技术领域

背景技术
σ受体(SR)是发现于包括脑在内的许多组织中的整体膜蛋白(Guitart等人,2004;Rousseaux和Greene,2016)。在细胞水平,它们主要分布在细胞质线粒体、内质网、核膜等中(van Waarde等人,2015)。中枢神经系统(CNS)中广泛存在的SR参与调节神经保护和许多其他行为,包括记忆和运动(Walker等人,1993)。它们在胚胎发生的所有阶段都会过度表达(Langa等人,2003),在增殖活跃的细胞中也是如此(Van Waarde等,2010年)。据报道,SR的失调和过度表达与乳腺癌、结直肠癌、前列腺癌和脑癌等侵袭性癌症的侵袭和转移表型有关(Vilner等人,1995)。由于SR在肿瘤细胞中的表达量高出10倍,SR配体可选择性地靶向肿瘤细胞并诱导肿瘤细胞选择性凋亡(Van Waarde等人,2010)。

发明内容

发明目的
本发明的主要目的是开发一种用于胶质瘤治疗的纳米制剂及其制备方法。本发明提供了一种针对侵袭性胶质母细胞瘤的强治疗策略。在本发明中,由葡萄糖基碳纳米球(CSP)制成的载体可穿过血脑屏障(BBB)到达大脑,通过使用具有足够抗癌活性的修饰的σ配体H8,纳米球诱导靶向杀死中度表达σ受体的胶质瘤细胞。
此外,纳米球靶向表达σ受体的肿瘤相关巨噬细胞的靶向能力并将其杀死,导致荷胶质瘤小鼠的存活率提高。
发明概述

本发明的肿瘤肿块靶向组合物包含碳纳米球,碳纳米球携带阳离子σ配体,阳离子σ配体是阳离子脂质和作为σ受体靶向配体的氟哌啶醇的共轭物,可同时靶向脑内胶质母细胞瘤肿块中的肿瘤上皮细胞(TEC)和肿瘤相关巨噬细胞(TAM)。由于该组合物具有可携带附加抗癌药物的额外优势,因此该组合物显示出了靶向和杀死TEC和TAM的能力,从而展示了有效消退肿瘤和提高存活率的双靶向策略。
本发明提供了一种双重药物递送策略,可更有效地用于表达σ受体的癌症中的肿瘤有效消退。更进一步的是,附加药物的共轭可能会带来显著的疗效。
本发明的发明人在进行了涉及大量的人工和技术干预的重要实验后,能够意外地提供一种物质,通过将σ受体靶向配体H8与葡萄糖衍生的碳纳米球CSP共轭,将药物分子特异性地输送到肿瘤部位。本发明提供了一种如此发明的物质和技术诀窍,用于向所有类型的SR表达癌症高度选择性地递送药物。在癌症化疗领域,医学科学领域很可能从本发明受益最多。
本发明的优势在于通过BBB向胶质瘤区域递送药物的特异性。此外,纳米共轭物对表达σ受体的肿瘤上皮细胞和肿瘤相关巨噬细胞表现出靶向能力。这些发现是迄今为止未知的,因此也是本发明的发明人的预料不到的发现。

实施例1
二辛基甘氨酸甲酯(a)的合成
在装有回流冷凝器的50mL圆底烧瓶中,将甘氨酸甲酯(0.5g,5.6mmol)溶解在20mL干燥的乙酸乙酯中。加入碳酸钾(1.9g,13.77mmol)和1-溴辛烷(4.278g,22.4mmol),将所得混合物在70-80℃的油浴中回流12小时。然后冷却,用水(2×20mL)和盐水(1×20mL)洗涤,用无水Na2SO4干燥,蒸发。使用60-120目硅胶和10%乙酸乙酯-正己烷(v/v)作为洗脱剂对残留物进行柱层析纯化,得到化合物(1.29g,收率73.3%,在10%乙酸乙酯-正己烷(v/v)中的Rf=0.6)。

实施例2
N-(2-甲氧基-2-氧代乙基)-N-辛基辛烷-1-氯化铵(b)的合成
在25mL圆底烧瓶中,将化合物(a)(0.3g,0.95mmol)溶于5mL干燥的二甲基甲酰胺中,然后加入过量的碘甲烷(3mL)。将反应混合物在室温下搅拌4小时。然后浓缩,使用60-120目硅胶和2%甲醇-氯仿(体积分数)作为洗脱剂对残留物进行柱层析纯化后,通过对残留物进行氯离子交换色谱(使用Amberlite IRA-400氯离子树脂和甲醇作为洗脱剂),得到淡黄色胶状液体化合物(0.1g,收率87%,在5%甲醇-氯仿(v/v)中Rf=0.4)。

在25mL的圆底烧瓶中,将化合物(b)(0.2g,0.55mmol)溶解在10mL的1:1比例的THF和H2O中,加入10当量的氢氧化锂,进一步使得在室温下搅拌3小时。然后用2N-HCl洗涤(2×30mL),并用乙酸乙酯萃取。使用60-120目硅胶和8%甲醇-氯仿(v/v)作为洗脱剂,通过冲洗柱色谱对残留物进行纯化,得到化合物(c),为黄色胶状液体(收率69.7%,10%甲醇-氯仿(v/v)中的Rf=0.5)。

实施例4
N-叔丁氧羰基-β-丙氨酸-氟哌啶醇共轭物(d)的合成
在50mL圆底烧瓶中,将N-Boc-β-丙氨酸(1.35g,7.17mmol)、氟哌啶醇(2.24g,5.9mmol)和N,N-二甲基氨基吡啶(DMAP)(0.53g,2.92mmol)的混合物溶解在10mL干燥的二氯甲烷(DCM)中,并在冰中搅拌半小时。向混合物中加入溶于5mL的干躁DCM的DCC(1.5g,7.29mmol),并在冰浴中搅拌1小时。反应混合物在室温下继续搅拌24小时。然后过滤反应混合物。滤液用水(2×30mL)和盐水(1×30mL)洗涤,用无水Na2SO4干燥,然后蒸发。使用60-120目硅胶和1%甲醇-氯仿(v/v)作为洗脱剂对残留物进行柱层析纯化,得到化合物(d),为微红黄色胶状物(2.85g,收率85.8%,在5%甲醇-氯仿(v/v)中的Rf=0.6)。

β-丙氨酸-氟哌啶醇共轭物(e)的合成
在50mL的圆底烧瓶中,将化合物(d)(1.0g,1.83mmol)溶于18mL的干躁DCM中,并在冰浴中搅拌15分钟。然后向溶液中滴加2mL三氟乙酸。反应混合物在冰浴中继续搅拌4小时,然后用饱和NaHCO3溶液中和溶液。混合物用DCM(2×15mL)萃取,有机层用Na2SO4干燥。蒸发有机层,得到化合物(e),为微红黄色胶状物质(0.75g,收率92%,10%甲醇-氯仿(v/v)中Rf=0.4,在茚三酮炭化中具有活性)。由于化合物(e)的纯度为95%(通过TLC显示),因此可直接用于下一步。

H8(f)的合成
在25mL圆底烧瓶中,将化合物(c)(0.250g,0.56mmol)溶于5mL干燥的DMF中,并在冰浴中搅拌15分钟。然后加入N-[(二甲基氨基)-1H-1,2,3-三唑并[4,5-b]吡啶-1-基亚甲基]-N-甲基甲铵六氟磷酸盐N-氧化物(N-[(Dimethylamino)-1H-1,2,3-triazolo-[4,5-b]pyridin-1-ylmethylene]-N-methyl methanaminium hexafluorophosphate N-oxide,HATU)(0.231g,0.61mmol),继续搅拌30分钟。然后加入溶解在2mL干燥DMF中的化合物(e)(0.195.5g,0.56mmol),接着滴加二异丙基乙胺(DIPEA),直到反应混合物变成微碱性。所得混合物搅拌48小时。然后将反应混合物溶解在20mL DCM中,用1N-HCl(2×20mL)、水(1×20mL)和盐水(1×20mL)洗涤,用无水Na2SO4干燥并蒸发。柱色谱纯化(使用100-200目硅胶和2%甲醇-氯仿(v/v)作为洗脱剂)得到H8,为无色胶状固体(96mg,收率22%,在10%甲醇-氯仿(v/v)中的Rf=0.5)。

实施例8
CSP共轭物的制备:在5mL甲醇(HPLC级)中分别制备3mM的H8和Q8储备液。将这些储备液单独加入到CSP(10mg)中,在水浴超声下保持5分钟,然后在室温(RT)下搅拌12小时。纳米共轭物混合物在27℃、10,000rpm转速下离心10分钟,得到的CSP纳米共轭物颗粒用于进一步的表征研究。
H8、Q8及其CSP共轭物的表征
H8和Q8阳离子分子的化学结构(图1)。CSP的合成和化合物在CSP上的结合是按照之前描述的方案完成的(Selvi等人,2008)。然后按照之前讨论的方案(印度专利申请号201841009113;2018年3月13日提交),将CSP与H8或Q8结合,并分别标记为CH8(CSP-H8)和CQ8(CSP-Q8)。将这些CSP共轭物分散在水溶液中,并使用zeta测定仪(zeta sizer)测量它们的水动力直径(范围为340至380nm)、多分散指数(PDI<0.3)和表面电荷(-37至-21mV)(表1)。zeta电位测量值分别从-37mV上升到-23mV和-22mV,这清楚地表明阳离子脂质H8和Q8分别吸附在CSP上。将纯CSP和CSP共轭物的FT-IR光谱与纯脂质的FT-IR光谱进行比较,表明脂质在CSP上的吸附/结合情况。流体力学直径和PDI值表明CSP共轭物分布均匀,FE-SEM图像中球形纳米共轭物的均匀分布进一步支持了这一点。在27℃条件下,使用高效液相色谱法测量了吸附在CSP上的H8和Q8的量。数据显示,10mg CSP上吸附了约1.8mg(2.3μmol)H8和0.65mg(1.9μmol)Q8。
DLS研究:使用Anton-Paar litesizer-500仪器测量了CSP、CH8(CSP-H8)和CQ8(CSP-Q8)的流体力学直径和表面电荷。简单地说,将20μL的CSP纳米共轭物悬浮液进一步稀释到1mL的去离子水中。然后测量水动力直径和表面电荷(表1)。
实施例9
H8赋予GL261和U87细胞对CSP的细胞摄取具有SR靶向性
为了检查在GL261(小鼠胶质瘤)、U87细胞(人类胶质母细胞瘤)以及非癌细胞CHO和HEK293中的摄取研究,将纳米球CH8和CQ8进一步与Rh-PE共轭,并处理这些细胞4小时。FACS摄取分析表明,CH8处理的脑癌细胞比CQ8处理的脑癌细胞中的荧光向右偏移更多。因此,H8配体的存在有助于CH8在细胞GL261和U87两者中获得更好的摄取。很明显,细胞测量数据表明,脑肿瘤细胞对CH8-Rh-PE的摄取率更高。在这些细胞中,U87对CH8的摄取率高于GL261细胞。这与GL261和U87的SR表达几乎相似,而U87的SR表达略高于GL261的事实是一致的。不过,为了了解脑癌细胞的摄取是否由SR辅助,在CH8处理之前,先用SR拮抗剂氟哌啶醇预处理GL261和U87细胞。
实施例10
CH8显示出对癌细胞具有高效的选择性杀伤作用:通过3-(4,5-二甲基噻唑-2-基)-2,5-二苯基溴化四氮唑(MTT)测定,分别检测和比较了H8、CH8和CQ8在GL261、U87、HEK293和CHO细胞中的细胞毒性。CH8的IC50值为1.9-2.3μM,而原始H8分子的IC50值为2.0-2.6μM,CQ8在这些浓度范围内对GL261和U87细胞的杀伤作用不明显,表明CH8在杀死癌细胞方面比CQ8显著有效。此外,CH8、H8和CQ8在非癌细胞(如CHO、HEK293)中在这些浓度范围内的细胞毒性作用可以忽略不计,因为这些细胞中SR的表达量很低或可以忽略不计。
ROS生成和细胞凋亡
CH8处理后会在GL261和U87细胞中产生ROS,且随着CH8浓度的增加,ROS的产生量也增加(图3)。此外,与使用CQ8处理的细胞相比,使用CH8处理的GL261和U87细胞显著地显示出更多数量的晚期凋亡细胞。然而,CH8处理导致非癌CHO和HEK293细胞中的晚期凋亡细胞数量微乎其微。这些数据(图4)总体上表明了CH8在癌细胞中的选择性凋亡诱导能力。

实施例11
CH8在小鼠原位胶质瘤中的积累效率高于CSP
在体外研究中证实了CH8对癌细胞的选择性细胞毒性后,在小鼠原位胶质瘤模型中检测了CH8的积累情况。为此,将CSP和CH8表面与近红外染料(DiR)共轭,表示为CSP(CSP-DiR)和CH8(CH8-DiR)。在两个不同的时间点(8小时和24小时)对携带原位GL261肿瘤的C57BL/6J小鼠进行腹膜内注射后,检查(CSP-DiR)和(CH8-DiR)的组织分布。背面图上的体内图像数据显示,有更多的纳米物质在大脑中聚集(图5)。在两个时间点,CH8-DiR在大脑中的累积量都明显高于CSP-DiR。虽然在24小时后,两个处理组脑中的近红外强度都有所下降,但CH8-DiR在脑中的保留量明显高于CSP-DiR在脑中的累积量。体外胶质瘤脑图像进一步支持了这一点(图6)。显然,与非靶向CSP相比,CH8在大脑中的渗透性和保留量都有提高,这表明CH8可能更倾向于与原位胶质瘤发生相互作用。由于H8(一种SR配体)的存在和不存在导致了积累的明显差异,因此可以得出结论,SR在大脑中的表达可用于将纳米粒子有效靶向到大脑中。
实施例12
肿瘤生长抑制和存活率研究
此外,为了观察目前所描述的SR靶向CH8对正位脑肿瘤生长的抑制效率,我们检测了不同治疗组交替给药5次后带有脑肿瘤的小鼠的存活率。在这里,腹腔注射自基于立体定向将GL261细胞接种到脑部的第4天开始,至第12天结束(共注射5次)。经CH8处理的小鼠与经游离H8处理的小鼠和未处理组相比,存活率显著提高(图7a)。在另一组实验中,在携带脑内原位GL261细胞并具有5次不同处理注射的动物在第12天被处死,以观察从各处理组获得的全脑中肿瘤病变的情况。来自UT和H8处理的小鼠的大脑显示出肿瘤部位(黑点)的相当效果,与从CH8处理的小鼠获得的大脑中的肿瘤部位(黑点)相比,来自UT和H8处理的小鼠的大脑中的肿瘤部位(黑点)在视觉上要大得多(图7b)。显然,各种治疗方法的视觉效果反映了治疗小鼠的总体存活率。
如前所证实的,H8本身具有抗癌作用,但可能无法穿越BBB,以在原位胶质瘤小鼠中显示其抗肿瘤作用。如果这一观点是正确的,那么在GL261皮下肿瘤模型中,H8和CH8的抗肿瘤作用应该是相同的。为了证明这一假设,我们建立了皮下模型,并按照同样的5次注射治疗模式,从接种GL261细胞后的第11天开始。从肿瘤消退曲线(图7c)可以看出,H8和CH8的抗肿瘤效果相似。此外,在接种后第20天处死小鼠后获得的不同处理组肿瘤的视觉图像(图7d)和各自的体积(图7e)也证实了H8和CH8具有相似的抗肿瘤效果。这些效应累积起来影响了不同治疗组小鼠的总体存活率,其中,我们看到分别用H8和CH8治疗的各组小鼠的存活率相似(图7f)。
实施例13
从肿瘤微环境中分离TAM并估算从肿瘤微环境中获得的TAM群体中的SR表达量
由于从经CH8处理的携带正位肿瘤的小鼠获得的离体脑中实现的肿瘤存活率较高,且可观察到肿瘤病变的面积相对较小,我们推测抗癌作用应累积作用于肿瘤微环境(TME)相关的肿瘤上皮细胞以及在TME中释放有利因子的其他促肿瘤细胞。TME包含由包括肿瘤相关巨噬细胞(TAM)在内的各种细胞分泌的多种促肿瘤因子。因此,我们选择破解TAM是否也会被CH8靶向并杀死。这只有在CH8能被有效摄取的情况下才有可能,如果在TAM上表达,可能是通过其同源受体SR。
为此,作为概念验证,使用抗皮下GL261肿瘤的肿瘤裂解液CD11b的微珠分离出了TAM。CD11b是TAM的表面标记物之一。磁性分离出的CD11b+TAM部分首先使用抗各种表面标记物(如F4/80、CD68)的抗体来检测它们是否主要是促肿瘤生成的M2亚型(图8a),F4/80、CD68是众所周知的M2-巨噬细胞标记物。显然,F4/80和CD68在TAM中有更多的表达。此外,还记录到缺乏LY6C和较低的MHCII表面标志物表达,这典型地表示如此分离出的TAM不存在抗肿瘤巨噬细胞M1的标记物或其水平较低。很明显,这样获得的TAM群体中含有活化的巨噬细胞(M2或TAM),它们能够抑制抗肿瘤免疫并促进肿瘤生长。
在分离出TAM并分离出不含TAM的肿瘤细胞部分后,我们通过FACS分析检测了SR在TAM和分离出的肿瘤细胞中的表达情况。我们发现,SR在两种细胞类型中都有表达。但令人惊讶的是,与分离的肿瘤细胞相比,SR在TAM中的表达量更高(图8b)。
实施例14
CH8在体内TAM中的积累:为此,在GL261皮下肿瘤接种后,当肿瘤体积达到1500mm3时,将小鼠分成两组,分别用Rh-PE共轭的CH8和CSP处理。8小时后,从不同处理组中被处死小鼠的皮下肿瘤中收集TAM和肿瘤细胞。FACS研究显示,CH8处理小鼠的TAM中Rh-PE的摄取高于分离的肿瘤细胞(图9a)。图9b清楚地表明,TAM比CSP摄取更多的CH8,这表明由于TAM表达SR,SR靶向纳米球在特定时间点驻留在TAM中的能力增强。
CH8-DOX对GL261细胞的细胞毒性:通过3-(4,5-二甲基噻唑-2-基)-2,5-二苯基溴化四唑(MTT)测定,检测并比较了DOX、CSP-DOX(C-DOX)、CH8和CH8-DOX在GL261细胞中的细胞毒性。对于给定浓度的DOX,CH8-DOX的细胞毒性效果相对高于DOX、C-DOX,在有相应浓度的CH8纳米共轭物存在的情况下也是如此(图10)。
本发明的优势
开发出一种强效靶向载体,对胶质母细胞瘤等致命癌症具有增强的疗效。穿越BBB到达脑癌细胞在制造高效治疗药物中发挥关键作用。在这里,受体靶向配体在低浓度下对脑癌细胞具有抗增殖作用,有助于选择性地杀死肿瘤细胞。此外,这种受体靶向配体还能容纳已获批准的抗癌药物,并在极低浓度下显示出突出的细胞毒性作用。
Claims (18)
1.一种具有抗癌活性的纳米制剂,包含比例为1:0.08至1:0.2的碳纳米球(CSP)和σ受体靶向配体(H8)的复合物。
2.一种制备具有抗癌活性的纳米制剂的方法,所述纳米制剂包含比例为1:0.08至1:0.2的碳纳米球(CSP)和σ受体靶向配体(H8)的复合物,所述方法包括以下步骤:
i.)提供:
a.N-(羧甲基)-N-甲基-N-辛基辛烷-1-氯化铵
b.N-[(二甲基氨基)-1H-1,2,3-三唑并[4,5-b]吡啶-1-基亚甲基]-N-甲基甲铵六氟磷酸盐N-氧化物(HATU)
c.β-丙氨酸-氟哌啶醇共轭物
d.CSP(碳纳米球)
ii.)将步骤(i)(a)中得到的N-(羧甲基)-N-甲基-N-辛基辛烷-1-氯化铵溶于干燥的二甲基甲酰胺(DMF)中,并在冰浴中搅拌,得到混合物;
iii.)将步骤(i)(b)中得到的N-[(二甲基氨基)-1H-1,2,3-三唑并[4,5-b]吡啶-1-基亚甲基]-N-甲基甲铵六氟磷酸盐N-氧化物(HATU)加入到步骤(ii)中得到的混合物中,得到反应混合物;
iv)将步骤(i)(c)中得到的β-丙氨酸-氟哌啶醇共轭物溶于干燥的二甲基甲酰胺(DMF)中,得到共轭物混合物;
v.)向步骤(iv)中得到的共轭物混合物中加入二异丙基乙胺(DIPEA),然后将混合物滴加到步骤(iii)中得到的反应混合物中,直到反应混合物变成微碱性;
vi)将步骤(iv)中得到的反应混合物搅拌40-50小时;
vii.)将二氯甲烷(DCM)溶于步骤(v)中得到的反应混合物中,然后用1N-HCl、水和盐水洗涤,用无水Na2SO4干燥、蒸发和纯化,得到H8;
viii.)将步骤(vi)中得到的H8溶于甲醇中,然后将溶液加入步骤(i)(d)中得到的CSP中,将共轭物在水浴超声下保持5-10分钟,然后在室温下搅拌10-12小时,得到纳米共轭物混合物;
ix.)将步骤(vii)中得到的纳米共轭物在20-30℃下离心10分钟,得到CSP纳米共轭物颗粒。
3.根据权利要求1所述的纳米制剂,其中,所述纳米制剂与附加药物共轭,其中,所述附加药物选自包含多柔比星、吉西他滨、替莫唑胺、卡莫司汀和依维莫司的抗癌药物的组。
4.根据权利要求1所述的纳米制剂,其中,所述纳米制剂对于靶向胶质母细胞瘤肿块中的肿瘤上皮细胞(TEC)和肿瘤相关巨噬细胞(TAM)是有用的。
5.一种下述通式的复合物:
CSP-H8-D
其中,CSP代表碳纳米球;H8代表σ受体靶向配体;D代表强效药物;
其中,CSP与H8共轭;CSP-H8共轭物与强效药物D共价或非共价连接。
6.根据权利要求5所述的复合物,其中,所述强效药物D是亲水性或疏水性抗癌剂,所述亲水性或疏水性抗癌剂选自包含多柔比星、吉西他滨、卡莫司汀、依维莫司和替莫唑胺的组。
7.根据权利要求5所述的复合物,其中,所述碳纳米球(CSP)和所述σ受体靶向配体(H8)的比例为1:0.08至1:0.2。
8.根据权利要求5所述的复合物,其中所述复合物对于靶向肿瘤或胶质母细胞瘤肿块中的肿瘤上皮细胞和肿瘤相关巨噬细胞是有用的。
9.一种制备具有下述通式的复合物的方法:
CSP-H8-D,
所述方法包括以下步骤:
(iii)将碳纳米球(CSP)与σ受体靶向配体(H8)共轭,形成CSP-H8或CH8纳米共轭物;
(iv)通过将CSP-H8或CH8纳米共轭物与强效药物D的乙醇溶液混合,并搅拌足以确保D与CH8连接的时间,最大限度地保持CH8:D的比例为1:0.2,从而将强效药物D与CH8共轭。
10.根据权利要求9所述的方法,其中,所述搅拌的时间为7-15小时,优选为8-12小时。
11.根据权利要求9所述的方法,其中,所用的醇为C1至C3醇。
12.一种靶向肿瘤或胶质母细胞瘤肿块的组合物,包含:
a)碳纳米球(CSP),其携带阳离子σ配体,所述配体是阳离子脂质的共轭物;和
b)作为σ受体靶向配体(H8)的氟哌啶醇衍生物。
13.根据权利要求12所述的组合物,其中,所述碳纳米球(CSP)和所述σ受体靶向配体(H8)的比例为1:0.08至1:0.2。
14.根据权利要求12所述的组合物,其中,所述组合物对于靶向胶质母细胞瘤肿块中的肿瘤上皮细胞(TEC)和肿瘤相关巨噬细胞(TAM)是有用的。
15.一种将药物分子特异性递送至肿瘤部位的药物递送试剂盒,所述试剂盒具有通过将σ受体靶向配体(H8)与葡萄糖衍生的碳纳米球(CSP)共轭制备的复合物。
16.根据权利要求15所述的试剂盒,其中,所述复合物进一步与附加药物共轭,其中,所述附加药物选自包含多柔比星、吉西他滨、替莫唑胺、卡莫司汀和依维莫司的抗癌药物的组。
17.根据权利要求15所述的试剂盒,其中,所述试剂盒对于靶向肿瘤上皮细胞和肿瘤相关巨噬细胞,用于治疗胶质母细胞瘤或肿瘤肿块是有用的。
18.一种治疗肿瘤或胶质母细胞瘤肿块的方法,所述方法分别用权利要求1、12和15所述的纳米制剂或组合物靶向肿瘤上皮细胞(TEC)和肿瘤相关巨噬细胞(TAM)。
