Detailed Description
In order to make the objects, technical solutions and advantages of the present invention more apparent, the present invention is further described in detail with reference to the following embodiments. It should be understood that the specific embodiments described herein are merely illustrative of the invention and are not intended to limit the invention.
Aiming at the problems in the prior art, the invention provides a preparation method of a traditional Chinese medicine for treating diabetes, and the invention is described in detail with reference to the accompanying drawings.
The traditional Chinese medicine for treating diabetes provided by the embodiment of the invention comprises, by mass, 20-50 parts of a bitter gourd extract, 20-35 parts of a corydalis tuber extract, 15-30 parts of astragalus membranaceus, 15-30 parts of coptis chinensis, 15-30 parts of radix rehmanniae recen, 10-20 parts of poria cocos, 8-15 parts of a red peony root extract, 6-12 parts of cornus officinalis, 5-8 parts of oldenlandia diffusa, 3-5 parts of hairyvein agrimony, 2-4 parts of uncaria and the balance of sterilized deionized water.
As shown in fig. 1, the preparation method of the traditional Chinese medicine for treating diabetes provided by the embodiment of the invention comprises the following steps:
s101, obtaining fresh bitter gourd, corydalis tuber decoction pieces and red paeony root; slicing fresh fructus Momordicae Charantiae, drying, pulverizing, adding water, extracting with low temperature water in extraction tank, concentrating under reduced pressure, separating, and adsorbing to obtain fructus Momordicae Charantiae extract;
s102, carrying out reflux extraction on the obtained rhizoma corydalis decoction pieces by using ethanol, concentrating, centrifuging, loading by using macroporous resin, eluting, concentrating and drying to obtain a rhizoma corydalis extract; soaking radix Paeoniae Rubra in water, decocting, extracting, adsorbing, eluting, distilling under reduced pressure to remove solvent, and drying to obtain radix Paeoniae Rubra extract;
s103, weighing radix astragali, rhizoma coptidis, radix rehmanniae, poria cocos, cornus officinalis, oldenlandia diffusa, hairyvein agrimony, uncaria, bitter gourd extract, rhizoma corydalis extract and red paeony root extract in proportion;
s104, crushing and extracting the weighed astragalus, coptis, radix rehmanniae, poria, cornus officinalis, oldenlandia diffusa, hairyvein agrimony and uncaria, removing impurities, adding the balsam pear extract, the corydalis tuber extract, the red paeony root extract and sterilized deionized water, uniformly stirring, drying and granulating to obtain the traditional Chinese medicine for treating diabetes.
As shown in fig. 2, the preparation of the momordica charantia extract in step S101 provided by the embodiment of the present invention includes:
s201, screening fresh bitter gourds, slicing the whole fruits, drying, and crushing to 80-150 meshes to obtain bitter gourd powder;
s202, uniformly mixing the bitter gourd powder with water, and placing the mixture in an extraction tank for low-temperature water extraction to obtain an extracting solution;
s203, filtering the extracting solution under reduced pressure to obtain filtrate, concentrating the filtrate under reduced pressure to obtain extract of the bitter gourd extract, performing ceramic membrane separation by using a ceramic pot, and adsorbing impurities to obtain the bitter gourd extract.
The bitter gourd powder and water provided by the embodiment of the invention are mixed according to the weight ratio of 1: and (3) uniformly mixing the components in a mass ratio of 2-5.
The filtrate provided by the embodiment of the invention is decompressed and concentrated to the relative density of 1.15-1.20, so as to obtain the extract of the balsam pear extract.

The concentration of the ethanol provided by the embodiment of the invention is 45-90 wt%.
As shown in fig. 4, the preparation of the red peony root extract in step S102 provided by the embodiment of the present invention includes:
s401, taking red paeony root, adding water with the weight 8-10 times that of the red paeony root, soaking for 2-4 hours, and then decocting;
s402, sequentially extracting the decoction obtained in the step S401 by using petroleum ether, ethyl acetate and n-butanol, and collecting extract liquor;
and S403, adsorbing and eluting the extract liquor by macroporous resin, then carrying out reduced pressure distillation to remove the solvent, and drying to obtain the red paeony root extract.
The decoction frequency provided by the embodiment of the invention is 3-5 times.
As shown in fig. 5, the preparation of the traditional Chinese medicine for treating diabetes in step S104 according to the embodiment of the present invention includes:
s501, respectively crushing astragalus membranaceus, coptis chinensis, radix rehmanniae, poria cocos, cornus officinalis, oldenlandia diffusa, hairyvein agrimony and uncaria rhynchophylla, sieving the crushed materials with a 120-200-mesh sieve, and uniformly mixing the materials to obtain mixed raw material powder;
s502, adding 8-12 times of ethanol by weight into the mixed raw material powder, extracting, concentrating under reduced pressure, performing ceramic membrane separation by using a ceramic tank, and adsorbing by using macroporous resin to remove impurities;
s503, adding the balsam pear extract, the corydalis tuber extract, the red paeony root extract and the sterilized deionized water into the extract after impurity removal in sequence, stirring uniformly, drying and granulating to obtain the traditional Chinese medicine for treating diabetes.
The technical effects of the present invention will be further described below in conjunction with pharmacological analysis.
The pulp of dogwood fruit and the tuckahoe can promote the brain health, the uncaria can relax blood vessels and prevent thrombosis, and the uncaria has obvious effect particularly on brain parts.
The bitter gourd extract: contains steroid saponin such as momordicin, insulin-like peptide and alkaloid, which can endow Momordica charantia with hypoglycemic activity. The main component of the momordica charantia extract can stimulate the release of insulin and reduce the concentration of glucose in blood, so the momordica charantia extract can play a good role in controlling blood sugar of type 2 diabetes patients.
Corydalis tuber extract: has the double functions of promoting insulin secretion and improving insulin resistance.
Astragalus root: hepatic glucose output can be inhibited by increasing hepatic glycogen synthesis and down-regulating gluconeogenic enzyme expression, thereby improving hepatic insulin resistance in type 2 diabetes.
Coptis chinensis: has the effects of clearing heat, eliminating dampness, purging pathogenic fire, removing toxic substances, and thickening intestines and stomach, and is matched with the pathogenesis of type 2 diabetes with diabetes, qi deficiency, dryness-heat as the main target, berberine serving as the main component in coptis can increase short-chain fatty acids in intestinal tracts by adjusting the intestinal flora structure, reduce the chronic inflammation state of organisms, and improve insulin resistance, thereby realizing the purpose of treating type 2 diabetes.
Dried rehmannia root: has effects of clearing heat, cooling blood, promoting fluid production, quenching thirst, improving pancreatic islet beta cell function, improving insulin resistance of peripheral tissue, regulating cell glucose homeostasis, reducing hepatic glucose-6-phosphatase activity, and improving glycolipid metabolism disorder.
Red peony root extract: increasing sensitivity of fat and skeletal muscle cells to insulin, and inhibiting hepatic glucose output, thereby improving insulin resistance and reducing blood glucose.
The oldenlandia diffusa and the hairyvein agrimony can inhibit the depolarization of islet beta cells, stimulate the islet beta cells to release insulin, inhibit the islet alpha cells to release glucagon, improve the sensitivity of peripheral tissues of type 2 diabetes patients to insulin and achieve the purpose of controlling blood sugar.

step four, crushing and extracting the weighed astragalus, coptis, radix rehmanniae, poria, cornus officinalis, oldenlandia diffusa, agrimony and uncaria, removing impurities, adding the balsam pear extract, the corydalis tuber extract, the red paeony root extract and sterilized deionized water, uniformly stirring, drying and granulating to obtain the traditional Chinese medicine for treating type 2 diabetes.

weighing astragalus, coptis, radix rehmanniae, poria cocos, cornus officinalis, oldenlandia diffusa, hairyvein agrimony, uncaria, bitter gourd extract, corydalis tuber extract and red paeony root extract according to a proportion;
step four, crushing and extracting the weighed astragalus, coptis, radix rehmanniae, poria, cornus officinalis, oldenlandia diffusa, agrimony and uncaria, removing impurities, adding the bitter gourd extract, the corydalis tuber extract, the red paeony root extract and sterilized deionized water, uniformly stirring, drying and granulating to obtain the traditional Chinese medicine for treating diabetes.

(3) filtering the extractive solution under reduced pressure to obtain filtrate, concentrating the filtrate under reduced pressure to obtain extract of fructus Momordicae Charantiae extract, performing ceramic membrane separation with ceramic tank, and adsorbing impurities to obtain fructus Momordicae Charantiae extract.
3. The preparation method of the traditional Chinese medicine for treating diabetes according to claim 2, wherein the bitter gourd powder and the water in the step (2) are mixed according to a ratio of 1: and (3) uniformly mixing the components in a mass ratio of 2-5.
4. The preparation method of the traditional Chinese medicine for treating type 2 diabetes according to claim 2, wherein the filtrate obtained in the step (3) is concentrated under reduced pressure until the relative density is 1.15-1.20, so as to obtain an extract of momordica charantia.
