CN106619358A discloses a natural, mild and non-irritating herbal skin care preparation, which scientifically matches plant components from the factor of inducing skin sensitivity and allergy, and improves skin sensitivity and allergy conditions. Each plant extract contains unique skin care effect. The baicalin and flavonoid substances in the baical skullcap root extract have the effects of resisting inflammation, inhibiting histamine release, resisting inflammation and inhibiting bacteria of naringin and flavonoid glycoside in the naringin extract, inhibiting histamine release, repairing and strengthening the barrier by the total saponins in the asiatic centella extract, moisturizing and relieving the polysaccharide and sterol in the cactus extract, and the flavonoid substances in the glabrous greenbrier rhizome extract, relieving allergy and immunosuppression, and the synergistic effects of several plant components, resisting inflammation and relieving, inhibiting the release of inflammatory mediators, removing excessive free radicals, repairing skin and restoring skin health activities. The use of multiple plant extracts has high production cost, and the components in the extracts are complex, which may bring about certain side effects.
Disclosure of Invention
The invention aims to overcome the defects of the background technology and provides American elder fermentation liquor with a repairing effect, and a preparation process and application thereof. The preparation process of the invention uses a fermentation substrate containing Sambucus nigra L, and uses lactobacillus plantarum as a microbial strain for fermentation, and the obtained fermentation liquid can synergistically increase the three dimensions of strengthening the barrier, scavenging free radicals and inhibiting inflammatory injury, and can be used for preventing and repairing the skin barrier in an omnibearing way and can be added into cosmetics as an efficacy raw material.
In order to achieve the aim of the invention, the first aspect of the invention provides a American elder fermentation broth with repairing effect, which comprises components of American elder stem branches, microorganism strains, a culture medium and pure water.
The American elder fermentation liquor is rich in a plurality of plant secondary metabolites mainly comprising polysaccharide, and is converted into active ingredients with smaller molecular weight under the fermentation condition of microorganism strains, so that the active ingredients are easier to be absorbed and utilized by human bodies.
The microorganism strain comprises one or more of Lactobacillus delbrueckii subsp bulgaricus (Lactobacillus delbrueckii subsp. Bulgaricum), lactobacillus casei (Lactobacillus casei), saccharomyces cerevisiae (Saccharomyces cerevisiae), lactobacillus plantarum (Lactobacillus plantarum) and acetic acid bacteria (Acetobacter aceti).
When the microorganism strain is Lactobacillus plantarum, the effect of scavenging free radicals of the fermentation broth can be improved, the content of hydroxycinnamic acid, flavonol and anthocyanin in the sambucus nigra is higher, and the active substances such as phenolic acids, flavonoids and the like capable of scavenging free radicals can be increased after the lactobacillus plantarum is fermented. Meanwhile, researches show that the lactobacillus plantarum can reduce the growth of other miscellaneous bacteria by generating various antibacterial compounds, and the lactobacillus plantarum fermentation broth is presumed to be more acidic and is not beneficial to the survival of microorganisms sensitive to the acidity.
Preferably, the microorganism strain is Lactobacillus plantarum (Lactobacillus plantarum), available from Shanghai-associated Biotechnology Inc., model number LZ-J63226.
When the weight ratio of the elder stems to the pure water is 1:10-30, the method is favorable for fermentation process, water is added for dilution, and the sugar concentration of a fermentation system is reduced, so that the influence of high osmotic pressure in a high sugar environment on yeast propagation and fermentation is reduced, the number of yeasts is obviously increased, but the supplied sugar amount is higher than the assimilation rate of yeast cells, and the uneven fermentation caused by the subsequent accumulation of alcohol is unfavorable for the progress of each reaction in the fermentation process. s1, drying, crushing and sieving stems and branches of sambucus nigra, adding pure water for dispersion, and sterilizing to obtain a fermentation substrate;
s2, inoculating a microbial strain into a culture medium, and performing shake culture to obtain a seed culture solution;
s3, adding a certain amount of seed culture solution into a fermentation substrate, and fermenting to obtain an initial fermentation solution;
and S4, sequentially sterilizing the initial fermentation liquor, performing wall breaking treatment, filtering, and collecting filtrate to obtain the American elderberry fermentation liquor with repairing effect.
Preferably, the particle size after sieving in the step S1 is 500-1000 meshes.
Further preferably, the particle size after sieving in S1 is 800 mesh.
Preferably, the sterilization of S1 and S4 is performed at 110-130 ℃ for 20-60min.
Further preferably, the sterilization of S1 and S4 is performed at 121 ℃ for 20min.
Preferably, the microbial strain inoculation amount in the S2 is 0.5-3% of the mass percentage of the culture medium, the rotation speed of a shaking table is 100-200rpm, the culture temperature is 35-45 ℃, and the culture time is 18-48h.
Further preferably, the microbial strain inoculation amount in the S2 is 1.5% of the mass percentage of the culture medium, the rotation speed of a shaking table is 150rpm, the culture temperature is 37 ℃, and the culture time is 36 hours.
Preferably, the medium in S2 is an MRS liquid medium, purchased from beijing solebao science and technology, model: m8540.
The seed culture solution is inoculated in an amount of 1-5% by mass of the fermentation substrate, and the fermentation solution has excellent strengthening barrier effect, and probably improves the content of substances such as citric acid, malic acid and the like in the fermentation solution, so that the moisture retention of the fermentation solution is improved, and the pH value of skin can be improved when the fermentation solution is added into cosmetics. Too small inoculation amount, insufficient strain in fermentation broth to raise active matter content, too large inoculation amount, easy excessive fermentation of fermentation broth and too high acidity, can enrich impurity protein and is unfavorable for absorption. Further research shows that when the seed culture solution is inoculated in an amount of 1.5-3.5% of the fermentation substrate, the odor of the American elder fermentation broth can be further improved, and the American elder fermentation broth is more acceptable when the American elder fermentation broth is applied to cosmetics, and the odor of the fermentation broth is mutually synergistically improved due to the fact that a large amount of glycolytic enzymes, lipolytic enzymes and proteolytic enzymes convert some nutrient substances of the culture medium into compounds with sensory characteristics and are mixed with special musk odor of the American elder.
Preferably, the seed culture solution is inoculated in an amount of 1-5% by mass of the fermentation substrate.
Further preferably, the seed culture solution in the step S3 is inoculated in an amount of 1.5-3.5% by mass of the fermentation substrate.
Still more preferably, the seed culture solution in S3 is inoculated in an amount of 3% by mass of the fermentation substrate.
When the fermentation temperature is 35-45 ℃, the fermentation efficiency can be improved by cooperating with the lactobacillus plantarum, due to the higher temperature fitness with the proper growth of the lactobacillus plantarum, further research discovers that when the fermentation temperature is 36-39 ℃ and the fermentation time is 24-72 hours, the clearance of the fermentation liquor to DPPH (1, 1-diphenyl-2-trinitrophenylhydrazine) free radicals is further improved, as the fermentation is carried out, the acidity of the system is reduced, the concentration of flavone in the fermentation liquor is continuously increased, the antioxidation activity is continuously strong, the antioxidation activity begins to be weakened after the fermentation time is continuously prolonged, and presumably, the change of the active components of the fermentation liquor in the later fermentation period is caused, and when the acidity is too high, the cyanogenic glycoside substances in the sambucus chinensis are decomposed into aldehyde ketone and hydrocyanic acid, so that certain side effects are generated on a human body.
Preferably, the fermentation temperature in the step S3 is 35-45 ℃, and the rotation speed of the shaking table is 100-200rpm when the fermentation time is 24-72 hours.
Further preferably, the fermentation temperature in S3 is 38 ℃, the fermentation time is 48 hours, and the rotation speed of the shaking table is 200rpm.
The high-pressure homogenizer is adopted for wall breaking treatment, the process can be simplified, the production efficiency is improved, the high-speed material has better wall breaking effect when the pressure is 400-600bar, strong shearing force can be generated when the high-speed material passes through a valve cavity channel and a current limiting gap, cells are broken, the lactobacillus plantarum belongs to gram positive bacteria, the cell wall has very thick peptidoglycan components, the wall breaking is difficult, the wall breaking efficiency is not high enough, the temperature is improved to 70-90 ℃ through research, the wall breaking efficiency can be improved, but the wall breaking is carried out at high temperature for a long time, meanwhile, friction heat among the high-speed material is generated, the local temperature of a system is increased, the effect of partial active ingredients in fermentation liquid can be lost due to high temperature, and further research finds that the wall breaking of gaps can be solved, particularly, the wall breaking is carried out for 4-6min, and the better effect can be achieved through repeated 3-5 times.
Preferably, the pressure of the high-pressure homogenizing wall breaking treatment is 400-600bar, the temperature is 70-90 ℃, and the wall breaking method comprises the following steps: breaking wall for 4-6min, and repeating for 3-5 times at a gap of 4-6min
Further preferably, the high-pressure homogenizing wall breaking treatment in S4 is performed at a pressure of 500bar and a temperature of 75 ℃, and the wall breaking method comprises the following steps: breaking wall for 5min, and repeating for 3 times at 5 min.
The third aspect of the invention provides an application of American elder fermentation liquor with a repairing effect, which is applied to cosmetics.
Preferably, the cosmetic comprises one or more of water, emulsion, spray, cream and facial mask.
Preferably, the American elder fermentation liquor is added into the cosmetic in a mass percentage of 0.1-10%.
Further preferably, the American elder fermentation broth is added into the cosmetic in a mass percentage of 1-5%.
The beneficial effects are that:
1. when the weight ratio of the elder stems to the pure water is 1:10-30, the fermentation process is facilitated, and the effect of inhibiting inflammatory injury of the fermentation liquid can be improved.
2. When the microorganism strain is lactobacillus plantarum, the effect of scavenging free radicals of the fermentation liquor can be improved, and the growth of other miscellaneous bacteria can be reduced.
3. The seed culture solution is inoculated in an amount of 1-5% by mass of the fermentation substrate, and the fermentation solution has excellent strengthening barrier effect.
4. The fermentation temperature is 35-45 ℃, the fermentation time is 24-72h, and the clearance of the fermentation liquor to DPPH (1, 1-diphenyl-2-trinitrophenylhydrazine) free radicals is improved.
5. The wall breaking treatment is carried out by a high-pressure homogenizer at 70-90deg.C under 400-600bar for 4-6min and 4-6min for 3-5 times, thereby simplifying process and improving production efficiency.

Performance test method
The antioxidant DPPH clearance (%) test is carried out as follows: a) 0.2g of fermentation broth was weighed, 98.8g of water was added, and a 0.2% fermentation broth sample was prepared.
b) A40. Mu.l sample of the fermentation broth was removed by pipetting, and 160. Mu.l of DPPH (1, 1-diphenyl-2-trinitrophenylhydrazine) ethanol solution (200. Mu.g/ml) was added.
c) After mixing, reacting for 30min at room temperature under the dark condition, measuring absorbance A at 517nm, setting 3 groups of parallel experiments for each group, and calculating the average value, wherein the calculation method is as follows:

d) After the completion of the addition, the 6-well plate was placed in a cell incubator (5% CO2, 37 ℃) and cultured for 2 hours.
e) LPS stimulation: after 2h of incubation, 200. Mu.l of the LPS-containing working solution was added to each well, and the mixture was placed in a cell incubator (5% CO2, 37 ℃) and incubated for 22h.
f) And (3) detection: cell culture supernatants were collected and assayed by ELISA according to the ELISA kit instructions.

b) Screening 30 healthy volunteers, aged 30-45 years, using 2% fermentation broth samples, 1 time each day in the morning and evening, and continuously using for 4 weeks;
c) Skin moisture content and transepidermal moisture loss values were measured using a german CK Corneometer CM825 skin moisture tester.
