Abstract
本发明提供了抗炎因子在促进外泌体产生的应用,所述抗炎因子选自IL‑4、IL‑10和IL‑13中的一种或多种。本发明还提供了一种制备外泌体的方法,该方法包括(1)将编码抗炎因子的核苷酸序列导入宿主细胞中,以及(2)从所述宿主细胞的培养基中分离出外泌体,由该方法制备的外泌体具有表达水平升高的CD81。
Description
技术领域
本发明涉及一种富含CD81的外泌体的制备方法及其应用。
背景技术
外泌体是最小的细胞外膜泡,直径约为40~100nm,内含蛋白质、脂质、信使RNA和微小RNA(miRNAs)。它们是在细胞内吞内化过程中由细胞膜产生的,释放到细胞外空间影响受体细胞的功能和生理。因此,它们是细胞间通信的关键串扰因子。外泌体可以在血液和滑膜液中稳定存在。
间充质干细胞具有自我更新和多谱系分化的潜力,对于组织修复和基因治疗有很大作用。间充质干细胞也表现出免疫细胞的某些特征,能够通过旁分泌作用释放大量的细胞因子和生长因子。因此,间充质干细胞分泌的外泌体具有组织修复的作用。但是,间充质干细胞分泌的外泌体量,以及外泌体所表达的各种细胞因子和生长因子量均有限,无法满足临床应用。
因此需要开发一种能够提高所需的外泌体的产量,且提高其表达所需细胞因子表达水平的方法。
发明内容
为了解决现有技术中存在的上述技术问题之一,本发明提供了抗炎因子IL-4、IL-10和IL-13在促进外泌体产生的应用,以及制备外泌体的方法。
根据一个方面,提供了抗炎因子在促进外泌体产生的应用,其中所述抗炎因子选自IL-4、IL-10和IL-13中的一种或多种。
在一些实施方式中,所述外泌体具有表达水平升高的CD81。
在一些实施方式中,所述外泌体可以由宿主细胞,例如哺乳动物细胞产生。在一些实施方式中,所述宿主细胞可以包括,但不限于,人胚胎肾(HEK)细胞、中国仓鼠卵巢(CHO)细胞、HT-1080细胞、HeLa细胞、PERC-6细胞、CEVEC细胞、成纤维细胞、羊膜细胞、上皮细胞和间充质干细胞(MSC)细胞。在优选的实施方式中,所述宿主细胞可以为间充质干细胞。在具体的实施方式中,所述间充质干细胞可以包括骨髓来源的MSCs、脂肪来源的MSCs、脐带血来源的MSCs。
在一些实施方式中,所述抗炎因子能够促进CD81阳性外泌体的产生。在一些实施方式中,所述抗炎因子能够富集CD81阳性外泌体。
根据另一方面,提供了一种制备外泌体的方法,所述方法包括以下步骤:(1)将编码抗炎因子的核苷酸序列导入宿主细胞中;以及,(2)从所述宿主细胞的培养基中分离出外泌体。
在一些实施方式中,所述编码抗炎因子的核酸序列位于慢病毒载体中。在具体的实施方式中,所述慢病毒载体包括含有ψ序列的5’LTR,3’LTR,位于5’LTR和3’LTR之间的所述编码抗炎因子的核苷酸序列,以及与所述编码抗炎因子的核苷酸序列可操作连接的启动子和翻译起始序列。
在一些实施方式中,在步骤(1)中,使用所述慢病毒载体感染所述宿主细胞。
在一些实施方式中,所述5’LTR和所述3’LTR的U3可以被缺失或突变。在优选的实施方式中,所述3’LTR是缺失了U3区域的3’LTR(ΔU3/3’LTR)。在一些实施方式中,所述5’LTR是缺失形式的5’LTR(Δ5’LTR)。
在一些实施方式中,所述慢病毒载体的5’LTR的启动子被替换成异源启动子,例如巨细胞病毒(CMV)启动子、劳斯肉瘤病毒(RSV)启动子或猿猴病毒40(SV40)启动子。
在一些实施方式中,与所述编码抗炎因子的核苷酸序列可操作地连接的启动子选自:短延伸因子1α(EF1α)启动子、RSV启动子、猿猴病毒40(SV40)启动子或其转录活性片段。
在一些实施方式中,与所述编码抗炎因子的核苷酸序列可操作地连接有RSV启动子,EF1α启动子和Kozak翻译起始序列。
在一些实施方式中,所述慢病毒载体还可包括筛选标记物。在一些实施方式中,所述筛选标记物可以选自荧光素酶、(增强的)绿色荧光蛋白、链霉亲和素结合肽、嘌呤霉素抗性基因、氨苄抗性基因、卡那霉素抗性基因和新霉素抗性基因中的一种或多种。在一些实施方式中,所述筛选标记物可以包括增强的绿色荧光蛋白(EGFP)和嘌呤霉素抗性基因(Puro)。
在一些实施方式中,所述慢病毒载体还可包括SV40早期pA。
在一些实施方式中,所述慢病毒载体的转录后调控元件包括土拨鼠肝炎病毒转录后调控元件(WPRE)。
在一些实施方式中,所述慢病毒载体包含逆转录病毒输出元件。“逆转录病毒输出元件”是指调节RNA转录物从细胞核到细胞质的转运的顺式作用转录后调节元件。优选地,所述逆转录病毒输出元件包括但不限于人类免疫缺陷病毒(HIV)rev应答元件(RRE)和乙型肝炎病毒转录后调节元件(HPRE)。
在一些实施方案中,所述慢病毒载体可以包含中央多聚嘌呤区(cPPT)或中心终止序列(CTS)作为顺式作用元件,cPPT/CTS序列可以是HIV1的cPPT/CTS,能够提高载体整合和转导效率。
在一些实施方式中,所述慢病毒载体从5’LTR区到3’LTR区依次包括:RSV启动子,5’LTR,Ψ序列,EF1α启动子,Kozak翻译起始序列,编码抗炎因子的核苷酸序列,SV40,Kozak翻译起始序列,EGFR/Puro,WRPE,ΔU3/3’LTR和SV40早期pA。在一些实施方式中,所述抗炎因子包括IL-10,IL-13和IL-4。在优选的实施方式中,编码IL-10、IL-13和IL-4的核苷酸序列分别如SEQ ID NO:1,SEQ ID NO:2和SEQ ID NO:3所示。
本领域技术人员应理解,编码IL-10、IL-13和IL-4的核苷酸序列的顺序可以改变,只要是IL-10、IL-13和IL-4可以在宿主细胞中同时表达即可。
在一些实施方式中,编码IL-10、IL-13和IL-4的核苷酸序列之间被编码2A肽的核苷酸序列间隔开。在一些实施方式中,所述2A肽为T2A肽、P2A肽、E2A肽或F2A肽。优选地,所述2A肽为T2A或P2A肽。所述2A肽可以避免产生IL-10、IL-13和IL-4的融合蛋白。
在一些实施方式中,所述EGFR/Puro的编码核苷酸中,EGFR和嘌呤霉素抗性基因之间以编码2A肽的核苷酸序列间隔开。
在优选的实施方式中,通过上述慢病毒载体感染宿主细胞,可以在所述宿主细胞中同时过表达IL-10、IL-13和IL-4三种抗炎因子。
在一些实施方案中,所述宿主细胞能够表达所述慢病毒载体中携带的目的核苷酸。
在一些实施方案中,所述宿主细胞为哺乳动物细胞。在一些实施方式中,所述宿主细胞可以包括,但不限于,人胚胎肾(HEK)细胞、中国仓鼠卵巢(CHO)细胞、HT-1080细胞、HeLa细胞、PERC-6细胞、CEVEC细胞、成纤维细胞、羊膜细胞、上皮细胞和间充质干细胞(MSC)细胞。在优选的实施方式中,所述宿主细胞可以为间充质干细胞。在具体的实施方式中,所述间充质干细胞可以包括骨髓来源的MSCs、脂肪来源的MSCs、脐带血来源的MSCs。
本文的上述慢病毒载体无需分别制备IL-10、IL-13和IL-4三种载体质粒,仅需要一种载体质粒同时携带编码IL-10、IL-13和IL-4的编码核苷酸,简化了操作成本,减少了转染多个病毒载体对细胞的损伤,有利于后续作为细胞药物制剂的质量控制。
在一些实施方式中,所述方法制备的外泌体具有高表达的CD81。因此,通过本公开的方法能够有效富集CD81阳性的外泌体。
根据又一方面,提供了通过上述方法制备的外泌体,所述外泌体具有表达水平升高的CD81。
根据又一方面,提供了本公开制备的外泌体在治疗和/或预防炎性疾病中的应用。在一些实施方式中,所述外泌体可以用于创面修复、组织修复、伤口愈合等。
在一些实施方式中,本公开制备的外泌体可用于加快伤口愈合速度,提高伤口恢复质量,减小疤痕面积等。
本公开通过基因编辑,提高了宿主细胞,特别是间充质干细胞分泌的特定亚型(CD81+外泌体)的比例,从而提取富含这一亚型的外泌体,能有效治疗和/或预防炎性疾病。

具体实施方式
为使本发明的目的、技术方案及优点更加清楚明白,以下结合实施例,对本发明进行进一步的详细说明。此处所描述的具体实施例仅用于解释本发明,并不用于构成对本发明的任何限制。此外,在以下说明中,省略了对公知结构和技术的描述,以避免不必要地混淆本公开的概念。这样的结构和技术在许多出版物中也进行了描述。
定义
除非另有定义,否则本发明使用的所有技术术语和科技术语都具有如在本发明所属领域中通常使用的相同含义。出于解释本说明书的目的,将应用以下定义,并且在适当时,以单数形式使用的术语也将包括复数形式,反之亦然。
除非上下文另有明确说明,否则本文所用的表述“一种”和“一个”包括复数指代。例如,提及“一个细胞”包括多个这样的细胞及本领域技术人员可知晓的等同物等等。
外泌体(exosome)是由细胞内多泡体(multivesicular body,MVB)与细胞膜融合后,释放到细胞外基质中的膜性囊泡。几乎所有类型的细胞,都可以产生并释放外泌体。它是一种直径为30~100nm的纳米级脂质包裹体结构,内部包裹了蛋白、mRNA和microRNA等物质。外泌体天然存在于体液中,包括血液、唾液、尿液和母乳,被分泌出的外泌体会进入血液、唾液、尿液、及乳汁等体液中,通过循环系统到达其他细胞与组织,产生远程调控作用。大部分外泌体含有MHC I类和/或II类分子、热休克蛋白(hsp)、四跨膜蛋白、整合素、细胞骨架蛋白、mRNA和其他非编码RNA以及各种代谢酶。外泌体的膜上包含不同的表面标记物,其中典型的是四跨膜蛋白CD9、CD63和CD81。四跨膜蛋白可用于各种肿瘤和传染病的诊断。
慢病毒载体系统包括包装质粒,包膜质粒和载体质粒,并且可以为二质粒系统,三质粒系统或四质粒系统。三质粒系统是将慢病毒基因组中负责包装,逆转录和整合所需要的顺式作用序列结构和编码反式作用蛋白的序列分离,再克隆到三个独立的质粒中,并去除了所有的辅助序列。四质粒系统是在三质粒系统的基础上改进而来的,与三质粒系统相比,第一个变化是将rev基因放在一个单独的表达质粒上,新增一个质粒更增加了系统的安全性,第二个变化是将tat基因去除,并在载体质粒上增添了与异源启动子融合的嵌合5'LTR,以启动载体质粒的表达。此外,三质粒系统和四质粒系统还有一个可以放置目的基因序列的载体,称作载体质粒。
IL-4主要由活化T细胞产生,其可抑制内皮细胞及单核细胞合成分泌IL-1、IL-6和TNF-α等炎性细胞因子,起到抑制促炎细胞因子合成,发挥其抑制炎症反应的作用。IL-10主要由Th2细胞、活化的单核细胞和上皮细胞分泌产生,具有明确的免疫抑制活性。IL-13是由活化T细胞产生的一种新的细胞因子,其与IL-4的受体α链具有同源性,信号转导途径和生物学活性也非常相似,具有抑制炎症反应的功能。
ψ序列是慢病毒基因组的衣壳化所需要的最小包装信号。
2A肽是来源于病毒的短肽通常为18~25个氨基酸长,通常被称作自我剪切肽,能使得一条转录物产生多种蛋白。
下面提供实施例和附图以帮助理解本发明。但应理解,这些实施例和附图仅用于说明本发明,但不构成任何限制。本发明的实际保护范围在权利要求书中进行阐述。应理解,在不脱离本发明精神的情况下,可以进行任何修改和改变。
实施例1:基因编辑和感染效率鉴定
首先设计合成了过表达三个抗炎因子IL-4,IL-10和IL-13的慢病毒载体,命名为pLV-hIL10(SEQ ID NO:1)-hIL13(SEQ ID NO:1)-hIL4(SEQ ID NO:1),载体设计图谱如图1所示。所构建的慢病毒载体同时载体携有增强型GFP和抗生素Puro序列,方便转染效率的观察。通过Sanger测序对核酸序列进行鉴定,比对完全正确。
病毒载体质粒构建成功后,进行病毒包装,转染脐带来源的间充质干细胞。选择P4-P8代的细胞,将细胞按1.2X105/孔的密度接种到6孔板中,培养基为DMEM/F12+10%FBS,在37℃5%CO2的培养箱中培养18-20h后,密度融合为30%-50%时进行转染。转导当天,在冰上解冻病毒,按照MOI 20吸取病毒数,加入培养基混合均匀。培养基用量为1mL/孔。同时向每孔中加入6g/mL的辅助转染试剂Polybrene,培养6-8h。同时将带有空载体Vector的病毒液作为空白对照以同样的方法进行转导。第二天,吸出含有病毒的培养基,加入新鲜的培养基过夜培养。在72h后共聚焦显微镜观察到GFP的绿色荧光蛋白的强表达(参见图2),计数绿色荧光的百分比,同时流式分析确认感染效率(参见图3),结果显示MOI为20时有效感染率为96.9%。
实施例2:外泌体的收集
将转染病毒的间充质干细胞接种到15cm培养皿中,培养基为DMEM/F12+10%FBS,待细胞融合密度为80%时,弃掉培养基,用PBS冲洗两边,然后加入不含血清的DMEM/F12培养基,继续培养40h,收集细胞上清,提取外泌体。2000g离心20min去除细胞和死细胞,然后10000g离心30min去除细胞碎片。去上清加到超离管中167000g离心70min后,取沉淀加入PBS重悬,然后167000g离心70min去上清,获取的沉淀即为间充质干细胞分泌的外泌体。通过Western Blot对收集到的外泌体进行亚型的鉴定(参见图4)。结果发现,与空载细胞的外泌体相比,过表达抗炎因子的细胞分泌的外泌体的标志物CD81表达高,说明转染病毒后,细胞分泌的外泌体CD81亚型提高。
本发明的技术方案不限于上述具体实施例的限制,凡是根据本发明的技术方案做出的技术变形,均落入本发明的保护范围之内。
Claims (10)
1.抗炎因子在促进外泌体产生的应用,其中所述抗炎因子选自IL-4、IL-10和IL-13中的一种或多种,优选地,所述外泌体具有表达水平升高的CD81。
2.一种制备外泌体的方法,所述方法包括以下步骤:(1)将编码抗炎因子的核苷酸序列导入宿主细胞中;以及,(2)从所述宿主细胞的培养基中分离出外泌体。
3.根据权利要求2所述的方法,其特征在于,所述编码抗炎因子的核酸序列位于慢病毒载体中,优选地,所述慢病毒载体包括含有ψ序列的5’LTR,3’LTR,位于5’LTR和3’LTR之间的所述编码抗炎因子的核苷酸序列,以及与所述编码抗炎因子的核苷酸序列可操作连接的启动子和翻译起始序列。
4.根据权利要求2所述的方法,其特征在于,所述5’LTR和所述3’LTR的U3被缺失或突变,
优选地,所述3’LTR是缺失了U3区域的3’LTR(ΔU3/3’LTR),
优选地,所述5’LTR是缺失形式的5’LTR(Δ5’LTR),
优选地,所述5’LTR的启动子被替换成异源启动子,优选被替换成巨细胞病毒(CMV)启动子、劳斯肉瘤病毒(RSV)启动子或猿猴病毒40(SV40)启动子,

5.根据权利要求2所述的方法,其特征在于,所述慢病毒载体还包括筛选标记物的编码核苷酸,
优选地,所述筛选标记物选自荧光素酶、绿色荧光蛋白、链霉亲和素结合肽、嘌呤霉素抗性基因、氨苄抗性基因、卡那霉素抗性基因和新霉素抗性基因中的一种或多种,
优选地,所述筛选标记物选自增强的绿色荧光蛋白(EGFP)和嘌呤霉素抗性基因(Puro),
优选地,所述慢病毒载体还包括土拨鼠肝炎病毒转录后调控元件WPRE、逆转录病毒输出元件、中央多聚嘌呤区cPPT或中心终止序列CTS。
6.根据权利要求2所述的方法,其特征在于,所述慢病毒载体从5’LTR区到3’LTR区依次包括:RSV启动子,5’LTR,Ψ序列,EF1α启动子,Kozak翻译起始序列,编码抗炎因子的核苷酸序列,SV40,Kozak翻译起始序列,EGFR/Puro,WRPE,ΔU3/3’LTR和SV40早期pA。
7.根据权利要求2所述的方法,其特征在于,所述抗炎因子包括IL-10,IL-13和IL-4,
优选地,编码IL-10、IL-13和IL-4的核苷酸序列之间被编码2A肽的核苷酸序列间隔开,
优选地,所述2A肽为T2A肽、P2A肽、E2A肽或F2A肽。
8.由权利要求2-7中任一项所述的方法制备的外泌体。
9.根据权利要求8所述外泌体,其特征在于,所述外泌体具有表达水平升高的CD81。
10.权利要求2-7中任一项所述的方法制备的外泌体在治疗和/或预防炎性疾病、创面修复、组织修复或伤口愈合中的应用。
