(1) Mixing surfactant polyoxyethylene hydrogenated castor oil, co-surfactant anhydrous alcohol and aromatic Chinese medicinal essential oil at a certain proportion, stirring for 10min to mix well;
(2) Adding the alcohol extract of guarana, and stirring for 10min until the mixture is clear and transparent;
(3) Adding purified water with the prescription amount into the obtained product, and continuously stirring for 20min to obtain the compound essential oil guarana composite nanoemulsion with clear color.
The stirring speed is 600rpm/min.
The application of the disinfection composition containing the aromatic Chinese medicinal essential oil and the guarana extract comprises the application of disinfection on kitchen boards, hand sterilization and disinfection, sterilization cream, sterilization gel and the like.
Compared with the prior disinfection technology, the disinfection composition containing the aromatic traditional Chinese medicine essential oil and the guarana extract has the beneficial effects that: the composition has the effects of remarkable antibacterial effect, strong antibacterial effect and good application prospect. The pure aromatic Chinese medicinal essential oil disinfectant can completely kill common pathogenic bacteria within 30s and dominant putrefying bacteria within 3 h; compared with the disinfection effect of the pure essential oil disinfectant, the compound aromatic Chinese medicine essential oil compatible guarana air disinfectant has the effect of 100% inactivation on common pathogenic bacteria in the air within 30s and completely killing dominant putrefying bacteria within 30min, and has the inhibition effect on the pathogenic bacteria within 15s better than that of the pure essential oil disinfectant, so that the disinfection effect is quick and strong, and the compound aromatic Chinese medicine essential oil compatible guarana air disinfectant is a green strong disinfection product. The materials are natural, nontoxic, high in safety factor, capable of being used in human environment and free of residue after being acted. Wherein, the alcohol content is below 30%, effectively prevents the disinfectant from mildewing, is easy to transport, and avoids accidents such as explosion caused by overhigh alcohol concentration of the disinfectant. In addition, the multiple applications provided by the invention provide products with the functions of sterilization, bacteriostasis and drug resistance for different disinfection channels, and meet the requirements of different industries.
The technical scheme of the invention has the following functions of various aromatic traditional Chinese medicines:
folium artemisiae argyi: the dry leaves of the Artemisia argyi of the Compositae family are bitter, warm and pungent in nature, have the functions of expelling cold and dampness, warming channels, regulating qi and blood, stopping bleeding, preventing miscarriage and the like, and have various clinical effects of antibiosis, anti-inflammation, antioxidation and the like. The main chemical components of the composition comprise volatile oil, flavonoid, tannin, triterpenes, eudesmane, polysaccharide, trace elements and the like. Researches show that the folium artemisiae argyi has the effect of preventing diseases and plagues, and has obvious inhibition effects on tubercle bacillus, bacillus subtilis, escherichia coli, staphylococcus aureus and the like. Modern researches have considered that the folium artemisiae argyi has certain analgesic effect besides inhibiting or killing various fungi, bacteria and viruses.
Rhizoma atractylodis: dried rhizome of Atractylodes lancea or Atractylodes lancea of Compositae. Is pungent and bitter in flavor, and has the effects of eliminating dampness, strengthening spleen, dispelling wind and cold and improving eyesight. Research shows that the main components of the rhizoma atractylodis include volatile oil components such as atractylone, paleoxol and beta-cineol, and the rhizoma atractylodis has an inhibiting effect on the growth of pathogenic bacteria such as staphylococcus aureus, escherichia coli, candida albicans, trichophyton rubrum and the like. Rhizoma Atractylodis can destroy organelles, change cell structure, and kill bacteria. Experimental data show that the rhizoma atractylodis volatile oil spray, fumigation liquid and fumigant can inhibit bacteria such as staphylococcus aureus, escherichia coli, pseudomonas aeruginosa and the like in the air. Meanwhile, the rhizoma atractylodis can also play a role in killing influenza viruses by inhibiting virus DNA.
Patchouli: the dry aerial parts of Pogostemon cablin of Labiatae, pungent in flavor and slightly warm in nature, have the effects of aromatizing, resolving dampness, regulating the middle warmer, relieving vomit, relieving exterior syndrome and relieving summer-heat. The main components of the patchouli are volatile oil: patchouli alcohol, alpha-patchouli alkene, beta-patchouli alkene, and the like. Modern researches show that the product has the effects of resisting bacteria, fungi, plasmodium and plant pathogenic fungi, regulating gastrointestinal motility, promoting secretion of digestive juice, protecting intestinal barrier function, regulating immunity, etc. It has broad-spectrum antibacterial activity, and has effects of inhibiting and even killing various bacteria and fungi, and has certain inhibiting ability on Escherichia coli, bacillus subtilis, staphylococcus albus, and Staphylococcus aureus.
Herba Eupatorii: the Eupatorium fortunei of Compositae has dry aerial parts, pungent taste, and effects of eliminating dampness with aromatics, activating spleen, stimulating appetite, relieving exterior syndrome, and relieving summer-heat. Pharmacological research shows that the medicine has the functions of resisting inflammation, inhibiting bacteria, resisting tumor, eliminating phlegm, resisting tumor, strengthening immunity, exciting stomach smooth muscle, etc. The traditional Chinese medicine composition is clinically used for treating coronary heart disease, summer-heat and dampness cold, infant rotavirus enteritis, infant diarrhea and the like. The eupatorium contains essential oil: para-cymene, aurantiol acetate, thymol methyl ether, etc. Research shows that the water decoction has the effect of inhibiting diphtheria, staphylococcus aureus, sarcina, proteus and typhoid bacillus. The volatile oil and monomer components including cymene and neryl acetate have direct inhibitory effect on influenza virus.
Clove: the dried flower bud of Eugenia caryophyllata of Myrtaceae has pungent and warm natured taste, and has effects of warming spleen and stomach, lowering adverse qi, dispelling cold, relieving pain, warming kidney and tonifying yang. Modern research shows that the essential oil mainly contains the following volatile oil: eugenol, acetyl eugenol, beta-caryophyllene, methyl n-amyl ketone, methyl salicylate, etc., wherein eugenol is one of the effective components in clove volatile oil, and the decoction has the inhibiting effect on staphylococcus, streptococcus and bacilli such as diphtheria, deformity, pyocyanea, large intestine, etc. The clove bacteriostasis mechanism is that the permeability of bacteria is increased, the thallus morphology is abnormal, cell structures such as cell walls, cell membranes and the like are damaged, a large amount of cell contents are leaked out, even cell malformation occurs, and the clove bacteriostasis mechanism has the inhibiting effect on bacteria such as escherichia coli, staphylococcus aureus, candida albicans, helicobacter pylori, escherichia coli and the like and viruses.
Heartleaf houttuynia herb: fresh whole plant or dried aerial parts of houttuynia cordata of Saururaceae, is pungent and slightly cold in nature, and has effects of clearing heat and detoxicating, resolving carbuncle and expelling pus, and inducing diuresis for treating stranguria. Modern researches show that the cordate houttuynia mainly contains volatile oil, and the houttuynine sodium bisulfite contained in the volatile oil has different degrees of inhibition effects on various gram positive and negative bacteria such as staphylococcus aureus, diplococcus pneumoniae, streptococcus a, influenza bacillus and the like.
Cinnamon: the dry bark of cinnamon belonging to Lauraceae family is hot, pungent and sweet in flavor. Modern researches show that the main components of the traditional Chinese medicine composition are cinnamyl aldehyde, salicylaldehyde and the like, and the traditional Chinese medicine composition has the effects of dispelling wind and invigorating stomach, promoting blood circulation to remove blood stasis, dispelling cold and relieving pain, diminishing inflammation and sterilizing and the like.
Melon and sodium: also known as balsamine, belongs to the family of soapberry, is produced in the tropical plants of the brazilian amazonian basin, is used in the fields of medical care, food industry, cosmetics and the like, and has a wide range of consumer groups. Mainly contains chemical components such as alkaloid, polyphenol, tannic acid, etc., and the main medicinal parts are fruits and seeds. Modern researches show that the guarana has antioxidation and antibacterial effects, and has certain effects in resisting fatigue, improving memory, resisting depression, resisting tumor, etc.

Adding water to make up the balance, magnetically stirring at 600rpm for 20min, mixing, sterilizing, and bottling to obtain the compound essential oil air spray disinfectant.

Adding the 15% alcohol extract of guarana, and magnetically stirring at 600rpm for 10 min. Adding water to supplement the balance, magnetically stirring at 600rpm for 20min, mixing, sterilizing, and bottling to obtain the compound essential oil guarana air spray disinfectant.
The following are toxicity evaluation, irritation and sterilization experiments performed on the disinfectants prepared in the above examples to illustrate the effects of the present invention:
1. acute toxicity test animals: 40 SPF-grade KM mice are purchased from the animal experiment center of the university of traditional Chinese medicine in Jiangxi, the male and female are half, and the weight is 18-22g.
The experimental method comprises the following steps: one-time maximum test is carried out according to the specification of 2002 edition 'disinfection technical specification', for example, the one-time gavage dosage of 5000mg/kg & BW for 20 animals (each half of a male and a female), no death is caused in 14 days, and LD can be judged50Greater than 5000mg/kg body weight. The evaluation rules are: LD (laser diode)50The people with the weight more than 5000mg/kg are actually nontoxic and LD50The patients with more than 501mg/kg-5000mg/kg body weight belong to low toxicity and LD50The body weight of 51mg/kg-500mg/kg belongs to the medium toxicity and LD501mg/kg-50 mg/kg of body weight belongs to high toxicity and LD50Those with the weight less than 1mg/kg are extremely toxic.
The experimental steps are as follows: the test was randomized into 3 test groups (examples 1, 5, 7) and 1 blank control group, and fasted overnight prior to testing using a one-time maximal test. The gavage dose of the test group was 6000mg/kg BW, and the same amount of sterile water was given to the control group. After gavage, the animals were observed for signs of intoxication and number of deaths and time of death, recorded for 14d, and dead animals and animals after the observation were necropsied and histopathologically examined.
As a result: no death occurred in 14d, and no other abnormal conditions existed. According to the classification standard of acute toxicity of the disinfectant in 2002 edition of disinfection technical Specification of the Ministry of public health of the people's republic of China, the tested samples 1, 5 and 7 are all practically nontoxic.
2. Skin and mucosa irritation test the irritation/corrosion effect and strength of the disinfectant on the skin of the test animals. Experimental animals: healthy rabbits or guinea pigs with intact skin are purchased from the animal experiment center of the university of traditional Chinese medicine in Jiangxi.
The experimental steps are as follows: referring to the disinfection technical Specification, 2002 edition, ministry of health of the people's republic of China, new Zealand rabbits were used for a complete skin irritation test and a skin breakage test, respectively, and for an acute eye irritation test. At 24h before the test, hairs on both sides of the spine of the rabbit or guinea pig are removed by depilatory, the hair removal range is about 3cm × 3cm on the left and right, respectively, and the skin is not damaged. The next day, the test substance (stock solution 0.5 ml) was directly dropped on the intact skin with hair removed on one side of 3cm × 3cm area, covered with a layer of gauze, and fixed with non-irritating adhesive tape. The depilated skin on the other side was used as a blank control and the application time was 4h. After the test is completed, the residual test substance is removed with warm water or a non-irritating solvent. Local skin reactions were observed at 1h, 24h and 48h after removal of the test substance, respectively, and stimulation responses were scored for performance. Each 3 new zealand rabbits were grouped. Setting an experimental group and a blank control group, wherein 0.5mL of stock solution is smeared on the experimental group of the skin irritation test, and 0.1mL of stock solution is dripped on the experimental group of the acute eye irritation test.
As a result: in a one-time medicine application complete skin experiment, local skin reactions of New Zealand rabbits for 1 hour, 24 hours and 48 hours are observed, and no erythema or edema phenomenon is found; in a skin damage experiment of a New Zealand rabbit, the back wound of the New Zealand rabbit is found to be normally scabbed, and the wound recovery is not influenced; eye mucosa stimulation is carried out on New Zealand rabbits, liquid medicines are respectively dripped, and observation is carried out after 1h, 24h, 48h, 72h and 7d of eye dripping, and the observation shows that the experimental group is normal in the 1h period except for the blood streak of eyes of individual rabbits. It can thus be determined that: the disinfectants prepared in the examples 1, 5 and 7 of the present invention have no irritation to the skin and mucous membranes.
The effect of the invention is further illustrated by combining the experiment of the bacteriostatic rate of the disinfectant:
7 antibacterial zone test experiment of essential oil
Pouring agar cooled to 40 ℃ into the plate, standing and condensing for later use. Formulation 1X105Sucking 200ul of the strain suspension (see Table 1 for specific strains), putting into a plate, coating with a coating rod, standing for 5min, and naturally drying at room temperature. 10ul of essential oil is dripped into each filter paper sheet, and the dripped surface is uniformly placed on a bacteria-containing plate with 3 sheets per plate. And (4) carrying out inverted culture at 37 ℃ for 16-18h, observing the result, and measuring the diameter of the inhibition zone. The test results are shown in table 1.

as can be seen from the in vitro antibacterial data in Table 1 and FIG. 1, the volatile oils have good antibacterial effect, wherein the clove has the strongest antibacterial ability, has good killing effect on Escherichia coli and Staphylococcus aureus, has the same antibacterial effect as the positive drug gentamicin, has the diameter of the antibacterial zone of more than 20, and shows high sensitivity to bacteria. Therefore, through orthogonal design experiments, the essential oil is mixed in proportion, and test results show that the folium artemisiae argyi essential oil can be obtained: patchouli essential oil: eupatorium essential oil: houttuynia essential oil: clove essential oil: cinnamon essential oil =3:1:1:3:3:4, mixing according to the volume ratio, and providing prescription data support for the preparation of the subsequent Chinese herbal compound essential oil melon radon antibacterial spray, wherein the mixture is a better antibacterial compatibility combination.
3. Disinfection Effect test
The sterilization effect of the traditional Chinese medicine disinfectant on selected pathogenic bacteria is tested by adopting the standard WS/T650-2019 in the above embodiments 1,3, 5 and 7, the stock solution of the disinfectant is used for actual measurement according to the standard so as to ensure the consistency of the concentrations of the samples, and the sterilization effect of each embodiment is compared and analyzed.
The bacterial suspension prepared in advance was taken and diluted to about 5.0X 10 with PBS5CFU/mL~4.5×106CFU/mL bacterial suspension is ready for use. Taking a sterile test tube, adding 5.0mL of sample (using stock solution), placing the sterile test tube in a water bath kettle at the temperature of 20 +/-1 ℃ for 5min, then adding 0.1mL of test bacterial suspension, quickly mixing uniformly and immediately timing. After the test bacteria and the sample interact for 15s, 0.5min, 1.5min, 2.5min and 5min, respectively sucking 1.0mL of test bacteria and sample mixed solution for inoculation2 plates, and pouring the culture medium (when the bacterial count can not be counted, performing 10-fold serial dilution by PBS, selecting appropriate dilution, respectively sucking 1.0mL to inoculate 2 plates, and performing live bacterial culture counting). At the same time, PBS was used in place of the sample for parallel testing as a positive control. The number of colonies recovered from the positive control was 1.0X 104CFU/mL~9.0×104CFU/mL. And (5) inoculating the PBS and the culture medium of the same batch into a culture dish. All test samples and control samples are cultured at 36 +/-1 ℃, and bacterial propagules are cultured for 48h to observe the final result; the Candida albicans is cultured for 72h to observe the final result. The test is repeated for 3 times, and the bacteriostasis rate is calculated.
The results of the 15s bacteriostasis test of examples 1,3, 5 and 7 on 4 bacteria are as follows:

According to the in vitro bacteriostasis rate test experiment result, the aromatic Chinese medicinal essential oil air spray disinfectant and the aromatic Chinese medicinal essential oil compatible guarana air disinfectant prepared by the method have excellent bactericidal effects, and have obvious killing effects on common pathogenic bacteria white staphylococcus in the air, common pathogenic bacteria escherichia coli and staphylococcus aureus on the surface of an object. The disinfectant stock solutions and the bacterial suspensions in the embodiments 1,3, 5 and 7 are respectively acted for different times of 0.5min, 1.5min, 2.5min and 5min, and the results show that all pathogenic bacteria can be killed within 0.5min, which indicates that the compound essential oil and the compound essential oil compatible guarana alcohol extract both have extremely excellent bactericidal action. On the basis of the research, the disinfection effect of the 4 examples on several common pathogenic bacteria is researched by shortening the time (15 s) and diluting the stock solutions (the dilution times of examples 1 and 3 are 2; and the dilution times of examples 5 and 7 are 5), the results are shown in table 2, after the disinfectants are diluted, the disinfectants still have strong inhibition effect on the 4 pathogenic bacteria, and the two concentrations of the essential oil compatible guarana extract disinfectants have higher bacteriostasis rate and better effect than pure essential oil disinfectants.
According to the invention, five common aromatic traditional Chinese medicines of folium artemisiae argyi, clove, patchouli, fortune eupatorium herb and cinnamon are preferably selected from ancient and modern sachets, and the heartleaf houttuynia herb essential oil and the guarana alcohol extract are optimally added on the basis of compatibility of the traditional Chinese medicines.
4. Test of Corrosion prevention Effect
Inhibiting effect of disinfectant on dominant putrefying bacteria
The disinfectant prepared in examples 5 and 6 and a normal saline control group are respectively subjected to determination of bacteriostatic efficacy test method of lactobacillus according to China pharmacopoeia 1121 of 2020 edition by using stock solution.
The experimental steps are as follows: preparing lactobacillus (Lactobacillus acidophilus) within 5 generations with sterile normal saline solution to obtain a solution containing 10 bacteria per 1ml8Bacterial suspension of cfu. The disinfectant prepared in the 2 examples and the control group are respectively absorbed by 5.0ml in a centrifugal tube, different bacterial strains are added into each tube, and the inoculation bacterial quantity in 1g or 1ml of test article is 105-106cfu, the volume of the inoculated bacterial liquid is not more than 1 percent of the volume of the test sample, and the inoculated bacterial liquid is fully mixed and then placed at 20 to 25 ℃ for dark storage. Then, the number of viable bacteria was measured, and 1ml of the sample was taken out from the centrifugal tube at intervals shown in the graph, and the number of bacteria contained in the sample was measured. MRS agar was poured. The invention adopts the common aromatic traditional Chinese medicine essential oil of ancient and modern sachets to be compatible with the guarana alcohol extract, and the guarana alcohol extract is uniformly mixed with the polyoxyethylene hydrogenated castor oil and the absolute ethyl alcohol to prepare the nano-emulsion disinfectant, and the effect of the traditional Chinese medicine compound essential oil disinfectant of different embodiments is evaluated through the sterilization effect on 4 different pathogenic bacteria of escherichia coli, staphylococcus aureus, staphylococcus albus and candida albicans. The experimental result shows that the disinfectant containing aromatic Chinese medicinal essential oil with the concentration of 3 percent or more has obvious bacteriostatic action.
