(2) Adding a mixture of pectase, cellulase and protease into the raw material powder of the honey Luo Mu obtained in the step (1), and adding water to react for 1-3 days at the temperature of 30-40 ℃ to obtain an enzymolysis product;
(3) Adding a mixed solvent into the enzymolysis product obtained in the step (2) for ultrasonic extraction treatment, centrifuging after ultrasonic extraction is finished, taking supernatant and filtering to obtain filtrate;
(4) Concentrating the filtrate obtained in the step (3) under reduced pressure to obtain a dense Luo Mu extract.
In the preparation method, the raw material of the honey Luo Mu is decomposed by adopting a mixture of pectase, cellulase and protease, and the pectase and the cellulase are matched for use, so that components such as pectin and cellulose in the cell wall of the honey Luo Mu can be degraded, starch, lipids, vitamins and proteins in the cell of the honey Luo Mu are released, and the starch can be decomposed to generate oligosaccharides and monosaccharides. The added protease can decompose the protein released by the density Luo Muzhong to generate polypeptide and amino acid. The small molecular nutrient substances generated by the complex enzyme hydrolysis density Luo Mu can be better absorbed by skin, so that the effect of improving skin erythema is achieved.
In a specific embodiment, the temperature at which the seal Luo Mu is dried in step (1) is from 40 to 60 ℃.
In a specific embodiment, the mass ratio of the raw material powder of Mi Luo Mu, pectase, cellulase and protease is 8-10:1.5:1:0.5.
In a specific embodiment, the mixed solvent is an ethanol aqueous solution, and preferably, the volume fraction of ethanol in the ethanol aqueous solution is 50% -70%.
In a specific embodiment, the ratio of the enzymatic hydrolysate to the mixed solvent is 1:60-80 g/ml, preferably 1:65-75 g/ml, and more preferably 1:70 g/ml.
In a specific embodiment, the ultrasonic power of ultrasonic extraction in the step (3) is 400-600W, the ultrasonic extraction time is 20-30 min, and the ultrasonic extraction temperature is 60-80 ℃;
preferably, the power of the ultrasonic wave is 450-550W, the ultrasonic extraction time is 22-28 min, and the ultrasonic extraction temperature is 65-75 ℃.
The enzymolysis product of the known high-density Luo Mu contains various nutritional components and has very complex components, so that a proper solvent is required to extract the effective components in the enzymolysis product. In the invention, 50-70% ethanol water solution is used as a mixed solvent for ultrasonic extraction of the enzymolysis product of the honey Luo Mu, most of the effective components in the enzymolysis product of the honey Luo Mu can be extracted by the operation, and ethanol in the enzymolysis product can be removed by decompression concentration, so that the extract of the honey Luo Mu can be obtained.
In a specific embodiment, the reduced pressure concentration in step (4) is performed at a temperature of 40 to 60 ℃ and a pressure of-0.2 to-0.1 MPa.
In one embodiment of the invention, a Mi Luo Mu extract prepared from Mi Luo Mu extract with erythema dispelling effect is provided.
In one embodiment of the invention, the application of the extract of the honey Luo Mu in the field of skin care products is provided.
The present invention will be further illustrated with reference to the following specific examples, but the present invention is not limited to the following examples. The methods are conventional methods unless otherwise specified.
Example 1
(1) Drying and crushing the honey Luo Mu at the temperature of 40 ℃ to obtain honey Luo Mu raw material powder;
(2) Adding a mixture of pectase, cellulase and protease with the mass ratio of 8:1.5:1:0.5 into the raw material powder of the honey Luo Mu obtained in the step (1), and adding water to react for 1 day at the temperature of 30 ℃ to obtain an enzymolysis product;
(3) Adding 50% ethanol water solution into the enzymolysis product obtained in the step (2) to carry out ultrasonic extraction treatment, wherein the ultrasonic power of ultrasonic extraction is 400W, the ultrasonic extraction time is 20min, the ultrasonic extraction temperature is 60 ℃, centrifuging treatment is carried out after ultrasonic extraction is completed, and supernatant liquid is taken and filtered to obtain filtrate;
(4) Concentrating the filtrate obtained in the step (3) under reduced pressure at 40 ℃ and under-0.2 MPa to obtain the extract of Mi Luo Mu.
Example 2
(1) Drying and crushing the honey Luo Mu at 50 ℃ to obtain honey Luo Mu raw material powder;
(2) Adding a mixture of pectase, cellulase and protease with the mass ratio of 9:1.5:1:0.5 into the raw material powder of the honey Luo Mu obtained in the step (1), and adding water to react for 2 days at the temperature of 35 ℃ to obtain an enzymolysis product;
(3) Adding an ethanol water solution with the ethanol volume fraction of 60% into the enzymolysis product obtained in the step (2) for ultrasonic extraction treatment, wherein the ultrasonic power of ultrasonic extraction is 500W, the ultrasonic extraction time is 25min, the ultrasonic extraction temperature is 70 ℃, centrifuging treatment is carried out after ultrasonic extraction is completed, and supernatant liquid is taken and filtered to obtain filtrate;
(4) Concentrating the filtrate obtained in the step (3) under reduced pressure at 50deg.C under-0.15 MPa to obtain extract of Mi Luo Mu.
Example 3
(1) Drying and crushing the honey Luo Mu at 55 ℃ to obtain honey Luo Mu raw material powder;
(2) Adding the raw material powder of the density Luo Mu obtained in the step (1) into the raw material powder of the density Luo Mu in a mass ratio of 9.5:1.5:1: 0.5, adding water to react for 2.5 days at 38 ℃ to obtain an enzymolysis product;
(3) Adding ethanol water solution with ethanol volume fraction of 68% into the enzymolysis product obtained in the step (2) for ultrasonic extraction treatment, wherein the ultrasonic power of ultrasonic extraction is 550W, the ultrasonic extraction time is 28min, the ultrasonic extraction temperature is 75 ℃, centrifuging treatment is carried out after ultrasonic extraction is completed, and supernatant is taken and filtered to obtain filtrate;
(4) Concentrating the filtrate obtained in the step (3) under reduced pressure at 55deg.C under-0.2 MPa to obtain extract of Mi Luo Mu.
Example 4
(1) Drying and crushing the honey Luo Mu at 60 ℃ to obtain honey Luo Mu raw material powder;
(2) Adding the raw material powder of the density Luo Mu obtained in the step (1) into the raw material powder of the density Luo Mu in a mass ratio of 10:1.5:1:0.5, adding water to react for 3 days at the temperature of 40 ℃ to obtain an enzymolysis product;
(3) Adding 50-70% ethanol water solution into the enzymolysis product obtained in the step (2) to carry out ultrasonic extraction treatment, wherein the ultrasonic power of ultrasonic extraction is 600W, the ultrasonic extraction time is 30min, the ultrasonic extraction temperature is 80 ℃, centrifuging treatment is carried out after ultrasonic extraction is completed, and supernatant liquid is taken and filtered to obtain filtrate;
(4) Concentrating the filtrate obtained in the step (3) under reduced pressure at 60 ℃ and under-0.1 MPa to obtain the extract of Mi Luo Mu.
Test examples
Anti-inflammatory efficacy test of extract of mi Luo Mu:
the testing method comprises the following steps: the macrophages of the mice are obtained by culturing, and lipopolysaccharide LPS is used for stimulating the macrophages of the mice, so that the macrophages of the mice secrete inflammatory factor TNF-alpha. The extract of Mi Luo Mu prepared by the embodiment of the invention can inhibit macrophage from secreting inflammatory factor TNF-alpha. The inhibition of inflammatory factor TNF- α release by the extract of mi Luo Mu was evaluated by measuring the amount of inflammatory factor TNF- α in the supernatant of the mouse macrophage culture fluid. The evaluation criterion was calculated to show that the higher the inhibition ratio (%) of inflammatory factor TNF-alpha, the better the inflammation inhibition effect of the extract of Mi Luo Mu was. The test results are shown in Table 1.

As can be seen from Table 1, the inhibition ratio of the extract of Mi Luo Mu prepared in examples 1-4 of the present invention to inflammation was higher than 50%, which indicates that the extract of Mi Luo Mu prepared in the present invention has good anti-inflammatory effect.
Antiallergic efficacy test of Mi Luo Mu extract:
histamine is a small molecular weight amine, mast cells stored in tissues and basic cells in the circulation. When the human body develops antigen-antibody reaction or anaphylaxis, the mast cells are stimulated to release histamine, so that the antiallergic effect of the extract of Mi Luo Mu can be evaluated by detecting the release of histamine. This example uses KU812 cells, which are highly similar to mast cells, to conduct an antiallergic efficacy test of the extract of mi Luo Mu. KU812 cells were cultured to adhere, the culture solution was added with the extract of mi Luo Mu prepared in the example of the invention for further culture, the test group was added with C5a to stimulate the release of histamine from mast cells, and a blank group was set. After completion of the reaction, the supernatant of the culture broth and the cell pellet were separated, the histamine content was measured by a fluorescence method, and the resultant histamine inhibition was calculated, and the results were shown in Table 2.

As can be seen from Table 2, the extract of Mi Luo Mu prepared in the example of the present invention has high histamine inhibition rate, and can achieve antiallergic effect, thereby effectively inhibiting erythema caused by anaphylaxis.
Spot-removing efficacy test of mi Luo Mu extract:
the testing method comprises the following steps: 20 volunteers with skin erythema of different degrees caused by acne were equally divided into four groups, wherein two random groups of volunteers were applied with the Luo Mimu extract prepared by the example of the present invention after cleansing with clear water daily in the morning and evening, the other two groups of volunteers were applied with some existing brand of freckle-removing skin care product after cleansing with clear water daily in the morning and evening, the number of skin erythema of the volunteers was detected from day 0 and day 7 of the beginning of use, respectively, and the reduction rate of erythema was calculated. The test results are shown in Table 3.

As can be seen from table 3, luo Mimu extract prepared according to the present invention was effective in reducing the number of erythema on the skin surface of volunteers.

Claims (4)
1. Use of a mi Luo Mu extract for the preparation of a medicament or cosmetic for combating erythema, characterized in that: the preparation method of the extract of Mi Luo Mu comprises the following steps:
(1) Drying and crushing the honey Luo Mu to obtain honey Luo Mu raw material powder;
(2) Adding a mixture of pectase, cellulase and protease into the raw material powder of the honey Luo Mu obtained in the step (1), and adding water to react for 1-3 days at the temperature of 30-40 ℃ to obtain an enzymolysis product;
(3) Adding a mixed solvent into the enzymolysis product obtained in the step (2) for ultrasonic extraction treatment, centrifuging after ultrasonic extraction is finished, taking supernatant and filtering to obtain filtrate;
(4) Concentrating the filtrate obtained in the step (3) under reduced pressure to obtain a dense Luo Mu extract;
wherein, the mass ratio of the raw material powder of the honey Luo Mu to the pectase to the cellulase to the protease is 8-10: 1.5:1:0.5;
the mixed solvent is ethanol water solution, and the volume fraction of ethanol in the ethanol water solution is 50% -70%;
the feed liquid ratio of the enzymolysis product to the mixed solvent is 1: 60-80 g/ml.
2. The use according to claim 1, wherein: and (2) drying the density Luo Mu at 40-60 ℃ in the step (1).
3. The use according to claim 1, wherein the ultrasonic power of the ultrasonic extraction in the step (3) is 400-600W, the ultrasonic extraction time is 20-30 min, and the ultrasonic extraction temperature is 60-80 ℃.
4. The use according to claim 1, wherein the reduced pressure concentration in step (4) is carried out at a temperature of 40-60 ℃ and a pressure of-0.2 to-0.1 MPa.
