Detailed Description
The invention provides a nutrition composition for pregnant and lying-in women, which comprises the following raw materials in parts by mass: 28 to 50 parts of protein, 0.2 to 1 part of mineral nutrition, 0.2 to 0.6 part of nutrition supplement, 8 to 23 parts of prebiotics, 7 to 18 parts of skim milk powder, 5 to 15 parts of water-soluble starch and 20 to 35 parts of glucose;
the nutritional supplement comprises taurine, folic acid and DHA algae powder.
The protein of the present invention preferably includes a vegetable protein and an animal protein, and the mass ratio of the vegetable protein to the animal protein is preferably (25 to 35): (3 to 15), more preferably (28 to 35): (3 to 10), more preferably (28 to 30): (5-8), most preferably 30:5. The vegetable protein of the present invention preferably comprises soy protein isolate that is readily absorbable by the human body, and the animal protein preferably comprises whey protein.
The mineral nutrition according to the invention preferably comprises calcium, iron and zinc, and the calcium is preferably derived from calcium lactate, the iron is preferably derived from ferric pyrophosphate, and the zinc is preferably derived from zinc gluconate. The mass ratio of the calcium lactate, the ferric pyrophosphate and the zinc gluconate is preferably (0.25-0.95): (0.038-0.088): (0.01 to 0.018), more preferably (0.25 to 0.8): (0.038-0.068): (0.01 to 0.0178), more preferably (0.5 to 0.8): (0.038-0.048): (0.01-0.0175), most preferably 0.5:0.048:0.0175.
The weight ratio of taurine, folic acid and DHA algae powder is preferably (0.11-0.14): (0.0003 to 0.0006): (0.1 to 0.45), more preferably (0.13 to 0.14): (0.0004 to 0.0006): (0.1 to 0.25), more preferably (0.13 to 0.135): (0.0005 to 0.0006): (0.1 to 0.15), most preferably 0.135:0.0005:0.15.
The prebiotics according to the invention preferably comprise fructo-oligosaccharides and galacto-oligosaccharides; and the mass ratio of the fructo-oligosaccharide and the galacto-oligosaccharide is preferably (3-10): (5 to 13), more preferably (5 to 10): (8 to 13), more preferably (5 to 7): (8-10), most preferably 7:9.
The nutritional composition is nutritional powder for pregnant and lying-in women, each raw material is nutritional elements required by the pregnant and lying-in women, and two large prebiotics are added specifically aiming at the trouble of constipation during pregnancy.
The invention also provides a preparation method of the nutritional composition, which comprises the following steps: mixing protein, mineral nutrition, nutritional supplement, prebiotics, skimmed milk powder, water soluble starch and glucose, and packaging to obtain the nutritional composition.
The raw materials are preferably screened to remove impurities, and then mixed and stirred, wherein the stirring speed is preferably 20r/min, and the stirring time is preferably 30min.
The invention divides the stirred semi-finished product into sub-packages, for example, when the semi-finished product is divided into sub-packages according to the specification of 15 g/bag, one bag is used once, and 1 time a day.
The invention also provides application of the nutritional composition in preparing a solid beverage rich in nutrition of pregnant and lying-in women.
The nutritional composition (or called pregnant and lying-in women nutritional powder) is safe and nontoxic, and in the embodiment, feed addition experiments are carried out by taking zebra fish as an example, and the embodiment proves that the MTC of the pregnant and lying-in women nutritional powder to 2 month old zebra fish is 1.0mg/ml and LD 10 0.33-3.25 mg/ml and LD 50 7.45-48.33 mg/ml; the pregnant and lying-in women nutrition powder with the concentration of 0.25mg/ml, 0.5mg/ml and 1.0mg/ml is added into the feed, so that the weight gain rate, the specific growth rate and the feed efficiency of the zebra fish of 2 months old can be obviously improved; the pregnant and lying-in woman nutrition powder with the concentration of 0.50mg/ml and 1.0mg/ml is added into the feed, so that the gonadal index of the zebra fish of 2 months old can be obviously improved; the feed is added with nutrition powder for pregnant and lying-in women, and the content of 17 amino acids and the content of essential amino acids (P) in the back muscle of the zebra fish of 2 months age are obviously improved<0.05 0.50mg/ml and 1.0mg/ml of the nutrition powder for pregnant and lying-in women has the best effect of improving essential amino acids, and no difference (P)>0.05)。
For further explanation of the present invention, a nutritional composition for pregnant and parturients, a preparation method and application thereof, provided by the present invention, will be described in detail with reference to the accompanying drawings and examples, which should not be construed as limiting the scope of the present invention.
Example 1
30g of soybean protein isolate, 10g of skim milk powder, 5g of whey protein, 9g of galacto-oligosaccharide, 7g of fructo-oligosaccharide, 0.0005g of folic acid, 0.15g of DHA algae oil, 0.135g of taurine, 0.5g of calcium lactate, 0.048g of ferric pyrophosphate, 0.0175g of zinc gluconate, 8.149g of water-soluble starch and 30g of glucose are mixed in a three-dimensional mixer for 30min to obtain a semi-finished product to be packaged.
Example 2
28g of soybean protein isolate, 8g of skim milk powder, 10g of whey protein, 8g of galacto-oligosaccharide, 5g of fructo-oligosaccharide, 0.0003g of folic acid, 0.11g of DHA algae oil, 0.135g of taurine, 0.5g of calcium lactate, 0.048g of ferric pyrophosphate, 0.0175g of zinc gluconate, 10.1892g of water-soluble starch and 30g of glucose are mixed in a three-dimensional mixer for 30min to obtain a semi-finished product to be packaged.
Example 3
25g of soybean protein isolate, 10g of skim milk powder, 10g of whey protein, 8g of galacto-oligosaccharide, 8g of fructo-oligosaccharide, 0.0005g of folic acid, 0.130g of taurine, 0.6g of calcium lactate, 0.055g of ferric pyrophosphate, 0.0155g of zinc gluconate, 9.199g of water-soluble starch and 29g of glucose are mixed in a three-dimensional mixer for 30min to obtain a semi-finished product, and the semi-finished product is packaged.
Example 4
30g of soybean protein isolate, 10g of skim milk powder, 5g of whey protein, 9g of galacto-oligosaccharide, 9g of fructo-oligosaccharide, 0.0005g of folic acid, 0.15g of DHA algae oil, 0.135g of taurine, 0.5g of calcium lactate, 0.048g of ferric pyrophosphate, 0.0175g of zinc gluconate, 6.077g of water-soluble starch and 30g of glucose are mixed in a three-dimensional mixer for 30min to obtain a semi-finished product to be packaged.
Animal experiments were carried out using the maternal nutrition powder prepared in example 1
1. Laboratory animals and feeding management
Wild type zebra fish AB strain, 2 months old, 150 zebra fish were put into a zebra fish culture system (14 h light, 10h dark), and 30 zebra fish with 2 months old were placed in each culture unit.
Feeding nutrition powder feed for pregnant and lying-in women with different concentrations according to 0.3% of the body weight of the zebra fish, and simultaneously setting a negative control group (feed without the nutrition powder for the pregnant and lying-in women) and a positive control group (positive control group only feeds artemia). Feeding for three times every day, feeding a proper amount of the bait to enable the bait to be ingested freely, observing ingestion condition within 20min, fishing out residual bait after feeding for 1h, drying and deducting. The feeding period was 42 days, and water and dissolved oxygen were recorded daily.
2. Experimental method
The experiment sets the adding level of 3 nutrition powder for pregnant and lying-in women, prepares the nutrition powder solution for pregnant and lying-in women according to the concentration of 1/4MTC (0.25 mg/ml), 1/2MTC (0.50 mg/ml) and MTC (1.0 mg/ml), and evenly mixes the 3 concentration solutions with feed (Shandong's fish feed research center), and air-dries in the shade at low temperature.
After MTC determination (1.0 mg/ml), maternal nutrition powder feed was prepared according to maximum tolerance of 1/4MTC (0.25 mg/ml), 1/2MTC (0.5 mg/ml) and MTC (1 mg/ml), and a negative control group and a positive control group were set.
After 42 days of culture, each treated zebra fish was collected and sacrificed with MS-222 anesthetic, and the procedure for the anesthesia and the sacrifice met the requirements of the American veterinary Association (AMVA) for the specification of the anesthesia and the sacrifice of animals. Distilled water is washed 3 times for 1min each time. The washed zebra fish sample is dried by paper towel and then weighed, and the body length is measured while photographing.
After weighing, zebra fish were dissected on ice, the gonads of each treated zebra fish were collected and weighed to calculate sex-to-body ratio.
The back muscle above the lateral line of zebra fish is placed in an EP tube for preservation at-20deg.C.
The muscle of the back of zebra fish of each treatment group was weighed and then the moisture of the samples was determined by direct drying according to the national standard of GB 5009.3-2016.
The dried sample was ground and crushed in a mortar, and then 17 amino acid contents in zebra fish muscle were measured in GB 5009.124-2016 national standard using an amino acid automatic analyzer L-8900, tryptophan was destroyed during acid hydrolysis, and not otherwise measured. Amino acid content is a percentage of fresh mass.
3. Statistical analysis
The data were analyzed using the spss18.0 One-way ANOVA method and Duncan's comparison, the results were expressed as mean ± standard error, P <0.05 indicated significant differences and statistical significance.
2. Experimental results
1. Influence of maternal nutrition powder on growth and gonad index of zebra fish
The nutrition powder of pregnant and lying-in women with different concentrations is fed to the zebra fish for 42 days, and the zebra fish has no death condition. And collecting a sample to detect the influence of the nutrition powder of the pregnant and lying-in women on the growth performance and gonad index of the zebra fish.
As shown in fig. 1 and 2, the effect of the nutrition powder of the pregnant and lying-in women on the feeding rate of the zebra fish is not obvious, and the negative control group and the nutrition powder adding group of the pregnant and lying-in women have no obvious difference (P > 0.05), so that the effect of the nutrition powder of the pregnant and lying-in women on the feeding rate of the zebra fish can be presumed. The positive control group is fed with artemia, and the comparative feeding rate between the artemia and the feed has no great significance because of the difference between the artemia and the feed.
Feed efficiency reflects the quality and utilization of feed. The higher the feed efficiency, the higher the feed utilization. After the nutrition powder for pregnant and lying-in women is added, the feed efficiency is obviously increased along with the increase of the additive amount (P < 0.05), wherein the 1.0mg/ml group feed efficiency is highest, and the 0.25mg/ml group is combined with 0.5mg/ml group, and the feed efficiency is obviously higher than that of a negative control group and a positive control group (P < 0.05). The pregnant and lying-in women nutrition powder is added into the feed, so that the weight gain rate, the specific growth rate and the feed efficiency of the zebra fish can be obviously improved, and the growth performance is obviously improved along with the increase of the addition proportion.
The nutrition powder for pregnant and lying-in women has obvious influence on the gonadal development and index of the zebra fish. As shown in fig. 3, the addition of the nutrition powder for pregnant and parturient women increases the volume of the zebra fish egg nest. The gonadal index of the 0.25mg/ml, 0.5mg/ml and 1.0mg/ml groups were significantly higher than that of the negative and positive control groups (P < 0.05). The pregnant and lying-in woman nutrition powder with the concentration of 0.25mg/ml, 0.5mg/ml and 1.0mg/ml can be added into the feed to obviously promote the gonadogenesis of the zebra fish, wherein the group with the concentration of 0.5mg/ml and 1.0mg/ml has the best effect.
2. Influence of nutrition powder of pregnant and lying-in women on muscle amino acid content of zebra fish
The amino acid composition and content of the back muscle of the zebra fish treated by the nutrition powder of pregnant and lying-in women with different concentrations are measured, and the results are shown in fig. 4 and table 1. The results show that: the addition of the nutrition powder for pregnant and lying-in women has remarkable improvement effect on the total amino acid and essential amino acid content of fish bodies, and the total amino acid and essential amino acid content is remarkably improved (P < 0.05) along with the increase of the concentration of the nutrition powder for pregnant and lying-in women.
Of the 17 amino acids in the 5 treatment groups, lysine, phenylalanine, methionine, threonine, isoleucine, leucine and valine were essential amino acids, with arginine and histidine being the essential (semi-essential) amino acids. The content of 7 essential amino acids and conditional essential amino acids is obviously higher than that of the control group and the positive group (P < 0.05) along with the increase of the concentration of the nutrition powder of the pregnant and lying-in women, wherein the content of the essential amino acids in the group with 1.0mg/ml is the highest, but the essential amino acids in the group with 1.0mg/ml and the group with 0.5mg/ml are not obviously different.

Note that: the letter differences indicate that there is a significant difference (P < 0.05).
In summary, the nutrition powder for pregnant and lying-in women can be obtained by using the invention:
1. the MTC of the nutrition powder for pregnant and lying-in women to the 2-month-old zebra fish is 1.0mg/ml. LD (laser diode) 10 0.33-3.25 mg/ml and LD 50 7.45-48.33 mg/ml.
2. The pregnant and lying-in women nutrition powder with the concentration of 0.25mg/ml, 0.5mg/ml and 1.0mg/ml is added into the feed, so that the weight gain rate, the specific growth rate and the feed efficiency of the zebra fish of 2 months old can be obviously improved.
3. The pregnant and lying-in woman nutrition powder with the concentration of 0.50mg/ml and 1.0mg/ml can obviously improve the gonadal index of the zebra fish of 2 months of age.
4. The feed can be added with the nutrition powder for pregnant and lying-in women, which can obviously improve the content of 17 amino acids and the content of essential amino acids (P is less than 0.05) of the back muscles of the zebra fish of 2 months of age, and the nutrition powder for pregnant and lying-in women with 0.50mg/ml and 1.0mg/ml has the best effect of improving the essential amino acids, and has no difference (P is more than 0.05).
Although the foregoing embodiments have been described in some, but not all, embodiments of the invention, it should be understood that other embodiments may be devised in accordance with the present embodiments without departing from the spirit and scope of the invention.
Claims (10)
1. The nutritional composition for the pregnant and lying-in women is characterized by comprising the following raw materials in parts by mass: 28 to 50 parts of protein, 0.2 to 1 part of mineral nutrition, 0.2 to 0.6 part of nutrition supplement, 8 to 23 parts of prebiotics, 7 to 18 parts of skim milk powder, 5 to 15 parts of water-soluble starch and 20 to 35 parts of glucose;
the nutritional supplement comprises taurine, folic acid and DHA algae powder.
2. The nutritional composition according to claim 1, wherein the protein comprises a vegetable protein and an animal protein in a mass ratio of (25-35): (3-15).
3. Nutritional composition according to claim 1 or 2, characterized in that the proteins comprise soy protein isolate and whey protein.
4. The nutritional composition according to claim 1, wherein the mineral nutrition comprises calcium, iron and zinc.
5. The nutritional composition according to claim 4, wherein the calcium is derived from calcium lactate, the iron is derived from ferric pyrophosphate, and the zinc is derived from zinc gluconate;
the mass ratio of the calcium lactate to the ferric pyrophosphate to the zinc gluconate is (0.25-0.95): (0.038-0.088): (0.01-0.018).
6. The nutritional composition according to claim 1, wherein the mass ratio of taurine, folic acid and DHA algae powder is (0.11-0.14): (0.0003 to 0.0006): (0.1-0.45).
7. The nutritional composition according to claim 1, wherein the prebiotic comprises fructo-oligosaccharides and galacto-oligosaccharides;
the mass ratio of the fructo-oligosaccharide to the galacto-oligosaccharide is (3-10): (5-13).
8. A method of preparing a nutritional composition according to any one of claims 1 to 7, comprising the steps of: mixing protein, mineral nutrition, nutritional supplement, prebiotics, skimmed milk powder, water soluble starch and glucose, and packaging to obtain the nutritional composition.
9. The method of claim 8, wherein the dispensing comprises dispensing at a 5 g/bag format.
10. Use of a nutritional composition according to any one of claims 1 to 7 for the preparation of a solid beverage enriched in maternal nutrition.
