Abstract
The invention belongs to the technical field of A61P17/14, and particularly relates to an anti-hair loss and hair-nourishing composition and a hair washing and hair care preparation thereof. The paint at least comprises the following components in percentage by weight: a) at least 0.05 wt% of a pyrrolidinyl diaminopyrimidine; (b) at least 0.05 wt% adenosine; (c) at least 0.05 wt% oligopeptide; (d) at least 1.0 wt% lecithin; (e) at least 5 wt% of a polyglyceryl fatty acid ester; and the balance of a dispersion medium; wherein the lecithin is egg yolk lecithin; the saponification value of the egg yolk lecithin is 195-212 mgKOH/g. In the anti-hair loss and hair growth composition, under the interaction of pyrrolidinyl diaminopyrimidine, adenosine, oligopeptide, egg yolk lecithin and the like, the damage of components such as hydrogen peroxide and testosterone to HDPC (high-density polyethylene carbonate) cells and HaCaT (HaCaT) cells is effectively inhibited, and the growth, proliferation and differentiation of fibroblast growth factors in dermal papilla cells are accelerated, so that hair loss is reduced, and the hair growth is improved.
Description
Technical Field
The invention belongs to the technical field of A61P17/14, and particularly relates to an anti-hair loss and hair nourishing composition and a hair washing and protecting preparation thereof.
Background
With the increasing of working pressure, the accelerating of life rhythm, the environmental impact, the use and the popularization of hair-dressing and shaping products such as hair dyeing and perm and the like, the number of the hair loss patients is increased in a great proportion every year. Meanwhile, investigation shows that the alopecia population accounts for a higher degree than the normal population due to the negative effect of the alopecia on the appearance, and the alopecia symptoms are further aggravated to a certain degree.
In order to solve the above problems, chinese patent CN108210937A discloses a pyrrolidinyl diaminopyrimidine oxide beta-cyclodextrin base clathrate, which uses cyclodextrin to wrap the pyrrolidinyl diaminopyrimidine oxide drug component in the composition, thereby increasing the solubility of pyrrolidinyl diaminopyrimidine oxide and improving the bioavailability thereof. However, the target composition is prepared by using a large amount of cyclodextrin components, and the target composition is prepared by preparing hydroxypropyl-beta-cyclodextrin or dimethyl-beta-cyclodextrin in a specific manner when the target composition is used, so that the production cost is high. In addition, although it is pointed out that the water solubility of the composition prepared by the method of the present invention is improved, the components prepared therefrom are solid powders, and it is necessary to prepare the composition into a specific product before it is absorbed, thereby achieving the anti-hair loss effect, however, since the patent does not disclose a specific use method of the composition or a related product, nor the anti-hair loss effect of the composition, further development of related products is required to solve the above problems.
In addition, chinese patent CN201910036145 discloses a nano composition containing pyrrolidinyl diaminopyrimidine oxide for preparing a medicament for treating androgenetic alopecia, alopecia areata, telogen effluvium, and anagen-phase osteoporosis syndrome. On the one hand, however, although the composition disclosed therein contains pyrrolidinyl diaminopyrimidine oxide, it also requires 5 α -reductase inhibitors, androgen receptor antagonists, vasodilators, anti-inflammatory bacteriostats, and the like to interact together to achieve the effects described herein, and is complicated and expensive. On the other hand, a large amount of alcohol solvent such as propylene glycol is needed, and the anti-alopecia effect can be realized by using the alcohol solvent in a specific dosage and period even though the alcohol solvent is prepared into products such as hair growth promoting liquid. However, when such a composition with a high alcohol solvent content is prepared into products such as shampoo and the like, and is frequently used, the micro environment of the scalp of a user is seriously affected, the scalp and hair quality are dried, and even a sebum membrane of some users is damaged, so that unnecessary troubles are brought.
Disclosure of Invention
In view of the above technical problems, the first aspect of the present invention provides an anti-alopecia hair-growing composition, which at least comprises the following components by weight: a) at least 0.05 wt% of a pyrrolidinyldiaminopyrimidine; (b) at least 0.05 wt% adenosine; (c) at least 0.05 wt% oligopeptide; (d) at least 1.0 wt% lecithin; (e) at least 5 wt% of a polyglyceryl fatty acid ester; and the balance of a dispersion medium; wherein the lecithin is egg yolk lecithin; the saponification value of the egg yolk lecithin is 195-212 mgKOH/g.
The yolk lecithin is prepared by removing neutral fat from yolk powder by using acetone and extracting the yolk lecithin from the yolk powder by using absolute ethyl alcohol. The egg yolk lecithin is a mixture of phospholipids, and the components, namely the contents, of the egg yolk lecithin are different according to different extraction methods, raw material sources and the like, so that the egg yolk lecithin has different corresponding physicochemical effects.
The term "saponification number" in the present invention means the number of milligrams of potassium hydroxide consumed for the hydrolysis of 1g of lecithin, a physical quantity that is used to some extent to characterize its molecular weight.
The term "acid value" in the present invention means the number of milligrams of potassium hydroxide required for neutralization of the free fatty acids in the lecithin compound.
The term "iodine value" in the present invention means the amount of iodine consumed per 100g of lecithin in the case where an addition reaction of an unsaturated bond in the structure of the lecithin compound with a halogen is carried out.
The saponification value of the egg yolk lecithin is 195-212 mgKOH/g.
Further, the acid value of the egg yolk lecithin is not higher than 30 mgKOH/g.
Further, the iodine value of the egg yolk lecithin is 65-73 gI/100 g.
Further, the content of phosphatidylcholine in the egg yolk lecithin is not less than 75 wt%; further, the content of phosphatidylcholine in the egg yolk lecithin is not less than 85 wt%; further, the content of phosphatidylcholine in the egg yolk lecithin is not less than 90 wt%.
The content of the egg yolk lecithin in the invention is at least 1.0 wt%, and further, the content is 2-5 wt%. The egg yolk lecithin of the present invention may be a commercially available related product such as E-90 lecithin from North and America industries, Inc.
In the process of the invention, the applicant finds that the selection of lecithin has important influence on the hair loss prevention and hair growth of the composition, if the specific components of the lecithin and the content of the lecithin in the composition are not optimized, the hair loss prevention and hair growth effects are easily reduced, and the specific results show that the subjects have no feedback on the improvement of hair growth after 2 months of use, and the number of people for improving the hair loss during hair washing and hair combing is small. In addition, the applicant also found that when different lecithins are adopted, the results of the relevant samples are greatly different after the foam test is carried out, and that the foam value of the sample is obviously influenced by improper lecithin component, and the phenomena of insufficient and dense foam, insufficient foam amount and the like occur. In combination with the above results, the applicant speculates that the composition mainly utilizes the inhibition effect of the components such as pyrrolidinyl diaminopyrimidine, adenosine, oligopeptide and the like on the damage of HDPC cells, HaCaT cells and the like, thereby reducing alopecia, but because the water solubility of the pyrrolidinyl diaminopyrimidine is poor, the pyrrolidinyl diaminopyrimidine cannot be directly absorbed by the scalp, and when the components such as lecithin and the like are not selected, the pyrrolidinyl diaminopyrimidine cannot form a strong effect with the components such as adenosine, oligopeptide and the like, thereby failing to exert a sufficient anti-alopecia effect. The egg yolk lecithin with a specific structure can improve the mutual diffusion capacity of components such as the pyrrolidinyl diaminopyrimidine, the adenosine, the oligopeptide and the like to a certain extent, enhances the interaction force between the components, generates more thick foam when in use, reduces the surface tension of the composition, and enables the composition to permeate into the scalp more quickly, so that the composition can be fully absorbed to play a corresponding role. When egg yolk lecithin with low phosphatidylcholine content or other kinds of lecithin (such as soybean lecithin) is used, the interaction between lecithin and components such as pyrrolidinyl diaminopyrimidine, adenosine, oligopeptide, etc. is changed, and the balance therebetween may be broken, thereby reducing the efficacy while affecting the amount of foam thereof.
The oligopeptide is oligopeptide, the content of the oligopeptide is at least 0.05 wt%, and further the content of the oligopeptide is 0.05-3 wt%.
As a preferable technical scheme, the molecular weight of the oligopeptide is 100-1000 daltons.
Further, the oligopeptide is acetyl tetrapeptide-3, i.e. it is tetrapeptide.
The anti-hair loss and hair-growing composition comprises a proper amount of polyglycerol fatty acid ester which is an ester compound obtained by the reaction of glycerol and different fatty acids. According to the structure of the fatty acid which reacts with glycerol, the polyglyceryl fatty acid ester has different structures, correspondingly has different HLB values and different physical and chemical properties, and plays a vital role in dispersing the components such as the pyrrolidinyl diaminopyrimidine, the adenosine, the lecithin and the like in the application, the scalp absorption speed of the shampoo and hair care preparation in use and the like.
As a preferable technical solution, the polyglycerin fatty acid ester includes decaglyceryl monolaurate and triglycerol diisostearate. The decaglycerol monolaurate is polyglycerol-10 laurate (CAS: 34406-66-16), and the triglycerol diisostearate is polyglycerol-3 diisostearate (CAS: 66082-42-6).
Further, the content of the triglycerol diisostearate is at least 35 wt% of the polyglycerin fatty acid ester.
Further, the mass ratio of the decaglycerol monolaurate to the triglycerol diisostearate is (1-2): (0.8-1.8).
Further, the mass ratio of the decaglycerol monolaurate to the triglycerol diisostearate is 1.2: 1.
in the process of completing the invention, the applicant finds that the foam amount of the composition can be obviously improved when the two polyglyceryl fatty acid esters with different specific structures are compounded for use, so that the composition can generate more and more continuous foams when in use, and therefore, the components such as the pyrrolidinyl diaminopyrimidine, the lecithin, the adenosine, the oligopeptide and the like in the composition can be more fully contacted with the scalp and the hair, and the hair conditioning and growing effects of the composition can be improved. Meanwhile, because the chain lengths of the fatty acids connected with the glycerol in the structures of the decaglycerol monolaurate and the triglycerol diisostearate are different, a plurality of different physical entanglement networks can be formed when the micelle for stabilizing the components such as the pyrrolidinyl diaminopyrimidine, the adenosine and the like is formed in a dispersion medium, so that the effective components with different sizes are wrapped, and the transdermal effect is accelerated.
As a preferred embodiment, it further comprises at least 0.1% by weight panthenol and 0.1% by weight cholesterol.
Further, the content of panthenol is 0.1 to 1 wt%, and more preferably, the content of panthenol is 0.1 to 0.8 wt%.
Further, the content of cholesterol is 0.1-1 wt%; more preferably, the cholesterol content is 0.3 to 0.7 wt%.
As a preferable technical solution, the dispersion medium in the present invention includes a solvent and a dispersion aid.
Further, the dispersing aid comprises caprylic-capric triglyceride and polyglycerol ricinoleate (PGPR).
Further, the weight ratio of the caprylic-capric triglyceride to the polyglycerol ricinoleate is (1-2): (1-2); further, the weight ratio of the caprylic-capric triglyceride to the polyglycerol polyricinoleate is 1: 1.8.
The polyglycerol ricinoleate is prepared by esterification reaction of polyglycerol and condensed castor oil fatty acid. The content of the polyglycerol ricinoleate in the composition is 2.5-4.5 wt%; further, the content of the polyglycerol ricinoleate in the composition is 3.2-3.9 wt%.
The applicant finds that while using a specific combination of polyglyceryl fatty acid esters (in particular, 1.2:1 by weight of decaglyceryl monolaurate and triglyceryl diisostearate), using a specific ratio of caprylic-capric triglyceride and polyglyceryl ricinoleate as part of the dispersion medium helps to further improve the foam level of the composition, even in water with higher hardness and more mineral components, the excellent foam level can still be ensured, and differences in efficacy due to different water used in the product can be avoided. Probably, when the specific polyglycerin fatty acid ester and the dispersing aid are used, micelles formed in the system reach the most stable state, and inorganic ions such as magnesium ions and calcium ions in water cannot effectively destroy the micelles, so that the foam amount of a sample cannot be obviously influenced. In addition, the applicant also finds that the excellent hair loss preventing and hair growth promoting effects can be still ensured on the premise of reducing the alcohol solvents (mainly ethanol and propylene glycol) in the composition to be not higher than 10 wt% by regulating and controlling the components and the proportion of the lecithin, the polyglycerol fatty acid ester and the dispersing auxiliary agent. And the reduction of alcohol solvents such as ethanol, propylene glycol and the like greatly reduces the use limit of the composition, so that the composition can be used for a long time by some people with sensitive skin, and the application range of the composition can be expanded.
Further, the solvent in the present invention includes water and an alcohol solvent.
Further, the content of the alcohol solvent in the composition of the present invention is not more than 15 wt%.
Further, the alcohol solvent comprises propylene glycol, ethanol and glycerol.
Further, the total content of propylene glycol and ethanol in the composition of the present invention is not higher than 12 wt%.
Further, the total content of propylene glycol and ethanol in the composition of the present invention is not higher than 10 wt%.
As a preferable technical scheme, the content of the pyrrolidinyl diaminopyrimidine is 1.2-3.5 wt%.
Further, the content of the adenosine is 0.05-3 wt%.
The preparation method of the hair-loss-preventing and hair-nourishing composition is not particularly limited, and the hair-loss-preventing and hair-nourishing composition can be prepared according to a conventional mode, for example, the pyrrolidinyl diaminopyrimidine oxide, lecithin and cholesterol are mixed and homogenized, then the mixture is added into the mixture of adenosine, panthenol, deionized water and the rest raw materials, the mixture is stirred and mixed, the mixture is filtered, the mixture is homogenized to about 100nm by high-pressure microjet, and the hair-loss-preventing and hair-nourishing composition is obtained by filtering and filling.
In a second aspect, the present invention provides a hair care shampoo formulation comprising, as components, the above hair loss prevention and hair growth promotion composition; the hair loss preventing and hair growing composition accounts for at least 5 wt% of the shampoo preparation; furthermore, the anti-hair loss and hair growth promoting composition accounts for 10-50 wt% of the shampoo preparation.
As a preferable technical scheme, the hair washing and caring preparation comprises a liquid preparation and an ointment.
Further, the hair washing and protecting preparation can be shampoo, hair conditioner, essence and the like; it is preferably a shampoo.
Compared with the prior art, the invention has the following beneficial effects:
(1) in the anti-hair loss and hair-growing composition, under the interaction of pyrrolidinyl diaminopyrimidine, adenosine, oligopeptide, egg yolk lecithin and the like, the damage of components such as hydrogen peroxide, testosterone and the like to HDPC (high density polyethylene) cells and HaCaT (HaCaT) cells is effectively inhibited, the growth, proliferation and differentiation of fibroblast growth factors in dermal papilla cells are accelerated, and therefore hair loss is reduced, and the hair quantity is improved.
(2) The anti-hair loss and hair-growing composition adopts the egg yolk lecithin with specific phosphatidylcholine, specific iodine value and specific saponification value, improves the mutual diffusion capacity of components such as pyrrolidinyl diaminopyrimidine, adenosine, oligopeptide and the like, strengthens the interaction force among the components, generates more dense foam when in use, reduces the surface tension of the composition, and enables the foam to permeate into the scalp more quickly, so that the composition can be absorbed fully, and the treatment effect of the composition on the symptoms of hair loss is improved.
(3) The hair loss preventing and hair growing composition disclosed by the invention adopts two polyglycerol fatty acid esters with different specific structures to be compounded for use, so that the foam amount of the composition is obviously improved, and more dense foams are generated when the composition is used, and therefore, the components such as pyrrolidinyl diaminopyrimidine, lecithin, adenosine and oligopeptide in the composition are more fully contacted with scalp and hair, and the hair loss preventing and hair growing effect of the composition is improved.
(4) In the hair loss preventing and hair nourishing composition, the specifically compounded polyglycerol fatty acid ester (especially the decaglycerol monolaurate and the triglycerol diisostearate with the weight ratio of 1.2: 1) is adopted, and the caprylic-capric triglyceride and the polyglycerol ricinoleate with the specific ratios are adopted as a part of the dispersion medium, so that the foam amount of the composition is further improved, the composition can still ensure excellent foam amount in water with higher hardness and more mineral components, and the difference of effects caused by different water adopted in the product during use is avoided.
(5) In the hair loss preventing and hair growing composition, the content of alcohol solvents (mainly ethanol and propylene glycol) in the composition is effectively reduced by regulating and controlling the components and the proportion of lecithin, polyglycerol fatty acid ester and dispersing auxiliary agent, so that the excellent hair loss preventing and hair growing effect can still be ensured on the premise that the content of the alcohol solvents (mainly ethanol and propylene glycol) in the composition is not higher than 10 wt%. And the reduction of alcohol solvents such as ethanol, propylene glycol and the like greatly reduces the use limit of the composition, so that the composition can be used for a long time by people with sensitive skin, and the application range of the composition can be expanded.

Detailed Description
The technical features of the technical solutions provided by the present invention will be further clearly and completely described below with reference to the specific embodiments, and it should be apparent that the described embodiments are only a part of the embodiments of the present invention, and not all of the embodiments. All other embodiments, which can be derived by a person skilled in the art from the embodiments given herein without making any creative effort, shall fall within the protection scope of the present invention.
Example 1: the embodiment provides an anti-hair loss and hair growth composition, which comprises the following components in percentage by weight:
a)3.0 wt% pyrrolidinyl diaminopyrimidine;
b)0.10 wt% adenosine;
c)0.50 wt% oligopeptide;
d) 3.3% by weight lecithin;
e)8 wt% of polyglycerin fatty acid ester;
f)0.3 wt% panthenol;
g)0.7 wt% cholesterol;
and the balance of a dispersing medium;
the dispersing medium is a mixture of a solvent and a dispersing aid, wherein the dispersing aid consists of caprylic-capric triglyceride and polyglycerol ricinoleate, the weight ratio of the dispersing aid to the polyglycerol ricinoleate is 1:1.8, and the content of the polyglycerol ricinoleate in the composition is 3.6 wt%; the solvent consists of deionized water and an alcohol solvent, wherein the content of the alcohol solvent in the composition is 12 wt%, the alcohol solvent consists of propylene glycol, ethanol and glycerin, the mass ratio of the propylene glycol to the ethanol is 3:1, and the content of the glycerin in the composition is 4 wt%; the lecithin is yolk lecithin; the average saponification value of the egg yolk lecithin is 205 plus or minus 2mgKOH/g, the acid value is 28 plus or minus 1mgKOH/g, the iodine value is 68 plus or minus 1gI/100g, the content of phosphatidylcholine is about 92 plus or minus 1 wt%, and E-90 lecithin of North America industry vitamin technology Limited company is adopted; the polyglycerol fatty acid ester consists of decaglycerol monolaurate and triglycerol diisostearate, and the weight ratio of the polyglycerol fatty acid ester to the triglycerol diisostearate is 1.2: 1; the oligopeptide is acetyl tetrapeptide-3.

Performance testing
The applicant takes 6.3g of samples in the above examples, mixes and stirs with 93.7g of hard water (wherein the content of magnesium ions is 92mg/L, and the content of calcium ions is 71mg/L) to obtain samples S-1, S-2, S-3, S-4, S-5 and S-6, and carries out foam test on the samples, specifically, the samples are added into an electric foam instrument respectively, the electric foam instrument is opened, the stirring is carried out for one minute, the foaming is finished, and the foam is poured into a measuring cylinder immediately to measure the foam amount and record data. The procedure was repeated three times, and the average value was taken, and the test results thereof are shown in table 1 below.

The raw material components of the control sample are as follows: 2.3 parts by weight of pyrrolidinyl diaminopyrimidine oxide, 100ppm of oligopeptide (acetyl tetrapeptide-3) and 1000ppm by weight of adenosine, 0.5 parts by weight of panthenol, 7.5 parts by weight of polyglycerol-10 laurate, 2.5 parts by weight of caprylic-capric triglyceride, 2.5 parts by weight of ethanol, 7.5 parts by weight of propylene glycol, 2.5 parts by weight of glycerol, 2.5 parts by weight of lecithin (phosphatidylcholine content about 26. + -.1, soybean lecithin PC-25 by North America vitamin technology Co., Ltd.), 0.5 parts by weight of cholesteryl alcohol and 67 parts by weight of deionized water.
In addition, the applicant takes the samples in the above examples, adds deionized water to prepare samples S-1 ', S-2', S-3 ', S-4', S-5 'and S-6' with the composition content of 6 wt%, and invites 30 qualified male/female subjects to take the test for each sample, during which the subjects use the above samples to shampoo at a frequency of once every 48 hours, continuously use for 8 weeks, and after the experiment is finished, perform questionnaire survey, collect effective data (wherein the data according to the test conditions are discarded), wherein the effective data is less than 83% (i.e. the data of more than 5 subjects are not available), and then the samples are subjected to the relevant test again. During the test, the subjects had to use no wash, hair care and styling products (gels, sprays, etc.) other than the test product; and the hair length, the hairstyle and the color (namely, the hair must not be permed or dyed) must not be changed in the test process. The test results are shown in the following table 2, wherein the actual valid data of the samples S-3 'and S-6' are less than 65%, the use experience of the samples cannot be accurately reflected, and thus no record is recorded.

Claims (10)
1. The anti-hair loss and hair-growing composition is characterized by comprising the following components in percentage by weight: a) at least 0.05 wt% of a pyrrolidinyldiaminopyrimidine; (b) at least 0.05 wt% adenosine; (c) at least 0.05 wt% oligopeptide; (d) at least 1.0 wt% lecithin; (e) at least 5 wt% of a polyglyceryl fatty acid ester; and the balance of a dispersing medium; wherein the lecithin is egg yolk lecithin; the saponification value of the egg yolk lecithin is 195-212 mgKOH/g.
2. The hair loss prevention and hair growth promotion composition according to claim 1, wherein the iodine value of the egg yolk lecithin is 65-73 gI/100 g.
3. The hair loss prevention and hair growth promotion composition of claim 2, wherein the phosphatidylcholine content in the egg yolk lecithin is not less than 75 wt%.
4. The hair loss prevention and hair growth promotion composition according to claim 1, wherein the oligopeptide has a molecular weight of 100 to 1000 daltons.
5. The anti-hair loss and hair growth composition according to claim 4, wherein the oligopeptide comprises acetyl tetrapeptide-3.
6. The hair loss preventing and growing composition according to any one of claims 1 to 5, wherein the polyglycerin fatty acid ester comprises decapolyglycerin monolaurate and triglycerol diisostearate; preferably, the triglycerol diisostearate accounts for at least 35 wt% of the polyglyceryl fatty acid ester.
7. The hair loss preventing and hair growth promoting composition according to any one of claims 1 to 5, wherein the content of the pyrrolidinyl diaminopyrimidine is 1.2 to 3.5 wt%.
8. The anti-hair loss and hair growth composition according to claim 7, further comprising at least 0.1 wt% panthenol and 0.1 wt% cholesterol.
9. A hair washing and conditioning preparation characterized by comprising the hair loss preventing and hair growing composition according to any one of claims 1 to 8; the hair loss preventing and hair growing composition accounts for at least 5 wt% of the shampoo preparation.
10. The hair care shampoo preparation according to claim 9, which is in a dosage form comprising a liquid dosage form and a cream.
