The invention discovers that the asparagus extract can obviously improve the transdermal delivery rate of the oily purslane extract containing the unsaturated fatty acid active ingredient, thereby improving the application prospect and value of the oily purslane extract in the fields of cosmetics and medicines by percutaneous absorption.
The invention provides a method for improving transdermal delivery rate of an oily purslane extract, which is characterized in that the oily purslane extract is mixed with an asparagus extract or a solution containing the asparagus extract.
The invention further provides a composition containing the purslane extract, which is characterized by comprising the oily purslane extract and the asparagus extract, and the composition has an improved transdermal delivery rate of the oily purslane extract.
The oily purslane extract is prepared by lipophilic solvents such as: diethyl ether, ethyl acetate, propane, butane fluids and supercritical CO 2 Fluid extraction is carried out.
The asparagus extract is alcohol or water or alcohol/water extract.
The asparagus extract is present in the form of a solution.
The concentration of the asparagus extract solution is 10-20 mg/ml.
The present invention advantageously and surprisingly eliminates or reduces the disadvantages of poor transdermal delivery of existing oily purslane extracts. In particular, an alcohol or water or alcohol/water extract of asparagus is used as a dispersion medium to enhance transdermal delivery of the oily purslane extract and to enhance transdermal delivery of a composition comprising a dispersion of the oily purslane extract. The dispersion medium of the present invention is formed of natural products and can even be edible.
Detailed Description
The invention is further illustrated by the following examples, which are not intended to limit the scope of the invention. The experimental methods without specifying specific conditions in the following examples were selected according to the conventional methods and conditions, or according to the commercial instructions.
Example 1
Preparation method of oily herba Portulacae extract containing unsaturated fatty acid
Fresh purslane and drinking water are used as main raw materials, the fresh purslane is selected, cleaned and cut up, water is added according to the weight ratio of 1:3 for soaking for 12 hours, the purslane is decocted for 6 hours at 80 ℃, and filtration is carried out to obtain purslane residues.
Extracting purslane residues by using a subcritical isobutane extraction technology, extracting an oily purslane extract: the subcritical fluid extraction process is set under the conditions that the extraction pressure is 0.8MPa, the extraction temperature is 40 ℃, extraction is carried out for 150min, purslane oil-immersed paste is obtained, after three times of extraction, the purslane oil-immersed paste obtained each time is combined, and the crude oil purslane extract is prepared after centrifugation at 4000r/min for 15min and filtration.
Mixing the crude oil purslane extract with 0.5mol/L KOH-ethanol solution according to the weight ratio of 1: 20, mixing, then placing in a water bath at 75 ℃, stirring and refluxing for 120min to obtain a saponified solution; adding ice distilled water equivalent to 0.5mol/L KOH-ethanol solution for quick cooling, extracting saponified substance with petroleum ether equivalent to 0.5mol/L KOH-ethanol solution for multiple times, taking a water layer, adding water for dilution twice, slowly adding 10% HCl, adjusting pH to 2-3, and immediately extracting mixed fatty acid with petroleum ether equivalent to 0.5mol/L KOH-ethanol solution for multiple times to obtain a petroleum ether layer through a separating funnel; washing the petroleum ether layer with 5% NaCl solution to neutral, drying with anhydrous sodium sulfate, filtering, and rotary evaporating to obtain purified oily herba Portulacae extract with light yellow color.
Example 2
Preparation method of Germinatus Phragmitis extract
(1) Drying and pulverizing Germinatus Phragmitis, sieving with 10-40 mesh sieve, adding 70% ethanol at solid/liquid ratio of 1: 5-1: 20(W/V), and ultrasonic treating at power of 20-800W/cm 2 Extracting at 20-60 deg.C for 40 min, performing liquid-solid separation to obtain extractive solution of Germinatus Phragmitis, and distilling the extractive solution under reduced pressure to recover ethanol;
(2) adding water saturated n-butanol into the concentrated solution obtained in the step (1) according to the volume ratio of 1: 0.5 for extraction, repeating the extraction process for 2-3 times according to the same method, combining the extraction solutions, and recovering the extractant under reduced pressure to obtain a purified asparagus extract;
(3) preparing the extract obtained in the step (2) into aqueous solution according to the concentration of 10mg/ml, 15mg/ml and 20mg/ml respectively.
Examples 3 to 6
Preparation of purslane essential oil composition
Mixing the components according to the proportion shown in the table 1 according to the weight percentage, heating to 60-70 ℃, and uniformly mixing to obtain the purslane essential oil composition:

Example 7
Transdermal delivery assay
Calculating the diffusion rate of different oily purslane extract solutions through a diffusion cell simulating human skin, and specifically comprises the following steps:
testing in an in vitro Franz diffusion cell with healthy male Wistar rat dorsal skin as a permeation barrier;
fixing the skin between the receiving tank and the supply tank (the inner layer of the skin faces the receiving tank) to ensure that no air bubbles exist between the skin and the receiving liquid;
diffusion cell parameters: effective diffusion area of about 3.14cm 2 The volume of the receiving tank was about 8.5mL, the magnetic stirring speed was 600rpm, and the temperature of the receiving solution was 37.0. + -. 0.1 ℃.
Filling a receiving pool with 1% SDS-10% ethanol-physiological saline as a release medium, removing bubbles, starting stirring, keeping the temperature to (37.0 +/-0.1) DEG C, uniformly coating about 1g of sample on the surface of skin, sucking 0.5mL of receiving solution after 1, 2, 4, 6, 8, 10 and 12 hours, and immediately adding the release medium with the same volume and temperature after sampling. The concentration of purslane extract in the receiving solution filtered through a 0.22 μm filter was determined by HPLC. And calculating the cumulative permeation amount of the medicaments at different times.
The cumulative transdermal quantity per unit area of the purslane extract is calculated according to the following formula:
in the above formula: q: accumulating transdermal volume; s: an effective diffusion area; v: receiving the volume of the normal saline in the pool; c i : the concentration of the drug in the receiving solution from the 1 st to the last sampling; c n : the concentration of the drug in the receiving fluid during the sub-sampling; m is 0 : theoretically weighing the sample; m: the sample is actually weighed.

From the experimental results shown in table 2, the transdermal permeation amount of the essential oil of the purslane extract in example 3 is small, and the transdermal permeation amount of the purslane extract is remarkably increased by using the asparagus extract solution as a dispersing agent in examples 4-5, and the increase amount is in positive correlation with the concentration of the asparagus extract solution.
Claims (6)
1. A composition of purslane extract, comprising an oily purslane extract and an asparagus extract.
2. The composition of claim 1, wherein the oily purslane extract is obtained by extraction with a lipophilic solvent.
3. The composition of claim 1, wherein the asparagus extract is an alcoholic or aqueous or alcoholic/aqueous extract.
4. The composition of claim 1, wherein the asparagus extract is present as a solution.
5. The composition of claim 4, wherein the concentration of the asparagus extract solution is 10-20 mg/ml.
6. A method for improving the transdermal delivery rate of an oily purslane extract is characterized in that the oily purslane extract is mixed with an asparagus extract or a solution containing the asparagus extract.
