Abstract
本发明涉及一种贻贝粘蛋白头皮修护组合物及其制备方法和应用,所述组合物中成分包含贻贝粘蛋白、芦荟提取物、牛磺酸、乙酰谷氨酰胺、柠檬酸三乙酯、表面活性剂、柠檬酸、氯化钠、异鲸蜡醇硬脂酸酯、甲基丙二醇、C11‑15仲链烷醇聚醚‑12、PEG‑10聚二甲基硅氧烷、乙基己基甘油、维生素E和苯氧乙醇;本发明以贻贝粘蛋白和芦荟提取液为主要修复活性成分,通过对组合物pH和离子浓度的调节,使得组合物保持较好的渗透性,该组合物形成一层薄膜,增加有效活性成分的吸收,提高贻贝粘蛋白的修护效果。

技术领域
本发明属于洗护日用品技术领域,具体涉及一种贻贝粘蛋白头皮修护组合物及其制备方法和应用。
背景技术
随着现在生活环境的影响包括生活压力的上升,头皮油脂分泌过多、头屑、头皮敏感等问题越来越常见,使得消费者的对洗护产品的功效需求也逐渐增大,其中头皮的油脂分泌的失衡,在产品功能上,洗发护发产品的功能不断延伸并逐渐细化,从最初的清洁头发功能,逐步延伸为控油、去屑、护理和滋润等功能,更进一步延伸到乌发、防脱和生发等概念。
贻贝是一种广泛分布于沿海和近海的甲壳类海洋生物,其足丝腺能分泌足丝,并通过足丝形成的足丝盘将贻贝固定在各种固体基材表面,研究表明,这与贻贝分泌的足丝有关,其足丝的主要成分为贻贝足丝蛋白也称为贻贝粘蛋白。目前,现有技术中已报道贻贝粘蛋白具有一定的修复效果,主要用于创面敷料中,而在洗护产品中使用较少,中国专利CN115120549A公开了一种贻贝头皮护养精华及其制备方法,该精华中包含贻贝提取物,同时还添加了其他包括中药材提取物、蛋白类物质等改善头皮微生态环境,修复头皮屏障功能,但该精华中的中药材提取物,蛋白等物质,此类物质的吸收效果并不好,储存稳定性也较差,而该专利技术并未解决该技术问题;中国专利CN108771651A公开了一种防止断发的修护型洗发水,该洗发水中含有贝类活性肽提取液可加快蛋白质的合成,增强免疫力、抗菌等特性,但并未给出贝类活性肽在促进有效成分吸收和渗透的作用和机理,以及如何而提高贝类活性肽的生物利用度,
基于现有技术很少涉及对贻贝及其提取物在洗护产品中的应用,制备一种利用贻贝及其提取物清洁毛囊、修复头皮的洗护产品对其开发与利用具有重要的意义。
发明内容
针对现有技术中存在的问题,本发明提供了一种贻贝粘蛋白头皮修护组合物及其制备方法和应用,本发明制备的护发组合物具有良好的防脱修护效果,渗透效果好,无刺激性,尤其适用于头发、头皮受损人群。
第一方面,本发明提供了一种贻贝粘蛋白头皮修护组合物,所述组合物中成分包含贻贝粘蛋白、芦荟提取物、牛磺酸、乙酰谷氨酰胺、柠檬酸三乙酯、表面活性剂、柠檬酸、氯化钠、异鲸蜡醇硬脂酸酯、甲基丙二醇、C11-15仲链烷醇聚醚-12、PEG-10聚二甲基硅氧烷、乙基己基甘油、维生素E和苯氧乙醇。
进一步地,按重量份数计,所述贻贝粘蛋白头皮修护组合物中成分包含贻贝粘蛋白0.1~2份、芦荟提取物1~10份、牛磺酸0.1~1份、乙酰谷氨酰胺0.5~1份、柠檬酸三乙酯0.5~3份、表面活性剂1~5份、柠檬酸0.1~1份、氯化钠0.01~1份、异鲸蜡醇硬脂酸酯1~15份、甲基丙二醇1~10份、C11-15仲链烷醇聚醚-12 1~10份、PEG-10聚二甲基硅氧烷1~5份、乙基己基甘油0.1~0.5份、维生素E0.5~3份和苯氧乙醇0.01~0.2份。
优选地,按重量份数计,所述贻贝粘蛋白头皮修护组合物中成分包含贻贝粘蛋白0.1~1份、芦荟提取物2~6份、牛磺酸0.1~0.5份、乙酰谷氨酰胺0.5~0.6份、柠檬酸三乙酯0.5~2份、表面活性剂1.5~3.5份、柠檬酸0.1~0.5份、氯化钠0.1~0.55份、异鲸蜡醇硬脂酸酯5~12份、甲基丙二醇2~6份、C11-15仲链烷醇聚醚-12 2~6份、PEG-10聚二甲基硅氧烷1~3份、乙基己基甘油0.2~0.5份、维生素E1.5~3份和苯氧乙醇0.01份。
进一步地,所述组合物中表面活性剂选自甲基椰油酰基牛磺酸钠、牛脂基三甲基氯化铵和/或椰油酰氨基丙酸钠中的一种或组合。
进一步地,所述表面活性剂为质量比(1~5):0.1:(0.5~2)的甲基椰油酰基牛磺酸钠、牛脂基三甲基氯化铵和椰油酰氨基丙酸钠复合物。
第二方面,本发明提供了所述芦荟提取物的制备方法为:将芦荟去皮后捣碎成浆状,加入5~15倍质量的乙醇浸泡、超声、过滤,浓缩。
第三方面,本发明提供了贻贝粘蛋白头皮修护组合物制备方法,包括以下步骤:
S1、将异鲸蜡醇硬脂酸酯、柠檬酸三乙酯、C11-15仲链烷醇聚醚-12、PEG-10聚二甲基硅氧烷、乙基己基甘油、苯氧乙醇和氯化钠混合,开启搅拌,加热至75℃~90℃;
S2、将贻贝粘蛋白、芦荟提取物、牛磺酸、乙酰谷氨酰胺、柠檬酸、维生素E、甲基丙二醇、柠檬酸三乙酯和表面活性剂混合,开启搅拌,加热至55℃~85℃。
S3、将步骤S2制备的混合物加入至步骤S1物料中,继续搅拌,混合温度为60~80℃,然后均质后得到头皮修护组合物。
第四方面,本发明还提供了所述贻贝粘蛋白头皮修护组合物在头皮、毛发或毛囊修复护理品中的应用。
进一步地,所述头皮、毛发或毛囊修复护理品为液体、膏状、乳液、泡沫或喷雾的任一种形态,但产品并不局限于所述形态。
与现有技术相比,本发明的有益效果如下:
本发明制备的贻贝粘蛋白头皮修护组合物使用过程中能在头皮上形成一层薄膜,由于添加了贻贝粘蛋白具有优异的粘附效果,能显著提高组合物中其他调理成分的渗透和吸收,同时,另外,贻贝黏蛋白本身具有良好的修复效果,因此,本发明制备的贻贝粘蛋白头皮修护组合物有显著的头皮修护性能,修复头皮毛囊受损,有效减少头发脱落,使用15日约有9%~11%的防脱效果,使用30日有23%~26%的防脱效果,使用45日有40%~46%的防脱效果,使用60日有62%~71%的防脱效果。

除非另有定义,本发明所使用的所有的技术和科学术语与属于本发明的技术领域的技术人员通常理解的含义相同。本发明的说明书中所使用的术语只是为了描述具体的实施例的目的,不用于限制本发明。

实施例1贻贝粘蛋白头皮修护组合物的制备
本实施例制备的贻贝粘蛋白头皮修护组合物具体组成配方如表1所示,表1中贻贝粘蛋白头皮修护组合物主要成分的添加量均为质量份;

本实施例的贻贝粘蛋白头皮修护组合物采用表1中的成分制备得到,具体步骤为:
S1、将异鲸蜡醇硬脂酸酯、柠檬酸三乙酯、C11-15仲链烷醇聚醚-12、PEG-10聚二甲基硅氧烷、乙基己基甘油、苯氧乙醇、氯化钠和2/3质量份的水混合,开启搅拌,加热至90℃;
S2、将贻贝粘蛋白、芦荟提取物、牛磺酸、乙酰谷氨酰胺、柠檬酸、维生素E、甲基丙二醇、柠檬酸三乙酯、表面活性剂(甲基椰油酰基牛磺酸钠、牛脂基三甲基氯化铵和椰油酰氨基丙酸钠的质量比为1.5:0.1:1.8)和1/3质量份的水混合,开启搅拌,加热至75℃。
S3、将步骤S2制备的混合物加入至步骤S1物料中,继续搅拌,混合温度为70℃,适量补充混合过程中因加热散失的水分,20000r/min转速下均质2min得到护发组合物。
所述芦荟提取物的制备方法为:将芦荟去皮后捣碎成浆状,加入8倍质量的乙醇浸泡12h、超声30min、过滤,浓缩至干。
实施例2贻贝粘蛋白头皮修护组合物的制备
本实施例制备的贻贝粘蛋白头皮修护组合物具体组成配方如表2所示,表2中贻贝粘蛋白头皮修护组合物主要成分的添加量均为质量份;

本实施例的贻贝粘蛋白头皮修护组合物采用表2中的成分制备得到,具体步骤为:
S1、将异鲸蜡醇硬脂酸酯、柠檬酸三乙酯、C11-15仲链烷醇聚醚-12、PEG-10聚二甲基硅氧烷、乙基己基甘油、苯氧乙醇、氯化钠和2/3质量份的水混合,开启搅拌,加热至90℃;
S2、将贻贝粘蛋白、芦荟提取物、牛磺酸、乙酰谷氨酰胺、柠檬酸、维生素E、甲基丙二醇、柠檬酸三乙酯、表面活性剂(甲基椰油酰基牛磺酸钠、牛脂基三甲基氯化铵和椰油酰氨基丙酸钠的质量比为3.2:0.1:1.2)和1/3质量份的水混合,开启搅拌,加热至75℃。
S3、将步骤S2制备的混合物加入至步骤S1物料中,继续搅拌,混合温度为70℃,适量补充混合过程中因加热散失的水分,20000r/min转速下均质2min得到护发组合物。
所述芦荟提取物的制备方法为:将芦荟去皮后捣碎成浆状,加入12倍质量的75%乙醇浸泡12h、超声30min、过滤,浓缩至干。
实施例3贻贝粘蛋白头皮修护组合物的制备
本实施例制备的贻贝粘蛋白头皮修护组合物具体组成配方如表3所示,表3中贻贝粘蛋白头皮修护组合物主要成分的添加量均为质量份;

本实施例的贻贝粘蛋白头皮修护组合物采用表3中的成分制备得到,具体步骤为:
S1、将异鲸蜡醇硬脂酸酯、柠檬酸三乙酯、C11-15仲链烷醇聚醚-12、PEG-10聚二甲基硅氧烷、乙基己基甘油、苯氧乙醇、氯化钠和2/3质量份的水混合,开启搅拌,加热至90℃;
S2、将贻贝粘蛋白、芦荟提取物、牛磺酸、乙酰谷氨酰胺、柠檬酸、维生素E、甲基丙二醇、柠檬酸三乙酯、表面活性剂(甲基椰油酰基牛磺酸钠、牛脂基三甲基氯化铵和椰油酰氨基丙酸钠的质量比为4.5:0.1:0.5)和1/3质量份的水混合,开启搅拌,加热至75℃。
S3、将步骤S2制备的混合物加入至步骤S1物料中,继续搅拌,混合温度为70℃,适量补充混合过程中因加热散失的水分,20000r/min转速下均质2min得到护发组合物。
所述芦荟提取物的制备方法为:将芦荟去皮后捣碎成浆状,加入10倍质量的95%乙醇浸泡12h、超声30min、过滤,浓缩至干。
试验例一、本发明的贻贝粘蛋白头皮修护组合物的修护性能评价
评价前先对实施例1~3制备的贻贝粘蛋白头皮修护组合物进行相关的毒理学试验,经急性眼刺激性试验(30s)冲洗,结果为无刺激性;经急性皮肤刺激性试验,结果显示无刺激性;经皮肤光毒性试验,结果显示为未见皮肤光毒性;经皮肤变态反应试验,结果表明为未见皮肤变态反应。
修护性能评价:选择年龄在25-45周岁,头部皮肤健康无过敏或伤痕及无局部系统性其他疾病的脱发患者30名,男性15名,女性15名,分成三组,每组5名男性和5名女性,分别使用实施例1、2、3的贻贝粘蛋白头皮修护组合物,连续使用2个月,每两天清洁一次头发和头皮后使用,每次使用2~5mL,均匀涂抹至头发和头皮,保持2~3分钟,冲洗即可,使用前记录三次清洁头发和头皮后的头发脱落数(根)并取平均值,记录第15天、30天、45天和60天使用后的头发脱落数(根),计算使用后头发脱落数与使用前脱落数的差值,每组取平均值,记录在表4中;

由表4结果可知,本发明制备的贻贝粘蛋白头皮修护组合物对头皮的修护性能显著,由于脱发问题主要原因是头皮毛囊受损导致,因此,本发明的组合物对头皮毛囊具有修复作用,其中实施例1~3制备的组合物具有显著的防脱发作用,使用15日即约有9%~11%的防脱效果,使用30日约有23%~26%的防脱效果,使用45日约有40%~46%的防脱效果,使用60日约有62%~71%的防脱效果,其中以实施例2的防脱、修护效果最佳。
对比例1~2
对比例1与实施例2的区别在于,不添加贻贝粘蛋白,用水补足重量份,其余组分和制备步骤与实施例2一致。
对比例2与实施例2的区别在于,贻贝粘蛋白采用水解角蛋白,其余组分和制备步骤与实施例2一致。
对比例3~6
对比例3与实施例2的区别在于,不添加柠檬酸三乙酯,采用水补足重量份,其余组分和制备步骤与实施例2一致。
对比例4与实施例2的区别在于,柠檬酸添加量为1.5份,水为62.92份,其余组分和制备步骤与实施例2一致。
对比例5与实施例2的区别在于,不添加氯化钠,水补足重量份,其余组分和制备步骤与实施例2一致。
对比例6与实施例2的区别在于,氯化钠添加量为2份,水为62.34份,其余组分和制备步骤与实施例2一致。
对比例7~9
对比例7与实施例2的区别在于,表面活性剂(甲基椰油酰基牛磺酸钠和椰油酰氨基丙酸钠的质量比为3.2:1.2),其余组分和制备步骤一致。
对比例8与实施例2的区别在于,表面活性剂(甲基椰油酰基牛磺酸钠、牛脂基三甲基氯化铵和椰油酰氨基丙酸钠的质量比为3.2:1:1.2),其余组分和制备步骤一致。
对比例9与实施例2的区别在于,表面活性剂(牛脂基三甲基氯化铵和椰油酰氨基丙酸钠的质量比为1.4:4),其余组分和制备步骤一致。
试验例二、对比例1~9制备的组合物的修护性能评价
与试验例一评判方式一致,将对比例1~9进行相关的毒理学试验,经急性眼刺激性试验(30s)冲洗,结果为无刺激性;经急性皮肤刺激性试验,结果显示无刺激性;经皮肤光毒性试验,结果显示为未见皮肤光毒性;经皮肤变态反应试验,结果表明为未见皮肤变态反应。
使用评价:与试验例一评判方式一致,选择年龄在25-45周岁,头部皮肤健康无过敏或伤痕及无局部系统性其他疾病的脱发患者90名,男性45名,女性45名,分成9组,每组5名男性和5名女性,分别使用对比例1~9中的一组组合物样品,连续使用2个月,每两天清洁一次头发和头皮后使用,每次使用2~5mL,均匀涂抹至头发和头皮,保持2~3分钟,冲洗即可,使用前记录三次清洁头发和头皮后的头发脱落数(根)并取平均值,记录第15天、30天、45天和60天使用后的头发脱落数(根),计算使用后头发脱落数与使用前脱落数的差值,每组取平均值,记录在表5中;

由表5结果可知,相比实施例2,对比例1~9制备的组合物的修护性能明显降低,其中对比例1~2的防脱修护效果最差,表明本发明的组合物中的贻贝粘蛋白有显著的防脱修护效果;对比例4~6结果表明组合物中的柠檬酸和氯化钠对组合物的修护效果也有明显的影响,本发明采用的甲基丙二醇和C11-15仲链烷醇聚醚-12协同作为渗透成分,仅仅是通过分子链长以及空间位阻的层面提高的渗透作用,进一步基于贻贝黏蛋白的吸附方式,采用柠檬酸和氯化钠通过调节组合物的pH和离子浓度进而增强了组合物中有效活性成分的渗透作用;对比例7~9表明本发明采用的甲基椰油酰基牛磺酸钠、牛脂基三甲基氯化铵和椰油酰氨基丙酸钠复合表面活性剂对组合物的修护作用有显著影响,同时本发明通过氯化钠和柠檬酸的添加发现,对于该复合离子表面活性剂的作用有明显的影响。
试验例三、组合物的稳定性测定
对本发明实施例1~3制备的贻贝粘蛋白头皮修护组合物、对比例1~9制备的组合物的稳定性进行考察,具体结果见表6。

由表6结果看出,本发明制备的贻贝粘蛋白头皮修护组合物稳定性好,耐热、耐寒,而对比例3~9均出现不同程度的不稳定现象,其中柠檬酸三乙酯和氯化钠能显著提高组合物在低温下的稳定性,柠檬酸对组合物高温、低温下的稳定性均有影响,表面活性剂对组合物的高温下稳定性有影响。

Claims (4)
1.一种贻贝粘蛋白头皮修护组合物,其特征在于,按重量份数计,所述组合物中成分包含贻贝粘蛋白0.1~2份、芦荟提取物1~10份、牛磺酸0.1~1份、乙酰谷氨酰胺0.5~1份、柠檬酸三乙酯0.5~3份、表面活性剂1~5份、柠檬酸0.1~1份、氯化钠0.01~1份、异鲸蜡醇硬脂酸酯1~15份、甲基丙二醇1~10份、C11-15仲链烷醇聚醚-12 1~10份、PEG-10聚二甲基硅氧烷1~5份、乙基己基甘油0.1~0.5份、维生素E 0.5~3份和苯氧乙醇0.01~0.2份;
所述芦荟提取物的制备方法为:将芦荟去皮后捣碎成浆状,加入5~15倍质量的乙醇浸泡、超声、过滤,浓缩;
所述表面活性剂为质量比(1~5):0.1:(0.5~2)的甲基椰油酰基牛磺酸钠、牛脂基三甲基氯化铵和椰油酰氨基丙酸钠复合物。
2.根据权利要求1所述的贻贝粘蛋白头皮修护组合物,其特征在于,按重量份数计,所述组合物中成分包含贻贝粘蛋白0.1~1份、芦荟提取物2~6份、牛磺酸0.1~0.5份、乙酰谷氨酰胺0.5~0.6份、柠檬酸三乙酯0.5~2份、表面活性剂1.5~3.5份、柠檬酸0.1~0.5份、氯化钠0.1~0.55份、异鲸蜡醇硬脂酸酯5~12份、甲基丙二醇2~6份、C11-15仲链烷醇聚醚-122~6份、PEG-10聚二甲基硅氧烷1~3份、乙基己基甘油0.2~0.5份、维生素E1.5~3份和苯氧乙醇0.01份。
3.如权利要求1或2所述贻贝粘蛋白头皮修护组合物在头皮、毛发或毛囊修复护理品中的应用。
4.根据权利要求3所述的应用,其特征在于,所述头皮、毛发或毛囊修复护理品为液体、膏状、乳液、泡沫或喷雾的任一种形态。
