到细菌细胞中。这些结果证明了氟化的磷酸己糖可以用作载体分子,通过UhpT将抗生素转运到细菌细胞中的观点。这将使因目前疗效低下和/或窄谱的抗菌活性而被放弃的抗生素能够被重新使用。
实施例3:通过与3FG6P缀合来扩大利奈唑胺的抗菌活性
利奈唑胺是3-芳基-2-噁唑烷酮(3-aryl-2-oxazolidinones)家族的成员,其具有连接到噁唑烷酮环的5位的乙酰氨基甲基和在苯基的3位的氟取代。如本文所用,利奈唑胺是指(S)-N-[[3-[3-氟-4-(N-1-哌嗪基)苯基]-2-氧代-5-噁唑烷基]甲基]乙酰胺。利奈唑胺在很早的阶段就抑制细菌核糖体蛋白的合成。利奈唑胺与50S核糖体亚基的23S结合,从而阻止与30S亚基、fMet-RNA、起始因子(initiation factors)IF2和IF3以及mRNA形成功能性70S起始复合物(initiation complex)。
利奈唑胺对所有临床上重要的革兰氏阳性菌,如MIC90为1-4μg/ml和2μg/ml的耐甲氧西林金黄色葡萄球菌(methicillin-resistant Staphylococcus aureus,MRSA)、MIC90为1-4μg/ml和1μg/ml的表皮葡萄球菌(Staphylococcus epidermidis,MRSE)、MIC90为1-4μg/ml和2μg/ml的耐万古霉素肠球菌(vancomycin-resistant Enterococcus,VRE)中的粪肠球菌和屎肠球菌有效。然而,利奈唑胺由于其快速外排机制(efflux mechanisms),对需氧革兰氏阴性病原体的效果较差。利奈唑胺对不动杆菌属(Acinetobacter spp)、大肠杆菌、肺炎克雷伯氏菌、彭氏变形菌(Proteus penneri)、铜绿假单胞菌和嗜麦芽寡养单胞菌(Stenotrophomonas maltophilia)没有活性。利奈唑胺对流感嗜血杆菌(Haemophilusinfluenzae)和淋病奈瑟菌(Neisseria gonorrhea)的活性最低,MIC90为16μg/ml。参见SeeJones RN,Johnson DM,Erwin ME.In vitro antimicrobial activities and spectra ofU-100592 and U-100766,two novel fluorinated oxazolidinones.Antimicrob AgentsChemother 1996;40(3):720-726和Zhanel GG,Karlowsky JA,Low DE,HobanDJ.Antibiotic resistance in respiratory tract isolates of Haemophilusinfluenzae and Moraxella catarrhalis collected from across Canada in 1997-1998.J Antimicrob Chemother 2000;45(5):655-66。
与3FG6P缀合的利奈唑胺的合成
为了证明氟化的磷酸己糖可以改善利奈唑胺的功效和/或抗菌活性谱,将利奈唑胺与3FG6P缀合。与3FG6P(17)缀合的利奈唑胺的合成分三部分进行,首先由3,4-二硝基苯(1)经一系列8步合成利奈唑胺部分(S)-N-[[3-[3-氟-4-(N-1-哌嗪基)苯基]-2-氧代-5-噁唑烷基]甲基]乙酰胺(11)。3,4-二硝基苯(1)首先与哌嗪反应得到1-(2-氟-4-硝基苯基)哌嗪,1-(2-氟-4-硝基苯基)哌嗪随后反应得到N-保护的(protected)衍生物(5)。然后使用正丁基锂(n-BuLi)将N-保护的衍生物(5)锂化,随后与丁酸(R)-缩水甘油酯(6)反应以获得噁唑烷基衍生物(7),然后将噁唑烷基衍生物(7)与对甲苯磺酰氯反应以提供O-甲苯磺酰化产物(8)。使O-甲苯磺酰化产物(8)与邻苯二甲酰亚胺钾进行取代反应,得到(R)-N-[[3-[3-氟-4-[N-1-(4-苄氧羰基)哌嗪基]-苯基]-2-氧代-5-噁唑烷基]甲基]邻苯二甲酰亚胺(9)。将(9)脱保护为伯胺,并用乙酸酯进一步保护,得到N-乙酰基产物(10),该产物用H2和钯碳脱保护,得到图7中所示的所需产物(11)。
在4-二甲氨基吡啶(DMAP)碱的存在下,通过3-氟-葡萄糖与氯磷酸二苯基酯反应,合成3-氟-葡萄糖-6-苯基化磷酸部分(13)(图8)。
然后将产物(11)与γ-丁内酯反应转化为中间体酰胺(14),中间体酰胺(14)随后通过酸催化的糖基化反应与3-氟-葡萄糖-6-苯基化磷酸部分(13)偶联,以得到产物(15)。最终脱保护产生所需的产物(16)(图9)。
实施例4:利奈唑胺和与3FG6P缀合的利奈唑胺的体外抗菌活性
为了测试利奈唑胺和与3FG6P缀合的利奈唑胺(Lzd-3FG6P)对革兰氏阳性和革兰氏阴性病原体的抗菌活性,使用的病原体为金黄色葡萄球菌ATCC 25923、肺炎克雷伯氏菌ATCC 35657、鲍氏不动杆菌ATCCBAA1605、大肠杆菌ATCC 25922、阴沟肠杆菌ATCC 13047、产气肠杆菌ATCC13048和鼠伤寒沙门氏菌ATCC 14028。根据临床和实验室标准研究所(Clinical and Laboratory Standards Institute,CLSI)文件M07-A9(Clinical andLaboratory Standards Institute.2012.M07-A9.Methods for dilution antimicrobialsusceptibility tests for bacteria that grow aerobically:approved standard,9thed.Clinical and Laboratory Standards Institute,Wayne,PA)的说明,使用液体培养基微量稀释法测定最小抑制浓度(minimal inhibitory concentration,MIC)。简单地说,这些菌株在MullerHinton肉汤(MHB)中生长,直到指数期(OD600<1.0)。指数生长的测试细菌在MHB中稀释到OD600为0.01,并在96孔板中等分。在DMSO中以10mg/ml的浓度制备利奈唑胺(Lzd)和与3FG6P缀合的利奈唑胺(Lzd-3FG6P)的贮备液(Stocks),通过从64μg/ml到2μg/ml(最终浓度)的两倍系列稀释进行稀释,并添加到96孔板中。包括DMSO对照(图中为0μg/ml)。将板在37℃下孵育,并通过使用Cytation 5板读取器(BioTek公司)测量OD600来监测细菌的生长。
虽然利奈唑胺成功抑制了MIC低于2μg/ml的革兰氏阳性病原体金黄色葡萄球菌ATCC 25923的生长,但即使在64μg/ml的浓度下,利奈唑胺也无法抑制革兰氏阴性病原体的生长(图10)。相比之下,与3FG6P(Lzd-3FG6P)缀合的利奈唑胺在2-8μg/ml的浓度范围内成功抑制了革兰氏阳性和革兰氏阴性病原体的生长(图10和下表2)。这些结果清楚地表明,利奈唑胺的抗菌活性通过与3FG6P缀合而扩展到革兰氏阴性病原体。

实施例5:利奈唑胺和与3FG6P缀合的利奈唑胺的体内抗菌活性
为了测试利奈唑胺和与3FG6P缀合的利奈唑胺的体内抗菌活性,使用pLuxABCDE质粒对临床大肠杆菌菌株进行构建,使其组成型地表达生物发光信号。参见Karsi A,Lawrence ML.Broad host range fluorescence and bioluminescence expressionvectors for Gram-negative bacteria.Plasmid.2007;57(3):286-95.Epub 2007/01/09.doi:10.1016/j.plasmid.2006.11.002.PubMed PMID:17207855,用于实时监测感染进展。从哈兰实验室(Harlanlaboratory)购买6至8周龄雌性C57BL/6小鼠,并根据密西西比州立大学(Mississippi State University)机构动物护理和使用委员会批准的方案进行饲养和维护。用悬浮在PBS中的50μl生物发光大肠杆菌菌株(2×109CFU/ml)经尿路感染动物(n=6/组)。在感染后2、24和48小时,用腹膜内注射利奈唑胺(80mg/kg)或与3FG6P缀合的利奈唑胺(80mg/kg)或PBS对照处理动物。通过使用IVIS Lumina XR小动物成像系统测量生物发光信号来监测感染的进展。感染72小时后,对动物实施人道安乐死,并收集膀胱和肾样本,匀浆并连续稀释,将连续稀释液平板涂布在血液琼脂上以确定细菌负荷。在没有抗生素处理(PBS对照组)的情况下,经尿道接种大肠杆菌在肾和膀胱中建立了持续感染。肾和膀胱中的平均细菌计数分别为log106.328±0.132和log106.171±0.155 CFU/g(图12)。利奈唑胺治疗并没有减少肾和膀胱中的细菌计数,而与3FG6P缀合的利奈唑胺(Lzd-3FG6P)处理完全清除了肾脏的感染,几乎完全清除了膀胱的感染。这些结果清楚地表明,3FG6P的缀合使利奈唑胺对控制由大肠杆菌引起的尿路感染非常有效。

为了测试3FG6P和4FG6P是否能增强磷霉素的功效,获得了携带fosA基因的临床耐磷霉素UTI大肠杆菌分离株和金黄色葡萄球菌COL菌株。使这些细菌的过夜培养物在稀释至0.5 McFarland浊度的脑心浸液(BHI)肉汤中生长,并接种到补充有不同浓度的单独磷霉素的新鲜BHI肉汤或对于大肠杆菌而言,补充有3FG6P(50μM)的磷霉素的新鲜BHI肉汤;或对于金黄色葡萄球菌而言,补充有4FG6P(50μM)的磷霉素的新鲜BHI肉汤。
如图13A至图13D所示,耐磷霉素的大肠杆菌和耐磷霉素的金黄色葡萄球菌都允许在单独补充高达128μg/ml磷霉素的BHI中生长。相比之下,在补充了50μM 3FG6P或4FG6P的BHI中,这两种细菌的生长在低浓度的磷霉素下都受到显著抑制。特别是,使用50μM的3FG6P,即使使用2μg/ml的磷霉素,大肠杆菌的生长也会被完全抑制。同样,32μg/ml磷霉素可完全抑制耐磷霉素金黄色葡萄球菌的生长,而在较低浓度时可显著延迟生长。
这些结果清楚地表明,通过激活三组分调控系统诱导UhpT表达,显著增强了磷霉素的功效,达到足以逆转磷霉素修饰酶的抗性机制的水平。
实施例7:磷霉素和磷霉素与3FG6P共同给药的体内抗菌活性
为了测试磷霉素和磷霉素与4FG6P共同给药的体内抗菌活性,使用pLuxABCDE质粒产生组成型地表达生物发光信号的金黄色葡萄球菌COL菌株,用于实时监测感染进展。从哈兰实验室(Harlan laboratory)购买6至8周龄雌性C57BL/6小鼠,并根据密西西比州立大学(Mississippi State University)机构动物护理和使用委员会批准的方案进行饲养和维护。
用悬浮在PBS中的50μl生物发光金黄色葡萄球菌菌株(2×109CFU/ml)腹膜内感染动物(n=3/组)。在感染后2小时和24小时,用腹膜内注射单独的磷霉素(3 mg/kg)或磷霉素与4FG6P(50μg/kg)共同给药或PBS对照处理动物。通过使用IVIS Lumina XR小动物成像系统测量生物发光信号来监测感染的进展。
感染48小时后,对动物实施人道安乐死,并测定肺、肾、肝、脾和腹膜灌洗液中的细菌负荷。单独用PBS或磷霉素处理的动物的平均细菌计数范围分别为5.34±0.154 CFU/g至6.47±0.115CFU/g。相比之下,磷霉素与4FG6P共同给药的处理完全清除了腹膜灌洗、肺和肾的感染,并显著降低了肝和脾的细菌负荷,超过4个对数刻度(图14)。

14.一种将3-氟-葡萄糖-6-磷酸部分与利奈唑胺部分缀合的方法,所述方法包括以下步骤:

18.根据权利要求17所述的方法,其中,所述磷酸己糖或所述氟化的磷酸己糖选自3-氟-葡萄糖-6-磷酸、4-氟-葡萄糖-6-磷酸、3-脱氧-3,3-二氟葡萄糖、2-脱氧-2-氟葡萄糖、下式(B2)的2-脱氧-2-氟葡萄糖的2,2-二氟化衍生物:
2,3-双脱氧-2,3-二氟葡萄糖、下式(C2)的2,3-双脱氧-2,2,3,3-四氟化类似物:
