Abstract
本发明涉及医药技术领域,公开了一种聚合物核酸递送载体及药物组合物应用。该聚合物核酸递送载体的结构如式(I)所示,其制备方法包括:在反应溶剂I存在的条件下,将式(II)所示的化合物、式(III)所示的化合物、L‑赖氨酸二异氰酸酯与2,2‑双(溴甲基)‑1,3‑丙二醇混合进行反应I,再与甲氧基聚乙二醇‑羟基混合进行反应II得到式(IV)所示的反应物P1;在反应溶剂II存在的条件下,将反应物P1与式(V)所示的化合物混合进行反应III。所述药物组合物含有siRNA以及聚合物核酸递送载体。该聚合物核酸递送载体在光刺激下可有效诱发免疫原性细胞死亡效应,药物组合物具有较好的内涵体逃逸效果和体内外安全性,可有效抑制多种肿瘤的生长。

背景技术
非侵入性治疗方法为肿瘤治疗开辟了新的途径。光动力疗法(PhotodynamicTherapy,PDT)作为美国食品和药物管理局批准的一种非侵入性治疗方法,已广泛应用于临床各种病理治疗。传统的光动力治疗的穿透深度有限(1-3mm),以及肿瘤深部缺少氧气导致产生较少活性氧/自由基,均会降低光动力的治疗效果。此外,光动力疗法的光敏剂自身经静脉注射时对肿瘤的选择性较低,会引起全身的毒性,影响光敏剂的临床效果。与NIR-I窗口(650-900nm)相比,在NIR-II窗口(1000-1700nm)光谱区域的光动力治疗具有更深穿透(5mm)、最小化组织散射和吸收。
尽管光动力疗法的效果令人鼓舞,但在肿瘤复发和转移方面存在局限性,而超过90%的癌症相关死亡是转移造成的,光动力疗法通过免疫原性细胞死亡可触发肿瘤免疫,但其效果不足以完整地消除肿瘤。
肿瘤微环境是肿瘤发生转移、免疫抑制功能的主要场所。越来越多的数据表明,肿瘤细胞通过上调与免疫抑制和血管生成相关的基因来促进肿瘤的生长和转移。光动力产生的活性氧会诱导肿瘤细胞免疫抑制和耐受,释放多种免疫抑制因子,甚至会主动上调多种基因来辅助肿瘤转移和抑制T细胞的免疫功能。增强的免疫抑制微环境可逐渐限制免疫细胞功能,导致肿瘤适应性免疫阻滞。因此,光动力疗法和其他治疗方式的结合是提高PDT抗癌治疗成功的可行策略。其中,siRNA基因治疗(阻断免疫检查点)与光动力的联合治疗因其协同治疗效果而成为一种很有前景的治疗策略。
PD-1/PD-L1(Programmed Death-1,Programmed Death-ligand 1)是近几年备受关注的免疫检查点。PD-1在免疫细胞中广泛表达,如毒性T淋巴细胞、自然杀伤细胞、B细胞、树突状细胞、巨噬细胞等,具有免疫抑制功能。当受到细胞毒性T淋巴细胞分泌的细胞因子作用时,为实现免疫逃逸,肿瘤细胞会上调PD-L1的表达。值得注意的是,毒性T淋巴细胞会被肿瘤细胞(如CT26和4T1)高表达的PD-L1特异性结合,而使自身的免疫功能受到抑制。因此,肿瘤通过PD-L1的高表达实现免疫逃逸。
在将siRNA基因治疗与光动力的联合治疗的载体中,聚合物纳米颗粒是应用较为广泛的联合治疗载体。研究表明,可通过在聚合物中修饰肿瘤微环境响应型基团来实现在肿瘤部位的siRNA与光敏剂的特异性释放。与非响应型聚合物相比,肿瘤微环境响应型聚合物显然更有优势,因为后者容易在生理环境下泄露引起全身的毒性,带来生物安全性问题。使用RNAi技术(siRNA)阻断肿瘤细胞高表达的PD-L1,打破肿瘤微环境免疫抑制,增强细胞毒性T细胞浸润,释放细胞因子杀伤肿瘤可实现协同放大的治疗效果。
但是,目前关于肿瘤微环境响应型聚合物的研究较少,其细胞毒性与使用安全性还有待提升。
发明内容
本发明的目的是为了克服现有技术存在的肿瘤微环境响应型聚合物的细胞毒性与使用安全性有待提升的问题,提供一种聚合物核酸递送载体及药物组合物应用,该聚合物核酸递送载体在光刺激下可有效诱发免疫原性细胞死亡效应,药物组合物具有较好的内涵体逃逸效果和体内外安全性,可有效抑制多种肿瘤的生长。
为了实现上述目的,本发明第一方面提供一种聚合物核酸递送载体,该聚合物核酸递送载体的结构如式(I)所示,
式(I),其中,m、n、p各自独立地为3-15的正整数,q为40-120的正整数。
本发明第二方面提供一种聚合物核酸递送载体的制备方法,包括以下步骤:
(1)在反应溶剂I存在的条件下,将式(II)所示的化合物、式(III)所示的化合物、L-赖氨酸二异氰酸酯与2,2-双(溴甲基)-1,3-丙二醇混合进行反应I,再与甲氧基聚乙二醇-羟基混合进行反应II得到式(IV)所示的反应物P1;
(2)在反应溶剂II存在的条件下,将反应物P1与式(V)所示的化合物混合进行反应III,得到式(I)所示的聚合物;
本发明第三方面提供一种药物组合物,含有siRNA以及前述的聚合物核酸递送载体和/或前述的制备方法制得的聚合物核酸递送载体。
本发明第四方面提供一种制备药物组合物的方法,包括以下步骤:将前述的聚合物核酸递送载体和/或前述的制备方法制得的聚合物核酸递送载体与胶束剂进行混合反应I得到聚合物胶束,将所述聚合物胶束与siRNA进行混合反应II。
本发明第五方面提供前述的聚合物核酸递送载体、前述的制备方法制得的聚合物核酸递送载体、前述的药物组合物和前述的方法制得的药物组合物中的至少一者在制备抗肿瘤药物中的应用。
通过上述技术方案,本发明的有益效果为:
本发明提供的聚合物核酸递送载体,在NIR-I区具有宽吸收峰(700-900nm)并在NIR-II区具有较强的荧光发射(900-1100nm),其侧链上阳离子单元可有效负载siRNA,主链上的缩硫醇键在肿瘤微环境高ROS水平下响应断裂,在808nm激光照射下可有效产生活性氧,具有良好的细胞毒性;该聚合物与siRNA形成的药物组合物具有理想的粒径、电势,能够有效地被细胞内吞,具有较好的内涵体逃逸效果,实现荷载核酸的逃逸与释放;该药物组合物在体内外同时具有较好的安全性,且在长时间下依然保持较高的基因沉默效率,能够有效抑制多种肿瘤的生长、促进肿瘤细胞的凋亡。

本发明提供的如式(I)所示的聚合物核酸递送载体属于阳离子聚合物,基于NIR-II Aza-BODIPY(氮杂氟硼二吡咯),主链上的缩硫醇键在肿瘤微环境高ROS水平下响应断裂,侧链修饰的胆固醇季铵化阳离子单体能够有效负载siRNA;该聚合物核酸递送载体在NIR-I区具有宽吸收峰(700-900nm)并在NIR-II区具有较强的荧光发射(900-1100nm),具有光敏效应,在808nm激光照射下,可有效产生活性氧,进而具有良好的细胞毒性。

本发明第二方面提供一种聚合物核酸递送载体的制备方法,包括以下步骤:
(1)在反应溶剂I存在的条件下,将式(II)所示的化合物、式(III)所示的化合物、L-赖氨酸二异氰酸酯与2,2-双(溴甲基)-1,3-丙二醇混合进行反应I,再与甲氧基聚乙二醇-羟基混合进行反应II得到式(IV)所示的反应物P1;
(2)在反应溶剂II存在的条件下,将反应物P1与式(V)所示的化合物混合进行反应III,得到式(I)所示的聚合物;
本发明中,式(II)所示的化合物、式(III)所示的化合物、L-赖氨酸二异氰酸酯、2,2-双(溴甲基)-1,3-丙二醇和甲氧基聚乙二醇-羟基可以分别商购获得,也可以根据现有技术中公开的方法制备获得。其中,式(III)所示的化合物为2,2'-(propane-2,2-diylbis(sulfanediyl))bis(ethan-1-ol),简称DSB。
根据本发明,步骤(1)中各原料的用量可在较宽的范围内选择。优选地,步骤(1)中所述式(II)所示的化合物、式(III)所示的化合物、L-赖氨酸二异氰酸酯、2,2-双(溴甲基)-1,3-丙二醇与甲氧基聚乙二醇-羟基的重量比为1:0.8-1.2:2-2.8:0.9-1.3:5-6。发明人发现,在该优选实施方式下,有利于提高式(I)所示的聚合物的制备效率,提升其荧光效应和细胞毒性。
根据本发明,所述甲氧基聚乙二醇-羟基可以选用常规聚合度的甲氧基聚乙二醇-羟基(mPEG-OH)产品,优选地,所述甲氧基聚乙二醇-羟基的分子量为2000-5000,例如mPEG2000-OH、mPEG5000-OH。
根据本发明,在上述式(I)所示的聚合物的制备方法中,反应I的条件可在较宽的范围内选择。优选地,所述反应I的条件包括:温度为45-55℃,具体可以为45℃、47℃、49℃、51℃、53℃、55℃,或者上述两个数值之间的任意值;时间为10-15h,具体可以为10h、11h、12h、13h、14h、15h,或者上述两个数值之间的任意值。发明人发现,在该优选实施方式下,有利于提高式(I)所示的聚合物的产率。

根据本发明,优选地,所述反应溶剂I和所述反应溶剂II分别独立地选自无水N,N-二甲基甲酰胺、无水二氯甲烷、无水四氢呋喃和无水二甲基亚砜中的至少一种。更优选地,所述反应溶剂I和所述反应溶剂II分别为无水N,N-二甲基甲酰胺。发明人发现,在该优选实施方式下,有利于提高式(I)所示的聚合物制备时的反应效率。
根据本发明,可以选用本领域常规的分离方法将反应物P1从所述反应II得到的反应液中分离出。优选地,所述步骤(1)还包括:将所述反应II得到的反应液进行透析I得到透析液I,将所述透析液I进行干燥得到所述式(IV)所示的反应物P1。示例性地,可以将反应II得到的反应液与少量水混合后,采用7000MW透析袋截留目标反应物。
根据本发明,可以选用本领域常规的分离方法将聚合物从所述反应III得到的反应液中分离出。优选地,所述步骤(2)还包括:将所述反应III得到的反应液进行透析II得到透析液II,将所述透析液II进行干燥得到所述式(I)所示的聚合物。
本发明中,所述干燥可以选用本领域内常规的干燥方法,例如真空冷冻干燥、热风干燥、冷冻干燥等,优选地,所述透析液I和所述透析液II的干燥采用冷冻干燥,具体的冷冻干燥的过程包括:将所述透析液I或者所述透析液II分别在-90℃至-70℃条件下预冷25-35min,再进行冷冻干燥40-60h。
本发明第三方面提供一种药物组合物,含有siRNA以及前述的聚合物核酸递送载体和/或前述的制备方法制得的聚合物核酸递送载体。
本发明所提供的聚合物核酸递送载体与siRNA形成的药物组合物具有理想的粒径、电势,能够通过降解靶基因导致mRNA基因沉默,通过钙网蛋白的暴露、三磷酸腺苷的分泌和高迁移率组蛋白B1的释放引起免疫原性细胞死亡,从而介导机体的免疫应答;在CT26结直肠癌、4T1-Luc乳腺癌、肝癌患者癌组织异种移植肿瘤模型中PTSQ/siRNA复合物均可有效抑制肿瘤生长,促进癌细胞凋亡;该药物组合物具有较好的内涵体逃逸效果,实现荷载核酸的逃逸与释放,在体内外同时具有较好的安全性,且在长时间下依然保持较高的基因沉默效率,能够有效抑制多种肿瘤的生长、促进肿瘤细胞的凋亡,实现光动力肿瘤治疗。
根据本发明,所述药物组合物中聚合物核酸递送载体与siRNA的用量可以在较宽的范围内选择。优选地,所述聚合物核酸递送载体与所述siRNA的重量比为5-50:1。发明人发现,在该优选实施方式下,有利于提高药物组合物对肿瘤细胞生长的抑制性能。
根据本发明,所述siRNA可以选用本领域内能够负载于胆固醇季铵化阳离子上,且具有特异性靶向治疗靶点的RNA药物,例如siRNA为选择性沉默肿瘤细胞高表达的PD-L1蛋白的RNA药物(siRNA对)。优选地,所述siRNA含有完全反向互补的正义链和反义链,所述正义链和所述反义链的核苷酸序列分别如SEQ ID NO.1和SEQ ID NO.2所示;
正义链(SEQ ID NO.1):5’-AGACGUAAGCAGUGUUGAA-3’;

本发明第四方面提供一种制备药物组合物的方法,包括以下步骤:将前述的聚合物核酸递送载体和/或前述的制备方法制得的聚合物核酸递送载体与胶束剂进行混合反应I得到聚合物胶束,将所述聚合物胶束与siRNA进行混合反应II。
本发明中,将聚合物核酸递送载体形成聚合物胶束后,与siRNA通过静电吸附作用结合,以得到药物组合物,制备过程工艺简单、便于操作。
根据本发明,优选地,所述聚合物核酸递送载体与所述siRNA的重量比为5-50:1。
根据本发明,优选地,所述siRNA含有完全反向互补的正义链和反义链,所述正义链和所述反义链的核苷酸序列分别如SEQ ID NO.1和SEQ ID NO.2所示。
根据本发明,所述胶束剂可以选用本领域内能够使得式(I)所示的聚合物形成胶束的胶束剂。优选地,所述胶束剂为超纯水;相对于1mg所述聚合物核酸递送载体,所述胶束剂的用量为0.1-0.3mL。发明人发现,在该优选实施方式下,有利于提高对式(I)所示的聚合物的胶束效率,且胶束效果更优。
根据本发明,优选地,所述混合反应I的过程包括:将所述聚合物核酸递送载体溶于溶剂后,滴入所述胶束剂中,再在温度为5-40℃、转速为80-150rpm的条件下搅拌25-35min得到混合反应液,将所述混合反应液进行透析III得到聚合物胶束。发明人发现,在该优选实施方式下,有利于优化对聚合物核酸递送载体形成胶束的效果,进而提高药物组合物的产率。
本发明中,所述聚合物核酸递送载体的溶剂可以是本领域内常规能够溶解该聚合物核酸递送载体的溶剂,优选地,所述溶剂选自无水N,N-二甲基甲酰胺、无水二氯甲烷、无水四氢呋喃和无水二甲基亚砜中的至少一种,更优选为N,N-二甲基甲酰胺。
根据本发明,所述混合反应II的条件可在较宽的范围内选择。优选地,所述混合反应II的条件包括:温度为5-40℃,具体可以为5℃、10℃、15℃、20℃、25℃、30℃、35℃、40℃,或者上述两个数值之间的任意值;时间为25-35min,具体可以为25min、27min、29min、31min、33min、35min,或者上述两个数值之间的任意值。
根据本发明一种特别优选的实施方式,制备药物组合物的方法包括以下步骤:
(1)在反应溶剂I存在的条件下,将式(II)所示的化合物、式(III)所示的化合物、L-赖氨酸二异氰酸酯与2,2-双(溴甲基)-1,3-丙二醇以重量比为1:0.8-1.2:2-2.8:0.9-1.3混合,在温度为45-55℃条件下反应10-15h,再与甲氧基聚乙二醇-羟基(式(II)所示的化合物与甲氧基聚乙二醇-羟基的重量比为1:5-6)混合,在温度为45-55℃条件下反应10-15h得到反应液I,将反应液I进行透析I得到透析液I,将透析液I进行冷冻干燥得到式(IV)所示的反应物P1;
(2)在反应溶剂II存在的条件下,将反应物P1与式(V)所示的化合物以重量比为2.5-3.5:1混合,在温度为45-55℃条件下反应10-15h得到反应液II,将反应液II进行透析II得到透析液II,将所述透析液II进行冷冻干燥得到式(I)所示的聚合物;
(3)将式(I)所示的聚合物溶于溶剂后,滴入胶束剂中,再在温度为5-40℃、转速为80-150rpm的条件下搅拌25-35min得到混合反应液,将混合反应液进行透析III得到聚合物胶束,将所述聚合物胶束与核苷酸序列如SEQ ID No.1所示和SEQ ID No.2的siRNA混合后在温度为5-40℃的条件下静置25-35min得到药物组合物,其中,聚合物与siRNA的重量比为5-50:1。
本发明第五方面提供前述的聚合物核酸递送载体、前述的制备方法制得的聚合物核酸递送载体、前述的药物组合物和前述的方法制得的药物组合物中的至少一者在制备抗肿瘤药物中的应用。
本发明中,所述抗肿瘤药物除含有前述的聚合物核酸递送载体、前述的制备方法制得的聚合物核酸递送载体、前述的药物组合物和前述的方法制得的药物组合物中的至少一者,还可以含有药学上可接受的辅料组成和/或其他能够抑制肿瘤生长的化合物。
其中,所述辅料为药学上可接受的辅料,比如稳定剂、赋形剂、溶剂等。可以通过本领域已知的方法可将抗肿瘤药物制成以下形式:片剂、胶囊剂、水性或油性溶液剂、混悬剂、乳剂、乳膏剂、软膏剂、凝胶剂、喷鼻剂、栓剂、用于吸入的细小分散的粉剂或气雾剂或喷雾剂、用于胃肠道外(包括静脉内、肌内或输注)的无菌水性或油性溶液或混悬剂或无菌乳剂。
根据本发明,优选地,所述肿瘤的类型选自结直肠癌、乳腺癌和肝癌中的至少一种。

以下实施例中,紫外吸收光谱采用日本岛津仪器有限公司生产的紫外分光光度计(UVPC1601)测得;荧光发射光谱采用美国瓦里安有限公司公司生产的荧光光谱仪(CaryEclipse)测得;粒径和电势采用英国马尔文仪器有限公司生产的动态光散射仪(MalvernNano-s)测得;1H NMR图谱和13C NMR采用瑞士布鲁克公司生产的液体核磁共振波谱仪器(Bruker Avance 400)测得;其他参数检测所采用的仪器和方法均采用本领域常规的仪器和方法。

以下实施例中,在无特殊说明的情况下,室温为25±5℃。
实施例1
(1)准确称取20mg式(II)所示的化合物、20.72mg的DSB、47.81mg的L-赖氨酸二异氰酸酯、22.15mg的2,2-双(溴甲基)-1,3-丙二醇加入到5mL无水N,N-二甲基甲酰胺中,在温度为50℃条件下反应12h,再加入110.67mg的m-PEG5000-OH,在温度为50℃条件下继续反应12h得到反应液I,向反应液I中加入少量水,使用7000MW透析袋截留目标反应物,透析结束后将透析液I放进-80℃冰箱中预冷30min,然后冷冻干燥48h,得到式(IV-1)所示的反应物P1;
(2)称取60mg步骤(1)得到的反应物P1、20mg式(V)所示的化合物加入到5mL无水N,N-二甲基甲酰胺中,在温度为50℃条件下反应12h得到反应液II,向反应液II中加入少量水,使用7000MW透析袋截留目标聚合物,透析结束后将透析液II放进-80℃冰箱中预冷30min,然后冷冻干燥48h,得到式(I-1)所示的聚合物PTSQ;
实施例1中式(I-1)所示的聚合物PTSQ的合成路线如图1所示,式(IV-1)所示的反应物P1和式(I-1)所示的聚合物PTSQ的结构式及聚合度采用核磁共振波谱法确定。式(IV-1)所示的反应物P1的1HNMR图谱如图2所示,式(I-1)所示的聚合物PTSQ的1H NMR图谱如图3所示。从图2可以看出,反应物P1的1H NMR图谱中出现了式(II)所示的化合物(峰a和b)、m-PEG5000(峰c)、DSB(峰d)的特征峰,证实了这些单体成功连接在反应物P1的聚合物链上;从图3可以看出,聚合物PTSQ的1H NMR图谱中除了式(II)所示的化合物(峰a和b)、m-PEG5000(峰c)、DSB(峰d),还出现了式(V)所示的化合物(峰e)的特征峰,证实聚合物PTSQ成功合成。
实施例2
(1)准确称取20mg式(II)所示的化合物、16mg的DSB、40mg的L-赖氨酸二异氰酸酯、18mg的2,2-双(溴甲基)-1,3-丙二醇加入到5mL无水N,N-二甲基甲酰胺中,在温度为45℃条件下反应15h,再加入100mg的m-PEG5000-OH,在温度为45℃条件下继续反应15h得到反应液I,向反应液I中加入少量水,使用7000MW透析袋截留目标反应物,透析结束后将透析液I放进-70℃冰箱中预冷35min,然后冷冻干燥40h,得到式(IV-1)所示的反应物P1;
(2)称取50mg步骤(1)得到的反应物P1、20mg式(V)所示的化合物加入到5mL无水N,N-二甲基甲酰胺中,在温度为45℃条件下反应15h得到反应液II,向反应液II中加入少量水,使用7000MW透析袋截留目标聚合物,透析结束后将透析液II放进-70℃冰箱中预冷35min,然后冷冻干燥40h,得到式(I-1)所示的聚合物PTSQ。
实施例3
(1)准确称取20mg式(II)所示的化合物、24mg的DSB、56mg的L-赖氨酸二异氰酸酯、26mg的2,2-双(溴甲基)-1,3-丙二醇加入到5mL无水N,N-二甲基甲酰胺中,在温度为55℃条件下反应10h,再加入120mg的m-PEG5000-OH,在温度为55℃条件下继续反应10h得到反应液I,向反应液I中加入少量水,使用7000MW透析袋截留目标反应物,透析结束后将透析液I放进-90℃冰箱中预冷25min,然后冷冻干燥60h,得到式(IV-1)所示的反应物P1;
(2)称取70mg步骤(1)得到的反应物P1、20mg式(V)所示的化合物加入到5mL无水N,N-二甲基甲酰胺中,在温度为55℃条件下反应10h得到反应液II,向反应液II中加入少量水,使用7000MW透析袋截留目标聚合物,透析结束后将透析液II放进-90℃冰箱中预冷25min,然后冷冻干燥60h,得到式(I-1)所示的聚合物PTSQ。
实施例4
称取10mg实施例1制得的PTSQ聚合物,加入500μL的无水N,N-二甲基甲酰胺溶液,溶解后缓慢滴入到2mL超纯水中,室温下使用磁力搅拌器以转速100rpm旋转30min得到混合反应液,将混合反应液使用7000MW分子量的透析袋进行透析12h,收集透析溶液得到PTSQ胶束;将PTSQ胶束和siRNA(采用siNC)在室温下混合(PTSQ聚合物与siRNA的重量比分别为1:1、5:1、10:1、20:1、35:1、50:1),静置30min,得到PTSQ/siRNA药物组合物,简称PTSQ/siNC。
实施例4制得的药物组合物PTSQ/siRNA的结构示意图如图4所示。
实施例5
称取10mg实施例1制得的PTSQ聚合物,加入500μL的无水N,N-二甲基甲酰胺溶液,溶解后缓慢滴入到1mL超纯水中,室温下使用磁力搅拌器以转速150rpm旋转25min得到混合反应液,将混合反应液使用7000MW分子量的透析袋进行透析12h,收集透析溶液得到PTSQ胶束;将PTSQ胶束和siRNA(采用siNC)在室温下混合(PTSQ聚合物与siRNA的重量比分别为10:1、20:1、30:1、50:1、70:1、100:1),静置25min,得到PTSQ/siRNA药物组合物,简称PTSQ/siNC。
实施例5制得的药物组合物PTSQ/siRNA采用动态光散射仪检测粒径和电势,结果如图5所示,制得的药物组合物粒径均一、大小和电势合适,有利于药物的递送。
实施例6
称取10mg实施例1制得的PTSQ聚合物,加入500μL的无水N,N-二甲基甲酰胺溶液,溶解后缓慢滴入到3mL超纯水中,室温下使用磁力搅拌器以转速80rpm旋转35min得到混合反应液,将混合反应液使用7000MW分子量的透析袋进行透析12h,收集透析溶液得到PTSQ胶束;将PTSQ胶束和siRNA(采用siNC)在室温下混合(PTSQ聚合物与siRNA的重量比分别为10:1、25:1、50:1、75:1、100:1),静置35min,得到PTSQ/siRNA药物组合物,简称PTSQ/siNC。
测试例1
将实施例1制得的PTSQ聚合物进行紫外吸收光谱和荧光发射光谱分析,结果如图6所示。从图6可以看出,实施例1制得的PTSQ聚合物在808nm有强的紫外吸收峰,同时在900-1100nm处有强的宽荧光发射,在980nm有最强荧光发射,这表明PTSQ聚合物在光动力治疗中会具有更深的穿透深度。
测试例2PTSQ/siRNA的凝胶阻滞实验和物理特性
称取0.8g琼脂糖,加入40mL的TBE缓冲液,在微波炉中高火加热溶解成2重量%琼脂糖凝胶;然后加入4μL的1/10000的Super Red Gel RedTM(擎科,中国北京),用玻璃棒轻轻搅拌混匀;倒入模具中,在室温静置30min;待冷却凝固后,取10μL实施例4中制得的PTSQ/siNC溶液(颗粒的siRNA终浓度为25pmol),加入2μL上样缓冲液,吹打混匀后加入琼脂糖凝胶上样孔中;用1×TBE缓冲液(Tris/Acetate/EDTA缓冲液)作为电泳缓冲液,100V下电泳20min,然后通过凝胶成像仪进行成像拍照,以游离siRNA(Naked siNC)作为对照,结果如图7所示。
通过凝胶阻滞实验分析实施例4中PTSQ聚合物与siRNA为不同重量比时PTSQ聚合物结合负载siRNA药物的能力。如图7所示,游离的siRNA条带在琼脂糖凝胶上没有受到阻滞,出现了明显的迁移且条带亮度较高,当PTSQ聚合物与siRNA重量比为5:1,泳道中的siRNA条带完全消失,siRNA被滞留在凝胶上样孔内,PTSQ聚合物将siRNA完全组装在纳米颗粒中,siRNA与PTSQ聚合物完全结合形成稳定的复合物。基于凝胶阻滞结果可知,PTSQ聚合物与siRNA的重量比为5-50:1时,PTSQ聚合物材料已经完全负载siRNA。以下测试例中,在无特殊说明的情况下,PTSQ/siRNA均采用实施例4中以PTSQ聚合物与siRNA的重量比为50:1时制得的PTSQ/siRNA复合物进行。
测试例3药物组合物产生活性氧能力评价
1,3二苯基异苯并呋喃(DPBF)是一种常用的1O2检测指示剂,在420nm有紫外吸收峰,能够快速与1O2进行反应,反应后紫外吸收值减少。因此使用DPBF进行活性氧(ROS)检测,具体步骤如下:取10μL母液DPBF溶液,加入0.99mL的DMF,混合均匀后,在超声状态下加入到19mL水中,得到工作液;取3mg实施例4制得的PTSQ/siRNA(PTSQ与siRNA的重量比为50:1),溶解于1mL的DMF中,得到浓度为3mg/mL的PTSQ/siRNA溶液,取96孔板,加入190μL工作液,再加入10μL的PTSQ/siRNA溶液,立马用酶标仪检测300-600nm的紫外吸收值,作为0s吸收对照曲线,然后在新的孔中加入190μL工作液和10μL的PTSQ/siRNA溶液,使用808nm激光器进行照射,强度为2W/cm2,照射10s后进行检测,重复上一步操作,改变光照射时间(0-300s),记录检测数据,结果如图8所示,图8中从上至下的曲线依次与时间0s、10s、20s、30s、60s、75s、120s、180s、240s、300s对应。
利用CLSM图像评价游离siRNA(siNC组)、Lipo/siNC组、实施例4制得的PTSQ/siNC组(PTSQ与siRNA的重量比为50:1)以及PTSQ/siNC+L组在细胞系CT26产生活性氧能力,使用DCFH-DA探针检测,DCFH-DA被细胞酯酶水解后可被活性氧氧化为绿色荧光DCF,因此,可以用绿色荧光的强度来表示活性氧的水平。具体步骤如下:将CT26细胞铺在共聚焦显微镜培养皿内,每皿细胞密度为1×105/mL,24h后,吸弃原有培养基,加入分别含有PTSQ/siNC复合物(实施例4制得,PTSQ与siRNA的重量比为50:1)、游离siRNA(Naked siNC)或者Lipo/siNC(将siNC利用转染试剂Lipofectamine 2000进行转染获得的复合物,作为阳性对照)的Opti-MEM培养基,于37℃细胞培养箱内继续培养3h;将DCFH-DA(10mM)探针使用Opti-MEM培养基稀释到10μM作为工作液,吸弃掉培养皿中的所有液体,加入上述稀释好的DCFH-DA工作液,于37℃细胞培养箱内避光孵育30min,30min后使用Opti-MEM培养基洗涤细胞2次,使用808nm激光器照射PTSQ/siNC+L组,光强强度2W/cm2,光照时间4min,将Hoechst 33342染料1:1000稀释后加入培养皿中10min,1×PBS缓冲液洗涤3次;使用共聚焦激光扫描显微镜(CLSM)观察、记录,结果如图9所示,其中,Nuclei为nucleus细胞核图像,Merge为合并后图像。
图8和图9的结果表明PTSQ/siRNA复合物在光照条件下可有效产生具有细胞毒性的活性氧。
测试例4药物组合物的细胞活力和毒性评价
使用MTT方法分析PTSQ/siRNA复合物对CT26细胞的毒性和评价,在光照条件下PTSQ/siRNA复合物对CT26细胞的杀伤能力。MTT是水溶性的化合物,其可被线粒体膜上的琥珀酸脱氢酶还原为不溶性的甲瓒,具体步骤如下:将CT26细胞铺在96孔板上,每孔1×104个细胞,装有细胞的96孔板在37℃,5体积%CO2的培养箱中培养24h,将实施例6制得的不同重量比例的PTSQ/siNC复合物、游离siRNA(Naked siNC)和Lipo/siNC(将siNC利用转染试剂Lipofectamine 2000进行转染获得的复合物,作为阳性对照)用转染培养基(Opti-MEM)稀释到相应的体积(每孔100μL总体积),然后将96孔板中的1640培养液吸弃,换成上述含有PTSQ/siNC、Naked siNC或者Lipo/siNC的Opti-MEM培养基,转染4h后,每个孔补充100μL1640完全培养基,并继续培养20h,使用1640培养基将配置好的MTT母液(5mg/mL)稀释成100μg/mL的工作液,将96孔板中的培养液吸弃,加入上述配置好的工作液,放入培养箱中,4h后,吸弃每个孔中的所有液体,加入100μL DMSO,在37℃孵育10min,使生成的甲瓒充分溶解,使用酶标仪在波长540和650nm处检测吸光度,结果见图10A。
在评价PTSQ/siRNA复合物对CT26细胞的杀伤能力时,在转染完3h后对细胞进行光照处理,光强强度2W/cm2,光照时间4min,其他各步骤与上述MTT方法一致,结果见图10B。图10表明PTSQ/siRNA复合物在光照的条件下对CT26细胞显示出强烈的细胞毒性,且呈剂量依赖关系。
测试例5药物组合物的基因沉默效果评价
评价PTSQ/siRNA复合物介导的细胞PD-L1基因沉默效率,首先参照实施例4的方法,将siRNA采用siPD-L1,以PTSQ与siRNA的重量比为50:1制得PTSQ/siPD-L1复合物,将PTSQ/siPD-L1、游离siRNA(Naked siPD-L1)和Lipo/siPD-L1(将siPD-L1利用转染试剂Lipofectamine 2000进行转染获得的复合物,作为阳性对照)分别按照测试例4中的方法进行CT26细胞系和4T1-Luc细胞培养后,形成Naked siPD-L1组、Lipo/siPD-L1组、PTSQ/siPD-L1组和PTSQ/siPD-L1+L组,提取各组的总RNA,使用Nanodrop测定RNA浓度;使用快速逆转录试剂盒合成cDNA,配制gDNA去除反应体系,模板mRNA 1μg/管,在42℃下加热3min;然后配制逆转录反应体系,加入到gDNA去除反应体系中,在42℃下加热15min,继续在95℃加热3min进行逆转录;qPCR:用SuperReal荧光定量预混试剂盒配制反应体系,设置反应程序为95℃,15min;95℃,3s;60℃,20s;循环40次,进行qPCR反应,以GAPDH为内参,对靶基因PD-L1 mRNA的相对含量进行检测。
通过qRT-PCR检测PTSQ/siPD-L1在CT26细胞系和4T1-Luc细胞系的基因的沉默效果,结果如图11A和图11B所示。相比Mock组来说,PTSQ/siPD-L1能有效抑制/调节PD-L1的mRNA的表达水平。同时,光照射PTSQ/siPD-L1+L组的基因沉默效率优于无光照射PTSQ/siPD-L1组,推测是由于光照射可能通过促进细胞对PTSQ/siPD-L1复合物的摄取和溶酶体逃逸来抑制PD-L1的mRNA的表达,从而发挥双重作用。光照射PTSQ/siPD-L1+L组的PD-L1mRNA表达较Mock对照组下降60%,与脂质阳性Lipo/siPD-L1组相似,这说明在近红外光诱发下,PTSQ/siRNA复合物更容易从溶酶体中逃逸出来,这进一步突出了PTSQ聚合物的优势。
采用Western blot方法分析上述各组转染CT26细胞系24h后PD-L1蛋白含量情况,依次进行蛋白的收集,总蛋白浓度检测,SDS-PAGE及酶联免疫反应来定量蛋白含量,结果如图11C所示。PD-L1蛋白水平明显降低,对这一结果使用Image J软件进行蛋白灰度定量分析,结果如图11D所示,该结果与PD-L1的mRNA表达水平基本一致。
测试例6药物组合物的细胞胞吞及胞吞机制研究
采用测试例4中的方法进行CT26细胞培养,采用Cy5标记的siRNA(Cy5-siRNA)与实施例1制得的PTSQ聚合物按照实施例4所述的方法(PTSQ与siRNA的重量比为50:1)进行复合形成PTSQ/siRNA,并将Cy5-siRNA利用转染试剂Lipofectamine 2000进行转染获得的复合物Lipo/siRNA,作为阳性对照,转染3h后,对PTSQ/siRNA组进行808nm光照/不光照处理,转染4h后,用流式细胞仪检测细胞摄取情况,结果如图12A-12B所示。如图12A所示,从上到下的曲线依次与Mock、Lipo/siRNA、PTSQ/siRNA、PTSQ/siRNA+L对应,PTSQ/siRNA组的平均荧光强度始终低于Lipo 2000阳性对照。但我们发现,Lipo/siRNA组和PTSQ/siRNA复合物组的细胞摄取多,这表明转染效率高。另外,值得注意的是,在近红外光照射下PTSQ/siRNA复合物组的平均荧光强度高于未施加光处理PTSQ/siRNA复合物组,同样,细胞摄取的曲线也稍微上升佐证了这一点,这说明给PTSQ/siRNA复合物处理过的细胞施加近红外光照射能够有效促进细胞对PTSQ/siRNA复合物的摄取。
参照测试例4中的方法进行CT26细胞培养,采用多种抑制剂来阻断细胞内化通路,包括氯丙嗪(Chlorpromazine,阻断网格蛋白介导的内吞作用),阿米洛利(Amiloride,阻断大胞饮作用),染料木素(Genistein,阻断囊泡介导的内吞作用)和脂筏(Lipidraft,抑制脂筏依赖性内吞作用)以期找到PTSQ/siRNA复合物进入细胞的依赖方式,同时将细胞置于4℃下,阻断细胞依赖能量的内化方式,不同内吞抑制剂预孵育的CT26细胞经过PTSQ/siRNA复合物处理4h后,采用共聚焦显微镜和流式细胞术检测Cy5标记的siRNA荧光,结果如图12C-12E所示,共聚焦显微镜数据显示出氯丙嗪、染料木素和脂筏均能有效抑制PTSQ/siRNA复合物的内化,同时,在4℃作用下,细胞内化也受到影响,这说明PTSQ/siRNA复合物进入细胞也需要消耗能量。
测试例7药物组合物的免疫原性细胞死亡研究
参照测试例4中的方法进行CT26细胞培养,然后依据萤光素产生萤光需要ATP间接检测,具体步骤如下:将ATP标准液用检测液稀释到适当的浓度梯度,配置ATP检测工作液,每个样品和标准品各100μL,放置在冰上,向检测板里加入100μL ATP检测工作液,室温下放置5min;取细胞培养皿中的悬液培养基和标准品各20μL,加入到检测孔中进行检测,结果如图13A所示,光照射PTSQ/siRNA复合物组(PTSQ/siNC+L组)中ATP的胞外分泌量是未处理Mock对照组的8.35倍。
参照测试例4中的方法进行CT26细胞培养,对于流式细胞术检测CRT暴露,需要先将细胞消化下来,用4体积%的多聚甲醛固定细胞10min,对于HMGB1,需要使用0.5%的Trito-100,将细胞通透20min,向培养皿中加入含有1%的BSA的1×PBS,在室温下封闭30min,加入含1%BSA的1×PBS稀释的抗体,流式细胞术检测细胞膜表面钙网蛋白(CRT)暴露,将上述加过抗体的细胞放置于室温4h即可,将培养皿放置于4℃冰箱中,过夜孵育;用1×PBS洗涤细胞2-3次,加入CRT和高迁移率族蛋白B1(HMGB1)相关的二抗溶液,室温避光孵育1h,对于流式细胞术检测,可直接当天孵育4h一抗后,直接孵育二抗,进行流式检测;而共聚焦检测需要对细胞核染色,加入DAPI染料5min,使用1×PBS洗涤细胞2-3次,上述操作中,加入二抗后,需进行避光,结果如图13所示。在光照处理下,PTSQ/siRNA复合物组可以观察到明显的CRT表面暴露(绿色荧光),同时,HMGB1从胞内释放到胞外,经过光照射后,加速了HMGB1的释放,这表明在近红外808nm光照射下,PTSQ/siRNA复合物能够引起细胞免疫原性细胞死亡,促进CRT的表面暴露,HMGB1的释放和ATP的分泌。
测试例8药物组合物治疗结直肠癌模型中的研究
在6-8周的雌/雄性BALB/c小鼠的腋下接种小鼠结肠癌细胞CT26约100μL,或者移植3-5mm3的肿瘤块,当肿瘤长至100mm3左右时,将小鼠随机分成5组,G1:PBS组、G2:PTSQ/siNC组、G3:PTSQ/siNC+L组、G4:PTSQ/siPD-L1组、G5:PTSQ/siPD-L1+L组。在第1、4d将各组对应的药物瘤内注射到小鼠体内,剂量siRNA为0.5mg/kg;6h后,使用808nm激光对肿瘤部位照射3min,1W/cm2,期间,每天记录小鼠体重和肿瘤体积,体积计算公式=1/2×长×宽2;当PBS组小鼠的肿瘤体积达到2000mm3时安乐死小鼠,收集小鼠的肿瘤、心、肝、肺、肾、脾和血液,记录小鼠的肿瘤、脾、肝的重量,剩余小鼠留作生存时间观察;取一部分做血细胞分析,将剩余的全血经离心后取上清,做血清生化分析,将上述脏器多聚甲醛固定/OCT胶包埋冷冻后进行苏木精/伊红染色分析,结果如图14所示。
图14B结果表明,相比G1、G2组,G3、G4、G5组表现出不同程度的肿瘤生长抑制,与PTSQ/siNC治疗组(G2)相比,PTSQ/siPD-L1(G4组)下调PD-L1基因显著并抑制了肿瘤的生长。同时,我们可以注意到,808nm光照射组(G3、G5)与其他各组相比,尤其是未施加激光照射组(G2、G4),对肿瘤生长有明显的抑制作用,表明PTSQ对实体瘤也具有显著的光动力治疗作用。因此siPD-L1在小鼠结直肠癌模型体内中起到了抑瘤作用,这突出了光动力结合基因治疗的优势。在治疗期间,如图14C所示,G3-G5任何一组小鼠的体重都没有明显变化,G1和G2组由于肿瘤未得到抑制,小鼠体重稍微有一些上升。如图14D所示,G5组的肿瘤重量和大小明显小于其他组。同时,对肝和脾称重得到肝/体重比和脾/体重比,图14E可以看到肝/体重比无明显变化。如图14G生存曲线所示,发现G5治疗组无小鼠死亡,肿瘤在治疗期间已经全部消除,无复发现象,而其他各组在20d内相继死亡。表明PTSQ/siRNA复合物在结直肠癌中具有良好的治疗效果。
测试例9
在对小鼠治疗时,采用肿瘤光动力方法和免疫检查点抑制方法,取测试例8中治疗后第6d的小鼠肿瘤、脾、引流淋巴结进行免疫分析。具体步骤如下:取治疗第6d小鼠的肿瘤、脾、引流淋巴结,放置于1×PBS缓冲液中,使用剪刀把上述组织剪碎,放置于200目分子筛中研磨,制备单细胞悬液,收集上述各组织的研磨液,300g,离心5min,倒掉上清液,保留底部细胞沉淀,加入1-3mL红细胞裂解液,静置20min;加入两倍裂解液体积的1×PBS,使用吸管混合均匀,300g,离心5min,倒掉上清液,保留细胞沉淀,确保红细胞去除干净,若未去除干净,重复操作;加入3-5mL 1×PBS缓冲液,转移到新的分子筛,收集过滤单细胞悬液,300g,离心5min;加入含有1%BSA的1×PBS 1-3mL,室温下放置在摇床上封闭1h,随后,用各种抗体对细胞进行染色,用于区分免疫细胞群的抗体如下:CD3+、CD4+、CD8-,CD3+、CD4-、CD8+,F4/80+、CD80+、CD206+,CD11c+、CD80+、CD86+,室温避光1h,用1×PBS缓冲液洗涤细胞2-3次,加入300μL 1×PBS缓冲液进行流式细胞仪分析,评估PTSQ/siRNA复合物治疗后肿瘤微环境和免疫器官的免疫状态变化结果如图15所示。
如图15A-15C所示,G3、G4组的CD3+CD4+T细胞显著增多,G4、G5组的CD3+CD8+T细胞也都增多,尤其是G5治疗组的细胞毒性T淋巴细胞浸润最多,这是光动力的ICD效应和siRNA的协同治疗效果。如图15D-15F所示,进一步用流式细胞仪检测了肿瘤细胞中M1、M2型巨噬细胞的百分比和M1/M2巨噬细胞的比例,M1巨噬细胞比例增多。如图15G-15I所示,通过CD11c+CD80+CD86+标记肿瘤、淋巴结和脾的DC细胞,表明光照可显著诱导DC细胞的成熟。综上所述,表明PTSQ/siRNA复合物可以在光照射下诱导CT26荷瘤小鼠的适应性免疫,结合免疫检查点siPD-L1疗法进一步放大机体的免疫调节作用。
测试例10
为评价经过PTSQ/siRNA复合物治疗后小鼠的恢复正常水平,在参照测试例8中的方法治疗的第11d后,采集荷瘤小鼠的全血进行血液分析,评价PTSQ/siRNA复合物体内安全系数,结果如图16所示。如图16A-16H所示,包括WBC(白细胞计数)、RBC(红细胞计数)、Lym(淋巴细胞比例)、HCT(红细胞压积)、MCV(平均红细胞体积)、MCH(平均红细胞血红蛋白含量)、MCHC(平均红细胞血红蛋白浓度)、RDW-CV(红细胞分布宽度变异系数)等参数。图中灰色区域为Balb/c健康小鼠8周的全血分析标准生理生化数据,数据来源于斯贝福(北京)生物技术有限公司官网,可以从图中看到,小鼠的WBC、RBC、Lym、HCT数值受肿瘤的影响较大,但是,明显可以看到,治疗组的各项指标更接近/处于正常范围。如图16I-16L所示,包括ALT(谷丙转氨酶)、AST(谷草转氨酶)、CK-MB(肌酸激酶)、UREA(尿素氨)等参数,同样可以从图中看到,治疗组的各项指标更接近/处于正常范围,表明经过PTSQ/siRNA复合物治疗有助于小鼠恢复健康,这也从侧面表明治疗组小鼠的肿瘤得到控制。
测试例11
为评价经过PTSQ/siRNA复合物治疗后小鼠组织脏器的安全水平,在参照测试例8中的方法治疗的第11d后,采集荷瘤小鼠的心、肝、脾、肺、肾组织做H&E染色分析,PTSQ/siRNA复合物体内抗肿瘤安全系数评价结果如图17所示。从图17可以看出,小鼠的各项组织脏器未见异常。
测试例12
参照测试例8中的方法,进行PTSQ/siRNA复合物在治疗乳腺癌模型中的研究,乳腺癌肿瘤细胞为小鼠乳腺癌细胞4T1,实验分为5组,G1:PBS组、G2:PTSQ/siNC组、G3:PTSQ/siNC+L组、G4:PTSQ/siPD-L1组、G5:PTSQ/siPD-L1+L组,结果如图18所示。
如图18B和18D所示,相比G1生理盐水组,G2、G3、G4、G5组都有不同程度的肿瘤抑制现象。其中相比较G2、G4组,G3和G5组显示出良好的肿瘤抑制效果,这表明光动力在肿瘤治疗方面具有良好的治疗效果。G4和G2组相比,显示出siPD-L1的治疗效果。对于G3和G5组,在两次给药后,均明显抑制了肿瘤的生长。治疗后第12d,进一步证实PTSQ/siRNA复合物治疗对肿瘤生长的抑制作用。治疗过程中,荷瘤小鼠的体重变化如图15C所示,各小组小鼠体重无明显变化。如图18E,治疗过程中各组小鼠肝/体重指数均无显著差异。但如图18F所示,令人惊讶的是,G1、G2和G4组小鼠的脾显著增大,而G3和G5组没有出现这种现象,猜测这可能与肿瘤及肿瘤的转移有关。
测试例13
参照实施例4的方法,将siRNA采用siRRM2,以PTSQ与siRNA的重量比为50:1制得PTSQ/siRRM2复合物。参照测试例8中的方法,进行PTSQ/siRNA复合物在治疗PDX患者来源肝癌模型中的研究,肝癌肿瘤细胞为患者来源的异种移植肝癌细胞,实验分为5组:G1:PBS,G2:PTSQ/siNC,G3:PTSQ/siNC+L,G4:PTSQ/siRRM2,G5:PTSQ/siRRM2+L,结果如图19所示。
从肿瘤生长曲线图19B可知,肝脏PDX肿瘤恶性程度较高,对照组G1肿瘤生长速度最快,G4组肿瘤大小明显受到抑制,这表明肝脏PDX肿瘤对基因干扰治疗方法比较敏感。光照射G3和G5组肿瘤生长受到强烈抑制,在第二次治疗后,G5组肿瘤已经有完全抑制的趋势。与CT26结直肠癌模型相似,G3组肿瘤在第二次治疗停止后有复发的趋势,并于第5d开始重新生长。图19C表明,在治疗期间,各小组小鼠体重无明显变化。如图19D所示,切除Balb/C裸鼠肿瘤,并称重记录。可以看到,G3和G5治疗组肿瘤重量和大小明显小于其他组,尤其是G5治疗组,这与肿瘤体积生长曲线数据图19B一致。在治疗的第6d,各组分别处死3只小鼠,并取小鼠相关组织和血液进行后续分析,其血生化指标如图19F所示各组均无显著差异,剩余6只小鼠持续监测健康状况,其生存曲线如图19E所示。这表明该PTSQ/siRNA复合物可有效抑制肿瘤生长,促进癌细胞凋亡,并延长小鼠生存周期。
以上详细描述了本发明的优选实施方式,但是,本发明并不限于此。在本发明的技术构思范围内,可以对本发明的技术方案进行多种简单变型,包括各个技术特征以任何其它的合适方式进行组合,这些简单变型和组合同样应当视为本发明所公开的内容,均属于本发明的保护范围。
Claims (10)
3.根据权利要求2所述的制备方法,其特征在于,步骤(1)中所述式(II)所示的化合物、式(III)所示的化合物、L-赖氨酸二异氰酸酯、2,2-双(溴甲基)-1,3-丙二醇与甲氧基聚乙二醇-羟基的重量比为1:0.8-1.2:2-2.8:0.9-1.3:5-6;
优选地,所述甲氧基聚乙二醇-羟基的分子量为2000-5000;
优选地,所述反应I的条件包括:温度为45-55℃,时间为10-15h;
优选地,所述反应II的条件包括:温度为45-55℃,时间为10-15h。
4.根据权利要求2或3所述的制备方法,其特征在于,步骤(2)中式(IV)所示的反应物P1与式(V)所示的化合物的重量比为2.5-3.5:1;
优选地,所述反应III的条件包括:温度为45-55℃,时间为10-15h。
5.根据权利要求2或3所述的制备方法,其特征在于,所述反应溶剂I和所述反应溶剂II分别独立地选自无水N,N-二甲基甲酰胺、无水二氯甲烷、无水四氢呋喃和无水二甲基亚砜中的至少一种;
优选地,所述步骤(1)还包括:将所述反应II得到的反应液进行透析I得到透析液I,将所述透析液I进行干燥得到所述式(IV)所示的反应物P1;
优选地,所述步骤(2)还包括:将所述反应III得到的反应液进行透析II得到透析液II,将所述透析液II进行干燥得到所述式(I)所示的聚合物。
6.一种药物组合物,其特征在于,该药物组合物含有siRNA以及根据权利要求1所述的聚合物核酸递送载体和/或根据权利要求2至5中任意一项所述的制备方法制得的聚合物核酸递送载体。
7.根据权利要求6所述的药物组合物,其特征在于,所述聚合物核酸递送载体与所述siRNA的重量比为5-50:1;
优选地,所述siRNA含有完全反向互补的正义链和反义链,所述正义链和所述反义链的核苷酸序列分别如SEQ ID NO.1和SEQ ID NO.2所示。
8.一种制备药物组合物的方法,其特征在于,包括以下步骤:将根据权利要求1所述的聚合物核酸递送载体和/或根据权利要求2至5中任意一项所述的制备方法制得的聚合物核酸递送载体与胶束剂进行混合反应I得到聚合物胶束,将所述聚合物胶束与siRNA进行混合反应II。
9.根据权利要求8所述的方法,其特征在于,所述聚合物核酸递送载体与所述siRNA的重量比为5-50:1;
优选地,所述siRNA含有完全反向互补的正义链和反义链,所述正义链和所述反义链的核苷酸序列分别如SEQ ID NO.1和SEQ ID NO.2所示;
优选地,所述胶束剂为超纯水;相对于1mg所述聚合物核酸递送载体,所述胶束剂的用量为0.1-0.3mL;
优选地,所述混合反应I的过程包括:将所述聚合物核酸递送载体溶于溶剂后,滴入所述胶束剂中,再在温度为5-40℃、转速为80-150rpm的条件下搅拌25-35min得到混合反应液,将所述混合反应液进行透析III得到聚合物胶束;
优选地,所述混合反应II的条件包括:温度为5-40℃,时间为25-35min。
10.根据权利要求1所述的聚合物核酸递送载体、根据权利要求2至5中任意一项所述的制备方法制得的聚合物核酸递送载体、根据权利要求6或7所述的药物组合物和根据权利要求8或9所述的方法制得的药物组合物中的至少一者在制备抗肿瘤药物中的应用;
优选地,所述肿瘤的类型选自结直肠癌、乳腺癌和肝癌中的至少一种。
