Abstract
The invention relates to a traditional Chinese medicine extract composition capable of preventing and treating diabetes, which comprises the following components in parts by weight: 1-2 parts of ganoderma lucidum spore powder, 1-2 parts of bitter gourd extract, 1-2 parts of huperzia serrata extract, 2-3 parts of rhizoma corydalis extract, 1-2 parts of coptis chinensis extract, 1-2 parts of astragalus extract, 1-2 parts of salvia miltiorrhiza extract and 2-3 parts of red paeony root extract. The invention is based on the pathogenesis of diabetes which is disclosed by modern science, namely promoting the release of insulin and improving the insulin resistance. The traditional Chinese medicine extract composition has the effects of obviously blocking an ATP sensitive potassium ion channel, exciting islet beta cells and promoting insulin release, and can obviously improve insulin resistance of diabetes model animals, so that the blood sugar level of the diabetes model animals is reduced. The traditional Chinese medicine extract composition prepared by the invention can be used for preventing and treating type 2 diabetes. The traditional Chinese medicine extract composition can be prepared into capsules, tablets, granules, oral liquid, oral spray and the like.
Description
Technical Field
The invention belongs to the technical field of traditional Chinese medicine extract formulas, and particularly relates to a traditional Chinese medicine extract composition capable of preventing and treating diabetes.
Background
Diabetes mellitus is a serious chronic disease characterized by hyperglycemia that endangers human health. Hyperglycemia is caused by a defect in insulin secretion or an impaired biological action, or both. Patients with type 1 diabetes have abnormal immune systems, and the autoimmune reaction is caused after infection of certain viruses such as coxsackie virus, rubella virus, mumps virus and the like, so that insulin beta cells are damaged, and the absolute deficiency of insulin is shown. The pharmacological mechanism of each Chinese medicinal extract is as follows.

(2) The bitter gourd extract: contains steroid saponin such as momordicin, insulin-like peptide and alkaloid, which can endow Momordica charantia with hypoglycemic activity. (7) The red sage root extract: can improve diabetic microcirculation disturbance, thereby relieving liver, kidney, eye and neuropathy.
(8) Red peony root extract: promote glucose absorption and inhibit liver glucose regeneration, thereby improving insulin resistance and lowering blood sugar.
In the rat pancreas beta cell in-vitro experiment, the traditional Chinese medicine extract composition capable of preventing and treating diabetes has the effects of obviously blocking an ATP sensitive potassium ion channel, exciting islet beta cells and promoting insulin release, and in the whole diabetes animal model experiment, the traditional Chinese medicine extract composition can obviously improve the insulin resistance of animals so as to reduce the blood sugar level of the diabetes animals. Embodiment 1 (for diabetes of more than moderate degree)
A traditional Chinese medicine extract composition capable of preventing and treating diabetes comprises the following components in parts by weight: 2 parts of ganoderma lucidum spore powder, 2 parts of balsam pear extract, 2 parts of huperzia serrata extract, 3 parts of corydalis tuber extract, 2 parts of coptis chinensis extract, 2 parts of astragalus extract, 2 parts of salvia miltiorrhiza extract and 3 parts of red paeony root extract.
Embodiment 2 (for incipient, mild diabetes)
A traditional Chinese medicine extract composition capable of preventing and treating diabetes comprises the following components in parts by weight: ganoderma lucidum spore powder 1, balsam pear extract 1, huperzia serrata extract 1, corydalis tuber extract 2, coptis chinensis extract 1, astragalus extract 1, salvia miltiorrhiza extract 1 and red paeony root extract 2.
The traditional Chinese medicine extract composition for preventing and treating diabetes mellitus in the embodiment 2 is uniformly mixed according to a proportion, and dissolved and prepared into three concentrations of 1%, 5% and 10% in an experiment.
Test and data (taking embodiment 2 as an example)
1. Effect on rat pancreatic beta cells
As can be seen from A in FIG. 1, under the conditions recorded by patch clamp and current clamp, rat pancreatic beta cells can cause cell membrane depolarization and cell excitation to generate action potential when perfused with high concentration glucose (22.5 mM). As can be seen from B in fig. 1: under the same experimental recording conditions, if a 10% concentration of the Chinese medicinal composition (ZYZH) is applied to pancreatic beta cells, it can also cause depolarization of cell membranes and excitation of cells, resulting in action potentials.
2. Effect on ATP-sensitive Potassium ion channels in rat pancreatic beta cells
As can be seen from a in fig. 2: in isolated rat pancreatic beta cells, ATP-sensitive potassium channels were opened by repeated triangular wave depolarization under patch clamp whole cell, voltage clamp recording conditions, and thus potassium channel currents were suppressed using ATP-sensitive potassium channel specific blockers (tolbutamides). As can be seen from B in fig. 2: the magnitude of the current can be enhanced with an ATP-sensitive potassium channel specific opener (diazolidine). As can be seen from C in fig. 2: under the same conditions, the current of ATP sensitive nail channel can be inhibited by using 10% of the traditional Chinese medicine composition. As can be seen from D in fig. 2: the composition of the extract of the chinese traditional medicine for preventing and treating diabetes mellitus of example 2 can inhibit the current of ATP-sensitive nail channel, and the inhibitory effect thereof is concentration-dependent.
3. Effect on insulin Release from rat pancreatic beta cells
It is shown from the left-hand bar of FIG. 3 that perfusing high concentration glucose (22.5mM) can increase insulin release. As shown in FIG. 5, in the diabetic model rat, there was a decrease in insulin receptor sensitivity as shown by the black bars. After the Chinese medicinal composition is injected into the abdominal cavity, especially when the concentration reaches 5 percent (right oblique column) and 10 percent (net column), the sensitivity of insulin receptors can be obviously reduced.
6. Effect on insulin resistance in diabetic rats
As shown in fig. 6, diabetic rats developed significant Insulin Resistance (IR), which was found to be significantly reduced by 5% and 10% of the herbal extracts after treatment with the herbal composition.
7. Effect on rat insulin Release levels
As shown in fig. 7, the level of insulin secretion was increased in the diabetic rats because of the development of insulin resistance (black bars) compared to the normal rats. On the basis, 1% of the traditional Chinese medicine composition can further promote insulin release (vertical bars, the concentration of 1% does not reach the concentration for improving insulin resistance, see figure 6), but the promotion effect disappears along with the further increase of the concentration of the traditional Chinese medicine composition, and when the concentration of the traditional Chinese medicine composition reaches 5%, the insulin resistance is improved (see figure 6), so that the high insulin release level of the diabetic rat is reduced (right oblique bars).
The experiments show that the traditional Chinese medicine extract composition for preventing and treating diabetes can promote the release of insulin and improve insulin resistance, thereby effectively reducing and controlling the blood sugar level of diabetic rats, and can be used as a novel high-efficiency low-toxicity new scheme for preventing and treating diabetes with clear pharmacological action targets and mechanisms.
