Description

Essence water with antioxidant and anti-inflammatory effects
Technical Field
The invention belongs to the field of cosmetics, and particularly relates to essence water with antioxidant and anti-inflammatory effects.
Background
There are a variety of active substances in natural plants, such as: polysaccharides, polypeptides, flavones, polyphenols, amino acids, and the like. The traditional process is to extract active ingredients in plants by using an organic solvent or an aqueous solution, but the obtained active ingredients have low content and sometimes cannot meet the expected efficacy. Research shows that the active ingredients can be enriched after the plant raw materials are fermented. For example, when the lactobacillus acidophilus is used for fermenting tricholoma matsutake, more or stronger whitening substances can be extracted from the tricholoma matsutake, and the whitening effect of the tricholoma matsutake fermentation liquid is better than that of the aqueous extract and the alcohol extract of the tricholoma matsutake.
Chinese plant resources are rich, the difference of planting environments in different regions is large, active substances in plants are complex, a few plant components may have certain toxic and side effects, and effective utilization of a part of plant resources is limited. In addition, the content of toxic components such as alkaloid, lactone substances, glycoside compounds, toxic protein, aristolochic acid, anthraquinone, tannin, heavy metal and the like in plants can be reduced by utilizing a microbial fermentation technology.
Populus tomentosa is a plant of Populus genus of Populicaceae family, also called Populus trifoliata, and is distributed in Liaoning, North China, northwest China, east China, etc. Generally, the cultivation is carried out, the collection can be carried out all the year round, and the resources are extremely rich. Poncirus trifoliata is an important traditional Chinese medicinal material, has wide source, low price and high medicinal value, contains abundant flavonoid compounds, has better free radical scavenging effect and anti-inflammatory effect, and has obvious inhibition effect on bacteria. The active ingredients of the Poncirus trifoliata flower are applied to cosmetics, so that a new choice is brought to the formula design, and meanwhile, the method has certain reference significance for the expansion of the utilization direction of Poncirus trifoliata flower resources.

the results of the experiment are shown in table 1.
TABLE 1 free radical scavenging rates of different Populus trifoliata extracts
Figure BDA0003626634850000042
From the experimental data in table 1, it can be found that the free radical clearance of the extract obtained by fermenting the poplar flower with the microorganism is higher than that of the poplar flower extract obtained by the conventional water extraction method and alcohol extraction method, wherein when the microorganism adopted is saccharomycetes and lactobacillus plantarum (the ratio of viable bacteria is 2: 3), the free radical clearance is higher and reaches 86.8%, which indicates that the poplar flower extract prepared by fermenting with the microorganism has excellent antioxidant activity.
Anti-inflammatory experiments: 60 mice were randomly divided into 12 groups. The blank control group is physiological saline, the positive control group is hydrocortisone 0.02 g/kg-1The remaining groups were treated with the Populus trifoliata fermentation extract, the aqueous extract and the alcoholic extract in the formulation table of example 1. The groups were administered by intragastric administration 1 time in the morning for 7 days, and the positive control group was administered by intraperitoneal injection. 30min after the last administration, 0.2mL of xylene was evenly smeared on both sides of the mouse right auricle with a cotton ball to cause inflammation, and the left auricle was not smeared with the cotton ball to serve as a control. After 1h, the mice were sacrificed by decapitation and both ears were cut off. Round lugs are respectively punched at the same position by a puncher with the diameter of 7mm, the round lugs are weighed, the difference value of the weights of the left and right lugs is used as swelling degree, and the swelling inhibition percentage is calculated. The experimental results are shown in table 2, which shows that the Populus deltoides fermented extract has good anti-inflammatory effect and stronger anti-inflammatory effect on inflammation compared with an aqueous extract and an alcohol extract.

Figure BDA0003626634850000051
The antibacterial ability test method comprises the following steps: coating a layer of diluted experimental bacteria on an agar plate, inversely placing the agar plate in a constant-temperature incubator at 37 ℃, vertically placing an oxford cup on the surface of the agar plate by using an aseptic forceps after the surface is dried, slightly pressing the oxford cup, adding a sample to be detected, and setting a blank bacterial growth control and a positive growth control. Culturing in a constant-temperature incubator at 37 ℃ for 24h, taking out the plate and measuring the diameter of the inhibition zone around each oxford cup. The experimental bacteria include various common strains such as escherichia coli, staphylococcus aureus and aspergillus niger, and the data in this embodiment take escherichia coli as an example.
TABLE 3 antibacterial Capacity of different Populus extracts
Figure BDA0003626634850000052
As can be seen from Table 3, compared with the Populus trifoliata extract obtained by the conventional water extraction method and the alcohol extraction method, the diameter of the inhibition zone of the Populus trifoliata fermented extract is slightly reduced, but the inhibition zone is basically kept in a higher range, and the inhibition effect on experimental bacteria is hardly influenced, wherein the data of the Populus trifoliata fermented extract 7 and the Populus trifoliata alcohol extract are relatively close, which indicates that the Populus trifoliata extract obtained by the formula has relatively strong inhibition effect and relatively good antibacterial effect.
Example 3 moisturizing ability test of Populus trifoliata fermenting extract-containing essence liquid
The experimental conditions are as follows: the test environment temperature is 20 ℃, the relative humidity is 45-55%, the same indoor light condition is kept, and real-time dynamic monitoring is carried out.
Preparation before testing: four groups of volunteers were selected, aged 20-45 years, and tested for skin non-traumatic. The test site cannot use any cosmetics or external drugs 2-3 days before the test. Before the test, a tested part needs to be cleaned by a tested subject, a plurality of areas can be marked simultaneously after the cleaning, the interval between every two testing areas is at least 1cm, basic data is tested after the tested subject sits statically for 30min in a constant-temperature and constant-humidity environment, water and beverage cannot be drunk in the period, the tested subject is placed in a testing state, and a relaxation state is kept.
The method comprises the following steps:
refer to QB/T4256-.
Firstly, a test sample is coated for one time according to a certain dosage, a latex finger sleeve is used for uniformly coating the sample in a test area, and the actual sample coating quantity is recorded.
Measuring the tested part with the skin analyzing and testing instrument for at least 3 times. The initial value of each test area (before sample application) was measured and then the skin moisture content of the test area was measured after a set time.
Measuring the skin water content of the tested area after 1h, 2h and 4h after smearing respectively. The test of the same subject must be done by the same measuring person using the same instrument, and the measuring probe should be cleaned between the two measurements.
The results of the experiment are shown in table 4:
TABLE 4 Effect of essential waters containing different Populus trifoliata fermentation extracts on skin Water content
Figure BDA0003626634850000061
Note: 1-9 parts of Populus trifoliata fermenting extract is correspondingly added, and 10 parts of essence water is essence water containing Populus trifoliata alcohol extract.
From the experimental data in table 4, it can be found that the essence containing the citric flower fermentation extract has a significant moisturizing effect after being used for 1 hour, can still achieve a significant moisturizing effect until 4 hours, is maintained for a long time, and has a significantly improved moisturizing rate compared with an essence sample containing no citric flower fermentation extract.
