Description
Technical Field
The invention relates to enterococcus faecium JYEF-355 for promoting cat hair removal and application thereof, and belongs to the technical field of microorganisms.
Background
Since cats are habitual to licking Mao Shumao and have many barbs on the tongue, hair can not be spit out and can be rolled into the abdomen and gradually clustered into a small sphere. Some of these undigested hairs are expelled with the feces, and some are expelled from the mouth in the form of vomiting. However, a great amount of nondigestible hair is involved in the digestive tract of the cat and when a great amount of hair gathers to form a bulb which is difficult to discharge, the intestinal tract is blocked, so that the cat has symptoms of mental depression, inappetence, sleepiness, vomiting, abdominal distension, abdominal pain, constipation and the like, and the healthy growth of the cat is seriously affected, if the cat is not treated in time, the cat life is even threatened.
In order to prevent or treat hair bulb diseases and help cats discharge hair from the body, hair bulb products such as hair melting paste, hair melting chips, catfish and the like need to be fed to the cats regularly to assist the cats in discharging hair bulbs from the body.
The hair-dissolving paste mainly enables hair to slide out of the alimentary canal through the lubrication action of grease, simultaneously can also cause nutrient substances to slide out together, is easy to cause malnutrition of cats, affects the absorption of nutrient substances by intestines and stomach after long-term eating, and can possibly cause diarrhea; the hair-dissolving tablet contains a large amount of food-calling components and filling agents, has few effective components, can cause a cat to ingest a large amount of unnecessary filling agents when being eaten frequently, causes a certain burden on intestines and stomach, causes the weakening of stomach function when being eaten for a long time, and is not beneficial to the health of the cat; the catgrass is not convenient enough to obtain, the palatability is general, some cats do not like to eat plants, the problems of insufficient feed intake, poor hair removal effect and the like can be caused, and the catgrass mainly promotes the cats to discharge hair balls in a vomit mode, so that painless hair removal cannot be achieved.
Probiotics are microorganisms existing in the intestinal tracts of cats and beneficial to the health of the cats, are applied to the directions of treating diarrhea, improving soft stool, improving immunity and the like of the cats, but strains capable of helping the cats to convert hair and discharge hair are not reported at present.

Example 3 in vitro bionic digestion test of cat hair
1. Test materials: 3 cats with good skin health conditions (the varieties are respectively puppets, silver gradual layers and short, and the cat hair is collected by 1g of each cat) and placed in a 50mL centrifuge tube. And taking out the cat hair, and drying the cat hair in an oven at 65 ℃ until the weight is constant.
2. Preparation of simulated gastric fluid and simulated intestinal fluid
(1) Preparation of simulated gastric fluid: weighing 0.2g of sodium chloride and 0.32g of pepsin, adding 100mL of distilled water, stirring for dissolution, adjusting the pH to 2.5 by using 1mol/L hydrochloric acid solution, and filtering with a 0.2 mu m filter membrane for later use.
(2) Preparation of simulated intestinal juice: 0.68g of potassium dihydrogen phosphate was weighed, dissolved in 50mL of distilled water, and pH was adjusted to 6.75 with 0.1mol/L sodium hydroxide solution. 1g of pancreatin is weighed, and 50mL of distilled water is added for stirring and dissolution. Mixing the above two solutions to obtain simulated intestinal juice.
3. The test method comprises the following steps:
(1) 6 identical beakers (3 each for the experimental group and the control group) were taken, 0.3g of the cat hair after constant weight in step 1 (0.1 g each for 3 cat hairs) was added to each beaker, 20mL of the bacterial liquid prepared in step (2) of example 2 was added to the experimental group, and 20mL of the sterile MRS liquid medium was added to the control group.
(2) 50mL of simulated gastric fluid was added to each beaker, shaken well, and placed on a shaker to simulate gastric digestion for 4h (60 rpm at 37 ℃). The gastric simulation product was removed, pH was adjusted to 6.75 using 1mol/L sodium hydroxide solution, 40mL of simulated intestinal fluid was added, sealing was performed, and the shaker intestinal simulated digestion was allowed to stand for 9h (60 rpm at 37 ℃), after which the product was removed after completion of the gastrointestinal simulation.
(3) The cat hair was filtered through an 80 mesh nylon mesh, washed three times with distilled water, and dried in an oven at 65 ℃ to constant weight.
(4) The weight of cat hair after constant weight was weighed, and the weight loss ratio was calculated, and the results are shown in Table 1.
TABLE 1 weight loss of cat hair
Note that: * Shows significant difference compared with the control groupP<0.05)。
As can be seen from Table 1, the weight loss rate of cat hair increased after the enterococcus faecium JYEF-355 bacterial liquid was added. The weight loss rate of the cat hair test group is improved by 5.43 percent compared with that of the control group, and the cat hair test group has obvious change. The enterococcus faecium JYEF-355 can promote the decomposition of cat hair in the gastrointestinal simulated environment, and has a strong promoting effect on the cat hair decomposition.
EXAMPLE 4 enterococcus faecium JYEF-355 in vivo hairing test
(1) The test selects 20 healthy adult puppet cats with the weight of 3.5-4.5 kg, the weight is randomly divided into 2 groups (test group and control group), each group is 10 repeated, each group is repeated by 1 cat (each group comprises 1/2 of male and female cats), and all cats do not take any fur-dissolving product within half a year before the test starts.
(2) Necessary immunization and insect repellent treatment were performed before the test, the cat litter was replaced daily, and the cat house was cleaned daily.
(3) During the test period, each cat was fed in a single cage, the control group and the test group were fed with the same amount of basal diet daily, the control group was supplemented with 2g of glucose daily, and the test group was supplemented with 2g of the microbial inoculum prepared in example 2 daily.
(4) The pre-test period is 1 week, the positive test period is 8 weeks, on the 7 th day, the 14 th day, the 21 st day, the 28 th day, the 35 th day, the 42 th day, the 49 th day and the 56 th day, all the feces of the cat on the day are collected, whether the cat hair is discharged or not is observed, the cat hair in the feces is separated, the cat hair is dried to constant weight in a drying box at 65 ℃ after being washed by clear water, and the data are weighed and recorded, and the result is shown in figure 1.
As can be seen from fig. 1, during the test period, the fecal hair output of the cats in the test group tended to rise and then fall and then tended to be gentle, whereas the fecal hair output of the cats in the control group did not change much throughout the process. On test day 7, the cats in the test group discharge a large amount of hair through defecation, the weight of the hair bulb in the feces is increased until day 21, and the weight of the hair bulb in the feces of the cats in the control group is stable, which means that a large amount of hair bulb is accumulated in the abdomen of the cats, and if no product which is helpful for hair melting is taken, a small amount of hair can be discharged through defecation every day, and the intestinal tract remains, so that the health is influenced over time. After the microbial inoculum prepared in the example 2 is taken for a period of time, the hair discharge amount in the cat feces of the test group is obviously higher than that of the control group, which indicates that the microbial inoculum is helpful for cats to discharge hair balls in the bellies in a defecation mode, has the effect of promoting hair discharge, takes effect quickly and is beneficial to health. On day 28, the weight of the fecal hair bulb began to decrease in the test group, and after day 42, the fecal hair output tended to stabilize, probably by having previously discharged most of the hair bulb accumulated in the abdomen, but since cats were habituated to lick Mao Shumao, hair was constantly being discharged, and the fecal hair output of the test group was always higher than that of the control group during the test. Since cats in the control group do not take the wool-dissolving effect product for a long time, only a small amount of wool balls can be discharged daily, a large amount of wool balls are left in the abdomen, and the wool balls cannot be digested and cannot be discharged. On day 60, 20 cats were palpated by a professional feline doctor, more than half of the cats in the control group had soft foreign bodies in their small intestines, and were subsequently treated.
Example 5 test of the Effect of enterococcus faecium JYEF-355 on the body Condition of cats
When a large amount of hair gathers or has globus fulgidus in the cat digestive tract, the cat is often accompanied by symptoms such as vomiting, anorexia, camping, constipation, listlessness, hair loss, rough hair, no luster and the like. Cat health assessment criteria (table 2) were formulated based on the above symptoms and the cats in example 4 were scored for health by a professional assessor for a period of time of day 56 of example 4, with the assessment results shown in figures 2 and 3.
TABLE 2 Cat health assessment criteria
TABLE 3 enterococcus faecium JYEF-355 influence on cat physical condition scoring results
As can be seen from fig. 2 and table 3, the test group has higher score values in terms of feces, vomiting degree, mental state, hair loss degree, hair smoothness, appetite, activity and abdominal state than the control group, which indicates that the enterococcus faecium JYEF-355 microbial agent has an improving effect on the health condition of cats. Wherein, the method is particularly prominent in improving the mental state of cats, and the scores of cats in the test group on the mental state are significantly different from those of cats in the control groupP<0.001). In addition, the cat in the test group had the hair falling degree [ ]P=0.039), appetite%P=0.044) and abdominal stateP=0.014) the score values were significantly higher than the control group.
