Abstract
The invention discloses a Chinese herbal medicine additive for preventing and treating bacterial rotten body disease of alepidote fish and application thereof, wherein the Chinese herbal medicine additive comprises the following components in parts by weight: 0.1-1.5 parts of Chinese herbal medicine mixture, 0.2-1.0 part of organic acid and 0.03-0.1 part of vitamin K; wherein the Chinese herbal medicine mixture comprises the following components in parts by weight: 20-25 parts of dandelion, 20-25 parts of wormwood, 15-20 parts of houttuynia cordata, 15-20 parts of honeysuckle and 15-20 parts of cinnamon. The Chinese herbal medicine additive provided by the invention is especially suitable for preventing and treating bacterial body rot of fish without scales, and is especially suitable for freshwater fishes without scales, such as channel catfish, yellow catfish, clarias fuscus and the like. Can reduce the use of antibiotics, improve the antibacterial and bactericidal performance, improve the immunity of the fry and promote the growth of the fry.
Description
Technical Field
The invention belongs to the technical field of aquaculture, and particularly relates to a Chinese herbal medicine additive for preventing bacterial canker of alepidote fish and application thereof.
Background
The non-scale fish refers to a fish species without scales or with very small scales, most of the non-scale fish in the sea live in deep sea with the depth of more than 500 meters, and mainly refers to fish species of eel order, such as eel, eel and the like; the freshwater fish without scales includes loach, finless eel, catfish, whitebait, etc.
The conventional species of aquaculture comprises pelteobagrus fulvidraco, namely yellow diced fish, angci fish and the like, and is a common freshwater fish of the genera of the families of the bagrus and the pelteobagrus fulvidraco. The yellow catfish is rich in amino acids, tender in meat quality, delicious in taste, high in nutritive value, and has nourishing effect and medicinal value, and has no muscle thorns.
The catfish, also called the beard fish and the clarias fuscus, is characterized in that the whole body has no scales, the body surface has more mucus, the head is flat and wide, the upper jaw and the lower jaw have 4 beards respectively, the distribution is wide, the catfish is mainly produced in Yangtze river and Yangtze river basin, and the best season is from spring to summer (4-7 months). The catfish is a carnivorous fish, has tender and less meat, is delicious and rich in protein and fat, is rich in nutrition, and is particularly suitable for people with weak constitution and malnutrition.
The channel catfish with channel catfish and large-scale freshwater fish have high meat content, tender meat quality and delicious taste, are widely popular with consumers of various countries, are main freshwater fish breeding varieties in the United states, and are widely introduced into multi-country breeding in the world
The silurus meridionalis belongs to the order silurus, the family silurus, and the genus silurus. Has the advantages of high growth speed, high culture benefit, less diseases, low temperature resistance and the like, and is a famous fish variety suitable for popularization and culture.
These fish species are all susceptible to pathogenic bacterial infections during the artificial breeding stage, for example fish diseases caused by parasites, including trichodiniasis, ichthyophthiriasis, for example fish diseases caused by fungi, including saprolegniasis, for example fish diseases caused by bacteria, including bacterial septicaemia, bacterial gill rot and intestinal inflammation. Serious diseases such as bacterial body rot and body rot, skin ulceration of fish with body rot is clear, a square ulceration focus is often formed, and skin is completely peeled off to expose muscles when the skin is serious (the fish generally dies at this time). Some diseased fish are also accompanied by internal bleeding and visceral diseases. The 'square' rotten body has fast onset of disease and extremely high mortality rate, and the death rate develops from several jin to very high every day within a few days.
At present, inorganic acid is commonly used for detoxifying for treating bacterial canker of freshwater fish without scales, and the freshwater fish is disinfected by disinfectant water and taken orally with antibiotic for protecting liver, but the long-term use of the antibiotic can cause the destruction of intestinal flora of fish body and produce the symptom of frequent bleeding. Meanwhile, the residue of antibiotics in the fish body can directly enter the human body through eating, so that the immunity of the human body is reduced, and therefore, the problem that the aquaculture industry needs to solve is to find an additive capable of reducing or completely replacing the antibiotics.
Disclosure of Invention
The invention aims to provide a Chinese herbal medicine additive for preventing and treating bacterial canker of fish without scales and application thereof, and the Chinese herbal medicine additive is particularly suitable for freshwater fishes without scales, such as channel catfish, pelteobagrus fulvidraco, clarias fuscus and the like, can reduce the use of antibiotics in the breeding process, improve the immunity of the fishes, and has the functions of strongly inhibiting bacteria and killing bacteria.
In order to achieve the purpose, the invention provides a Chinese herbal medicine additive for preventing and treating bacterial rot of alepidote fish, which comprises the following components in parts by weight: 0.1-1.5 parts of Chinese herbal medicine mixture, 0.2-1.0 part of organic acid and 0.03-0.1 part of vitamin K, wherein the Chinese herbal medicine mixture comprises the following components in parts by weight: 20-25 parts of dandelion, 20-25 parts of wormwood, 15-20 parts of houttuynia cordata, 15-20 parts of honeysuckle and 15-20 parts of cinnamon;
every 100 parts of organic acid comprises 70-80 parts of formic acid, 5-10 parts of malic acid, 5-10 parts of butyric acid and 5-10 parts of other organic acid.
Further, the Chinese herbal medicine mixture is prepared by the following method:
weighing corresponding Chinese herbal medicines according to a proportion, mixing, crushing, and sieving with a 100-mesh sieve to obtain the traditional Chinese medicine.
Further, the application of the Chinese herbal medicine additive for preventing and treating bacterial rotten body disease of the alepidote fishes is used for mixing and feeding the materials in the fish breeding period, the fattening period and the disease development period.
Furthermore, the weight standard of the fish seeds in the fish seed culture period is 150 kg/kg-5 kg/kg, and the weight standard of the fish seeds in the fattening period is more than 100g per tail.
Further, the feeding standard of the fish species in the culture period is as follows: 100 parts of fish feed, 0.1-0.3 part of Chinese herbal medicine mixture, 0.2-0.4 part of organic acid and 0.03-0.05 part of vitamin K.
Further, the feeding standard of the fattening period is as follows: 100 parts of the bred feed, 0.3-0.5 part of the Chinese herbal medicine mixture and 0.3-0.5 part of the organic acid, 0.05-0.1 part of the vitamin K.
Further, the feeding standard in the disease period is as follows: 100 parts of fish feed or breeding feed, 1-1.5 parts of Chinese herbal medicine mixture, 0.5-1.0 part of organic acid and 0.1 part of vitamin K, and continuously feeding for 5-7 days.
Further, the fish pond is required to be disinfected in the disease period, and the method comprises the following steps: the whole pond is sprayed after mixing and diluting enrofloxacin and vitamin K, and the bottom is changed by adopting potassium hydrogen persulfate every other day after the whole pond is disinfected by povidone iodine solution the next day.

dandelion is bitter, sweet and cold. It enters liver and stomach meridians. Has the effects of clearing away heat and toxic materials, relieving swelling, dispersing pathogenic accumulation, inducing diuresis, and treating stranguria. It can be used for treating furuncle, swelling, acute mastitis, scrofula, conjunctival congestion, pharyngalgia, pulmonary abscess, intestinal abscess, jaundice due to damp-heat pathogen, and stranguria with damp-heat pathogen.
Mugwort is bitter, pungent and warm in nature and enters spleen, liver and kidney. The whole herb can be used as medicine, and has the effects of warming channels, removing dampness, dispelling cold, stopping bleeding, diminishing inflammation, relieving asthma, relieving cough, preventing miscarriage, resisting allergy, etc.
Yu xing Cao is pungent in flavor and cold in nature and enters lung meridian. Has effects in clearing away heat and toxic materials, relieving swelling, treating sore, promoting urination, removing dampness, clearing away heat, relieving dysentery, invigorating stomach, and promoting digestion, and can be used for treating lung abscess, pyocutaneous disease, hematochezia, and heat accumulation in spleen and stomach due to excess heat, heat toxin, dampness, and disease heat.
Honeysuckle flower is sweet and cold in nature. It enters lung and stomach meridians. Has the effects of clearing away heat and toxic materials, relieving inflammation, and relieving swelling, and can be used for treating affection of exogenous wind-heat or epidemic febrile disease, fever, heatstroke, toxic heat, dysentery, carbuncle, furuncle, pharyngitis, and various infectious diseases.
Rou Gui is pungent and sweet in flavor and strongly hot in nature. It enters kidney, spleen, heart and liver meridians. Has the effects of supplementing fire and supporting yang, guiding fire to the origin, dispelling cold and relieving pain, and warming and dredging channels and collaterals. Can be used for treating sexual impotence, cold womb, psychroalgia of waist and knee, asthma due to kidney deficiency, floating of deficient yang, giddiness, conjunctival congestion, psychroalgia of heart and abdomen, vomiting and diarrhea due to deficiency cold, abdominal pain due to cold hernia, dysmenorrhea, and amenorrhea.
2. The traditional Chinese medicine mixture disclosed by the invention is reasonable and scientific in formula, sweet in taste and bitter in matching, and does not cause discomfort in taste in the using process so that the scale-free freshwater fish cannot eat; the combination of the upper temperature and the upper cold of the air can not cause too hot and too cold to the scale-free freshwater fish to influence the growth of the fish body; in the aspect of medicine property, the formula is more reasonable, the cinnamon with the insecticidal function and the houttuynia, the honeysuckle and the dandelion with the antiviral function are matched, so that the formula has the effects of focus convergence hemostasis, bacteriostasis, antivirus and immunity improvement, has obvious inhibition effects on bacterial protein isolate and enzyme activity, and can prevent bacterial cell division.
3. The organic acid (malic acid, formic acid, butyric acid and the like) in the invention has the effects of inhibiting and killing gram-negative bacteria (Edwardsiella, Aeromonas hydrophila and the like) commonly seen in aquaculture, and simultaneously has the effects of improving the intestinal health of cultured fish and promoting the growth of fish bodies.
4. The additive disclosed by the invention adopts the synergistic effect of the Chinese herbal medicines and the organic acid, so that the effects of inhibiting bacteria, sterilizing, improving immunity and promoting growth are better, and the additive is one of the most effective schemes for reducing the use of antibiotics or replacing antibiotics in the aquaculture process.
5. The invention assists in sterilizing the fish pond during the treatment period, can quickly kill harmful bacteria in the culture environment, inhibits the regeneration of the harmful bacteria in a short period, reduces the secondary invasion of the bacteria to the fish body, and improves the culture survival rate.

The invention provides a Chinese herbal medicine additive for preventing and treating bacterial rotten body disease of alepidote fish, which comprises the following components in parts by weight: 0.1-1.5 parts of Chinese herbal medicine mixture, 0.2-1.0 part of organic acid and 0.03-0.1 part of vitamin K, wherein the Chinese herbal medicine mixture comprises the following components in parts by weight: 20-25 parts of dandelion, 20-25 parts of wormwood, 15-20 parts of houttuynia cordata, 15-20 parts of honeysuckle and 15-20 parts of cinnamon;
every 100 parts of organic acid comprises 70-80 parts of formic acid, 5-10 parts of malic acid, 5-10 parts of butyric acid and 5-10 parts of other organic acid.
The Chinese herbal medicine mixture is prepared by the following method: weighing corresponding Chinese herbal medicines according to a proportion, mixing, crushing, and sieving with a 100-mesh sieve to obtain the traditional Chinese medicine.
The invention also provides the application of the Chinese herbal medicine additive, namely the Chinese herbal medicine additive is used for mixing and feeding in the fish culture period, the fattening period and the disease development period.
Wherein the weight standard of the fish seeds in the fish seed culture period is 150-5 kg/kg, and the weight standard of the fish seeds in the fattening period is more than 100g per tail.
The feeding standard in the breeding period of the fish seeds is as follows: 100 parts of fish feed, 0.1-0.3 part of Chinese herbal medicine mixture, 0.2-0.4 part of organic acid and 0.03-0.05 part of vitamin K. The fish feed is a conventional breeding feed, and meets the weight standard of fish species, for example, the clarias fuscus needs to be fed with the special fish feed for the clarias fuscus, the feed can contain cicada pupa powder, peanut bran, flour, shell powder, compound probiotics and the like, and can also contain corn protein powder, cuttlefish cream, fish oil, soybean phospholipid oil and the like, the proportion of the components is selected according to the reasons of self breeding level, season, water quality and the like, and the components do not belong to the protection scheme of the invention, and do not influence the effect of the Chinese herbal medicine additive of the invention, and the feed can be sold in the common markets.
The feeding standard in the fattening period is as follows: 100 parts of the bred feed, 0.3-0.5 part of the Chinese herbal medicine mixture and 0.3-0.5 part of the organic acid, 0.05-0.1 part of the vitamin K. The breeding feed is a conventional breeding feed, and only meets the weight standard of fish species, for example, the breeding feed special for breeding the pelteobagrus fulvidraco needs to be fed with the pelteobagrus fulvidraco, the feed contains fish meal, soybean meal, rapeseed meal, bran, wheat, corn, inorganic salt and the like, the feed standards of all farmers are different, and the breeding feed can be selected according to self conditions, water quality, seasons and the like, but does not influence the efficacy of the traditional Chinese medicine additive provided by the invention.
The feeding standard in the disease onset stage is as follows: 100 parts of fish feed or breeding feed, 1-1.5 parts of Chinese herbal medicine mixture, 0.5-1.0 part of organic acid and 0.1 part of vitamin K, and continuously feeding for 5-7 days. Meanwhile, the fish pond is required to be disinfected in the onset period, and Chinese herbal medicine additives are used in an auxiliary manner, and the method comprises the following steps: the whole pond is sprayed after enrofloxacin and vitamin K are mixed and diluted, the dilution multiple is based on the dilution multiple of a conventional disinfection and sterilization product, and after the whole pond is disinfected by povidone iodine solution the next day, the bottom of the pond is changed by adopting potassium hydrogen persulfate every other day.

Test example 1
200 pelteobagrus fulvidraco are purchased, averagely divided into 20 groups, 10 fish in each group, and the experimental animal model is established by artificially infecting Edwardsiella sp.
The 20 groups were a control group (blank group, not treated by artificial infection), an infected group, an infection prevention group (administered 24 hours after infection), and an infection treatment group (administered when meat rot symptoms were found on part of the fish body surface after infection).
The drugs used in the infection prevention group and the infection treatment group were the drugs prepared in examples 1 to 3 and comparative examples 1 to 4, respectively, and the results of the prevention rate and the cure rate are shown in Table 1.

Test example 2
The initial weight of the ictalurus punctatus is as follows: 16.5 plus or minus 0.3 g;
experiment feed: control group (longsnout catfish species basal diet), test group a (example 1), test group B (example 3);
the experimental method comprises the following steps: 3 replicates were set for each test group for 9 treatment groups, each holding 30 fish. Stopping feeding for 24h before the experiment begins, then selecting juvenile fish with healthy body surface and regular specification to group, feeding corresponding test feed for each test group, and culturing for 8 weeks. The water temperature is 28.5 +/-0.5 ℃ during the experimental cultivation period, and the pH value is maintained at 7.6-8.0.
Sampling in an experiment: after the breeding experiment is finished, each processing group measures the number (used for calculating the survival rate) and the total weight (used for calculating the growth rate) of the channel catfish, each processing group takes 3 channel catfish to dissect and take the liver tissue of the channel catfish and store the liver tissue at the temperature of-20 ℃ for later use (used for measuring the nonspecific immunity index), each processing group takes 3 channel catfish to dissect and take the foregut of about 2cm, and the channel catfish is placed in HE staining stationary liquid (10% formaldehyde solution) for later use (used for observing intestinal histology).
The experimental process comprises the following steps: 20 channel catfish punctatus are taken from each treatment group to carry out the virus attacking test, and muscle and liver tissues of the channel catfish suffering from the Edwardsiella disease and having serious rottenness are ground (diseased fish is obtained from a terminal diseased catfish farmer and is separated and identified as Edwardsiella infection) and are respectively put into each virus attacking treatment. On the day of challenge, no feed was fed on the next day, on the third day, basal feed was fed to each treatment group and infection and death of the test longsnout catfish (occurrence of fins and portions of fins, congestion around the mouth and oral cavity, and red swelling of anus were all regarded as diseased) were recorded, and the challenge test lasted for 10 days, with the results shown in tables 2 to 3 and fig. 1 to 3.

As can be seen from tables 2 and 3, the Chinese herbal medicine additive is added to the longsnout catfish to apply the combination scheme, so that the growth performance of the longsnout catfish can be promoted, and the terminal weight and the survival rate of the longsnout catfish can be improved; can obviously improve the immunity and the oxidation resistance of the longsnout catfish and improve the body health of the longsnout catfish.
FIG. 1 is a photograph showing the control foregut tissue structure (HE, 100X) showing the vacuolar degeneration of the epithelial cells of the foregut mucosa, the detachment of a part of the striated margin, the proliferation of the goblet epithelial cells, the loosening of the connective tissue of the natural membrane, the irregular growth and the short length of the villi, and the pathological damage of the intestinal tract.
Fig. 2 shows the tissue structure of foregut (HE, 100 x) of test group a, fig. 3 shows the tissue structure of foregut (HE, 100 x) of test group B, and it can be seen from fig. 2 and fig. 3 that the application scheme of the combination of Chinese herbal medicine additives can promote the length and surface area of intestinal villus of longsnout catfish, improve intestinal muscle layer and intestinal cells, and promote healthy development and growth of intestinal tract. Meanwhile, as can be seen from table 4, the Chinese herbal medicine additive of the present invention can be fed differently according to different growth periods, and can also be fed in selectively increased dosage according to onset of disease or non-onset of disease, but during the onset of disease, the combined formula of the corresponding onset of disease is fed, and the effect is better.

Claims (8)
1. The Chinese herbal medicine additive for preventing and treating bacterial rot of alepidote fish is characterized by comprising the following components in parts by weight: 0.1-1.5 parts of Chinese herbal medicine mixture, 0.2-1.0 part of organic acid and 0.03-0.1 part of vitamin K, wherein the Chinese herbal medicine mixture comprises the following components in parts by weight: 20-25 parts of dandelion, 20-25 parts of wormwood, 15-20 parts of houttuynia cordata, 15-20 parts of honeysuckle and 15-20 parts of cinnamon;
every 100 parts of organic acid comprises 70-80 parts of formic acid, 5-10 parts of malic acid, 5-10 parts of butyric acid and 5-10 parts of other organic acid.
2. The herbal additive of claim 1 for controlling bacterial rot of alepidote fish, wherein the herbal mixture is prepared by the following method:
weighing corresponding Chinese herbal medicines according to a proportion, mixing, crushing, and sieving with a 100-mesh sieve to obtain the traditional Chinese medicine.
3. The use of the Chinese herbal medicine additive for preventing and treating bacterial rotten body disease of alepidote fish as claimed in claim 1 or 2, characterized in that the additive is used for mixing and feeding the fish in the breeding period, fattening period and disease period.
4. The use of claim 3, wherein the standard weight of the fish species in the breeding period is 150/jin to 5/jin, and the standard weight of the fish species in the fattening period is more than 100g per tail.
5. The use according to claim 3, wherein the feeding criteria during the fingerling raising period are: 100 parts of fish feed, 0.1-0.3 part of Chinese herbal medicine mixture, 0.2-0.4 part of organic acid and 0.03-0.05 part of vitamin K.
6. The use according to claim 3, wherein the feeding criteria for the fattening period are: 100 parts of the bred feed, 0.3-0.5 part of the Chinese herbal medicine mixture and 0.3-0.5 part of the organic acid, 0.05-0.1 part of the vitamin K.
7. The use according to claim 3, wherein the feeding criteria for the onset are: 100 parts of fish feed or breeding feed, 1-1.5 parts of Chinese herbal medicine mixture, 0.5-1.0 part of organic acid and 0.1 part of vitamin K, and continuously feeding for 5-7 days.
8. The use according to claim 3, wherein the outbreak period further requires a disinfection treatment of the fish pond, comprising the steps of: the whole pond is sprayed after mixing and diluting enrofloxacin and vitamin K, and the bottom is changed by adopting potassium hydrogen persulfate every other day after the whole pond is disinfected by povidone iodine solution the next day.
