Abstract
本发明公开了一种清咽润喉和增强免疫力的中药提取物、药剂及其制备方法。该中药组合物由以下重量份的成分组成:罗汉果9~15份、麦冬6~12份、桔梗3~10份、百合6~12份、金银花6~15份、红景天1~6份、贝母5~10份、薄荷油1~3份。本发明的中药组合物配方严谨,原料都属于药食同源的中药,安全无毒。经动物功能试验结果证明,本发明所提供的中药组合物具有清咽润喉、增强免疫力的作用。可应用于药物或保健品的制备,具有广阔的市场前景。

技术领域
本发明涉及中药领域,具体而言,涉及一种清咽润喉和增强免疫力的中药提取物、药剂及其制备方法。
背景技术
咽喉是进食吞咽、呼吸和发音的部位,极易受到外界环境和内部因素的影响而感染发炎,从而引起一系列的咽喉问题。由于近年来空气质量的持续恶化,以及人们工作压力的增大和不良生活习惯的增多,咽喉炎的发病率明显增高。尤其是在过多有害气体环境下工作的人群和用嗓过多的人群,例如交警、教师、话务员等,经常会出现咽喉问题。清咽功能是原卫生部批准的保健食品功能项目之一,它所适用的主要人群便是日益增多的咽炎患者。由于目前咽炎患者群体庞大,加上人们的预防保健意识的上升,使得清咽功能保健食品市场前景广阔。
免疫是机体识别并排除抗原性异物,维护自身平衡和稳定的一种功能。它主要由免疫系统来完成,免疫系统具有识别“自我”和“非己”的特点,主要担负着免疫防御、免疫稳定、免疫监视等功能。正常时可防御病原微生物的感染,清除体内衰老、死亡细胞及靶细胞,防止机体细胞癌变,发挥免疫调节等作用,从而保护机体不受外邪影响。异常时,可引起免疫缺陷病、感染性疾病、超敏反应性疾病、自身免疫性疾病和恶性肿瘤等。因此,人体的免疫系统与身体健康密切相关。中医认为疾病的发生、发展和转归取决于正邪的消长,与机体的免疫功能有关,从而提出了扶正祛邪的主要治病原则。扶正是指扶助机体正气,增强体质、提高机体抗邪、抗病能力的一种治疗原则。通过增强正气的方法,祛邪外出,恢复健康状况。中药在免疫低下的治疗效果突出,能根据人们不同体质辨证论治,充分调动机体免疫力。
中药是中华名族的瑰宝,自古以来就有“药食同源”理论。目前增强免疫力、改善咽喉不适已成为日益关注的焦点,基于国家卫健委公布的药食同源目录以及古代经典名方为基础,科学组方并优选出最佳配比。研究开发出一种具有清咽润喉、增强免疫力作用,又无毒副作用的中药组合物及其最佳比例具有重要意义。

一种清咽润喉和增强免疫力的中药提取物,按照重量份计,其原料包括:罗汉果9~15份、麦冬6~12份、桔梗3~10份、百合6~12份、金银花6~15份、红景天1~6份、贝母5~10份、菊花6~15份、薄荷油1~3份。
一种上述清咽润喉和增强免疫力的中药提取物的制备方法,其特征在于,包括:
将罗汉果、麦冬、桔梗、百合、金银花、红景天、贝母、菊花、薄荷油混合得到提取原料;
将提取原料用水进行提取。
一种清咽润喉和增强免疫力的药剂,其包括上述清咽润喉和增强免疫力的中药提取物,以及药学上可接受的辅料。

一种清咽润喉和增强免疫力的中药提取物,按照重量份计,其原料包括:罗汉果9~15份、麦冬6~12份、桔梗3~10份、百合6~12份、金银花6~15份、红景天1~6份、贝母5~10份、菊花6~15份、薄荷油1~3份。
优选地,按照重量份计,其原料包括:罗汉果12~15份、麦冬8~12份、桔梗5~10份、百合6~10份、金银花8~13份、红景天3~5份、贝母7~10份、菊花8~12份、薄荷油1~2份。
更为优选地,按照重量份计,其原料包括:罗汉果15份、麦冬12份、桔梗5份、百合8份、金银花13份、红景天5份、贝母10份、菊花11份、薄荷油1份。
其中,罗汉果为葫芦科植物罗汉果的成熟果实,性凉,味甘,性质清凉、甘甜爽口,为名贵水果和补品。具有清肺止咳、润肠通便、生津润喉等功效。
麦冬,别名沿阶草,为百合科植物麦冬的干燥块根。性微寒,味甘、微苦,归肺经、胃经、心经。具有益胃生津、清心除烦、养阴润肺等功效。
桔梗为桔梗科桔梗属多年生草本植物桔梗的嫩茎叶及根。性平、味苦,辛,归胃经。具有宣肺、利咽、祛痰、排脓等功效。
百合为百合科多年生草本植物百合或细叶百合肉质鳞茎。性微寒,味甘,具有润肺止咳、清心安神等功效。主要用于燥热咳嗽,劳嗽咯血,虚烦惊悸,失眠多梦。
金银花为忍冬科植物忍冬、黄褐毛忍冬的花蕾。性寒,味甘,归肺、心、胃经。具有清热解毒,疏散风热等功效。主要用于痈肿疔疮,喉痹,丹毒,热毒血痢,风热感冒,温病发热。
红景天为景天科属植物红景天或大花红景天的根茎。性寒,味甘,归脾、肺经。具有健脾益气,清肺止咳,活血化瘀等功效。主要用于气虚血瘀,胸痹心痛,中风偏瘫,脾肺气虚,倦怠气喘。
贝母有川贝母和浙贝母两种,川贝母为百合科多年生草本植物川贝母、暗紫贝母等的地下鳞茎。浙贝母为百合科多年生草本植物浙贝母的地下鳞茎。贝母性微寒,味苦、甘,归肺经。具有清肺化痰止咳,清热散结等功效,常用治痰热咳嗽、肺痈、肺痿以及瘰疬、痈肿等证。
菊花为菊科植物菊的干燥白色头状花序。性微寒,味甘、苦,归肺、肝经。具有疏风清热、平肝明目、解毒消肿的功效,主治风热感冒、头痛眩晕、目赤肿痛、眼目昏花、疔疮肿毒等症。
薄荷油为唇形科植物薄荷的鲜茎叶经蒸馏而得的挥发油。性凉、味辛,归肝、肺经。具有疏风、清热的功效。主治外感风热、头痛目赤、咽痛、牙疼、皮肤风热。
以上药材中包含有多种具有清咽利嗓、润肺止咳作用的中药材,经过发明人的精心搭配后,按照特定的比例进行组合,使的各成分之间的药性得以充分发挥,起到协同增效的作用,从而达到对咽喉问题具有疗效好,见效快,无毒副作用的效果。除此之外,该中药提取物中的罗汉果、桔梗、贝母、麦冬、百合、菊花、金银花及红景天均具有增强免疫力的作用。长期使用下,可以有效提高人体免疫力,减少感染疾病的概率。
进一步地,本发明实施例还提供了一种上述清咽润喉和增强免疫力的中药提取物的制备方法,其包括:
S1. 将罗汉果、麦冬、桔梗、百合、金银花、红景天、贝母、菊花、薄荷油混合得到提取原料;
S2. 将提取原料用水进行提取。
其中,提取原料与水的质量比为1:10~15。在上述比例范围内,谁可以将药用成分充分地从原料中提取出,从而提高产品的治疗效果。
可选地,提取过程中,先家那个提取原料在冷水中浸泡0.5~1 h,再加热提取1~1.5h。将原料在冷水中浸泡后在热水中提取,可以促进药用成分的释放,提高提取效率。
提取结束后,趁热过滤,在滤渣中加水进行二次提取,过滤,合并两次提取后的滤液,减压浓缩即得。二次提取可以回收滤渣中的药用成分,对原料充分利用,避免浪费。
进行二次提取时,滤渣和水的质量比为1:5~8;在加热下提取0.5~1 h。在上述条件下,对于滤渣的提取效率较高,可以更好地对药用成分进行回收。
本发明实施例还提供了一种清咽润喉和增强免疫力的药剂,其包括上述清咽润喉和增强免疫力的中药提取物,以及药学上可接受的辅料。
具体地,根据药剂剂型的不同,辅料可以是溶剂、抛射剂、增溶剂、助溶剂、乳化剂、着色剂、黏合剂、崩解剂、填充剂、润滑剂、润湿剂、渗透压调节剂、稳定剂、助流剂、矫味剂、防腐剂、助悬剂、包衣材料、芳香剂、抗黏合剂、整合剂、渗透促进剂、pH值调节剂、缓冲剂、增塑剂、表面活性剂、发泡剂、消泡剂、增稠剂、包合剂、保湿剂、吸收剂、稀释剂、絮凝剂与反絮凝剂、助滤剂、释放阻滞剂等。
可选地,该药剂为粉剂、颗粒剂、胶囊剂、片剂、口服液剂中的任一种。上述剂型的药剂可以不仅便于患者服用吸收,而且更加利于储存和运输,可以实现商业化生产。

罗汉果13份、麦冬9份、桔梗4份、百合7份、金银花6份、红景天2份、贝母8份、菊花13份、薄荷油1份。

按上述配比称取各原料,加入15倍药材量的蒸馏水,浸泡60 min,加热提取90min,趁热过滤。再将滤渣加入8倍药材量的蒸馏水加热提取60min,过滤。合并两次滤液,减压浓缩。

本实施例提供了一种清咽润喉和增强免疫力的中药提取物,按照重量份数计,其原料包括:
罗汉果9份、麦冬10份、桔梗6份、百合12份、金银花7份、红景天4份、贝母7份、菊花6份、薄荷油1份。

按上述配比称取各原料,加入12倍药材量的蒸馏水,浸泡45 min,加热提取80min,趁热过滤。再将滤渣加入6倍药材量的蒸馏水加热提取40min,过滤。合并两次滤液,减压浓缩。

罗汉果11份、麦冬8份、桔梗10份、百合6份、金银花11份、红景天6份、贝母5份、菊花7份、薄荷油1份。

试验例1
本试验例采用实施例1~6所提供的清咽润喉和增强免疫力的中药提取物,对其增强免疫力功能进行测试,具体实验材料和实验方法如下:
1)实验材料:
动物:小鼠,ICR,70只,雄,18-22 g。分笼饲养于中国药科大学中药学院动物实验中心清洁级动物实验室内。
受试药物:本发明实施例1~6的中药组合物(分别对应给药1~6组)。
材料:游标卡尺(精密度0.02mm)、SRBC(绵羊红细胞)、微量注射器(50μL)、生理盐水、微量血凝实验板、离心机。
2)实验方法
1.动物分组:随机分为7组,每组10只。分别为空白对照组,给药1-6组。
2.给药方法:各给药组灌胃给药。给药1-6组,分别予以实施例1-6的中药提取物,给药量为19.2g生药/kg/d。均为每日1次,共15天。空白对照组给予同体积蒸馏水。
3. 脏器/体重比值测定:小鼠称重后颈椎脱臼处死,取其胸腺、脾脏,去其筋膜,用滤纸吸干脏器表面血污后分别称重,根据公式计算脏器指数。脏器指数=脏器质量(mg)/小鼠体质量(g)。
4.迟发型变态反应(DTH)实验:小鼠用2%(v/v)SRBC腹腔或静脉免疫,每只鼠注射0.2mL。免疫后4天,测量左后足跖部厚度,然后在测量部位皮下注射20%(v/v)SRBC,每只鼠20μL,注射后于24h测量左后足跖部厚度,同一部位测量三次,取平均值。
5.血清溶血素的测定-血凝法:将绵羊红细胞缓慢注入离心管内,盖好盖子后将离心管放入离心机,配平后离心(2000r/min)10min离出抗凝剂等。第一次离心结束后,将上层液体吸出,加入适量生理盐水,配平后进行第二次离心,(2000r/min)10min,进一步清洗红细胞。第二次离心结束后,同样吸出上层液体,再加入适量生理盐水。配平后进行第三次离心,2000r/min离心10min,进一步清洗红细胞。离心结束后一次吸出上层清液。最后将压积SRBC用生理盐水配成2%(v/v)的细胞悬液。每只鼠腹腔注射0.2mL进行免疫。4~5天后,摘除眼球取血于离心管内,放置约1h,将凝固血与管壁剥离,使血清充分析出,2000r/min离心10min,收集血清。用生理盐水将血清倍比稀释,将不同稀释度的血清分别置于微量血凝实验板内,每孔100μL,再加入100μL 0.5%(v/v)的SRBC悬液,混匀,装入湿润的平盘内加盖,于37℃温箱孵育3h,观察血球凝集程度。实验结果如表1~表3所示。

由表1可以看出,与空白组比较,各给药组均差异无统计学意义(P>0 .05),表明实施例1~6所提供的中药提取物对小鼠脾脏、胸腺质量无显著影响。

由表2和图1可以看出,与空白组比较,各给药组均差异有统计学意义(P<0 .05),表明实施例1~6所提供的中药提取物能增强小鼠迟发型变态反应能力,其中实施例5所提供的中药提取物对增强小鼠迟发型变态反应能力最强。

由表3和图2可以看出,与空白组比较,各给药组均差异有统计学意义(P<0 .05),表明实施例1~6所提供的中药提取物能具有促进小鼠抗体生成的作用,其中实施例5所提供的中药提取物促进小鼠抗体生成能力最强。
试验例2
本试验例采用实施例1~6所提供的清咽润喉和增强免疫力的中药提取物,根据《允许保健食品声称的保健功能目录(2020年版)》中清咽润喉功能检验方法项对其清咽润喉功能进行测试,具体实验材料和实验方法如下:
1)实验材料
动物:小鼠,ICR,70只,雄,18-22 g。分笼饲养于中国药科大学中药学院动物实验中心清洁级动物实验室内。
受试药物:本发明实施例1~6的中药提取物(分别对应给药1-6组)。
材料:直径9mm打孔器、致炎剂(二甲苯)、微量加样器、分析天平
2)实验方法
1.动物分组:随机分为7组,每组10只。分别为空白对照组,给药1-6组。
2.给药方法:各给药组灌胃给药。给药1-6组,分别予以实施例1-6处方提取物,给药量为19.2g生药/kg/d。均为每日1次,共15天。空白对照组给予同体积蒸馏水。
3.小鼠耳肿胀实验:汲取二甲苯20μL,滴加在小鼠右耳外侧面耳廓的中央,让其自由扩散,30分钟后,将小鼠脱颈椎处死,剪下双耳,用9mm直径打孔器在两耳相同部位打下耳片并称重,以两耳重量之差为耳廓肿胀值,计算耳廓肿胀率。
4.体重测定方法:将小鼠适应性饲养1周后,第一次灌胃前称重为初始体重。第八次灌胃前称重为中期体重,小鼠处死前称重为结束体重。增长值=结束体重(g)—初始体重(g)。实验结果如表4~5:

由表4和图3可以看出,与空白组比较,各给药组均差异有统计学意义(P<0 .05),表明实施例1~6所提供的中药提取物可明显抑制小鼠耳廓肿胀,其中实施例3和实施例5所提供的中药提取物抑制小鼠耳廓肿胀的效果最好。

由表5和图4、图5可以看出,与空白组比较,各给药组均差异有统计学意义(P<0.05),表明实施例1~3所提供的中药提取物可显著影响小鼠体重增长,其中实施例4~6所提供的中药提取物对体重增长有轻微影响。
试验例3
本试验例采用实施例5所提供的中药提取物,对其总皂苷、粗多糖含量测定,测试方法如下:
1)实验材料
试剂:正丁醇、无水乙醇、甲醇、氨水、高氯酸、冰乙酸、香草醛(5%)、人参皂苷Re标准储备液(0.2 mg/ml)、葡萄糖标准液(0. 1mg/ml)、蒽酮硫酸溶液(0.2%)。
仪器:紫外分光光度计、天平、超声波清洗器、离心机、恒温水浴锅、离心管。
2)实验方法
1.总皂苷含量测定:吸取混合均匀的待测试样10ml,加入100ml容量瓶中,再取1ml加入10ml容量瓶,混匀,制得供试品溶液。将10ml供试品溶液用水全部转移至分液漏斗中并加水至约25ml。加入20ml水饱和正丁醇振摇萃取,分取正丁醇液(必要时可离心),重复操作3次,合并正丁醇液用20 ml氨试液洗涤,重复操作2次,弃去氨试液,正丁醇液水浴蒸干,残渣用甲醇溶解并转移至10ml量瓶中,加甲醇定容至刻度,摇匀,滤过,取续滤液,备用。
标准曲线的制作:吸取人参皂苷Re标准溶液0 ml、0.4 ml、0.6ml、0.8 ml、1.0 ml、1.2 ml于10 ml具塞比色管中,置水浴中挥干溶剂,加入0.2 ml香草醛冰乙酸溶液,再加入0.8 ml高氯酸,混匀,使残渣全部溶解,置60℃水浴中加热10min,取出,冰浴冷却后,加入5.0 mL冰乙酸,摇匀后,立即于560 nm波长处测定吸光度。试液测定:吸取续滤液1.0ml于10ml具塞比色管中,置水浴中挥干溶剂,加入0.2 ml香草醛冰乙酸溶液,再加入0.8 ml高氯酸,混匀,使残渣全部溶解,置60℃水浴中加热10min,取出,冰浴冷却后,加入5.0ml冰乙酸,摇匀后,立即于560 nm波长处测定吸光度。
结果计算:
Xi—试样中总皂苷的含量(以人参皂苷Re计)
Ci—经试样背景校正后,由标准曲线算得被测液中人参皂苷Re质量(mg);
V—被测定样液的定容体积(ml);
V0—被测定样液的显色体积(ml);
m—试样的称样质量(g)。
2.粗多糖含量测定:准确吸取1.5 ml待测液于10ml 离心管中,加入7.5ml无水乙醇,加盖反复倾倒管子数次。在离心机中以4000r/min离心10min,并小心弃去上清液,再加1.5ml80%乙醇溶液冲洗离心管中沉淀物,重复一次后,再以4000r/min离心10min,直到上清夜滴加到蒽酮试剂中不显色为止,小心地用吸管将上层液体吸去。然后用热水分次溶解沉淀并稀释定容至250ml。过滤,弃去初滤液即为待测液。准确吸取葡萄糖标准液(0.1mg/mL)0、0.1、0.2、0.4、0.6、0.8、1.0ml于10ml具塞比色管中,加水至1.0ml,加入蒽酮试剂5ml充分混匀,在沸水浴中加热10min,取出在流水中冷却20min后,在620m波长下,以试剂空白调零,测定各管的吸光度值绘制标准曲线。准确吸取样品待测液1.0ml按标准曲线绘制步骤,于620m波长下测定吸光度值并求出样品含葡萄糖量。
m1—由标准曲线查得样品液含葡萄糖质量(mg);
m—样品质量(g);
n—稀释倍数。
实验结果如表6~7所示。

由表6和表7,结合图6、图7的标准曲线可以计算出,中药组合物实施例5总皂苷含量(以人参皂苷Re计)为23.43%~26.03%。粗多糖含量(以葡萄糖计)为15.63%-20.84%。总皂苷可以起到营养神经、活血化瘀等功效,也具有止咳化痰等作用。而粗多糖可以起到促进消化,促进自身免疫力,降低血压血脂,抗氧化等。从总皂苷和粗多糖的含量上,也可以证明本发明实施例所提供的中药提取物,在增强免疫力和清咽润喉上具有较佳的功效。
综上所述,本发明公开了一种清咽润喉和增强免疫力的中药组合物、提取物及其制法和应用。该中药组合物由以下重量份的成分组成:罗汉果9~15份、麦冬6~12份、桔梗3~10份、百合6~12份、金银花6~15份、红景天1~6份、贝母5~10份、薄荷油1~3份。本发明的中药组合物配方严谨,原料都属于药食同源的中药,安全无毒。经动物功能试验结果证明,本发明所提供的中药组合物具有清咽润喉、增强免疫力的作用。可应用于药物或保健品的制备,具有广阔的市场前景。
Claims (10)
1.一种清咽润喉和增强免疫力的中药提取物,其特征在于,按照重量份计,其原料包括:罗汉果9~15份、麦冬6~12份、桔梗3~10份、百合6~12份、金银花6~15份、红景天1~6份、贝母5~10份、菊花6~15份、薄荷油1~3份。
2.根据权利要求1所述的中药,其特征在于,按照重量份计,其原料包括:所述罗汉果12~15份、所述麦冬8~12份、所述桔梗5~10份、所述百合6~10份、所述金银花8~13份、所述红景天3~5份、所述贝母7~10份、所述菊花8~12份、所述薄荷油1~2份。
3.根据权利要求2所述的中药,其特征在于,按照重量份计,其原料包括:所述罗汉果15份、所述麦冬12份、所述桔梗5份、所述百合8份、所述金银花13份、所述红景天5份、所述贝母10份、所述菊花11份、所述薄荷油1份。
4.一种如权利要求1~3任一项所述的清咽润喉和增强免疫力的中药提取物的制备方法,其特征在于,包括:
将所述罗汉果、所述麦冬、所述桔梗、所述百合、所述金银花、所述红景天、所述贝母、所述菊花、所述薄荷油混合得到提取原料;
将所述提取原料用水进行提取。
5.根据权利要求4所述的制备方法,其特征在于,所述提取原料与所述水的质量比为1:10~15。
6.根据权利要求4所述的制备方法,其特征在于,提取过程中,先家那个所述提取原料在冷水中浸泡0.5~1 h,再加热提取1~1.5 h。
7.根据权利要求4所述的制备方法,其特征在于,提取结束后,趁热过滤,在滤渣中加水进行二次提取,过滤,合并两次提取后的滤液,减压浓缩即得。
8.根据权利要求7所述的制备方法,其特征在于,进行二次提取时,所述滤渣和水的质量比为1:5~8;在加热下提取0.5~1 h。
9.一种清咽润喉和增强免疫力的药剂,其特征在于,包括权利要求1~3任一项所述的清咽润喉和增强免疫力的中药提取物,以及药学上可接受的辅料。
10.根据权利要求1所述的药剂,其特征在于,所述药剂为粉剂、颗粒剂、胶囊剂、片剂、口服溶液剂中的任一种。
