Abstract

背景技术

近年来,基于铁离子的定量检测及其机制的探究开发了电化学、离子色谱、阴离子交换液相色谱-电感耦合等离子体质谱法等检测手段,然而由于仪器昂贵、操作繁琐,限制了其普遍应用。相较于上述方法,荧光法是一种高效、便捷、低廉的检测方式之一。它是指荧光探针与分析物特异结合前后探针分子的光物理性质发生明显变化,从而检测荧光信号的变化,实现对生物体或环境中不同分子或离子含量的检测。目前,越来越多的研究者开发荧光探针用于铁离子的检测。然而,这些荧光探针仍面临着一些难题,如稳定性差、特异性低、毒性高,无法实现铁离子的高灵敏、高特异性检测以及活体内的荧光成像。为了克服这些难题,亟需开发一种化学结构稳定、生物相容性高的新型荧光探针,构建简单、高灵敏和高特异性的铁离子分析平台,实现活体内荧光成像。
共价有机框架是一类高度结晶的多孔有机聚合物,由共价键连接的轻质元素构成。由于具有高比表面积、良好的稳定性、多变拓扑结构和易功能化修饰等优势,共价有机框架在包括气体储存、分子吸附/分离、药物运输和传感等应用方面显示出巨大潜力。此外,共价有机框架内部的π堆积层促进了电子转移,并且基于固有的孔隙结构,为合适的分析物提供了足够的纳米空间。基于这些优点,多种共价有机框架荧光探针已被开发用于检测铁离子。但目前报道的共价有机框架荧光探针存在量子产率低、生物相容性差且铁离子选择性有待改善等问题,无法运用于活体内铁离子水平的动态监测。
因此,急需提供一种基于共价有机框架的荧光成像探针、制备方法及应用以解决现有技术中存在的问题。
发明内容
本发明的主要目的是提供一种基于共价有机框架的荧光成像探针、制备方法及应用,以解决相关技术中共价有机框架荧光探针存在量子产率低、生物相容性差且铁离子选择性有待改善的技术问题。
为实现上述目的,本发明提供了一种基于共价有机框架的荧光成像探针,所述荧光成像探针的结构式如下:
其中,Me表示甲基,n表示聚合度,所述荧光成像探针包括共价有机框架以及聚乙烯二氧噻吩。
优选的,所述聚合度的范围为1000-4000。
本发明的有益效果是:
(1)本发明提供的荧光成像探针中,由于聚乙烯二氧噻吩中噻吩单元富含电子,因此赋予共价有机框架高电子密度环境,且噻吩上S原子的孤对电子可以在高度富电子的共轭体系中扩散,从而实现有效的电子转移。而铁离子能够提供未被占据的d轨道,是理想的电子受体。因此,当荧光成像探针与铁离子混合时,能够通过噻吩位点上S原子的孤对电子和铁离子之间发生反应,导致荧光猝灭,实现对铁离子检测的特异性和高灵敏性。
(2)本发明提供的荧光成像探针相比于相关技术中的荧光探针具有更高的量子产率,荧光强度更高。通过聚乙烯二氧噻吩提高共价有机框架结构的刚性,增加π电子共轭,减少非辐射跃迁的损失,提高荧光效率;此外聚乙烯二氧噻吩为共价有机框架提供了高电子密度环境,且硫原子的孤对电子在高度富电子的共轭体系中扩散,增加了电子跃迁数量,进而提高了荧光强度。
(3)本发明通过控制合适的聚乙烯二氧噻吩聚合度,使得聚乙烯二氧噻吩与共价有机框架之间能够形成牢固的空间缠绕结构,使得得到的荧光成像探针更加稳定,减少由于荧光成像探针中空间结构失效而出现的实验误差,提高实验结果可靠性。
本发明还提供一种用于制备上述荧光成像探针的制备方法,包括:
将2,5-二溴-3,4-乙烯基二氧噻吩溶于有机溶剂B中,与共价有机框架粉末混合,搅拌,真空蒸发有机溶剂A得到半成品;
对半成品进行提纯得到荧光成像探针。
优选的,所述共价有机框架的合成包括:将1,3,5-三(4-氨基苯基)苯和2,5-二甲氧基对苯二甲醛加入有机溶剂B中,超声,搅拌并将乙酸注入混合液中,搅拌,离心,洗涤,真空干燥制得共价有机框架粉末。
优选的,所述有机溶剂A包括丙酮以及乙醚中的一种或多种;所述有机溶剂B包括乙腈以及丙腈中的一种或多种。
优选的,所述共价有机框架粉末与2,5-二溴-3,4-乙烯基二氧噻吩的加入质量比为1:(1.4-2.2)。
优选的,所述提纯步骤包括:冲洗半成品;保护气下密封,并在50-70℃下加热2.5-3天,75-90℃下加热0.5-1.5天;冷却,多次洗涤,干燥得到荧光成像探针。
本发明的有益效果是:
通过本发明提供的制备方法得到荧光成像探针灵敏度较高。通过该制备方法得到的荧光成像探针含有的杂质较少,因此当将其用于实验时,也会减少由于杂质带来的对于实验结果的影响,提高了实验结果的可靠性。
本发明还提供一种上述的荧光成像探针或如上述制备方法制得的荧光成像探针在检测铁离子的应用。
优选的,所述检测铁离子的过程包括:
获取荧光成像探针溶液荧光强度与铁离子浓度之间的线性方程;
将荧光成像探针溶液与待测铁离子溶液混合,测定荧光成像探针溶液荧光强度,通过线性方程计算得到待测铁离子溶液的浓度。
本发明的有益效果是:通过获取荧光成像探针溶液荧光强度与铁离子浓度之间的线性方程,然后测定荧光强度,即可根据线性方程确定待测铁离子溶液的浓度,不仅丰富了测定铁离子浓度的手段,而且极大的简化了浓度测定的步骤,减少了浓度测定过程中的时间成本。
本发明还提供一种上述的荧光成像探针或上述制备方法制得的荧光成像探针在活体荧光成像中的应用。
本发明的有益效果在于:本发明用于活体荧光成像的荧光成像探针采用聚乙烯二氧噻吩和共价有机框架混合制得,由于聚乙烯二氧噻吩和共价有机框架分别具有较好的生物相容性,因此荧光成像探针也具有较好的生物相容性,使得本发明提供的荧光成像探针能够用于活体荧光成像。

本发明提供了一种基于共价有机框架的荧光成像探针,所述荧光成像探针(可简称为PEDOT@COF)的结构式如下:
其中,Me表示甲基,n表示聚合度,所述荧光成像探针包括共价有机框架(英文名称COF)以及聚乙烯二氧噻吩(英文名称PEDOT)。
聚乙烯二氧噻吩具有分子结构简单、能隙小、电导率高等特点,而共价有机框架(COF)是一类高度结晶的多孔有机聚合物,由共价键连接的轻质元素构成。由于具有高比表面积、良好的稳定性、多变拓扑结构和易功能化修饰等优势,共价有机框架在气体储存、分子吸附/分离、药物运输和传感等应用方面显示出巨大潜力。此外,共价有机框架内部的π堆积层促进了电子转移,并且基于固有的孔隙结构,为合适的分析物提供了足够的纳米空间。本发明通过添加用于形成聚乙烯二氧噻吩的原料(例如2,5-二溴-3,4-乙烯基二氧噻吩)以共价有机框架内部的孔隙结构作为聚合场所,形成聚乙烯二氧噻吩,从而得到聚乙烯二氧噻吩与共价有机框架的缠绕空间结构,即荧光成像探针。聚乙烯二氧噻吩和共价有机框架结合方式参见图1。
荧光成像探针中,由于聚乙烯二氧噻吩中噻吩单元富含电子,因此赋予共价有机框架高电子密度环境,且噻吩上S原子的孤对电子可以在高度富电子的共轭体系中扩散,从而实现有效的电子转移。而铁离子能够提供未被占据的d轨道,是理想的电子受体。因此,当荧光成像探针与铁离子混合时,能够通过噻吩位点上S原子的孤对电子和铁离子之间发生反应,导致荧光猝灭,实现对铁离子的高特异性和高灵敏性检测。
相比于相关技术中的荧光探针具有更高的量子产率,荧光强度更高。通过聚乙烯二氧噻吩提高共价有机框架结构的刚性,增加π电子共轭,减少非辐射跃迁的损失,提高荧光效率;此外聚乙烯二氧噻吩为共价有机框架提供了高电子密度环境,且硫原子的孤对电子在高度富电子的共轭体系中扩散,增加了电子跃迁数量,进而提高了荧光强度。
所述聚合度的范围为1000-4000。通过控制合适的聚乙烯二氧噻吩聚合度,使得聚乙烯二氧噻吩与共价有机框架之间能够形成牢固的空间缠绕结构,使得得到的荧光成像探针更加稳定,减少由于荧光成像探针中空间结构失效而出现的实验误差,提高实验结果可靠性。
本发明还提供一种用于制备上述荧光成像探针的制备方法,包括:
将2,5-二溴-3,4-乙烯基二氧噻吩(英文名称为DBrEDOT)溶于有机溶剂B中,与共价有机框架粉末混合,搅拌,真空蒸发有机溶剂A得到半成品;
对半成品进行提纯得到荧光成像探针。
在一些实施例中,所述共价有机框架的合成包括:将1,3,5-三(4-氨基苯基)苯(英文名称为TAPB)和2,5-二甲氧基对苯二甲醛(英文名称为DMTP)加入有机溶剂B中,超声,搅拌并将乙酸注入混合液中,搅拌,离心,洗涤,真空干燥制得共价有机框架粉末。
在一些实施例中,所述有机溶剂A包括丙酮以及乙醚中的一种或多种;所述有机溶剂B包括乙腈以及丙腈中的一种或多种。
在一些实施例中,所述共价有机框架粉末与2,5-二溴-3,4-乙烯基二氧噻吩的质量比为1:(1.4-2.2),优选的,共价有机框架粉末与2,5-二溴-3,4-乙烯基二氧噻吩的质量比为1:1.8。通过合适的质量比,能够提高共价有机框架粉末以及2,5-二溴-3,4-乙烯基二氧噻吩的转化率,从而减少在生成产物荧光成像探针中的杂质含量,有利于减少后续提纯步骤中冲洗剂的用量,减少制备成本,此外通过合适质量比得到的荧光成像探针的荧光强度更高,能更好的用于荧光成像。
在一些实施例中,所述提纯包括:用冲洗剂(包括正己烷、正庚烷中的一种或多种)冲洗半成品;冲洗过的半成品在氮气作为保护气的条件下密封,在60℃和85℃下分别加热3天和1天,冷却后,用丙酮、亚硫酸钠水溶液、超纯水依次洗涤。最后将所得到的粉末真空干燥过夜,得到荧光成像探针。
通过本发明提供的制备方法得到荧光成像探针灵敏度较高。通过该制备方法得到的荧光成像探针含有的杂质较少,因此当将其用于实验时,也会减少由于杂质带来的对于实验结果的影响,提高了实验结果的可靠性。

荧光成像探针采用聚乙烯二氧噻吩和共价有机框架混合制得,由于聚乙烯二氧噻吩和共价有机框架分别具有较好的生物相容性,因此荧光成像探针也具有较好的生物相容性,使得本发明提供的荧光成像探针能够用于活体荧光成像。
实施例1:荧光成像探针的制备
参见图1将1,3,5-三(4-氨基苯基)苯(20mg)和2,5-二甲氧基对苯二甲醛(20mg)溶解乙腈(40mL)中,超声30秒后,在搅拌条件下将乙酸(1mL)注入混合液中,在室温下保持剧烈搅拌。随后,离心收集沉淀物,并用无水乙醇洗涤数次,50℃下真空干燥制得共价有机框架粉末。
将2,5-二溴-3,4-乙烯基二氧噻吩(36mg)溶于丙酮(1.5mL)中,与共价有机框架粉末(20mg)混合,在25℃下搅拌,真空条件下蒸发丙酮。对半成品进行提纯得到荧光成像探针,所述提纯步骤包括:用正己烷(5mL)冲洗粉末。冲洗过的粉末在氮气作为保护气的条件下密封,在60℃和85℃下分别加热3天和1天。冷却后,用丙酮(10mL)、亚硫酸钠水溶液(1.0M20mL)、超纯水(20mL)依次洗涤。最后将所得到的粉末真空干燥过夜,得到荧光成像探针。
实施例2:与实施例1不同之处在于,2,5-二溴-3,4-乙烯基二氧噻吩用量为28mg,共价有机框架粉末用量为20mg。
实施例3:与实施例1不同之处在于,2,5-二溴-3,4-乙烯基二氧噻吩用量为44mg,共价有机框架粉末用量为20mg。
对比例1:与实施例1不同之处在于,2,5-二溴-3,4-乙烯基二氧噻吩用量为20mg,共价有机框架粉末用量为20mg。
对比例2:与实施例1不同之处在于,2,5-二溴-3,4-乙烯基二氧噻吩用量为52mg,共价有机框架粉末用量为20mg。
对比例3:与实施例1不同之处在于,2,5-二溴-3,4-乙烯基二氧噻吩用量为60mg,共价有机框架粉末用量为20mg。
实施例1-3与对比例1-3得到的荧光成像探针的荧光强度参见图2。可见实施例1-3荧光成像探针的荧光强度相比于对比例1-3的荧光成像探针更高,能够更好的适用于荧光成像,其中实施例1得到荧光成像探针的荧光强度最高,是最优选的实施方式。
实施例4:实施例1中制备的荧光成像探针的表征
实施例1中制备的荧光成像探针的粒径大小、分子结构、结晶度分别通过电子透射电镜(TEM)、傅里叶红外光谱(FT-IR)、X射线衍射测试。荧光成像探针和共价有机框架的电子透射电镜图如图3中所示,荧光成像探针的形状大小与共价有机框架一致,呈球形,粒径大小约为850nm,但球形有阴影边缘,这是由于聚乙烯二氧噻吩的包裹。傅里叶红外光谱如图4所示,荧光成像探针的光谱显示了共价有机框架和聚乙烯二氧噻吩的特征吸收峰,表明共价有机框架在固相聚合过程中得到了保留。1617cm-1、1353cm-1和1117cm-1处的吸收峰对应于不对称C=N、C=C拉伸和环间C-C拉伸。此外,1070cm-1、976cm-1和629cm-1的吸收峰分别对应于C-O-C弯曲振动和C-S-C拉伸振动,这表明聚乙烯二氧噻吩成功聚合于共价有机框架孔径中。X射线衍射结果图如图5所示,实施例1中制备的荧光成像探针相较于共价有机框架,单个衍射峰的相对强度降低,这是由于共价有机框架孔径中存在的聚乙烯二氧噻吩为无序的客体。
实施例5:实施例1中制备的荧光成像探针吸收-发射光谱的测定
用无水乙醇溶剂将实施例1中合成的荧光成像探针配成浓度为0.24g·L-1的溶液。用200-800nm波长范围内用荧光可见分光光度计在365nm激发下收集得到荧光发射光谱,结果如图6所示,荧光成像探针在乙醇溶液中的激发峰位于365nm,发射峰位于475nm,荧光成像探针在无水乙醇溶液中的激发-发射光谱(左侧部分为激发光谱,右侧部分为发射光谱)。说明荧光成像探针的激发-发射峰位。
实施例6:实施例1中制备的荧光成像探针检测铁离子
用实施例1中制备的0.24g·L-1的荧光成像探针溶液中加入不同浓度(浓度根据需求选择)的铁离子溶液,用荧光可见分光光度计在365nm激发下收集得到荧光发射光谱的变化,用Origin软件处理数据得到如图7所示的线性方程。说明实施例1中制备的荧光成像探针的荧光强度与铁离子浓度呈现良好的线性关系;检测范围在0-1mM时,线性方程为:y=1.0804+0.00512x,R2=0.994。
实施例7:实施例1中制备的荧光成像探针在细胞中的荧光成像
1.细胞培养:HeLa细胞株是在37℃、CO2浓度为5%的培养箱中进行培养,培养液为含有10%胎牛血清和1%青链霉素混合液的高糖DMEM。
2.细胞毒性:利用CCK8实验评估实施例1中制备的荧光成像探针的细胞毒性具体是:在含10%胎牛血清的DMEM细胞培养基(200μL)的96孔板中培养HeLa细胞,5%CO2,37℃,潮湿环境下培养24h,原密度为5000个/孔。随后,去除培养基,加入不同浓度(包括0.0g·L-1、0.1g·L-1、0.2g·L-1、0.3g·L-1、0.4g·L-1以及0.5g·L-1)的荧光成像探针和培养基,孵育24h。随后,每孔中加入CCK8试剂(20μL),再次在37℃和5%CO2下孵育0.5h,然后记录每孔吸光度。结果如图8所示,0.5g·L-1的荧光成像探针在24h内不会影响HeLa细胞的正常生长,说明该荧光成像探针具有较好的生物相容性,
3.细胞荧光成像:以HeLa细胞为测试细胞,在玻璃底培养皿中培养细胞进行细胞成像。培养皿中含有2mL培养基,其中包括10%胎牛血清和90%DMEM细胞培养基,在37℃和5%CO2湿度环境下培养。随后,弃掉培养基,然后用含有荧光成像探针(0.24g·L-1)的全新培养基加入培养皿中(2mL)。
培养24h后,用PBS溶液清洗三次以去除任何残留的荧光成像探针。然后,用含铁离子的培养基培养2h。用PBS溶液洗涤细胞3次,用溶酶体红色荧光探针(LysoTracker RedDND-99)复染HeLa细胞溶酶体1h后,用PBS溶液去除残留的LysoTracker Red DND-99。随后,使用40×共聚焦显微镜观察HeLa细胞的荧光。结果如图9所示(A)的荧光图像收集在红色通道(λex=561nm)中,(B)的荧光图像收集在蓝色通道(λex=405nm)中,(D)的荧光图像收集在蓝色通道(λex=405nm)中,(E)的荧光图像收集在红色通道(λex=561nm)中。(A)和(B)的合并后的图像(C),(D)和(E)的合并图像(F)。(A)、(B)和(C)显示仅用0.24g·L-1荧光成像探针孵育的HeLa细胞。(D)、(E)和(F)显示HeLa细胞与0.24g·L-1荧光成像探针和500μM铁离子一起孵育。比例尺为50μm),可以清晰观察到实施例1中制备的荧光成像探针与溶酶体探针LysoTracker Red ddn-99能很好地实现共定位,说明实施例1中的制备的荧光成像探针内化于细胞溶酶体中。此外,加入铁离子后,细胞内荧光减弱,说明实施例1中的制备的荧光成像探针能够对细胞内的铁离子响应。
实施例8:实施例1中制备的荧光成像探针在斑马鱼中的荧光成像
1.斑马鱼孵育:鱼卵置于26℃下恒温孵育。
2.斑马鱼毒性:配制不同浓度的荧光成像探针溶液加进六孔板中,每个浓度组孵育20条斑马鱼1天,记录斑马鱼的生存能力。结果如图10所示,实施例1中制备的荧光成像探针对斑马鱼毒性较低,当浓度达到0.5g·L-1时,在24h内也不会影响斑马鱼的生存能力。
3.斑马鱼荧光成像:孵化后的斑马鱼首先在荧光成像探针溶液(0.24g·L-1)中培养2h,然后用蒸馏水清洗。最后,将斑马鱼浸入铁离子溶液中30min。用荧光显微镜拍摄荧光图像。结果如图11所示,斑马鱼体内显示出蓝色荧光,说明实施例1中制备的荧光成像探针能够成功进入斑马鱼体内。此外,引入铁离子后,斑马鱼的蓝色荧光减弱,说明实施例1中制备的荧光成像探针能够成功进入斑马鱼体内,可以动态监测斑马鱼中的铁离子浓度。
实施例9:实施例1中制备的荧光成像探针在小鼠体内的荧光成像
1.溶血率实验:将2%红细胞(20μL)与实施例1中制备的荧光成像探针溶液(1mL)混合,使其最终浓度分别为0.2g·L-1,0.4g·L-1,0.6g·L-1,0.8g·L-1和1.0g·L-1。对照组分别设阳性对照(含水和2%红细胞)和阴性对照(含0.1MPBS和2%红细胞)。孵育1h后,将样品离心。使用酶标仪测定上清液545nm处光密度(OD)。结果如图12所示,当实施例1中制备的荧光成像探针浓度达到1g·L-1时,溶血率仅达2.68%。说明荧光成像探针具有良好的血液相容性,可以进行动物实验。
2.小鼠体内荧光成像:C57小鼠用乙醚麻醉,然后将实施例1中制备的荧光成像探针通过尾静脉注射到C57小鼠中(200μL),培养2h。随后,注射铁离子溶液,并用小动物活体成像仪捕获荧光图像。结果如图13所示,实施例1中制备的荧光信号主要在心脏、肺、肾脏中聚集,加入铁离子后,荧光信号减弱,说明实施例1中制备的荧光成像探针能够成功进入小鼠体内,表明荧光探针可以可视化小鼠体内铁离子的变化。由实施例1-7可知,本发明成功制备得到了荧光成像探针,且本发明制备的荧光成像探针的荧光强度与铁离子浓度的呈现良好的线性关系,能够用于铁离子的浓度检测;此外经过细胞毒性测试、斑马鱼毒性测试以及溶血率测试验证了本发明提供的荧光成像探针具有优异的生物相容性,并在后续实验中将本发明提供的荧光成像探针成功用于HeLa细胞、斑马鱼以及C57小鼠的体内荧光成像。
本发明提供的荧光成像探针,不仅丰富了测定铁离子浓度的手段,简化了浓度测定的步骤,减少了浓度测定过程中的时间成本,而且由于其具有优异的生物相容性,使得其能够用于活体荧光成像。
综上所述,本发明提供的基于共价有机框架荧光成像探针、制备方法及应用解决了相关技术中共价有机框架荧光探针存在量子产率低、生物相容性差且铁离子选择性有待改善的技术问题。
本发明的上述技术方案中,以上仅为本发明的优选实施例,并非因此限制本发明的专利范围,凡是在本发明的技术构思下,利用本发明说明书及附图内容所作的等效结构变换,或直接/间接运用在其他相关的技术领域均包括在本发明的专利保护范围。
Claims (10)
1.基于共价有机框架的荧光成像探针,其特征在于,所述荧光成像探针的结构式如下:
其中,Me表示甲基,n表示聚合度,所述荧光成像探针包括共价有机框架以及聚乙烯二氧噻吩。
2.根据权利要求1所述的荧光成像探针,其特征在于,所述聚合度的范围为1000-4000。
3.一种用于制备如权利要求1或2所述的荧光成像探针的制备方法,包括:
将2,5-二溴-3,4-乙烯基二氧噻吩溶于有机溶剂B中,与共价有机框架粉末混合,搅拌,真空蒸发有机溶剂A得到半成品;
对半成品进行提纯得到荧光成像探针。
4.根据权利要求3所述的制备方法,其特征在于,所述共价有机框架的合成包括:将1,3,5-三(4-氨基苯基)苯和2,5-二甲氧基对苯二甲醛加入有机溶剂B中,超声,搅拌并将乙酸注入混合液中,搅拌,离心,洗涤,真空干燥制得共价有机框架粉末。
5.根据权利要求4所述的制备方法,其特征在于,所述有机溶剂A包括丙酮以及乙醚中的一种或多种;所述有机溶剂B包括乙腈以及丙腈中的一种或多种。
6.根据权利要求3所述的制备方法,其特征在于,所述共价有机框架粉末与2,5-二溴-3,4-乙烯基二氧噻吩的质量比为1:(1.4-2.2)。
7.根据权利要求3所述的制备方法,其特征在于,所述提纯步骤包括:冲洗半成品;保护气下密封,并在50-70℃下加热2.5-3天,75-90℃下加热0.5-1.5天;冷却,多次洗涤,干燥得到荧光成像探针。
8.如权利要求1或2所述的荧光成像探针或如权利要求3-7任意一项所述的制备方法制得的荧光成像探针在检测铁离子的应用。
9.根据权利要求8所述的应用,其特征在于,所述检测铁离子的过程包括:
获取荧光成像探针溶液荧光强度与铁离子浓度之间的线性方程;
将荧光成像探针溶液与待测铁离子溶液混合,测定荧光成像探针溶液荧光强度,通过线性方程计算得到待测铁离子溶液的浓度。
10.如权利要求1或2任意一项所述的荧光成像探针或如权利要求3-7任意一项所述的制备方法制得的荧光成像探针在活体荧光成像中的应用。
