本发明提供了软骨组织脱细胞水凝胶支架及其制备方法与应用,本发明制备了软骨组织脱细胞水凝胶支架,并成功构建了兔髌骨沟缺损模型,通过软骨组织脱细胞水凝胶支架移植进入模型缺损处,可实现对缺损的有效改善。

软骨组织脱细胞水凝胶支架及其制备方法与应用
技术领域
本发明属于软骨组织工程技术领域,涉及软骨组织脱细胞水凝胶支架及其制备方法与应用。
背景技术
髌骨位于膝关节前方,股骨的下端前面,是人体内最大的籽骨,包埋于股四头肌腱内,为三角形的扁平骨。底朝上,尖向下,前面粗糙,后面为光滑的关节面,与股骨的髌面相关节,参与膝关节的构成。可在体表摸到。髌骨具有保护膝关节,增加膝关节回转能力的功能。髌骨缺损如果不能及时修复将影响膝关节的功能,进而影响生活质量。
目前组织工程学方法为解决关节骨缺损的修复提供了新的途径,如何更接近正常关节软骨的组织结构则是组织工程支架构建过程中的一个重要因素。
发明内容
本发明的目的在于提供软骨组织脱细胞水凝胶支架及其制备方法与应用。
为了实现本发明的目的,本发明采用的技术方案为:
本发明提供了软骨组织脱细胞水凝胶支架的制备方法,包括:
1)制备软骨组织脱细胞,包括:
1-1)将软骨组织切成小块,以纯水冲洗去除杂质;
1-2)洗净后的标本置于液氮10min,结束后在37℃环境下溶解10min,完成5个循环,超声清洗;
1-3)4℃下,将标本在PBS中用0.25%胰蛋白酶处理24小时,每6小时替换一次胰蛋白酶溶液,然后用高渗缓冲溶液洗涤胰蛋白酶消化的软骨标本浆液;
1-4)将标本在37℃下用核酸酶溶液在10mM Tris-HCl,pH 7.5中处理4h;
1-5)将酶处理过的软骨浆液用1%Triton X-100溶液处理24小时后,再用低渗的Tris-HCL溶液洗涤20小时,以除去所有酶;
1-6)1×PBS冲洗标本,纯水冲洗标本,液氮30min,低温粉碎组织,以100目滤网筛选软骨微粒,真空冷冻干燥;
2)制备凝胶:将步骤1)制备好的软骨组织脱细胞支架粉末添加到1mg·mL-1的胃蛋白酶的0.01M HCl溶液中,支架粉末:胃蛋白酶的重量比为10:1,并在室温下以恒定速度搅拌预定的消化时间,从48小时支架粉末的部分消化产生pH值2~3的中等粘度溶液,通过在冰上中和之前添加适量的1×PBS,制备出浓度较低的支架粉末溶液,通过加入0.01M NaOH溶液和1×PBS分别提供生理酸性和盐度,将中和的胃蛋白酶消化液在37℃下孵育以完成凝胶化;
3)制备凝胶复合支架:在室温下将支架浸泡在制备好的凝胶中4-6h完成复合。
优选地,步骤1-1)中,将软骨组织切成2mm3小块,以纯水冲洗5×3min,以去除杂质。
优选地,步骤1-2)中,超声清洗机功率100W,20℃,20min,清洗3遍。
优选地,步骤1-3)中,高渗缓冲溶液含有1.5M NaCl的50mM Tris-HCl,pH 7.6.
优选地,步骤1-4)中,核酸酶溶液含有50U ml-1DNAse和1U ml-1RNAse A。
优选地,步骤1-6)中,1×PBS冲洗标本,5×6min;纯水冲洗标本,5×6min。
本发明还提供了制备方法制备得到的软骨组织脱细胞水凝胶支架。
本发明又提供了上述软骨组织脱细胞水凝胶支架在兔髌骨沟缺损模型修复中的应用。
优选地,所述应用包括:在髌骨沟处用牙科钻做一个直径5mm,长3mm的缺损,然后把软骨组织脱细胞水凝胶支架移植进入缺损处。
优选地,所述兔髌骨沟缺损模型通过以下方法构建:常规麻醉,麻醉起效后,对动物右后肢进行备皮,动物仰卧位固定在手术台上,对右后肢进行造模,先碘伏消毒,髌骨内侧打开关节囊,分离肌肉和韧带,暴露关节腔,然后在髌骨沟处用牙科钻做一个直径5mm,长3mm的缺损。

具体实施方式
为了更清楚地说明本发明,下面结合实施例并对照附图对本发明作进一步详细说明。本领域技术人员应当理解,下面所具体描述的内容是说明性的而非限制性的,不应以此限制本发明的保护范围。
实施例
一、软骨组织脱细胞水凝胶支架制备
1.实验目的
将大鼠软骨组织进行脱细胞处理后制成凝胶复合到3D打印支架上。
2.实验材料和方法
材料(货号,厂家):大鼠软骨组织(动物房提供);液氮;PBS(KGB5001,凯基生物);胰蛋白酶(T8150,索莱宝);Tris-HCl(T8230,索莱宝);DNaseI(CW2090S,康为世纪);RNaseA(CW0601S,康为世纪);灭菌超纯水;胃蛋白酶(Sigma,P7125);
仪器(货号,厂家):超声清洗机(YM-020S,语盟);冷冻干燥机(FD-1A-50,上海比朗);100目滤网(YA0945,索莱宝)。
3.实验步骤
3.1软骨组织脱细胞制备:将软骨组织切成2mm3小块,以纯水冲洗5×3min,以去除杂质。洗净后的标本置于液氮10min,结束后在37℃环境下溶解10min。完成5个循环(即置于液氮10min,结束后在37℃环境下溶解10min)。超声清洗机清洗(功率100W,20℃,20min),清洗3遍。4℃下,将标本在PBS中用0.25%胰蛋白酶处24小时,每6小时替换一次胰蛋白酶溶液。用高渗缓冲溶液(含有1.5M NaCl的50mM Tris-HCl,pH 7.6)洗涤胰蛋白酶消化的软骨标本浆液,接下来将标本在37℃下用核酸酶溶液(50U ml-1DNAse和1U ml-1RNAse A)在10mMTris-HCl,pH 7.5中处理4h。将酶处理过的软骨浆液用1%Triton X-100溶液处理24小时后,再用低渗的Tris-HCl溶液洗涤20小时,以除去所有酶。1×PBS冲洗标本,5×6min;纯水冲洗标本,5×6min。液氮30min,组织粉碎机低温粉碎组织,以100目滤网筛选软骨微粒。未过滤则再次研磨粉碎,直至尽可能的通过滤网。真空冷冻干燥机,24h。
3.2凝胶制备:将上述制备好的软骨组织脱细胞支架粉末添加到1mg·mL-1的胃蛋白酶(Sigma,P7125)的0.01M HCl溶液中,支架粉末:胃蛋白酶的重量比为10:1,并在室温下以恒定速度搅拌预定的消化时间,从48小时支架粉末的部分消化产生pH值约为2.8的中等粘度溶液。通过在冰上中和之前添加适量的1×PBS,可以制备出浓度较低的支架粉末溶液。支架粉末溶液的凝胶化是通过加入0.01M NaOH溶液和1×PBS分别提供生理酸性和盐度。将中和的胃蛋白酶消化液在37℃下孵育以完成凝胶化。通过观察颜色从清澈到浑浊的变化以及在将玻璃瓶上下颠倒放置在玻璃瓶底部的完整结构来确认凝胶化。
3.3凝胶复合支架制备:在室温下将支架浸泡在制备好的凝胶中4-6h完成复合。
4.实验结果
制备得到的软骨组织脱细胞水凝胶支架外观如图1所示,扫描电镜照片如图2所示,可见支架空隙均匀。
如图3和4所示,经生物力学检测可知,最大力为173.022N;压缩强度7.61MPa。
二、兔髌骨沟缺损模型构建
1.实验材料和方法
材料(货号,厂家):32只(多的用于备用)新西兰大白兔,2.2-2.5kg,雄性,厂家:北京华阜康生物科技股份有限公司,许可证号:SCXK(京)2019-0008,饲养条件(普通饲料饲养,温度20~26℃,湿度40%~70%)。
手术器械:刀片、手术剪、持针器、手术镊、缝合线、缝合针、显微剪、牙科钻等。
2.实验方法
具体造模方法:常规麻醉,麻醉起效后,对动物右后肢进行备皮。动物仰卧位固定在手术台上,对右后肢进行造模,先碘伏消毒,髌骨内侧打开关节囊,分离肌肉和韧带,暴露关节腔,然后在髌骨沟处用牙科钻做一个直径5mm,长3mm的缺损,如图5所示。
3.实验结果
HE染色及甲苯胺蓝染色结果分别如图6和图7所示,可见正常组关节软骨结构完整,关节面光滑,结构清晰,潮线完整;模型组关节软骨侵蚀,软骨面不平整,表层缺失,潮线严重缺失。
油红O染色和Masson结果分别如图8和9所示,可见正常组成纤维细胞数目少,密度低,胶原纤维排列整齐;模型组大量成纤维细胞及胶原纤维堆积,胶原纤维排列紊乱并伴有炎细胞浸润。
三、软骨组织脱细胞支架对兔髌骨沟缺损的影响
1.具体方法:在髌骨沟处用牙科钻做一个直径5mm,长3mm的缺损。然后把软骨组织脱细胞支架移植进入缺损处,如图10所示。
2.实验结果
HE染色和甲苯胺蓝染色结果分别如图11和图12所示,可见正常组关节软骨结构完整,关节面光滑,结构清晰,潮线完整;模型组关节软骨侵蚀,软骨面不平整,表层缺失,潮线严重缺失;水凝胶支架组关节软骨轻微损伤,见软骨面较不平整,潮线不完整。
油红O染色和Masson结果分别如图13和图14所示,可见正常组成纤维细胞数目少,密度低,胶原纤维排列整齐;模型组大量成纤维细胞及胶原纤维堆积,胶原纤维排列紊乱并伴有炎细胞浸润;水凝胶支架组相对模型组成纤维细胞及炎细胞减少,胶原纤维排列较紊乱。

1-4)将标本在37℃下用核酸酶溶液在10 mM Tris-HCl,pH =7.5中处理4h,核酸酶溶液含有50 U ml-1 DNase I和1 U ml-1RNase A;
1-5)将步骤1-4)中酶处理过的软骨浆液用1%Triton X-100溶液处理24小时后,再用低渗的Tris-HCl溶液洗涤20小时,以除去所有酶;
1-6)1×PBS冲洗步骤1-5)中制得的标本,再纯水冲洗标本,液氮30min,低温粉碎组织,以100目滤网筛选软骨微粒,真空冷冻干燥;
2)制备凝胶:将步骤1)制备好的软骨组织脱细胞支架粉末添加到1 mg·mL-1 的胃蛋白酶的0.01 M HCl溶液中,支架粉末:胃蛋白酶的重量比为10:1,并在室温下以恒定速度搅拌预定的消化时间,从48小时支架粉末的部分消化产生pH值为2~3的中等粘度溶液,通过在冰上中和之前添加适量的1×PBS,制备出浓度较低的支架粉末溶液,通过加入0.01 M NaOH溶液和1×PBS分别提供生理酸性和盐度,将中和的胃蛋白酶消化液在37℃下孵育以完成凝胶化;
3)制备凝胶复合支架:在室温下将支架浸泡在步骤2)中制备好的凝胶中4-6 h完成复合。
2.根据权利要求1所述的软骨组织脱细胞水凝胶支架的制备方法,其特征在于,步骤1-1)中,将软骨组织切成2mm3小块,以纯水冲洗5×3min,以去除杂质。
3.根据权利要求1所述的软骨组织脱细胞水凝胶支架的制备方法,其特征在于,步骤1-2)中,超声清洗机功率100%,20℃,20min,清洗3遍。
4.根据权利要求1所述的软骨组织脱细胞水凝胶支架的制备方法,其特征在于,步骤1-6)中,1×PBS冲洗标本,5×6min;纯水冲洗标本,5×6min。
5.权利要求1-4任意一项所述的软骨组织脱细胞水凝胶支架的制备方法制备得到的软骨组织脱细胞水凝胶支架。
