天然产物是生物体经过长期进化形成的自身组成成分或其代谢产物,是生物活性物质和实用药物研发的重要对象。微生物是天然产物的主要来源之一,近几十年来的研究表明已经很难从土壤中分离到新的菌种,并提取出新的具有生物活性的次生代谢产物。然而,从动物体内分离新的菌种,尤其是昆虫体内分离新的菌种是另一个重要渠道。共生菌与宿主昆虫之间形成了密切关系,能帮助宿主昆虫消化食物,抵御病原菌侵染,提供食物来源,以及为宿主昆虫提供营养物质等。共生菌对昆虫的作用主要集中在消化食物、抵抗病原菌侵染、保护幼虫、协同进化等方面,而这些共生菌生物活性的表现主要是利用其次生代谢产物来实现的。
CN106635885A公开了一种昆虫病原线虫共生菌及其应用,该共生菌为致病杆菌(Xenorhabdus budapestensis)HEB02,保藏号为CGMCC No.12352,其发酵液具有较强的抑制大豆立枯病原菌丝生长的能力,且随着体积浓度的增加而增大,抑菌效果越明显。CN108676734A公开了一株昆虫病原真菌及其应用,该共生菌为球孢白僵菌(Beauveriabassiana)HUBB,保藏号为GMCC No.15866,其发酵液对马铃薯早疫病有很好的拮抗作用。CN111876337A公开了一种尖孢镰刀菌及其发酵方法与应用,该尖孢镰刀菌(Fusarium oxysporum),保藏号为GMCC No.19031,发酵液有显著的抑菌活性,能用于白粉病、灰霉病及霜霉病等植物病害的防治。
氟唑菌酰羟胺(pydiflumetofen)是作用于琥珀酸脱氢酶(SDHI)的新型吡啶酰胺类杀菌剂。氟唑菌酰羟胺具有广谱、高效的特征,适用于许多作物,例如玉米、小粒谷物、大豆、花生、油菜籽、藜麦等。在所有化学类型的产品中,氟唑菌酰羟胺对叶斑病和白粉病活性最高,对难以防治的病害,如葡萄孢菌(Botrytis spp.)、核盘菌(Sclerotinia spp.)和棒孢菌(Corynespora spp.)等病原菌引起的病害亦有较好的防治效果;同时该杀菌剂还突破性地防治谷物上由镰刀菌(Fusarium)引起的病害,如赤霉病(Fusarium headblight)等。
当前,农林业病虫害防治的主要方式是化学防治,但是长期使用性质相对稳定的化学药剂,会增强害虫、病原菌的抗药性,降低其防治效果。因此,开发及应用高效、安全、不易产生抗药性的药剂来防治植物病虫害,已成为农药及其制剂开发技术领域的研究热点。
发明内容
本发明的目的在于将昆虫共生菌次生代谢产物与农药复配,探究昆虫共生菌次生代谢产物与农药复配用于防治植物病虫害所产生的有益效果。
一方面,本发明提供了一种包含昆虫共生菌次生代谢产物的农用组合物,其以质量份计,有效成分由致病杆菌HEB02的发酵液提取物5-60份、球孢白僵菌HUBB的发酵液提取物10-50份、尖孢镰刀菌的发酵液提取物9-50份、氟唑菌酰羟胺30-80份构成;致病杆菌HEB02的菌种保藏号为CGMCC No.12352,球孢白僵菌HUBB的菌种保藏号为CGMCC No.15866,尖孢镰刀菌的菌种保藏号为CGMCCNo.19031。
优选地,本发明提供的包含昆虫共生菌次生代谢产物的农用组合物中,以质量份计,有效成分由致病杆菌HEB02的发酵液提取物39份、球孢白僵菌HUBB的发酵液提取物32份、尖孢镰刀菌的发酵液提取物21份、氟唑菌酰羟胺68份构成。
优选地,本发明提供的包含昆虫共生菌次生代谢产物的农用组合物中,以质量份计,有效成分由致病杆菌HEB02的发酵液提取物44份、球孢白僵菌HUBB的发酵液提取物43份、尖孢镰刀菌的发酵液提取物34份、氟唑菌酰羟胺75份构成。
优选地,本发明提供的包含昆虫共生菌次生代谢产物的农用组合物中,以质量份计,有效成分由致病杆菌HEB02的发酵液提取物41份、球孢白僵菌HUBB的发酵液提取物39份、尖孢镰刀菌的发酵液提取物28份、氟唑菌酰羟胺68份构成。
进一步地,本发明提供的包含昆虫共生菌次生代谢产物的农用组合物中,所述致病杆菌HEB02的发酵液提取物的制备方法为:将致病杆菌HEB02菌种接种于NA培养液中,于28℃180r/min振荡培养24h后,配制成悬浮液,悬浮液中致病杆菌HEB02浓度控制在106个/mL数量级;以1%体积比例的接种量,将致病杆菌HEB02接种于NA培养液中,在28℃180r/min的条件下,振荡发酵培养48h,倒入离心管中冷冻离心后,取上清液,获得无菌体发酵液,将无菌体发酵液经乙酸乙酯萃取干燥,即为所述致病杆菌HEB02的发酵液提取物。
进一步地,本发明提供的包含昆虫共生菌次生代谢产物的农用组合物中,球孢白僵菌HUBB的发酵液提取物的制备方法为:将球孢白僵菌HUBB菌种接种于液体培养基中,于28℃180r/min振荡培养24h后,配制成悬浮液,悬浮液中球孢白僵菌HUBB浓度控制在106个/mL数量级;按体积比1%接种于液体培养基中,在28℃180r/min的条件下,振荡发酵培养48h,倒入离心管中冷冻离心后,取上清液,获得无菌体发酵液,将无菌体发酵液经乙酸乙酯萃取干燥,即为所述球孢白僵菌HUBB的发酵液提取物。
进一步地,本发明提供的包含昆虫共生菌次生代谢产物的农用组合物中,尖孢镰刀菌的发酵液提取物的制备方法为:将尖孢镰刀菌菌种接种于液体培养基,32℃震荡培养3d,配制成悬浮液,悬浮液中尖孢镰刀菌浓度控制在106个/mL数量级;以5%体积比例的接种量接入液体培养基中,30℃120r/min振荡培养72h,倒入离心管中冷冻离心后,取上清液,获得无菌体发酵液,将无菌体发酵液经乙酸乙酯萃取干燥,即为尖孢镰刀菌的发酵液提取物。
另一方面,本发明涉及包含昆虫共生菌次生代谢产物的农用组合物作为杀菌剂的用途。
具体地,本发明提供的包含昆虫共生菌次生代谢产物的农用组合物用于防治白粉病或赤霉病。
根据实际应用需要,本发明提供的包含昆虫共生菌次生代谢产物的农用组合物按照本领域技术人员公知的方法,可调整制备成便于农业上施用的制剂剂型。本领域技术人员易于知晓,除含有有效成分外,制剂体系中还含有配制农药制剂所需的助剂。助剂可以选用表面活性剂、润湿剂、稳定剂、分散剂、增稠剂、pH调节剂、消泡剂、防冻剂、填料等中的一种或几种的混合。本发明的助剂可以均为已知物质,比如农药制剂中常用的各种助剂,根据不同情况可以有所变化,并无特别限定。
具体地,本发明提供的包含昆虫共生菌次生代谢产物的农用组合物制成悬浮剂,悬浮剂的制备选用气相SiO2作为增稠剂。
与现有技术相比,本发明的农用组合物,至少具有下述的优点或
有益效果:
本发明将昆虫共生菌次生代谢产物与氟唑菌酰羟胺复配后,发现其在防治白粉病和赤霉病表现优异,表现出协同增效的效果。本发明将各有效成分制成悬浮剂时,选用气相SiO2作为增稠剂,使得该组合物农药持效期得到一定延长。
具体实施方式
下面,结合实施例对本发明的技术方案进行说明,但是,本发明并不限于下述的实施例。
下述各实施例中实验方法和检测方法,如无特殊说明,均为常规方法;所述药剂和材料,如无特殊说明,均可在市场上购买得到;所述指标数据,如无特殊说明,均为常规测量方法。
制剂实施例1
本实施例提供了包含昆虫共生菌次生代谢产物的农用组合物的制备试验。
5.5%包含昆虫共生菌次生代谢产物的农用组合物悬浮剂的制备,选用气相SiO2为增稠剂。实施例所用气相SiO2购自江西大凯新材料有限公司。
各组分的重量百分比为:
致病杆菌HEB02的发酵液提取物0.5%
球孢白僵菌HUBB的发酵液提取物1%
尖孢镰刀菌的发酵液提取物1%
氟唑菌酰羟胺原药3%
润湿分散剂-烷基酚聚氧乙烯基醚甲醛缩合物磷酸盐5%
增稠剂-气相SiO23%
稳定剂-苯甲酸钠0.5%
防冻剂-乙二醇2%
消泡剂-二甲基硅油0.2%
去离子水补足100%。
致病杆菌HEB02的菌种保藏号为CGMCC No.12352,球孢白僵菌HUBB的菌种保藏号为CGMCC No.15866,尖孢镰刀菌的菌种保藏号为CGMCC No.19031。
致病杆菌HEB02的发酵液提取物的制备:将致病杆菌HEB02菌种接种于NA培养液(牛肉膏5g、蛋白胨10g和蒸馏水1000mL)中,于28℃180r/min振荡培养24h后,配制成悬浮液,悬浮液中致病杆菌HEB02浓度控制在106个/mL数量级;以1%体积比例的接种量,将致病杆菌HEB02接种于NA培养液中,在28℃180r/min的条件下,振荡发酵培养48h,倒入离心管中冷冻离心后,取上清液,获得无菌体发酵液,将无菌体发酵液经乙酸乙酯萃取干燥,即为所述致病杆菌HEB02的发酵液提取物。
球孢白僵菌HUBB的发酵液提取物的制备:将球孢白僵菌HUBB菌种接种于改良马丁氏培养液(过滤后马铃薯蒸煮液1000mL、葡萄糖20g、KH2PO43g、MgSO4·7H2O 1.5g、维生素B18mg)中,于28℃180r/min振荡培养24h后,配制成悬浮液,悬浮液中球孢白僵菌HUBB浓度控制在106个/mL数量级;按体积比1%接种于改良马丁氏培养液中,在28℃180r/min的条件下,振荡发酵培养48h,倒入离心管中冷冻离心后,取上清液,获得无菌体发酵液,将无菌体发酵液经乙酸乙酯萃取干燥,即为所述球孢白僵菌HUBB的发酵液提取物。
尖孢镰刀菌的发酵液提取物的制备:将尖孢镰刀菌菌种接种于接入PDA培养基(马铃薯200g、葡萄糖20g、蒸馏水1L),32℃震荡培养3d,配制成悬浮液,悬浮液中尖孢镰刀菌浓度控制在106个/mL数量级;以5%体积比例的接种量接入PDA培养基中,30℃120r/min振荡培养72h,倒入离心管中冷冻离心后,取上清液,获得无菌体发酵液,将无菌体发酵液经乙酸乙酯萃取干燥,即为尖孢镰刀菌的发酵液提取物。
将全部物料混合搅拌,搅拌均匀后把物料加入到砂磨机进行砂磨,砂磨2-3个小时后使物料达到要求粒剂,即得到5.5%包含昆虫共生菌次生代谢产物的农用组合物悬浮剂。经检验有效成分含量合格,悬浮率95%,通过75μm筛的筛分率为99.5%,1min持久起泡性20mL,倾倒后残余物3.4%,洗涤后残余物0.3%,40℃42d热贮稳定性合格。
制剂实施例2
本实施例提供了包含昆虫共生菌次生代谢产物的农用组合物的制备试验。
5.5%包含昆虫共生菌次生代谢产物的农用组合物悬浮剂的制备,选用黄原胶为增稠剂。实施例所用黄原胶购自久辉生物科技有限公司。
制备方法同制剂实施例1,区别在于,选用黄原胶为增稠剂。
实施例1
本实施例提供了包含昆虫共生菌次生代谢产物的农用组合物的防治草莓白粉病的试验。
试验地点为运城市草莓种植基地,试验草莓品种为红颜。种植采用标准八型钢管塑料大棚,水肥一体化种植,双行栽培,畦宽60cm,沟宽30cm,株行距20cm×25cm。试验大棚历年均有草莓白粉病发生,常年采用氟唑菌酰羟胺防治白粉病。
试验设有5组试验组,设有20组单剂对照组,设有1组清水对照组,各试验组制剂均采用制剂实施例1的制剂方法配置,区别在于有效成分与配比不同,各试验组有效成分与配比如表1所示。
本试验采用Colby法计算农药组合物的理论防效E0,当E-E0>0时,说明混配组合物产生增效作用;当E-E0<0时,说明混配组合物产生拮抗作用当E-E0值等于0时,说明混配组合物产生相加作用,公式中的E表示各处理的实测防效%。
根据防效对E0进行换算,E0的计算公式为:
其中,X为致病杆菌HEB02的发酵液提取物用量为x g/hm2的防治效果,Y为球孢白僵菌HUBB的发酵液提取物用量为y g/hm2的防治效果,Z为尖孢镰刀菌的发酵液提取物用量为z g/hm2的防治效果,N为氟唑菌酰羟胺用量为n g/hm2的防治效果。在采用Colby法计算中,E为包含昆虫共生菌次生代谢产物的农用组合物用量为(x+y+z+n)g/hm2的防治效果。
叶片病情分级标准:0级,叶片无病斑;1级,叶片病斑面积占总叶面积的5%以下;3级,病斑面积占总叶面积6%~15%;5级,病斑面积占总叶面积16%~25%;7级,病斑面积占总叶面积26%~50%;9级,病斑面积占总叶面积的51%以上。
病情指数的计算公式为:
防治效果的计算公式为:
其中,CK0为空白对照区施用前病情指数;CK1为空白对照区施用后病情指数;PT0为处理区施用前病情指数;PT1为处理区施用后病情指数。
各试验小区面积15m2,种植150株草莓,每组处理重复4次,随机区组排列。各小区栽培管理条件一致。草莓白粉病发病初期施药1次,对草莓叶片正反面均匀喷雾,试验期间未施用其他药剂。试验期间观察各药剂处理区草莓叶片是否产生药害现象。在施药前和施药后7d分别调查草莓叶片发病情况。每小区定点调查15株,每株调查全部叶片,试验结果如表1所示。

根据Colby法,表1数据显示,本发明提供的包含昆虫共生菌次生代谢产物的农用组合物表现出协同增效的作用,E-E0最大为19.92%,最小为11.67%,均大于10%。
实施例2
本实施例提供了包含昆虫共生菌次生代谢产物的农用组合物的防治小麦赤霉病的试验。
试验地点为河南省新乡市,试验小麦品种为小偃22号。在小区试验的基础上,选择长势均匀整齐、易发病的田块进行示范性试验。试验设有5组试验组(增稠剂选为气相SiO2,制剂参考制剂实施例1),5组对照试验组(增稠剂选为黄原胶,制剂参考制剂实施例2),以不喷药为对照,共11个处理,每处理重复3次,小区面积6m2,随机区组排列。小麦扬花初期喷孢子悬浮液(浓度为5×105/mL)接种,保护作用试验于接种前2d喷药,治疗作用试验于接种后2d喷药。分别于接种后田间病情扩展稳定时,调查小麦的发病情况。
病情指数级别为四级:0级:无病1级:发病麦穗面积占总面积25%以下;2级:发病麦穗面积占总面积25%~50%;3级:发病麦穗面积占总面积50%~75%;4级:发病麦穗面积占总面积75%以上。防效计算方式同实施例1。
接种后2d喷药,各试验喷药有效成分使用量均为10g/hm2,分别于喷药后7d、14d和21d调查各试验小区内病情,病情指数调查时每小区随机取3个1m2内所有的麦穗,按病情逐穗记录,计算病情指数及相对防效,试验结果如表2所示。

如表2所示,本发明所提供的包含昆虫共生菌次生代谢产物的农用组合物选用气相SiO2为增稠剂时,在单次施药后21d依然保有较好的药效,而选用常规增稠剂黄原胶时,在单次施药后21d防效不佳。由此可见,本发明所提供的包含昆虫共生菌次生代谢产物的农用组合物选用气相SiO2为增稠剂时,农药持效期得到一定延长。
如上所述,即可较好地实现本发明,上述的实施例仅仅是对本发明的优选实施方式进行描述,并非对本发明的范围进行限定,在不脱离本发明设计精神的前提下,本领域普通技术人员对本发明的技术方案做出的各种改变和改进,均应落入本发明确定的保护范围内。
Claims (6)
1.一种包含昆虫共生菌次生代谢产物的农用组合物,其特征在于,以质量份计,有效成分由致病杆菌HEB02的发酵液提取物5-60份、球孢白僵菌HUBB的发酵液提取物10-50份、尖孢镰刀菌的发酵液提取物9-50份、氟唑菌酰羟胺30-80份构成;
所述致病杆菌HEB02的菌种保藏号为CGMCC No.12352,所述球孢白僵菌HUBB的菌种保藏号为CGMCC No.15866,所述尖孢镰刀菌的菌种保藏号为CGMCC No.19031;
所述致病杆菌HEB02的发酵液提取物的制备方法为:将致病杆菌HEB02菌种接种于NA培养液中,于28℃ 180r/min振荡培养24h后,配制成悬浮液,悬浮液中致病杆菌HEB02浓度控制在106个/mL数量级;以1%体积比例的接种量,将致病杆菌HEB02接种于NA培养液中,在28℃ 180r/min的条件下,振荡发酵培养48h,倒入离心管中冷冻离心后,取上清液,获得无菌体发酵液,将无菌体发酵液经乙酸乙酯萃取干燥,即为所述致病杆菌HEB02的发酵液提取物;
所述球孢白僵菌HUBB的发酵液提取物的制备方法为:将球孢白僵菌HUBB菌种接种于液体培养基中,于28℃ 180r/min振荡培养24h后,配制成悬浮液,悬浮液中球孢白僵菌HUBB浓度控制在106个/mL数量级;按体积比1%接种于液体培养基中,在28℃ 180r/min的条件下,振荡发酵培养48h,倒入离心管中冷冻离心后,取上清液,获得无菌体发酵液,将无菌体发酵液经乙酸乙酯萃取干燥,即为所述球孢白僵菌HUBB的发酵液提取物;
所述尖孢镰刀菌的发酵液提取物的制备方法为:将尖孢镰刀菌菌种接种于液体培养基,32°C震荡培养3d,配制成悬浮液,悬浮液中尖孢镰刀菌浓度控制在106个/mL数量级;以5%体积比例的接种量接入液体培养基中,30°C120r/min振荡培养72h,倒入离心管中冷冻离心后,取上清液,获得无菌体发酵液,将无菌体发酵液经乙酸乙酯萃取干燥,即为尖孢镰刀菌的发酵液提取物。
2.根据权利要求1所述的包含昆虫共生菌次生代谢产物的农用组合物,其特征在于,以质量份计,有效成分由致病杆菌HEB02的发酵液提取物39份、球孢白僵菌HUBB的发酵液提取物32份、尖孢镰刀菌的发酵液提取物21份、氟唑菌酰羟胺68份构成。
3.根据权利要求1所述的包含昆虫共生菌次生代谢产物的农用组合物,其特征在于,以质量份计,有效成分由致病杆菌HEB02的发酵液提取物44份、球孢白僵菌HUBB的发酵液提取物43份、尖孢镰刀菌的发酵液提取物34份、氟唑菌酰羟胺75份构成。
4.根据权利要求1所述的包含昆虫共生菌次生代谢产物的农用组合物,其特征在于,以质量份计,有效成分由致病杆菌HEB02的发酵液提取物41份、球孢白僵菌HUBB的发酵液提取物39份、尖孢镰刀菌的发酵液提取物28份、氟唑菌酰羟胺68份构成。
5.权利要求1-4任一项所述的包含昆虫共生菌次生代谢产物的农用组合物在草莓白粉病防治中的应用。
6.根据权利要求5所述的应用,其特征在于,所述农用组合物制成悬浮剂,所述悬浮剂的制备选用气相SiO2作为增稠剂。
