Abstract

现有技术公开了一些治疗角膜新生血管的策略,例如CN114931637A公开了抗PDL1抗体在用于治疗角膜新生血管性疾病中的应用,通过局部应用抗PDL1抗体后进行裂隙灯检查评估角膜新生血管、角膜血管组织免疫荧光染色等实验发现,抗PDL1抗体治疗组小鼠的角膜新生血管明显减少,说明局部应用抗PDL1抗体可抑制角膜新生血管。证实了PDL1抗体可显著减少中性粒细胞亚群的浸润,抑制其分泌的一系列炎症因子和血管生长因子,从而发挥抑制病理性新生血管的作用。例如CN114917230A提供了CB-839在制备抑制角膜新生血管生成的药物中的应用,其发现负载CB-839的药物能够明显抑制小鼠角膜新生血管生成,炎症细胞浸润减少,并且没有发现明显副作用。
针对角膜新生血管的治疗,究其根本,还是抑制新生血管的生成,具体来说,就是通过抑制新生血管形成的信号通路的激活来实现对内皮细胞迁移、新生血管生成的抑制。新生血管形成的信号通路包括常见的血管内皮生长因子(Vascular endothelial growthfactor,VEGF)与其受体,如血管内皮生长因子受体(VEGFR)、神经菌毛素受体(neuropilin-1,NRP-1)之间的信号通路,也有其它细胞外基质蛋白或膜上受体参与。其中,整合素αVβ3受体对新生血管生成具有较大的贡献。
由于存在病灶,导致巨噬细胞等免疫细胞到达该部位,可能会导致炎症反应。适当的炎症反应有助于实现对病灶的清除,但是过量的炎症反应可能不利于疾病的治愈,因此,需要在一定程度抑制炎症反应。整合素αVβ3受体介导的PI3 kinase/Akt/NF-κB信号通路可以调控炎症反应,受到了研究人员的广泛关注。
纳米尺寸颗粒会增加在体内的循环时间;其也可以被动靶向作用富集到肿瘤等病灶部位。依靠多肽分子间的非共价键作用(氢键、疏水、π-π等相互作用),可通过对外源性条件包括时间、温度,或内源性条件包括酶、pH及配体-受体相互作用等的调控,使多肽分子发生结构上的变化。例如具有β-sheet结构的纳米纤维相比于纳米颗粒,可以通过多价键效应实现更强的受体结合效果和结合时间,更长效地阻止整合素αVβ3受体的下游信号通路。
因此,开发一种可以在角膜新生血管处实现靶向并原位转化形成纳米纤维的多肽材料,长效抑制整合素αVβ3受体介导的新生血管生成以及降低病灶部位的炎症反应,具有广泛的应用研究意义。
发明内容
针对现有技术的不足,本发明的目的在于提供一种多肽纳米材料及其制备方法和应用,具体提供一种靶向治疗角膜新生血管的多肽纳米材料及其制备方法和应用,尤其提供一种靶向治疗角膜新生血管且降低病灶部位炎症反应的多肽纳米材料及其制备方法和应用。该多肽纳米材料具有角膜新生血管内皮细胞的靶向性,同时兼具与角膜新生血管内皮细胞上受体蛋白之间的配体-受体相互作用(Receptor-ligand interaction),能够在内皮细胞表面由纳米颗粒转化生成纳米纤维。
为达到此发明目的,本发明采用以下技术方案:
第一方面,本发明提供一种靶向治疗角膜新生血管的多肽纳米材料,所述多肽纳米材料包括依次连接的疏水单元、组装单元、亲水单元和靶向单元;所述靶向单元包括靶向新生血管内皮细胞且与整合素受体特异性结合的多肽序列。
本发明所涉及的多肽纳米材料包括四部分功能结构,分别为疏水单元、组装单元、亲水单元和靶向单元,四者相辅相成、共同配合、发挥药效。其中,靶向单元不仅具有角膜新生血管内皮细胞的靶向性,同时与角膜新生血管内皮细胞上的整合素受体蛋白具有配体-受体相互作用,即双方特异性基团的具有互补性且空间结构适配,通过此相互作用,受体蛋白诱导纳米材料的形貌发生转化,形成纳米纤维。纳米纤维覆盖在内皮细胞表面,通过长效结合整合素受体抑制内皮细胞的迁移,同时负调控炎症下游信号通路,最终实现对角膜新生血管的治疗并降低病灶部位的炎症反应。该多肽纳米材料为角膜新生血管的治疗及降低病灶部位的炎症反应提供了新的靶向治疗策略,具有广泛的应用前景。

疏水单元能够帮助所述多肽纳米材料在从有机相到水相的分散过程中利用疏水相互作用自组装形成聚集体纳米颗粒。

上述特定结构的多肽纳米材料具有角膜新生血管内皮细胞的靶向性,同时兼具与角膜新生血管内皮细胞上的整合素αVβ3受体之间的配体-受体相互作用,通过此相互作用,多肽纳米材料具有从纳米颗粒转化生成纳米纤维的能力,并以纳米纤维形式覆盖在内皮细胞表面。相较于纳米颗粒,纳米纤维具有更好的稳定性,在生物体内具有较长的滞留时间;纳米纤维占据整合素αVβ3受体,抑制了整合素下游信号通路的传导,因而有效地抑制了内皮细胞的迁移,进而抑制了新生血管的生成并降低了病灶部位的炎症反应。
第二方面,本发明提供一种根据第一方面所述的靶向治疗角膜新生血管的多肽纳米材料的制备方法,所述制备方法包括:
以树脂为载体,以疏水单元、组装单元、亲水单元和靶向单元的组成氨基酸为原料,利用固相合成方法制备得到所述多肽纳米材料。
本发明所涉及的多肽纳米材料可以采用本领域常规的技术手段进行制备,其中,最常用的方法即为固相合成法。以Wang树脂为载体材料,按照多肽的氨基酸连接顺序,通过偶联剂进行偶联,其中有机化合物也同样按照氨基酸的偶联方法进行反应,最后经三氟乙酸裂解、氮气吹干和乙醚纯化等步骤制备即可得到。
第三方面,本发明提供根据第一方面所述的靶向治疗角膜新生血管的多肽纳米材料在制备抑制角膜新生血管生成及降低病灶部位炎症的药物中的应用。
第四方面,本发明提供第一方面所述的靶向治疗角膜新生血管的多肽纳米材料组装形成纳米纤维的方法,所述方法包括:
将溶于有机溶剂的多肽纳米材料与水相溶剂混合,使多肽纳米材料自组装成纳米颗粒;然后与钙离子和/或整合素αVβ3受体混合,组装形成纳米纤维。
本发明所涉及的多肽纳米材料在PBS缓冲溶液中能够自组装形成纳米颗粒,在整合素αVβ3蛋白的作用下,诱导转化生成纳米纤维。
优选地,所述有机溶剂包括二甲基亚砜、N,N-二甲基甲酰胺或1,4-二氧六环中的任意一种或至少两种的组合。
优选地,所述多肽纳米材料在有机溶剂中的浓度为10-4~10-2M,例如10-4M、5×10- 3M、10-3M、5×10-2M、10-2M等。
优选地,所述水相溶剂包括PBS缓冲液。
优选地,所述与钙离子和/或整合素αVβ3受体混合在超声条件下进行,超声的频率为20-60kHz,例如20kHz、30kHz、40kHz、50kHz、60kHz等;超声的时间为0-8min,例如1min、2min、3min、4min、5min、6min、8min等。
优选地,所述超声后还包括静置的操作。
优选地,所述静置为在20~30℃(例如可以是22℃、25℃、28℃、30℃、32℃、35℃或38℃等)下保持0~1h(例如可以是0.2h、0.5h或1h)。

相对于现有技术,本发明具有以下有益效果:
本发明所涉及的多肽纳米材料包括四部分功能结构,分别为疏水单元、组装单元、亲水单元和靶向单元,四者相辅相成、共同配合、发挥药效。其中,靶向单元不仅具有角膜新生血管内皮细胞的靶向性,同时与角膜新生血管内皮细胞上的整合素受体蛋白具有配体-受体相互作用,即双方特异性基团的具有互补性且空间结构适配,通过此相互作用,受体蛋白诱导纳米材料的形貌发生转化,形成纳米纤维。纳米纤维覆盖在内皮细胞表面,通过长效结合整合素受体抑制内皮细胞的迁移,同时负调控炎症下游信号通路,最终实现对角膜新生血管的治疗并降低病灶部位的炎症反应。该多肽纳米材料为角膜新生血管的治疗及降低病灶部位的炎症反应提供了新的靶向治疗策略,具有广泛的应用前景。本发明所涉及的多肽纳米材料可以采用本领域常规的技术手段进行制备,制备工艺简单易操作,适合于工业化生产,具有实用性。

下面通过具体实施方式来进一步说明本发明的技术方案。本领域技术人员应该明了,所述实施例仅仅是帮助理解本发明,不应视为对本发明的具体限制。
以下实施例中,未注明具体技术或条件者,按照本领域内的文献所描述的技术或条件,或者按照产品说明书进行;所用试剂或仪器未注明生产厂商者,均为可通过正规渠道获得的常规产品。
制备例
本制备例提供一种靶向治疗角膜新生血管的多肽纳米材料,该多肽纳米材料由疏水单元、组装单元、亲水单元和靶向单元组成,其中疏水单元的的供体为有机化合物棕榈酸,组装单元的供体为KLVFF,亲水单元的供体为GSG,靶向单元的供体为靶向多肽HSDVHK。其分子结构如下所示:
该多肽纳米材料采用本领域常规的固相合成方法来合成,以Wang树脂为载体,用N,N-二甲基甲酰胺对其进行溶胀,按照多肽顺序,通过偶联剂(氮甲基吗啉:DMF=5:95,体积比)进行偶联,其中棕榈酸也同样按照氨基酸的偶联方法进行反应,使用水合肼脱除K氨基酸侧链的Dde保护基并继续进行合成,最后经三氟乙酸裂解、氮气吹干、乙醚纯化而得。
对制得的多肽纳米材料进行质谱表征,结果如图1所示,由该图可知,图中所出现的离子峰满足材料分子的分子量的特征峰,表明上式结构的多肽纳米材料被成功合成。
实施例1
本实施例探究多肽纳米材料的蛋白诱导形变组装过程,具体如下:
称取制备例制得的多肽纳米材料7.0mg溶于1mL DMSO溶剂中,再将此溶液用注射器快速注射到49mL的超纯水中,表征多肽纳米材料在超纯水中的透射电镜图,如图2所示,材料在水中自组装形成纳米球状颗粒。
然后滴加整合素αVβ3蛋白1mL,浓度为80μM,超声5分钟,静置2小时,使用透射电镜对所得溶液进行观察,如图3所示,材料发生形貌转换,呈现纳米纤维状。
实施例2
本实施例探究多肽纳米材料在内皮细胞表面的滞留和转化情况,具体如下:
(1)准备人脐静脉内皮细胞(HUVECs)悬浮液,每个共聚焦培养皿1mL,培养过夜;取出培养基,加入含有摩尔浓度为80μM的Cy3标记的多肽纳米材料的培养基1mL,培养4小时,用PBS清洗3遍,进行共聚焦成像实验,采用550nm激光通道,收集红光波段(570nm);所得单光子激光共聚焦照片如图4所示。
(2)准备人脐静脉内皮细胞(HUVECs)细胞悬浮液于12孔板中,每个孔1.5mL,加入表面用等离子体处理的硅板,培养过夜;取出培养基,加入含有摩尔浓度为80μM的多肽纳米材料的培养基1.5mL,培养4小时,用PBS清洗3遍,再使用质量分数为4%的多聚甲醛溶液(即多聚甲醛与PBS缓冲液的体积比为1:24)固化1h,再依次用PBS稀释好的10%、30%、50%、70%、90%、100%的乙醇溶液脱水处理,每个浓度脱水两次,每次10分钟;然后,用叔丁醇溶液置换出水和乙醇,置换一次,每次10分钟,喷金,进行扫描电镜实验。
图5为制备例制得的多肽纳米材料在内皮细胞表面发生原位形貌转化的扫描电镜照片,图6为图5中黑色框线所选位置的放大图。由图可知,多肽纳米材料能够在内皮细胞表面滞留,且发生原位转化后生成纳米纤维。
实施例3
本实施例探究多肽纳米材料对内皮细胞迁移抑制的情况,具体如下:
准备人脐静脉内皮细胞(HUVECs)细胞悬浮液于6孔板中,每个孔3mL,过夜培养;使用移液枪枪头在孔板中部自上而下划出伤口,再用PBS洗去没有贴壁的细胞,加入多肽纳米材料浓度为160μM的培养基3mL,培养24小时;同时设置低浓度多肽纳米材料浓度组(80μM)和PBS组。PBS组中不添加多肽纳米材料,即空白组。最后用PBS清洗3遍,质量浓度为4%的多聚甲醛溶液(即多聚甲醛与PBS缓冲液的体积比为1:24)固化2h,再用PBS清洗3遍,进行伤口愈合实验拍摄。
如图7为拍摄图,其中a为高浓度实验组,b为低浓度实验组,c为PBS组。图8为各组中伤口范围大小对比统计图,由图可知,本发明所涉及的多肽纳米材料可以有效的抑制内皮细胞的迁移,且高浓度抑制效果相比于低浓度多肽纳米材料和PBS组具有显著性差异。
实施例4
本实施例探究多肽纳米材料在小动物角膜区域的滞留效果,具体如下:新西兰白兔(雄性,体重在2-3kg之间)角膜区域使用浸润了NaOH溶液(1M)的滤纸处理30秒,之后使用生理盐水冲洗,开始用于材料滞留效果的验证。
将所有兔随机的分为2组,每组4只,通过结膜下注射实验材料,注射频次为仅第一天一次;其中,实验组注射100μL、含有摩尔浓度为167.9μM的多肽纳米材料的PBS溶液(同时,保证DMSO体积分数不超过2.5%);同时设置PBS组,PBS组中不添加多肽纳米材料,即空白组。距离第一次注射7天后将兔安乐死,取其眼部组织进行固定并切片,对切片进行生物TEM切片。
结果如图9所示,由图可知,实验组中(a)存在纤维状结构,证明多肽纳米材料在该区域以纳米纤维形式存在。PBS组中(b)并没有纤维存在。
实施例5
本实施例探究多肽纳米材料对角膜新生血管的治疗效果,具体如下:
新西兰白兔(雄性,体重在2-3kg之间)角膜区域使用浸润了NaOH溶液(1M)的滤纸处理30秒,之后使用生理盐水冲洗,用于建立角膜新生血管兔模型。造模成功后开始用于材料对角膜新生血管的治疗效果的验证。
将所有兔随机的分为4组,每组4只,通过玻璃体内注射多肽纳米材料,注射频次为仅第一天一次。高浓度实验组注射100μL,浓度为167.9μM的多肽纳米材料的PBS溶液(保证DMSO体积分数不超过2.5%)。低浓度实验组注射100μL,浓度为83.9μM的多肽纳米材料的PBS溶液(保证DMSO体积分数不超过2.5%)。对照组注射100μL,浓度为167.9μM的单克隆抗体贝伐单抗(上海芮晖化工,R190605)。PBS组注射100μL的PBS溶液。
距离第一次注射14天后对角膜新生血管区域进行拍照,如图10所示,由图可知,高浓度实验组(a)的兔角膜新生血管区域面积明显小于低浓度对照组(b)、单抗对照组(c)和PBS组(d),新生血管面积统计图如图11所示,由图可知这种面积的差异具有显著性。
之后将兔安乐死,取其眼部组织进行固定并切片,对切片进行CD31免疫荧光测试,如图12所示,由图可知,高浓度实验组(a)的兔角膜新生血管数量明显小于低浓度实验组(b)、单抗对照组(c)和PBS组(d),新生血管数量统计图如图13所示,由图可知,各组新生血管数量的差异具有显著性,证明了本发明所涉及的多肽纳米材料具有抑制角膜新生血管的效果。
实施例6
本实施例探究多肽纳米材料对降低炎症反应的治疗效果,具体如下:
在本实施例中考察实施例1制备得到的用于治疗角膜新生血管的多肽纳米材料对角膜新生血管的治疗效果,方法如下:
新西兰白兔(雄性,体重在2-3kg之间)角膜区域使用浸润了NaOH溶液(1M)的滤纸处理30秒,之后使用生理盐水冲洗,用于建立角膜新生血管兔模型。造模成功后开始用于材料对降低炎症反应的治疗效果的验证。
将所有兔随机的分为4组,每组4只,通过玻璃体内注射多肽纳米材料,注射频次为仅第一天一次。高浓度实验组注射100μL,浓度为167.9μM的多肽纳米材料的PBS溶液(保证DMSO体积分数不超过2.5%)。低浓度实验组注射100μL,浓度为83.9μM的多肽纳米材料的PBS溶液(保证DMSO体积分数不超过2.5%)。对照组注射100μL,浓度为167.9μM的单克隆抗体贝伐单抗(上海芮晖化工,R190605)。PBS组注射100μL的PBS溶液。
距离第一次注射14天后将兔安乐死,取其眼部组织进行固定并切片,对切片进行F4/80阳性巨噬细胞观察和IL-1β免疫荧光观察。F4/80阳性巨噬细胞观察图如图14所示,高浓度实验组(a)的F4/80阳性巨噬细胞数量明显小于低浓度对照组(b)、单抗对照组(c)和PBS组(d),各组F4/80阳性巨噬细胞数量统计图如图15所示,由图可知,这种数量的差异具有显著性。
IL-1β免疫荧光观察图如图16所示,高浓度实验组(a)的多肽纳米材料的病灶部位IL-1β表达量明显小于低浓度实验组(b)、单抗对照组(c)和PBS组(d),且各组IL-1β表达量统计图如图17所示,由图可知,各组IL-1β表达量具有显著性,表明本发明所涉及的多肽纳米材料具有抑制炎症反应的能力。
申请人声明,本发明通过上述实施例来说明本发明的一种靶向治疗角膜新生血管的多肽纳米材料及其制备方法和应用,但本发明并不局限于上述实施例,即不意味着本发明必须依赖上述实施例才能实施。所属技术领域的技术人员应该明了,对本发明的任何改进,对本发明产品各原料的等效替换及辅助成分的添加、具体方式的选择等,均落在本发明的保护范围和公开范围之内。

1.一种靶向治疗角膜新生血管的多肽纳米材料,其特征在于,所述多肽纳米材料包括依次连接的疏水单元、组装单元、亲水单元和靶向单元;所述靶向单元包括靶向新生血管内皮细胞且与整合素受体特异性结合的多肽序列。
2.根据权利要求1所述的靶向治疗角膜新生血管的多肽纳米材料,其特征在于,所述靶向单元为靶向新生血管内皮细胞且与整合素αVβ3受体特异性结合的多肽序列;
优选地,所述靶向单元来自于多肽序列HSDVHK。
3.根据权利要求1或2所述的靶向治疗角膜新生血管的多肽纳米材料,其特征在于,所述亲水单元来自于多肽序列GSG。
4.根据权利要求1-3中任一项所述的靶向治疗角膜新生血管的多肽纳米材料,其特征在于,所述组装单元来自于多肽序列KLVFF。
5.根据权利要求1-4中任一项所述的靶向治疗角膜新生血管的多肽纳米材料,其特征在于,所述疏水单元来自于C10-C20烷基羧酸;
优选地,所述疏水单元来自于月桂酸和/或棕榈酸。
7.根据权利要求1-6中任一项所述的靶向治疗角膜新生血管的多肽纳米材料的制备方法,其特征在于,所述制备方法包括:

9.权利要求1-6中任一项所述的靶向治疗角膜新生血管的多肽纳米材料组装形成纳米纤维的方法,其特征在于,所述方法包括:
将溶于有机溶剂的多肽纳米材料与水相溶剂混合,使多肽纳米材料自组装成纳米颗粒;然后与钙离子和/或整合素αVβ3受体混合,组装形成纳米纤维。
10.根据权利要求9所述的方法,其特征在于,所述有机溶剂包括二甲基亚砜、N,N-二甲基甲酰胺或1,4-二氧六环中的任意一种或至少两种的组合;
优选地,所述多肽纳米材料在有机溶剂中的浓度为10-4~10-2M;
优选地,所述水相溶剂包括PBS缓冲液;
优选地,所述与钙离子和/或整合素αVβ3受体混合在超声条件下进行,超声的频率为20-60kHz,超声的时间为0-8min。
