Abstract
本发明属于生物技术领域,公开了包含木瓜提取物的生物制剂,其由如下重量份的原料制备而得:木瓜提取物5份、沙棘提取物4份、山楂提取物3份、辅料A?2份、辅料B?2份、稳定剂1份以及牛奶100份。本发明生物制剂采用全天然生物原料,毒副作用小,应用前景广阔。
Description
技术领域
本发明属于生物技术领域,具体地涉及包含木瓜提取物的生物制剂及其应用。
背景技术
木瓜是蔷薇科植物贴梗海棠的干燥近成熟果实,味酸、温,归肝、脾经。随着中药药理学研究的深入,发现木瓜含有多种药理活性成分,包括三萜类、有机酸类、黄酮类以及维生素C等。主要的三萜类成分为五环三萜类化合物,包括齐墩果酸(oleanolicacid)以及熊果酸(ursolicacid)等。
沙棘是一种落叶性灌木,其特性是耐旱、抗风沙,可以在盐碱化土地上生存,因此被广泛用于水土保持。中国西北部大量种植沙棘,用于沙漠绿化。沙棘果实中维生素C含量高,素有维生素C之王的美称。沙棘是植物和其果实的统称。植物沙棘为胡颓子科沙棘属,是一种落叶性灌木。国内分布于华北、西北、西南等地。沙棘为药食同源植物。沙棘的根、茎、叶、花、果,特别是沙棘果实含有丰富的营养物质和生物活性物质,可以广泛应用于食品、医药、轻工、航天、农牧鱼业等国民经济的许多领域。沙棘可降低胆固醇和血糖,缓解心绞痛发作,还有防治冠状动脉粥样硬化性心脏病的作用。
山楂核果类水果,核质硬,果肉薄,味微酸涩。果可生吃或作果脯果糕,干制后可入药,是中国特有的药果兼用树种,具有降血糖血脂、血压、强心、抗心律不齐等作用,同时也是健脾开胃、消食化滞、活血化痰的良药,对胸膈脾满、疝气、血淤、闭经等症有很好的疗效。山楂内的黄酮类化合物牡荆素,是一种抗癌作用较强的药物,其提取物对抑制体内癌细胞生长、增殖和浸润转移均有一定的作用。
糖尿病是由遗传因素、免疫功能紊乱、微生物感染及其毒素、自由基毒素、精神因素等等各种致病因子作用于机体导致胰岛功能减退、胰岛素抵抗等而引发的糖、蛋白质、脂肪、水和电解质等一系列代谢紊乱综合征,临床上以高血糖为主要特点,典型病例可出现多尿、多饮、多食、消瘦等表现,即“三多一少”症状,糖尿病(血糖)一旦控制不好会引发并发症,导致肾、眼、足等部位的衰竭病变,且无法治愈。长期服药对身体会有较大的毒副作用,并且影响胃、肝、肾的功能。如果不需要服药即可控制血糖,而且能够参与正常饮食,那么糖尿病患者将会大大减轻痛苦,并且生活质量大大提高。
发明内容
为了克服现有技术的不足,本发明提供了包含木瓜提取物的生物制剂,该制剂采用全天然生物原料,毒副作用小,可有效地控制血糖和血脂,提高人体免疫机能。
为实现上述目的,本发明采用的技术方案如下:
包含木瓜提取物的生物制剂,其由如下重量份的原料制备而得:
木瓜提取物5份、沙棘提取物4份、山楂提取物3份、辅料A2份、辅料B2份、稳定剂1份以及牛奶100份;
上述生物制剂的制备工艺包括如下步骤:
1)制备木瓜提取物,2)制备沙棘提取物,3)制备山楂提取物,4)制备辅料A;5)制备辅料B;6)制备生物制剂。
具体地,包括如下步骤:
1)制备木瓜提取物:将木瓜洗净后剖开去籽,切成碎块,风干,粉碎成粉末,然后放入体积分数为70%的乙醇中浸泡30分钟,以没过为准,减压旋转蒸发去除乙醇;随后紫外照射15min,再添加体积分数为85%的乙醇浸泡30min,然后1000转/min离心收集上清液,将上清液浓缩成密度为1.2g/ml的浸膏,80℃干燥后,粉碎成粉末,即得;
2)制备沙棘提取物:将沙棘用高速组织捣碎机打浆,然后加水煮沸20min,用三层纱布过滤去渣,收集滤液即得;所述沙棘和水的质量比为1:3;
3)制备山楂提取物:将山楂切成片状,在炒锅中翻炒十分钟,加水煎煮30min后倒出煎煮液,冷却后,离心3分钟,收集离心液,然后浓缩成1.2g/ml的浸膏,即得;所述山楂和水的质量比为1:3;
4)制备辅料A:将山药、枸杞以及牛蒡根按照3:2:1的重量比混合均匀,加5倍重量的水煎煮1小时,过滤收集滤液,用1mol/L的盐酸溶液调整滤液的pH为6.5,然后将滤液浓缩成密度为1.1g/ml的浸膏,80℃干燥后,粉碎成粉末即得;
5)制备辅料B:将粳米和薏苡仁按照2:1的重量比混合均匀,然后研磨成粉末状,过200目筛即得;
6)制备生物制剂:按照重量份取木瓜提取物5份、沙棘提取物4份、山楂提取物3份、辅料A2份、辅料B2份、稳定剂1份以及牛奶100份,混合均匀,然后置于95℃消毒160秒,冷却降温到32℃后,加入保加利亚乳杆菌,发酵12小时,终止发酵,最后冷却降温到4℃,包装密封即得;其中,保加利亚乳杆菌的浓度为5×108个/ml,加入量与稳定剂重量相同。
所述稳定剂由六偏磷酸钠、黄原胶以及海藻酸钠按照5:1:3的质量比混合而得。
本发明取得的有益效果主要包括:
本发明制备的木瓜提取物含有齐墩果酸、熊果酸以及维生素C等物质,能够有效地降低血糖和血脂,提高身体机能;本发明制备的沙棘提取物含有丰富的维生素以及沙棘黄酮,具备较好的降血糖降血脂等功效;山楂提取物中含有丰富的山楂黄酮和有机酸;本发明通过添加多种名贵中药提取物,提高了生物制剂的药用价值;本发明含有益生菌,促进肠道吸收效果好;本发明制备的生物制剂可以有效地预防和治疗高血糖高血脂,无需服药,避免了服用药物带来的毒副作用,并且还可以补充能量,一举两得。本发明制备工艺独特,不同的原料采用不同的处理方式,避免了单一处理方式造成的营养成分浪费,可大规模工业化生产,前景较好。

根据下述实施例,可以更好地理解本发明。然而,本领域的技术人员容易理解,实施例所描述的具体的物料配比、工艺条件及其结果仅用于说明本发明,而不应当也不会限制权利要求书中所详细描述的本发明。
实施例1
包含木瓜提取物的生物制剂,其由如下重量份的原料制备而得:
木瓜提取物5份、沙棘提取物4份、山楂提取物3份、辅料A2份、辅料B2份、稳定剂1份以及牛奶100份;
上述生物制剂的制备工艺包括如下步骤:

实施例2
动物学实验
试验动物选择:选取健康的Wistar雄性大鼠50只,(200±10)g,山东大学实验动物中心提供。大鼠在动物房适应喂养3天后,随机抽取10只作为正常对照组喂以普通维持饲料。高糖高脂饲料配方:大鼠基础饲料68%,蔗糖10%,蛋黄粉9%,猪油8%,胆固醇1%,胆盐0.2%,余量为其他物质。
实施方式:其余40只采用高糖高脂饲料喂养4周。4周后大鼠经禁食18h后,链脲佐菌素(STZ)静脉注射20mg/Kg,每两天一次,使用两次后,血糖血脂居高不下者为造模成功。选取造模成功的大鼠,随机分为3组,每组10只,即模型组(不施用任何药物)、本发明组和对照组:均施用格列奇特(10mg/kg),施用方式均为口服,早晚各一次,进食前半小时服用;本发明组额外每天施用实施例1制备的生物制剂50ml。连续服用10天后检测。
统计学分析:数据资料采用x±s表示,使用SPSS17.0统计软件进行数据分析,多组间比较采用方差分析,两组间比较采用t检验。P<0.05有统计学意义。
治疗效果:检测了空腹血糖(FBG)、总胆固醇(TC)、甘油三酯(TG)等生化指标,单位均为mmol/L,具体结果见表1。

结论:通过上表可知,本发明实施例1制备的生物制剂能够有效地配合西药,显著降低对FBG、TC、TG。
实施例3
典型病例
宋某某,女53岁,兰山区人,空腹血糖7.7mmol/L,餐后两小时血糖9.3mmol/L,总胆固醇6.93mmol/L服用本发明实施例1制备的生物制剂一个月,每天早晚各一次,每次250ml,空腹血糖降至6.0mmol/L,餐后两小时血糖降至7.6mmol/L,总胆固醇也恢复至正常水平,坚持食用本发明的生物制剂,随访半年,无反弹现象。
李某某,男69岁,蒙阴县人,患糖尿病多年。服用本发明实施例1制备的生物制剂一个月后,空腹血糖降至6.0mmol/L,餐后两小时血糖降至7.9mmol/L,而且不用同时服用其他治疗药物。继续食用本发明的生物制剂,随访3个月,无反弹现象,并且身体状况较以前明显改善,精神矍铄,可从事一定的体力劳动。
上述采用具体实施例对本发明进行了详尽描述,但并非对本发明保护范围的限制,所属领域技术人员应该明白,在本发明的技术方案的基础上,本领域技术人员不需要付出创造性劳动即可做出的各种修改或变形仍在本发明的保护范围以内。

1.包含木瓜提取物的生物制剂,其由如下重量份的原料制备而得:
木瓜提取物5份、沙棘提取物4份、山楂提取物3份、辅料A2份、辅料B2份、稳定剂1份以及牛奶100份。
2.根据权利要求1所述的生物制剂,其特征在于,所述生物制剂的制备工艺包括如下步骤:
1)制备木瓜提取物,2)制备沙棘提取物,3)制备山楂提取物,4)制备辅料A;5)制备辅料B;6)制备生物制剂。
3.根据权利要求1或2所述的生物制剂,其特征在于,所述生物制剂的制备工艺包括如下步骤:
1)制备木瓜提取物:将木瓜洗净后剖开去籽,切成碎块,风干,粉碎成粉末,然后放入体积分数为70%的乙醇中浸泡30分钟,以没过为准,减压旋转蒸发去除乙醇;随后紫外照射15min,再添加体积分数为85%的乙醇浸泡30min,然后1000转/min离心收集上清液,将上清液浓缩成密度为1.2g/ml的浸膏,80℃干燥后,粉碎成粉末,即得;
