丙二酸又称缩苹果酸,是一种具有三个碳原子的二羧酸,为无色片状晶体,易溶于水,且溶于丙酮、醇和醚等有机试剂中。丙二酸也是制药和化妆品工业中许多产品和工艺的标准组分,主要用于医药中间体,也用于香料、粘合剂、树脂添加剂、电镀抛光剂等。传统方法上丙二酸的合成,主要是通过氯乙酸与碳酸钠反应制得的钠盐与氰化钠反应生成氰基乙酸盐,然后通过氢氧化钠将腈基水解为丙二酸钠,最后用酸化法获得丙二酸。但这种方法获得的丙二酸纯度不高,回收率低,并且在此过程中所使用的的氰化物为剧毒物质,造成严重的环境污染。目前丙二酸主要通过水解丙二酸二甲酯或丙二酸二乙酯来生产,这种方法虽然可以提高产物的收率和纯度,但是原料成本较高,不适合大规模工业化生产。而生物法制备有机酸具有反应条件温和,绿色环保等优势,但由于对其合成途径及关键酶了解有限,目前生物发酵法制备丙二酸的较少。WO2021133722A1公开了能够产生丙二酸、丙二酸酯、丙二酸酯或其混合物的工程微生物,其中工程改造微生物包括丙二酸-半醛脱氢酶,能够产生约9g/L至约250g/L的丙二酸、丙二酸酯、丙二酸酯或其混合物。所述微生物还包括能够将磷酸烯醇式丙酮酸(PEP)转化为草酰乙酸(OAA)的多肽;能够将丙酮酸转化为草酰乙酸(OAA)的多肽;能够将草酰乙酸 (OAA) 转化为天冬氨酸的多肽;能够将天冬氨酸转化为β丙氨酸的多肽;能够将β丙氨酸转化为丙二酸-半醛或其混合物的多肽。该文献中以草酰乙酸为底物合成丙二酸-半醛也需要经过多个步骤,并不经济。
因此,亟待开发高效的丙二酸生物合成途径及生产菌株,为丙二酸的高效绿色合成提供基础。
发明内容
本发明经过广泛而深入的研究,设计了全新的丙二酸合成途径,并鉴定了途径中所涉及的两个关键酶,酮酸脱羧酶和丙二酸半醛脱氢酶,在体外能够催化草酰乙酸合成丙二酸,并将该新途径导入微生物后,能够以简单碳源葡萄糖为底物合成丙二酸,如此形成了本发明。
为此,本发明采用以下技术方案。
本发明第一方面,提供了一种合成丙二酸的方法,其以草酰乙酸为底物,在体外通过两步酶催化反应合成丙二酸。其特征在于,以草酰乙酸为底物,加入酮酸脱羧酶和丙二酸半醛脱氢酶,进行多酶催化反应合成丙二酸。其中,酮酸脱羧酶催化草酰乙酸生成丙二酸半醛,而后丙二酸半醛脱氢酶催化丙二酸半醛生成丙二酸。
在具体实施方式中,底物草酰乙酸可以是草酰乙酸纯品也可以是采用酸解、酶转化或生物发酵法转化单糖、多糖或其组合获得的草酰乙酸。优选地,所述的单糖选自葡萄糖,木糖,阿拉伯糖或其组合;所述的聚糖选自所述的聚糖包括纤维素、半纤维素、蔗糖、淀粉或其组合,优选的为结晶纤维素和木聚糖,或组合。
在另一优选方式中,底物草酰乙酸可以通过植物生物质发酵转获得,具体地植物生物质包括农作物秸秆、林业废弃物、能源植物或其部分或全部分解产物;其中,所述农作物秸秆包括玉米秸秆,小麦秸秆,水稻秸秆,高粱秸秆,大豆秸秆,棉花秸秆,甘蔗渣,玉米芯;所述林业废弃物包括枝叶,锯末;所述能源植物包括甜高粱,柳枝稷,芒草,芦苇或其组合。
在另一优选方式中,多酶催化反应的反应温度为10-95℃,进一步优选为20-80℃,更优选为30-60℃,最优选为37℃。
在具体实施方式中,多酶催化反应的体系中还加入缓冲液、磷酸盐、金属离子。
在另一优选方式中,所述酮酸脱羧酶如SEQ ID NO.1所示;或其氨基酸序列与SEQID NO.1所示氨基酸序列具有至少60%,优选至少80%,更优选至少90%,最优选至少95%的同一性;或SEQ ID NO.1所示的序列经一个或几个氨基酸的缺失、添加或取代而形成的、具有催化草酰乙酸合成丙二酸半醛的一种或多种由衍生的多肽。
在另一优选方式中,所述丙二酸半醛脱氢酶如SEQ ID NO.3或SEQ ID NO.21所示;或其氨基酸序列与SEQ ID NO.3或SEQ ID NO.21所示氨基酸序列具有至少60%,优选至少80%,更优选至少90%,最优选至少95%的同一性;或SEQ ID NO.3或 SEQ ID NO.21所示的序列经一个或几个氨基酸的缺失、添加或取代而形成的、具有催化草酰乙酸合成丙二酸半醛的一种或多种由衍生的多肽。
本发明第二方面,提供了一种可合成丙二酸的微生物重组菌株的构建方法,其特征在于,在微生物中引入酮酸脱羧酶和丙二酸半醛脱氢酶基因,通过表达所述基因的酶,以将胞内的草酰乙酸催化合成丙二酸。
所述微生物重组菌株的构建方法适用于多种但不限于以下微生物,脉孢菌(Neurospora)、曲霉(Aspergillus)、木霉(Trichoderma)、青霉(Penicillium)、毁丝霉(Myceliophthora)、侧孢霉(Sporotrichum)、镰孢菌(Fusarium)、根霉(Rhizopus)、毛霉(Mucor)和拟青霉(Paecilomyces)。优选地,所述菌株是嗜热毁丝霉。
在具体实施方式中,所述编码酮酸脱羧酶基因编码如SEQ ID NO.1所示氨基酸廒;或编码的氨基酸序列与SEQ ID NO.1所示氨基酸序列具有至少60%,优选至少80%,更优选至少90%,最优选至少95%的同一性;或者编码了SEQ ID NO.1所示的序列经一个或几个氨基酸的缺失、添加或取代而形成的、具有催化草酰乙酸合成丙二酸半醛的一种或多种由衍生的多肽;所述编码丙二酸半醛脱氢酶的基因编码了如SEQ ID NO.3或 SEQ ID NO.21所示的氨基酸序列;或编码了氨基酸序列与SEQ ID NO.3或SEQ ID NO.21所示氨基酸序列具有至少60%,优选至少80%,更优选至少90%,最优选至少95%的同一性;或者编码了SEQ IDNO.3或SEQ ID NO.21所示的序列经一个或几个氨基酸的缺失、添加或取代而形成的、具有催化草酰乙酸合成丙二酸半醛的一种或多种由衍生的多肽。
本发明第三方面,提供了一种发酵法合成丙二酸的方法,其特征在于,将上述所述构建方法得到的可合成丙二酸的微生物重组菌株,以单糖、聚糖和/或植物生物质为底物发酵生产有机酸。
在另一优选方式中,所述的底物还包括单糖、多糖或其组合。优选地,所述的单糖所述的单糖选自葡萄糖,木糖,阿拉伯糖或其组合;所述的聚糖选自所述的聚糖包括纤维素、半纤维素、蔗糖、淀粉或其组合,优选的为结晶纤维素和木聚糖,或组合;所述的植物生物质包括农作物秸秆、林业废弃物、能源植物或其部分或全部分解产物;其中,所述农作物秸秆包括玉米秸秆,小麦秸秆,水稻秸秆,高粱秸秆,大豆秸秆,棉花秸秆,甘蔗渣,玉米芯;所述林业废弃物包括枝叶,锯末;所述能源植物包括甜高粱,柳枝稷,芒草,芦苇或其组合。
本发明还提供酮酸脱羧酶或其编码基因在催化草酰乙酸生成丙二酸半醛和/或在制备丙二酸中的应用,其特征在于,所述酮酸脱羧酶具有如SEQ ID NO.1所示;或其氨基酸序列与SEQ ID NO.1所示氨基酸序列具有至少60%,优选至少80%,更优选至少90%,最优选至少95%的同一性;或者SEQ ID NO.1所示的序列经一个或几个氨基酸的缺失、添加或取代而形成的、具有催化草酰乙酸合成丙二酸半醛的一种或多种由衍生的多肽。
本发明还提供丙二酸半醛脱氢酶或其编码基因在催化丙二酸半醛生成丙二酸和/或在制备丙二酸中的应用,其特征在于,所述丙二酸半醛脱氢酶具有如SEQ ID NO.3或SEQID NO.21所示;或其氨基酸序列与SEQ ID NO.3或SEQ ID NO.21所示氨基酸序列具有至少60%,优选至少80%,更优选至少90%,最优选至少95%的同一性;或者SEQ ID NO.21所示的序列经一个或几个氨基酸的缺失、添加或取代而形成的、具有催化草酰乙酸合成丙二酸半醛的一种或多种由衍生的多肽。
应理解,在本发明范围内中,本发明的上述各技术特征和在下文(如实施例)中具体描述的各技术特征之间都可以互相组合,从而构成新的或优选的技术方案。限于篇幅,在此不再一一累述。

为更进一步阐述本发明所采取的技术手段及其效果,以下结合本发明的优选实施例来进一步说明本发明的技术方案,应理解,这些实施例仅用于说明本发明而不用于限制本发明的范围。
下述实施例中所用方法如无特别说明均为常规方法,例如Sambrook 等人所著的《分子克隆:实验室手册》(New York: Cold SpringHarbor Laboratory Press,1989)中所述的条件,或按照制造厂商所建议的条件。所用试剂或仪器未注明生产厂商者,均为可通过正规渠道商购获得的常规产品。
下面结合具体实施例来进一步描述本发明,本发明的优点和特点将会随着描述而更为清楚。但是应理解所述实施例仅是范例性的,不对本发明的范围构成任何限制。本领域技术人员应该理解的是,在不偏离本发明的精神和范围下可以对本发明技术方案的细节和形式进行修改或替换,但这些修改或替换均落入本发明的保护范围。
实施例1、丙二酸合成途径的设计及体外测试
根据前期研究基础,本发明设计了全新的丙二酸合成途径(图1所示),该途径中将草酰乙酸经过两步反应合成丙二酸,其中所涉及的关键酶与关键步骤包括:(1)草酰乙酸脱羧酶,用于催化草酰乙酸生成丙二酸半醛;(2)丙二酸半醛脱氢酶,用于催化丙二酸半醛生成丙二酸。随后发明人针对数据库中注释为酮酸脱酸酶以及琥珀酸半醛脱氢酶的序列进行了筛选,最后获得了关键酶,使得丙二酸合成新途径得以在体内和体外进行验证完成,具体如下:
其中酮酸脱羧酶基因kde委托江苏金唯智生物技术有限公司合成来后直接连接至质粒pET-21a(+)中;丙二酸半醛脱氢酶基因Msh片段或丙二酸半醛脱氢酶基因MDE片段,以大肠杆菌基因组为模板,用引物MSH-F/R,或MDE-F/R通过PCR获得,而后连接入质粒pET-21a(+)中,将这两个质粒分别转化至大肠杆菌表达菌BL21(DE3) (Invitrogen ,Carlsbad,CA)中,进行蛋白表达与纯化。

PCR反应体系为:2×Phanta® Max Buffer 25μL,10 mM dNTPs 1μL,上游/下游引物各1.5μL,模板1μL,Phanta® Max Super-Fidelity DNA Polymerase 1μL,水19μl。
PCR反应条件为:先95 ºC 30s;然后95℃ 15s,58ºC 1s,72℃ 45s,32个循环;最后72 ºC 5min,4 ºC 10min。
以草酰乙酸(OAA)为底物,酶反应组分如表所示,在37ºC下检测OD340值变化:
结果显示体外酶反应液Kde+Mde(图2)和酶反应液Kde+Msh(图3),在340nm下吸光值随时间持续升高,说明两组酶均能催化草酰乙酸合成丙二酸;而后通过GC-MS检测检测了酶反应液(Kde+Msh)的产物,进一步证实了反应液中有丙二酸生成(图4和图5)。
综合可知,利用所设计的路线及所选用的酶,能够催化草酰乙酸合成丙二酸,证实草酰乙酸脱羧酶Kde能够催化草酰乙酸(OAA)为丙二酸半醛,丙二酸半醛脱氢酶Msh和MDE都能够催化第二反应,以丙二酸半醛为底物合成丙二酸。
实施例2 嗜热毁丝霉重组菌株的构建
通过PCR扩增分别获得组成型启动子Ppgk片段和Msh片段,过Gibson Assembly的方法连接到由限制性内切酶BiglII和XbaI酶切的线性化载体pAN52-TB-Intron(Liu Q, LiJ, Ying S, Wang J, Sun W, Tian C, Feng M. 2014. Unveiling equal importance oftwo 14-3-3 proteins for morphogenesis, conidiation, stress tolerance andvirulence of an insect pathogen. Environ Microbiol. doi: 10.1111/1462-2920.12634),获得Msh过表达载体pAN52-Ppgk-MSH。
类似的,通过PCR扩增分别获得组成型启动子Pap片段和kde片段,过GibsonAssembly的方法连接到由限制性内切酶BiglII和XbaI酶切的线性化载体pAN52-TB-Intron,获得kde过表达载体pAN52-Pap-kde。
将表达载体pAN52-Ppgk-MSH和pAN52-Pap-kde线性化后,通过原生体质转化的方法同时导入嗜热毁丝霉ATCC 42464中,随后PCR验证所获得的转化子,获得阳性嗜热毁丝霉重组菌株SYG01。

实施例3嗜热毁丝霉重组菌株在丙二酸合成中的应用
将嗜热毁丝霉重组菌株SYG01接种于丙二酸发酵培养基中葡萄糖75 g/L,蛋白胨6.0 g/L,0.15 g/L KH2PO4,0.15 g/L K2HPO4,0.10 g/L CaCl2•2H2O,0.10 g/L MgSO4•7H2O,10.0 g/L碳酸钙,1 ml/L 微量元素液(5g NaCl,5 g FeSO4•7H2O,1 g citric acid / L),在45ºC条件,培养2天后,检测培养基中丙二酸含量,GC-MS检测结果显示在培养基含有丙二酸,说明在嗜热毁丝霉体内成功构建了本发明所设计的丙二酸合成途径,能够转化葡萄糖合成丙二酸。

Claims (9)
1.一种合成丙二酸的方法,其特征在于,以草酰乙酸为底物,利用酮酸脱羧酶和丙二酸半醛脱氢酶催化合成丙二酸。
2.如权利要求1所述的方法,其特征在于,所述底物草酰乙酸为草酰乙酸纯品,或采用酸解、酶转化或植物生物质发酵法转化单糖、多糖或其组合衍生获得产物中的草酰乙酸;进一地,所述的单糖,选自葡萄糖、木糖、阿拉伯糖,或其组合;所述的聚糖,选自结晶纤维素、半纤维素,或其组合;所述的植物生物质,选自玉米秸秆,小麦秸秆,水稻秸秆,高粱秸秆,大豆秸秆,棉花秸秆,甘蔗渣,玉米芯,优选为玉米芯。
3.如权利要求1-2任一所述的方法,其特征在于,所述酮酸脱羧酶如SEQ ID NO.1所示;或其氨基酸序列与SEQ ID NO.1所示氨基酸序列具有至少60%,优选至少80%,更优选至少90%,最优选至少95%的同一性;或者SEQ ID NO.1所示的序列经一个或几个氨基酸的缺失、添加或取代而形成的、具有催化草酰乙酸合成丙二酸半醛的一种或多种由衍生的多肽;所述丙二酸半醛脱氢酶如SEQ ID NO.3或SEQ ID NO.21所示;或其氨基酸序列与SEQ IDNO.3或SEQ ID NO.21所示氨基酸序列具有至少60%,优选至少80%,更优选至少90%,最优选至少95%的同一性;或者SEQ ID NO.21所示的序列经一个或几个氨基酸的缺失、添加或取代而形成的、具有催化草酰乙酸合成丙二酸半醛的一种或多种由衍生的多肽。
4.一种构建可生产丙二酸重组菌株的方法,其特征在于,在菌株中导入编码酮酸脱羧酶和后丙二酸半醛脱氢酶的基因,所获得的能够合成丙二酸的重组菌株。
5.如权利要求4所述的方法,其特征在于,所述菌株为丝状真菌细胞,选自脉孢菌(Neurospora)、曲霉(Aspergillus)、木霉(Trichoderma)、青霉(Penicillium)、毁丝霉(Myceliophthora)、侧孢霉(Sporotrichum)、镰孢菌(Fusarium)、根霉(Rhizopus)、毛霉(Mucor)和拟青霉(Paecilomyces),更优选地,所述毁丝霉菌选自嗜热毁丝霉(Myceliophthora thermophila)、异梭毁丝霉(Myceliophthora heterothallica)。
6.如权利要求5所述的方法,其特征在于,所述编码酮酸脱羧酶基因编码如SEQ IDNO.1所示氨基酸廒;或编码的氨基酸序列与SEQ ID NO.1所示氨基酸序列具有至少60%,优选至少80%,更优选至少90%,最优选至少95%的同一性;或者编码了SEQ ID NO.1所示的序列经一个或几个氨基酸的缺失、添加或取代而形成的、具有催化草酰乙酸合成丙二酸半醛的一种或多种由衍生的多肽;所述编码丙二酸半醛脱氢酶的基因编码了如SEQ ID NO.3或SEQ ID NO.21所示的氨基酸序列;或编码了氨基酸序列与SEQ ID NO.3或SEQ ID NO.21所示氨基酸序列具有至少60%,优选至少80%,更优选至少90%,最优选至少95%的同一性;或者编码了SEQ ID NO.21所示的序列经一个或几个氨基酸的缺失、添加或取代而形成的、具有催化草酰乙酸合成丙二酸半醛的一种或多种由衍生的多肽。
7.一种发酵生产丙二酸的方法,其特征在于,利用如权利要求4至6任一项所述方法获得的重组菌株以单糖、聚糖和/或植物生物质为底物发酵生产丙二酸;优选地,所述的单糖,选自葡萄糖、木糖、阿拉伯糖,或其组合;所述的聚糖,选自结晶纤维素、半纤维素,或其组合;所述的植物生物质,选自玉米秸秆,小麦秸秆,水稻秸秆,高粱秸秆,大豆秸秆,棉花秸秆,甘蔗渣,玉米芯,优选为玉米芯。
8.酮酸脱羧酶或其编码基因在催化草酰乙酸生成丙二酸半醛和/或在制备丙二酸中的应用,其特征在于,所述酮酸脱羧酶具有如SEQ ID NO.1所示;或其氨基酸序列与SEQ IDNO.1所示氨基酸序列具有至少60%,优选至少80%,更优选至少90%,最优选至少95%的同一性;或者SEQ ID NO.1所示的序列经一个或几个氨基酸的缺失、添加或取代而形成的、具有催化草酰乙酸合成丙二酸半醛的一种或多种由衍生的多肽。
9.丙二酸半醛脱氢酶或其编码基因在催化丙二酸半醛生成丙二酸和/或在制备丙二酸中的应用,其特征在于,所述丙二酸半醛脱氢酶具有如SEQ ID NO.3或SEQ ID NO.21所示;或其氨基酸序列与SEQ ID NO.3或SEQ ID NO.21所示氨基酸序列具有至少60%,优选至少80%,更优选至少90%,最优选至少95%的同一性;或者SEQ ID NO.21所示的序列经一个或几个氨基酸的缺失、添加或取代而形成的、具有催化草酰乙酸合成丙二酸半醛的一种或多种由衍生的多肽。
