Pitaya (scientific name: hylocereusundulanusBritt.) is a perennial climbing fleshy shrub plant of the family Cactaceae, genus Petasites, the fruit of which is a common, delicious fruit. The dragon fruit stems are rich in mucilage, polysaccharide and flavonoid components, but the medicinal use of the dragon fruit stems is not wide.
Luffa cylindrica (a scientific name: luffaaegyptiaca Miller) is an annual climbing vine plant of Cucurbitaceae, and the tissue exudate after cutting Luffa cylindrica contains rich components such as polysaccharide, plant mucus, vitamins, minerals and the like, is a natural moisturizing factor, and can relieve dry skin. Contains natural medicinal components, has the functions of clearing heat and diminishing inflammation, and is a good raw material of skin care products.
Based on the reasons, the invention provides a traditional Chinese medicine composition containing Ligustrum quihoui, dragon fruit stem extract and towel gourd tissue liquid, wherein the extract has good activities of resisting oxidation and inhibiting tyrosinase, and can be used in various skin care products such as facial masks, essence solutions, toilet water, skin cream and the like.
Disclosure of Invention
The invention mainly aims to provide a natural and safe traditional Chinese medicine composition for resisting oxidation and inhibiting tyrosinase, which has definite curative effect and few side effects, so as to solve the problems of poor anti-aging and whitening effects and the like of common skin care products in the prior art.
In order to achieve the above object, according to one aspect of the present invention, there is provided a Chinese medicinal composition comprising 1 to 10 parts of ligustrum quihoui extract, 1 to 10 parts of dragon fruit stem extract and 1 to 10 parts of towel gourd tissue fluid.
Further, the traditional Chinese medicine composition preferably comprises 4 parts of ligustrum quihoui extract, 5 parts of dragon fruit stem extract and 1 part of towel gourd tissue fluid.
Further, in the traditional Chinese medicine composition, the content of total flavonoids in the Ligustrum quihoui extract is more than or equal to 30%, the content of total triterpenoids is more than or equal to 20%, and the content of total phenylethanoid glycosides is more than or equal to 10%; the content of total polysaccharides in the dragon fruit stem extract is more than or equal to 30 percent, and the content of total flavonoids is more than or equal to 20 percent; the content of total polysaccharide in the towel gourd tissue fluid is more than or equal to 55 percent.
According to another aspect of the present invention, there is provided a method for preparing a ligustrum quihoui extract, comprising the steps of:
taking Ligustrum quihoui leaves, crushing into coarse powder, adding 20 times volume of extraction solvent (containing 0.3% betaine, 0.25% citric acid, 0.3% 1, 3% butanediol aqueous solution), reflux extracting for 2 hours, filtering, loading filtrate on D101 macroporous adsorbent resin, eluting with 5 times column volume of water, 10% ethanol and 70% ethanol, collecting 70% ethanol eluate, recovering ethanol, concentrating, and drying to obtain Ligustrum quihoui extract.
According to another aspect of the present invention, there is provided a method for preparing a dragon fruit stem extract, the method comprising the steps of:
squeezing fresh stems of pitaya to obtain juice, filtering, diluting filtrate with 8 times of water, applying the filtrate to D101 macroporous adsorption resin, sequentially eluting with 5 times of column volume of water and 50% ethanol, collecting water and 50% ethanol eluent, mixing, recovering ethanol, concentrating and drying to obtain the pitaya stem extract.
According to another aspect of the present invention, there is provided a method for preparing a luffa tissue fluid, comprising the steps of:
cutting off the part 30 cm above the root of luffa stem, inserting the upper end of stem into a clean bottle, sealing the bottle, allowing luffa solution to flow into the bottle, collecting, aging, concentrating, filtering, and lyophilizing.
According to another aspect of the invention, the application of the traditional Chinese medicine composition in the aspects of antioxidation and whitening efficacy and the application of the traditional Chinese medicine composition in various skin care products such as facial masks, essence solutions, toilet water, skin cream and the like are provided.
The invention has the beneficial effects that:

Examples
Example 1 preparation of Ligustrum quihoui extract

Through detection, the content of the total flavone in the ligustrum quihoui extract is 33.8%, the content of the total triterpene is 25.3%, and the content of the total phenylethanoid glycoside is 17.2%.
Example 2 preparation of dragon fruit stem extract
Squeezing 5Kg of fresh stem of dragon fruit to obtain juice, filtering, diluting the filtrate with 8 times of water, loading on D101 macroporous adsorbent resin, eluting with 5 times of column volume of water and 50% ethanol in sequence, collecting water and 50% ethanol eluate, mixing, recovering ethanol, concentrating, and drying to obtain 56.9g of dragon fruit stem extract. Through detection, the content of total polysaccharide in the dragon fruit stem extract is 35.1%, and the content of total flavone is 26.8%.
EXAMPLE 3 preparation of towel gourd tissue fluid
Cutting off 30 cm above the root of 100 luffa stems, inserting the upper end of stem into a clean bottle, sealing the bottle, allowing the luffa solution to flow into the bottle, collecting, aging, concentrating, filtering, and lyophilizing to obtain 31.9g of luffa tissue solution. Through detection, the content of total polysaccharide in the towel gourd tissue fluid is 55.3 percent.
Example 4 evaluation of antioxidant Activity of Chinese medicinal composition

Experimental data show that each extract group shows different degrees of antioxidation, and cells respectively subjected to extract predosing can resist cell death caused by peroxide to different degrees.
The antioxidant effect of the traditional Chinese medicine composition prepared by the three traditional Chinese medicine extracts in different proportions is greatly improved, and after the traditional Chinese medicine composition is used for pretreating cells, the cell activity is obviously higher than the sum of the effects of the respective single extracts. After the three traditional Chinese medicine extracts are mixed according to different proportions, the respective abilities of inhibiting cell death caused by peroxide are also higher than the sum of the respective effects of the corresponding respective concentrations.
The results suggest that the three traditional Chinese medicine extracts of ligustrum quihoui, dragon fruit stem and towel gourd may act on different ways or different action sites when having the antioxidation effect, the combined application of the three has a synergistic effect, and the combined application effect of the three is superior to the single use effect.
Example 5 in vitro tyrosinase inhibition evaluation of Chinese medicinal compositions
Mixing 10 μ L10% PBS buffer solution with 90 μ L tyrosinase (100U/mL), incubating at 37 deg.C for 30min, adding 100 μ L5 mg/mL-dopa solution, incubating at 37 deg.C for 30min, measuring absorbance at 475nm, and recording as A; replacing the tyrosinase solution with PBS to measure the absorbance, and recording as B; the absorbance of each group of extract (same as example 4) was measured by using the solution in place of PBS and was designated as C; the absorbance was measured by replacing PBS with each group of extract solution and replacing tyrosinase solution with PBS, and recorded as D. 10 μ L of 0.3% kojic acid was used as a positive control instead of PBS buffer. The tyrosinase inhibition was calculated in parallel 3 times according to the following formula.

The above experimental data show that each extract group shows tyrosinase inhibitory activity of different degrees, the tyrosinase inhibitory activity of the traditional Chinese medicine composition prepared from the three traditional Chinese medicine extracts in different proportions is greatly improved, the above results suggest that the three traditional Chinese medicine extracts of ligustrum quihoui, dragon fruit stem and towel gourd have the tyrosinase inhibitory activity and a synergistic effect, and the combined application effect of the three extracts is superior to that of the single application effect.
Example 6 preparation of a facial cream containing Ligustrum quihoui, pitaya stem extract and Luffa cylindrica tissue fluid
1) Preparation of face cream
The formula of the face cream comprises: 66.1% of water, 4.0% of privet leaflet extract, 5.0% of dragon fruit stem extract, 1.0% of towel gourd tissue fluid, 4.0% of glycerol, 4.0% of polydimethylsiloxane, 3.0% of 1, 3-propylene glycol, 2.0% of hydroxyethyl acrylate/sodium acryloyldimethyl taurate copolymer, 1.0% of caffeine, 1.0% of cyclodextrin, 1.0% of trehalose, 0.5% of acetyl hexapeptide, 1.76% of cyclopenta dimethyl siloxane, 0.39% of sodium hyaluronate, 0.2% of jasmine extract, 0.05% of disodium EDTA, 0.5% of p-hydroxyacetophenone, 0.15% of 1, 2-hexanediol and 0.35% of 1, 3-butanediol.
The preparation process comprises the following steps: adding water, ligustrum quihoui extract, pitaya canker extract, loofah tissue fluid, glycerol and polydimethylsiloxane into a main pot, stirring and heating to 85 deg.C for fully dissolving; then adding 1, 3-propylene glycol, hydroxyethyl acrylate/acryloyl dimethyl sodium taurate copolymer and EDTA disodium, and continuously stirring uniformly; cooling to 45 deg.C, sequentially adding the rest components, slowly stirring, inspecting, packaging, inspecting, and storing.
