Abstract
The invention provides a skin composition for promoting skin barrier repair function, and a preparation method and application thereof. The skin composition comprises active ingredients, and the content of the active ingredients is 0.021-50%, preferably 0.1-25%, more preferably 0.5-10% based on the total mass of the skin composition as 100%; wherein the active ingredients comprise chia seed oil, sunflower seed oil unsaponifiables and ceramides. The skin composition with the function of promoting the skin barrier repair has the effects of calming, relieving, resisting bacteria and inflammation, resisting oxidation, resisting ultraviolet and infrared radiation and repairing skin injury, and achieves the effects of enhancing the skin resistance, maintaining the health and stability of the skin and relieving skin discomfort.
Description
Technical Field
The invention relates to a skin composition for promoting skin barrier repair function, a preparation method and application thereof, and belongs to the field of cosmetics.
Background
The skin is the largest organ of human body contacting with external environment, covers the whole body, and is the protective umbrella at the outermost layer of other organs of the body. The surface of the skin-moistening cream is provided with a sebaceous membrane formed by fusing sebaceous gland lipid, sweat and keratinocyte lipid, so that the skin-moistening cream can reduce the water evaporation on the surface of the skin, prevent external dust dirt, irritants and the like from invading the skin, provide a natural barrier protective layer for the skin and is an important structure for maintaining the health of the skin.
In recent years, more and more people suffer from skin barrier damage caused by over-cleaning, facial mask application, cutin removal and the like, and once the skin barrier is damaged, the moisture retention capacity of the skin is reduced, so that the symptoms of tight, dry, peeling, erythema and the like after washing are caused, and even sensitive skin is easily generated. The formation of sensitive skin is due to problems with the natural defense system of the skin, which loses its barrier capacity to external stimuli. The first warning of a decrease in the natural barrier function of the skin is the breakdown of the sebaceous membrane.
In human skin, 75-90% of skin lipids are secreted from sebaceous glands, and the remaining 10-25% of lipids are derived from intercellular lipids, mainly synthesized from ceramides, cholesterol and free fatty acids. Among them, fatty acids cannot be synthesized by the human body itself, and unsaturated fatty acids are particularly more advantageous to the human body. The unsaturated fatty acid can inhibit the inflammatory action of various mediators and cytokines which cause inflammation and does not bring side effects, thereby bringing a new scheme for treating the inflammation.
However, some natural and non-irritant skin care products are needed for nursing the sensitive skin, but the natural and non-irritant skin care products do not have the efficacy, and the components added into the product after being compounded do not necessarily achieve the expected effect of repairing the sensitive skin, so that the efficacy is inhibited mutually, and the effect of repairing the sensitive skin is not good.

Problems to be solved by the invention
In view of the technical problems in the prior art, for example: the invention firstly provides a skin composition for promoting the skin barrier repair function, and solves the problems of poor absorption of effective components, allergy, unobvious effect, skin care product oil components which are not friendly to skin and the like. The skin composition can relieve allergy, improve skin dryness, and repair skin barrier effectively.
Furthermore, the invention also provides a preparation method of the skin composition for promoting the skin barrier repair function, which is simple and feasible in preparation method, easy in acquisition of raw materials and suitable for mass production.
Means for solving the problems
The invention provides a skin composition, which comprises active ingredients, wherein the content of the active ingredients is 0.021-50%, preferably 0.1-25%, more preferably 0.5-10% based on the total mass of the skin composition as 100%; wherein the content of the first and second substances,
the active ingredients include chia seed oil, sunflower seed oil unsaponifiable matter and ceramide.
The skin composition comprises 0.01-30%, preferably 1-25% of chia seed oil, 0.01-30%, preferably 1-25% of sunflower seed oil unsaponifiables and 0.001-10%, preferably 0.01-8% of ceramides, based on the total mass of the skin composition being 100%.
The skin composition provided by the invention is characterized in that the mass ratio of chia seed oil to sunflower seed oil unsaponifiable matter to ceramide is 1-10:1-5:1, preferably 2-8:1.5-4: 1.
The composition for skin according to the present invention, wherein the ceramide is one or a combination of two or more selected from the group consisting of ceramide 1, ceramide 1A, ceramide 2, ceramide 3, ceramide 4, ceramide 6II, ceramide 6, ceramide 7, ceramide 8, ceramide NS, ceramide NP, ceramide EOS, ceramide EOP, ceramide NG, and ceramide AP.
The skin composition of the present invention further comprises one or a combination of two or more of a humectant, a thickener, an emulsifier, a chelating agent, a preservative, and a solvent.
The skin composition comprises 0.01-50% of the moisturizing agent, 0.01-20% of the thickening agent, 0.01-20% of the emulsifying agent, 0.001-2% of the chelating agent, 0.01-5% of the preservative and 0-98% of the solvent.
The present invention also provides a method of preparing the dermatological composition according to the present invention, comprising the step of mixing the components of the dermatological composition.
The invention also provides the application of the skin composition in preparing an external preparation for promoting or improving or strengthening the barrier function of the skin.
The invention further provides the use of the dermatological composition according to the invention for the preparation of a skin care product.
The use of the invention, wherein the skin care product comprises one of emulsion, cream, essence, facial mask, skin lotion and skin oil.

The skin composition with the function of promoting the skin barrier repair has the effects of calming, relieving, resisting bacteria and inflammation, resisting oxidation, resisting ultraviolet and infrared radiation and repairing skin injury, and achieves the effects of enhancing the skin resistance, maintaining the health and stability of the skin and relieving skin discomfort.
The preparation method of the skin composition for promoting the skin barrier repair function is simple and feasible, and raw materials are easy to obtain, so that the skin composition is suitable for mass production.

<First aspect>
The first aspect of the present invention provides a skin composition comprising an active ingredient in an amount of 0.021 to 50%, preferably 0.1 to 25%, more preferably 0.5 to 10%, based on 100% by mass of the total skin composition, for example: 0.05%, 0.2%, 0.8%, 1%, 3%, 5%, 7%, 9%, 11%, 15%, 20%, 25%, 30%, 35%, 40%, 45%, etc.; wherein the content of the first and second substances,
the active ingredients include chia seed oil, sunflower seed oil unsaponifiable matter and ceramide.
The skin composition has the effects of calming, relieving, resisting bacteria and inflammation, resisting oxidation, resisting ultraviolet and infrared radiation and repairing skin injury, and achieves the effects of enhancing the skin resistance, maintaining the health and stability of the skin and relieving skin discomfort.
Chia seed oil
The chia seed oil is vegetable oil obtained by squeezing. Chia seed is seed of herba Menthae plant such as Gordonia euryales. Chia seeds are rich in polyphenols and isoflavones, and these antioxidant compounds have the function of scavenging free radicals, reducing the risk of chronic diseases and preventing certain diseases (such as diabetes, alzheimer's disease and parkinson's disease). Meanwhile, the fat composition in chia seeds is mostly unsaturated fatty acid, the content of Omega-3 and Omega-6 is very rich, and Omega-3 has excellent anti-inflammatory effect, can inhibit inflammation and quicken inflammation repair. Omega-6 is involved in three phases of wound healing: inflammatory phase, proliferation phase, remodeling phase. In the invention, chia seed oil has a relatively obvious relieving effect.
In some specific embodiments, the chia seed oil is present in an amount of 0.01% to 30%, preferably 1% to 25%, for example: 0.5%, 1%, 3%, 5%, 7%, 9%, 10%, 12%, 15%, 18%, 20%, 22%, 25%, 28%, etc., and is effective in soothing the skin when the chia seed oil is present in an amount of 0.01% to 30%, particularly 1% to 25%. If the content of chia seed oil is too high, the skin-soothing effect is not increased more obviously, and the price is higher.
In some specific embodiments, the chia seed oil can be obtained commercially or prepared in the present invention.
Specifically, the chia seed oil can be prepared by adopting a squeezing method. Specifically, the chia seed oil can be squeezed by a screw, and during the squeezing process, the temperature during squeezing is maintained at 1-10 ℃ by adding liquid nitrogen, and then the chia seed oil is prepared by filtering.
Unsaponifiable sunflower seed oil
The action mechanism of the sunflower seed oil unsaponifiable matter is to inhibit the release of transcription factor NF-kB & cell factor to reduce skin cell inflammation. Inhibiting the amount of inflammatory cytokines IL1b, IL6, IL8 and TNF alpha, thereby achieving the anti-inflammatory effect. Activates the synthesis of key lipids in an in vitro model of the skin, and enhances skin barrier function without altering the total lipid content.
In some specific embodiments, the sunflower seed oil unsaponifiable matter is present in an amount of 0.01% to 30%, preferably 1% to 25%, based on 100% of the total mass of the dermatological composition, for example: 0.5%, 1%, 3%, 5%, 7%, 9%, 10%, 12%, 15%, 18%, 20%, 22%, 25%, 28%, etc., and is effective for soothing the skin when the content of the sunflower seed oil unsaponifiable matter is 0.01% to 30%, particularly 1% to 25%. If the content of the unsaponifiable sunflower seed oil is too high, the skin-soothing effect is not increased more remarkably, and the price is high.
The sunflower seed oil unsaponifiable matter can be obtained commercially or prepared by a molecular distillation technology.
Ceramide
Ceramide (Ceramide) is a class of amide compounds formed by dehydrating long-chain fatty acids and the amino groups of sphingosine, mainly comprises Ceramide phosphorylcholine and Ceramide phosphorylethanolamine, phospholipid is the main component of cell membranes, 40-50% of sebum in horny layer is composed of Ceramide, Ceramide is the main part of intercellular matrix, and plays an important role in keeping the moisture balance of horny layer. Ceramide can significantly enhance adhesion between keratinocytes, improve skin dryness, and reduce desquamation of skin. Ceramide can significantly enhance adhesion between keratinocytes, improve skin dryness, and reduce desquamation of skin. Ceramides play a very important role in maintaining the skin barrier function.
In some specific embodiments, the ceramide is present in an amount ranging from 0.001% to 10%, preferably from 0.01% to 8%, for example: 0.005%, 0.05%, 0.1%, 0.5%, 1%, 2%, 3%, 4%, 5%, 6%, 7%, 8%, 9%, etc., and is effective for soothing the skin when the ceramide content is 0.01% to 10%, particularly 0.01% to 8%. If the content of ceramide is too high, the skin-soothing effect is not more obvious and gradually increased, and the price is higher.
Further, the type of ceramide in the present invention is not particularly limited, and may be those commonly used in the art. Specifically, the ceramide is selected from one or a combination of more than two of ceramide 1, ceramide 1A, ceramide 2, ceramide 3, ceramide 4, ceramide 6II, ceramide 6, ceramide 7, ceramide 8, ceramide NS, ceramide NP, ceramide EOS, ceramide EOP, ceramide NG and ceramide AP.
The present inventors have found that a skin composition containing chia seed oil, sunflower seed oil unsaponifiable matter and ceramide as active ingredients can effectively repair the skin barrier by synergistically enhancing the skin composition, relieving allergy, and improving the degree of skin dryness.
Further, in the present invention, the content of the active ingredient is 0.01 to 50%, preferably 0.1 to 25%, more preferably 0.5 to 10%, for example: 1%, 3%, 5%, 7%, 9%, 11%, 13%, 15%, 17%, 19%, 21%, 23%, 25%, 27%, 29%, 31%, 33%, 35%, 37%, 39%, 41%, 43%, 45%, 47%, 49%, etc. When the content of the active ingredients is 0.01-50%, the functions of chia seed oil, sunflower seed oil unsaponifiable matter and ceramide can be effectively exerted. When the content of the active ingredient is less than 0.01%, a function of repairing a skin barrier cannot be realized, and when the content of the active ingredient is more than 50%, the cost is too high, and more excellent effects cannot be obtained.
In some specific embodiments, the mass ratio of chia seed oil, sunflower seed oil unsaponifiable matter and ceramide is 1-10:1-5:1, preferably 2-8:1.5-4:1, for example: 2.5-8.5:1.8-3.8:1, 3-7:2-3.5:1, 3.5-6.5:2.2-3.2:1, 4-6:2.5-3:1, 4.5-5.5:2.6-2.8:1, 5:2.5:1, etc. When the mass ratio of chia seed oil to sunflower seed oil unsaponifiable matter to ceramide is 1-10:1-5:1, the functions of chia seed oil, sunflower seed oil unsaponifiable matter and ceramide can be more effectively exerted, so that the function of repairing the skin barrier is realized.
Other ingredients
The skin composition of the present invention may further include one or a combination of two or more of a humectant, a thickener, an emulsifier, a chelating agent, a preservative, and a solvent.
The content of the humectant is 0.01-50% based on 100% of the total mass of the skin composition, and specifically, the content of the humectant can be, for example: 1%, 3%, 5%, 7%, 9%, 11%, 13%, 15%, 17%, 19%, 21%, 23%, 25%, 27%, 29%, 31%, 33%, 35%, 37%, 39%, 41%, 43%, 45%, 47%, 49%, etc. In the invention, when the addition amount of the humectant is 0.01-50%, the humectant can play a role in moisturizing.
Specifically, in the present invention, the humectant includes one or a combination of two or more of butylene glycol, panthenol, dipropylene glycol, glycerin, betaine, xylitol, sodium hyaluronate, trehalose, pentanediol, hexanediol, and 1, 3-propanediol.
The content of the thickening agent is 0.01-20% based on the total mass of the skin composition as 100%. Specifically, the content of the thickener may be, for example: 1%, 3%, 5%, 7%, 9%, 11%, 13%, 15%, 17%, 19%, etc. The consistency of the emulsifying system of the invention can be adapted by adding a thickener.
Specifically, in the invention, the thickener comprises one or a combination of more than two of carbomer, carbomer salt, xanthan gum, sodium polyacrylate, carrageenan, gellan gum, hydroxyethyl cellulose, acryloyl dimethyl ammonium taurate/VP copolymer, polyacrylate cross-linked polymer-6 and hydroxyethyl acrylate/acryloyl dimethyl sodium taurate copolymer.
The content of the emulsifier is 0.01-20% based on the total mass of the skin composition as 100%. Specifically, the content of the emulsifier may be, for example: 0.05%, 0.1%, 0.3%, 0.4%, 0.5%, 0.7%, 0.8%, 1%, 3%, 5%, 7%, 9%, 11%, 13%, 15%, 17%, 19%, etc. When the dosage of the emulsifier is less than 0.01%, the emulsification is insufficient, so that the system is unstable; when the dosage of the emulsifier is more than 20 percent, the irritation of the product can be increased, and meanwhile, certain influence can be caused on the stability of the product.
Specifically, in the present invention, the emulsifier may include one or a combination of two or more of cetearyl alcohol, hydrogenated lecithin, cetearyl glucoside, PEG-100 stearate, polyglycerol-10 stearate, glycerol stearate/PEG-100 stearate, sorbitan sesquioleate, isosteareth-20, PEG-10 campesterol, laureth-7, polyglycerol-3 methylglucose distearate, PEG/PPG-10/1 polydimethylsiloxane, polyglycerol-3 diisostearate, glycerol stearate, steareth-20, ceteth-20, polysorbate-60, cetearyl olive oleate, sorbitan olive oleate.
The skin composition of the present invention may further contain a chelating agent. The addition amount of the chelating agent is 0.001-2% of the total mass of the emulsifying system. The chelating agent may be EDTA-2Na and/or EDTA-4 Na.
The skin composition of the invention can also comprise a preservative and/or a solvent, wherein the preservative is added in an amount of 0.01-5% and the solvent is contained in an amount of 0-98% based on the total mass of the emulsifying system; specifically, the preservative may include one or a combination of two or more of phenoxyethanol, methylparaben, propylparaben, ethylhexylglycerin, benzoic acid, p-hydroxyacetophenone, and salts thereof, and the solvent is water.
Further, the first aspect of the present invention also provides a method for preparing the dermatological composition according to the present invention, comprising the step of mixing the components of the dermatological composition.
The skin composition of the present invention can effectively alleviate the problems of redness, stinging and the like caused by skin sensitivity. According to the invention, the chia seed oil, the sunflower seed oil unsaponifiable matter and the ceramide are compounded, so that the effect is obviously superior to that of a single or simple combination, the synergistic effect can be achieved, and the effects of calming, relieving, resisting bacteria, resisting inflammation, resisting oxidation, resisting ultraviolet and infrared radiation and repairing skin injury are realized. The effects of enhancing the skin resistance, maintaining the health and stability of the skin and relieving the skin discomfort are realized.
<Second aspect of the invention>
In a second aspect, the invention provides the use of a dermatological composition according to the invention for the preparation of an external preparation for promoting or enhancing or strengthening the barrier function of the skin.
Further, the second aspect of the present invention also provides a use of the dermatological composition according to the present invention for the preparation of a skin care product. Specifically, the skin care product comprises one of emulsion, cream, essence, facial mask, skin lotion and skin oil. The product prepared by the invention is safe and non-irritant, can effectively relieve discomfort caused by skin sensitivity, and is suitable for people of all ages.
Examples
Embodiments of the present invention will be described in detail below with reference to examples, but those skilled in the art will appreciate that the following examples are only illustrative of the present invention and should not be construed as limiting the scope of the present invention. The examples, in which specific conditions are not specified, were conducted under conventional conditions or conditions recommended by the manufacturer. The reagents or instruments used are not indicated by the manufacturer, and are all conventional products commercially available.
The preparation method of chia seed oil comprises the following steps:
squeezing with screw while adding liquid nitrogen to maintain the squeezing temperature at 4 deg.C, and filtering.
The sunflower seed oil unsaponifiable is purchased from laboraires expansciene, france.
Examples 1 to 10
The skin compositions of examples 1 to 10 were prepared according to the following preparation methods based on the mass percentages of the components in tables 1 and 2 below.
Wherein, the preparation method of the skin composition comprises the following steps:
1) adding the sterilized solvent into a reaction kettle, adding the chelating agent, the humectant and the thickening agent while stirring, uniformly stirring, heating to 82 ℃, adding the active ingredient and the emulsifier, homogenizing, and uniformly stirring.
2) Cooling to 45 deg.C, adding antiseptic, stirring, cooling to 35 deg.C, inspecting, and discharging.
Comparative examples 1 to 7 and 9
Skin compositions of comparative examples 1 to 9 were prepared according to the preparation methods of examples 1 to 10 based on the mass percentages of the components in tables 3 and 4 below.
Comparative example 8
The skin composition was prepared according to the following method based on the mass percentages of the components in table 4 below.
1) Adding the sterilized solvent into the main kettle, adding the chelating agent, the humectant and the thickening agent while stirring, uniformly stirring, heating to 82 ℃, adding the mineral wax and the emulsifier, homogenizing and uniformly stirring.
2) Cooling to 45 deg.C, adding antiseptic, stirring, cooling to 35 deg.C, inspecting, and discharging.

Performance testing
First, analysis of water loss rate of skin through epidermis
The Trans-epidermal water loss (TEWL) is measured by the german CK skin water loss (TEWL) tester Tewameter, TM300, and is an important parameter for reflecting skin water loss and barrier. Lower values for the variability of the transepidermal water loss rate of cheek skin indicate better barrier function of the skin.
1. The detection method for analyzing the water loss rate of the skin through the epidermis comprises the following steps:
1.1 subject screening criteria
A healthy female subject between the ages of 20 and 50 years; and the skin of the subject is positive after the test of 10% lactic acid stabbing pain (judged as sensitive skin); slight redness of the face; the number of people is 95, and each sample is randomly 5.
1.2 site of use: face part
1.3 frequency of use: it is administered 1 time each in the morning and evening, and for 28 days, 1.2g each time.
1.4 test environmental conditions
The temperature is 21 +/-1 ℃, and the relative humidity is 50 +/-5%.

As is clear from the results in Table 5, in the case of using examples 1 to 10, the transepidermal water loss rate of the skin after 28 days of continuous use was smaller than that of comparative examples 1 to 9, and the transepidermal water loss was smaller, indicating that the skin barrier was improved by using the composition of the present invention.
Further, as can be seen from examples 1 to 9, comparative examples 1 to 3, and comparative example 8, the skin composition of the present invention has a significantly improved effect on skin barrier repair as compared with chia seed oil, sunflower seed oil unsaponifiable matter and ceramide NP, vaseline alone, and thus chia seed oil, sunflower seed oil unsaponifiable matter and ceramide have a significant synergistic effect.
Second, analysis of degree of skin redness
The skin redness parameter was obtained by acquiring facial images by Visia-CR and analyzing the skin a value of the cheek area using Image-Pro-Plus Image analysis software on the pictures under Standard2 light source, and the smaller the difference of the degree of redness of cheek area, the less redness of skin.
1. The detection method for analyzing the degree of skin redness is as follows:

As is apparent from the results in Table 6, in the case of using examples 1 to 10, the degree of skin redness after 28 days of continuous use was more reduced than in the case of comparative examples 1 to 9, and the skin barrier was improved with the composition of the present invention as shown by the result that the skin moisture loss was less.
As can be seen from examples 1 to 9, comparative examples 1 to 3 and comparative example 8, the dermatological composition of the present invention has a significantly improved effect on the degree of redness of the skin compared to chia seed oil, sunflower seed oil unsaponifiable and ceramide NP, petrolatum alone, and thus chia seed oil, sunflower seed oil unsaponifiable and ceramide have a significant synergistic effect. Therefore, the composition of the present invention can be used to promote the repair of skin barrier.
The above examples of the present invention are merely examples for clearly illustrating the present invention and are not intended to limit the embodiments of the present invention. Other variations and modifications will be apparent to persons skilled in the art in light of the above description. This need not be, nor should it be exhaustive of all embodiments. Any modification, equivalent replacement, and improvement made within the spirit and principle of the present invention should be included in the protection scope of the claims of the present invention.
Claims (10)
1. A skin composition characterized by comprising an active ingredient in an amount of 0.021 to 50%, preferably 0.1 to 25%, more preferably 0.5 to 10%, based on 100% by mass of the total skin composition; wherein the content of the first and second substances,
the active ingredients include chia seed oil, sunflower seed oil unsaponifiable matter and ceramide.
2. The dermatological composition according to claim 1, wherein the chia seed oil is present in an amount of 0.01% to 30%, preferably 1% to 25%, the sunflower seed oil unsaponifiable matter is present in an amount of 0.01% to 30%, preferably 1% to 25%, and the ceramide is present in an amount of 0.001% to 10%, preferably 0.01% to 8%, based on 100% of the total mass of the dermatological composition.
3. The dermatological composition according to claim 1 or 2, wherein the weight ratio of chia seed oil, sunflower seed oil unsaponifiable matter and ceramide is 1-10:1-5:1, preferably 2-8:1.5-4: 1.
4. The dermatological composition according to any one of claims 1 to 3, wherein the ceramide is selected from the group consisting of ceramide 1, ceramide 1A, ceramide 2, ceramide 3, ceramide 4, ceramide 6II, ceramide 6, ceramide 7, ceramide 8, ceramide NS, ceramide NP, ceramide EOS, ceramide EOP, ceramide NG, and ceramide AP.
5. The dermatological composition of any one of claims 1 to 4, further comprising one or a combination of two or more of a humectant, a thickener, an emulsifier, a chelating agent, a preservative, and a solvent.
6. The dermatological composition of claim 5, wherein the humectant is present in an amount of 0.01% to 50%, the thickener is present in an amount of 0.01% to 20%, the emulsifier is present in an amount of 0.01% to 20%, the chelating agent is present in an amount of 0.001% to 2%, the preservative is present in an amount of 0.01% to 5%, and the solvent is present in an amount of 0% to 98%.
7. A method of preparing a dermatological composition according to any one of claims 1 to 6, comprising the step of mixing the components of the dermatological composition.
8. Use of the dermatological composition according to any one of claims 1 to 6 for the preparation of an external preparation for promoting or enhancing or strengthening the barrier function of the skin.
9. Use of a dermatological composition according to any one of claims 1 to 6 for the preparation of a skin care product.
10. The use according to claim 9, wherein the skin care product comprises one of a lotion, cream, serum, mask, lotion, oil.
