Abstract
本发明公开了血小板衍生生长因子‑BB在制备治疗骨关节炎药物中的应用和药物组合物,将PDGF‑BB应用在骨关节炎治疗过程中,用于促进软骨细胞增殖、胞外基质修复及其软骨形成作用。PDGF‑BB能够显著促进ATDC5细胞增殖及胞外基质的合成,促进ATDC5细胞的软骨形成作用。预示PDGF‑BB有望作为促进软骨修复的药物,为开发骨关节炎治疗药物奠定基础。
Description
技术领域
本发明涉及医疗领域,具体是指血小板衍生生长因子-BB在制备治疗骨关节炎药物中的应用和组合物。
背景技术
骨关节炎(osteoarthritis,OA)是一种常见的、致残慢性病,被世界卫生组织称为“不死的癌症”。OA是老年人致残的首要因素,给患者带来了痛苦、造成患者行动不便,最终严重降低了患者的生活质量。此外,世界上每年花在治疗OA上的相关医疗费用达到3000亿美元左右,给整个患者和社会带来沉重的经济负担。
OA实际上是一种软骨退化性疾病,最终常常引起包括滑膜增生、软骨下骨硬化及关节边缘骨质增生等在内的整个关节的病变。目前临床上尚缺乏有效的OA治疗手段。当前临床上无论是口服非甾体抗炎药、还是关节腔注射皮质类固醇药物,透明质酸等药物,均只能减轻症状和改善关节功能,无法阻止OA病情进展。鉴于软骨退变是OA的主要病理改变,因此,亟需开发有效促进软骨修复的治疗药物以阻遏OA的进展。
血小板衍生生长因子(Platelet Derived Growth Factor,PDGF)是多种间充质来源细胞包括成纤维细胞、血管平滑肌细胞及软骨细胞等的有丝分裂原,能促进其分裂、迁移及增殖。其中,血小板衍生生长因子-BB(Platelet Derived Growth Factor-BB,PDGF-BB),被发现能促进软骨细胞的增殖、迁移以及胞外基质(extracellular matrix,ECM)的合成并且能抑制IL-1β (interleukin-1β)引起的软骨降解。此外,PDGF-BB还能够减少体内软骨细胞的凋亡、促进体内软骨再生及缺损软骨的修复。经检索,有关PDGF-BB在促进软骨细胞增殖,促进软骨细胞胞外基质合成及在促进软骨修复的药物中应用的专利尚未见报道。
发明内容
针对现有技术中的上述不足,本发明提出了血小板衍生生长因子-BB(PDGF-BB)在制备促进软骨细胞增殖、促进细胞ECM合成,治疗骨关节炎药物中的应用。
血小板衍生生长因子-BB在制备治疗骨关节炎药物中的应用,将PDGF-BB应用在骨关节炎治疗过程中,用于促进软骨细胞增殖、胞外基质修复及其软骨形成作用。
作为优选的,所述PDGF-BB能够有效促进软骨细胞增殖的浓度为10ng/ml。
作为优选的,所述PDGF-BB能够有效促进软骨细胞软骨形成作用的浓度为2-10ng/ml。
一种治疗骨关节炎的药物组合物,上述任一项的PDGF-BB的试剂或药物,以及至少一种药学上可接受的载体或赋形剂。
上述生长因子PDGF-BB,促进软骨细胞增殖、促进软骨细胞胞外基质合成的细胞生物学实验及结果如下:
1.生长因子PDGF-BB对ATDC5软骨细胞系存活率的影响。
实验组:无血清培养的条件下,分别加入浓度为1,2,5,10ng/ml的PDGF-BB处理小鼠软骨发生细胞系ATDC5细胞48h后,cck-8实验检测细胞存活率变化。阴性对照为等量的0.1%BSA。
结果表明:浓度为1,2,5,10ng/ml的PDGF-BB处理ATDC5细胞48h后能显著提高ATDC5细胞的存活率。
2.生长因子PDGF-BB对ATDC5细胞增殖的影响。
实验组:无血清培养的条件下,加入浓度为10ng/ml的PDGF-BB处理ATDC5细胞24h后,Edu实验检测细胞增殖变化。阴性对照为等量的0.1%BSA。
结果表明:浓度为10ng/ml的PDGF-BB处理ATDC5细胞24h后能显著促进ATDC5细胞增殖。
3.生长因子PDGF-BB对ATDC5细胞软骨形成作用的影响。
实验组:(alcian blue实验)无血清培养的条件下,分别加入浓度为2,5ng/ml的PDGF-BB 处理小鼠软骨发生细胞系ATDC5细胞72h后,alcian blue实验检测蛋白多糖聚糖含量变化; (western blot实验)无血清培养的条件下,分别加入浓度为2,5,10ng/ml的PDGF-BB处理ATDC5细胞30min或24h后,western blot实验检测软骨标志物的蛋白水平变化;(qPCR 实验)无血清培养的条件下,分别加入浓度为2,5ng/ml的PDGF-BB处理ATDC5细胞24h 后,qPCR实验检测软骨标志物的mRNA水平变化。阴性对照均为等量的0.1%BSA。
结果表明:在上述浓度和条件下,PDGF-BB能显著促进ATDC5细胞外基质的合成。
本发明的有益效果为:
PDGF-BB能够显著促进ATDC5细胞增殖及胞外基质的合成,促进ATDC5细胞的软骨形成作用。预示PDGF-BB有望作为促进软骨修复的药物,为开发骨关节炎治疗药物奠定基础。

为使本发明实施例的目的、技术方案和优点更加清楚,下面将结合本发明实施例中的附图,对本发明实施例中的技术方案进行清楚、完整地描述,显然,所描述的实施例是本发明一部分实施例,而不是全部的实施例。通常在此处附图中描述和示出的本发明实施例的组件可以以各种不同的配置来布置和设计。
实施例一:PDGF-BB处理ATDC5细胞后cck-8法检测细胞的存活率。
(1)以3000个/孔的密度将ATDC5细胞均匀铺于96孔板中。
(2)第二天将孔内的培养基换为无血清培养基,并加入不同浓度的PDGF-BB处理细胞 48h(每24h更换一次培养基并重新添加PDGF-BB)。
(3)每孔加入10ul cck-8试剂,37°培养箱继续孵育3-4h。
(4)用酶标仪测定450nm波长处光吸收的OD值。
结果表明:用浓度为1,2,5,10ng/ml的PDGF-BB处理ATDC5细胞48h后能显著提高ATDC5细胞的存活率,见图1。
实施例二:PDGF-BB处理ATDC5细胞后Edu实验检测细胞的增殖情况。
(1)以5*105个/孔的密度将ATDC5细胞均匀铺于12孔板中。
(2)第二天将孔内的培养基换为无血清培养基,并加入不同浓度的PDGF-BB处理细胞24h。
(3)按照Edu增殖检测试剂盒说明书进行Edu实验。
结果表明:用浓度为10ng/ml的PDGF-BB处理ATDC5细胞24h后能显著显著促进ATDC5 细胞增殖,见图2、图3。
实施例三:PDGF-BB处理ATDC5细胞后alcian blue实验检软骨细胞分泌的蛋白多糖聚糖的含量变化。
(1)以5*10^5个/孔的密度将ATDC5细胞均匀铺于24孔板中。
(2)第二天待细胞达到100%的汇合度时,将孔内的培养基换为无血清培养基,并加入不同浓度的PDGF-BB处理细胞72h(每24h更换一次培养基并重新添加PDGF-BB)。
(3)弃培养基,4%多聚甲醛室温固定15min。
(4)1x PBS漂洗3遍。
(5)1x alcian blue染液室温染30min。
(6)1x PBS漂洗3遍,把多余的染料洗掉。
(7)普通光学显微镜下拍照。
(8)加入200μl的6M的盐酸胍溶液,室温溶解8h,用酶标仪测定620nm波长处光吸收的OD值。
结果表明:用浓度为2,5ng/ml的PDGF-BB处理ATDC5细胞72h后能显著促进软骨细胞分泌蛋白多糖聚糖,见图4、图5。
实施例四:PDGF-BB处理ATDC5细胞后western blot实验检测软骨标志物的蛋白水平变化。
(1)以合适的密度将ATDC5细胞均匀铺于6cm细胞培养皿中。
(2)第二天将皿内的培养基换为无血清培养基,并加入不同浓度的PDGF-BB处理细胞 30min或24h。
(3)收集细胞,RIPA细胞裂解液裂解细胞。
(4)根据试剂盒说明,BCA法测定样品的蛋白浓度。
(5)取适量的蛋白样品,通过western blot实验检测软骨标志物Sox9及Acan的蛋白水平。
结果表明:用浓度为2,5,10ng/ml的PDGF-BB处理ATDC5细胞24h能后能显著提高Sox9及Acan的蛋白水平,见图6。
实施例五:PDGF-BB处理ATDC5细胞后qPCR实验检测软骨标志物的mRNA水平变化。
(1)以合适的密度将ATDC5细胞均匀铺于6cm培养皿中。
(2)第二天将皿内的培养基换为无血清培养基,并加入不同浓度的PDGF-BB处理细胞 24h。
(3)收集细胞,用TRIzol试剂盒裂解细胞,并根据说明书提取RNA。
(4)根据反转录试剂盒说明书将RNA反转录为cDNA。
(5)通过用SYBR Green Master Mix试剂盒进行qPCR实验,检测软骨标志物Sox9、Acan 及Col2a1的mRNA水平。
结果表明:用浓度为2,5ng/ml的PDGF-BB处理ATDC5细胞24h后能显著提高软骨标志物Sox9、Acan及Col2a1的mRNA水平,见图7。
上述的实验数据统计学处理:实验数据以平均值±标准误差表示,经t检验:“*,P<0.05”,为有统计学差异;“**,P<0.01”,为有显著统计学差异;“***,P<0.001”,为有极其显著的统计学差异。
由上述实验及其结果,可以得出如下结论:
PDGF-BB能够显著促进ATDC5细胞增殖及胞外基质的合成,促进ATDC5细胞的软骨形成作用。预示PDGF-BB有望作为促进软骨修复的药物,为开发骨关节炎治疗药物奠定基础。

1.血小板衍生生长因子-BB在制备治疗骨关节炎药物中的应用,其特征在于:将PDGF-BB应用在骨关节炎治疗过程中,用于促进软骨细胞增殖、胞外基质修复及其软骨形成作用。

4.根据权利要求1所述的血小板衍生生长因子-BB在制备治疗骨关节炎药物中的应用,其特征在于:所述PDGF-BB促进ATDC5细胞外基质合成有效浓度至少为2ng/ml。
5.根据权利要求1所述的血小板衍生生长因子-BB在制备治疗骨关节炎药物中的应用,其特征在于:所述PDGF-BB提高ATDC5细胞软骨形成作用的有效浓度至少为2ng/ml。
6.一种治疗骨关节炎的药物组合物,其特征在于,包括权利要求1-5任一项所述的PDGF-BB的试剂或药物,以及至少一种药学上可接受的载体或赋形剂。
