Abstract
本发明提供了一种磺酰胺‑萘酰亚胺‑TB衍生物及其合成方法和应用,选用对溴苯胺、多聚甲醛、对甲苯磺酰氯、乙二胺、4‑溴‑1,8‑萘酰亚胺等为原料,通过多步反应合成得到。其结构式如下式第一衍生物(10)和第二衍生物(11)所示:产物在溶液和固态中均具有较大的Stokes位移(均大于100nm);具有显著的AIE性质;具有广泛的pH适用范围,可应用于人生理环境中;与在纯甲醇中相比,在甲醇/丙三醇=1/9(v/v)时荧光分别增强了1.6倍和4.3倍,均对黏度有较好的响应能力;均易进入活的A549细胞中,对其内质网均具有较强的靶向能力(皮尔逊系数Pr分别为0.81和0.71);对A549细胞和HpeG2细胞的光毒性和暗毒性均较低,表明其生物相容性高,具有作为肿瘤细胞的黏度探针和内质网探针的潜力。

技术领域
本发明属于化学合成领域,具体涉及磺酰胺-萘酰亚胺-base衍生物及其合成方法及其在黏度响应、内质网靶向中的应用。
背景技术
内质网(Endoplasmic Reticulum,ER)是真核细胞内重要的细胞器,主要负责脂质合成,蛋白质的合成、加工和修饰以及新生肽链的折叠、组装和运输。其状态和结构是动态的,易受环境因素影响。一些外界诱因如缺氧、病毒感染、氧化还原反应、葡萄糖缺失等均会引发ER稳态失衡,导致ER腔内错误和未折叠蛋白的聚集以及钙离子平衡紊乱,进而产生ER应激(ERS)。ERS发生后,细胞启动相应的保护反应,即未折叠蛋白反应(UPR)来恢复ER稳态,但当应激过强或持续时间过久时,会导致ER程序性坏死或凋亡;促使ER与其他细胞器形成互作。所以严重的ERS会导致心脏病、糖尿病、神经退行性疾病和癌症等疾病。
虽然陆续有ER定位荧光探针的报道,但还存在两个问题:其内在分子机制不明确,导致种类有限;在水性介质中具有强的聚集荧光淬灭(ACQ)效应,导致成像效果差。因此,开发种类多、选择性高、ACQ效应弱甚至无ACQ效应、高效低毒的ER荧光探针是该领域的难点和热点。到目前为止,尚未见基于base(TB)骨架的ER荧光探针的报道。
1,8-萘酰亚胺是重要的发光团,具有良好的光稳定性、大的Stokes位移以及较高的荧光量子产率,且具有双光子发射。含有1,8-萘酰亚胺(NI)的分子是有机光子学和电子学等领域快速发展的研究热点,在荧光染料、激光染料、金属传感器、pH传感器、生物成像、有机发光二极管等领域均有广泛应用。NI的光物理和电化学性质取决于取代模式和供体单元的性质,对NI单元的亚胺位点进行修饰,不仅可以生产大量的NI衍生物,还可以调整其光物理和电子特性。
因此,本发明通过在TB骨架上依次引入1,8-萘酰亚胺和ER定位基对甲苯磺酰胺片段,设计、合成了具有内质网靶向功能的磺酰胺-萘酰亚胺base衍生物。/>base(简称TB)及其衍生物具有独特的V-型骨架和较长的共轭结构,在光子激发下具有多种跃迁方式(π-π*、n-π*、空间跃迁),理论上具有较大摩尔吸光系数,是一种优异的紫外光吸收材料基础骨架。
发明内容
技术问题:本发明的目的在于提供一种磺酰胺-萘酰亚胺-TB衍生物及其合成方法和应用,以对溴苯胺、多聚甲醛、对甲苯磺酰氯、乙二胺、4-溴-1,8-萘酰亚胺等为原料,通过多步反应合成得到磺酰胺-萘酰亚胺base衍生物,并将其应用于黏度的识别和ER定位等领域。

以下有:4-溴苯胺1,多聚甲醛2,第一中间体3,第二中间体4,甲苯磺酰氯5,乙二胺6,第三中间体7,4-溴-1,8萘酰亚胺8,第四中间体9,第一衍生物10,第二衍生物11。

本发明还提供了上述新型磺酰胺-萘酰亚胺base衍生物的制备方法,包括:
本实施例中,以对溴苯胺、多聚甲醛、正丁基锂、4溴-1,8萘酰亚胺和乙二胺等为原料通过多步反应制得。包括以下步骤:
4-溴苯胺与多聚甲醛反应得到第一中间体3,第一中间体3与正丁基锂反应得到第二中间体4,第二中间体4与第四中间体9通过偶联反应得到第一衍生物10和第二衍生物11。
通过上述合成方法,制备实施例中的第一衍生物10和第二衍生物11:
将4-溴苯胺(50.0mmol)和多聚甲醛(100.0mmol)依次加入200.0mL圆底烧瓶中,置于低温槽中调温至-15℃,搅拌下向烧瓶内缓慢滴加三氟乙酸(100.0mL,约30min滴加完毕)后,室温下反应7天。反应完全后(TLC追踪),将混合物倒入冰水中,氨水调节pH=9-10,冷却至室温,二氯甲烷萃取(50.0mL×3),旋干得粗产物。加入丙酮,加热至粗产物全部溶解,室温下重结晶,抽滤,丙酮洗涤,得第一中间体3。

(3)将第一中间体3(5.0mmol)加入100mL圆底烧瓶内,抽换气三次后,置于低温槽中调温至-78℃,搅拌下向烧瓶内加入20.0mL无水四氢呋喃,滴加2.5mL正丁基锂,在氩气保护下反应1h后滴加0.6mL硼酸三甲酯,之后置于室温下反应4h。TLC追踪至反应完全后,二氯甲烷萃取(30.0mL×3),旋干得粗产物。粗产物经柱层析提纯(VPE:VEA=5:1)得第二中间体4(65%)。

(4)将对甲苯磺酰氯(1.0mmol)溶于二氯甲烷(5mL)后缓慢加入到二氯甲烷溶解的乙二胺(10mmol)溶液中,40℃搅拌15分钟后,将混合物用水(25mL)洗涤两次并用Na2SO4干燥。真空除去溶剂,得到第三中间体7,为白色固体产物(83%)。

(5)将第三中间体7(1.0mmol)和4-溴-1,8-萘酰亚胺(0.28g,1.0mmol)溶于乙醇(20mL)中。将混合物在氩气保护下在80℃搅拌4小时,反应完全后(TLC追踪),加水淬灭过滤后,滤饼用CH2Cl2/CH3OH=25/1(v/v)作为洗脱液,经柱层析纯化得第四中间体9(72%)。

(5)取第一中间体3(1.0mmol)、第四中间体9(1.2mmol)、四(三苯基磷)钯(20%mmol,0.03g)和K2CO3(0.2mmol)依次加入100mL圆底烧瓶中,氩气保护下加入20mL无水甲苯,108℃反应24h。反应完全后(TLC追踪),加水淬灭,二氯甲烷萃取(10.0mL×3),有机相经Na2SO4干燥后,旋干得到的粗产物经柱层析提纯(V石油醚:V乙酸乙酯=7:1),得第一衍生物10(38%)和第二衍生物11(33%)。

光学性能测试
测试了本发明化合物的溶剂化效应,具体试验方案如下:
将第一衍生物10和第二衍生物11分别用甲醇(Methanol)、二甲基亚砜(DMSO)、N,N-二甲基甲酰胺(DMF)、四氢呋喃(THF)、乙酸乙酯(ethyl acetate)、三氯甲烷(CHCl3)、甲苯(Toluene)、正己烷(n-Hexane)配制成浓度为1×10-5mol·L-1的工作液,并测试其紫外吸收光谱和荧光发射光谱。如图7a-7d所示。
从图7a-7d可看出,第一衍生物10和第二衍生物11的紫外吸收波长随着溶剂极性的增加λem发生明显的蓝移,这归因于杂原子的R带吸收。随着极性增加,第一衍生物10和第二衍生物11的n-π*的跃迁增强,基态极性比激发态大,因此基态能够与极性溶剂之间产生较强的氢键,能量下降较大,而激发态能量下降较小,故跃迁能量增加,荧光发射波长蓝移。
测试了第一衍生物10和第二衍生物11在THF溶液中的紫外吸收和荧光发射光谱及其固态荧光发射光谱,具体试验方案如下:
称取10-5mol的4-溴苯胺,第二中间体4,第一衍生物10和第二衍生物11,用THF溶液定容至浓度为1×10-5mol/L,测试其紫外吸收、荧光发射及固态荧光发射光谱。
4-溴苯胺,第二中间体4,第一衍生物10和第二衍生物11的光谱数据如表2所示。
由表2可知,与4-溴苯胺和第二中间体4相比,第一衍生物10和第二衍生物11的荧光性质具有以下变化:
(1)第一衍生物10的溶液及固态λem均发生明显红移,溶液和固态Stokes位移(分别为103、182nm)明显增大;
(2)第二衍生物11的溶液及固态λem均发生明显红移,溶液和固态Stokes位移(分别为125、173nm)明显增大;
(3)第二衍生物11与第一衍生物10相比,第二衍生物11的溶液及固态荧光强度均有所增加,这可能是因为增加的萘酰亚胺数目时荧光量子产率,发光强度增强,因此第二衍生物11的荧光发射强度大于第一衍生物10。
以上结果说明,将TB骨架与1,8-萘酰亚胺片段结合可放大二者在发光性能上的优势,是获得具有优异发光性能的产物的新途径。

pH响应
pH响应的合适范围是探针在生物体系中成功应用的关键因素之一,因此我们探索了第一衍生物10的pH适用范围。以THF作为溶剂将6配制成浓度为1×10-4mol·L-1的工作溶液,分别量取1.0mL第一衍生物10工作液于9个10.0mL容量瓶中,随后分别加入1.0mLpH值为2.2-10.0的缓冲溶液(pH为2.2-8.0时选取柠檬酸/磷酸氢二钠体系,pH为9.0-10.0时选取碳酸氢钠/碳酸钠体系),THF定容,使其浓度均为1×10-5molL-1,测得其荧光发射光谱(λex=370nm,狭缝:5/5nm)。
第二衍生物11的试剂配制方法与第一衍生物10一致。第一衍生物10和第二衍生物11的荧光强度在pH2.2-10.0的范围内几乎保持恒定,表明第一衍生物10和第二衍生物11具有广泛的pH适用范围。
AIE特性
由于第一衍生物10易溶于THF,难溶于水,将10配制成浓度为1×10-4mol·L-1的工作溶液,分别量取1.0mL工作液于10个10.0mL容量瓶中,随后在10个10.0mL容量瓶中分别加入0.0-9.0mL双蒸水,THF定容,使其浓度均为1×10-5mol·L-1(THF/H2O(v/v)依次为1/9-9/1),将第一衍生物10配制成浓度为1×10-3mol·L-1的工作溶液,量取100.0μL工作液于10.0mL容量瓶中,随后在10.0mL容量瓶中加入9.9mL双蒸水,THF定容,使其浓度为1×10- 5molL-1,使得THF/H2O=1/99(v/v)。测得荧光发射光谱如7a-7b所示(λex=370nm,狭缝:5/5nm)。
第二衍生物11的试剂配制方法与第一衍生物10一致(λex=380nm,狭缝:5/5nm,图7c-7d)。
由7a-7d可知,第一衍生物10在水含量为10%-80%时,荧光逐渐降低,当含水量从80%继续增加到99%时,由于分子内运动受限导致荧光不断增强,在水含量为99%荧光强度达到峰值,且荧光发生红移,到达红光区域,有利于细胞成像。第二衍生物11随着溶液极性的增加,荧光降低,出现ACQ现象,可能是由于第二衍生物11具有两条长脂肪链,降低了分子内刚性。
为了探究第一衍生物10的AIE性能的形成机制,我们采用扫描电子显微镜(SEM)观察了第一衍生物10在THF/H2O=2/8(v/v)和THF/H2O=1/99(v/v)时的形貌特征,如图8a-8b所示。
由图8a-8b可知:第一衍生物10在THF/H2O=2/8(v/v)时分子呈现出无定型的状态,而在THF/H2O=1/99(v/v)时分子呈现均匀的片状聚集,其荧光急剧增强,体现出AIE特性,而该纳米聚集体可能是赋予其AIE性能的原因。第一衍生物10在THF/H2O=1/99(v/v)时分子的平均直径为2μm。
黏度响应
对第一衍生物10和第二衍生物11的黏度响应性进行了测试:以甲醇为溶剂将10配制成浓度为1×10-4mol·L-1的工作液。取10个10.0mL容量瓶,分别加入0.0-9.0mL丙三醇,量取1.0mL工作液于容量瓶中,甲醇定容,使其浓度均为1×10-5mol·L-1,测得荧光发射光谱(甲醇/丙三醇的体积比依次为1/9-10/0),如9a-9b所示(λex=370nm,狭缝:5/5nm)。
第二衍生物11的试剂配制方法与10一致(λex=430nm,狭缝:5/10nm,图7c-7d)。
由图9可知,随着黏度的增大,第一衍生物10和第二衍生物11的荧光强度增强。这可能是随着黏度的升高,导致分子内运动受阻,这表明第一衍生物10和第二衍生物11有可能针对内质网中粘度的变化产生高水平响应,可作为多功能内质网荧光探针做进一步研究。
干扰实验
检查了常见阳离子Fe3+、Al3+、Na+、Ca2+、Cu2+、Cr3+和K+;阴离子CO3 2-、HCO3 -、CH3COO-、PO4 2-、SO4 2-、SCN-和HS-;生物硫醇Cys、Hcy和GSH以及90%丙三醇(从左至右2-18,1为空白对照)对第一衍生物10荧光发射光谱的影响,如图10所示(λex=370nm,狭缝:5/10nm)。
同理,测试了各不同离子和分子对第二衍生物11的影响(λex=380nm,狭缝:5/10nm,图11)。
由结果可知,加入各种阴阳离子、生物硫醇后,第一衍生物10和第二衍生物11的荧光强度基本上没有发生变化,并且与第一衍生物10和第二衍生物11在90%丙三醇溶剂中的荧光强度差别明显。因此,第一衍生物10和第二衍生物11均对黏度具有特异性响应,能够在复杂的生物环境中实现对黏度的特异性检测。
蛋白质聚集实验
蛋白质错误折叠和聚集过程中形成的致密多肽链会导致黏度发生变化[74]。而蛋白质变性后理化性质会发生改变,并暴露出分子内部的疏水基团,使其凝聚速度加快,从水溶液中析出,导致其黏度增加。因此我们采用蛋清热变性过程模拟蛋白质聚集过程中的黏度变化,并用第一衍生物10对其变性时的黏度变化进行监测,观察其荧光变化。
首先测试了第一衍生物10(图12a,λex=370nm,狭缝:5/10nm)和(图12b,λex=380nm,狭缝:5/10nm)在0-100℃时的荧光变化,结果表明第一衍生物10和第二衍生物11在0-100℃时荧光强度无明显的变化,表明其具有良好的热稳定性(λex=430nm,狭缝:5/10nm)。
将第一衍生物10和第二衍生物11与适量蛋清混合,随着温度升高,蛋白质逐渐聚集、黏度增加,第一衍生物10和第二衍生物11的荧光随之逐渐增强,表明其可用来监测蛋白质聚集时的黏度变化,具有成为蛋白质聚集的黏度响应探针的潜能(图12c-12d)。
ER定位实验
ER作为细胞重要的细胞器,在细胞中将细胞核、细胞质、细胞膜有机地连接成一个整体,并负责物质在细胞中的转运,同时也是蛋白质、脂类的合成基地。而氰基脂溶性好,理论上可定位于ER。因此,设计了合理的共定位实验进行验证。
首先分别将ER商业探针ER-Tracker Green分别与第一衍生物10和第二衍生物11在适宜的条件下与HeLa细胞共孵育30分钟后,用磷酸缓冲盐溶液(PBS)清洗两次,加入PBS进行共聚焦成像,结果如图13-14所示。
第一衍生物10和第二衍生物11均能易进入活的HeLa细胞中,第一衍生物10的Pr=0.81,第二衍生物11的Pr=0.71,表明第一衍生物10和第二衍生物11均能准确定位于ER并对ER进行荧光成像,具有较强的ER靶向能力。
体外光动力治疗
以人非小细胞肺癌(A549)细胞和人肝癌(HepG2)细胞为模型,采用MTT法检测化合物第一衍生物10和第二衍生物11对A549和HepG2细胞的细胞毒性。将A549和HepG2细胞分别接种在96微孔板中(1×10-5个/mL),每个孔中加入100μL的培养基,在37℃的CO2培养箱中培养24h后,加入不同浓度的第三中间体7到接种好的细胞中孵育24h。然后用PBS缓冲溶液冲洗微孔板3次,在每个孔中加入10μL的MTT溶液继续培养4h。移除孔内的培养基,在每个孔中加入150μL的DMSO用以溶解细胞内的蓝紫色甲臜(Formazam)晶体,并将其置于摇床上,低速震荡5-7min,使结晶物质充分溶解。最后采用酶联免疫检测仪测量各个孔在560nm和670nm处的吸光度值。细胞毒性通过下列公式计算:

式中Ai分别为不同浓度化合物的吸光度值;A0为无添加化合物的对照孔的平均吸光度值;n(=3)表示三次平行实验。
采用MTT法检测第一衍生物10和第二衍生物11对HpeG2细胞以及A549细胞的暗毒性和光毒性。光源为430nm,对照组不进行光照处理,并采用酶联免疫检测仪测量各个孔在560nm和670nm处的吸光度值。第一衍生物10和第二衍生物11对A549和HpeG2细胞的暗毒性和光毒性如图15a-15d所示。
第一衍生物10和第二衍生物11对HepG2和A549细胞的半数抑制率如表3所示,结果表明,第一衍生物10和第二衍生物11对A549细胞和HpeG2细胞的光毒性和暗毒性均较低;表明其对这两种细胞的生物相容性高,适用于进行癌症细胞的黏度变化监测、内质网定位等。

以上所述仅是本发明的优选实施方式,应当指出:对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。
