Observation experiment for improving sleep effect of dendrobium nobile flower CO2 supercritical extract
1) Experimental method
ICR mice (20-24 g) were purchased from sbefu (beijing) biotechnology limited [ license number: SCXK (Beijing) 2019-0010], which is bred in an environment with room temperature of 20 +/-2 ℃, relative humidity of 50-70% and 12h alternating illumination and darkness, and during which people can drink water and eat freely.
The mice were randomly divided into 5 groups of 10 mice each, namely a blank control group, a positive control group, an extract high dose group, an extract medium dose group and an extract low dose group. Perfusing distilled water with corresponding volume in the blank control group; a positive control group of perimidine (1.5 mg/kg); the mice in each group were subjected to intraperitoneal injection of 40mg/kg sodium pentobarbital to cause the mice to sleep after the intragastric administration for 1h in the extract high dose group (100 mg/kg), the extract medium dose group (50 mg/kg) and the extract low dose group (25 mg/kg). The sleep latency and sleep duration of each group of mice were recorded during the experiment.
2) Results of the experiment
The experimental results are shown in fig. 1 and fig. 2, the experimental data are expressed as mean ± standard deviation (x ± s), fig. 1 is the experimental result of the sleep latency of the mouse, and fig. 2 is the experimental result of the sleep duration of the mouse. Wherein, compared to the blank control group, indicates P <0.05; * Denotes P <0.01.
The results show that: compared with a blank control group, the sleep latency of the positive control group is obviously shortened, the sleep duration is obviously prolonged, and the positive control group has extremely obvious difference (P < 0.01). In the aspect of sleep latency, compared with a blank control group, the sleep latency of the dendrobium nobile lindl flower extract high-dose group and the dendrobium nobile lindl flower extract medium-dose group is obviously shortened (P is less than 0.05); in terms of sleep duration, the sleep duration of the dendrobium nobile flower extract high-dose group and the dendrobium nobile flower extract medium-dose group was significantly prolonged (P < 0.05) compared with the blank control group. The experimental results show that the CO2 supercritical extract of dendrobium nobile flowers has an exact sleep improving effect.
Claims (6)
1. A traditional Chinese medicine extract with the function of improving sleep is characterized in that: comprises the following components in percentage by mass: 6-methoxyquinoline nitroxide, 2-fluoro-6-trifluoromethyl-3, 5-difluorophenyl ester and n-heptacosane, wherein the mass percentage of the substances is not less than 5.0%.
2. The preparation method of the traditional Chinese medicine extract with the sleep improving effect according to claim 1, which is characterized by comprising the following steps:
s1: taking dendrobium stem flower, filling CO 2 Extracting in an extraction kettle of a supercritical extractor, wherein the pressure of the extraction kettle is 27.5MPa, the temperature is 46-47 ℃ and the extraction is carried out for 180min;
s2: from CO after extraction 2 Separating out dendrobium nobile flower CO in separating kettle of supercritical extraction apparatus 2 Supercritical extract, separation at I part pressure: 6.9MPa; i minute temperature: 38-39 ℃; II partial pressure: 5.9MPa; II, temperature division: 44-45 ℃.

The traditional Chinese medicine extract with the sleep improving effect as claimed in claim 1, is characterized in that: the Chinese medicinal extract is used for preparing medicinal preparations, such as capsule, tablet, granule, oral liquid, unguent, misty preparation, dripping pill, mixture, slurry, powder or teabag.
4. The traditional Chinese medicine extract with the sleep improving effect as claimed in claim 3, is characterized in that: the pharmaceutical preparation also comprises pharmaceutic adjuvants.

The traditional Chinese medicine extract with sleep improvement effect according to claim 4, is characterized in that: the pharmaceutic adjuvant comprises one or more of a diluent, a disintegrating agent, an adhesive, a preservative, an antioxidant, a flavoring agent, a suspending agent and an emulsifying agent;
the diluent is one or more of mannitol, sucrose, lactose, sorbitol, xylitol, polyethylene glycol, propylene glycol, vegetable oil and mineral oil;
the disintegrant is one or more of croscarmellose sodium, colloidal silicon dioxide and citric acid;
the adhesive is one or more of starch slurry, ethanol, water and polyvidone alcohol solution;
the preservative is one or more of ethyl paraben, propyl paraben, sorbic acid, potassium sorbate, calcium propionate, sodium dehydroacetate, sodium diacetate and sodium lactate;
the antioxidant is one or more of ethylenediamine tetraacetic acid, disodium ethylenediamine tetraacetic acid, dibutyl hydroxy toluene, glycine, inositol, ascorbic acid, sodium ascorbate, lecithin, malic acid, hydroquinone, citric acid, succinic acid, sodium pyrosulfite;
the flavoring agent is one or more of aspartame, sucrose, xylitol, stevioside, sodium cyclamate, sorbitol, cacao, pure vanilla, vanillin, ethyl vanillin, chocolate, fructus Hordei Germinatus, and herba Menthae;
the suspending agent is one or more of xanthan gum, polyvinylpyrrolidone, sodium alginate, aluminum stearate, and hydrogenated vegetable oil;
the emulsifier is one or more of alkyl sulfate, soap, dodecyl benzene sulfonate, lactate, sulfosuccinate, monoglyceride sulfonate, phosphate, siloxane, and taurate.
6. The extract of a Chinese herb with sleep improving effect according to any one of claims 3 to 5, wherein: the pharmaceutical preparation can be added into medicine, food or health product for preventing or treating sleep disorder or promoting sleep.
