本发明涉及一种复方茶油消斑油的制备方法,包含以下原料:人参、当归、白芷、白及、僵蚕、马齿苋、紫草、珍珠母。其步骤为:将复配中药研磨成分,过筛,采用索式提取法,对复配中药的脂溶性成分进行提取。将脂溶性活性成分添加于茶油中,并辅以硬脂酰表没食子儿茶素为油脂抗氧化剂,制备消斑油。本发明绿色、天然、无毒,具有很高的食用及药用价值。同时,所用中药材皆具有排毒、养颜、调节新陈代谢的作用,诸药配伍祛斑作用快、效果显著,通过调节表皮细胞的新陈代谢,提高面部肌肤的活力。

技术领域
本发明涉及一种复方茶油消斑油的制备方法。
背景技术
面部色素斑种类繁多,比如:黄褐斑、雀斑、老年斑等,是一种常见的发生于面部的后天性色素沉着过渡性皮肤病。本病主要与内分泌失调有关,由于雌激素分泌紊乱,导致皮肤代谢减弱,血液循环不畅,造成黑色素沉积,最终导致皮肤变得昏暗,衰老,甚至产生褶皱。
然而,目前的皮肤增白护肤品大多将避光剂涂抹于皮肤表层,以达到美白祛斑的作用,该方法“治标不治本”,效果不佳,且具有一定的副作用。
发明内容
本发明要解决的技术问题是:提供一种复方茶油消斑油的制备方法,该方法以茶油为基质,辅以脂溶性黄酮为油脂抗氧化剂,并添加多位中草药,以期达到协同祛斑的作用;该方法制备工艺简单、成本低廉、使用方便,所添加物皆为天然中草药成分,祛斑效果明显。
解决上述技术问题的技术方案是:一种复方茶油消斑油的制备方法,包括如下重量组分:人参0.1~20份,当归0.2~25份,白芷0.5~20份,白及0.2~25份,僵蚕0.1~20份,马齿苋0.5~20份,紫草2~20份,珍珠母1~45份;
包括如下制备步骤:
(a)按以上配比取人参、当归、白芷、白及、僵蚕、马齿苋、紫草和珍珠母粉碎,过筛,得混合药粉备用;
(b)将混合药粉置于索式提取装置中,随后以乙醇、石油醚或乙醚作为提取溶剂,利用冷凝回流法于45~65℃条件下提取8~18小时,提取混合药粉中的脂溶性活性成分,得酯溶性化合物LSCs;
(c)将10~30mg表没食子儿茶素与15~50mg硬脂酸混合,再加入酶催化剂得反应混合物,所述酶催化剂为Lipozyme CALB脂肪酶、Novozyme435脂肪酶或Lipozyme TLIM脂肪酶,酶催化剂的用量为反应混合物总质量的1.0~5.0%;在温度为40~50℃情况下催化反应12~48小时,得硬脂酸表没食子儿茶素;
(d)按酯溶性化合物LSCs∶硬脂酸表没食子儿茶素∶茶油的质量比为1:1:100~1:1:50计,将酯溶性化合物LSCs及硬脂酸表没食子儿茶素添加于茶油中混合均匀,得复方茶油消斑油。
进一步的,所述一种复方茶油消斑油的制备方法,优选如下重量组分:人参1~15份,当归2~20份,白芷2~20份,白及5~25份,僵蚕1~10份,马齿苋0.5~10份,紫草5~20份,珍珠母2~30份。
进一步的,所述一种复方茶油消斑油的制备方法,优选如下重量组分:如下重量组分:人参1~10份,当归5~10份,白芷2~10份,白及5~15份,僵蚕5~10份,马齿苋4~10份,紫草5~10份,珍珠母10~30份。

茶油又名茶子树、茶油树,因其种子可榨茶油供食用而得名,具有清热解毒,活血散瘀,止痛等功效。在民间,茶油可被用于治疗少儿尿布疹,亦可用于治疗烧烫伤,具有很高的药用价值。
硬脂酸表没食子儿茶素为表没食子儿茶素经硬脂酸经酯化改性后的产物。表没食子儿茶素为黄烷三醇类物质,是葡萄籽原花青素中的一种单体,具有很高的抗氧化活性。有研究表明,其抗氧化活性为抗坏血酸的20倍(Uchida et al., Med Sci Res, 1980)。以硬脂酸为酯化剂,取代表没食子儿茶素分子结构中的羟基,以增强分子的脂溶性,可拓展其在油脂领域的应用。提高茶油稳定性的同时,增强了其抗氧化、抗炎、抗菌等活性。
人参属多年草本植物,分布于我国东北部高海拔地区,具有很强的药用价值,内服可治疗心血管疾病、肝、胃及消化系统疾病,亦可治疗癌症。外敷可促进伤口愈合,具有消炎、消肿等功效。
当归别名干归、秦哪,其根可入药。具有补血、活血、调经止痛、润燥滑肠等功效。当归中含有的丁基苯酞可增加血液流动,从而促进细胞代谢。
白芷产于我国西南、东北、华北等地,其根可入药,有祛病除湿、排脓生肌、活血止痛等功效,可改善人体微循环,促进皮肤新陈代谢。
白及属地生草本植物,广阔分布于江苏、安徽、浙江等地,味苦,性微寒,外用可治出血,疮疡肿毒,皮肤皲裂,具有收敛止血、消肿生肌的功效。
僵蚕别名天虫、姜蚕,是一味常用中药,具有祛风解痉,化痰散结的功效。古籍记载,其通散之能,可治疗风疮瘾疹、皮肤瘙痒等。
马齿苋广泛分布于全世界温带和热带地区,其草可供药,具有清热利湿、解毒消肿、消炎、止渴、利尿等作用。
紫草是多年草本植物,味苦,性寒,具有凉血,活血,清热,解毒的功效,可治治温热斑疹,湿热黄疸,紫癜等症。
珍珠母又名珠牡丹、珠母。据《中国医学大辞典》记载,其具有滋肝阴,清肝火的功效。以其涂面,可令人润泽,祛斑,逆转肌肤。
本发明将复配中药研磨成粉,过筛,采用索式提取法,对复配中药的脂溶性成分进行提取。将脂溶性活性成分添加于茶油中,并辅以硬脂酰表没食子儿茶素为油脂抗氧化剂,制备消斑油。本发明绿色、天然、无毒,具有很高的食用及药用价值。同时,所用中药材皆具有排毒、养颜、调节新陈代谢的作用,诸药配伍祛斑作用快、效果显著,通过调节表皮细胞的新陈代谢,提高面部肌肤的活力。
下面,结合实施例对本发明之一种复方茶油消斑油的制备方法的技术特征作进一步的说明。
具体实施方式
实施例1
(a)取人参5份,当归20份,白芷15份,白及10份,僵蚕15份,马齿苋20份,紫草8份,珍珠母25份,粉碎,过筛(用80目筛网过滤3次),得混合药粉备用。
(b)将混合药粉置于索式提取装置中,随后以乙醇作为提取溶剂,料液比1:10,利用冷凝回流法于65℃提取8小时,得到混合药粉中的LSCs。
(c)将15 mg表没食子儿茶素与30 mg硬脂酸混合,再加入固定化脂肪酶CALB(Lipozyme CALB脂肪酶)为酶催化剂,得反应混合物,反应混合物中酶催化剂浓度为2.0%(w/w),采用酶法酯化表没食子儿茶素,于40℃下反应12小时,定向引入硬脂酸酯,合成脂溶性黄酮(即硬脂酸表没食子儿茶素),并以此作为油脂抗氧化剂,提高茶油的货架期及生物活性。
(d)将LSCs及硬脂酸表没食子儿茶素按一定质量比例添加于茶油中(1:1:100)混合均匀,制备得复方茶油消斑油。
实施例2
(a)取人参15份,当归10份,白芷5份,白及20份,僵蚕10份,马齿苋18份,紫草14份,珍珠母40份,粉碎,过筛,得混合药粉备用。
(b)将混合药粉置于索式提取装置中,随后以石油醚作为提取溶剂,料液比1:10,利用冷凝回流法于55℃下提取10小时,得到混合药粉中的LSCs。
(c)将20 mg表没食子儿茶素与20 mg硬脂酸混合,再加入诺维信脂肪酶435(Novozyme435脂肪酶)为酶催化剂,得反应混合物,反应混合物中酶催化剂浓度为3.0%(w/w),采用酶法酯化表没食子儿茶素,于50 ℃下反应18小时,定向引入硬脂酸酯,合成脂溶性黄酮(即硬脂酸表没食子儿茶素),并以此作为油脂抗氧化剂,提高茶油的货架期及生物活性。
(d)将LSCs及硬脂酸表没食子儿茶素按一定质量比例添加于茶油中(1:1:75)混合均匀,制备得复方茶油消斑油。
实施例3
(a)取人参10份,当归10份,白芷10份,白及10份,僵蚕10份,马齿苋10份,紫草10份,珍珠母30份,粉碎,过筛,得混合药粉备用。
(b)将混合药粉置于索式提取装置中,随后以乙醚作为提取溶剂,料液比1:10,利用冷凝回流法于45℃下提取12小时,得到复配药粉中的LSCs。
(c)将15 mg表没食子儿茶素与15 mg硬脂酸混合,再加入诺维信脂肪酶TLIM(Lipozyme TLIM脂肪酶)为酶催化剂,得反应混合物,反应混合物中酶催化剂浓度为2.5%(w/w),采用酶法酯化表没食子儿茶素,于50 ℃下反应24小时,定向引入硬脂酸酯,合成脂溶性黄酮(即硬脂酸表没食子儿茶素),并以此作为油脂抗氧化剂,提高茶油的货架期及生物活性。
(d)将LSCs及硬脂酸表没食子儿茶素按一定质量比例添加于茶油中(1:1:50)混合均匀,制备得复方茶油消斑油。
本发明各实施例所用茶油为广西三江茶油。作为一种变换,也可以采用其他地区生产的茶油。
试用效果统计
1、一般数据:
选择90名实验者,均为女性患者,年龄在25岁~45岁之间,平均(35.1±1.5)岁;将90例女性患者随机分为治疗组和对照组。
治疗组:采用本发明实施例1制备得到的复方茶油消斑油均匀涂抹于色斑处,每晚1次,第二天洗净,连续用三十天,
对照组:外用维生素E软胶囊(广州白云山星群药业,规格100 mg/粒),将维生素E软胶囊挤出,均匀涂抹于色斑处,具体用法同治疗组。
2、疗效评定标准:
主要的判定依据为中国中西医结合学会皮肤性病专业委员会色素病学组制定的黄褐斑的疗效判定标准。
基本痊愈:采用肉眼视色斑面积消退>90%,颜色基本消退。
显效:采用肉眼视色斑面积消退>60%,颜色呈现明显变淡的趋势。
有效:采用肉眼视色斑面积消退>30%,颜色变淡。
无效:采用肉眼视色斑面积消退<30%,颜色基本无明显的变化。
3、结果统计

治疗组总有效率显著高于对照组,P<0.005,表示差异具有统计学意义。
典型病例
(1)覃某,女,30岁,自妊娠后颜面颊部出现黄褐色斑点,3年间逐渐扩大成片。使用本发明实施例1制备得到的复方茶油消斑油产品,于晚睡前涂抹于色斑处,第二天洗去,一月后皮损逐渐消退,颜面肤色正常。
(2)汤某,女,45岁,两年前因单位部门人员调整,加之经常熬夜加班,致面部出现黄褐色斑片,使用本发明实施例1制备得到的复方茶油消斑油产品,于睡前涂抹于患处,并遵医嘱调节作息,一月后面部黄褐斑基本消退。
货架期对比试验
对照例1、2、3的制备过程分别与本发明实施例1、2、3相同,不同之处在于:对照例1、2、3中省略步骤(c),步骤(d)是将LSCs按一定质量比例添加于茶油中混合均匀,制备得复方茶油消斑油。
分别对本发明实施例1、2、3和对照例1、2、3的货架期进行了考察,考察方法参照GB/T 21121-2007标准,结果如下表所示。

从上表可以看出,本发明加入合成脂溶性黄酮(即硬脂酸表没食子儿茶素)后,能够显著延长产品的货架期。

Claims (4)

1.一种复方茶油消斑油的制备方法,其特征在于:包括如下重量组分:人参0.1~20份,当归0.2~25份,白芷0.5~20份,白及0.2~25份,僵蚕0.1~20份,马齿苋0.5~20份,紫草2~20份,珍珠母1~45份;
包括如下制备步骤:
(a)按以上配比取人参、当归、白芷、白及、僵蚕、马齿苋、紫草和珍珠母粉碎,过筛,得混合药粉备用;
(b)将混合药粉置于索式提取装置中,随后以乙醇、石油醚或乙醚作为提取溶剂,利用冷凝回流法于45~65℃条件下提取8~18小时,提取混合药粉中的脂溶性活性成分,得酯溶性化合物LSCs;
(c)将10~30mg表没食子儿茶素与15~50mg硬脂酸混合,再加入酶催化剂得反应混合物,所述酶催化剂为Lipozyme CALB脂肪酶、Novozyme435脂肪酶或Lipozyme TLIM脂肪酶,酶催化剂的用量为反应混合物总质量的1.0~5.0%;在温度为40~50℃情况下催化反应12~48小时,得硬脂酸表没食子儿茶素;
(d)按酯溶性化合物LSCs∶硬脂酸表没食子儿茶素∶茶油的质量比为1:1:100~1:1:50计,将酯溶性化合物LSCs及硬脂酸表没食子儿茶素添加于茶油中混合均匀,得复方茶油消斑油。
2.根据权利要求1所述的一种复方茶油消斑油的制备方法,其特征在于:包括如下重量组分:人参1~15份,当归2~20份,白芷2~20份,白及5~25份,僵蚕1~10份,马齿苋0.5~10份,紫草5~20份,珍珠母2~30份。
3.根据权利要求2所述的一种复方茶油消斑油的制备方法,其特征在于:包括如下重量组分:人参1~10份,当归5~10份,白芷2~10份,白及5~15份,僵蚕5~10份,马齿苋4~10份,紫草5~10份,珍珠母10~30份。
4.根据权利要求1-3任一项所述的一种复方茶油消斑油的制备方法,其特征在于:步骤(a)中,过筛的具体操作为用80目筛网过滤3次。
