Example 1 preparation method of houttuynia cordata polysaccharide
1. Materials and instruments
Houttuynia cordata thunb is purchased from Yixintang pharmacy of Kunming, yunnan province, and contains analytical reagents such as 95% ethanol, phenol, concentrated sulfuric acid, glucose, acetone and the like.
UV-5500PC type spectrophotometer (Shanghai chromatography Instrument Co., ltd.); RE-52A rotary evaporator (Shanghai Yangrong Biochemical Instrument factory); a medicinal grinder (large mechanical limited, forest, greens); model 101-2 electrothermal constant temperature air-blast drying oven (Beijing Kewei Instrument Co., ltd.).
2. Extraction of houttuynia cordata crude polysaccharide
Drying houttuynia cordata at 60 ℃, crushing and sieving the dried houttuynia cordata, accurately weighing 4.0g of powder, putting the powder into a 500mL beaker, adding a certain volume of pure water to soak for 24 hours, carrying out ultrasonic extraction for a certain time, filtering and removing residues, repeating the extraction for 2 times, combining filtrates, concentrating, adding 95% ethanol, standing and precipitating, using a centrifuge (4000 r/30 min) to remove upper-layer suspended matters, combining precipitates, dissolving the precipitates in acetone, fully washing and standing overnight, fully washing the precipitates again in absolute ethyl alcohol, standing, heating the precipitates to a certain volume in a rotary evaporator after the precipitates are completely precipitated, and carrying out vacuum drying to obtain crude polysaccharide of houttuynia cordata.
3. Polysaccharide content determination method
The determination principle is as follows: the method for measuring the content of polysaccharide by adopting a phenol-sulfuric acid method has the basic principle that sucrose is concentrated in H 2 SO 4 Under the action of the reaction (as shown in figure 3), furfural or hydroxymethyl furfural generated by intramolecular dehydration can be condensed with phenol to form an orange red compound, and has a maximum absorption peak at 490nm wavelength, so that the absorbance can be measured at the wavelength by a colorimetric method. 4. Determination of polysaccharides
Accurately weighing vacuum-dried herba Houttuyniae polysaccharide, dissolving in pure water, adding into 100mL volumetric flask, diluting to desired volume, and shaking to obtain polysaccharide stock solution. Accurately measuring 2mL of polysaccharide stock solution, placing the polysaccharide stock solution in a 25mL measuring flask, then adding 1mL of 4% phenol solution, shaking uniformly, rapidly adding 7mL of concentrated sulfuric acid, fixing the volume, placing the solution at room temperature for 30min, taking a corresponding reagent as a blank control, and measuring the absorbance at 490 nm.
5. The extraction rate is calculated by the formula that the extraction rate of polysaccharide is% = the mass (g) of polysaccharide in the extracting solution/the mass (g) of houttuynia cordata used multiplied by 100%.
Example 2 Effect of houttuynia cordata polysaccharide extract on sleep in mice at subthreshold dose of sodium pentobarbital
The male ICR mice are divided into houttuynia cordata polysaccharide high-low dose groups (2, 1 g/kg), blank groups and positive control groups, wherein each group comprises 12 mice. The mice in each group were administered by gavage for 14 days continuously, the blank group was administered with 0.5% carboxymethylcellulose solution, and the positive group was administered with diazepam (2 mg/kg). 30min after the last gastric lavage, the abdominal cavity of each mouse is injected with a subthreshold dose of pentobarbital sodium solution (30 mg/kg), and the number of mice in each group falling asleep within 15min is observed and recorded (the mice fall asleep when the righting reflex disappears for more than 1 min).
The influence of the houttuynia cordata polysaccharide extract on the sleep of the mice caused by the threshold dose of the sodium pentobarbital is divided into groups and administered in the same way, after each group of mice are continuously subjected to intragastric administration for 3 days and 30min after the last intragastric administration, the sodium pentobarbital solution (50 mg/kg) with the threshold dose is injected, and the sleep latency period and the sleep duration time of the mice are observed and recorded.
All data in the experiment were analyzed using SPSS 18.0 statistical software, including unpaired t-test, one-way ANOVA, chi-square test 2 ) Two-way ANOVA, and the like, as P<A difference of 0.05 was significant.
