Abstract
本发明提供了一种新型荧光传感体系及其在PTP‑1B检测中的应用,涉及PTP‑1B检测技术领域,一种TiO2‑SWCNHs复合纳米材料,由钛酸四丁酯通过水热合成法制备。一种TiO2‑SWCNHs在PTP‑1B抑制药物中的应用;TiO2‑SWCNHs用于在制备检测或治疗T2DM的药物中的用途。TiO2‑SWCNHs在血浆中PTP‑1B活性的测定中的应用,将血浆样本稀释100倍后直接用于测定,不做预处理;血浆样品中加入不同浓度PTP‑1B,将每种溶液在37℃温育30min,记录荧光光谱,用于比对和鉴定。由钛酸四丁酯通过水热合成法首次制备二氧化钛修饰的单壁碳纳米角,并对其进行了表征;建立了一种基于新型TiO2‑SWCNHs复合纳米材料的荧光传感体系用于PTP‑1B活性的测定,该方法简单、灵敏,为临床检测试剂盒的开发和PTP‑1B抑制剂的筛选提供了新思路。
Description
技术领域
本发明涉及PTP-1B检测技术领域,尤其涉及一种新型荧光传感体系及其在PTP-1B检测中的应用。
背景技术
糖尿病作为一种常见的代谢紊乱疾病已经成为全球关注的健康问题。随着人们生活水平的提高、饮食结构的改变和体力劳动的减少,糖尿病成为继癌症、心血管疾病之后的第三大慢性非传染性疾病,其发病率在全球呈逐年递增的趋势。其中二型糖尿病(T2DM)约占患者人数的90%,其特征是外周对胰岛素产生抵抗作用,在分子水平表现为胰岛素与胰岛素受体结合后信号转导缺失。
蛋白酪氨酸磷酸酶1B(protein tyrosine phosphatase 1B)是蛋白酪氨酸磷酸酶(protein tyrosine phosphatases,PTPs)家族中的一员,与胰岛素抵抗有着十分密切的关系。研究发现,PTP-1B在胰岛素信号转导中起负调控作用,它的过度表达会使PTP-1B的催化活性增高,从而导致胰岛素抵抗和胰岛素信号传导受损,对T2DM的发生、发展有着重要的作用,被认为是治疗T2DM的一个重要靶点。细胞中PTP-1B水平的升高可以作为疾病诊断的生物标志物,因此其特异性检测成为临床诊断的迫切需要。
目前,已经报道的检测PTP-1B的方法包括荧光磷酸酶法或比色磷酸酶法等。这些方法是通过使用磷酸酯为伪底物发展而来的,上述方法对于PTP-1B来说不具有特异性,也会影响酶底物在生理条件下对PTP-1B性质的特异性测量。
其中,最常用的方法是磷钼酸盐法,该方法是用孔雀绿比色法测定PTP-1B活性的最常用方法,但是该方法容易受到细胞裂解物等原始样品的高背景的影响,灵敏度不能满足生物样品中低含量PTP-1B的检测要求。因此,开发简单、敏感、廉价的PTP-1B测定方法仍然是一个巨大的挑战。
发明内容
本发明针对现有技术的不足,提供了一种新型荧光传感体系及其在PTP-1B检测中的应用。
纳米材料具有量子尺寸效应、小尺寸效应、宏观量子隧道效应和表面与界面效应等特点,目前已广泛用于细胞荧光成像、分析检测、药物释放及治疗等领域。单壁碳纳米角(single-walled carbon nanohorns,SWCNHs)是一种新型纳米材料,具有比表面积大、内部空间较大等特殊的结构,使其在电子、材料和生物医药等多个领域具有广阔的应用前景。另外,SWCNHs作为一种荧光淬灭剂被广泛地应用于光学体系的构建中。
例如:用于构建基于SWCNHs的DNA、小分子和大分子等物质的检测体系。基于此,利用SWCNHs的淬灭性质发展了一种检测PTP-1B的荧光体系。
由于二氧化钛(TiO2)与磷酸基团具有强烈的特异性吸附作用。
为了增强SWCNHs与磷酸化肽段和非磷酸化肽段的吸附区别,将TiO2混合在SWCNHs中,成功地合成了TiO2修饰的SWCNHs复合纳米材料(TiO2-SWCNHs)。该纳米材料具有了TiO2和SWCNHs二者的性质,既可以特异性吸附磷酸化肽段又可以有效淬灭肽段的荧光。
因此,本申请采用一种基于TiO2-SWCNHs的荧光传感体系用于PTP-1B的简单、灵敏的检测。
在用于本申请中产品的制备和测定需要用到以下的仪器和试剂,由于这些仪器和试剂都是实验室较为常见的,因此,仅仅进行公开,而不做进一步的赘述。
仪器部分:
F97型荧光分光光度计、Delta 320型pH计、Milli-Reference超纯水净化系统、恒温振荡器:SHA-B、原子力显微镜(AFM):Nanoscope IIIa instrument、拉曼光谱仪:DXR2xi、X-射线衍射仪(XRD):D8-ADVANCE、X-射线光电子能谱(XPS):ESCALAB 250Xi。
试剂部分:
单壁碳纳米角,浓硫酸,分析纯,无水乙醇,分析纯;肽段FAM-DADE(pY)LIPQQG;蛋白酪氨酸磷酸酶1B(PTP-1B),牛血清白蛋白、人血清白蛋白、碱性磷酸酶、二肽基肽酶Ⅳ,分析纯;4-羟乙基哌嗪乙磺酸(HEPES),分析纯;钛酸四丁酯、MgCl2、KCl,NaH2PO4,FeCl3,ZnSO4,分析纯;CuSO4,分析纯;天冬氨酸、苯丙氨酸、色氨酸、组氨酸、天冬酰胺、酪氨酸、谷氨酸、丝氨酸、精氨酸、谷氨酰胺、苏氨酸、脯氨酸、蛋氨酸,分析纯;半胱氨酸、高半胱氨酸、还原型谷胱甘肽(GSH),分析纯。
需要说明的是:实验中使用的缓冲溶液为HEPES缓冲溶液(10mmol/L,pH=7.5);实验中使用的水为Milli-Q超纯水(电导率超过18MΩ·cm);所用的其它试剂均为分析纯。
本发明通过以下技术手段实现解决上述技术问题的:
一种TiO2-SWCNHs复合纳米材料,由钛酸四丁酯通过水热合成法制备。
一种TiO2-SWCNHs在PTP-1B抑制药物中的应用;TiO2-SWCNHs用于在制备检测或治疗T2DM的药物中的用途。
一种TiO2-SWCNHs复合纳米材料的制备方法,包括如下步骤:
S1、称取3mg干燥的单壁碳纳米角放于250mL圆底烧瓶中,取150mL无水乙醇加入圆底烧瓶,并将160μL钛酸四丁酯,120μL浓硫酸溶于上述无水乙醇中;
所得混合物于80℃油浴锅中回流反应8h;
S2、将反应后的溶液倒入1.5mL离心管内以12000r/min离心4min;
其中,离心后上层是白色固体,下层是黑色固体,去掉上层反应剩余的钛酸四丁酯后再加入水混匀离心去掉上层固体;
如此反复至溶液pH近中性或者为中性,得到二氧化钛修饰的单壁碳纳米角。
一种基于TiO2-SWCNHs复合纳米材料的荧光传感体系,其特征在于,制备方法包括以下步骤,
将TiO2-SWCNHs储备溶液加入到FAM-DADE(pY)LIPQQG的缓冲溶液中,得到肽段浓度为2μmol/L;
TiO2-SWCNHs的浓度在0-44μg/mL的溶液,然后记录每种溶液的荧光光谱,用于比对和鉴定;
将TiO2-SWCNHs储备溶液加入到FAM-DADE(pY)LIPQQG缓冲溶液中;
得到TiO2-SWCNHs的最大淬灭浓度为40μg/mL的溶液,并分别记录0-60min时,溶液的荧光光谱,用于比对和鉴定。
进一步的,所述FAM-DADE(pY)LIPQQG的最终浓度为2μmol/L。
进一步的,所述TiO2-SWCNHs储备溶液的浓度为200μg/mL。
进一步的,荧光强度和PTP-1B浓度的线性关系为(F-F0)/F0=3.995[PTP-1B](ng/mL)+16.3034(R2=0.998);线性范围为0.02-10ng/mL;检测限为0.015ng/mL。
进一步的,将不同体积的PTP-1B储备溶液(0.1μg/mL)加入到TiO2-SWCNHs/FAM-DADE(pY)LIPQQG的HEPES缓冲溶液中;
得到浓度在0.02-10ng/mL的若干种不同种浓度的PTP-1B溶液;
将每种溶液在37℃温育30min,记录荧光光谱,用于比对和鉴定。
TiO2-SWCNHs在血浆中PTP-1B活性的测定中的应用,将血浆样本稀释100倍后直接用于测定,不做预处理;
血浆样品中加入不同浓度PTP-1B,将每种溶液在37℃温育30min,记录荧光光谱,用于比对和鉴定。
本发明的有益效果:
本发明中,由钛酸四丁酯通过水热合成法首次制备二氧化钛修饰的单壁碳纳米角,并对其进行了表征;建立了一种基于新型TiO2-SWCNHs复合纳米材料的荧光传感体系用于PTP-1B活性的测定,该方法简单、灵敏,为临床检测试剂盒的开发和PTP-1B抑制剂的筛选提供了新思路。

实施例1
如图1至7所示,一种TiO2-SWCNHs复合纳米材料的制备方法;
本实施例所述的一种二氧化钛修饰的单壁碳纳米角的制备方法,包括如下步骤:
S1、称取3mg干燥的单壁碳纳米角放于250mL圆底烧瓶中,取150mL无水乙醇加入圆底烧瓶,并将160μL钛酸四丁酯,120μL浓硫酸溶于上述无水乙醇中;
混合物于80℃油浴锅中回流反应8h。
S2、将反应后的溶液倒入1.5mL离心管内以12000r/min离心4min;
离心后上层是白色固体,下层是黑色固体,去掉上层反应剩余的钛酸四丁酯后再加入水混匀离心去掉上层固体;
如此反复至溶液pH近中性,既得二氧化钛修饰的单壁碳纳米角。
实施例2
如图1至7所示,本实施例中所述的一种基于TiO2-SWCNHs复合纳米材料的荧光传感体系,其制备方法包括以下步骤:
将不同体积的TiO2-SWCNHs储备溶液(浓度为200μg/mL)加入到FAM-DADE(pY)LIPQQG的缓冲溶液中,得到肽段浓度为2μmol/L,
TiO2-SWCNHs的浓度在0-44μg/mL的溶液,然后记录每种溶液的荧光光谱,基于获得的光谱,能够对物质进行比对和鉴定,确定物料的成分,以下相同。
将TiO2-SWCNHs储备溶液(200μg/mL)加入到FAM-DADE(pY)LIPQQG(最终浓度为2μmol/L)的缓冲溶液中;
得到TiO2-SWCNHs的最大淬灭浓度为40μg/mL的溶液,并分别记录0-60min时,溶液的荧光光谱。
实施例3
如图1至7所示,本实施例中所述的一种PTP-1B活性的测定方法,包括以下步骤,
将不同体积的PTP-1B储备溶液(0.1μg/mL)加入到TiO2-SWCNHs/FAM-DADE(pY)LIPQQG的HEPES缓冲溶液中;
得到浓度在0.02-10ng/mL的不同浓度的PTP-1B溶液。
将每种溶液在37℃温育30min,记录荧光光谱。
实施例4
如图1至7所示,本实施例所述的TiO2-SWCNHs在PTP-1B活性的测定中的应用。
使用时,在最佳条件下评价所提出的方法的准确性,血浆样品稀释100倍后直接用于测定,不需要进行其他预处理过程。
向健康受试者稀释后的血浆样品中加入不同浓度PTP-1B,将每种溶液在37℃温育30min,记录荧光光谱。
实施例5
TiO2-SWCNHs用于在制备检测或治疗T2DM的药物中的用途;以及TiO2-SWCNHs在PTP-1B抑制药物中的应用。
实验部分:
由于TiO2与磷酸化基团具有较强的吸附作用,因此,对TiO2-SWCNHs进行了合成,配置的该纳米材料可以与荧光团修饰肽段具有强烈的结合作用,从而荧光团的荧光被SWCNHs淬灭。
实验设计了FAM标记的磷酸化肽段FAM-DADE(pY)LIPQQG作为PTP-1B的特异性底物肽段。该肽段对应的非磷酸化肽段DADEYLIPQQG的pI约为3.49(通过ProtParam软件计算得到)。
在pH 7.5的溶液中,非磷酸化肽段带负电荷。因此,非磷酸化肽段与纳米材料TiO2-SWCNHs不发生静电吸引作用。
工作原理部分:
如图1所示,当体系中不存在PTP-1B时,FAM-DADE(pY)LIPQQG与TiO2-SWCNHs强烈吸附,发生荧光淬灭;
当体系中加入PTP-1B时,由于PTP-1B与肽段的特异性识别作用,可以发生去磷酸化,故生成FAM-DADEYLIPQQG,该肽段与TiO2-SWCNHs不发生吸附作用,故TiO2-SWCNHs远离肽段,实现荧光的增强,从而能够定量检测PTP-1B。
参考图2A,这是是纳米复合材料TiO2-SWCNHs的AFM表征;从图中可以看出TiO2修饰的SWCNHs表面均匀分布着细小的TiO2纳米颗粒(~9.4nm)。
参考图2B,从拉曼光谱中可以看出,碳纳米角有两个特征峰,1332cm-1处的峰为D波段,1577cm-1处的峰为G波段;而二氧化钛修饰后的碳纳米角增加了387cm-1,509cm-1和632cm-1三个峰,这是TiO2典型的特征峰。
参考图2C,25.2°、37.8°、48.0°、62.3°的值分别对应锐钛矿型TiO2的(101)、(004)、(200)、(213),与标准的TiO2锐钛矿相匹配(JCPDS No.21-1271)。
上述结果证明,实验成功地合成了TiO2-SWCNHs纳米复合材料。
参考图3,由图3可以看出,C 1s、O 1s核电子的结合能和Ti 2p电子的结合能分别为285.2eV、530.0eV和461.7eV。
除了284.0eV处对应的C-C峰,还有286.0eV和288.3eV两处分别对应的C-O和O-C=O峰。由于表面存在这些含氧官能团,TiO2-SWCNHs的分散性明显优于SWCNHs。
531.6eV和529.9eV的峰值分别归因于水和二氧化钛的O1s。Ti 2p3/2和Ti 2p1/2自旋轨道分裂光电子的束缚能分别为464.4eV和458.6eV。
关于TiO2-SWCNHs的荧光淬灭作用
在本部分中,主要是验证淬灭剂TiO2-SWCNHs的用量对体系荧光强度的影响。
图4显示了在向溶液中加入TiO2-SWCNHs后FAM-DADE(pY)LIPQQG的荧光强度逐渐降低。
在不存在TiO2-SWCNHs的情况下,FAM-DADE(pY)LIPQQG的溶液显示出强的荧光光谱。然而,在加入TiO2-SWCNHs(40μg/mL)后,观察到可发生高达98.5%的荧光淬灭。
从图4内插图可以看出,随着TiO2-SWCNHs浓度的逐渐增大,体系的荧光强度逐渐降至最低。当TiO2-SWCNHs达到40μg/mL时,体系荧光强度几乎完全淬灭。这可能是因为TiO2-SWCNHs与FAM标记引物之间的电子转移引起的。
因此,可以选择FAM标记肽段的浓度为2μmol/L,TiO2-SWCNHs的最佳用量为40μg/mL。
在本部分中;验证了体系加入TiO2-SWCNHs后,荧光强度随时间的变化曲线。
参考图5A,加入TiO2-SWCNHs后,荧光强度迅速降至最低,在1min内荧光强度达到恒定水平,且不随时间变化,这表明在TiO2-SWCNHs表面上吸附磷酸化肽段是一个快速且稳定的过程。
参考图5B,可以得出,Stern-Volmer曲线的斜率Ksv值为0.0410(μg/mL)-1,由此得出,1/Ksv的数值即50%的荧光强度被淬灭时淬灭剂的浓度为24.39μg/mL。
另外,Stern-Volmer图显示出向上弯曲的趋势,说明荧光团FAM-DADE(pY)LIPQQG与TiO2-SWCNHs同时发生了动态和静态淬灭,FAM-DADE(pY)LIPQQG和TiO2-SWCNHs可形成静态配合物。
本部分中,验证了传感体系对不同浓度的PTP-1B的荧光响应。
参考图6中,可以看出,在没有PTP-1B的情况下,体系具有较低的荧光强度。
当体系中加入PTP-1B后,荧光强度逐渐增强,并且随PTP-1B浓度的增大而逐渐增大,这是由于PTP-1B对FAM标记磷酸化肽段的特异性识别作用引起,使肽段发生去磷酸化作用,从而远离TiO2-SWCNHs,致使体系的荧光增强。
在上述的最优条件下,我们得到荧光强度和PTP-1B浓度的线性关系:
为(F-F0)/F0=3.995[PTP-1B](ng/mL)+16.3034(R2=0.998);线性范围为0.02-10ng/mL。检测限为0.015ng/mL。(3S/m,S是试剂空白的标准偏差,n=11,m是线性方程中的斜率)
在本部分中,验证了该荧光传感体系对PTP-1B活性检测的选择性研究。
如图7所示,体系考察了二肽基肽酶Ⅳ(DPP-4),碱性磷酸酶(ALP),胰蛋白酶(TRY),溶菌酶(Lys),乙酰胆碱酯酶(AchE),牛血清白蛋白(BSA),人血清白蛋白(HSA)的浓度是PTP-1B浓度十倍量时对该体系的荧光的影响;
从图中可以看出,PTP-1B的荧光恢复效果远超其他物质。
上述结果表明本测定系统对PTP-1B含量检测具有较高的选择性。
在本部分在中,验证了考察该荧光传感检测体系对PTP-1B活性检测的抗干扰能力。
如表1所示,常见的共存物质对该荧光传感检测体系荧光几乎无影响。
因此,该荧光检测体系对PTP-1B活性的检测具有较强的抗干扰能力,有望用于实际样品中PTP-1B活性的检测。

实际样品的测定
为了验证实验建立方法的实际应用价值,实验选择健康受试者血浆样品稀释100倍后作为样品,没有进行其他预处理过程。
通过标准加入法,测定了实际样品中PTP-1B的含量。
表2可以看出,当PTP-1B浓度很低时,测量的回收率仍然取得了满意效果。这说明我们建立的传感体系能够用于血浆样中PTP-1B含量。

本实验由钛酸四丁酯通过水热合成法首次制备二氧化钛修饰的单壁碳纳米角,并对其进行了表征;建立了一种基于新型TiO2-SWCNHs复合纳米材料的荧光传感体系用于PTP-1B活性的测定,该方法简单、灵敏,为临床检测试剂盒的开发和PTP-1B抑制剂的筛选提供了新思路。

Claims (5)
1.一种TiO2-SWCNHs复合纳米材料的荧光传感体系,其特征在于:SWCNHs为单壁碳纳米角,复合纳米材料由钛酸四丁酯通过水热合成法制备,包括如下制备步骤:
S1、称取3mg干燥的单壁碳纳米角放于250mL圆底烧瓶中,取150mL无水乙醇加入圆底烧瓶,并将160μL钛酸四丁酯,120μL浓硫酸溶于上述无水乙醇中;
所得混合物于80℃油浴锅中回流反应8h;
S2、将反应后的溶液倒入1.5mL离心管内以12000r/min离心4min;
其中,离心后上层是白色固体,下层是黑色固体,去掉上层反应剩余的钛酸四丁酯后再加入水混匀离心去掉上层固体;
制备荧光传感体系包括以下步骤,将不同体积的TiO2-SWCNHs储备溶液加入到FAM-DADE(pY)LIPQQG的缓冲溶液中,得到肽段浓度为2μmol/L;
TiO2-SWCNHs的浓度在0-44μg/mL的溶液,然后记录每种溶液的荧光光谱,用于比对和鉴定;将TiO2-SWCNHs储备溶液加入到FAM-DADE(pY)LIPQQG缓冲溶液中;
得到TiO2-SWCNHs的最大淬灭浓度为40μg/mL的溶液,并分别记录0-60min时,溶液的荧光光谱,用于比对和鉴定。
2.根据权利要求1所述的一种TiO2-SWCNHs复合纳米材料的荧光传感体系,所述TiO2-SWCNHs储备溶液的浓度为200μg/mL。
3.根据权利要求1所述的一种TiO2-SWCNHs复合纳米材料的荧光传感体系,其特征在于:荧光强度和PTP1-B浓度的线性关系为(F-F0)/F0=3.995[PTP1-B](ng/mL)+16.3034(R2=0.998);线性范围为0.02-10ng/mL;检测限为0.015ng/mL。
4.TiO2-SWCNHs在制备治疗二型糖尿病T2DM药物中的用途,SWCNHs为单壁碳纳米角。
5.TiO2-SWCNHs在制备PTP1-B抑制药物中的应用,SWCNHs为单壁碳纳米角。
