Abstract
The invention belongs to the technical field of A61P17/14, and particularly relates to an anti-hair loss hair-care composition and a hair-washing and hair-care preparation thereof. It at least comprises the following components in percentage by weight: a) At least 0.05wt% of pyrrolidinyl diaminopyrimidine; (b) at least 0.05wt% adenosine; (c) at least 0.05wt% oligopeptide; (d) at least 1.0wt% lecithin; (e) at least 5wt% polyglycerol fatty acid ester; a dispersion medium for the remainder; wherein the lecithin is egg yolk lecithin; the saponification value of the egg yolk lecithin is 195-212 mgKOH/g. In the hair loss prevention and hair growth composition, under the interaction of pyrrolidinyl diaminopyrimidine, adenosine, oligopeptide, egg yolk lecithin and the like, the damage of components such as hydrogen peroxide, testosterone and the like to HDPC cells and HaCaT cells is effectively inhibited, the growth and proliferation differentiation of fibroblast growth factors in dermal papilla cells are accelerated, so that the alopecia is reduced, and the hair quantity is improved.

Technical Field
The invention belongs to the technical field of A61P17/14, and particularly relates to an anti-hair loss hair-care composition and a hair-washing and hair-care preparation thereof.
Background
With the increasing of working pressure, the accelerating of life rhythm, environmental influence, the use and popularization of hair styling products such as hair dyeing and hair waving, etc., the number of hair loss patients is greatly increased each year. Meanwhile, the investigation shows that the alopecia has a higher proportion of depression than the normal people due to the negative influence of the alopecia on the external image, and the alopecia symptoms are further aggravated to a certain extent.
In order to solve the above problems, chinese patent CN108210937a discloses a β -cyclodextrin inclusion compound of pyrrolidinyl diaminopyrimidine oxide, wherein the cyclodextrin is used to encapsulate the pharmaceutical ingredient of pyrrolidinyl diaminopyrimidine oxide in the composition, thereby increasing the solubility of pyrrolidinyl diaminopyrimidine oxide and improving the bioavailability thereof. However, the target composition is prepared by adopting a large amount of cyclodextrin components, preparing the cyclodextrin components into hydroxypropyl-beta-cyclodextrin or dimethyl-beta-cyclodextrin in use and then preparing the cyclodextrin components in a specific mode, wherein the production cost is high. In addition, although it is pointed out that the composition prepared by the method of the present invention has improved water solubility, the component prepared therefrom is still solid powder, and it is possible to absorb it after being prepared into a specific product, thereby achieving the effect of preventing hair loss, however, since the specific use method of the composition or the related product is not disclosed in the patent, the effect of preventing hair loss of the composition is not disclosed, and thus further development of the related product is required to solve the above-mentioned problems.
In addition, chinese patent CN201910036145 discloses a nano composition comprising pyrrolidinyl diaminopyrimidine oxide for the preparation of a medicament for the treatment of androgenetic alopecia, alopecia areata, telogen effluvium, anagen hair loss syndrome. However, on the one hand, although the disclosed compositions comprise pyrrolidinyl diaminopyrimidine oxides, they also require 5 alpha-reductase inhibitors, androgen receptor antagonists, vasodilators, anti-inflammatory bacteriostats, etc. components to interact together to achieve their stated efficacy, which is complex and costly. On the other hand, it requires a large amount of alcohol solvents such as propylene glycol, and the hair growth promoting liquid can be prepared, and the hair loss preventing effect can be achieved by using the same in specific amounts and for specific periods. However, when the composition with high alcohol solvent content is prepared into products such as shampoo, the micro-environment of the scalp of a user can be seriously affected when the composition is frequently used, so that the scalp and the hair become dry, and even the sebum membrane of some users is damaged, unnecessary trouble is brought.
Disclosure of Invention
In view of the above technical problems, a first aspect of the present invention provides an anti-hair loss hair-growing composition, which at least comprises the following components in percentage: a) At least 0.05wt% of pyrrolidinyl diaminopyrimidine; (b) at least 0.05wt% adenosine; (c) at least 0.05wt% oligopeptide; (d) at least 1.0wt% lecithin; (e) at least 5wt% polyglycerol fatty acid ester; a dispersion medium for the remainder; wherein the lecithin is egg yolk lecithin; the saponification value of the egg yolk lecithin is 195-212 mgKOH/g.
The egg yolk lecithin is prepared from eggs by removing neutral fat in egg yolk powder by using acetone, and extracting the egg yolk lecithin by using absolute ethyl alcohol. The yolk lecithin is a mixture of phospholipids, and the components, namely the contents, are different according to the extraction method, the raw material sources and the like, and correspondingly have different physicochemical effects.
The term "saponification value" in the present invention refers to the milligrams of potassium hydroxide consumed by hydrolysis of 1g of lecithin, and is a physical quantity to some extent used to characterize its molecular weight.
The term "acid value" as used herein refers to the milligrams of potassium hydroxide required to neutralize the free fatty acids in the lecithin compound.
The term "iodine value" in the present invention means the number of grams of iodine consumed per 100g of lecithin when an unsaturated bond in the structure of lecithin compound undergoes an addition reaction with halogen.
The saponification value of the egg yolk lecithin is 195-212 mgKOH/g.
Further, the acid value of the egg yolk lecithin is not higher than 30mgKOH/g.
Further, the iodine value of the egg yolk lecithin is 65-73 gI/100g.
Further, the content of phosphatidylcholine in the egg yolk lecithin is not less than 75wt%; further, the content of phosphatidylcholine in the egg yolk lecithin is not less than 85wt%; further, the content of phosphatidylcholine in the egg yolk lecithin is not less than 90wt%.
The egg yolk lecithin content in the invention is at least 1.0wt%, and further, the content is 2-5 wt%. The egg yolk lecithin of the present invention may be used as a commercially relevant product such as E-90 lecithin from North America vitamin technology Co.
The applicant found during the completion of the present invention that the selection of lecithin has an important effect on preventing hair loss and hair growth of the composition of the present invention, and if the specific components of lecithin and the content in the composition are optimized improperly, the hair loss prevention and hair growth effects thereof are easily reduced, particularly in that the subjects have substantially no improvement in feedback hair regeneration after 2 months of use, and the number of improvement in hair loss during hair washing and hair combing is small. In addition, the applicant also found that the results obtained after foam testing of the relevant samples using different lecithins are also greatly different, showing that the improper composition of the lecithins used can significantly affect the foam value of the samples, and the phenomena of insufficient foam, insufficient foam amount and the like occur. In view of the above, the applicant speculates that the composition mainly uses the inhibitory effect of components such as pyrrolidinyl diaminopyrimidine, adenosine, and oligopeptides on the damage of components such as hydrogen peroxide and testosterone to HDPC cells and HaCaT cells, thereby reducing alopecia, but because pyrrolidinyl diaminopyrimidine has poor water solubility, it cannot be directly absorbed by the scalp, and when components such as lecithin are not selected properly, it cannot exert a strong effect with components such as adenosine and oligopeptides, thereby failing to exert a sufficient alopecia preventing effect. The egg yolk lecithin with a specific structure can improve the mutual diffusion capability of components such as pyrrolidinyl diaminopyrimidine, adenosine, oligopeptide and the like to a certain extent, strengthen interaction force between the components, generate more dense foam when in use, reduce the surface tension of the composition, and enable the composition to permeate into the scalp more quickly, so that the composition can be fully absorbed to play a corresponding role. When egg yolk lecithin with low phosphatidylcholine content or other types of lecithins (such as soybean lecithin) are adopted, interaction between the lecithin and components such as pyrrolidinyl diaminopyrimidine, adenosine, oligopeptide and the like is changed, and balance among the components is possibly broken, so that the foam amount is influenced and the efficacy is reduced.
The oligopeptide is oligopeptide, the content of the oligopeptide is at least 0.05wt%, and further, the content of the oligopeptide is 0.05-3 wt%.
As a preferred embodiment, the oligopeptide has a molecular weight of 100-1000 daltons.
Further, the oligopeptide is acetyl tetrapeptide-3, i.e., it is a tetrapeptide.
The anti-hair loss and hair growth promoting composition disclosed by the invention is wrapped with a proper amount of polyglycerol fatty acid ester, which is an ester compound obtained by reacting glycerol with different fatty acids. According to the difference of fatty acid structures reacting with glycerol in the structure, the polyglycerol fatty acid ester has different structures, correspondingly has different HLB values and different physicochemical properties, and has a vital effect on the dispersion of components such as pyrrolidinyl diaminopyrimidine, adenosine, lecithin and the like, the scalp absorption speed of the shampoo and hair care preparation during use and the like.
As a preferred embodiment, the polyglyceryl fatty acid ester includes decapolyglyceryl monolaurate and triglyceryl diisostearate. The decapolyglycerol monolaurate is polyglycerol-10 laurate (CAS: 34406-66-16), and the triglycerol diisostearate is polyglycerol-3 diisostearate (CAS: 66082-42-6).
Further, the content of the triglycerol diisostearate is at least 35wt% of the polyglyceryl fatty acid ester.
Further, the mass ratio of the decaglycerol monolaurate to the triglycerol diisostearate is (1-2): (0.8-1.8).

the applicant finds that the foam amount of the composition can be obviously improved when the polyglycerol fatty acid esters with two different specific structures are compounded and used in the process of completing the invention, so that more and more dense foam is generated when the composition is used, and components such as pyrrolidinyl diaminopyrimidine, lecithin, adenosine, oligopeptide and the like in the composition are more fully contacted with scalp and hair, thereby being beneficial to improving Fan Tuoyo hair efficacy of the composition. Meanwhile, because the chain lengths of fatty acids connected with glycerol in the structures of the decaglycerol monolaurate and the triglycerol diisostearate are different, a plurality of different physical entanglement networks can be formed when micelles of components such as firm pyrrolidinyl diaminopyrimidine, adenosine and the like are formed in a dispersion medium, and the effective components with different sizes are wrapped, so that the transdermal effect is accelerated.
As a preferred embodiment, it further comprises at least 0.1% by weight of panthenol and 0.1% by weight of cholesterol.
Further, the content of the panthenol is 0.1 to 1wt%, and more preferably, the content of the panthenol is 0.1 to 0.8wt%.
Further, the cholesterol content is 0.1-1 wt%; further preferably, the cholesterol content is 0.3 to 0.7wt%.
As a preferred embodiment, the dispersion medium according to the invention comprises a solvent and a dispersing aid.
Further, the dispersing aid comprises caprylic-capric triglyceride and polyglycerol ricinoleate (PGPR).
Further, the weight ratio of the caprylic-capric triglyceride to the polyglycerol ricinoleate is (1-2): (1-2); further, the weight ratio of the caprylic-capric triglyceride to the polyglycerol ricinoleate is 1:1.8.
The polyglycerol ricinoleate is prepared by esterification reaction of polyglycerol and condensed castor oil fatty acid. The content of the polyglycerol ricinoleate in the composition is 2.5-4.5 wt%; further, the polyglycerol ricinoleate is contained in the composition of the present invention in an amount of 3.2 to 3.9wt%.
The applicant found that the use of specific proportions of caprylic-capric triglyceride and polyglycerol ricinoleate as part of the dispersion medium, while employing specific proportions of the polyglyceryl fatty acid esters (in particular, the decapolyglycerol monolaurate and the triglyceryl diisostearate in a weight ratio of 1.2:1), helps to further improve the foam content of the composition, and even in higher hardness, in water containing more mineral components, still ensures an excellent foam content, avoiding differences in efficacy due to the different water employed when the product is used. It is possible that when the specific polyglycerin fatty acid ester and the dispersing aid are adopted, the micelle formed in the system reaches the most stable state, and inorganic ions such as magnesium ions, calcium ions and the like in water cannot effectively destroy the micelle, so that the foam amount of the sample cannot be obviously influenced. In addition, the applicant also found that by controlling the components and proportions of lecithin, polyglycerol fatty acid ester and dispersing aid, excellent hair-loss preventing and hair-growing effects can be ensured on the premise of reducing the alcohol solvents (mainly ethanol and propylene glycol) in the composition to not more than 10wt%. The reduction of alcohol solvents such as ethanol, propylene glycol and the like greatly reduces the use limit of the composition, so that some people with sensitive skin can use the composition for a long time, and the application range of the composition can be enlarged.
Further, the solvent described in the present invention includes water and an alcohol solvent.
Further, the alcohol solvent is present in the composition of the present invention in an amount of no more than 15wt%.
Further, the alcohol solvent comprises propylene glycol, ethanol and glycerin.
Further, the total content of propylene glycol and ethanol in the composition of the present invention is not higher than 12wt%.
Further, the total content of propylene glycol and ethanol in the composition of the present invention is not more than 10wt%.
As a preferable technical scheme, the content of the pyrrolidinyl diaminopyrimidine is 1.2-3.5 wt%.
Further, the content of the adenosine is 0.05-3 wt%.
The preparation method of the hair-loss preventing and hair-growing composition is not particularly limited, and the hair-loss preventing and hair-growing composition can be prepared according to a conventional mode, for example, pyrrolidinyl diaminopyrimidine oxide, lecithin and cholesterol can be mixed and homogenized, and then added into the mixture of adenosine, panthenol, deionized water and the rest raw materials, stirred and mixed, filtered, homogenized into about 100nm by high-pressure microfluidics, filtered and filled.
In a second aspect, the present invention provides a hair care shampoo formulation comprising an anti-hair loss hair care composition as described above; the content of the anti-hair loss hair-growing composition in the shampoo preparation is at least 5wt%; further, the content of the hair loss preventing and hair growing composition in the shampoo preparation is 10-50wt%.
As a preferred technical scheme, the formulation of the hair care shampoo formulation comprises a liquid formulation and a paste.
Further, the hair care shampoo preparation can be shampoo, hair conditioner, essence and the like; it is preferably a shampoo.
Compared with the prior art, the invention has the following beneficial effects:
(1) In the hair loss prevention and hair growth composition, under the interaction of pyrrolidinyl diaminopyrimidine, adenosine, oligopeptide, egg yolk lecithin and the like, the damage of components such as hydrogen peroxide, testosterone and the like to HDPC cells and HaCaT cells is effectively inhibited, the growth and proliferation differentiation of fibroblast growth factors in dermal papilla cells are accelerated, so that the alopecia is reduced, and the hair quantity is improved.
(2) The hair loss prevention and hair growth composition adopts egg yolk lecithin with specific phosphatidylcholine, specific iodine value and specific saponification value, improves the mutual diffusion capability of components such as pyrrolidinyl diaminopyrimidine, adenosine, oligopeptide and the like, strengthens interaction force between the components, simultaneously generates more dense foam when in use, reduces the surface tension of the composition, and enables the composition to permeate into scalp more quickly, so that the composition can be fully absorbed, and improves the treatment effect of the composition on alopecia symptoms.
(3) The anti-hair loss hair-care composition disclosed by the invention is compounded by adopting two polyglycerol fatty acid esters with different specific structures, so that the foam quantity of the composition is obviously improved, and more dense foams are generated when the composition is used, so that the components such as pyrrolidinyl diaminopyrimidine, lecithin, adenosine, oligopeptide and the like in the composition are fully contacted with scalp and hair, and the Fan Tuoyo hair-care composition is improved.
(4) In the hair loss prevention hair-growing composition, specific compound polyglycerol fatty acid ester (especially, the ten polyglycerol monolaurate and the triglycerol diisostearate with the weight ratio of 1.2:1) is adopted, and meanwhile, when the caprylic-capric triglyceride and the polyglycerol ricinoleate with the specific ratio are adopted as part of a dispersion medium, the foam quantity of the composition is further improved, so that the composition has higher hardness, and excellent foam quantity can still be ensured in water containing more mineral components, and the difference in efficacy caused by different water adopted when the product is used is avoided.
(5) In the hair loss prevention and hair growth composition, the components and the proportion of lecithin, polyglycerol fatty acid ester and a dispersing auxiliary are regulated and controlled, so that the content of alcohol solvents (mainly ethanol and propylene glycol) in the composition is effectively reduced, and the excellent hair loss prevention and hair growth effects can be ensured on the premise that the content of alcohol solvents is not higher than 10wt%. The reduction of alcohol solvents such as ethanol, propylene glycol and the like greatly reduces the use limit of the composition, so that some people with sensitive skin can use the composition for a long time, and the application range of the composition can be enlarged.

Detailed Description
The technical features of the technical solution provided in the present invention will be further clearly and completely described in connection with the detailed description below, and it is apparent that the described embodiments are only some embodiments of the present invention, not all embodiments. All other embodiments, which can be made by those skilled in the art based on the embodiments of the invention without making any inventive effort, are intended to be within the scope of the invention.

Example 6: the embodiment provides an anti-hair loss and hair growth composition, which comprises the following components in percentage:

Performance testing
The applicant took 6.3g of the samples of the above examples and mixed and stirred with 93.7g of hard water (92 mg/L of magnesium ions and 71mg/L of calcium ions) to obtain samples S-1, S-2, S-3, S-4, S-5 and S-6, respectively, and foam test the samples, specifically, the samples were respectively added into an electric foam meter, the foam meter was turned on, stirring was completed for one minute, and the foam was immediately poured into a measuring cylinder to measure the amount of foam and record data. The procedure was repeated three times and averaged, and the test results thereof are shown in the following table 1.

The raw material composition of the control sample is as follows: 2.3 parts by weight of pyrrolidinyl diaminopyrimidine oxide, 100ppm of oligopeptide (acetyl tetrapeptide-3) and 1000ppm of adenosine, 0.5 part by weight of panthenol, 7.5 parts by weight of polyglycerol-10 laurate, 2.5 parts by weight of caprylic-capric triglyceride, 2.5 parts by weight of ethanol, 7.5 parts by weight of propylene glycol, 2.5 parts by weight of glycerol, 2.5 parts by weight of lecithin (phosphatidylcholine content about 26.+ -. 1, soybean lecithin PC-25 from Hebeige vitamin technology Co., ltd.), 0.5 parts by weight of cholesterol and 67 parts by weight of deionized water.
In addition, the applicant took samples of the above examples, respectively, added deionized water to prepare samples S-1', S-2', S-3', S-4', S-5', S-6' having a composition content of 6wt%, invited 30 eligible male/female subjects to participate in the test for each sample, and the subjects were shampooed at a frequency of 48 hours during the test, continuously used for 8 weeks, and after the end of the experiment, a questionnaire was conducted to collect valid data therein (which would be discarded according to the test conditions), and the sample was subjected to a correlation test again with less than 83% (i.e., more than 5 subjects were not available). During the test, the subject must not use any wash, hair care and styling products (gels, sprays, etc.) other than the test product; and the length (without trimming) and style and color (i.e. without perming or dyeing) of the hair should not be changed during the test. The test results are shown in Table 2 below, wherein the sample use experience cannot be accurately reflected, and thus is not recorded, since the actual effective data of the samples S-3 'and S-6' are already less than 65%.

Claims (3)
1. An anti-hair loss and hair-growing composition is characterized by comprising the following components in percentage by weight: 1.2-3.5wt% of pyrrolidinyl diaminopyrimidine; 0.05-3wt% of adenosine; 0.05-3wt% of oligopeptide; 2-5wt% of lecithin; at least 5wt% polyglycerol fatty acid ester; 0.1-0.8wt% of panthenol; 0.3-0.7wt% of cholesterol; a dispersion medium for the remainder;
the oligopeptide is acetyl tetrapeptide-3;
the lecithin is egg yolk lecithin;
the saponification value of the egg yolk lecithin is 195-212 mgKOH/g;
the iodine value of the egg yolk lecithin is 65-73 gI/100g;
the content of phosphatidylcholine in the egg yolk lecithin is not less than 75wt%;
the polyglyceryl fatty acid esters include decapolyglyceryl monolaurate and triglyceryl diisostearate;
the mass ratio of the decaglycerol monolaurate to the triglycerol diisostearate is 1.2:1, a step of;
the dispersion medium comprises a solvent and a dispersion auxiliary agent;
the dispersing aid comprises caprylic-capric triglyceride and polyglycerol ricinoleate;
the weight ratio of the caprylic acid-capric acid triglyceride to the polyglycerol ricinoleate is (1-2): (1-2);
the content of the polyglycerol ricinoleate in the composition is 2.5-4.5wt%;
the solvent includes water and alcohol solvents;
the alcohol solvent comprises propylene glycol, ethanol and glycerol;
the total content of propylene glycol and ethanol in the composition is not higher than 10wt%.
2. A hair care shampoo formulation comprising the anti-hair loss composition according to claim 1; the content of the anti-hair loss hair-growing composition in the shampoo preparation is at least 5wt%.
3. The hair care shampoo formulation according to claim 2, wherein the dosage form comprises a liquid dosage form and a paste.
