Abstract
The invention relates to a medicinal and edible composition with liver protection effect, which is prepared from 10-30% of black beans, 10-30% of oat, 10-30% of millet, 10-30% of astragalus mongholicus, 5-20% of codonopsis pilosula and 10-30% of Chinese angelica.
Description
Technical Field
The invention belongs to the technical field of foods, and particularly relates to a composition with an alcoholic liver injury reducing function, and a preparation method and application of the composition.
Background
The liver is an important organ in the body mainly having metabolism, has important physiological functions such as digestion, detoxification, metabolism, synthesis, secretion and the like, and plays an important role in maintaining metabolism of saccharides, proteins, fat, vitamins, hormone and the like. It can participate in biochemical reactions of organisms in the forms of oxidation, reduction, hydrolysis, combination and the like, and can realize the bioconversion of non-nutritive substances such as medicines, poisons and in-vivo metabolites from inside and outside bodies.
However, due to viral infection, excessive drinking, metabolic disorder, etc., damage to liver cells may be caused, thereby causing liver diseases such as liver abscess, alcoholic liver disease, drug-toxic liver disease, autoimmune hepatitis, etc.
Among them, the incidence of alcoholic liver disease such as alcoholic fatty liver, alcoholic hepatitis, alcoholic liver fibrosis and alcoholic cirrhosis is on the rise, and has become a serious disease affecting the physical and mental health of humans. Alcoholic liver disease has become a health problem worth the attention of the whole population.
The alcoholic liver disease is more progressive or hidden, and part of patients can have no symptoms for a long time, and can not be diagnosed as the alcoholic liver disease until glutamic pyruvic transaminase or glutamic oxalacetic transaminase is increased during physical examination or examination due to other diseases, and further liver protection medicines such as phosphatidylcholine, silybin, ursodeoxycholic acid, bicyclo alcohol, diammonium glycyrrhizinate, N-acetylcysteine and the like are adopted to carry out symptomatic treatment.
However, silybin has side effects such as allergy, nausea, chest distress, etc.; the medicine such as bicyclo-ethanol, N-acetylcysteine and the like has gastrointestinal side effects such as abdominal distension, nausea and the like after being taken; the glycyrrhizic acid preparation has glucocorticoid-like and mineralocorticoid-like effects, and can cause pseudoaldosteronism, raise blood glucose and blood pressure, and reduce blood potassium; moreover, the medicines are not suitable for simultaneous administration so as not to burden the liver or cause adverse reactions due to interaction among medicines.
Therefore, although drug therapy is an important therapeutic strategy for alcoholic liver disease, it is of great importance to reduce the incidence of alcoholic liver disease if early intervention can be performed on alcoholic liver injury.
Drinking has become a food culture in our country, has a unique role in interpersonal interaction, and has penetrated all aspects of social life. Since drinking is unavoidable in daily life, and drug treatment is often symptomatic treatment after explicitly diagnosing liver diseases and also has the problem of adverse drug reactions, how to protect the liver to the greatest extent without using drugs after drinking is clearly the best choice for a reasonable diet with liver protection function.
For example, schisandrin A in schisandra has been shown to regulate alcohol metabolism and inhibit liver oxidation and inflammatory injury; the water extract of the medlar polysaccharide plays a role in protecting liver in various liver disease models; the methanol extract of Corni fructus can improve alcohol-induced oxidative stress and lipid accumulation to exert liver protecting activity; the phenolic extract of litchi fruit has antioxidant activity, and can improve mitochondrial dysfunction.
Although there is a great deal of evidence that many medicinal and edible materials or foods have liver protecting function, most of the research is focused on a certain active ingredient or effective part of a traditional Chinese medicine or food, and reports on whether organic combinations of several materials or materials have or promote liver protecting activity are less. In addition, the current study subjects are single and are not enough to meet the nutrition collocation of daily diets.
Therefore, the food and the medicine-food homologous traditional Chinese medicine are reasonably combined, and the nutrition elements required by daily reasonable diet are satisfied while the pharmaceutical activity is exerted, so that the method has important significance for improving alcoholic liver diseases.

The invention is unexpectedly found in the food research process, when a plurality of foods are combined with the medicinal and edible traditional Chinese medicines, the antioxidant and anti-inflammatory activities of the foods can be obviously improved, and the oxidation and the inflammation are the important pathogenesis of alcoholic liver diseases, so that the medicinal and edible composition with the liver protection effect is obtained.
Specifically, the medicinal and edible composition with the liver protection effect is prepared from the following raw materials in percentage by mass: 10-30% of black beans, 10-30% of oat, 10-30% of millet, 10-30% of astragalus mongholicus, 5-20% of codonopsis pilosula and 10-30% of Chinese angelica.
Further, in the medicinal and edible composition with liver protection effect, the mass percentages of the raw materials are as follows: 20-30% of black beans, 10-20% of oat, 20-30% of millet, 10-20% of astragalus mongholicus, 5-20% of codonopsis pilosula and 10-25% of Chinese angelica.
Further, the mass percentages of the various raw materials are as follows: 20-28% of black beans, 10-15% of oat, 20-28% of millet, 10-15% of astragalus mongholicus, 6-20% of codonopsis pilosula and 12-24% of Chinese angelica.
The medicine and food homologous composition with the liver protection effect is preferably an active component obtained by extracting the raw materials with the mass percentage by water.
More specifically, the medicinal and edible composition with liver protection effect is prepared by adding 5-10 times of water into raw materials of black beans, oat, millet, astragalus mongholicus, codonopsis pilosula and angelica in percentage by mass, soaking for 10-15 hours, heating to 80-120 ℃ for extraction for 60-80min, and filtering and collecting filtrate; extracting the residue with the same method for 2 times, and collecting filtrate; mixing the filtrates for 3 times, and concentrating to obtain water extract.
Further, various food acceptable auxiliary materials can be added into the obtained water extract, and the water extract can be further prepared into various foods, such as, but not limited to, brewing powder, oral liquid and the like.
The medicine-food homologous composition with the liver protection effect can also be prepared by directly decocting the raw materials with the mass percentage content in water.
The medicinal and edible composition with liver protection function takes black beans, oat and millet which are conventional health foods as raw materials, and Chinese medicaments such as astragalus root, pilose asiabell root and Chinese angelica are added, so that the effects of health functions such as antioxidation and anti-inflammatory activity are greatly improved, particularly, the medicinal and edible composition has remarkable improvement effect on alcoholic liver injury, can be used as an auxiliary product for drinking for long-term drinking, and can play a role in protecting alcoholic liver diseases caused by long-term alcohol intake.

The medicinal and edible composition with the liver protection effect in the embodiment is prepared from the following raw materials in percentage by mass: 20-28% of black beans, 10-15% of oat, 20-28% of millet, 10-15% of astragalus mongholicus, 6-20% of codonopsis pilosula and 12-24% of Chinese angelica.
The preparation method comprises the steps of extracting the raw materials with water to obtain an active component, and adding food acceptable auxiliary materials into the active component to prepare preparations such as brewing powder, oral liquid and the like; or is prepared by directly decocting the raw materials with water according to the mass percentage.
The active components are prepared by weighing raw materials of black beans, oat, millet, astragalus mongholicus, codonopsis pilosula and angelica sinensis according to the mass percentage, adding 5-10 times of water, soaking for 10-15 hours, heating to 80-120 ℃ and extracting for 60-80min, and filtering and collecting filtrate; extracting the residue with the same method for 2 times, and collecting filtrate; mixing the filtrates for 3 times, and concentrating to obtain water extract.
Examples
Example 1
Weighing 40g of black beans, 20g of oat, 40g of millet, 20g of astragalus mongholicus, 10g of codonopsis pilosula and 20g of angelica sinensis, and soaking in water with the volume of 5 times for 12 hours; extracting at 100deg.C for 70min, filtering, and collecting filtrate; extracting the residue for 2 times, and collecting filtrate.
Combining the three filtrates, concentrating to 80mL to obtain the medicinal and edible homologous composition water extract with liver protecting effect.
Example 2
Weighing 40g of black beans, 20g of oat, 40g of millet, 20g of astragalus membranaceus, 30g of codonopsis pilosula and 40g of angelica sinensis, adding 5 times of water, and decocting with small fire for 1h to directly prepare the medicinal and edible composition with liver protection effect.
Example 3
Weighing 40g of black beans, 20g of oat, 40g of millet, 20g of astragalus mongholicus, 40g of codonopsis pilosula and 40g of angelica sinensis, and soaking in water with the volume of 5 times for 12 hours; extracting at 100deg.C for 70min, filtering, and collecting filtrate; extracting the residue for 2 times, and collecting filtrate.
Mixing the three filtrates, concentrating, adjusting pH to 6-8 with citric acid, adding a little xylitol for flavoring, filtering, quantifying to 70mL with purified water, sterilizing, and bottling according to 10 mL/bottle standard to obtain medicinal and edible composition oral liquid with liver protecting effect.
Example 4
According to the preparation method of the example 1, the aqueous extracts of the compositions of black beans, oat and millet (4:2:4) and the aqueous extracts of the compositions of astragalus, dangshen and angelica (2:1:2) are respectively prepared, and animal experiments are carried out on the aqueous extracts of the compositions of the medicine and food homology with the liver protection prepared in the example 1, so as to examine the liver protection effect of the composition of the medicine and food homology with the liver protection effect on alcoholic liver injury of rats.
SD male rats are randomly divided into a blank group, a model group, a food group, a traditional Chinese medicine group, a dosage group in the medicinal and edible composition, and 6 groups of high-dosage groups of the medicinal and edible composition, wherein 8 groups of the high-dosage groups are respectively arranged.
After the animals are adaptively raised for one week, the other groups except the blank group adopt a gradient ethanol method to irrigate the stomach to administer 52% of red star Erguotou, namely 4mL/kg/d for week 1, then sequentially increment 2mL/kg/d every week until the dosage is increased to 10mL/kg/d, and then molding is continued for 5 weeks for 8 weeks in total.

24h after the last gastric lavage, the uratam anesthetized rats, the abdominal aorta was bled, centrifuged at 3000rpm for 10min, the supernatant was aspirated, and the expression levels of ALT, AST, SOD, MDA, IL-6, TNF-alpha and IL-10 in the serum were determined according to the kit instructions, and their effects on oxidation and inflammation in alcoholic liver injury were evaluated.
In Table 1, after the black beans, the oat and the millet compositions or the astragalus, the dangshen and the angelica compositions are singly administered, the liver function indexes ALT and AST of rats are not remarkably improved, and the oxidation injury and the inflammatory injury of livers are not remarkably influenced, but after the medicinal and edible homologous composition with the liver protection function is used, all indexes such as liver function, oxidation and inflammation are basically recovered to normal levels.

1. A medicinal and edible composition with liver protection effect is prepared from the following raw materials in percentage by mass: 10-30% of black beans, 10-30% of oat, 10-30% of millet, 10-30% of astragalus mongholicus, 5-20% of codonopsis pilosula and 10-30% of Chinese angelica.

The medicinal and edible composition with liver protection effect according to claim 1, wherein the mass percentages of the raw materials are as follows: 20-30% of black beans, 10-20% of oat, 20-30% of millet, 10-20% of astragalus mongholicus, 5-20% of codonopsis pilosula and 10-25% of Chinese angelica.
