Description

Composition for promoting digestion and preparation method of beverage thereof
Technical Field
The invention belongs to the technical field of traditional Chinese medicine compositions, and particularly relates to a composition for promoting digestion and a preparation method of a beverage thereof.
Background
Dyspepsia is a disease caused by gastric motility disorder, a common internal digestive system disease, and also includes gastroparesis and esophageal reflux disease, which have poor gastric motility. The dyspepsia is wide in morbidity population, the symptoms of the dyspepsia are mainly manifested by intermittent epigastric discomfort or pain, early satiety, abdominal distension, acid regurgitation, eructation and indigestion, the patient is unwilling to eat or eat less as much as possible due to discomfort such as chest distress, early satiety, abdominal distension and the like, and many patients are accompanied with mental symptoms such as insomnia, anxiety, depression, headache, inattention and the like. In some patients, borborygmus is in the abdomen, and increased air discharge, and even undigested food residues are often found in the excrement.
At present, the types of western medicines for treating dyspepsia are various, and the western medicines are known to have quick response and good curative effect, but have certain side effects and cannot be taken for a long time. Functional dyspepsia belongs to the categories of epigastric fullness, stomachache, epigastric upset and the like in the traditional Chinese medicine, the diseases of the functional dyspepsia are in the stomach, relate to organs such as the liver and the spleen, and are treated according to syndrome differentiation, and the treatment methods of strengthening the spleen and the stomach, soothing the liver and regulating qi, promoting digestion and removing food stagnation and the like are performed. In the treatment with the traditional Chinese medicine, the traditional Chinese medicine has small toxic and side effects, and has the effects of nourishing the stomach and tonifying the spleen after being taken for a long time, so that the traditional Chinese medicine is gradually accepted by more people. The researches on traditional Chinese medicine and traditional Chinese medicine for aiding digestion and improving gastrointestinal motility are increased year by year, and all the researches have unique points. According to the five internal organs corresponding to the five elements in traditional Chinese medicine, the treatment of dyspepsia is not only limited to stomach, but also relates to intestinal tract. Liver and spleen.

The erythritol is purchased from Shanghai Michelin Biotechnology Limited and has the product number of SJ-E808834; the sorbitol is purchased from Shanghai Michelin Biotechnology, inc. under the product number D817607. Radix pseudostellariae: sweet, slightly bitter and mild in nature and taste. It is good for children's sweating due to deficiency, and is often combined with Bei Sha Shen, bai Dou and shan Yao to treat stomach weakness and poor appetite.
Kapok: is dried flower of bombax ceiba of bombax family, has sweet taste and cool property, and can be used for treating diarrhea, dysentery and hemorrhoid hemorrhage. Flos Bombacis Malabarici has effects of invigorating stomach, flavoring intestinal tract, and relieving diarrhea and chronic gastrointestinal abdominal distention. Common bombax flower and dried orange peel have the effects of tonifying spleen, clearing damp and nourishing stomach when eaten together.
White atractylodes rhizome: bitter, sweet and warm in nature. It enters spleen and stomach meridians. Is a deficiency tonifying medicine and is used for treating spleen deficiency, anorexia, abdominal distention and diarrhea. The bighead atractylodes rhizome has the functions of regulating spleen and stomach and preventing gastric ulcer, can be used for helping to improve the gastrointestinal dysfunction, regulating intestines and stomach and accelerating the digestion and absorption of food; after being taken, the health-care food can protect gastric mucosa, reduce food, stimulate the gastric mucosa, promote the secretion of gastric juice and accelerate the digestion and absorption of food.

Experiment 2 animal experiment-comparison of Small intestine Impulse Rate and stomach evacuation Rate
Test samples: drink test animals prepared with the compositions of examples 1-3 and comparative examples 1-6: 132 SPF-class mice, each half of male and female, weighing 18. + -.2 g, were randomly divided into 11 groups of 12 mice each, blank control group, model group, experimental group (examples 1-3 and comparative examples 1-6).
Constructing a Functional Dyspepsia (FD) mouse model of spleen-stomach qi deficiency. Mice develop the following symptoms: emotional agitation and irritability; squinting (eyes are only exposed by more than half); loss of appetite and decreased appetite; slow, even reduced, weight gain; bunching, bowing back and laziness; the hair color is dark yellow, and the hair is rough and disordered; when the excrement is dry, the excrement is thin, and the reaction of drawing the tail and discharging the excrement is positive; listlessness, lassitude and others indicate successful molding.
The Blank (BC) group was gavaged daily with 0.2ml of normal saline for 15 days.
Except for the blank group, the rest groups adopt diet failure (diet for daytime), exercise and qi consumption (swimming for 30 min) and feeding senna leaves for 0.4g/d (bitter and cold for purgation, qi consumption and qi exhaustion). Before daily feeding, the medicine is taken according to the body weight, the medicine is taken after swimming, the medicine is taken for a fixed time every day, and the stomach is continuously infused for 15 days to prepare a functional dyspepsia mouse model for deficiency of spleen-stomach qi.
The experimental method comprises the following steps: after the administration on the 29 th day, the mice in each group were administered with 0.5mL of powdered carbon semisolid paste (powdered carbon and peanut powder blended in a ratio of 1: 4) after fasting for 24 h. After 1h, the cells were sacrificed and the relevant organs and serum were collected and stored at-80 ℃. Anesthetizing the mouse, collecting blood from eyeball, standing in 1.5mL centrifuge tube at room temperature for 1h, centrifuging at 3000r/min at 4 deg.C for 15min to obtain serum, subpackaging, storing, and storing in-80 deg.C refrigerator for use. Immediately after blood collection, the mice were sacrificed by dislocation of cervical vertebrae, the abdomen of the mice was cut with surgical scissors, and the liver, stomach, small intestine, colon contents and colon were collected. Dissecting and ligating the cardia and pylorus of the stomach, taking the complete stomach, wiping the stomach with filter paper, weighing the mass as the mass of the whole stomach, then cutting the stomach body along the small curve of the stomach, washing the stomach contents with physiological saline, drying the stomach contents with the filter paper, weighing the mass as the mass of the empty stomach, and calculating the gastric emptying rate by using the difference of the weighed masses 2 times before and after. The entire small intestine will be removed from the pylorus to the ileocecal region. The total length of the small intestine and the moving distance of the carbon powder semisolid paste in the small intestine are respectively measured by a steel ruler, and the small intestine propulsion rate is calculated according to the length ratio of 2 times.
The results of the measurements of the rate of small intestine propulsion and the rate of gastric emptying are shown in Table 4.
The results of mouse feeding are shown in FIGS. 1 to 5, in which FIGS. 1 to 2 are mice of blank group and model group, respectively, and FIGS. 1 to 3 are mice of examples 1 to 3, respectively. After the administration of the mice for 29 days, the feces of the mice of examples 1-3 were returned from wet to a relatively dry, granulated state, indicating that the effect of promoting diuresis was good.
