Abstract
本发明公开了一种肠道益生菌组合物及其应用,所述肠道益生菌组合物,按重量份计包括:乳双歧杆菌0.8‑5份、唾液乳杆菌0.2‑4份、瑞士乳杆菌0.2‑1.8份、凝结芽孢杆菌1.5‑3份、嗜热链球菌0.3‑1.5份、酪酸梭状芽孢杆菌0.1‑2份、保护剂1‑3份和益生元混合物2‑8份。所述应用为肠道益生菌制剂在制备食品、保健品或天然产品生产中的应用。本发明通过益生菌、益生元和保护剂的合理搭配,可以有效调节肠道菌群,维持肠道平衡,降低腹泻风险。同时相比现有技术,本发明可以最大限度的减少活菌的损失,提高益生菌的存活率,稳定存储时间长。

技术领域
本发明属于肠道益生菌技术领域,涉及一种肠道益生菌组合物及其应用。
背景技术
肠道为人体免疫器官之一,其内环境中寄存着庞大、丰富的微生物群体,它们与宿主免疫、代谢等功能一同进化,因而对维持内环境系统的平衡具有至关重要的作用。
日常饮食可以影响肠道菌群的组成。目前我国调节肠道菌群类的食品从原料组成上可分以益生菌为主要原料的活菌类食品和以益生元为主要原料的非活菌类食品。益生菌(Probiotics)是指能够促进肠内菌群生态平衡,对宿主起有益作用的活的微生物制剂。摄取适量的益生菌,能对宿主产生一种或多种特殊的功能性健康益处,因此,益生菌在功能食品及药品中有着广阔的应用前景。益生元(Prebiotics)是指不易被消化的食品成分,通过选择性的刺激一种或几种细菌的生长与活性,而对寄主产生有益的影响,从而改善寄主健康的物质。
目前利用益生菌和益生元调节肠道菌群已成为一个研究热点。CN113750113A公开了一种益生菌和益生元的组合物及其应用,所述组合物包括抗性糊精、菊粉、玉米淀粉、麦芽糊精、水苏糖、植物乳杆菌、嗜酸乳杆菌、两歧双歧杆菌、副干酪乳杆菌和甜味剂。该发明的益生菌和益生元的组合物优化配比,具有调节肠道菌群平衡,改善肠道健康的作用。
CN113712207A公开了一种提高胃肠道免疫能力的益生菌益生元组合物及其应用,所述组合物是一种由副干酪乳杆菌(Lactobacillus paracasei)与母乳低聚糖(HumanMilk Oligosaccharides)组成的营养组合物,所述副干酪乳杆菌为保藏编号为CGMCCNo.15139或DSM27447、和/或保藏编号为CGMCC No.15077的副干酪乳杆菌,所述母乳低聚糖包括2’岩藻糖基乳糖。该发明的组合物可有效提高胃肠道免疫能力。
现有的益生菌和益生元调节肠道菌群的组合物存储周期短,且存储过程中益生菌的存活率低,严重影响了其功效的发挥。
发明内容
本发明的目的在于提供一种肠道益生菌组合物及其应用,本发明通过将益生菌、益生元和保护剂的合理搭配,最大限度的减少肠道益生菌组合物中活菌的损失,提高活菌的存活率,可有效延长稳定存储时间。
为了实现上述目的,本发明采用以下技术方案:
本发明提供了一种肠道益生菌组合物,按重量份计包括:乳双歧杆菌0.8-5份、唾液乳杆菌0.2-4份、瑞士乳杆菌0.2-1.8份、凝结芽孢杆菌1.5-3份、嗜热链球菌0.3-1.5份、酪酸梭状芽孢杆菌0.1-2份、保护剂1-3份和益生元混合物2-8份。
优选地,所述乳双歧杆菌、唾液乳杆菌和酪酸梭状芽孢杆菌的质量比为1-4:0.5-2:1;进一步优选为3:1:1。
优选地,所述益生元混合物,按重量份计包括:菊粉0.3-2份、抗性糊精0.3-0.5份、木糖醇0.5-3份。
优选地,所述保护剂,按重量份计包括,可溶性大豆多糖10-20份、海藻糖5-10份、普鲁兰多糖3-8份、甘露醇1-3份和脱脂奶粉10-20份。
进一步优选地,所述可溶性大豆多糖、普鲁兰多糖和甘露醇的质量比为10-20:3-8:1-3;最优选地,所述可溶性大豆多糖、普鲁兰多糖和甘露醇的质量比为15:5:2。
上述的肠道益生菌组合物每份含有50亿以上的活菌数量(CFU)。
本发明还提供了上述的肠道益生菌组合物的制备方法,包括以下步骤:
(1)将益生菌和保护剂混合,得到混合料;
(2)向步骤(1)得到的混合料中加入益生元混合物、制粒,得所述肠道益生菌组合物。
本发明还提供了一种益生菌制剂,包括上述的肠道益生菌组合物或上述的制备方法制备的肠道益生菌组合物。
优选地,所述肠道益生菌制剂为颗粒剂、粉剂、胶囊或片剂。
本发明还提供了一种上述的肠道益生菌组合物或益生菌制剂在食品、保健品或天然产品生产中的应用。
优选地,所述食品包括乳制品、果冻、糖果和饼干。
本发明的有益效果为:
(1)本发明的肠道益生菌组合物通过益生菌、益生元合理搭配,可以有效调节肠道菌群,维持肠道平衡,同时,可以降低腹泻风险。
(2)本发明的肠道益生菌组合物,通过添加保护剂,可以最大限度的减少了活菌的损失,提高了活菌的存活率,且稳定存储时间长。

实施例1一种肠道益生菌组合物
益生菌组合物按重量份数计包括:乳双歧杆菌0.8份、唾液乳杆菌0.2份、瑞士乳杆菌0.2份、凝结芽孢杆菌1.5份、嗜热链球菌0.3份、酪酸梭状芽孢杆菌0.1份、保护剂1份和益生元混合物2份;
所述益生元混合物,按重量份计包括:菊粉0.3份、抗性糊精0.3份、木糖醇0.5份;
所述保护剂,按重量份计包括,可溶性大豆多糖10份、海藻糖5份、普鲁兰多糖3份、甘露醇1份和脱脂奶粉10份;
辅料,按重量份计包括:硬脂酸镁2份和二氧化硅3份。
将上述益生菌和保护剂混合搅拌均匀后,再与益生元混合物和辅料混合,制粒、整粒、压片,得到片剂,每片0.25g。
实施例2一种肠道益生菌组合物
益生菌组合物按重量份数计包括:乳双歧杆菌5份、唾液乳杆菌4份、瑞士乳杆菌1.8份、凝结芽孢杆菌3份、嗜热链球菌1.5份、酪酸梭状芽孢杆菌2份、保护剂3份和益生元混合物8份;
所述益生元混合物,按重量份计包括:菊粉2份、抗性糊精0.5份、木糖醇3份;
所述保护剂,按重量份计包括,可溶性大豆多糖20份、海藻糖10份、普鲁兰多糖8份、甘露醇3份和脱脂奶粉20份;
辅料,按重量份计包括:硬脂酸镁2份和二氧化硅3份。
将上述益生菌和保护剂混合搅拌均匀后,再与益生元混合物和辅料混合,制粒、整粒、压片,得到片剂,每片0.25g。
实施例3一种肠道益生菌组合物
益生菌组合物按重量份数计包括:乳双歧杆菌1份、唾液乳杆菌0.5份、瑞士乳杆菌0.5份、凝结芽孢杆菌1.8份、嗜热链球菌0.5份、酪酸梭状芽孢杆菌1份、保护剂1.2份和益生元混合物4份

所述益生元混合物,按重量份计包括:菊粉1.5份、抗性糊精0.4份、木糖醇1.5份;
所述保护剂,按重量份计包括,可溶性大豆多糖15份、海藻糖5份、普鲁兰多糖5份、甘露醇2份和脱脂奶粉15份;
辅料,按重量份计包括:硬脂酸镁2份和二氧化硅3份。
将上述益生菌和保护剂混合搅拌均匀后,再与益生元混合物和辅料混合,制粒、整粒、压片(压力为15KN),得到片剂,每片0.25g。
实施例5一种肠道益生菌组合物
益生菌组合物按重量份数计包括:乳双歧杆菌3份、唾液乳杆菌3份、瑞士乳杆菌1份、凝结芽孢杆菌2份、嗜热链球菌1.2份、酪酸梭状芽孢杆菌1.5份、保护剂2.8份和益生元混合物7份;
所述益生元混合物,按重量份计包括:菊粉1.5份、抗性糊精0.4份、木糖醇1.5份;
所述保护剂,按重量份计包括,可溶性大豆多糖15份、海藻糖5份、普鲁兰多糖5份、甘露醇2份和脱脂奶粉15份;
辅料,按重量份计包括:硬脂酸镁2份和二氧化硅3份。
将上述益生菌和保护剂混合搅拌均匀后,再与益生元混合物和辅料混合,制粒、整粒、压片,得到片剂,每片0.25g。

上述实施例和对比例所述的肠道益生菌组合物每份含有50亿以上的活菌数量(CFU)。
实验例1肠道益生菌组合物对肠道菌群的影响
1.1受试者选取:
一个月内未患过胃肠疾病、未服用抗生素者的成人。志愿者年龄为30-60岁。实验期间不服用任何同类产品,要求每日喝水量不少于1500mL白开水。
1.2实验分组与给药剂量
将志愿者按年龄随机分为10组,每组10人,分别服用实施例1-6和对比例1-4制备的肠道益生菌组合物。每日早、晚餐前30分钟分别各服用1g,持续30天。
1.3实验方法
受试前和受试后2h,无菌采取受试者粪便,利用16SrRNA肠道基因检测盒(来源于中国科学院微生物研究所微生物基因组学研究中心)检测粪便肠道菌群。
1.4检测结果
粪便中拟杆菌、普氏菌和金黄色葡萄球菌的基因表达量如表1所示。

由表1可知,服用本发明肠道益生菌组合物的志愿者,其肠道拟杆菌和普氏菌的含量较服用前增加,同时,金黄色葡萄球菌含量较服用前降低。表明本发明的肠道益生菌组合物可以调节肠道菌群,同时还可以降低腹泻风险。
实验例2肠道益生菌组合物存储稳定性
将实施例和对比例制备的肠道益生菌组合物,置于恒温恒湿培养箱,温度设置25℃,湿度设置60%RH,放置3个月后,对其乳双歧杆菌、唾液乳杆菌和酪酸梭状芽孢杆菌三者的活菌数进行检验,根据肠道益生菌组合物初始的活菌总数和三个月后的活菌总数,计算三者益生菌的存活率,如表2所示。

由表2可知,本发明的肠道益生菌组合在温度25℃,湿度60%RH的存储条件下放置3个月,乳双歧杆菌、唾液乳杆菌和酪酸梭状芽孢杆菌三者的存活率大于80%,表明本发明通过益生菌、益生元和保护剂的合理搭配,尤其是保护剂的添加,可以最大限度的减少肠道益生菌组合物中活菌的损失,提高活菌的存活率,延长存储时间。

1.一种肠道益生菌组合物,其特征在于,按重量份计包括:乳双歧杆菌3份、唾液乳杆菌1份、瑞士乳杆菌1.2份、凝结芽孢杆菌2.2份、嗜热链球菌0.8份、酪酸梭状芽孢杆菌1份、保护剂2份和益生元混合物5份,其中,所述益生元混合物,按重量份计包括:菊粉1.5份、抗性糊精0.4份、木糖醇1.5份;所述保护剂,按重量份计包括,可溶性大豆多糖15份、海藻糖5份、普鲁兰多糖5份、甘露醇2份和脱脂奶粉15份。
2.权利要求1所述的肠道益生菌组合物的制备方法,包括以下步骤:
(1)将益生菌和保护剂搅拌混合,得到混合料;
(2)向步骤(1)得到的混合料中加入益生元混合物混合制粒,得所述益生菌组合物。
3.一种肠道益生菌制剂,其特征在于,包括权利要求1所述的肠道益生菌组合物或权利要求2所述的制备方法制备的肠道益生菌组合物。
4.根据权利要求3所述的肠道益生菌制剂,其特征在于,所述肠道益生菌制剂为颗粒剂、粉剂、胶囊或片剂。
