背景技术
动物类中药源于动物的整体、器官或组织、病理产物、排泄物或分泌物等,由于其大多含有蛋白质、脂肪酸、氧化三甲胺类成分,与植物类中药相比,一方面更容易发生霉变、虫蛀、变色等变质现象,另一方面由于贮存过程中蛋白质、脂肪酸等易发生分解,产生具有不良气味的生物胺、小分子醛、酮、醇、三甲胺、氨等物质,散发出腥臭气味,让患者在接触或服用过程中出现恶心、呕吐等不良反应。
目前动物类中药的贮存技术主要有低温贮存、中药同贮对抗养护及气调养护等方式。低温贮存的成本高,且存在出冷库后变色、变质加快的问题;传统的中药同贮对抗养护虽然成本较低,操作简便,但效果不佳,虫蛀发霉风险较高,难以在大规模储存中使用;气调养护的核心是降底空气中的氧气含量,杀灭害虫、减少部分蛋白质及脂肪酸的氧化分解,从而抑制虫蛀及产臭。但减低氧气含量只能对动物类中药表面的需氧菌进行抑制,无法阻碍厌氧菌及兼性厌氧菌的生长,而这类细菌往往存在动物中药的深层,难以杀灭,因此气调养护难以抑制或减缓动物类中药的腥臭味发生。
因此,研究开发一种用于动物类中药具降氧除湿祛臭协同作用的气调养护剂,对减少动物类中药因贮存过程中发生的霉变、虫蛀、变色、产臭等变质现象,具有重大意义。
发明内容
鉴于此,本发明的首要目的在于提供一种用于动物类中药的气调养护剂,该气调养护剂具有降氧除湿祛臭协同作用,可以抑制动物类中药贮存过程发生虫蛀、霉变、变色及产臭等变质现象。
本发明实现上述目的的技术方案是:
一种用于动物类中药的气调养护剂,包括以下重量份的原料:还原铁粉100-400份、氯化钙10-25份、硅藻土颗粒吸附剂5-300份、肉桂精油1-50份、花椒精油1-50份,吴茱萸精油1-50份。
作为优选,所述的用于动物类中药的气调养护剂,各原料药重量配比为:还原铁粉150-300份、氯化钙15-20份、硅藻土颗粒吸附剂10-150份、肉桂精油5-25份、花椒精油5-25份,吴茱萸精油5-25份。
作为优选,所述硅藻土颗粒吸附剂为通过10目筛但不通过80目筛的颗粒,选择该粒径范围内的硅藻土颗粒吸附剂的吸附率较好且不会由于粉末过细吸附精油后出现结团现象。
本发明所述的肉桂精油、花椒精油和吴茱萸精油可通过常规的水蒸气蒸馏技术从肉桂、花椒和吴茱萸提取制备得到,也可以通过市购获得。
本发明基于现有贮存技术进行创新,创新主要体现在两个方面,一是协同物理化学的降氧除湿与中药精油的抗菌及抗氧化活性,以强化抑制发霉、虫蛀及氧化的作用;二是借助中药精油的空气矫味作用,防止动物类中药在贮藏过程中产生异味对操作人员带来的不良感觉。
本发明气调养护剂中的还原性铁粉是一种主要含单质铁灰色或黑色粉末,可与空气中氧气发生化学反应达到降氧的目的;氯化钙是一种白色多孔、具有较强吸潮能力的强电解质盐类,可与空气中的水发生化学反应达到除湿的作用;通过肉桂精油、花椒精油及吴茱萸精油进行复合配比,结合三者良好的驱虫、抗菌作用以及特殊香气,达到杀虫抑菌除臭的作用。
花椒和吴茱萸是传统的中药同贮药物,其常用于部分动物类中药的保存贮藏。本发明将花椒和吴茱萸这两种具有特殊气味的中药以精油的形式加入本发明的气调养护剂,既吸收了传统不同品种中药同贮对抗养护在抑制中药发霉虫蛀的精华,又利用现代提取技术解决了传统贮存的操作不方便及特殊香气的浓度及稳定性不好控制的问题。
在抑制或矫正动物药腥臭味方面,本发明气调养护剂中的肉桂精油含有除臭活性成分(主要包括烯键、醛、酮及环氧烷烃基团),可以和臭气发生加成、缩合及中和反应生产挥发性低的无臭物质,从而实现祛臭。

(1)按照重量份分别称取还原铁粉、氯化钙、硅藻土颗粒吸附剂、肉桂精油、花椒精油和吴茱萸精油,待用;
(2)将肉桂精油、花椒精油和吴茱萸精油混匀后加入硅藻土颗粒吸附剂吸收精油;
(3)将步骤(2)吸收了精油的硅藻土颗粒吸附剂与还原铁粉和氯化钙混合混匀,装入无纺布袋中,封口密封,得到一种用于动物类中药的气调养护剂。
本发明还提供了上述的气调养护剂在动物类中药贮藏中的应用。本发明围绕动物药贮存过程防发霉、虫蛀、变色及抑制或矫正其腥臭味的核心诉求,开发了一种用于动物类中药的气调养护剂。可通过本发明的气调养护剂的成分比例调整及用量变化和外环境的控温实现协同养护动物类中药。
本发明与现有贮存技术相比,具有如下优点:
(1)协同物理化学的降氧除湿与中药精油的抗菌及抗氧化活性,以强化抑制发霉、虫蛀及氧化的作用;
(2)借助肉桂、吴茱萸、花椒精油的祛臭功能来抑制或矫正动物药腥臭味,减少贮藏环境的空气臭味,避免给操作人员带来的不良感觉;
(3)吸收了传统不同品种中药同贮对抗养护在抑制中药发霉虫蛀的精华,又利用现代提取技术解决了传统贮存的操作不方便及特殊香气的浓度及稳定性不好控制的问题;
(4)采用具有质轻、多孔、吸附性强特点的硅藻土颗粒吸附剂吸附肉桂、花椒及吴茱萸精油,起到增加精油挥发的表面积和稳定持续挥发精油的作用。
具体实施方式
下面通过具体实施方式来进一步说明本发明,以下实施例为本发明具体的实施方式,但本发明的实施方式并不受下述实施例的限制。
本发明实施例及对比例所采用的的原料均来源于市购。
硅藻土颗粒吸附剂:选用通过10目筛但不通过80目筛的硅藻土颗粒。
实施例1:
取1g肉桂精油、1g花椒精油和1g吴茱萸精油,混匀得到混合精油,加入6g硅藻土颗粒吸附剂吸收混合精油,再加入240g还原铁粉和15g氯化钙,混合均匀,装入无纺布袋中(装2袋),封口密封即得气调养护剂1。
实施例2:
取5g肉桂精油、5g花椒精油和5g吴茱萸精油,混匀得到混合精油,加入30g硅藻土颗粒吸附剂吸收混合精油,再加入240g还原铁粉和15g氯化钙,混合均匀,装入无纺布袋中(装2袋),封口密封即得气调养护剂2。
实施例3:
取25g肉桂精油、25g花椒精油和25g吴茱萸精油,混匀得到混合精油,加入150g硅藻土颗粒吸附剂吸收混合精油,再加入240g还原铁粉和15g氯化钙,混合均匀,装入无纺布袋中(装2袋),封口密封即得气调养护剂3。
实施例4:
取50g肉桂精油、50g花椒精油和50g吴茱萸精油,混匀得到混合精油,加入300g硅藻土颗粒吸附剂吸收混合精油,再加入240g还原铁粉和15g氯化钙,混合均匀,装入无纺布袋中(装2袋),封口密封即得气调养护剂4。
对比例1:
取5g肉桂精油,加入10g硅藻土颗粒吸附剂吸收精油,再加入240g还原铁粉和15g氯化钙,混合均匀,装入无纺布袋中(装2袋),封口密封即得气调养护对照剂1。
对比例2:
取5g花椒精油和5g吴茱萸精油,混匀得到混合精油,加入20g硅藻土颗粒吸附剂吸收精油,再加入240g还原铁粉和15g氯化钙,混合均匀,装入无纺布袋中(装2袋),封口密封即得气调养护对照剂2。
对比例3:
取5g肉桂精油和5g花椒精油,加入20g硅藻土颗粒吸附剂吸收精油,再加入240g还原铁粉和15g氯化钙,混合均匀,装入无纺布袋中(装2袋),封口密封即得气调养护对照剂3。
对比例4:
取5g肉桂精油和5g吴茱萸精油,加入20g硅藻土颗粒吸附剂吸收精油,再加入240g还原铁粉和15g氯化钙,混合均匀,装入无纺布袋中(装2袋),封口密封即得气调养护对照剂4。
下面将通过精油用量优化实验、协同祛臭对比实验、降氧除湿实验和抑菌实验对本发明精油的用量及协同作用作进一步的说明和证明本发明具有的技术效果。
一、精油用量优化实验
1实验材料及设备
本发明气调养护剂1-4;储物箱(86cm*63cm*48cm);蜈蚣(广州至信药业股份有限公司);带盖塑料瓶。
2实验方法
取五个储物箱,分为空白对照组和实验1-4组(精油组成情况见表1)。分别均匀摊开放入2kg的蜈蚣(有特殊刺鼻臭气的动物药),空白对照组不放入气调养护剂,实验1-4组分别在储物箱里蜈蚣对角线处各放入1袋相应的气调养护剂1-4。实验组与空白对照组置于25℃恒温环境中,放置30天后,按下述方法进行气味评价:空白对照组与实验1-4组各取3份蜈蚣(一份10条)置于干净无异味的瓶子里,密封置于25℃恒温环境放置24小时后按表2臭气评价等级标准进行盲试。对比5名人员对空白对照组和实验1-4组的气味评价结果,综合祛臭效果与精油使用量优选本发明气调养护剂精油的使用量。

3实验结果
如表3所示,与空白对照组比较,实验1-4组均能降低蜈蚣的特殊刺鼻臭气,差异均有统计学意义。随着精油用量的增加,祛臭效果相应增强。其中,实验1组对臭气刺鼻的缓解程度较小;实验4组的祛臭效果好但由于精油使用量较大,带有较浓的精油味道;实验2组的祛臭效果较好;实验3组的祛臭效果较好,但跟实验2组相比,祛臭效果差异不是很大。在本实验中,综合祛臭效果与精油使用量,实验2组的配比较优。此外,本发明气调养护剂精油的配比可根据具体动物药的情况进行调整。

二、协同祛臭对比实验
1实验材料及设备
本发明气调养护剂2,气调养护对照剂1-4;储物箱(86cm*63cm*48cm);蜈蚣(广州至信药业股份有限公司);带盖塑料瓶。
2实验方法
取五个储物箱,分为参照组和实验对比1-4组(精油组成情况见表4)。分别均匀摊开放入2kg的蜈蚣(有特殊刺鼻臭气的动物药)。参照组在储物箱里蜈蚣对角线处各放入1袋气调养护剂2;实验对比1-4组同法放入相应的气调养护对照剂1-4。参照组与实验对比1-4组置于25℃恒温环境中,放置30天后,按下述方法进行气味评价:参照组与实验对比1-4组各取3份蜈蚣(一份10条)置于干净无异味的瓶子里,密封置于25℃恒温环境放置24小时后按表2臭气评价等级标准进行盲试。对比5名人员对参照组与实验对比1-4组的气味评价结果,以此评价本发明气调养护剂协同祛臭的效果。

3实验结果
如表5所示,与参照组比较,实验对比1组的结果差异具有统计学意义,说明花椒精油和吴茱萸精油则发挥了协同祛臭作用;与参照组比较,实验对比2组的结果差异具有统计学意义,且结果差异较大,说明肉桂精油在祛臭效果上发挥了关键作用。这说明本发明气调养护剂的三种精油通过协同作用发挥祛臭效果。

三、降氧除湿实验
1实验材料及设备
本发明气调养护剂2;储物箱(86cm*63cm*48cm);温湿度计;氧气检测仪(希玛AR8100)。
2实验方法
取两个储物箱,分为空白对照组和实验组。将储物箱置于25℃恒温环境中,放入温湿度计,储物箱中预留一探测孔用于测量氧气含量,测量时打开,平时密封。待温湿度相对稳定后,实验组放入2袋本发明气调养护剂2,空白对照组不放气调养护剂。分别于记录0天,1天,3天,5天,10天,20天,30天,记录箱内温湿度,打开探测孔,测量氧气含量。对比空白对照组和实验组的氧气含量及湿度变化数据,以此评价本发明气调养护剂的降氧除湿效果。
3实验结果
3.1氧气降低效果
如表6所示,与空白对照组比较,本发明气调养护剂能显著降低密闭空间的氧含量,说明本发明气调养护剂具有明显的降氧效果。

3.2除湿效果
如表7所示,与空白对照组比较,本发明气调养护剂能显著降低密闭空间的湿度,说明本发明气调养护剂具有明显的除湿效果。

四、抑菌实验
1实验材料
本发明气调养护剂2;储物箱(86cm*63cm*48cm);电热恒温培养箱(XY-DR-600型,上海昕仪仪器仪表有限公司);75%乙醇消毒液(重庆市普康消毒用品有限公司);营养琼脂即用型无菌培养基(90mm,广东环凯微生物科技有限公司)。
2实验方法
将两个储物箱分为空白对照组和实验组,用75%乙醇消毒液对储物箱内表面进行消毒。置于自然通风处暴露2小时后,实验组加入2袋本发明的气调养护剂2,空白对照组不加。密闭储物箱72小时后,在储物箱四个角落及中央处放上营养琼脂即用型无菌培养基。继续密闭储物箱24小时后打开箱子,培养基加盖取出,置于37℃恒温培养箱培养24小时,计算菌落数。比较试验组与空白对照组空气菌落差异,以气调养护剂对空气中菌落数的影响评价其抑菌效果。
3实验结果
如表8所示,与空白对照组比较,本发明气调养护剂能显著抑制空气中的微生物的生存,差异有统计学意义(P<0.01),说明本发明气调养护剂具有明显的抑菌效果。

Claims (5)
1.一种用于动物类中药的气调养护剂,其特征在于,包括以下重量份的原料:还原铁粉100-400份、氯化钙10-25份、硅藻土颗粒吸附剂5-300份、肉桂精油1-50份、花椒精油1-50份,吴茱萸精油1-50份。
2.根据权利要求1所述的用于动物类中药的气调养护剂,其特征在于,各原料药重量配比为:还原铁粉150-300份、氯化钙15-20份、硅藻土颗粒吸附剂10-150份、肉桂精油5-25份、花椒精油5-25份,吴茱萸精油5-25份。
3.根据权利要求1或2所述的用于动物类中药的气调养护剂,其特征在于,所述硅藻土颗粒吸附剂为通过10目筛但不通过80目筛的颗粒。
4.权利要求1-3任一项所述的用于动物类中药的气调养护剂的制备方法,其特征在于,包括如下步骤:
(1)按照重量份分别称取还原铁粉、氯化钙、硅藻土颗粒吸附剂、肉桂精油、花椒精油和吴茱萸精油,待用;
(2)将肉桂精油、花椒精油和吴茱萸精油混匀后加入硅藻土颗粒吸附剂吸收精油;
(3)将步骤(2)吸收了精油的硅藻土颗粒吸附剂与还原铁粉和氯化钙混合混匀,装入无纺布袋中,封口密封,得到一种用于动物类中药的气调养护剂。
5.根据权利要求1-3任一项所述的气调养护剂在动物类中药贮藏中的应用。
