Abstract
本发明公开了鹿茸干细胞及其外泌体在制备抗疤痕产品中的应用,属于生物技术领域;通过将鹿茸干细胞以及由该鹿茸干细胞获取的鹿茸干细胞外泌体作用于伤口,并对伤口外观、伤口愈合速度、伤口皮肤再生、伤口中的胶原水平、创面中的肌成纤维细胞数量几项指标进行观察或检测,可知鹿茸干细胞与鹿茸干细胞外泌体均能够有效发挥抗疤痕效果,且鹿茸干细胞外泌体的抗疤恒效果远胜于骨髓间充质干细胞外泌体,由此体现鹿茸干细胞及鹿茸干细胞外泌体在抗疤痕方面能够发挥显著效果。

技术领域
本发明涉及生物技术领域,特别是鹿茸干细胞及其外泌体在制备抗疤痕产品中的应用。
背景技术
干细胞(stem cells,SC)是一类具有超强自我复制能力(self-renewing)的多潜能细胞,在一定条件下,它可以分化成多种功能细胞;具有修复、再生各种组织器官的潜在功能,医学界称之为“万用细胞”。
而干细胞外泌体是干细胞在生理活动过程中分泌出来的生理活性物质,其具有脂质膜结构,膜内包含多种生物活性物质,如蛋白质、多肽、microRNA以及DNA等。从干细胞培养基中提取出干细胞外泌体,并在最大程度上保存其活性,提取出来的外泌体活性几乎接近100%。研究表明,干细胞以及干细胞来源的外泌体在抗疤痕方面取得了不错的效果。
然而,不同动物以及不同组织中的干细胞及其外泌体在抗疤痕方面的作用效果差异很大,且因组织来源和伦理制约等因素,导致干细胞及其外泌体获取不便。
鹿茸作为哺乳动物中唯一能够周期性再生的复杂器官,可以作为研究附肢再生的动物模型。钙化的鹿茸(即鹿角)每年春天从角柄(鹿头上永久性的桩儿)脱落下来,在其顶端形成一个直径可超过10cm的伤口,这个伤口可在10天之内完成再生性愈合,几乎不留疤痕,实属罕见。研究发现,顶端皮肤伤口的再生性愈合,依赖于其下侧鹿茸再生芽基组织的存在,而鹿茸干细胞是此组织中最重要的细胞,因此鹿茸干细胞可能是伤口再生性愈合的关键刺激因素。同时,相较常用的骨髓间充质干细胞、脐带间充质干细胞等干细胞,鹿茸干细胞增殖速度更快(因为其支撑了鹿茸每天近2cm的生长速度)和更易于获取(因为鹿茸每年都可以再生,从而获取细胞)。
发明内容
本发明的目的在于提供鹿茸干细胞及其外泌体在制备抗疤痕产品中的应用,以最大程度实现抗疤痕效果。
本发明的目的是通过以下技术方案来实现的:
一种鹿茸干细胞在制备抗疤痕产品中的运用。
鹿茸顶端皮肤伤口的再生性愈合,依赖于其下侧鹿茸再生芽基组织的存在,而鹿茸干细胞是此组织中最重要的细胞,其再生能力强,本发明通过对鹿茸干细胞进行抗疤痕方面的测试,发现其在抗疤痕方面能够发挥显著的效果。
作为本发明的优选方案,所述鹿茸干细胞的制备方法如下:
取鹿茸再生芽基组织,将其切成组织块,并用PBS清洗,其后将组织块置于I/II型混合胶原酶中进行水浴消化,待消化结束后,去除混合胶原酶并用DMEM培养基清洗;其后用移液管将组织块转移分散至培养瓶底面,将培养瓶倒转底面朝上,进行细胞的原代培养;待细胞贴壁后,将培养瓶翻转正面朝上,并加入培养基,待细胞铺满瓶底时获得所述鹿茸干细胞(英文:Antler stem cells,简称:AnSCs)。
上述方案中,取鹿茸再生芽基组织,将其切成组织块,并经PBS清洗、水浴消化、DMEM培养基清洗后进行原代培养,获得鹿茸干细胞。通过选取鹿茸再生芽基作为制备鹿茸干细胞的原材料,能够使获得鹿茸干细胞具备更多的生物活性因子,在抗疤痕方面具有更加的效果;同时因鹿茸干细胞的增殖速度很快,为分泌鹿茸干细胞外泌体打下坚实的基础。
作为本发明的优选方案,所述I/II型混合胶原酶中I型胶原酶(200U/ml)与II型胶原酶(150U/ml)的质量比为80:65。通过I/II型混合胶原酶,能够提高组织块的消化速度,使其中的胶原纤维更容易从组织中分离出。
作为本发明的优选方案,所述原代培养中,细胞的培养体系为:10%FBS+DMEM培养基+5%双抗;培养条件为:37℃+5%CO2+饱和湿度。
通过进一步限定培养体系和培养条件,能够保证细胞有充足的营养和最佳的生长环境,同时能够抑制培养过程中细菌的产生,进而促进细胞的生长。
本发明还提供了一种鹿茸干细胞外泌体在制备抗疤痕产品中的运用。
鹿茸干细胞的增殖速度极快,通过鹿茸干细胞分泌的外泌体与鹿茸干细胞一样,均在抗疤痕方面发挥极为显著的效果。

S1将所述AnSCs在含10%FBS的完全培养基中进行培养,待细胞长至密度为70-80%时,更换为无血清培养基,继续进行培养;
S2待培养结束后将细胞培养上清液与外泌体提取试剂以4:1(v:v)的比例加入到离心管内,颠倒混匀后进行孵育,其后进行离心并收集离心后的沉淀物,用100μl PBS均匀吹打沉淀物,待充分混后转移至新的离心管中并进行离心,离心后的上清液即为所述鹿茸干细胞外泌体(英文:Antler stem cell-derived exosomes,简称:AnSC-exos)。
上述方案中,通过优化鹿茸干细胞外泌体制备过程中的参数和步骤,使制得的鹿茸干细胞外泌体纯度更高、更匀称,提高鹿茸干细胞外泌体的品质,进而提高其在抗疤痕方面的效果。
作为本发明的优选方案,所述孵育的温度为4℃,孵育时间为12h。
通过进一步限定孵育时间和孵育温度,能够提高外泌体的制备效率以及提高外泌体的品质和纯度,进而提高其在抗疤痕方面的效果。
本发明的有益效果是:
本发明是在鹿茸生物学特性及一系列鹿茸生物学研究的基础上提出来的,具有充实的理论基础和科学依据;本发明制备的鹿茸干细胞和鹿茸干细胞外泌体均能够提高伤口的愈合速度,且其能减少疤痕的形成和促进毛的再生;还能够促进损伤皮肤中皮肤附属器(毛囊和皮脂腺等)的再生;此外,鹿茸干细胞外泌体还能够降低愈合皮肤伤口中的胶原水平并改善其结构组成,包括诱导胶原呈不规则的网篮状排列、高表达促再生胶原基因、低表达促疤痕胶原基因;且无论在愈合中期还是后期中,均能减少创面中的肌成纤维细胞数量。

具体实施方式
下面结合实施例进一步详细描述本发明的技术方案,但本发明的保护范围不局限于以下。
实施例1AnSCs的制备
在鹿角自然脱落(角柄伤口形成)的第5天,切取角柄顶端组织(约1cm)。然后于无菌环境下,用手术刀及鼠牙镊剥离外侧皮肤,并摘除皮下结缔组织,暴露出再生芽基,更换新刀片轻轻切取芽基组织(避免切到其他组织),将芽基组织切成长约0.7mm的立方体组织块,PBS清洗3次;将组织块置于事先配好的I/II型混合胶原酶(配置方法见表1)中,37℃水浴消化30min后去除胶原酶,并用含10%FBS的DMEM培养基清洗3次灭活胶原酶;用移液管将这些组织块转移分散至T25培养瓶底面,将培养瓶倒转底面朝上(尽量少加培养基,防治其倒滴),进行细胞的原代培养,待细胞贴壁(12-24h)后,将培养瓶翻转正面朝上,加入正常量的培养基,7天后,细胞铺满瓶底,制得所述鹿茸干细胞。
其中,原代培养的细胞培养体系为:10%FBS+DMEM培养基+5%双抗;培养条件为:37℃+5%CO2+饱和湿度。
表1混合胶原酶配制
上述制备过程中:鹿茸再生芽基形成于角柄伤口出现(鹿角脱落)后2-5天,位于角柄顶端的皮肤内侧,呈新月形(图1A)。芽基组织块儿经胶原酶消化后,接种于培养瓶,显微镜下可观察到组织块边缘有重叠样的球形细胞,且培养基中有散在的细胞。培养24h后,可观察到细胞伸出伪足,攀附于培养瓶壁,呈成纤维样漩涡状生长(图1B)。流式细胞术及细胞免疫荧光结果显示,AnSCs高表达间充质干细胞标志物CD73、CD90和CD105以及胚胎干细胞标志物SOX2,但不表达血液细胞标志物CD34和CD45(图1C、图1D)。成骨诱导分化21-30天后,显微镜下观察,发现细胞呈漩涡状交织分布,其上散落矿化结晶,茜素红S染色后结晶呈橙红色有折光性,符合钙盐结晶特征,可认为AnSCs具有成骨分化能力(图1E)。成脂诱导分化21-30天后,显微镜下观察,发现细胞内出现圆形小泡,胞外出现大小不一的圆形集落,油红O染色后可见圆形小泡和集落均被染成透亮的红色,符合脂肪滴形态,可认为AnSCs具有成脂分化能力(图1F)。
综上结果说明,再生芽基来源的AnSCs可以被定义为干细胞。
实施例2AnSC-exos的制备
如图2A所示,将实施例1中制备的AnSCs培养1-3代,其后在含10%FBS的完全培养基中培养,待细胞长至密度为70-80%时(处于对数生长期),吸去完全培养基,用PBS冲洗3次,其后更换为无血清培养基,继续培养72h,再吸取细胞培养上清液于50ml离心管中。将所述细胞培养上清液与外泌体提取试剂(货号41201ES25,上海Yeasen)以4:1(v:v)的比例一起加入到50ml离心管内,颠倒混匀,4℃孵育12h;然后10,000g离心70min(4℃),轻轻倒掉上清液,收集沉淀;其后取100μl PBS均匀吹打沉淀物,使其充分混匀后,转移至新的1.5ml离心管中,并在4℃下,12,000g离心2min,弃沉淀,上清液即为AnSC-exos溶液,用BCA法测定浓度后,保存于-80℃冰箱中。
将制备的AnSC-exos溶液用BCA法蛋白定量检测,发现每离心500ml上清液可获得AnSC-exos约3000ug/ml;用粒径分析仪对粒径进行分析,结果显示AnSC-exos颗粒直径大小约在50-150nm范围内(图2B),符合外泌体规定的大小;透射电镜拍照观察AnSC-exos的形态,发现其为盘状囊泡,直径约在120nm左右(图2C),也符合外泌体的形态特征;Westernblot结果显示AnSC-exos表达外泌体的经典标志物CD9、CD63和TSG101(图2D)。
综上结果说明,所提取的物质确定为外泌体,可用于后续实验研究。
实验例
(1)大鼠皮肤全皮层缺损模型的建立
实验大鼠于SPF级动物饲养室中自由饮水采食,适应性生存一周后进行皮肤损伤建模。以3%的戊巴比妥钠麻醉大鼠(30mg/kg;腹腔注射),然后用宠物剃毛器剔除大鼠背毛,经1%碘伏及75%乙醇常规消毒后,用直径为12mm的环形皮肤取样器切除背部正中位置的全皮层皮肤,建立大鼠背部皮肤全皮层缺损模型。术后给予青霉素消炎(20000U/kg;肌注;每天一次,持续3天)和曲马多镇痛(50mg/kg;肌注;每天一次,持续3天)。
(2)分组与处理
建模后的大鼠随机分为4组,每组8只,分别为:
阴性对照组(CTRL),给予PBS处理;
鹿茸干细胞对照组(AnSCs),给予2×106AnSCs/只处理;
骨髓间充质干细胞外泌体对照组(BMSC-exos),给予50μg/只剂量的BMSC-exos处理;
鹿茸干细胞外泌体处理组(AnSC-exos),给予50μg/只剂量的AnSC-exos处理;
各组处理均每7天一次。
2.结果
(1)AnSCs和AnSC-exos促进皮肤伤口的愈合速度
首先,建立直径为12mm的大鼠背部皮肤全皮层缺损模型;然后给予AnSCs及AnSC-exos治疗,并以1×PBS溶液和BMSC-exos作为对照(图3A)。术后观察到试验大鼠伤口仅有少量组织液和血液渗出,无感染、化脓、水肿及坏死等病理反应发生。每4天拍照观察愈合过程中伤口外观形态的变化,随着时间延长所有伤口面积均逐渐减小,最后自然结痂愈合(图3B)。伤口面积的统计结果显示,相较于对照组,AnSCs和AnSC-exos治疗能显著促进伤口的愈合速度(图3C)。
进一步对比愈合后的伤口,发现AnSCs、BMSC-exos和AnSC-exos在外观形态上均能减少疤痕的形成和促进毛的再生,但是AnSCs和AnSC-exos的效果要强于BMSC-exos(图3B)。
综上结果说明,AnSC s和AnSC-exos均能够促进伤口的愈合速度,且其能减少疤痕的形成和促进毛的再生,且效果强于BMSC-exos。
(2)AnSCs和AnSC-exos诱导皮肤附属器的再生
HE染色结果表明,在POD28,所有组都已完成再上皮化,可以观察到表皮、真皮和皮下组织。值得注意的是,在CTRL组并未发现有明显的皮肤附属器再生;而在AnSCs组和AnSC-exos组则发现有大量的毛囊再生,其由不规则致密的真皮结缔组织包饶,毛囊显示髓质、皮质,含有表皮的内根鞘、外根鞘和玻璃膜;同时还发现有大量的皮脂腺再生,其由分泌部和排泄部组成,外由不规则致密的真皮结缔组织包饶。分泌部由腺细胞(内侧)和基细胞(外侧)组成,排泄部与毛囊等附属器相接(图4A)。统计附属器的数目(图4B),结果显示每HPF,CTRL、AnSCs、BMSC-exos和AnSC-exos分别有1.33±1.58、84.67±
5.03、13.00±1.00和23.00±2.65个。接下来,进一步利用免疫荧光染色对毛囊和皮脂腺的角色进行了确认,结果显示,CTRL组中几乎没有表达毛囊标志物CK14和皮脂腺标志物CK19,而在AnSCs组和AnSC-exos组则有大量的阳性表达(图4C-E)。qRT-PCR结果也显示AnSC组和AnSC-exos组CK14和CK19的相对表达量显著高于CTRL组和BMSC-exos组(图4F,图4G)。
综上结果说明,AnSC和AnSC-exos均能够促进损伤皮肤中皮肤附属器(毛囊和汗腺等)的再生,且效果强于BMSC-exos。
(3)AnSC和AnSC-exos改善胶原纤维的结构及组成
Masson染色结果显示(图5A),CTRL组愈合皮肤中的胶原呈横向平行的束状排列;而AnSC组和AnSC-exos组的胶原则为不规则的网篮状排列,一般包饶在皮肤附属器的周围。然后对胶原水平进行分析(图5A,图5B),CTRL组的愈合皮肤中累积了大量的胶原,其胶原水平显著高于其余3组(P<0.0001);但是这3组之间并无显著性差异。通过qRT-PCR法检测了胶原的主要组成(图5C,图5D),结果显示,Collagen I含量在各组之间无显著性差异;而Collagen III的含量却出现了显著性差异,其中CTRL组的含量显著高于AnSCs、BMSC-exos和AnSC-exos组(P<0.0001,P<0.001和P<0.001);进一步分析测Collagen I和CollagenIII的比值(图5E),结果显示CTRL组的比值显著高于其他3组(P<0.0001),但是这3组之间并无显著性差异。
随后通过qRT-PCR法检测了胶原合成主要诱导因子TGF-β1和TGF-β3的水平(图5F,图5G),结果显示,CTRL组中的TGF-β1含量显著高于AnSCs、BMSC-exos和AnSC-exos组(P<0.01,P<0.05和P<0.05);但是这3组之间并无显著性差异;TGF-β3含量显著低于AnSCs、BMSC-exos和AnSC-exos组(P<0.0001,P<0.0001和P<0.001);进一步分析TGF-β1和TGF-β3的比值(图5H),结果显示CTRL组的比值显著低于其余3组(P<0.01),但是这三组之间并无显著性差异。
最后通过qRT-PCR法检测了诱导胶原降解的主要基质酶MMP1和MMP3的水平(图5I,J),结果显示,CTRL组中的MMP1含量显著高于其余3组(P<0.0001),进一步发现AnSC-exos组中的含量显著高于BMSC-exos(P<0.01),但是却低于AnSCs组(P<0.001)。与MMP1相似,CTRL组中的MMP3含量也显著高于其余3组(P<0.0001,P<0.01和P<0.001)。
综上结果说明,AnSCs和AnSC-exos均能够降低愈合皮肤伤口中的胶原水平;并改善其结构组成,包括诱导胶原呈不规则的网篮状排列、高表达促再生胶原基因、低表达促疤痕胶原基因。
(4)AnSC和AnSC-exos抑制成纤维细胞向肌成纤维细胞的分化
对导致疤痕形成最主要的细胞,即肌成纤维细胞的含量进行了检测(通过检测其标志物α-SMA与TAGLN)。免疫荧光染色结果显示,在CTRL和BMSCs组中有大量α-SMA+细胞聚集,而在AnSCs组和AnSC-exos组中α-SMA+细胞数量则已经非常少,其显著少于CTRL和BMSC-exos组(图6A,B;P<0.0001)。与α-SMA+相似,TAGLN+细胞也在CTRL和BMSC-exos组中大量聚集,而在AnSCs组和AnSC-exos组则也非常少,也显著少于CTRL和BMSC-exos组(图6A,C;P<0.0001)。α-SMA+细胞和TAGLN+细胞数量在AnSCs和AnSC-exos组之间并无显著性差异。随后通过qRT-PCR和Western blot分别在mRNA转录水平和蛋白表达水平进行了验证。结果显示,相较于CTRL和BMSC-exos组,AnSCs和AnSC-exos均显著降低了α-SMA(图6D)和TAGLN(图6E)的mRNA水平。同时,相较于CTRL组,AnSC-exos显著降低了α-SMA(图6F,图6G;P<0.0001)和TAGLN(图6F,图6H;P<0.0001)的蛋白水平;但是α-SMA和TAGLN的蛋白水平在AnSCs、BMSC-exos和AnSC-exos组之间并无显著性差异。
综上结果说明,AnSCs和AnSC-exos治疗均能减少创面中的肌成纤维细胞数量,而这可能是其抑制疤痕形成和促进附属器再生的关键。
通过上述四个实验可知:AnSC-exos在抗疤痕方面的作用效果远胜于BMSC-exos,进而体现出不同的干细胞外泌体在抗疤痕方面的作用相差较远,由此体现本发明中的AnSC及AnSC-exos在抗疤痕方面的显著作用。
以上仅是本发明的优选实施方式,应当理解本发明并非局限于本文所披露的形式,不应看作是对其他实施例的排除,而可用于各种其他组合、修改和环境,并能够在本文构想范围内,通过上述教导或相关领域的技术或知识进行改动。而本领域人员所进行的改动和变化不脱离本发明的精神和范围,则都应在本发明所附权利要求的保护范围内。
Claims (6)
1.一种鹿茸干细胞外泌体在制备抗疤痕产品中的运用。
2.根据权利要求1所述的一种鹿茸干细胞外泌体在制备抗疤痕产品中的运用,其特征在于;所述鹿茸干细胞外泌体的制备方法包括如下步骤:
S1将鹿茸干细胞在含10%FBS的完全培养基中进行培养,待细胞长至密度为70-80%时,更换为无血清培养基,继续进行培养;
S2待培养结束后将细胞培养上清液与外泌体提取试剂以体积比4:1的比例加入到离心管内,颠倒混匀后进行孵育,其后进行离心并收集离心后的沉淀物,用100 μl PBS均匀吹打沉淀物,待充分混后转移至新的离心管中并进行离心,离心后的上清液即为所述鹿茸干细胞外泌体。
3.根据权利要求2所述的一种鹿茸干细胞外泌体在制备抗疤痕产品中的运用,其特征在于,所述孵育的温度为4℃,孵育时间为12 h。
4.根据权利要求1所述的一种鹿茸干细胞外泌体在制备抗疤痕产品中的运用,其特征在于;所述鹿茸干细胞的制备方法包括如下步骤:
取鹿茸再生芽基组织,将其切成组织块,并用PBS清洗,其后将组织块置于I/II型混合胶原酶中进行水浴消化,待消化结束后,去除混合胶原酶并用DMEM培养基清洗;其后用移液管将组织块转移分散至培养瓶底面,将培养瓶倒转底面朝上,进行细胞的原代培养;待细胞贴壁后,将培养瓶翻转正面朝上,并加入培养基,待细胞铺满瓶底时获得所述鹿茸干细胞。
5.根据权利要求4所述的一种鹿茸干细胞外泌体在制备抗疤痕产品中的运用,其特征在于,所述I/II型混合胶原酶中,200U/ml 的I型胶原酶与150U/ml 的II型胶原酶的质量比为80:65。
6.根据权利要求4所述的一种鹿茸干细胞外泌体在制备抗疤痕产品中的运用,其特征在于,所述原代培养中,细胞的培养体系为:10% FBS + DMEM培养基 + 5%双抗;培养条件为:37℃ + 5%CO2+ 饱和湿度。
