Abstract

Description
技术领域
本发明涉及基于特异性生物标志物的表达水平用于确定皮肤水分含量的方法。
背景技术
皮肤主要由三层组成,即从最表面开始,表皮、真皮和皮下组织。皮肤的外层(即表皮)由角质形成细胞(大部分)、黑素细胞(参与皮肤色素沉着)和朗格汉斯细胞组成。其功能是保护身体免受外部环境的影响并确保其完整性,特别是减缓微生物或化学物质的渗透,并防止包含在皮肤中的水蒸发。
为此,角质形成细胞经历连续定向的成熟过程,在该过程中位于表皮基底层的角质形成细胞在其分化的最终阶段形成角质形成细胞,该角质形成细胞是由蛋白质和脂质(诸如神经酰胺)组成的角质化包膜形式的完全角质化的死细胞。表皮的该最上层被称为角质层,且是表皮屏障功能的基本组成部分。
然而,表皮的屏障功能可在某些气候条件下(例如在寒冷和/或风的作用下)或在压力、疲劳或老化的作用下被破坏。皮肤屏障功能的破坏促进了过敏原、刺激剂或微生物的渗透,并直接导致皮肤干燥,这通常很少伴随不适感,诸如紧绷或发红。
为防止这种现象或纠正这种现象,已知的做法是将含有吸湿剂(诸如糖或多元醇)的化妆品组合物施用于皮肤,旨在捕获皮肤中存在的水,从而减缓其蒸发。
多种水合化合物是已知的和可获得的。然而,这些化合物通过不同的作用机制起作用,因此适合于不同的皮肤类型(例如干燥、敏感、混合型皮肤)。不幸的是,大多数人不知道他们的皮肤类型并使用不合适的产品。
皮肤干燥度通常在临床上通过视觉评估,或通过使用旨在测量上皮肤层的相对介电常数的corneometer来评估。不幸的是,这两种方法都面临严重限制。皮肤的视觉评估取决于进行该评估的专家,因此可能因人而异。类似地,corneometer评估在可重复性方面受到限制:结果与施加的压力和进行测量的大气条件密切相关。

发明简述

因此,本发明的第一方面涉及用于确定受试者中皮肤水分含量的方法,所述方法包括在从所述受试者获得的皮肤样品中测量至少一种蛋白质的表达水平的步骤,所述蛋白质选自下组:ESYT3、USP7、PPL和LCE1C。这种方法特别可以检测干燥皮肤,且因此可以适当地处理此类状况。
根据第二方面,本发明还涉及评估用于水合受试者皮肤的美容处理功效的方法,所述方法包括以下步骤:
a)在接受所述美容处理前从所述受试者获得的皮肤样品中测量

d)当在步骤b)中测量的表达水平低于在步骤a)中测量的表达水平时,确定所述美容处理是有效的,或者当在步骤b)中测量的表达水平类似于或高于在步骤a)中测量的表达水平时,确定所述美容处理是无效的。
最后,本发明还涉及筛选用于水合皮肤的候选化妆品化合物的方法,其中所述方法包括以下步骤:
a)在皮肤样品中测量至少一种蛋白质的表达水平,所述蛋白质选自下组:ESYT3、USP7、PPL和LCE1C;
b)使测试化合物与所述皮肤样品接触;

发明详述
本发明涉及与皮肤干燥特别相关的新的生物标志物的鉴定。这些生物标志物,即延长的突触结合蛋白-3(ESYT3)、泛素-羧基末端水解酶7(USP7)、旁血小板溶蛋白(PPL)和晚期角质化包膜蛋白1C(LCE1C),可用于准确评估皮肤的水分含量,特别可用于干躁皮肤的诊断。本发明人特别揭示,与正常皮肤相比,这些生物标志物在干燥皮肤中显著过表达,且它们的表达水平与皮肤干燥直接相关。本发明人进一步证明,保湿处理对根据本发明的生物标志物的表达水平具有直接影响,并且因此,它们代表了用于治疗干燥皮肤以及用于鉴定新的保湿剂的极有希望的工具。

ESYT3、USP7、PPL和LCE1C是本领域技术人员完全熟悉的已知蛋白质。
延伸突触结合蛋白-3(ESYT3)是延伸的突触结合蛋白(Esyt)家族的成员,其包含多结构域C2膜蛋白,其直向同源物存在于从酵母到人的生物体中。小鼠和人中存在三种Esyt基因。已知它们参与内质网和质膜之间连接的形成,以及参与Ca2+-依赖性膜磷脂的更新。ESYT3与桶状结构域中的甘油磷脂结合,且据报道可能在细胞脂质转运中起作用。人ESYT3的序列可在Uniprot数据库的条目A0FGR9下获得。
已显示泛素-羧基末端水解酶7(USP7)调节许多信号传导分子的更新,诸如p53和PTEN。Li等(EMBO Rep.2020May 6;21(5))已揭示,角质形成细胞中USP7的缺失导致IKKα(IκB激酶α)的泛素化增加,使得细胞分化期间IKKα水平降低。从移植的USP7 KO细胞再生的皮肤呈现显著的表皮异常,包括增厚的表皮和基底细胞扩张。表皮角质形成细胞中USP7的缺失导致IKKα水平降低和异常分化。人USP7的序列可在Uniprot数据库的条目Q93009下获得。
旁血小板溶蛋白(PPL)是角质形成细胞的角质化包膜的组分。角膜包膜由一系列在转谷氨酰胺酶作用下交联的前体蛋白组装而成。所得结构是具有共价连接的脂质的蛋白质的不溶性层,具有~15nm的厚度,且其沉积在角质形成细胞中的质膜的内表面。旁血小板溶蛋白所属的plakin蛋白家族的成员分别充当表皮细胞连结子和细胞-细胞和细胞-基质粘附复合物的组分,即桥粒和半桥粒。人PPL序列可在Uniprot数据库的条目O60437下获得。
晚期角质化包膜蛋白1C(LCE1C)是角质层角质化包膜的前体。LCE1C属于晚期角质化包膜家族,其成员在包膜成熟过程中通过转谷氨酰胺酶介导的交联整合到角质化包膜中。人LCE1C的序列可在Uniprot数据库中的条目Q5T751下获得。
在本发明的上下文中,测量这些生物标志物中至少一种的表达水平。这些生物标志物之一的表达水平足以确定皮肤的水分含量。然而,根据本发明的方法可以包括测量这4种生物标志物中的1、2、3或所有的表达水平。
本领域技术人员熟知每天用于确定蛋白质表达水平的众多技术。
蛋白质的表达水平可通过测量编码其的基因的表达水平来评估。基因的表达水平典型地通过分析从该基因转录的mRNA来确定。这些核酸分子可以典型地从获自受试者的生物样品中提取并通过标准方法分析。一种常用的方法是使用对目的基因特异的寡核苷酸引物对分离的mRNA进行逆转录(“RT”)和聚合酶链式反应(“PCR”)扩增。mRNA的定量典型地可以使用称为SYBR或/>的两种实时定量技术中的一种来进行。
还可通过测量样品中检测的所述蛋白质的量直接评估蛋白质表达水平。这些方法典型地包括将待分析的生物样品与能够特异性结合靶蛋白的试剂接触。该试剂通常是多克隆或单克隆抗体。然后,典型地在通过电泳分离蛋白质(也称为“Western blotting”的技术)后通过标准免疫检测方法或通过直接、间接、竞争或免疫捕获方法的免疫测定(也称为“ELISA”的技术)检测蛋白质的存在。通常通过测量产生有色、化学发光或荧光产物的酶促反应来检测和定量目标蛋白质和靶向所述蛋白质的抗体之间的复合物的形成。
“皮肤水分含量”在本文中是指包含在皮肤中,特别是包含在角质层中的水量。我们的皮肤由70%的水组成,其以从深度的70%到表面的15%递减的梯度分布。由于良好质量的皮肤屏障可以将水保留在皮肤内,这种称为皮肤出汗的自然现象是可能的。健康的皮肤屏障基于水(15%)和脂质(10-15%)间的微妙平衡。健康和正常的角质层含有约15-20%的水。在这些正常条件下,皮肤是柔软的。皮肤的水分含量可以根据其含水量进行皮肤分类。皮肤通常分为四组,即较湿润皮肤(well-moisturized skin)、正常/湿润皮肤、干燥皮肤和非常干燥的皮肤。当角质层中的水含量低于15%,特别是低于10%时,称为干燥皮肤,而当所述含量为5%及以下时,称为非常干燥皮肤。以类似的方式,正常/湿润皮肤在角质层中呈现15-20%的水含量,而在较湿润的皮肤中所述含量高于20%。干燥皮肤定义为上述平衡的失衡,特别是脂质的缺乏和其物理屏障的破坏导致水的损失和细胞成熟度下降。这种持久的皮肤状况的特征是紧绷、发红和鳞屑。保湿化合物通常旨在用神经酰胺或脲来补偿这种表面脂质的缺乏,以减少经表皮的水分流失。
皮肤的水分含量是皮肤科医师和皮肤专家常规评估的标准。目前使用几种技术来确定皮肤的水分含量。这些技术包括,例如使用corneometer,视觉评估或近红外(NIR)光谱(参见例如,综述Khazaka G.,Bioengineering of the skin:water and stratum corneum(2005):249;或Mohamad M.et al.Journal of Physics:ConferenceSeries.Vol.546.No.1.IOP Publishing,2014)。
根据本发明的方法可以确定皮肤的水分含量。因此,在本发明的上下文中,该方法有利地包括其中基于本发明生物标志物的表达水平,将皮肤分类为非常干燥、干燥、正常/湿润或较湿润(well-moisturized)的步骤。
如上所述,本发明人已显示根据本发明的生物标志物在干燥皮肤中过表达。发明人特别揭示,皮肤样品中表达的生物标志物越多,皮肤越干燥。因此,根据本发明的方法特别地可以诊断,即检测干燥皮肤(包括非常干燥的皮肤)。因此,根据特定实施方案,根据本发明的方法包括确定所述至少一种蛋白质的表达水平越高,受试者皮肤越干燥。
有利地,根据本发明的生物标志物的表达水平可以与参考值比较。对于每个生物标志物确定该参考值。其可以通过实验、经验或理论来确定,并设定以获得最佳的特异性和选择性。该参考值可以基于在对照群体中获得的结果来确定,例如在正常/湿润皮肤中,或者在干燥皮肤中。与基于正常/湿润皮肤确定的阈值相似的表达水平将与正常皮肤状况相关,而与该值显著不同,特别是高于该参考值的表达水平将与干燥皮肤状况相关。相反,与基于干燥皮肤确定的阈值相似的表达水平将与干燥皮肤状况相关,而显著不同且特别是低于该参考值的表达水平将与正常/湿润皮肤状况相关。
根据优选实施方案,基于在正常/湿润皮肤中测量的根据本发明的生物标志物的表达水平确定参考值。根据该实施方案,本发明涉及用于确定受试者中皮肤水分含量的方法,所述方法包括以下步骤:
a)在从所述受试者获得的皮肤样品中测量至少一种蛋白质的表达水平,所述蛋白质选自下组:ESYT3、USP7、PPL和LCE1C;
b)将所述至少一种蛋白质的表达水平与在正常/湿润皮肤中确定的参考值进行比较;以及
c)当所述至少一种蛋白质的表达水平高于参考值时,得出结论所述受试者的皮肤干燥或非常干燥。
根据本发明的“皮肤样品”可以是任何皮肤样品,其中可检测和测量根据本发明的蛋白质表达水平。典型地,根据本发明的皮肤样品是角质层样品。角质层样品可通过各种技术获得,包括活组织检查、皮肤刮擦或胶带剥离(参见Michaels,Barry综述,(2002).Chapter 26-Skin Sampling Techniques:Handbook of Topical Antimicrobials andTheir Applications)。根据本发明的样品优选通过使用非侵入性技术获得。根据优选实施方案,所述角质层样品是胶带剥离的角质层样品。典型地,将胶带压在皮肤表面上,随后突然将其除去。该技术通常用于皮肤病学研究领域(参见综述Hughes,A.J.,et al.BritishJournal of Dermatology185.1(2021):26-35)。

候选化妆品化合物可以是任何类型。其可以是天然来源的或可以通过化学合成产生。这可涉及结构上定义的化合物、未表征的化合物或物质,或化合物的混合物的库。
天然化合物包括植物来源的化合物,如植物。优选地,候选化妆品化合物是植物性的(vegetal),优选地选自植物提取物(botanical extracts)。
根据优选实施方案,在根据本发明的筛选方法的上下文中使用的皮肤样品是人造皮肤样品。人造皮肤主要用于化妆品研究领域(参见例如Brohem et al.Pigment cell&melanoma research 24.1(2011):35-50;或Yun et al.Journal of PharmaceuticalInvestigation 48.2(2018):215-223)且可以容易地从专门的制造商获得。
本公开还提供了用于治疗干燥皮肤的方法,所述方法包括用有效量的水合化合物处理根据本发明的方法鉴定为干燥皮肤的步骤。
通过以下非限制性实施例进一步说明本发明。
具体实施方式
实施例1:干燥皮肤生物标志物的鉴定:
对37位35-40岁女性进行了研究,其腿部的皮肤正常(17)或非常干燥(20)。按不同参数对女性皮肤状况进行分类;在皮肤病学对照下,通过使用干燥皮肤的等级标度,通过测量TWEL、电容和通过连续的胶带取样进行目测。胶带的染色可以根据角质细胞在胶带上的分布,它们的数量和颜色强度将不同等级的干燥皮肤分类。进行12条胶带的裂解用于蛋白质组鉴定和定量。这种蛋白质组学表征可以鉴定优先在干燥皮肤表面表达的四种新的生物标志物。
1-材料和方法
队列描述:本研究的目的在于揭示仅涉及干燥皮肤的靶标。为能够识别这些靶标,选择一组具有正常皮肤与非常干燥皮肤的女性。本研究汇集了37名35-40岁,其光型范围为1-3的志愿者,分为两个亚群:一组17名皮肤正常的女性,另一组20名皮肤明显非常干燥的女性。该分布由皮肤科医生进行,该医生评估腿前部的采样区域与其研究当天的视觉外观的关系。摄影支持的确认与生物物理测量(诸如TWEL和电容)相关。
取样方法描述:胶带剥离是一种非侵入性方法,可以除去角质层的第一层。将第一胶带放置在腿前部,然后施加标准重量作为恒定且可重复的压力。施加重量10秒,然后提起。画出胶带的轮廓,以便在相同位置施加随后的胶带。然后,使用夹子以直截了当地动作一次性去除该胶带。丢弃第一胶带,以便对随后的所取量进行标准化。然后,以与第一胶带类似的方式施加和移除下一个胶带。然后,将胶带储存在-80℃下。在3个不同区域上总共取样15个胶带。其中12个用于蛋白提取,并且1个用于形态学染色。
胶带染色:对胶带进行染色以突出每个志愿者的角质细胞的分布和形态。将胶带在室温下解冻,置于37℃的热板上,然后用染料PMS:碱性品红和甲苯胺蓝的混合物覆盖1小时。然后,将染色胶带在连续的自来水浴中漂洗并在露天晾干,然后置于载玻片和薄片(lamella)之间进一步分析。
图像分析:使用AT Turbo载玻片扫描仪进行载玻片扫描,放大倍数为x40,分辨率为0.25μm/pxl。基于颜色反卷积,已开发了两个算法来:
1)测量对染色阳性的组织表面并根据3类比色强度将阳性角质细胞的表面分类。该算法在其阳性组织表面测量中应用比色强度梯度。因此,足以将角质层细胞的表面分为三组:角质细胞弱,中等和强标记。最后,该算法为每个胶带确定对应于角质细胞平均强度的平均光密度的归一化值。用于角质细胞表面测量和根据颜色梯度分类的阈值和参数对于每个胶带是相同的。
2)计数由角质细胞形成的小岛并测量它们的面积。首先,确定分析面积和可能的排除区。然后,应用颜色反卷积以从图像的剩余部分提取阳性组织,从而使其自身摆脱可改变图像分析的伪影。最后,该算法将对小岛进行分割和个体化,从而可对它们进行计数,并测量它们的面积。所分析的总面积与小岛所占据的面积之间的比例可以将每个胶带的小岛密度量化为百分比值。
蛋白质提取:将每个胶带(粘合剂面朝上)放置在其自身的20mL硼硅酸盐闪烁小瓶(Wheaton;VWR)。将胶带浸泡在含有SDS(0.2%),丙二醇(0.5%)和HALT抗蛋白酶混合物(1X)的PBS缓冲液中。将小瓶置于超声浴中60分钟。从每个小瓶中收集细胞裂解物并根据供体的编号汇集。将蛋白质在90%甲醇的溶液中在-20℃下沉淀过夜。通过离心(15000g,20分钟,4℃)收集沉淀的蛋白质并溶解在含有尿素的Tris-HCl缓冲液(8M)中。使用BCA方法确定蛋白质浓度。
蛋白纯化和浓缩:根据FASP方法(过滤辅助样品制备)制备肽提取物。将50μg蛋白质加载到超滤装置(Amicon Ultra 0.5,10kDa MWCO)上,然后还原(二硫苏糖醇)、烷基化(碘乙酰胺)并用胰蛋白酶消化。通过SPE色谱法(C18)纯化肽,干燥并溶解在100μl 0.1%甲酸水溶液中。使用BCA方法测定肽浓度。
蛋白质分析LC-MS/MS《鸟枪法蛋白质组学》:当可能时,对每个样品注射250ng肽,一式三份。对于具有未确定肽浓度的样品,使用10μl未稀释的样品。色谱使用Ultimate3000(Dionex)设备进行,使用C18(75μm×50cm,2μm材料)柱,在预柱上3分钟捕获步骤后,施用2.5%-35%乙腈60分钟梯度,流速300nl/min。使用Q-Exactive Plus(Thermo)质谱仪获得数据。以70000的分辨率和60ms的累积时间进行MS扫描。MS/MS扫描以17500的分辨率对每个循环的10个最强离子进行,60ms的累积时间。进行5600个循环,因此每个色谱峰平均14个循环。应用以下实验设置:源电压(2300V)吹扫气体(0psi)转移管温度(275℃)归一化碰撞能量28*(*碰撞能量根据所分析化合物的质量和电荷自动确定)。
蛋白质鉴定:使用SEQUEST-HT算法和收集从UNIPROT挖掘的人参考蛋白质组的数据库鉴定蛋白质。检索参数为:酶=胰蛋白酶(全);允许的裂解=2;前体误差容限=10ppm;片段误差容限=0.02Da;动态修饰=氧化(M),脱酰胺(N/Q);蛋白末端修饰=乙酰化;静态修饰=氨基甲酰甲基(C)。使用Percolator算法确定错误发现率(FDR)。
蛋白定量:使用Minora和Proteome Discoverer 2.2软件的特征映射器处理数据。峰积分参数为:采集后重新校准=True(精细参数)最小轨迹长度=5;同位素的最小数量=2;同位素的最大ΔRT=0.2min积分的最小PSM置信水平=高。色谱校准参数为:RT校准=TRUE;参数tuning=Fine;最大RT位移=5min质量公差10ppm。特征映射参数为:RT容限(RTtolerance)=自动;质量公差=自动;S/N阈值=2。通过使用用于Proteome Discoverer2.2软件的前体离子量化器节点进行统计分析。一般量化设置为:使用的肽=Unique+RAZOR(Unique=不同蛋白质或蛋白质组不共有的肽;RAZOR=由多个蛋白质组共有但仅用于定量具有最大数目的独特肽和具有最长氨基酸序列的蛋白质的肽);考虑对于肽Uniqueness的蛋白质组=True;拒绝缺少通道的Quan结果=False。前体定量设置为:基于面积的前体丰度;最小数量重复特征=50%(必须在用于定量的一组样品的至少50%中检测肽)。标准化设置:总肽量(计算所有已识别肽的每次注射的丰度值总和,具有最高总丰度的注射用作参考,通过每次注射的恒定因子校正所有其他注射中的丰度值,以便最终所有注射的总丰度相同)。Quan Rollup假设检验设置:比例计算=基于总丰度(蛋白质比例由重复组的总样品丰度的中值计算)。插补模式=基于重复的重采样(缺失值被替换为从以(技术,生物学)重复检测值的中值为中心的分布中采样的随机值)。
假设检验=ANOVA(单个蛋白质)。
2/结果:
胶带染色:形态学分析。
胶带染色提供了广泛的信息,从整个胶带上角质细胞占据的表面到由它们形成的小岛(islet)数目以及相关的比色强度。实际上,干燥皮肤逐渐与胶带上角质细胞所占据的较大表面结合。干燥皮肤也逐渐与高比色浓度的小岛数量增加相关。角质细胞的比色密度和密集分布与皮肤的干燥程度直接相关。
染色胶带的定量分析
除了皮肤科医生进行的分析和所述分析与生物物理测量的相关性之外,还确定了正常皮肤的群体和具有干燥皮肤的人群体。在应用相同的分布之后,将这些结果与关于在整个胶带上获得的小岛的平均尺寸所得的结果相关联。
与正常皮肤相比,干燥皮肤角质细胞占据超过43%的胶带表面。在干燥皮肤中小岛的表面平均增加28%。干燥皮肤呈现高色度强度,比在正常皮肤中观察到的高255%。
干燥皮肤中的蛋白质组学表征:
对干燥和正常皮肤的分析表明,在所鉴定的613种蛋白质中,340种在干燥皮肤中被差异调节。在来自干燥皮肤组和正常皮肤组的样品中分别鉴定出平均227和212个蛋白质。从定量的角度来看,在340种可定量的蛋白质(整理后(after curation))中,88种被认为在敏感皮肤中过表达,而77种被认为表达不足(阈值:p值<0.05,倍数变化>1.5或<0.67)。
干燥皮肤的特异性蛋白质生物标志物:
通过对正常皮肤施用100%的归因百分比(imputation percentage),相比之下,我们在干燥皮肤中获得的百分比非常低。这种低百分比的归因与靶标在干燥皮肤中的表达成反比。因此,这意味着归因百分比越低,靶标在皮肤中表达就越多。这练习可以选择优先在干燥皮肤中表达的目标。仅存在于干燥皮肤中的5个主要靶标是:旁血小板溶蛋白、ESYT3、LCE1C、USP7和IL1RN。
这些不同靶标在角质化细胞水平的过表达可以建立用于研究干燥皮肤的新模型。然后,可评估所选活性成分对这些靶标的作用以评估它们的保湿效果。
实施例2:保湿剂对所选生物标志物的影响:
所述的四种蛋白质的过表达表征干燥皮肤。结果是,减少其过表达的活性成分将使干燥皮肤恢复到正常状态。针对LCE1C测试该假设。确定两种不同浓度0.1%和0.033%的活性绒毛花(Anthyllis)的全身治疗对黑化重建表皮的影响。处理5天后,与未处理的对照相比,LCE1C的表达分别降低4.1和11.8倍。
因此,根据本发明的生物标志物可以准确地诊断干燥皮肤并代表用于评估水合化合物的功效或用于监测水合处理的功效的极其相关的靶标。
Claims (10)
1.用于确定受试者中皮肤水分含量的方法,所述方法包括在从所述受试者获得的皮肤样品中测量至少一种蛋白质的表达水平的步骤,所述蛋白质选自下组:延长的突触结合蛋白-3(ESYT3)、泛素-羧基末端水解酶7(USP7)、旁血小板溶蛋白(PPL)和晚期角质化包膜蛋白1C(LCE1C)。
2.根据权利要求1所述的方法,所述方法还包括其中基于测量的表达水平,将所述皮肤分类为非常干燥、干燥、湿润或较湿润的步骤。
3.根据权利要求1或2的所述方法,其中确定,所述至少一种蛋白质的表达水平越高,受试者的皮肤越干燥。
4.根据权利要求1-3中任一项所述的方法,其中所述方法还包括在从所述受试者获得的所述皮肤样品中测量白介素-1受体拮抗剂蛋白(IL1RN)的水平的步骤。
5.根据权利要求1-4中任一项所述的方法,其中所述皮肤样品是角质层样品。
6.根据权利要求5所述的方法,其中所述角质层样品是胶带剥离的角质层样品。
7.用于评估水合受试者皮肤的美容处理功效的方法,所述方法包括以下步骤:
a)在接受所述美容处理前从所述受试者获得的皮肤样品中测量至少一种蛋白质的表达水平,所述蛋白质选自下组:延长的突触结合蛋白-3(ESYT3)、泛素-羧基末端水解酶7(USP7)、旁血小板溶蛋白(PPL)和晚期角质化包膜蛋白1C(LCE1C);
b)在接受所述美容处理后从所述受试者获得的皮肤样品中测量所述至少一种蛋白质的表达水平;
c)比较在步骤a)和b)中测量的表达水平;以及
d)当在步骤b)中测量的表达水平低于在步骤a)中测量的表达水平时,确定所述美容处理是有效的,或者当在步骤b)中测量的表达水平类似于或高于在步骤a)中测量的表达水平时,确定所述处理是无效的。
8.筛选用于水合皮肤的候选化妆品化合物的方法,其中所述方法包括以下步骤:
a)在皮肤样品中测量至少一种蛋白质的表达水平,所述蛋白质选自下组:延长的突触结合蛋白-3(ESYT3)、泛素-羧基末端水解酶7(USP7)、旁血小板溶蛋白(PPL)和晚期角质化包膜蛋白1C(LCE1C);
b)使测试化合物与所述皮肤样品接触;
c)测量所述皮肤样品中所述至少一种蛋白质的表达水平;以及
d)如果步骤c)中测量的表达水平低于步骤a)中测量的表达水平,则选择所述化合物。
9.根据权利要求8的方法,其中所述候选化妆品化合物是植物提取物。
10.根据权利要求8或9的方法,其中所述皮肤样品是人造皮肤样品。
