Description
Technical Field
The invention belongs to the technical field of medicines for poultry, and relates to a microemulsion preparation for expelling trichomonas pigeon, and a preparation method and application thereof.
Background
Trichomonas pigeon is the most common infectious protozoal disease of pigeons, and the trichomonas pigeon is mainly parasitic in the upper digestive tract, so that warm and humid weather is more frequent; the pigeons in any place, variety and age can be ill, and the pigeon is one of the most common diseases threatening the pigeon raising industry. About 80% of pigeon with insects, the symptoms of adult pigeons are not obvious, and the symptoms of young pigeons are prominent and even die. Even if the young pigeons with insects are slightly ill, the nutrition absorption is affected, the quality is reduced, medicine residues are easy to appear during the administration, the export is blocked, the benefit is reduced, and the disease attracts high importance.
Trichomonas is an avian trichomonas belonging to the genus trichomonas, subglothuridae, polyslotriales, trichomonadaceae. The insect body is pear-shaped, round or oval, has a length of 5-9 micrometers and a width of 2-9 micrometers, has 4 flagella at the front end, has a wave membrane extending along the longitudinal axis of the insect body at the outer side, and has 1 axial thorn extending out of the insect body and resembling a tail. The insect body moves in body fluid by means of flagella and a wave membrane. The insect body is bred in a longitudinal two-split mode, and is bred for one generation in about 4.5 hours, and the in-vitro culture of the insect body can be grown by adding serum.
The main infection route of trichomonas pigeon is through mouth feel. The most common parasitic and damaging site for worms is the digestive tract. The canker sore of the pigeon is the settlement point of trichomonas, and a large number of living bodies are also in saliva; both the affected pigeons and the pigeons with insects are sources of infection. The insect body can infect pigeons through drinking water, feed, wounds, unclosed umbilical rings and other ways. The adult pigeons can directly transfer the insect bodies to the companion or own offspring through kissing each other or through 'pigeon milk' feeding young pigeons. Infection and stress factors of other diseases can become causative factors of the disease.
There are different clinical manifestations depending on the location of the damage to the insect body. Of these, the throat is the most commonly affected part. The diseased pigeons are manifested by dysphagia and dyspnea, loss of appetite, increased thirst, digestive disorders, listlessness, emaciation, loose feathers, and yellowish green manure due to invasion of the mucous membranes of the mouth, pharynx, esophagus and crop. The sick pigeon dies the anterior conjunctiva and the oral mucosa to take cyanosis. Often mixed with candida albicans or co-infected, the morbidity and mortality are high. The insect bodies can also invade the adenosis stomach, myostomach, intestines, lungs, air bags, livers and even peritoneum, and the sick pigeons show mental depression, progressive emaciation and loose feathers; if the respiratory tract is damaged, the pigeon with the disease can open mouth or stretch neck to breathe, cough and wheeze; if the intestinal tracts are damaged, the sick pigeons are wasted and eat, the yellow green sticky paste thin manure is pulled, the perianal feathers are full of thin manure, and the sick pigeons are rapidly emaciated and killed; the morbidity and mortality of young pigeons of 7-30 days old are high. Sometimes, the unclosed navel holes of young pigeons can be infected by padding and the like which pollute the insect bodies, and the sick pigeons are in a state of mental dullness, loose and disordered feathers, emaciation, reluctant to lie down, inflamed and red swelling of umbilical regions, tenderness and even ulcer focus.
At present, the most common medicine for preventing and treating trichomonas of pigeons in China is metronidazole, and the best using method is to feed the pigeon with feed or drink the pigeon with water, and the pigeon with feed can be used as a common preventive medicine or be mixed with health care sand for feeding. However, the metronidazole has certain toxicity, and the dosage can not be increased or the treatment course can not be prolonged at will when the metronidazole is used, and generally one treatment course is preferably five days, otherwise the phenomenon of reduced egg laying or increased egg laying of the breeding pigeons can be caused. In addition, in the 2B cancerogenic substance list, the excessive use of the metronidazole medicine by edible birds can cause influence on the health of human beings, and the residual medicine does not accord with the sustainable development thinking of green health. Therefore, development of a nontoxic, green and healthy product for preventing and controlling trichomonas pigeon is urgent.
Disclosure of Invention
Aiming at the defects of the prior art, the invention aims to provide a microemulsion preparation for expelling trichomonas pigeon, and a preparation method and application thereof.
In order to achieve the aim of the invention, the invention adopts the following technical scheme:
in a first aspect, the present invention provides a microemulsion formulation for repelling trichomonas pigeon, the microemulsion formulation for repelling trichomonas pigeon comprising the following components: macleaya cordata extract, carvacrol, an emulsifying agent, a cosolvent and water.
The invention creatively combines the macleaya cordata extract and the carvacrol as active ingredients, and combines an emulsifying agent, a cosolvent and water to develop a microemulsion preparation for expelling trichomonas pigeon. The microemulsion preparation is red to reddish brown liquid, and the scientific cooperation of the components ensures that the product has stable property and can be diluted and dissolved with water in any proportion; the macleaya cordata extract and the carvacrol are both plant extracts, are green, low-toxic and safe, have a synergistic effect on the effect of preventing and controlling the trichomonas pigeon, have more remarkable prevention and control effect on the trichomonas pigeon, have unexpected beneficial effects on preventing and controlling the trichomonas pigeon, and have no adverse reaction compared with the single use of the carvacrol or the macleaya cordata extract or the combination of other substances.
Preferably, the macleaya cordata extract is 0.5-2% by mass, such as 0.5%, 0.8%, 1%, 1.2%, 1.4%, 1.5%, 1.6%, 1.8%, 2% and the like, in the microemulsion preparation; the carvacrol is 1-4% by mass, such as 1%, 1.5%, 2%, 2.5%, 3%, 3.5%, 4% and the like, in the microemulsion preparation; other specific point values in the numerical ranges are selectable, and will not be described in detail herein.
The microemulsion preparation provided by the invention enables the active ingredients of macleaya cordata extract and carvacrol to stably exist in the preparation in a larger concentration through scientific matching of specific components, and the preparation system is very stable, thus providing a foundation for further development and utilization of the microemulsion preparation.
The cosolvent is added into the microemulsion preparation, so that the pH value can be adjusted to increase the solubility of the macleaya cordata extract in water, the concentration of the macleaya cordata extract in a system is more than 0.5%, and the solution is easy to become turbid due to the fact that the addition amount of the cosolvent exceeds the range.
Preferably, the mass percentage of the emulsifier in the microemulsion preparation is 10-30%, for example 10%, 12%, 15%, 18%, 20%, 25%, 28%, 29%, 30%, etc., and other specific values in the numerical range can be selected, so that no further description is given here.
Preferably, the emulsifier comprises any one or a combination of at least two of tween 80, EL-40 or tween 20, more preferably a combination of tween 80 and EL-40.
The addition of an emulsifier is the basis for forming a microemulsion formulation, the choice of which type has a certain effect on the stability of the system, more preferably a combination of tween 80 and EL-40 as emulsifier.
Preferably, the cosolvent is an alkaline solution.
Preferably, the alkaline solution comprises 1-5% sodium hydroxide solution.
Preferably, the content of sanguinarine (C 20H18NO4 +) in the macleaya cordata extract is not less than 40%, and the total content of sanguinarine (C 20H18NO4 +) and chelerythrine (C 21H18NO4 +) is not less than 60%.
In a second aspect, the present invention provides a method of preparing a microemulsion formulation for repelling trichomonas pigeon according to the first aspect, the method comprising:
(1) Mixing the macleaya cordata extract with 30-50% of water and cosolvent, and stirring until the solution is clear to obtain an aqueous phase mixture; mixing and stirring carvacrol and an emulsifier to obtain an oil phase mixture;
(2) Mixing the water phase mixture and the oil phase mixture, stirring and emulsifying until the solution is clear, and then mixing with the rest water to obtain the microemulsion preparation for expelling trichomonas pigeon.
The preparation method of the microemulsion preparation for expelling trichomonas pigeon has the advantages of simple process, easy operation, simple production process and equipment, low production cost and convenient industrialization, and can be implemented by simple mixing and dissolving.
Preferably, the pH of the aqueous mixture is between 1.5 and 2.0, e.g. ph=1.5, ph=1.6, ph=1.7, ph=1.8, ph=1.9, ph=2.0, etc.
The proper pH value can increase the solubility of the macleaya cordata extract in the water phase, and the macleaya cordata extract is difficult to stably exist in a system due to the too low or too high pH value.
Preferably, the stirring or mixing is independently performed at 15-40 ℃, e.g., 15 ℃, 17 ℃, 20 ℃, 25 ℃, 30 ℃, 35 ℃, 40 ℃, etc.
Preferably, the mixing and stirring time of step (1) is 10-20min, such as 10min, 12min, 14min, 16min, 17min, 18min, 20min, etc.
Other specific point values within the above numerical ranges are all selectable, and will not be described in detail herein.
In a third aspect, the invention provides a microemulsion preparation diluent for expelling trichomonas pigeon, which is obtained by diluting the microemulsion preparation for expelling trichomonas pigeon in the first aspect with water.
The microemulsion preparation for expelling trichomonas pigeon can be diluted with water according to any proportion, can obtain clear and transparent solution, is convenient to use, and can not block a drinking line.
In a fourth aspect, the invention provides the use of the microemulsion preparation for expelling trichomonas pigeon according to the first aspect or the microemulsion preparation diluent for expelling trichomonas pigeon according to the third aspect in the preparation of a medicament for preventing or treating trichomonas pigeon infection.
In a fifth aspect, the invention provides the use of a microemulsion preparation for expelling trichomonas in the first aspect or a microemulsion preparation diluent for expelling trichomonas in the third aspect for preparing a pigeon feed additive for alleviating trichomonas infection.
In a sixth aspect, the present invention also provides a method for using the microemulsion formulation of the first aspect, comprising: adding 0.25-1mL of microemulsion preparation into 1L of drinking water for preventing trichomonas infection; 2.5-10mL of microemulsion preparation is added to 1L of drinking water for treating trichomonas infection.

Detailed Description
The technical scheme of the invention is further described by the following specific embodiments. It will be apparent to those skilled in the art that the examples are merely to aid in understanding the invention and are not to be construed as a specific limitation thereof.
The carvacrol used in the examples below was a product purchased from Jiangxi Hai Natural plants Co., ltd; macleaya cordata extract is a product purchased from long Sha Kunbo biosystems, inc; cinnamaldehyde is a product purchased from Jiangxi Hai Rui natural plant Co., ltd; thymol is a product purchased from Jiangxi Hai Rui Natural plant Co.
Example 1
The embodiment provides a microemulsion preparation for expelling trichomonas pigeon, which comprises the following components: 1% macleaya cordata extract, 2% carvacrol, 15% EL-40, 5% Tween 80, a proper amount of 4% sodium hydroxide aqueous solution (determined according to the required pH value) and the balance of purified water. The preparation method comprises the following steps:
(1) Weighing raw materials of the macleaya cordata extract, adding purified water accounting for 40% of the total amount, uniformly stirring, adding a 4% sodium hydroxide aqueous solution to adjust the pH value to 1.9, and continuously stirring until the solution is clear;
(2) Weighing carvacrol raw material, EL-40 and Tween 80, mixing and stirring for 15min to obtain a uniform substance;
(3) Adding the solution obtained in the step (1) into the uniform substance obtained in the step (2), stirring and emulsifying while adding until the solution is clear, adding the rest purified water to the full amount, and stirring uniformly to obtain the final product. The graph of the sample obtained is shown in FIG. 1.
Example 2
The embodiment provides a microemulsion preparation for expelling trichomonas pigeon, which comprises the following components: 2% macleaya cordata extract, 1% carvacrol, 10% EL-40, 10% Tween 80, a proper amount of 4% sodium hydroxide aqueous solution (determined according to the required pH value) and the balance of purified water. The preparation method comprises the following steps:
(1) Weighing raw materials of the macleaya cordata extract, adding purified water accounting for 40% of the total amount, uniformly stirring, adding a 4% sodium hydroxide aqueous solution to adjust the pH value to 1.8, and continuously stirring until the solution is clear;
(2) Weighing carvacrol raw material, EL-40 and Tween 80, mixing and stirring for 20min to obtain a uniform substance;
(3) Adding the solution obtained in the step (1) into the uniform substance obtained in the step (2), stirring and emulsifying while adding until the solution is clear, adding the rest purified water to the full amount, and stirring uniformly to obtain the final product.
Example 3
The embodiment provides a microemulsion preparation for expelling trichomonas pigeon, which comprises the following components: 0.5% macleaya cordata extract, 2.5% carvacrol, 5% EL-40, 15% Tween 80, a proper amount of 4% sodium hydroxide aqueous solution (determined according to the required pH value) and the balance of purified water. The preparation method comprises the following steps:
(1) Weighing raw materials of the macleaya cordata extract, adding purified water accounting for 40% of the total amount, uniformly stirring, adding a 4% sodium hydroxide aqueous solution to adjust the pH value to 1.6, and continuously stirring until the solution is clear;
(2) Weighing carvacrol raw material, EL-40 and Tween 80, mixing and stirring for 10min to obtain a uniform substance;
(3) Adding the solution obtained in the step (1) into the uniform substance obtained in the step (2), stirring and emulsifying while adding until the solution is clear, adding the rest purified water to the full amount, and stirring uniformly to obtain the final product.
Example 4
This example provides a microemulsion formulation for the repellency of trichomonas pigeon which differs from example 1 only in the composition and preparation method in that 20% of the emulsifier EL-40 is used, all other conditions remaining unchanged.
Example 5
This example provides a microemulsion formulation for the repellency of trichomonas pigeon which differs from example 1 only in the composition and preparation method in that 20% tween 80 is used as the emulsifier, all other conditions remaining unchanged.
Example 6
This example provides a microemulsion formulation for the repellency of trichomonas pigeon which differs from example 1 only in the components and preparation method using 15% EL-40 and 5% tween 20 as emulsifiers, all other conditions remaining unchanged.
Example 7
This example provides a microemulsion preparation for expelling trichomonas pigeon, which is prepared by the method which is different from example 1 only in that in the step (1), 4% sodium hydroxide aqueous solution is added to adjust the pH value to 2.5, and stirring is continued, and other conditions are kept unchanged.
Comparative example 1
This comparative example provides a microemulsion formulation for the control of trichomonas pigeon, which is prepared by a process differing from that of example 1 only in that no 4% aqueous sodium hydroxide solution is added in step (1), and the other conditions remain unchanged. The graph of the sample obtained is shown in FIG. 2.
Comparative example 2
This comparative example provides a microemulsion formulation for the repellency of trichomonas pigeon which differs from example 1 only in that it does not contain macleaya cordata extract, and is prepared by dispensing a reduced amount of macleaya cordata extract onto the mass of carvacrol, with the other components remaining unchanged, and the method of preparation is described in example 1.
Comparative example 3
This comparative example provides a microemulsion formulation for the repellency of trichomonas pigeon which differs from example 1 only in that it does not contain carvacrol, and is formulated in reduced amounts to the quality of the macleaya cordata extract, the other components remaining unchanged, and the method of preparation is described in example 1.
Comparative example 4
This comparative example provides a microemulsion formulation for the repellency of trichomonas pigeon which differs from example 1 only in the replacement of carvacrol with equivalent amounts of cinnamaldehyde, the other components remaining unchanged, and the preparation method is described in reference to example 1.
Comparative example 5
This comparative example provides a microemulsion formulation for the repellency of trichomonas pigeon which differs from example 1 only in the replacement of carvacrol with equal amounts of thymol, the other components remaining unchanged, the preparation being as described in example 1.
Test example 1
Stability evaluation test:
The microemulsion formulations prepared in examples 1 to 7 and comparative example 1 were tested with reference to an accelerated test assay in the "animal pharmacopoeia of the people's republic of China" 2020 edition one stability. Placing for 6 months at 40+/-2 ℃ under the condition that the relative humidity is 70% +/-5%, monitoring the temperature and the humidity in real time, sampling once respectively at the 1 st month, 2 months, 3 months and 6 months during the test period, detecting the macleaya cordata content according to the method of macleaya cordata extract in 2006-2011 edition of quality standard assembly of veterinary medicine, and detecting the content of carvacrol in the carvacrol according to the detection method of carvacrol in the standard Q/ZD 034-2018 mixed feed additive feeding flavoring agent of classical biotechnology company of the florida, and observing and detecting the properties of the macleaya cordata. The results are shown in tables 1,2, 3 and 4.

Note that: the content of macleaya cordata extract (total amount of sanguinarine C 20H14NO4 + and chelerythrine C 21H18NO4 +) should be 90-110% of the labeled amount, and the content of carvacrol should be 90-110% of the labeled amount.
As can be seen from the data in tables 1 to 4, the microemulsion preparation for expelling trichomonas pigeon according to the present invention has stable properties, no obvious change in properties and substantially stable content through an accelerated test at a temperature of 40+ -2 ℃ and a relative humidity of 70% + -5%. In the formula of the comparative example 1, no cosolvent is added to adjust the pH value, the macleaya cordata extract can not be completely dissolved during preparation, the sample solution is reddish brown clear turbid clear liquid, the sample solution is placed through an acceleration test and can be deposited at the bottom, and the content of the sample solution is not in a standard range after the second month of the acceleration test. In the formulation of example 7, the cosolvent is added too much, the macleaya cordata extract is not completely dissolved, and the content is not in the standard range after the third month of the accelerated test.
Test example 2
Pigeon trichomonas killing effect evaluation test:
The microemulsion formulations prepared in examples 1-5 and comparative examples 2-5 were evaluated as follows:
(1) Material preparation
(1.1) Preparation of culture medium: preparing culture medium according to the preparation requirement of liver soup liquid culture medium, sterilizing under high pressure, cooling to room temperature, adding 10% fetal bovine serum into the culture solution, and adding appropriate amount of antibiotics according to 2000IU of each culture solution containing green streptomycin. And subpackaging the prepared culture solution with 2ml centrifuge tubes, subpackaging 1ml of each tube, placing the tube on a centrifuge tube rack, avoiding inversion in the moving process, and ensuring that a tube cover is tightly covered.
(1.2) Sampling: after dipping physiological saline with a sterile cotton swab, rubbing back and forth for 5 times in the oral cavity of the pigeon, putting the cotton swab back into the centrifuge tube, and breaking the cotton swab from the cotton ball position to ensure that the centrifuge tube is covered.
(1.3) Culturing: the collected sample is placed under a 100 times microscope for observation, the oval or pear-shaped transparent insect bodies with swimming in the visual field can be cultivated, the sample is placed into a 37 ℃ incubator for cultivation for 12-24 hours (according to the quantity of trichomonas on the sample), a pipettor is used for blowing and beating the evenly mixed culture solution, 10 mu L of culture solution is sucked out, and the culture solution is dripped on a blood cell counting plate for counting.
(1.4) Drug sensitivity test: trichomonas (long fusiform, fast swimming) cultured to logarithmic growth phase was used for drug susceptibility testing. Divided into 10 groups, examples 1-5, comparative examples 2-5 and blank positive groups, respectively. 30 ten thousand trichomonas were pipetted into 1ml of liver broth, and 2 μl, 4 μl, 6 μl, 8 μl, 10 μl of sample groups were pipetted into the corresponding trichomonas-containing liver broth, respectively, and no drug was added to the blank group. The efficacy was evaluated on the basis of the change in trichomonas count at 1 hour intervals.
The results are shown in Table 5.

As can be seen from the data in table 5: the microemulsion preparation for expelling the trichomonas pigeon has remarkable killing effect on the trichomonas pigeon, and the effect of the macleaya cordata extract and the carvacrol compounded is better than that of single components and other substances compounded, and the effect of killing the trichomonas pigeon can be basically achieved after 1h of the microemulsion preparation.
Test example 3
Animal evaluation test:
the microemulsion formulations prepared in examples 1-3 were evaluated as follows:
(1) Selecting 40 pigeons with trichomonas pigeon, dividing the pigeons into 4 groups, dipping 10 pigeons in each group with sterile cotton swabs into normal saline, rubbing back and forth in the oral cavity of the pigeons for 5 times, putting the cotton swabs back into a centrifuge tube filled with 1ml of normal saline, breaking the cotton swabs from the cotton ball position, and ensuring that the centrifuge tube is covered. The centrifuge tubes were placed in a vortex mixer and mixed for 15 seconds, 10. Mu.L of the solution was aspirated, and the solution was dripped onto a hemocytometer to count, and the average number of trichomonas pigeon in each group was calculated.
(2) Samples were added to the pigeon's daily drinking water in groups as follows:
Group 1: 5ml of example 1 was added per 1L of drinking water for 2 days.
Group 2: 10ml of example 2 was added per 1L of drinking water for 2 days.
Group 3: 2.5ml of example 3 was added per 1L of drinking water for 2 days.
Group 4: the blank group normally drinks water without adding medicine.
(3) After 2 days, sampling and counting are carried out respectively, the average number of trichomonas of each group of pigeons is calculated, and the health condition of the pigeons is observed. The results are shown in Table 6.

From the data in Table 6, it is clear that the microemulsion preparation for expelling trichomonas in pigeons according to the present invention can thoroughly kill trichomonas in pigeons and is safe and non-irritating to pigeons when applied to pigeons in the amount of the usage provided.
Test example 4
Evaluation test of solubility of microemulsion preparation:
In order to prove that the microemulsion preparation for expelling trichomonas pigeon provided by the invention can be diluted with water in any proportion, can obtain clear and transparent solution, is convenient to use, does not block a drinking line, and performs the following tests:
1ml of each of the samples of examples 1 to 3 and comparative example 1 was diluted with tap water in the following ratio by the stepwise addition method, and the dissolution was observed, and the results are shown in Table 7.

From the results in Table 7, it can be seen that: the microemulsion preparation for expelling trichomonas pigeon provided by the invention can be diluted with water in any proportion, so that clear and transparent solution can be obtained, and the microemulsion preparation is convenient to use and can not block a drinking line. The sample solution without the cosolvent is turbid, cannot be diluted with water at will, is clear and transparent, and can be obtained by adding more than 1:100 of water.
The applicant states that the present invention illustrates a microemulsion formulation for the repellency of trichomonas pigeon and a method of preparing and using the same by the above examples, but the present invention is not limited to the above examples, i.e. it does not mean that the present invention must be practiced by relying on the above examples. It should be apparent to those skilled in the art that any modification of the present invention, equivalent substitution of raw materials for the product of the present invention, addition of auxiliary components, selection of specific modes, etc., falls within the scope of the present invention and the scope of disclosure.
The preferred embodiments of the present invention have been described in detail above, but the present invention is not limited to the specific details of the above embodiments, and various simple modifications can be made to the technical solution of the present invention within the scope of the technical concept of the present invention, and all the simple modifications belong to the protection scope of the present invention.
In addition, the specific features described in the above embodiments may be combined in any suitable manner, and in order to avoid unnecessary repetition, various possible combinations are not described further.

Use of a microemulsion formulation in the manufacture of a medicament for preventing or treating trichomonas infection in a pigeon or alleviating the condition of a pigeon infected with trichomonas, or in the manufacture of a pigeon feed additive for alleviating the condition of a pigeon infected with trichomonas, characterised in that the microemulsion formulation consists of: macleaya cordata extract, carvacrol, an emulsifying agent, a cosolvent and water;
Wherein, based on the total weight of the microemulsion preparation, the macleaya cordata extract accounts for 0.5 to 2 percent, the carvacrol accounts for 1 to 4 percent, and the emulsifier accounts for 10 to 30 percent;
the cosolvent is an alkaline solution;
the microemulsion preparation is prepared by the following steps:
(1) Mixing the macleaya cordata extract with 30-50% of water and cosolvent, and stirring until the solution is clear to obtain an aqueous phase mixture, wherein the pH value of the aqueous phase mixture is 1.5-2.0; mixing and stirring carvacrol and an emulsifier to obtain an oil phase mixture;
(2) Mixing the water phase mixture and the oil phase mixture, stirring and emulsifying until the solution is clear, and then mixing with the rest water to obtain the microemulsion preparation.
2. The use according to claim 1, wherein the emulsifier is any one or a combination of at least two of tween 80, EL-40 or tween 20.
3. The use according to claim 2, wherein the emulsifier is a combination of tween 80 and EL-40.
4. Use according to claim 1, characterized in that the alkaline solution is a 1-5% sodium hydroxide solution.
5. The use according to claim 1, wherein the macleaya cordata extract contains not less than 40% of sanguinarine and not less than 60% of total sanguinarine and chelerythrine.
