Abstract
本发明提供了调控HMGN5的活性分子在恶性肿瘤增殖和转移中的应用。本发明提供了靶向并高效抑制HMGN5的人工siRNA(siHMGN5)或其前体序列。实验表明,本发明的siRNA靶向HMGN5,并显著抑制HMGN5的转录与翻译,在细胞与动物水平调控乳腺癌等HMGN5高表达肿瘤的增殖、转移和迁移侵袭。乳腺癌裸鼠移植瘤模型中进一步证实了本发明siRNA可显著抑制裸鼠的肿瘤生长以及乳腺癌转移。本发明的siHMGN5可以用于治疗乳腺癌等HMGN5高表达肿瘤。

技术领域
本发明属于生物医药领域,具体地涉及调控HMGN5的活性分子在恶性肿瘤增殖和转移中的应用。
背景技术
RNA干扰(RNAi)在实验室中是一种强大的实验工具,利用具有同源性的双链RNA(dsRNA)诱导序列特异的目标基因的沉寂,迅速阻断基因活性。siRNA在RNA沉默通路中起中心作用,是对特定信使RNA(mRNA)进行降解的指导要素。
siRNA是RNAi途径中的中间产物,是RNAi发挥效应所必需的因子。siRNA的形成主要由Dicer和Rde-1调控完成。由于RNA病毒入侵、转座子转录、基因组中反向重复序列转录等原因,细胞中出现了dsRNA,Rde-1(RNAi缺陷基因-1)编码的蛋白质识别外源dsRNA,当dsRNA达到一定量的时候,Rde-1引导dsRNA与Rde-1编码的Dicer(Dicer是一种RNaseIII活性核酸内切酶,具有四个结构域:Argonaute家族的PAZ结构域,III型RNA酶活性区域,dsRNA结合区域以及DEAH/DEXHRNA解旋酶活性区)结合,形成酶-dsRNA复合体。Dicer切割后形成siRNA,然后,在ATP的参与下,细胞中存在的一种RNA诱导的沉默复合体RNA-inducedsilencing complex RNAi干涉的关键步骤是组装RISC和合成介导特异性反应的siRNA蛋白。siRNA并入RISC中,然后与靶标基因编码区或UTR区完全配对,降解靶标基因,因此说siRNA只降解与其序列互补配对的mRNA。其调控的机制是通过互补配对而沉默相应靶位基因的表达,所以是一种典型的负调控机制。
siRNA识别靶序列是有高度特异性的,因为降解首先在相对于siRNA来说的中央位置发生,所以这些中央的碱基位点就显得极为重要,一旦发生错配就会严重抑制RNAi的效应。siRNA作为一种新兴的治疗技术,也已经以一种前所未有的速度迈进了临床试验阶段。
高迁移率族核小体结合蛋白5(HMGN5)是高迁移率核小体结合蛋白(highmobility group nucleosome binding protein,HMGN)家族成员之一,通过高度保守的核小体结合结构域(NBD)与核小体结合,从而调节染色质结构,进而影响下游基因的转录,研究表明HMGN5参与包括细胞增殖、凋亡、自噬、迁移、侵袭和耐药等多种与肿瘤发生发展相关的生物学过程,在肿瘤发生发展中发挥重要的作用。HMGN5在多种人类肿瘤组织中呈现高表达,并被发现与乳腺癌,脑癌,前列腺癌,膀胱癌等肿瘤的病理分级,临床分期和预后等相关联,因此,HMGN5是一个具有开发潜力的癌症治疗的新靶点。
然而,目前虽然已经开发了一些针对HMGN5的抑制剂,然而其抑制效果尚难以令人满意,因此本领域迫切需要开发新的高效抑制HMGN5的抑制剂。
发明内容
本发明的目的就是提供一种高效抑制HMGN5的抑制剂及其在抑制恶性肿瘤增殖和转移的药物中的用途。
在本发明的第一方面,提供了一种人工siRNA或其前体序列,所述的siRNA靶向高迁移率族核小体结合蛋白5(HMGN5)并抑制HMGN5表达,
其中,所述siRNA对HMGN5表达的抑制率≥85%。

具体实施方式
本发明人经过广泛而深入地研究,首次设计制备了一类能够高效抑制HMGN5表达的siRNA及其前体序列。研究表明,本发明的高效抑制HMGN5表达的siRNA(简称为“siHMGN5”)可以极其有效地抑制HMGN5的表达,与阴性对照相比,本发明siHMGN5对mRNA表达的抑制率≥85%,甚至接近约99.8%,可用于治疗多种不同的HMGN5高表达的恶性肿瘤。在此基础上,完成了本发明。
具体地,本发明人在细胞水平评价了siHMGN5的抗肿瘤活性,采用人乳腺癌细胞株MDA-MB-231和T47D,转染siHMGN5序列,观察siHMGN5对细胞增殖、迁移和侵袭的作用,结果表明,siHMGN5可以高效抑制MDA-MB-231和T47D的细胞活力以及迁移侵袭能力。在动物水平的活性评价中,分别利用慢病毒建立MDA-MB-231和T47D的shHMGN5稳转细胞株,分别将shHMGN5稳转细胞株和对应的对照细胞株在裸鼠上进行皮下注射,发现无论是MDA-MB-231还是T47D的shHMGN5稳转细胞株的皮下移植瘤的生长速度相比对照组均显著减缓。此外,将MDA-MB-231的shHMGN5稳转细胞株和对照细胞株分别对裸鼠进行尾静脉注射,发现shHMGN5稳转细胞株体内肺转移概率降低并且程度减弱。此外,利用载体包裹siHMGN5对荷瘤小鼠进行尾静脉给药,也对小鼠乳腺癌皮下移植瘤生长具有显著抑制作用。
术语
除非另外定义,否则本文中所用的全部技术与科学术语均具有如本发明所属领域的普通技术人员通常理解的相同含义。如本文所用,在提到具体列举的数值中使用时,术语“约”意指该值可以从列举的值变动不多于1%。例如,如本文所用,表述“约100”包括99和101和之间的全部值(例如,99.1、99.2、99.3、99.4等)。
如本文所用,术语“有效量”可指为实现预期的效果所需的剂量和时段的有效的量。此有效量可能因某些因子而产生不同的变化,如疾病的种类或治疗时疾病的病症、被施用的特定标的器官的构造、病人个体大小、或疾病或症状的严重性。本领域具有通常知识者不需要过度实验即可凭经验决定特定化合物的有效量。
如本文所用,术语“本发明的siRNA”、“本发明的siHMGN5”、“本发明的靶向HMGN5的siRNA”可互换使用,指出本发明第一方面中所述的靶向靶向高迁移率族核小体结合蛋白5(HMGN5)并抑制HMGN5表达的siRNA。
siRNA及其前体
如本文所用,所述的“siRNA”是指一类RNA分子,从可形成siRNA前体的转录物加工而来。成熟的siRNA通常具有18-26个核苷酸(nt)(更特别的约19-22nt),也不排除具有其它数目核苷酸的siRNA分子。siRNA通常可被Northern印迹检测到。
人来源的siRNA可被从人细胞中分离。如本文所用,“分离的”是指物质从其原始环境中分离出来(如果是天然的物质,原始环境即是天然环境)。如活体细胞内的天然状态下的多聚核苷酸和多肽是没有分离纯化的,但同样的多聚核苷酸或多肽如从天然状态中同存在的其他物质中分开,则为分离纯化的。
优选地,本发明的siRNA在正义链和/或反义链在其3'端具有额外的0、1或2个核苷酸Z,所述的核苷酸Z选自胞嘧啶C、鸟嘌呤G、腺嘌呤A、胸腺嘧啶T、脱氧胞嘧啶dC、脱氧鸟嘌呤dG、脱氧腺嘌呤dA或脱氧胸腺嘧啶dT。
siRNA可从前体siRNA加工而来,所述的前体siRNA可折叠成一种稳定的茎环(发夹)结构,所述的茎环结构长度一般在50-100bp之间。所述的前体siRNA可折叠成稳定的茎环结构,茎环结构的茎部两侧包含基本上互补的两条序列。
在本发明中,所述的前体siRNA为人工合成的前体siRNA,且所述的前体siRNA具有式I所示的结构:
作为代表性的例子,B1为HMGN5siRNA正义链序列(或核心序列);
B2为与B1互补(包括基本互补和完全互补)的序列;
C为茎环结构序列;
A1和A2分别为无,或任选的由4个-5个碱基组成的核苷酸序列;
其中,所示的前体siRNA能在宿主中加工形成HMGN5siRNA。
优选地,在式I中,B2和B1为基本互补。如本文所用,“基本上互补”是指核苷酸的序列是足够互补的,可以以一种可预见的方式发生相互作用,如形成二级结构(如茎环结构)。通常,两条“基本上互补”的核苷酸序列互相之间至少有70%的核苷酸是互补的;优选的,至少有80%的核苷酸是互补的;更优选的,至少有90%的核苷酸是互补的。一般地,两条足够互补的分子之间可以具有最多8个不匹配的核苷酸,优选地,具有1、2、3、4、5个不匹配的核苷酸。
如本申请所用,“茎环”结构也被称作“发夹”结构,是指一种核苷酸分子,其可形成一种包括双链区域(茎部)的二级结构,所述的双链区域由该核苷酸分子的两个区域(位于同一分子上)形成,两个区域分列双链部分的两侧;其还包括至少一个“环”结构,包括非互补的核苷酸分子,即单链区域。即使该核苷酸分子的两个区域不是完全互补的,核苷酸的双链部分也可保持双链状态。例如,插入、缺失、取代等可导致一个小区域的不互补或该小区域自身形成茎环结构或其它形式的二级结构,然而,该两个区域仍可基本上互补,并在可预见的方式中发生相互作用,形成茎环结构的双链区域。茎环结构是本领域技术人员所熟知的,通常在获得了一条具有一级结构的核苷酸序列的核酸后,本领域技术人员能够确定该核酸是否能形成茎环结构。
本发明中,“茎环结构”可存在于式I所示前体siRNA的末端,例如由于B1和B2形成基本互补后,C会形成一固定的末端茎环结构;所述的“茎环结构”还可存在于式I所式前体siRNA内部,例如由于B1和B2之间并非完全互补,造成未互补结合的B1或B2的碱基会形成一内部的茎环(internal loop)。
根据本发明所提供的siRNA序列,可设计出在被导入后可被加工成可影响相应的mRNA表达的siRNA的多核苷酸构建物,也即所述多核苷酸构建物能够在体内上调相应的HMGN5siRNA的量,从而降低HMGN5的表达量。因此,本发明提供了一种分离的多核苷酸(构建物),所述的多核苷酸(构建物)可被人细胞转录成前体siRNA,所述的前体siRNA可被人细胞剪切且表达成所述的siRNA。
多核苷酸构建物
作为本发明的一种优选方式,所述的多核苷酸构建物自5'至3'端含有式II所示的结构:

通常,所述的多核苷酸构建物位于表达载体上。因此,本发明还包括一种载体,它含有所述的siRNA,或所述的多核苷酸构建物。所述的表达载体通常还含有启动子、复制起点和/或标记基因等。本领域的技术人员熟知的方法能用于构建本发明所需的表达载体。这些方法包括体外重组DNA技术、DNA合成技术、体内重组技术等。所述的表达载体优选地包含一个或多个选择性标记基因,以提供用于选择转化的宿主细胞的表型性状,如卡拉霉素、庆大霉素、潮霉素、氨苄青霉素抗性。
在本发明中,所述表达载体没有特别限制,包括市售的或用常规制备的表达载体。代表性的例子包括(但并不限于):pcDNATM6.2-GW/miR、pcDNA3、pMIR-REPORT miRNA、pAdTrack-CMV、pCAMBIA3101+pUC-35S、pCMVp-NEO-BAN、pBI121、pBin438、pCAMBIA1301、pSV2、CMV4表达载体、GV317、GV309、或其他GV系列表达载体。
在另一优选例中,在所述表达载体中,与所述表达所述前体siRNA多核苷酸操作性相连的启动子包括组成型启动子或组织特异性启动子,优选在肝脏组织中特异性启动的启动子。换言之,这些启动子用于驱动前体siRNA的表达。
代表性的启动子包括(但并不限于):Pcmv启动子、U6、H1、CD43启动子、CD45(LCA)启动子、CD68启动子、Endoglin(CD105)启动子、Fibronectin启动子、Flt-1(VEGFR-1)启动子、GFAP启动子、GPIIb(IntegrinαIIb)启动子、ICAM-2(CD102)启动子、MB(Myoglobin)启动子、NphsI(Nephrin)启动子、SPB启动子、SV40/hAlb启动子、SYN1启动子、WASP启动子或其组合。
药物组合物及施用方法
如本文所用,术语“有效量”或“有效剂量”是指可对人和/或动物产生功能或活性的且可被人和/或动物所接受的量。
如本文所用,术语“药学上可接受的”的成分是适用于人和/或哺乳动物而无过度不良副反应(如毒性、刺激和变态反应)的,即具有合理的效益/风险比的物质。术语“药学上可接受的载体”指用于治疗剂给药的载体,包括各种赋形剂和稀释剂。
本发明的药物组合物含有安全有效量的本发明的活性成分以及药学上可接受的载体。这类载体包括(但并不限于):盐水、缓冲液、葡萄糖、水、甘油、乙醇、及其组合。通常药物制剂应与给药方式相匹配,本发明的药物组合物的剂型为注射剂、口服制剂(片剂、胶囊、口服液)、透皮剂、缓释剂。例如用生理盐水或含有葡萄糖和其他辅剂的水溶液通过常规方法进行制备。所述的药物组合物宜在无菌条件下制造。
在本发明中,一种优选的药物组合物为脂质体制剂。
本发明所述的活性成分的有效量可随给药的模式和待治疗的疾病的严重程度等而变化。优选的有效量的选择可以由本领域普通技术人员根据各种因素来确定(例如通过临床试验)。所述的因素包括但不限于:所述的活性成分的药代动力学参数例如生物利用率、代谢、半衰期等;患者所要治疗的疾病的严重程度、患者的体重、患者的免疫状况、给药的途径等。通常,当本发明的活性成分每天以约0.00001mg-50mg/kg动物体重(较佳的0.0001mg-10mg/kg动物体重)的剂量给予,能得到令人满意的效果。例如,由治疗状况的迫切要求,可每天给予若干次分开的剂量,或将剂量按比例地减少。
本发明所述的药学上可接受的载体包括(但不限于):水、盐水、脂质体、脂质、微粒子(micro particle)、微泡(micro vesicle)、外泌体(exosomes)、脱落囊泡(sheddingvesicle)、纳米胶囊(Nanocapsules/Nanoparticles)、β环糊精胶囊(β-cyclodextriniclusion compound)蛋白、蛋白-抗体缀合物、肽类物质、纤维素、纳米凝胶、或其组合。载体的选择应与给药方式相匹配,这些都是本领域的普通技术人员所熟知的。
在本发明中,可将所述表达载体或siRNA直接施用于对象,也可将所述表达载体与药学上可接受的载体制备成药物组合后进行施用。优选的施用包括静脉注射。
治疗方法
本发明还提供了一种治疗HMGN5siRNA表达量相关疾病的方法,即,将安全有效量的本发明表达载体或药物组合物施用于所需对象,从而治疗HMGN5活性相关的疾病。
通常,“HMGN5siRNA表达量相关疾病”指的是患有所述疾病的患者中,HMGN5蛋白或mRNA的表达量(或活性)E1与正常组织(或癌旁组织)中HMGN5的量(或活性)E0相比具有显著性差异。较佳地,所述“高表达”指的是E1≥1.5E0,更佳地E1≥2E0。
可以通过常规方法检测HMGN5在肿瘤组织中是否高表达。通常,所述的HMGN5高表达肿瘤包括(但不限于):乳腺癌、脑癌、前列腺癌、膀胱癌、卵巢癌、子宫癌、结直肠癌、骨肉瘤、肺癌、胰腺癌。
本发明的主要优点包括:
(a)本发明的siHMGN5可以极其有效地抑制HMGN5的表达,与阴性对照相比,本发明siHMGN5对mRNA表达的抑制率≥85%,甚至接近约99.8%
(b)本发明的siHMGN5可以高效抑制乳腺癌细胞株MDA-MB-231和T47D的克隆形成能力,并在裸鼠移植瘤模型中显著抑制裸鼠的肿瘤生长。
(c)本发明的siHMGN5可以抑制乳腺癌细胞株的迁移侵袭能力,并在免疫缺陷鼠肺转移模型中显著抑制肿瘤肺转移。

本发明在乳腺癌细胞系MDA-MB-231和T47D中转入表1所示siHMGN5,通过实时荧光定量PCR检测siHMGN5对HMGN5mRNA的作用。

1)siHMGN5转染乳腺癌细胞:A、将100μL无血清培养基Opti-MEM(购自ThermoFisher Scientific公司,美国),1μL RNAimax(购自Invitrogen公司,美国)和30nM的siHMGN5混合避光静置20分钟作为反式转染的转染液加到24孔板的1个空孔内。B、胰酶消化细胞,计数,按照每孔50000个细胞铺板,和之前的转染液共培育72小时后收样检测。
2)提取RNA实验:总RNA抽提采用异丙醇沉淀法,具体步骤如下(以24孔板为例):A、每孔用500μL PBS清洗后,加入500μL RNAiso plus,吹打几次。将溶液转移至1.5mL EP管内。B、向1.5mL EP管内加入200μL氯仿,上下颠倒混匀30s,室温静置10分钟,放于4℃离心机内,12000rpm离心15分钟。C、将上清转移至新的1.5mLEP管内,加入等体积的异丙醇,用以沉淀RNA。上下颠倒混匀10次,室温静置10分钟后,放于4℃离心机内,12000rpm离心15分钟。D、弃去上清,可观测到管底有白色沉淀。加入1mL75%乙醇重悬沉淀,4℃离心机内,12000rpm离心15分钟。弃去上清,将EP管盖打开,室温挥发残余液体。15-20分钟后,加入适量DEPC水,溶解沉淀后,进行下一步实验。
3)实时定量荧光PCR实验:
A、利用酶标仪检测抽提的RNA浓度。
B、将RNA反转录为cDNA,反转录体系(20μL),见下表。
反转录体系:
RNA样品1000ng

DEPC水 up to 20μL
反转录过程:①37℃,15min;②85℃,5s;③保持10℃。
C、定量PCR反应体系(每孔):
定量PCR过程:①50℃,2min;②95℃,20s;③95℃,3s;④60℃,30s;③-④循环40次。
2.实验结果
结果如表2和图1A、B、D、E中所示,siHMGN5可以显著下调乳腺癌细胞中HMGN5的mRNA水平。

实施例2在乳腺癌细胞中,利用蛋白质免疫印迹(Western blot)技术,检测siHMGN5对HMGN5蛋白的作用
在本实施例中,在乳腺癌细胞系MDA-MB-231和T47D中转入siHMGN5_101、siHMGN5_102、siHMGN5_103,通过蛋白质免疫印迹法检测siHMGN5对HMGN5蛋白的作用。
1、实验材料和方法
1)siRNA转染细胞实验方法:详见实施例1。
2)蛋白质免疫印迹(Western blot)检测方法:
A、所用试剂:HMGN5抗体(Invitrogen,美国);GAPDH(Cell SignalingTechnology,美国);显影液(Immobilon ECL,Millipore,美国);溴酚蓝指示剂的1×SDS裂解液;5×Tris-甘氨酸电泳缓冲液(25mM Tris,250mM pH8.3甘氨酸,0.1%SDS);10×转膜缓冲液(39mM甘氨酸,48mMTris,0.037%SDS,20%甲醇);1×TBST(含0.5%Tween-20的TBS);蛋白marker(Thermo,美国)。
B、实验步骤:(A)制备蛋白样品:将24孔板中细胞培养液用真空泵吸出,PBS缓冲液润洗两次之后加入40μL左右1×SDS裂解液收取细胞,95℃加热10分钟将蛋白变性;(B)SDS-PAGE电泳:按蛋白大小配制不同浓度的SDS-PAGE胶,将1×电泳缓冲液加入电泳槽,设置电压为90V,30分钟后将电压调至150V,直至所需蛋白完全分开终止电泳;(C)湿转法转膜:将1×转膜缓冲液加入转膜槽,220mA,2小时,将蛋白转移到PVDF膜上;(D)封闭:配制5%的脱脂牛奶,将转膜完成后的PVDF膜置于其中室温封闭2小时;(E)一抗孵育:将一抗原液按合适比例用一抗稀释液稀释,4℃旋转孵育过夜;(F)洗膜:1×TBST洗膜三次,每次10分钟;(G)二抗孵育:将二抗按合适比例稀释于5%脱脂牛奶中,室温孵育1小时;(H)洗膜:1×TBST洗膜三次,每次10分钟;(I)ECL显影:将显影液A和B按1:1进行配置,将条带浸于混合液中,室温避光孵育1分钟后将条带放入显影仪中进行显影。
2、实验结果:
结果如图1C所示,siHMGN5_101、siHMGN5_102、siHMGN5_103可以显著抑制MDA-MB-231和T47D细胞中HMGN5的蛋白含量,且蛋白抑制率基本与mRNA抑制率相对应(表3)。

实施例3在人乳腺癌细胞中检测siHMGN5对细胞增殖的影响
在乳腺癌细胞系MDA-MB-231和T47D中转入siHMGN5_101、siHMGN5_102、siHMGN5_103,通过细胞增殖实验和平板克隆形成实验检测siHMGN5对细胞增殖的影响。
1、实验材料和方法
1)CCK-8试剂盒检测细胞活力:A、siHMGN5转染细胞实验方法:详见实施例1。B、转染完成后将细胞置于37℃恒温培养箱培养72小时后进行检测。C、将培养板中的细胞培养液吸出,每孔加入100μL含10%CCK-8的培养基后将培养板放回37℃恒温培养箱培养1个小时,利用多功能酶标仪检测波长为495nm时的吸光值。
2)平板克隆形成实验检测细胞克隆形成能力:A、siHMGN5转染细胞实验方法:详见实施例1。B、转染完成后将细胞置于37℃恒温培养箱培养10天左右直至细胞形成合适大小的克隆。C、将培养板中的细胞培养液吸出,用1mL-20℃预冷的甲醇固定细胞。D、用结晶紫染料对克隆进行染色。E、对克隆进行计数或用33%醋酸洗脱染料测量595nm处的吸光值。
2、实验结果:
结果如图2A和B所示,在MDA-MB-231和T47D细胞中转染30nM的siHMGN5_101、siHMGN5_102、siHMGN5_103,72小时后,和Ctrl组比显著抑制MDA-MB-231和T47D细胞的细胞活力;同时siHMGN5_101、siHMGN5_102、siHMGN5_103显著抑制了MDA-MB-231和T47D细胞克隆形成能力(图2C和D)。
实施例4在人乳腺癌细胞中检测siHMGN5对细胞迁移和侵袭能力的影响
在乳腺癌细胞系MDA-MB-231中转入siHMGN5_101、siHMGN5_102、siHMGN5_103,通过小室细胞迁移与侵袭实验检测siHMGN5对乳腺癌细胞迁移和侵袭能力的作用。
1、实验材料和方法
1)肿瘤细胞迁移与侵袭实验
肿瘤细胞迁移与侵袭实验是利用Transwell小室在有无基底膜基质的情况下考察肿瘤细胞从小室上表面穿过孔径至下表面的能力。细胞迁移实验不需要包被基底膜基质,细胞侵袭实验需要用基底膜基质包被Transwell小室,本实验利用24孔板体系操作,具体实验操作过程如下:
A、8μm孔径Transwell小室基底膜基质包被,冰上孵育基底膜基质包被缓冲液2小时,4℃解冻基底膜基质,用基底膜基质包被缓冲液稀释基底膜基质至终浓度为200μg/ml,注意稀释好的基质不能有气泡出现,如有气泡需4℃离心10分钟,转速为300×g。用-20℃预冷的枪头吸取100μl稀释好的基质小心加入每个小室底部,此步骤要避免基质与小室底部直接接触。B、将包被好的小室放置细胞培养箱孵育2小时,孵育期间不要晃动小室,加入细胞悬液之前吸去上层溶液。C、24孔板下室加入500μl新鲜预热的正常培养基,将小室放置在其中,轻轻拍打确保小室与培养基接触处无气泡产生。D、细胞在被转移至小室中培养之前要达到80%融合度,消化细胞并计数,用无血清的培养基重悬后按2.5×105个细胞/ml浓度稀释,每个小室加入200μl细胞悬液。E、组织培养箱中静置培养24小时,吸去培养基后用预热的1×PBS清洗一遍,4%多聚甲醛室温固定15分钟后,吸去多聚甲醛溶液并用棉棒轻轻将小室内上层未穿过的细胞擦拭掉,进而用苏木素溶液染色15分钟,流水返蓝后在显微镜下拍照。F、拍照后,Image J软件进行计数统计或者用1mL 33%醋酸洗脱染料后于595nm波长处测定吸光度,并分析各组间差异。
2、实验结果:
根据图3A的结果可见,siHMGN5_101、siHMGN5_102、siHMGN5_103均显著抑制了MDA-MB-231细胞的迁移能力。同时,图3B的结果表明siHMGN5_101、siHMGN5_102、siHMGN5_103对MDA-MB-231细胞的侵袭能力也产生了显著的抑制作用。
实施例5体内皮下移植瘤实验检测siHMGN5对乳腺癌增殖的影响
短发夹RNA(shRNA)可以通过慢病毒整合到细胞基因组上,稳定表达的shRNA在细胞内经过切割会形成相应的siRNA序列发挥靶向靶基因mRNA的作用。利用慢病毒感染细胞建立shHMGN5的乳腺癌稳转细胞系(MDA-MB-231和T47D),转录出的shHMGN5序列在体内经过切割会形成相应的siHMGN5序列发挥靶向靶基因mRNA的作用。将shHMGN5稳转细胞系和对照细胞系分别在裸鼠上进行皮下注射,观察siHMGN5对乳腺癌皮下移植瘤生长的作用。
1、实验材料和方法
1)shHMGN5稳转细胞系的构建:A、利用双酶切体系将shHMGN5序列(委托于泓讯合成,该shRNA对应于siHMGN5_104)克隆到慢病毒表达载体PLKO.1(购于Addgene)中,并进行质粒扩增;B、实验前一天将工具细胞293T接种于10cm培养皿中,保证次日细胞融合度约60%-70%;C、在转染前2h将10cm培养皿中待转染细胞培养液吸出,加入3mL预热的无血清培养基Opti-MEM饥饿细胞;D、将待转染的质粒DNA(实验组慢病毒包装质粒PLKO.1-shHMGN5/对照组慢病毒包装质粒PLKO.1-ctrl:5μg,辅助质粒PSPAX2和PMD2G分别为3.75μg和1.25μg)溶于1mL无血清培养基Opti-MEM中;E、将15μL Lipofectamine 2000溶于1mL无血清培养基Opti-MEM中,室温下孵育5分钟;F、迅速将以上两种混合物混匀,并于室温避光孵育20分钟;G、将混合物加入10cm培养皿中,随后放回37℃恒温培养箱培养6个小时;H、移去上清液,更换37℃预热的完全培养基进行培养;I、72小时后收上清,并用直径0.45μm的滤膜进行过滤,病毒直接使用或分装后冻存于-80℃;J、提前一天将待感染的MDA-MB-231或T47D细胞接种至6孔板中,使之在转染时达到30%-50%的融合度;K、将收集的病毒以1mL/孔加入细胞中,每孔另加1mL完全培养基和10μg/mL聚凝胺(polybrene),24小时后观察细胞状态,更换含2μg/mL嘌呤霉素的完全培养基筛选阳性细胞;L、筛选72小时后收样检测;M、在整个实验过程中注意观察细胞状态并每隔一段时间收集细胞样本并检测HMGN5的表达水平,当HMGN5表达被稳定抑制时,即认为已经获得了稳转细胞株。
2)裸鼠实验:将5×106个稳定表达shHMGN5的MDA-MB-231或T47D细胞及其对应的对照组细胞接种在裸鼠皮下,监测肿瘤生长体积,30天后处死实验裸鼠,拍照,取肿瘤组织以及称重,并通过蛋白免疫印迹检测肿瘤组织中HMGN5蛋白量。
2、实验结果:
结果显示与对照组相比,shHMGN5实验组裸鼠肿瘤体积和肿瘤重量显著降低,这表明,siHMGN5能够抑制乳腺癌移植瘤的体内生长。
实施例6体内免疫缺陷鼠肺转移实验检测siHMGN5对肿瘤细胞体内转移的影响
本发明将shHMGN5稳转乳腺癌细胞系和对照细胞系进行荧光素酶标记后从免疫缺陷鼠的尾静脉注射入体内,进行体内肺转移实验观察siHMGN5对乳腺癌转移的作用。
1、实验材料和方法
1)荧光素酶标记的shHMGN5稳转细胞系的构建方法:对实施例5中构建好的shHMGN5稳转细胞系进一步进行荧光素酶标记,即用含荧光素酶序列的慢病毒感染shHMGN5稳转细胞系和对照细胞系。注射前细胞与荧光素酶底物共孵育测量吸光值检测细胞是否成功过表达荧光素酶。
2)免疫缺陷鼠肺转移实验
A、将细胞消化并用生理盐水制备成5×106个/ml的细胞悬液,每只小鼠注射200ul细胞悬液(1×106个细胞),接种后6小时内观察小鼠状态,确保小鼠状态良好后方可离开。B、接种细胞后正常饲养环境饲养60天,期间每周称量体重一次,从第三周起,每周进行活体成像检测转移灶(拍摄前15min注射15mg/kg荧光素钠盐溶液)。C、注射细胞60天后麻醉和下腔静脉放血处死并取肺脏用10%中性福尔马林固定,石蜡包埋切片并进行HE染色以观察肿瘤细胞脏器内转移情况。
2.实验结果
图5A显示shHMGN5组的肺部结节数显著低于对照组,转移程度显著改善(图5B),并且肺转移发生率也明显降低(图5C)。
实施例7 siHMGN5载体药物给药实验检测对乳腺癌增殖的作用
利用聚合物囊泡包裹siHMGN5_011制备载体药物,之后对负荷MDA-MB-231细胞皮下瘤的裸鼠进行尾静脉给药。观察siHMGN5的载体药物对乳腺癌增殖的作用。
1、实验材料和方法
A、将2×106个MDA-MB-231细胞接种在裸鼠皮下,监测肿瘤生长体积,肿瘤生长十天后,按照皮下瘤体积将小鼠进行分成两组。B、一组小鼠尾静脉注射载体药物;另一组则给药空载,作为对照。每周给药两次。C、给药三周后处死实验裸鼠,拍照,取肿瘤组织以及称重,并通过蛋白免疫印迹检测肿瘤组织中HMGN5蛋白量。
2.实验结果
图6A-C结果表明siHMGN5的载体药物显著抑制了皮下移植瘤的生长。并且肿瘤组织中HMGN5的表达被siHMGN5显著下调(图6D和E)。
讨论
MGN5已被证实与多种肿瘤的病理分级,临床分期和预后等相关联。
在本发明中,本发明人开发了高效靶向并抑制HMGN5mRNA的小干扰RNA(siHMGN5),进一步开发了调控HMGN5的活性分子在肿瘤治疗中的应用。
上述实验表明,本发明的siHMGN5能够高效抑制HMGN5的转录与翻译,在细胞与动物水平调控乳腺癌增殖和转移。siHMGN5可以抑制乳腺癌细胞株MDA-MB-231和T47D的细胞活力和细胞迁移侵袭能力,并在乳腺癌裸鼠移植瘤模型中显著抑制裸鼠的肿瘤生长以及乳腺癌转移。
由于本发明的siHMGN5可极其高效抑制HMGN5的mRNA和蛋白表达,显著抑制乳腺癌的增殖和转移,因此有望开发为治疗肿瘤的药物,用于治疗乳腺癌,脑癌,前列腺癌,膀胱癌,卵巢癌,子宫癌,结直肠癌,骨肉瘤,肺癌,胰腺癌等HMGN5高表达肿瘤。

Claims (10)
1.一种人工siRNA或其前体序列,其特征在于,所述的siRNA靶向高迁移率族核小体结合蛋白5(HMGN5)并抑制HMGN5表达,
其中,所述siRNA对HMGN5表达的抑制率≥85%。
2.如权利要求1所述的人工siRNA或其前体序列,其特征在于,所述siRNA为选自表1中靶向HMGN5的siRNA(siHMGN5)或其经修饰的衍生物:

B1为第一核糖核酸序列,其中所述的第一核糖核酸序列为HMGN5 siRNA正义链序列;

C为茎环结构序列;
A1和A2分别为无,或任选的由4-5个碱基组成的RNA序列,
其中,所示的前体序列能在宿主中加工形成靶向HMGN5并抑制HMGN5的siRNA。
4.如权利要求3所述的人工siRNA或其前体序列,其特征在于,在前体序列中,所述的基本互补指所述B2与B1有2-8个碱基不互补,较佳地,所述B2与B1有3-5个碱基不互补;更佳地,所述的B2较B1缺失1-2个碱基。
5.一种多核苷酸,其特征在于,所述的多核苷酸能被宿主细胞转录形成权利要求1中所述的人工siRNA或其前体序列。

(b)药学上可接受的载体。
8.一种活性成分的用途,其特征在于,用于制备抗肿瘤的药物或制剂,其中,所述活性成分选自下组:权利要求1所述的权利要求1所述的人工siRNA或其前体序列、权利要求5所述的多核苷酸、或权利要求6所述的表达载体或其组合。
9.一种体外非治疗性抑制HMGN5高表达的肿瘤细胞生长的方法,包括步骤:
在第一活性成分存在下,培养HMGN5高表达的肿瘤细胞,从而抑制所述HMGN5高表达肿瘤细胞的生长,其中,活性成分选自下组:权利要求1所述的权利要求1所述的人工siRNA或其前体序列、权利要求5所述的多核苷酸、或权利要求6所述的表达载体或其组合。
10.一种治疗HMGN5高表达相关疾病的方法,其特征在于,包括步骤:将安全有效量的第一活性成分或含所述第一活性成分的药物组合物施用于所需对象,从而治疗HMGN5高表达相关疾病,其中,所述的第一活性成分选自下组:权利要求1所述的权利要求1所述的人工siRNA或其前体序列、权利要求5所述的多核苷酸、或权利要求6所述的表达载体或其组合。
