(2) The test method comprises the following steps:
(1) culturing, separating and identifying anaerobic bacteria: inoculating the sample obtained by separating skin lesions of acne patients into THl culture medium, culturing in anaerobic jar at 37deg.C for 72 hr by air bag method, and taking blue as indicator and palladium particles as catalytic thorns.
(2) Separation of anaerobic bacteria
Picking single bacterial colony, transferring to 2 flat plates, placing one plate into an anaerobic tank, and performing pure culture at 37 ℃ under anaerobic conditions; the other block was cultivated aerobically at 37 ℃; after 48h the results were observed.
(3) Identification of anaerobic bacteria no bacterial growth under aerobic culture; colonies with uniform, off-white, fine and round protrusions with diameters of about 1.0mm and slightly aligned edges appear on the anaerobic culture plates, and the colonies are subjected to microscopic examination to visualize the purple bacillus under the microscope. Microbending is in a rod shape or a rod shape, one end of the microbending is blunt, the other end of the microbending is thin, the cells are single, and the microbending can be arranged in pairs or in a shape like a human body or a v. And the propionibacterium acnes are identified by an oxygen-resistant test, a thixotropic enzyme test, an indole test and a biochemical series test.
(4) Anaerobic drug sensitivity test
Strain inoculation: 1 colony of the cultured colonies is picked and inoculated on an LB plate medium, aerobic culture is carried out for 24 hours at 370 ℃, propionibacterium acnes which are aerobically cultured for 24 hours is washed by sterile normal saline, and the washing liquid is put into a sterile test tube; turbidimetry is adopted, the turbidimetry is carried out for 12 times before the turbidimetry tube is used, the turbidimetry is carried out to 3X 108/mL, and then the turbidimetry is diluted to 106/mL for standby.
Inoculating LB plate medium: dipping sterilized cotton sticks in bacterial liquid, squeezing excessive bacterial liquid on the pipe wall, uniformly coating on an LB plate culture medium, sweeping a circle along the edge of a plate, covering the plate, and drying for 2min. The sterile steel cup (diameter of 6 mm) was placed on a plate culture medium, the essence obtained in examples and comparative examples was filled up, and the plate was placed in 37℃for aerobic culture for 24 hours, and no bacterial growth was found in the plate, and after continuing the aerobic culture at 37℃for 24 hours, a zone of inhibition was found to be formed. The diameter of the inhibition zone is measured, and the diameter values of the 3 groups of inhibition zones are averaged.
Test results:
the antibacterial sensitization of traditional Chinese medicines is classified according to corresponding antibiotic standards. The diameter of the inhibition ring is higher than 20mm, the inhibition ring is middle sensitivity and is 10-20 mm, the inhibition ring is larger in diameter, the better acne removing effect of the essence is shown in table 6.
