本发明提供了一种具有高皮肤穿透力的复方精油及其制备方法与应用,所述具有高皮肤穿透力的复方精油包括以下重量份的原料:人参6‑12份、狗脊8‑12份、细辛6‑10份、威灵仙6‑12份、乳香6‑12份、马钱子6‑10份、土虫5‑10份、白芍15‑25份、女贞子6‑10份、甘草5‑10份、渗透液20‑40份、促渗助剂5‑15份、杏仁30‑40份、仙人掌25‑35份和去离子水120‑180份。该具有高皮肤穿透力的复方精油穿透力强、疗效好、长期使用效果不衰减,无毒副作用,具有良好的应用前景。

技术领域
本发明涉及中医药技术领域,具体涉及一种具有高皮肤穿透力的复方精油及其制备方法与应用。
背景技术
植物精油在日常生活中常被称作“精油”,在其他领域也被称作芳香油、挥发性成分或挥发油,来源于芳香植物,常温常压状态下为油状液体,具有挥发性,为植物的次生代谢物,属于小分子化合物,挥发性较强,可蒸馏而出,多存在于植物果实、叶片、花和根中,对植物的芳香气味起到决定性作用。植物精油含有上百种成分,其中主要包括萜类、脂肪族类、芳香族类等化合物和少量含氮含硫化合物,在其中某一单独成分或某些成分的协同作用下,植物精油具有不同的功能,如湿度调节、疾病预防与治疗、抑菌杀菌、缓解情绪调节心理、消炎镇痛、保护植物免受病害。
精油在中医疗法中具有广泛的应用,中医的理论学说主要有阴阳五行学说、藏象学说、气血精液学说、经络学说等。经络学说是中医中研究人体经络的生理功能、病理变化及其与脏腑相互关系的学说。早在两千多年以前的中国,人们就已经知道人体皮肤上有着许多特殊的感觉点,《黄帝内经》指出:“气穴所发,各有处名”,并记载了160个穴位名称。
利用植物的精油来治疗人体的身、心、灵是辅助医学治疗的一种另类疗法,称为芳香疗法,芳香疗法期初在英国广为流传,目前所流行的芳疗信息均来源于英伦世界,虽然使用精油过程中的观点、方法和程序受到其影响,但是在世界范围内,不同国家、不同种族的文化对芳香疗法都独具匠心。
芳香疗法一般都是外用吸收,很少使用内服的方法,因为纯精油容易被胃酸所破坏,失去芳香疗法的效能。一般芳香疗法都是通过以下三种途径进入人体:1) 熏香吸收,2)按摩吸收,3) 沐浴吸收,而在现代美容领域,很多人将香薰按摩与中医的经络学说及穴位结合运用,不仅可用于面部护肤,也可用于全身,通过刺激我们人体的穴位,可缓解压力,放松神经,改善气滞血瘀,调节五脏六腑,使我们的身、心、灵得到放松。
多数精油配方在使用中是通过刺激神经达到疗效,初期效果良好,但反复刺激后,效果就会大减,疗效有限,原因在于这些精油大多难以穿透皮肤,直接作用于筋膜层穴位。因此,有必要提供一种皮肤穿透力强、疗效好、长期使用效果不衰减的复方精油。
发明内容
针对现有技术中存在的问题,本发明的目的在于提供一种皮肤穿透力强、疗效好、长期使用效果不衰减的复方精油。
为达此目的,本发明的技术方案如下:
本发明提供了一种具有高皮肤穿透力的复方精油,所述具有高皮肤穿透力的复方精油包括以下重量份的原料:人参6-12份、狗脊8-12份、细辛6-10份、威灵仙6-12份、乳香6-12份、马钱子6-10份、土虫5-10份、白芍15-25份、女贞子6-10份、甘草5-10份、渗透液20-40份、促渗助剂5-15份、杏仁30-40份、仙人掌25-35份和去离子水120-180份。
优选地,所述具有高皮肤穿透力的复方精油包括以下重量份的原料:人参10份、狗脊10份、细辛8份、威灵仙10份、乳香10份、马钱子8份、土虫7份、白芍20份、女贞子8份、甘草7份、渗透液30份、促渗助剂10份、杏仁35份、仙人掌30份和去离子水160份。
在本发明的一些实施方案中,所述渗透液包括琥珀提取物和天然冰片,所述琥珀提取物与天然冰片的质量比为3-5:1。
在本发明的一些实施方案中,所述琥珀提取物的制备方法包括以下步骤:
(1)将附子、薰衣草、川芎加入低共熔试剂中,加热,保持恒温,搅拌0.5-1h;
(2)将琥珀用水洗净、沥干,粉碎至10-20目,然后加入步骤(1)所得的液体中,升温至45-55℃,浸泡48-72h,用100-200目滤布过滤,再用孔径为0.1-0.15m的微滤氧化铝陶瓷膜过滤,收集液体,即得琥珀提取物。
在本发明琥珀提取物的制备方法中,步骤(1)中所述低共熔溶剂由氯化胆碱和1,2-丙二醇组成,氯化胆碱和1,2-丙二醇的质量比为1:2-4;所述附子、薰衣草、川芎的质量比为2-3:1-3:1;所述低共熔试剂的质量为附子、薰衣草、川芎总质量的5-10倍;所述加热温度为30-40℃。
在本发明琥珀提取物的制备方法中,步骤(2)中所述琥珀选自缅甸琥珀或抚顺琥珀,所述琥珀的质量为低共熔试剂的10-20%;所述微滤氧化铝陶瓷膜的操作压力为0.1-0.8Mpa,操作温度为25-35℃。
在本发明的一些实施方案中,所述促渗助剂由改性淀粉和香兰基丁基醚按质量比为1-2:1混合而成的混合物。
在本发明的一些实施方案中,所述改性淀粉的制备方法包括以下步骤:
(1)取淀粉加入水中,调节pH为8-9,搅拌均匀;
(2)在步骤(1)的溶液中,加入辛烯基琥珀酸酐和甘油,混匀后在120-130℃下熔融共混25-35min,冷却后即改性淀粉。
在本发明改性淀粉的制备方法中,步骤(2)中所述辛烯基琥珀酸酐的质量为淀粉的5-10%;所述甘油的质量为辛烯基琥珀酸酐的1-3倍。
一种具有高皮肤穿透力的复方精油的制备方法,所述具有高皮肤穿透力的复方精油的制备方法包括以下步骤:
(1)将人参、狗脊、细辛、威灵仙、乳香、马钱子、土虫、白芍、女贞子、甘草、杏仁、仙人掌加入去离子水中,浸泡48-72h;
(2)在步骤(1)得到的溶液中加入渗透液、促渗助剂,混匀,即得具有高皮肤穿透力的复方精油。
在本发明的一些实施方案中,所述具有高皮肤穿透力的复方精油在人体保健中的应用方法为:1)将1-1.5ml具有高皮肤穿透力的复方精油涂抹在脊柱部位,用刮板由下至上重复刮3-5min;2)将1.5-2ml具有高皮肤穿透力的复方精油涂抹在脊柱两侧的距离脊柱2-3cm的部位,由上至下用刮板重复刮10-15min,5-7次为一个疗程,3-4个疗程隔日一次,可长期使用。
脊柱是身体的支柱,位于背部正中,上端接颅骨,下端达尾骨尖,将精油用在人体的脊柱部位,能有效疏通全身的血脉,改善血液循环,使用后可立即改善精神状态,脊柱部位由下至上刮,而脊柱两侧的部位由上至下刮,顺应了人体的气血、能量循环方向,使精油的效果能良好的发挥出来。
与现有技术相比,本发明具有以下有益效果:
(1)本发明提供的具有高皮肤穿透力的复方精油,通过添加渗透液,使用自制的低共熔试剂将附子、薰衣草、川芎与琥珀共同进行提取、酿造,相较传统的萃取方式,以更低的成本得到琥珀提取物,同时搭配天然冰片发挥出较强的穿透性和溶解性,有效增强精油的皮肤穿透力,使精油中的有益成分顺利地透过皮肤、深入身体经络,修复老化的筋膜层,增加轻松愉悦感,长期使用效果不会衰减且无毒副作用。
(2)本发明提供的具有高皮肤穿透力的复方精油,通过使用辛烯基琥珀酸酐和甘油对淀粉进行改性,使其具有良好的稳定性和皮肤穿透力,搭配香兰基丁基醚制成促渗助剂能加大天然冰片的溶解量,与渗透液协同增强精油的皮肤穿透力,使其具有良好的渗透效果。
(3)本发明提供的具有高皮肤穿透力的复方精油,将人参、狗脊、细辛搭配可补气通络祛寒湿,将乳香、威灵仙、马钱子结合可活血化瘀,促进气血循环,舒筋活络,有利于心肺功能的改善,再协同白芍、女贞子可达到滋阴补肾的效果,君臣辅佐,可显著改善失眠、焦虑,有效提高免疫力,补气温阳去寒、活血化瘀,实现内病外调、内需外补。
具体实施方式
以下将结合具体实施方案来说明本发明。需要说明的是,下面的实施例为本发明的示例,仅用来说明本发明,而不用来限制本发明。在不偏离本发明主旨或范围的情况下,可进行本发明构思内的其他组合和各种改良。在以下实施例中,所使用到的化合物及相关试剂均可从市场购得,其中抚顺琥珀购自抚顺富绅琥珀有限公司,辛烯基琥珀酸酐选自武汉克米克生物医药技术有限公司,甘油购自广州达创贸易有限公司,天然冰片购自江西雪松天然药用油有限公司。
琥珀提取物Ⅰ的制备
(1)将30g附子、30g、薰衣草、10g川芎加入400g低共熔试剂(氯化胆碱和1,2-丙二醇的质量比为1: 2)中,加热至40℃,保持恒温,搅拌1h;
(2)将40g抚顺琥珀用水洗净、沥干,粉碎至20目,然后加入步骤(1)所得的液体中,升温至55℃,浸泡72h,用200目滤布过滤,再用孔径为0.15μm的微滤氧化铝陶瓷膜过滤,过滤操作的压力为0.8Mpa,操作温度为35℃,即得琥珀提取物Ⅰ。
琥珀提取物Ⅱ的制备
(1)将10g附子、20g、薰衣草、10g川芎加入200g低共熔试剂(氯化胆碱和1,2-丙二醇的质量比为1: 4)中,加热至30℃,保持恒温,搅拌0.5h;
(2)将20g抚顺琥珀用水洗净、沥干,粉碎至10目,然后加入步骤(1)所得的液体中,升温至45℃,浸泡48h,用100目滤布过滤,再用孔径为0.1μm的微滤氧化铝陶瓷膜过滤,过滤操作的压力为0.1Mpa,操作温度为25℃,即得琥珀提取物Ⅱ。
琥珀提取物Ⅲ的制备
(1)将10g抚顺琥珀用水洗净、沥干,粉碎至20目,然后加入去离子水中;
(2)升温至55℃,浸泡72h,用200目滤布过滤,再用孔径为0.15μm的微滤氧化铝陶瓷膜过滤,过滤操作的压力为0.8Mpa,操作温度为35℃,即得琥珀提取物Ⅲ。
改性淀粉Ⅰ的制备
(1)取100g淀粉加入水中,用0.015mmol/L的氢氧化钠水溶液调整pH为8,搅拌均匀,
(2)在步骤(1)的溶液中,加入10g辛烯基琥珀酸酐和300g甘油,混匀后在130℃下熔融共混35min,冷却后即改性淀粉Ⅰ。
改性淀粉Ⅱ的制备
(1)取100g淀粉加入水中,用0.015mmol/L的氢氧化钠水溶液调整pH为9,搅拌均匀,
(2)在步骤(1)的溶液中,加入5g辛烯基琥珀酸酐和100g甘油,混匀后在120℃下熔融共混25min,冷却后即改性淀粉Ⅱ。
实施例1
一种具有高皮肤穿透力的复方精油,所述具有高皮肤穿透力的复方精油包括以下重量份的原料:人参10份、狗脊10份、细辛8份、威灵仙10份、乳香10份、马钱子8份、土虫7份、白芍20份、女贞子8份、甘草7份、渗透液(琥珀提取物与天然冰片的质量比为5:1)30份、促渗助剂(改性淀粉Ⅰ和香兰基丁基醚的质量比为2:1)10份、杏仁35份、仙人掌30份和去离子水160份。
制备方法:
(1)将人参、狗脊、细辛、威灵仙、乳香、马钱子、土虫、白芍、女贞子、甘草、杏仁、仙人掌加入去离子水中,浸泡72h;
(2)在步骤(1)得到的溶液中加入渗透液、促渗助剂,混匀,即得具有高皮肤穿透力的复方精油。
实施例2
一种具有高皮肤穿透力的复方精油,所述具有高皮肤穿透力的复方精油包括以下重量份的原料:人参12份、狗脊12份、细辛10份、威灵仙12份、乳香12份、马钱子10份、土虫10份、白芍25份、女贞子10份、甘草10份、渗透液(琥珀提取物与天然冰片的质量比为5:1)40份、促渗助剂(改性淀粉Ⅱ和香兰基丁基醚的质量比为2:1)15份、杏仁40份、仙人掌35份和去离子水180份。
制备方法与实施例1相同。
实施例3
一种具有高皮肤穿透力的复方精油,所述具有高皮肤穿透力的复方精油包括以下重量份的原料:人参6份、狗脊8份、细辛6份、威灵仙6份、乳香6份、马钱子6份、土虫5份、白芍15份、女贞子6份、甘草5份、渗透液(琥珀提取物与天然冰片的质量比为5:1)20份、促渗助剂(改性淀粉Ⅱ和香兰基丁基醚的质量比为1:1)5份、杏仁30份、仙人掌25份和去离子水120份。
制备方法与实施例1相同。

与实施例1的区别在于:不添加渗透液。

与实施例1的区别在于:所添加的渗透液中缺少琥珀提取物。

对各实施例、对比例制备的具有高皮肤穿透力的复方精油进行功效试验,试验项目为失眠、焦虑的改善情况。
测试对象:具有失眠、焦虑、免疫力低下的30-50岁人群,随机分为18组,每组50人,进行本试验前3天不能使用其他同类产品。
测试方法:1)将1.5ml具有高皮肤穿透力的复方精油涂抹在脊柱部位,用刮板由下至上重复刮5min;2)将2ml具有高皮肤穿透力的复方精油涂抹在脊柱两侧的距离脊柱3cm的部位,由上至下用刮板重复刮15min,使用3个疗程,每位志愿者在疗程开始前和疗程结束后,按照SAS焦虑自评量表自我测试焦虑情况,计算焦虑程度改善比例(焦虑改善比例=(疗程前焦虑程度-疗程后焦虑程度)/疗程前焦虑程度);佩戴智能手环记录睡眠时长和深睡时长,计算睡眠时长增加比例(睡眠时长增加比例=(疗程后睡眠时长-疗程前睡眠时长)/疗程前睡眠时长)和深睡时长增加比例(深睡时长增加比例=(疗程后深睡时长-疗程前深睡时长)/疗程前深睡时长)。结果如表1所示:

由对比例1、对比例2、对比例3、对比例4与实施例1的测试结果比较可知,实施例1制备的具有高皮肤穿透力的复方精油选择琥珀粉和冰片搭配使用,可有效增强精油的穿透力,发挥长久有效的效果;由对比例5、对比例6、对比例7与实施例1的测试结果比较可知,通过使用促渗助剂能加大天然冰片的溶解量,与渗透液协同增强精油的皮肤穿透力,使其具有良好的渗透效果。综合比较各实施例与对比例测试结果可知,本发明提供的具有高皮肤穿透力的复方精油,具有较强的穿透力,使精油中的有益成分顺利地透过皮肤、深入身体经络,修复老化的筋膜层,增加轻松愉悦感,长期使用效果不会衰减且无毒副作用,通过选择人参、狗脊、附子、细辛为原料可补气通络祛寒湿,再配以乳香、威灵仙、马钱子活血化瘀促进气血循环,舒筋活络,协同白芍、女贞子滋阴补肾,君臣辅佐,具有良好的疗效。

1.一种增强中药复方皮肤穿透力的复方制剂,其特征在于,所述增强中药复方皮肤穿透力的复方制剂由以下重量份的原料制备而成:人参6-12份、狗脊8-12份、细辛6-10份、威灵仙6-12份、乳香6-12份、马钱子6-10份、土虫5-10份、白芍15-25份、女贞子6-10份、甘草5-10份、渗透液20-40份、促渗助剂5-15份、杏仁30-40份、仙人掌25-35份和去离子水120-180份;
所述渗透液由琥珀提取物和天然冰片制备而成,所述琥珀提取物与天然冰片的质量比为3-5:1;

(2)将琥珀用水洗净、沥干,粉碎至10-20目,然后加入步骤(1)所得的液体中,升温至45-55℃,浸泡48-72h,用100-200目滤布过滤,再用孔径为0.1-0.15m的微滤氧化铝陶瓷膜过滤,收集液体,即得琥珀提取物;
所述促渗助剂是改性淀粉、香兰基丁基醚按质量比为1-2:1混合而成的混合物;
所述改性淀粉的制备方法包括以下步骤:
(1)取淀粉加入水中,调节pH为8-9,搅拌均匀;
(2)在步骤(1)的溶液中,加入辛烯基琥珀酸酐和甘油,混匀后在120-130℃下熔融共混25-35min,冷却后即改性淀粉;
所述具有皮肤穿透力的复方制剂的制备方法包括以下步骤:(1)将人参、狗脊、细辛、威灵仙、乳香、马钱子、土虫、白芍、女贞子、甘草、杏仁、仙人掌加入去离子水中,浸泡48-72h;
(2)在步骤(1)得到的溶液中加入渗透液、促渗助剂,混匀,即得具有皮肤穿透力的复方制剂。
2.根据权利要求1所述的增强中药复方皮肤穿透力的复方制剂,其特征在于,所述增强中药复方皮肤穿透力的复方制剂由以下重量份的原料制备而成:人参10份、狗脊10份、细辛8份、威灵仙10份、乳香10份、马钱子8份、土虫7份、白芍20份、女贞子8份、甘草7份、渗透液30份、促渗助剂10份、杏仁35份、仙人掌30份和去离子水160份。
3.根据权利要求1所述的增强中药复方皮肤穿透力的复方制剂,其特征在于,所述琥珀提取物的制备方法中,步骤(1)中所述低共熔溶剂由氯化胆碱和1,2-丙二醇组成,氯化胆碱和1,2-丙二醇的质量比为1:2-4;所述附子、薰衣草、川芎的质量比为2-3:1-3:1;所述低共熔试剂的质量为附子、薰衣草、川芎总质量的5-10倍;所述加热温度为30-40℃;步骤(2)中所述琥珀选自缅甸琥珀或抚顺琥珀,所述琥珀的质量为低共熔试剂的10-20%;所述微滤氧化铝陶瓷膜的操作压力为0.1-0.8Mpa,操作温度为25-35℃。
4.根据权利要求1所述的增强中药复方皮肤穿透力的复方制剂,其特征在于,所述改性淀粉的制备方法中,步骤(2)中所述辛烯基琥珀酸酐的质量为淀粉的5-10%;所述甘油的质量为辛烯基琥珀酸酐的1-3倍。
