本发明属于敷料材料制备技术领域,具体涉及一种可吸收性止血敷料及其制备方法。
背景技术
皮肤创伤的救治和护理,如果症状较轻,可以采用“创口贴”贴敷即可,还可以使用康复新液或者云南白药等,康复新液适用于外伤、溃疡、瘘管等创面,可直接将纱布浸透,覆盖皮肤创伤,如果创面较大,则需要清创后再使用。云南白药适用于刀伤、枪伤及跌打损伤的止血、消肿,可直接用于患处。上述各种形式的止血敷料存在止血时间长、容易感染细菌发炎、伤口收组织液浸泡时间过长肿胀、止血敷料材料不易吸收、造成伤口愈合后伤疤较大等问题。
在民间,常见渔民采用鳝鱼血涂敷于纸上,晾干,用于治疗外创伤,具有快速止血的功效,但采用鳝鱼血虽然能快速止血,但腥气味很重,干鳝鱼血组分不易被人体吸收,堵塞于伤口处,造成伤疤面积增大。鳝鱼血必须彻底晾干,新鲜鳝鱼血具有活血化瘀的功效,晾干不彻底会起到反向效果。
土箭芪,中药名。为瑞香科荛花属植物长花荛花的根,长花荛花,分布于四川及云南,具有宽中理气,补脾益胃之功效,主治脘腹胀痛,食少便溏,虚肿不消。
目前,尚没有将鳝鱼血提取物与土箭芪提取物复合制备止血敷料的相关报道。
发明内容
本发明的目的在于提供一种可吸收性止血敷料及其制备方法。
一种可吸收性止血敷料,包括如下重量份数的原料组分:羟乙基甲壳素50-100份、骨胶原30-80份、鳝鱼血提取物3-10份、土箭芪提取物3-10份。
所述鳝鱼血提取物的制备方法为:
(1)取新鲜鳝鱼血烘干,磨成粉末;
(2)加3-6倍重量份数的40-80%的乙醇水溶液回流提取3次,合并滤液,蒸干制成。
所述土箭芪提取物的制备方法为:
(1)取土箭芪的根烘干,磨成粉末;
(2)加3-6倍重量份数的40-80%的乙醇水溶液回流提取3次,合并滤液,蒸干制成。
所述可吸收性止血敷料的制备方法,按照如下步骤进行:
(1)按照重量份数,取羟乙基甲壳素50-100份、骨胶原30-80份、鳝鱼血提取物3-10份、土箭芪提取物3-10份,搅拌条件下混合均匀;
(2)将步骤(1)制备的原料混合物在真空条件下,控制温度80-150℃进行加热发泡处理1-3h;
(3)将步骤(2)制备的物料,置于室温下冷却,得到止血棉片;
(4)将步骤(3)制备的止血棉片用蒸馏水进行清洗,加工成所需形状后干燥、灭菌,即得。
步骤(1)所述搅拌的速率为400-1000rpm,搅拌时间为10-30min。
步骤(2)所述述发泡处理为100℃的真空下,加热1.5h。
所述灭菌为121℃高温瞬时灭菌15s。
所述灭菌为在γ射线下均匀照射36-72h。
本发明的有益效果:本发明制备的止血敷料,可吸收性好,止血后能有效避免伤口肿胀、感染,治愈后留疤痕小;鳝鱼血提取物和土箭芪提取物的组合使用提高了止血时间,组合使用协同增效。

所述土箭芪提取物的制备方法为:
(1)取土箭芪的根烘干,磨成粉末;
(2)加4倍重量份数的50%的乙醇水溶液回流提取3次,合并滤液,蒸干制成。
所述可吸收性止血敷料的制备方法,按照如下步骤进行:
(1)按照重量份数,取羟乙基甲壳素80份、骨胶原50份、鳝鱼血提取物8份、土箭芪提取物8份,搅拌条件下混合均匀;所述搅拌的速率为600rpm,搅拌时间为20min;
(2)将步骤(1)制备的原料混合物在真空条件下,控制温度120℃进行加热发泡处理2h;
(3)将步骤(2)制备的物料,置于室温下冷却,得到止血棉片;
(4)将步骤(3)制备的止血棉片用蒸馏水进行清洗,加工成所需形状后干燥、灭菌,即得;所述灭菌为121℃高温瞬时灭菌15s。

所述土箭芪提取物的制备方法为:

一种可吸收性止血敷料,包括如下重量份数的原料组分:羟乙基甲壳素100份、骨胶原80份、鳝鱼血提取物10份、土箭芪提取物10份。
所述鳝鱼血提取物的制备方法为:
(1)取新鲜鳝鱼血烘干,磨成粉末;
(2)加6倍重量份数的70%的乙醇水溶液回流提取3次,合并滤液,蒸干制成。
所述土箭芪提取物的制备方法为:
(1)取土箭芪的根烘干,磨成粉末;
(2)加6倍重量份数的70%的乙醇水溶液回流提取3次,合并滤液,蒸干制成。
所述可吸收性止血敷料的制备方法,按照如下步骤进行:
(1)按照重量份数,取羟乙基甲壳素100份、骨胶原80份、鳝鱼血提取物10份、土箭芪提取物10份,搅拌条件下混合均匀;所述搅拌的速率为1000rpm,搅拌时间为10min;
(2)将步骤(1)制备的原料混合物在真空条件下,控制温度140℃进行加热发泡处理1h;
(3)将步骤(2)制备的物料,置于室温下冷却,得到止血棉片;
(4)将步骤(3)制备的止血棉片用蒸馏水进行清洗,加工成所需形状后干燥、灭菌,即得;所述灭菌为在γ射线下均匀照射48h。
对比例1
一种可吸收性止血敷料,包括如下重量份数的原料组分:羟乙基甲壳素80份、骨胶原50份、鳝鱼血提取物16份。
所述鳝鱼血提取物的制备方法为:
(1)取新鲜鳝鱼血烘干,磨成粉末;
(2)加4倍重量份数的60%的乙醇水溶液回流提取3次,合并滤液,蒸干制成。
所述可吸收性止血敷料的制备方法,按照如下步骤进行:
(1)按照重量份数,取羟乙基甲壳素80份、骨胶原50份、鳝鱼血提取物16份,搅拌条件下混合均匀;所述搅拌的速率为600rpm,搅拌时间为20min;
(2)将步骤(1)制备的原料混合物在真空条件下,控制温度120℃进行加热发泡处理2h;
(3)将步骤(2)制备的物料,置于室温下冷却,得到止血棉片;
(4)将步骤(3)制备的止血棉片用蒸馏水进行清洗,加工成所需形状后干燥、灭菌,即得;所述灭菌为121℃高温瞬时灭菌15s。
对比例2
一种可吸收性止血敷料,包括如下重量份数的原料组分:羟乙基甲壳素80份、骨胶原50份、土箭芪提取物16份。
所述土箭芪提取物的制备方法为:
(1)取土箭芪的根烘干,磨成粉末;
(2)加4倍重量份数的50%的乙醇水溶液回流提取3次,合并滤液,蒸干制成。
所述可吸收性止血敷料的制备方法,按照如下步骤进行:
(1)按照重量份数,取羟乙基甲壳素80份、骨胶原50份、土箭芪提取物16份,搅拌条件下混合均匀;所述搅拌的速率为600rpm,搅拌时间为20min;
(2)将步骤(1)制备的原料混合物在真空条件下,控制温度120℃进行加热发泡处理2h;
(3)将步骤(2)制备的物料,置于室温下冷却,得到止血棉片;
(4)将步骤(3)制备的止血棉片用蒸馏水进行清洗,加工成所需形状后干燥、灭菌,即得;所述灭菌为121℃高温瞬时灭菌15s。
实验例:
选取150只体重为6.0-6.3kg的比利时兔,随机分为5组,每组8只,分别标记为实施例1-5组,每只兔切断左耳耳缘静脉、右耳耳缘动脉的三分之二,自由出血5s,5s后分别用不同的止血敷料压敷伤口。其中,各组实验所用的止血敷料均统一裁切成长×宽×高=5cm×5cm×2cm的长方体块状,记录左耳耳缘静脉止血时间(s)、右耳耳缘动脉止血时间(s),实验结果见表1:

Claims (8)
1.一种可吸收性止血敷料,其特征在于,包括如下重量份数的原料组分:羟乙基甲壳素50-100份、骨胶原30-80份、鳝鱼血提取物3-10份、土箭芪提取物3-10份。
2.根据权利要求1所述可吸收性止血敷料,其特征在于,所述鳝鱼血提取物的制备方法为:
(1)取新鲜鳝鱼血烘干,磨成粉末;
(2)加3-6倍重量份数的40-80%的乙醇水溶液回流提取3次,合并滤液,蒸干制成。
3.根据权利要求1所述可吸收性止血敷料,其特征在于,所述土箭芪提取物的制备方法为:
(1)取土箭芪的根烘干,磨成粉末;
(2)加3-6倍重量份数的40-80%的乙醇水溶液回流提取3次,合并滤液,蒸干制成。
4.权利要求1所述可吸收性止血敷料的制备方法,其特征在于,按照如下步骤进行:
(1)按照重量份数,取羟乙基甲壳素50-100份、骨胶原30-80份、鳝鱼血提取物3-10份、土箭芪提取物3-10份,搅拌条件下混合均匀;
(2)将步骤(1)制备的原料混合物在真空条件下,控制温度80-150℃进行加热发泡处理1-3h;
(3)将步骤(2)制备的物料,置于室温下冷却,得到止血棉片;
(4)将步骤(3)制备的止血棉片用蒸馏水进行清洗,加工成所需形状后干燥、灭菌,即得。
5.根据权利要求4所述可吸收性止血敷料的制备方法,其特征在于,步骤(1)所述搅拌的速率为400-1000rpm,搅拌时间为10-30min。
6.根据权利要求4所述可吸收性止血敷料的制备方法,其特征在于,步骤(2)所述述发泡处理为100℃的真空下,加热1.5h。
7.根据权利要求4所述可吸收性止血敷料的制备方法,其特征在于,所述灭菌为121℃高温瞬时灭菌15s。
8.根据权利要求4所述可吸收性止血敷料的制备方法,其特征在于,所述灭菌为在γ射线下均匀照射36-72h。
