Abstract

背景技术
皮肤损伤伴随着皮肤正常结构的破坏、缺损和外界环境细菌的侵入,由此造成的伤口感染、伤口延迟愈合不仅给患者带来了肉体疼痛和精神创伤,还给医疗卫生系统带来了巨大的支出。以细菌感染为例,由于抗生素使用过程中细菌对抗生素的耐药性不断增加,探索有效的替代策略对抗细菌感染至关重要。自2007年首次报道了四氧化三铁纳米颗粒作为过氧化物酶模拟物的研究以来,纳米材料的靶向生物处理引起了人们的广泛关注。纳米酶作为下一代模仿天然酶,在伤口愈合应用中具有成本低、活性好、稳定性高等显著优势。特别是过氧化物酶类模拟物可以将过氧化氢分解为高活性的羟基自由基,对细菌具有毒害作用,抑制细菌耐药的发生。大量的纳米材料,包括金属纳米颗粒、二维材料和纳米碳,被认为具有本征酶样活性。虽然在纳米技术的推动下取得了很大的进展,但由于传统纳米酶结构的不均匀性,纳米酶的生物催化位点的定制和内在活性的进一步激发仍然是一个很大的挑战。利用稳定的结构载体使活性位点在纳米酶上均匀分散是一种有趣的方法。

发明内容
针对现有技术的不足,本发明利用杂原子掺杂策略,利用莫特-肖特基效应合成了超细锰负载的氮、氧掺杂纳米碳作为纳米酶。作为一种理想的半导体,原位锰纳米团簇可以锚定在氮、氧掺杂的纳米碳中,以避免活性位点聚集失活。因此,根据莫特-肖特基催化剂的结构,活性金属纳米团簇与纳米碳之间的电子密度可以有效降低分解过氧化氢的能垒。同时,掺杂氮、氧的纳米碳调节了金属位点的电荷态,使其易于释放羟基自由基。与没有金属活性位点的纳米碳相比,表现出突出的类过氧化物酶活性样,促进感染伤口高效愈合。

将甲壳素粉末和氯化钠的混合物加入球磨机中研磨,然后在氩气氛下的管式炉中程序升温热解,从25℃升温至700℃,随后用盐酸水溶液和去离子水清洗,产物经冷冻干燥得到黑色多孔纳米碳;
(2)纳米酶的制备
将四水合醋酸锰粉末加入到乙醇和去离子水的混合溶剂中,得到锰离子溶液,将多孔纳米碳粉末加入到锰离子溶液中,在室温下、在波长为365nm的紫外光下进行处理,以还原锰离子;然后将混合物依次进行离心、洗涤、冷冻干燥,得到所述甲壳素基多功能纳米酶。
根据本发明一种优选实施方式,步骤(1)中,所述甲壳素粉末与氯化钠的重量比为1:0.6-0.8。
根据本发明一种优选实施方式,步骤(1)中,所述研磨的条件包括:温度为室温,转速为300-500转/分,时间为1-3h,次数为2-3次。
根据本发明一种优选实施方式,步骤(1)中,所述热解的升温速率为8-12℃min-1,所述氩气的流速为40-60mL min-1。
根据本发明一种优选实施方式,步骤(1)中,所述盐酸的浓度为0.8-1.2M。
根据本发明一种优选实施方式,步骤(2)中,所述四水合醋酸锰在混合溶剂中的浓度为0.4-1.2mg/mL;所述乙醇与去离子水的体积比为8-10:1。
根据本发明一种优选实施方式,步骤(2)中,所述锰离子溶液与多孔纳米碳粉末的重量比为1:4-10。
根据本发明一种优选实施方式,步骤(2)中,紫外光处理的时间为0.8-1.2h。
本发明的第二方面提供由上述制备方法制得的甲壳素基多功能纳米酶。
本发明根据杂原子掺杂策略,利用莫特-肖特基效应合成了超细锰负载的氮、氧掺杂纳米碳作为纳米酶。原位锰纳米团簇可以锚定在氮、氧掺杂的纳米碳中,以避免活性位点聚集失活。因此,根据莫特-肖特基催化剂的结构,活性金属纳米团簇与纳米碳之间的电子密度可以有效降低分解过氧化氢的能垒。同时,掺杂氮、氧的纳米碳调节了金属位点的电荷态,使其易于释放羟基自由基,解决了传统纳米酶制备流程复杂、可控性差和催化活性低等问题。结合材料优异的光热效应,协同催化,表现出突出的类过氧化物酶活性样,兼具生物相容性合长效抗菌性能。因此,本发明的第三方面提供上述的甲壳素基多功能纳米酶在制备皮肤损伤愈合剂中的应用,该纳米酶在促进感染伤口愈合方面具有广阔的应用前景和市场价值。
本发明的纳米酶具有以下技术优势:
现有纳米酶的生物相容性极大地限制了其在生物医用领域的应用。本发明能够解决纳米酶大多潜在毒性大、可控性差等问题,利用自然界中含量最丰富的天然氨基多糖甲壳素。通过控制热解,长碳水化合物骨架转化为生物质衍生的多孔纳米碳,其有序的生物结构和特定的元素组成使多孔纳米碳具有独特的纳米结构和杂原子掺杂,制备一种生物相容性良好的纳米酶材料。
传统纳米酶结构大多不均匀,纳米酶的生物催化位点的定制和内在活性的进一步激发仍然是一个很大的挑战。本发明利用稳定的结构载体甲壳素衍生多孔纳米碳,其中含有大量氮、氧掺杂能提供锚固位点使活性位点在纳米酶上均匀分散解决其分散活性位点分散不均匀,活性不强的问题,制备高催化活性纳米酶材料。
现有的金属掺杂纳米酶大多制备过程复杂且催化活性低,根据莫特-肖特基催化剂的结构,活性金属纳米团簇与纳米碳之间的电子密度可以有效降低分解过氧化氢的能垒。同时,掺杂氮、氧的纳米碳调节了金属位点的电荷态,使其易于释放活性氧。本发明利用杂原子掺杂策略,合成了超细锰纳米粒子负载的氮、氧掺杂纳米碳作为纳米酶,解决制备复杂接活性低的问题。
本发明的其它特征和优点将在随后具体实施方式部分予以详细说明。

具体实施方式
下面将更详细地描述本发明的优选实施方式。虽然以下描述了本发明的优选实施方式,然而应该理解,可以以各种形式实现本发明而不应被这里阐述的实施方式所限制。
实施例1
(1)多孔纳米碳的制备
如图1所示,将甲壳素粉末(1.0g)和氯化钠(NaCl,0.7g)的混合物加入球磨机中,在室温下以400转/分的速度研磨2.0h,研磨两次。然后在氩气氛下的管式炉中程序升温热解,流速为50mL min-1。热解过程:从25℃到700℃,升温速率为10℃min-1,持续2小时。随后用1M盐酸水溶液(HCl,200mL)和去离子水清洗,最终产物经冷冻干燥得到黑色多孔纳米碳(PNCs)。
(2)纳米酶的制备
负载锰纳米粒子的多孔纳米碳的制备如图1所示:将27.0mg和54.0mg的四水合醋酸锰(Mn(OAc)2·4H2O)粉末分别加入到含有45mL乙醇和5mL去离子水的混合溶剂中,得到锰离子溶液。然后,将500mg多孔纳米碳粉末加入溶液中,在室温下,在波长为365nm的紫外线(UV)光下处理1小时,以还原锰离子。然后将混合物离心,用去离子水洗涤三次,冷冻干燥后得到制备好的样品低锰纳米粒子的纳米酶(L-Mn-PNCs)和高锰纳米粒子的纳米酶(Mn-PNCs)。
测试例1纳米酶的形貌表征
将纳米酶分散在样品台的导电胶上,利用扫描电镜(SEM,Zeiss)观察纳米酶表面形貌、将纳米酶分散在去离子水中,滴在铜网上,通过透射电镜(TEM,aJEM-2010)、高角环形暗场像扫描透射电镜(HADDF-STEM,JEOL JEM-ARM200F)观察纳米酶表面形貌以及负载的锰纳米粒子结构。
如图2所示,由于甲壳素层状结构之间存在较强的氢键相互作用,大量的官能团不能完全暴露,导致配位能力较弱,金属位点不稳定。根据盐模板法,NaCl在600℃以上开始分解,热解气体能剥离甲壳素层状结构。通过扫描显微镜可以清晰地呈现出多孔纳米碳的三维整体框架,其中间隔着巨大的亚微米级大孔。
如图3所示,透射电子图像和扫描电子图结果一致。多空纳米碳呈现出高度发达的、相互连接的、连续的三维网络结构,将有望提供快速的电荷转移路径和反应物扩散。
如图4所示,进一步在大尺度上进行纳米酶结构的准确观察,利用高角度环形暗场扫描透射电镜进行了表征。显示锰纳米粒子均匀分布在碳骨架中,与透射电子图像的观测结果吻合较好。
如图5所示,相关元素分布结果进一步验证了纳米酶中碳、氮、氧和锰元素的均匀分布。
如图6所示,从随机TEM区域的一些颗粒的样本量中获得了颗粒直径的直方图。可见,纳米酶平均粒径较小(3.0±1nm),粒径分布较窄。颗粒尺寸小可能是由于纳米碳网络的约束和氮、氧掺杂区域的存在,从而抑制了颗粒的团聚。
测试例2纳米酶的过氧化物酶样活性测定
将多孔纳米碳(PNCs)、低锰纳米粒子的纳米酶(L-Mn-PNCs)和高锰纳米粒子的纳米酶(Mn-PNCs)(30μL,100μg mL-1)分别与3,3',5,5'-四甲基联苯胺溶液(50μL,1mM)和过氧化氢溶液(100μL,100mM)在醋酸-醋酸钠缓冲液(120μL,pH 4.0)中在室温下混合。用紫外-可见光分光光谱仪记录吸收光谱。
如图7所示,过氧化氢在酸性溶液中首先吸附在纳米酶表面,获得过氧化物酶样活性。所以体系中过氧化氢分子倾向于破坏氧氧键来生成羟基自由基,它具有很强的氧化性,很容易从有机底物中提取氢原子,用作氧化剂氧化3,3',5,5'-四甲基联苯胺后,可与预吸附的氢离子结合生成水。生成的蓝色的3,3',5,5'-四甲基联苯胺氧化物,在652nm处的特征吸收峰强度与酶的活性成正比,其中负载锰纳米粒子的纳米酶类过氧化物酶活性最强,吸收峰最高。
测试例3纳米酶的光热效应检测
700至1000nm的近红外光因其能量低、穿透力强,对人体组织副作用小等优点而广泛使用。选择808nm激光器作为测试光源测试纳米酶的光热转换效率,具体地,用808nm近红外激光固定距离5cm照射含有不同样品的缓冲液(1mL)。以醋酸-醋酸钠缓冲液为空白对照,用FLIR E50红外摄像机监测和记录预定时间内,样品温度变化和红外热图像。
如图8所示,近红外照射3min后负载锰纳米粒子的纳米酶温度由20℃升至42℃,而纯醋酸缓冲液温度几乎没有变化,验证了纳米酶优异的光热性能。
测试例4纳米酶抗菌实验
通过细菌形貌结构评价纳米酶的抑菌能力。实验分为6组:(1)PBS组,(2)H2O2组,(3)PNCs组,(4)PNCs+H2O2+L组,(5)L-Mn-PNCs+H2O2+L组,(6)Mn-PNCs+H2O2+L组,L指808nm近红外激光照射3分钟,对照组采用相同的方法进行表面抗菌试验。然后用PBS冲洗细菌悬液三次。在2.5%戊二醛中固定12小时后,用一系列乙醇溶液(每种浓度为20%、40%、60%、80%、100%、100%,10分钟)对细菌样品进行脱水。最后,将细菌样品转移到样品台导电胶上,真空喷金并使用扫描电子显微镜(SEM)进行表征。
如图9所示,PBS组中所有细菌形态正常,PNCs、L-Mn-PNCs和Mn-PNCs组中细菌膜出现粗糙皱褶,细菌失去了原来的形状,不易识别,表明纳米酶催化产生的羟基自由基对细菌细胞膜具有很强的损伤作用。外源过氧化氢的加入和近红外辐射可增强对细菌形态的破坏作用,Mn-PNCs+H2O2+L处理组细菌几乎被完全破坏成碎片,说明纳米酶类过氧化物酶活性催化可急剧产生大量活性氧。而近红外辐射带来的光动力效应可以进一步促进活性氧的产生。
测试例5纳米酶的生物相容性实验
以L929细胞为模型细胞,每孔接种1×104细胞后,分别加入不同浓度的PNCs、L-Mn-PNCS、Mn-PNCs和对照组(空白组)在37℃共培养3天后,采用细胞计数试剂盒评价纳米酶的细胞相容性。
如图10所示,利用CCK-8法体外评估纳米酶对小鼠成纤维细胞的毒性情况。实验结果表明纳米酶在0~500μg mL-1浓度范围内培养3天,无明显细胞毒性,细胞存活率均高于96.3%,表现出优异的生物相容性。
以上已经描述了本发明的各实施例,上述说明是示例性的,并非穷尽性的,并且也不限于所披露的各实施例。在不偏离所说明的各实施例的范围和精神的情况下,对于本技术领域的普通技术人员来说许多修改和变更都是显而易见的。
Claims (10)
1.一种甲壳素基多功能纳米酶的制备方法,包括以下步骤:
(1)多孔纳米碳的制备
将甲壳素粉末和氯化钠的混合物加入球磨机中研磨,然后在氩气氛下的管式炉中程序升温热解,从25℃升温至700℃,随后用盐酸水溶液和去离子水清洗,产物经冷冻干燥得到黑色多孔纳米碳;
(2)纳米酶的制备
将四水合醋酸锰粉末加入到乙醇和去离子水的混合溶剂中,得到锰离子溶液,将多孔纳米碳粉末加入到锰离子溶液中,在室温下、在波长为365nm的紫外光下进行处理,以还原锰离子;然后将混合物依次进行离心、洗涤、冷冻干燥,得到所述甲壳素基多功能纳米酶。
2.根据权利要求1所述的制备方法,其特征在于,步骤(1)中,所述甲壳素粉末与氯化钠的重量比为1:0.6-0.8。
3.根据权利要求1所述的制备方法,其特征在于,步骤(1)中,所述研磨的条件包括:温度为室温,转速为300-500转/分,时间为1-3h,次数为2-3次。
4.根据权利要求1所述的制备方法,其特征在于,步骤(1)中,所述热解的升温速率为8-12℃min-1,所述氩气的流速为40-60mL min-1。
5.根据权利要求1所述的制备方法,其特征在于,步骤(1)中,所述盐酸的浓度为0.8-1.2M。
6.根据权利要求1所述的制备方法,其特征在于,步骤(2)中,所述四水合醋酸锰在混合溶剂中的浓度为0.4-1.2mg/mL;所述乙醇与去离子水的体积比为8-10:1。
7.根据权利要求1所述的制备方法,其特征在于,步骤(2)中,所述锰离子溶液与多孔纳米碳粉末的重量比为1:4-10。
8.根据权利要求1所述的制备方法,其特征在于,步骤(2)中,紫外光处理的时间为0.8-1.2h。
9.由权利要求1-8中任意一项所述的制备方法制得的甲壳素基多功能纳米酶。
10.权利要求9所述的甲壳素基多功能纳米酶在制备皮肤损伤愈合剂中的应用。
