Abstract
本发明涉及生物医药技术领域,更具体的说涉及一种中草药抗病毒槟榔及槟榔添加剂制备方法,该槟榔制品由槟榔、柠檬酸、陈醋、壳聚糖、活性氧化锌、独居石煅烧蒸馏水、牛骨蛋白肽、脂肪酶、低聚异麦芽糖、大豆卵磷脂、人参、白术、茯苓、甘草、黄芪、三七、陈皮、杜仲叶、香茅草、毛霉菌制备而成。试验研究表明,本发明槟榔制品具有很好的抗病毒等作用,对口腔疾病有很好的预防和治疗作用;致癌研究表明,本发明的槟榔制品比普通槟榔制品致癌性显著降低。

技术领域
本发明涉及生物医药技术领域,更具体的说涉及一种中草药抗病毒槟榔及槟榔添加剂制备方法。
技术背景
医学界认为经常嚼食槟榔会造成口腔溃疡、牙龈退变、黏膜下纤维化,进而导致口腔癌变。槟榔果中的槟榔素和槟榔碱具有潜在的致癌性,这些物质包括槟榔生物碱、槟榔鞣质、槟榔特异性亚硝胺和活性氧等。
槟榔还含有20多种微量元素,其中11种为人体必需的微量元素。槟榔种子含总生物0.3%-0.6%,主要为槟榔碱,并含有少量槟榔次碱、去甲基槟榔碱、异去甲基槟榔次碱、槟榔副碱及高槟榔碱等,均与鞣酸集合存在。还有鞣质、脂肪、甘露醇、半乳糖、蔗糖、儿茶精、表二茶精、无色花青素、槟榔红色素、皂苷及多种原矢车菊素的二聚体、三聚体、四聚体等。
据药监部门透露,我国有225个药品含有槟榔。传统医学认为,槟榔具有“杀虫,破积,降气行滞,行水化湿”的功效,曾被用来治疗绦虫、钩虫、蛔虫、绕虫、姜片虫等寄生虫感染。
槟榔作为一种传统食品,深受广大群众喜爱,已经形成了千亿市场,由于存在危害口腔健康和致癌的问题,2021年9月17日,中国国家广播电视总局下发通知,要求停止利用广播电视和网络视听节目宣传推销槟榔及其制品。
科学研究也需要取长补短,把有益的物质保留,把有害的物质去除,把有害变成有益,研发一种具有抗病毒功能的中草药槟榔和槟榔添加剂,改善、改良现有工艺流程和配方,这种槟榔可以杀灭多种有害细菌、灭活多种病毒,除了可以预防和治疗各种口腔疾病,槟榔汁吞服,还可以起到预防癌症的作用,适量咀嚼槟榔,保证黏膜不会发炎,实现非药物预防和治疗疾病的目的,有着巨大的市场价值和深远意义。
发明内容
基于上述原因,申请人经过若干次创造性的劳动,获得了得到一种新的槟榔制品,该槟榔制品由槟榔、柠檬酸、陈醋、壳聚糖、活性氧化锌、独居石煅烧蒸馏水、牛骨蛋白肽、脂肪酶、低聚异麦芽糖、大豆卵磷脂、人参、白术、茯苓、甘草、黄芪、三七、陈皮、杜仲叶、香茅草、毛霉菌制备而成。试验研究表明,本发明槟榔制品具有很好的抗病毒等作用,对口腔疾病有很好的预防和治疗作用;致癌研究表明,本发明的槟榔制品比普通槟榔制品致癌性显著降低。
本发明通过下述技术方案实现的。
一种槟榔制品,该槟榔制品由槟榔、柠檬酸、陈醋、壳聚糖、活性氧化锌、独居石煅烧蒸馏水、牛骨蛋白肽、脂肪酶、低聚异麦芽糖、大豆卵磷脂、人参、白术、茯苓、甘草、黄芪、三七、陈皮、杜仲叶、香茅草、毛霉菌制备而成。
所述的一种槟榔制品,其中柠檬酸100-200重量份、陈醋300-500重量份、壳聚糖20-40重量份、活性氧化锌5-15重量份、独居石煅烧蒸馏水5-15重量份、牛骨蛋白肽40-50重量份、脂肪酶50-70重量份、低聚异麦芽糖150-300重量份、大豆卵磷脂50-75重量份、人参50-80重量份、白术200-300重量份、茯苓100-150重量份、甘草300-400重量份、黄芪150-250重量份、三七90-130重量份、陈皮250-300重量份、杜仲叶350-450重量份、香茅草350-400重量份、毛霉菌150-200重量份。
所述的一种槟榔制品,其中柠檬酸150重量份、陈醋400重量份、壳聚糖30重量份、活性氧化锌10重量份、独居石煅烧蒸馏水10重量份、牛骨蛋白肽45重量份、脂肪酶60重量份、低聚异麦芽糖225重量份、大豆卵磷脂60重量份、人参65重量份、白术250重量份、茯苓125重量份、甘草350重量份、黄芪200重量份、三七110重量份、陈皮275重量份、杜仲叶400重量份、香茅草375重量份、毛霉菌175重量份。
所述的一种槟榔制品,其特征在于该槟榔制品的制备方法为:
(1)取柠檬酸,加入柠檬酸10倍重量的水,加热至100℃,冷却至50度,置于塑料桶内,将采摘的新鲜槟榔浸泡在水中,加入新鲜槟榔的量以水能没过槟榔,同时在水中加入陈醋,浸泡12小时-16小时后,取出槟榔烘干;
(2)取人参、白术、茯苓、甘草、黄芪、三七、陈皮、杜仲叶、香茅草洗净后,粉碎放在蒸笼里蒸30分钟,取出自然晾干后,撒上低聚异麦芽糖,翻动均匀,再撒上毛霉菌,置于发酵罐常温发酵30天;得到发酵后中药;
(3)在发酵后的中药里面添加牛骨蛋白肽、壳聚糖、脂肪酶、活性氧化锌、卵磷脂,再撒上无毒的独居石煅烧蒸馏水搅拌均匀,得到添加剂;
(3)将烘干槟榔切成2瓣,以烘干槟榔重量与添加剂重量为10:1的比例,添加中药添加剂,搅拌均匀,烘干,得到槟榔制品。

本发明独居石煅烧蒸馏水的制备方法为:将独居石粉碎;将粉置于1800-2200度高温炉中,烧制75-105分钟;冷却后,将烧制的粉置于800-1200目的布袋中,用蒸炉进行蒸溜,得到蒸馏液备用.
本发明独居石煅烧蒸馏水的制备方法为:将独居石粉碎;将粉置于2000度高温炉中,烧制90分钟;冷却后,将烧制的粉置于1000目的布袋中,用蒸炉进行蒸溜,得到蒸馏液备用。
本发明所述的脂肪酶(Lipase,甘油酯水解酶)隶属于羧基酯水解酶类,能够逐步的将甘油三酯水解成甘油和脂肪酸。脂肪酶存在于含有脂肪的动、植物和微生物(如霉菌、细菌等)组织中。包括磷酸酯酶、固醇酶和羧酸酯酶。脂肪酸广泛的应用于食品、药品、皮革、日用化工等方面。
本发明所述的毛霉菌(mucor)又叫黑霉、长毛霉。毛霉菌是接合菌亚门接合菌纲毛霉目毛霉科真菌中的一个大属。以孢囊孢子和接合孢子繁殖。毛霉在土壤、粪便、禾草及空气等环境中存在。在高温、高湿度以及通风不良的条件下生长良好。
本发明所述的活性氧化锌(ZnO):粒径介于1-100nm之间,是一种面向21世纪的新型高功能精细无机产品,表现出许多特殊的性质,如非迁移性、荧光性、压电性、吸收和散射紫外线能力等,利用其在光、电、磁、敏感等方面的奇妙性能,可制造气体传感器、荧光体、变阻器、紫外线遮蔽材料、图像记录材料、压电材料、压敏电阻、高效催化剂、磁性材料和塑料薄膜等。
本发明所述的低聚异麦芽糖(IMO)又称为异麦芽低聚糖、异麦芽寡糖、分枝低聚糖等,我国轻工行业标准定为低聚异麦芽糖。
本发明所述的陈醋为市售陈醋。本发明所述的牛骨蛋白肽有名为牛骨胶原蛋白肽,为市售牛骨胶原蛋白肽。
本发明先采用柠檬酸与陈醋对新鲜槟榔进行处理,去除有毒性的成分,再采用发酵中药和活性成分以及独居的蒸馏水,与处理的后的槟榔混合均匀,得到槟榔制品。

实施例1
槟榔制品:柠檬酸1000g、陈醋3000g、壳聚糖200g、活性氧化锌50g、独居石煅烧蒸馏水50g、牛骨蛋白肽400g、脂肪酶500g、低聚异麦芽糖1500g、大豆卵磷脂500g、人参500g、白术2000g、茯苓1000g、甘草3000g、黄芪1500g、三七900g、陈皮2500g份、杜仲叶3500g、香茅草3500g、毛霉菌1500g。
该槟榔制品的制备方法为:
(1)取柠檬酸,加入柠檬酸10倍重量的水,加热至100℃,冷却至50度,置于塑料桶内,将采摘的新鲜槟榔浸泡在水中,加入新鲜槟榔的量以水能没过槟榔,同时在水中加入陈醋,浸泡12小时后,取出槟榔烘干;
(2)取人参、白术、茯苓、甘草、黄芪、三七、陈皮、杜仲叶、香茅草洗净后,粉碎放在蒸笼里蒸30分钟,取出自然晾干后,撒上低聚异麦芽糖,翻动均匀,再撒上毛霉菌,置于发酵罐常温发酵30天;得到发酵后中药;
(3)在发酵后的中药里面添加牛骨蛋白肽、壳聚糖、脂肪酶、活性氧化锌、卵磷脂,再撒上无毒的独居石煅烧蒸馏水搅拌均匀,得到添加剂;
(3)将烘干槟榔切成2瓣,以烘干槟榔重量与添加剂重量为10:1的比例,添加中药添加剂,搅拌均匀,烘干,得到槟榔制品。

该槟榔制品的制备方法为:
(1)取柠檬酸,加入柠檬酸10倍重量的水,加热至100℃,冷却至50度,置于塑料桶内,将采摘的新鲜槟榔浸泡在水中,加入新鲜槟榔的量以水能没过槟榔,同时在水中加入陈醋,浸泡16小时后,取出槟榔烘干;

一种槟榔制品:柠檬酸1500g、陈醋4000g、壳聚糖300g、活性氧化锌100g、独居石煅烧蒸馏水100g、牛骨蛋白肽450g、脂肪酶600g、低聚异麦芽糖2250g、大豆卵磷脂600g、人参650g、白术2500g、茯苓1250g、甘草3500g、黄芪2000g、三七1100g、陈皮2750g、杜仲叶4000g、香茅草3750g、毛霉菌1750g。
该槟榔制品的制备方法为:
(1)取柠檬酸,加入柠檬酸10倍重量的水,加热至100℃,冷却至50度,置于塑料桶内,将采摘的新鲜槟榔浸泡在水中,加入新鲜槟榔的量以水能没过槟榔,同时在水中加入陈醋,浸泡14小时后,取出槟榔烘干;

该槟榔制品的制备方法为:

该槟榔制品的制备方法为:
(1)取新鲜槟榔烘干,备用;
(2)取人参、白术、茯苓、甘草、黄芪、三七、陈皮、杜仲叶、香茅草洗净后,粉碎放在蒸笼里蒸30分钟,取出自然晾干后,撒上低聚异麦芽糖,翻动均匀,再撒上毛霉菌,置于发酵罐常温发酵30天;得到发酵后中药;
(3)在发酵后的中药里面添加牛骨蛋白肽、壳聚糖、脂肪酶、活性氧化锌、卵磷脂,再撒上无毒的独居石煅烧蒸馏水搅拌均匀,得到添加剂;
(3)将烘干槟榔切成2瓣,以烘干槟榔重量与添加剂重量为10:1的比例,添加中药添加剂,搅拌均匀,烘干,得到槟榔制品。

(3)将烘干槟榔切成2瓣,以烘干槟榔重量与添加剂重量为10:1的比例,添加中药添加剂,搅拌均匀,烘干,得到槟榔制品。
独居石煅烧蒸馏水的制备方法为:将独居石粉碎;将粉置于2000度高温炉中,烧制90分钟;冷却后,将烧制的粉置于1000目的布袋中,用蒸炉进行蒸溜,得到蒸馏液备用。
试验例1
病毒灭活试验
将上述实施例1-5和对比例1-2制备得到的槟榔制品,进行病毒灭活试验。
试验方法参照《消毒技术规范》2020年版-2.1.1.10.7。
试验病毒:甲型H1N1流感病毒、甲型H5N1流感病毒、甲型H7N9流感病毒,宿主细胞:MDCK细胞;
试验药物浓度(以槟榔制品计):500mg/ml,溶剂为生理盐水(槟榔制品粉碎至100目,加入生理盐水中,制备成混悬液);作用时间为分别为3h。
试验结果如表1-3所示:

试验结论:上述试验表明,本申请槟榔制品具有很好的抗病毒作用,对于口腔中病毒具有很好的抑制作用,其灭活率能够达到99%;而对比例1和对比例2的槟榔制品,其抗病毒效果显著下降,充分说明本发明槟榔制品具有重要意义。
试验2
对鼠口腔的致癌研究
试验动物:SPF级健康金黄地鼠,体质量95g-105g。
试验方法:取金黄地鼠,随机分组:正常对照组、二甲基苯并蒽组、实施例3组、对比例1组、对比例2组;正常对照组不做任何处理;采用乙醚将金黄地鼠麻醉,用细砂纸将金黄地鼠两侧颊囊磨破,然后分别取实施例3槟榔制品、对比例槟榔制品置于金黄地鼠颊囊中,给药量以双侧颊囊填满为止,放置后不再取出,由金黄地鼠嚼食;二甲基苯并蒽组金黄地鼠双侧颊囊分别给予0.1mL浓度为5g/L的二甲基苯并蒽。每周打磨1次,每2天给药1次,连续18周。给药结束后,肉眼观察各组金黄地鼠颊囊的变化,处死,取其两侧颊囊,以40g/L多聚甲醛固定,以不同体积分数的乙醇和二甲苯逐级脱水透明,石蜡包埋切片后贴于载玻片上,60℃烘箱中烤2h备用。取切片二甲苯脱蜡,不同体积分数乙醇逐级脱水后,进行组织切片苏木素-伊红(HE)染色和微核染色,中性树胶封片,显微镜下观察、拍照,观测各组金黄地鼠颊囊有无病理变化。(注:该方法参考现有文献的方法进行实施,通过该试验方法来确定本发明的技术方案的技术效果)。
试验结果:见表4。

注:a、b:细胞排列规则,有极性;c、d:细胞排列紊乱,层次不清,癌细胞大量浸润。
试验结论:上述试验表明,本发明槟榔制品对鼠颊囊细胞损伤较小,细胞排列规则、有极性,几乎没有癌细胞;而对比例组对颊囊细胞排列紊乱、层次不清,癌细胞大量浸润。充分说明本申请槟榔制品相对安全性更好,致癌性显著降低。
以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。
Claims (7)
1.一种槟榔制品,其特征在于该槟榔制品由槟榔、柠檬酸、陈醋、壳聚糖、活性氧化锌、独居石煅烧蒸馏水、牛骨蛋白肽、脂肪酶、低聚异麦芽糖、大豆卵磷脂、人参、白术、茯苓、甘草、黄芪、三七、陈皮、杜仲叶、香茅草、毛霉菌制备而成。
2.根据权利要求1所述的一种槟榔制品,其中柠檬酸100-200重量份、陈醋300-500重量份、壳聚糖20-40重量份、活性氧化锌5-15重量份、独居石煅烧蒸馏水5-15重量份、牛骨蛋白肽40-50重量份、脂肪酶50-70重量份、低聚异麦芽糖150-300重量份、大豆卵磷脂50-75重量份、人参50-80重量份、白术200-300重量份、茯苓100-150重量份、甘草300-400重量份、黄芪150-250重量份、三七90-130重量份、陈皮250-300重量份、杜仲叶350-450重量份、香茅草350-400重量份、毛霉菌150-200重量份。
3.根据权利要求1所述的一种槟榔制品,其中柠檬酸150重量份、陈醋400重量份、壳聚糖30重量份、活性氧化锌10重量份、独居石煅烧蒸馏水10重量份、牛骨蛋白肽45重量份、脂肪酶60重量份、低聚异麦芽糖225重量份、大豆卵磷脂60重量份、人参65重量份、白术250重量份、茯苓125重量份、甘草350重量份、黄芪200重量份、三七110重量份、陈皮275重量份、杜仲叶400重量份、香茅草375重量份、毛霉菌175重量份。
4.根据权利要求1-3任一项所述的一种槟榔制品,其特征在于该槟榔制品的制备方法为:
(1)取柠檬酸,加入柠檬酸10倍重量的水,加热至100℃,冷却至50度,置于桶内,将采摘的新鲜槟榔浸泡在水中,加入新鲜槟榔的量以水能没过槟榔,同时在水中加入陈醋,浸泡12小时-16小时后,取出槟榔烘干;
(2)取人参、白术、茯苓、甘草、黄芪、三七、陈皮、杜仲叶、香茅草洗净后,粉碎放在蒸笼里蒸30分钟,取出自然晾干后,撒上低聚异麦芽糖,翻动均匀,再撒上毛霉菌,置于发酵罐常温发酵30天;得到发酵后中药;
(3)在发酵后的中药里面添加牛骨蛋白肽、壳聚糖、脂肪酶、活性氧化锌、卵磷脂,再撒上无毒的独居石煅烧蒸馏水搅拌均匀,得到添加剂;
(3)将烘干槟榔切成2瓣,以烘干槟榔重量与添加剂重量为10:1的比例,添加中药添加剂,搅拌均匀,烘干,得到槟榔制品。
5.根据权利要求1-4任一项所述的一种槟榔制品,该槟榔制品在制备治疗口腔疾病的药物中的应用。
6.根据权利要求1-4任一项所述的一种槟榔制品,该槟榔制品在制备治疗口腔炎的药物中的应用。
7.根据权利要求1-4任一项所述的一种槟榔制品,该槟榔制品在制备治疗口腔溃疡药物中的应用。
