Abstract
The invention provides a lentinan composite polysaccharide composition for improving immunity and a preparation method thereof, wherein the lentinan composite polysaccharide composition comprises the following raw materials in parts by weight: 25-40 parts of mushroom extract, 8-16 parts of lucid ganoderma extract, 15-30 parts of cordyceps militaris extract, 25-40 parts of astragalus extract, 4-12 parts of codonopsis pilosula extract, 4-12 parts of garlic extract, 1-5 parts of bifidobacterium powder, 0.005-0.01 part of vitamin A, 0.0001-0.0005 part of vitamin E, 0.01-0.03 part of vitamin C, 0.01-0.02 part of ferric gluconate and 0.001-0.002 part of zinc gluconate. The lentinus edodes compound polysaccharide composition provided by the invention has a good effect of improving the immunity of an organism, and has the effects of protecting the liver, detoxifying, relieving fatigue, increasing beneficial microbial flora in intestinal tracts and improving the intestinal tract function.
Description
Technical Field
The invention relates to the technical field of traditional Chinese medicine compositions, in particular to a lentinus edodes compound polysaccharide composition for improving immunity and a preparation method thereof.
Background
Immunity refers to the body's own ability to defend against diseases. It is the physiological response of identifying and eliminating invading foreign viruses, bacteria, fungi, etc., processing abnormal own cells, and identifying and eliminating "foreigners" by the human body. Low immunity means that the body's immune function is imbalanced or the immune system is not sound, and the body's defense capabilities are reduced. The "Emperor's Internal Path" records that "righteous qi is stored in the body, evil cannot be done, and where the evil is, the qi must be deficient." Righteous qi refers to the body's own immunity. Traditional Chinese medicine believes that "the insufficiency of righteous qi and the invasion of evil qi, and the imbalance of yin and yang of qi and blood" is the root cause of the weakened immune system. The Chinese patent publication No. CN107510059A discloses a preparation method and application of a polysaccharide composition, wherein the polysaccharide composition consists of black bean polysaccharide and oat polysaccharide, and the composition prepared by the invention has nutritive value, and has the functions of improving blood sugar regulation function and enhancing immunity.
However, the polysaccharide composition disclosed in the technical scheme has the defects of few polysaccharide source components and poor interaction between compound polysaccharides; the preparation process can not obtain high-purity polysaccharide, has the defects of single function and poor immunity enhancing function, and can not achieve the purpose of promoting immunity improvement by multiple purposes of one medicine.
Disclosure of Invention
The lentinus edodes compound polysaccharide composition has multiple polysaccharide source components, high polysaccharide purity and multi-path interaction, particularly strengthens the immune excitation effect of garlicin and improves the intestinal function, has good functions of enhancing immunity and multi-path metabolic regulation, and achieves the aim of multiple purposes of one medicine.
In order to achieve the purpose, the invention is realized by the following technical scheme:
the lentinan composite polysaccharide composition for improving the immunity comprises the following raw materials in parts by weight: 25-40 parts of mushroom extract, 8-16 parts of lucid ganoderma extract, 15-30 parts of cordyceps militaris extract, 25-40 parts of astragalus extract, 4-12 parts of codonopsis pilosula extract, 4-12 parts of garlic extract, 1-5 parts of bifidobacterium powder, 0.005-0.01 part of vitamin A, 0.0001-0.0005 part of vitamin E, 0.01-0.03 part of vitamin C, 0.01-0.02 part of ferric gluconate and 0.001-0.002 part of zinc gluconate.
Preferably, the lentinan composition comprises the following raw materials in parts by weight: 30-35 parts of mushroom extract, 10-15 parts of lucid ganoderma extract, 20-25 parts of cordyceps militaris extract, 30-35 parts of astragalus extract, 5-10 parts of codonopsis pilosula extract, 5-10 parts of garlic extract, 2-3 parts of bifidobacterium powder, 0.007-0.008 part of vitamin A, 0.0002-0.0003 part of vitamin E, 0.02-0.03 part of vitamin C, 0.015-0.02 part of ferric gluconate and 0.0015-0.002 part of zinc gluconate.
Preferably, the garlic extract is allicin.
The preparation method of the lentinus edodes compound polysaccharide composition for improving the immunity comprises the following steps:
(1) weighing 30-35 parts of shiitake mushroom, 10-15 parts of lucid ganoderma, 20-25 parts of cordyceps militaris and 30-35 parts of astragalus mongholicus according to parts by weight, crushing into 30-mesh coarse powder, and mixing to obtain mixture powder for later use;
(2) adding water into the mixture powder for extraction twice, wherein the mixture powder is extracted for the first time according to the mass ratio of M: adding water in a ratio of M water to 10, extracting for 2 hours under reflux, and taking primary filtrate; filtering residues for the second time according to the mass ratio M: adding water in a ratio of M water to 8: 1, extracting for 1.5h under reflux, taking secondary filtrate, and combining the two filtrates;

(5) weighing 5-10 parts of codonopsis pilosula according to parts by weight, cutting up the codonopsis pilosula, and then weighing the codonopsis pilosula in a mass ratio of M codonopsis pilosula: adding 95% ethanol into the mixture with the ratio of M ethanol to 1:6, extracting for 12h at the temperature of 40-50 ℃, filtering, washing filter residues with 95% ethanol for three times, recovering ethanol, and freeze-drying to obtain codonopsis pilosula ethanol extract freeze-dried powder;
(6) and (4) volatilizing ethanol from the codonopsis pilosula residue in the step (4), carrying out hot reflux water extraction, and mixing the medicine residues according to the mass M: extracting with water at ratio of 1:10 under reflux for 2 times, each time for 1 hr, mixing extractive solutions, concentrating under reduced pressure to relative density of 1.0-1.1, lyophilizing to obtain radix Codonopsis water extract lyophilized powder, and mixing the water extract lyophilized powder with ethanol extract lyophilized powder to obtain radix Codonopsis extract;
(7) and (2) uniformly mixing the lentinus edodes compound polysaccharide extract obtained in the step (1), the fresh garlic extract, the codonopsis pilosula extract and the rest other substances according to a formula ratio, and granulating to obtain the lentinus edodes compound polysaccharide composition.
Preferably, the pore size of the reverse osmosis concentration in the step (4) is 0.1-0.5 nm.
The lentinus edodes is a medicine-food homologous plant, has the effects of tonifying qi, not hunger, treating wind, breaking blood, benefiting stomach and assisting food, and is proved by modern medical research to contain a large amount of polysaccharide, protein, amino acid, multiple vitamins and the like, wherein the lentinus edodes polysaccharide is a key substance for improving the immunity, and the humoral immunity function of a human body is enhanced by improving the activity of auxiliary T cells. Typical cases

Experiment of
1. Experimental Material
Kunming mice, subject drug was the pharmaceutical composition prepared according to example 1. Shengmai oral liquid. Leukocyte diluent: prepared according to the proportion of glacial acetic acid 2ml distilled water 98ml and 1 percent methylene blue 2 drops before use.
2. Experimental methods and results
(1) Grouping and administration: kunming mice, male and female, each (15 + -2) g, were randomly divided into 8 groups of 10 mice each, namely normal group, high, medium and low dosage groups of tested drugs, pulse-activating oral liquid group, astragalus polysaccharide group and ganoderma polysaccharide group. The drug was dissolved or diluted with distilled water and each group of mice was gavaged at 0.5 ml/mouse/day. A test drug high dose group (1.20g/kg), a test drug medium dose group (0.60g/kg), a test drug low dose group (0.30g/kg), a ganoderma lucidum polysaccharide group (0.60g/kg), and an astragalus polysaccharide group (0.60 g/kg). The mixture is suspended by distilled water and then is intragastrically filled by 0.5 ml/tube. The dosage of the pulse-activating oral liquid is converted into 0.0026 of the dosage of adults; the normal group was also gavaged with distilled water at a rate of 0.5 ml/mouse. Animals were sacrificed 1 time daily for 7 consecutive days at 8 d. 20 μ L of blood was collected from the animal orbit and added to 380 μ L of leukocyte dilution, and leukocytes were counted under the microscope. Thymus and spleen of the mice were harvested, and after residual blood was blotted with filter paper, the weight (mg) was weighed and divided by the weight (g) of the mice, respectively, to obtain thymus index and spleen index.
The above indexes are calculated as mean and Standard Deviation (SD), statistical software SPSS11.0 is used for data analysis, and LSD method is used for comparison among groups, and the results are shown in Table 5.
(2) As a result: compared with the normal group, the pulse-activating oral liquid and each tested drug group have statistical significance on the influence of mouse peripheral leukocyte proliferation, and the astragalus polysaccharide group and the ganoderma polysaccharide group also have statistical significance on the influence of mouse peripheral leukocyte proliferation, but are weaker than the tested drug group (p is less than 0.05). Each dose group and pulse generation group of the tested medicaments can effectively inhibit the atrophy of the thymus of the mouse (p < 0.05); the tested drug dose groups and the pulse-activating oral liquid group have the effect of promoting the proliferation of the spleen of a mouse (p < 0.01). The experimental result shows that the tested medicine has the function of enhancing the immune function of mice, and the function is obviously stronger than that of the astragalus polysaccharide group and the ganoderma lucidum polysaccharide group.

(1) weighing 30-35 parts of shiitake mushroom, 10-15 parts of lucid ganoderma, 20-25 parts of cordyceps militaris and 30-35 parts of astragalus mongholicus according to parts by weight, crushing into 30-mesh coarse powder, and mixing to obtain mixture powder for later use;

(4) Weighing 5-10 parts of fresh garlic according to parts by weight, cutting into small fragments, and mixing the fresh garlic with the small fragments according to a mass ratio of M garlic: adding 75% ethanol into 1:2, homogenizing with colloid mill for 30min, standing for 2 hr, centrifuging to separate Bulbus Allii tissue residue, concentrating the supernatant by reverse osmosis, recrystallizing at 15-25 deg.C to obtain fresh Bulbus Allii extract;
(5) weighing 5-10 parts of codonopsis pilosula according to parts by weight, chopping, and then mixing the codonopsis pilosula and ginseng according to a mass ratio of M: adding 95% ethanol into 1:6, extracting at 40-50 deg.C for 12h, filtering, washing the residue with 95% ethanol for three times, recovering ethanol, and freeze drying to obtain ethanol extract lyophilized powder;
(6) and (5) volatilizing ethanol from the codonopsis pilosula residue in the step (5), carrying out hot reflux water extraction, and mixing the medicine residues according to the mass M: extracting with water at ratio of 1:10 under reflux for 2 times, each time for 1 hr, mixing extractive solutions, concentrating under reduced pressure to relative density of 1.0-1.1, lyophilizing to obtain radix Codonopsis water extract lyophilized powder, and mixing the water extract lyophilized powder with ethanol extract lyophilized powder to obtain radix Codonopsis extract lyophilized powder;
(7) and (2) uniformly mixing the lentinan composite polysaccharide extract obtained in the step (1), the fresh garlic extract, the codonopsis pilosula extract and the rest other substances according to a formula ratio, and granulating to obtain the lentinan composition.
5. The method for preparing lentinan complex composition for improving immunity as claimed in claim 4, wherein the reverse osmosis concentration pore size in the step (4) is 0.1-0.5 nm.
