背景技术
灵芝(Ganoderma Lucidum Karst)为多孔菌科真菌。灵芝的人工栽培始于上世纪六十年代。近十多年来,灵芝栽培技术的不断成熟,全球的灵芝产业发展极为迅速。但近年来由于人工栽培灵芝菌种退化,产量不稳,质量下降,很多灵芝和灵芝孢子粉质量达不到要求,市场上灵芝相关的产品质量参差不齐,严重阻碍了灵芝产业的进一步发展。药材好,药才好。为确保灵芝产品的质量,必须保证灵芝菌种的高质量。因此要选育优异的灵芝菌株,服务于灵芝生产。
灵芝所含的化学成分比较复杂,灵芝多糖是灵芝最主要的有效成分,具有提高机体的免疫力和耐缺氧能力、抑制肿瘤、消除自由基等药理活性。灵芝中的三萜类成分是灵芝中一类特有的成分,种类很多,具有解毒、保肝、抗肿瘤、抗炎等功效。此外,灵芝中含有一定量的核苷类、甾醇、氨基酸和无机元素,其中麦角甾醇具有调节新陈代谢、调节激素水平、预防心血管疾病等功能。因此灵芝多糖、灵芝三萜和麦角甾醇可作为评价灵芝及其相关产品质量的指标。

本发明提供的新菌株灵芝京诚1号(Ganoderma lucidum京诚1号,CCTCC NO:M2020875)可采用本领域常规方法及培养基进行菌种扩繁生产和栽培。例如,培养及栽培条件:菌丝生长温度10℃-38℃,最适23℃-28℃。
本发明提供的新菌株灵芝京诚1号(Ganoderma lucidum京诚1号,CCTCC NO:M2020875)孢子粉产量高,孢子粉和子实体产量比可达140%以上。

在本发明实施例中,所述代料培养基,包括麸皮15wt%,玉米粉5wt%,石膏粉1wt%,经灭菌的灰树花(Polyporus frondosus)发酵液3wt%;还包括三磷酸腺苷(ATP)、三磷酸鸟苷(GTP),以及余量的木屑;其中,以所述代料培养基的总量计,三磷酸腺苷的含量为10mg/kg,三磷酸鸟苷的含量为10mg/kg。
在本发明实施例中,制备灰树花发酵液的发酵培养基:葡萄糖20g,蛋白胨 4g,磷酸二氢钾1.5g,硫酸镁1g,维生素B1 1g,蒸馏水1000mL(pH自然)。
本发明还提供一种灵芝的栽培方法,包括采用上述代料培养基进行栽培。其中,优选采用上述新菌株灵芝京诚1号(Ganoderma lucidum京诚1号,CCTCC NO:M2020875)进行栽培。
本发明还包括上述栽培方法所得的灵芝孢子粉和/或灵芝子实体。该灵芝孢子粉中麦角甾醇含量显著高于现有产品。

具体实施方式
以下实施例用于说明本发明,但不用来限制本发明的范围。以下未特殊指明之处,均可采用本领域常规方法。
实施例1菌株筛选过程
2012年7月在江西省上饶市三清山一栎树树桩采集的一个野生灵芝,带回后进行组织分离和培养,获得纯化菌株(编号为G1)。将该菌株(G1)和灵芝主流栽培品种同期在实验大棚同样生长条件下进行栽培,获得灵芝子实体,根据其形态鉴定,结合分子鉴定,确定为灵芝(Ganoderma lucidum)。栽培发现该菌株(G1)性状优良,而后以此为出发菌株(G1),转接至装有200mL液体培养基(PDA)的500ml三角瓶中,26℃静置培养,无菌条件下将带有菌丝体的液体培养基(PDA)12000rpm离心12min,弃上清液,得到菌丝体,用无菌水清洗 (2次),再用0.6M甘露醇稳渗剂清洗菌丝体3次,每100mg湿菌丝体分别加入1mL酶液,并放入无菌玻璃珠,置于30℃恒温水浴锅中酶解2.5h,每隔 20min,摇匀1次,过滤,4000rpm离心10min,弃上清液,沉淀即为灵芝原生质体。紫外诱变该灵芝原生质体:取8×105个/mL该灵芝原生质体均匀涂于再生培养基中,在15W紫外灯下照射,照射距离为30cm,然后置于培养箱25℃避光培养。连续诱变12批,筛选出23株菌丝生长较快、较密的再生菌株进行生物学性质比较、遗传稳定性试验、大田栽培,系统选育筛选出新菌株(G2),编号为新菌株京诚1号,于2020年12月24日保藏于中国典型培养物保藏中心(简称CCTCC,地址:中国.武汉.武汉大学,邮编430072),分类命名为Ganoderma lucidum京诚1号,其保藏编号为:CCTCC NO:M2020875。
新菌株灵芝京诚1号(Ganoderma lucidum京诚1号,CCTCC NO:M2020875) 的形态特征如下:
子实体特征:菌盖近圆形,15×23cm,厚1.6-2.8cm,表面红褐色,有同心环沟和环带并有皱,有似漆样光泽;边缘锐,稍向内卷。菌肉淡白色,接近菌管处近褐色,厚约1cm。菌管淡褐色,长0.8-1.2cm。孔面初期白色,后渐变褐色。菌柄近圆柱形,近中生,长6.2-15.8cm,粗3.2-4.7cm,与菌盖同色,有光泽。
皮壳构造呈拟子实层型,淡褐色,组成菌丝棍棒状,顶端膨大部分宽6.5-8 μm,长18-25μm。
孢子卵形,顶端平截,双层壁,外壁透明,平滑,内壁淡褐色,有小刺,10-12 ×5-6μm,中间有油滴。
生物学特性:营养成分来源,可用PDA培养基或者加富PDA(马铃薯200g,麦麸50g,葡萄糖20g,磷酸二氢钾3g,硫酸镁1.5g,维生素B1 0.1g,琼脂粉15g,蒸馏水1000mL,pH自然)保存,适宜在4-8℃冰箱低温冷藏保存,每半年转接1次。
拮抗试验:将上述出发菌株(G1)和新菌株京诚1号在同一平板上接种(PDA 培养基),25℃恒温下培养。结果见图1。图1中,A为新菌株京诚1号,B 为出发菌株(G1)。由图1可见两菌株间产生拮抗反应,新菌株京诚1号菌丝生长速度快,且还能抑制出发菌株(G1)的生长。表明出发菌株(G1)和新菌株京诚1号存在差异。
实施例2
将实施例1出发菌株(G1)和新菌株京诚1号分别定量接种于平板上(PDA 培养基),25℃恒温下培养。每两天在培养皿底部划线标记,采用“十字交叉法”测量菌落直径。结果出发菌株(G1)的平均生长速度为1.641cm/d,而新菌株京诚1号平均生长速度为2.215cm/d。统计分析结果显示,新菌株京诚1号的菌丝生长速度显著高于出发菌株(G1)。
实施例3
将实施例1出发菌株(G1)和新菌株京诚1号分别按相同的方法进行代料栽培。所用代料培养基:木屑79wt%,麸皮15wt%,玉米粉5wt%,石膏粉1wt%。菌袋规格为33cm×17cm×0.025cm,平均装量1750克/袋。结果如下:

结果发现,新菌株京诚1号生长速度快,产量高,一级品率高,且袋污染率低,具有显著的生产优势。
结果还发现,与出发菌株(G1)相比,新菌株京诚1号在孢子粉和子实体产量比,子实体的灵芝三萜含量、灵芝多糖含量及孢子粉中麦角甾醇含量方面也具有显著的优势。
实施例4
将实施例1出发菌株(G1)和新菌株京诚1号分别按相同的方法进行代料栽培。栽培方法与实施例3的区别仅在于在所用代料培养基为:麸皮15wt%,玉米粉5wt%,石膏粉1wt%,经灭菌的灰树花(Polyporus frondosus)发酵液3wt%;还包括三磷酸腺苷(ATP)、三磷酸鸟苷(GTP),余量为木屑;以所述代料培养基的总量计,三磷酸腺苷的含量为10mg/kg,三磷酸鸟苷的含量为10mg/kg。其中,制备灰树花发酵液的发酵培养基:葡萄糖20g,蛋白胨4g,磷酸二氢钾 1.5g,硫酸镁1g,维生素B1 1g,蒸馏水1000mL,pH自然。
结果如下:孢子粉中麦角甾醇含量,出发菌株(G1)为1215μg/g,新菌株京诚1号为5510μg/g。
可见,在代料培养基中添加灰树花发酵液(经灭菌)、三磷酸腺苷(ATP) 和三磷酸鸟苷有利于进一步提高孢子粉中麦角甾醇含量。
另外,实验过程中还发现,本发明新菌株灵芝京诚1号(Ganoderma lucidum 京诚1号)尤其适用于代料栽培。该菌株代料栽培在产量、一级品率、抗污染方面相对于椴木栽培具有显著优势。
虽然,上文中已经用一般性说明及具体实施方案对本发明作了详尽的描述,但在本发明基础上,可以对之作一些修改或改进,这对本领域技术人员而言是显而易见的。因此,在不偏离本发明精神的基础上所做的这些修改或改进,均属于本发明要求保护的范围。

1.一种灵芝菌株,其特征在于,所述灵芝菌株为Ganoderma lucidum京诚1号,所述菌株的保藏编号为:CCTCC NO:M2020875。
2.一种灵芝孢子粉,其特征在于,由权利要求1所述的灵芝菌株栽培所得。
3.一种灵芝子实体,其特征在于,由权利要求1所述的灵芝菌株栽培所得。
4.权利要求2所述的灵芝孢子粉或权利要求3所述的灵芝子实体在制备食品或药物上的应用。
