According to a second aspect of the present invention, there is also provided a preparation method of the above health composition with effects of soothing nerves and aiding sleep, comprising the following steps:
mixing the raw materials to obtain the health-care composition with the effects of soothing nerves and helping sleep.
The preparation method has simple process and convenient operation, and is suitable for large-scale industrial production.
According to a third aspect of the invention, the invention also provides a health care product, which comprises the health care composition with the functions of soothing the nerves and helping sleep and auxiliary materials.
In view of the advantages of the health-care composition with the effects of soothing the nerves and aiding sleep, the health-care product prepared from the composition has the same advantages.
The specific type of the auxiliary materials is not limited, and those commonly used in the art can be used.
As an alternative embodiment of the invention, the auxiliary material comprises any one or a combination of at least two of a base, a suspending agent, a sweetener or a disintegrant.
The matrix can be used to solubilize a nutraceutical composition, including but not limited to vegetable oils. Examples of the vegetable oil include corn oil, sesame oil, palm oil, sunflower oil, peanut oil, walnut oil, olive oil, soybean oil, and rapeseed oil.
There are many types of suspending agents, including but not limited to beeswax.
The sweetener may be selected from the group of substances commonly used in the art, including but not limited to erythritol, maltitol, xylitol, stevia, sucrose, white sugar, fructose, brown sugar or maltose.
Disintegrants include, but are not limited to, gelatin.
The health-care composition with the functions of soothing the nerves and helping sleep is matched with auxiliary materials and can be prepared into health-care products of different formulations.
As an alternative embodiment of the present invention, the formulation of the health product includes any one of capsules, tablets, pills, granules or oral liquid.
As a preferred embodiment of the present invention, the health product is a soft capsule health product, comprising a health composition and an auxiliary material;
the auxiliary material is peanut oil, and the mass of the health-care composition accounts for 10-40% of the total mass of the health-care composition and the auxiliary material.
The proportion of the mass of the nutraceutical composition to the total mass of the nutraceutical composition and the auxiliary material is typically, but not limited to, 10%, 12%, 14%, 15%, 18%, 20%, 22%, 24%, 25%, 28%, 30%, 32%, 34%, 35%, 38% or 40%.
As a preferred embodiment of the present invention, the preparation method of the health product (soft capsule) comprises the following steps:
mixing the health composition with adjuvants, and filtering to obtain content;
and pelleting the content and the glue solution for forming the soft capsule, shaping and drying to obtain the health-care product in the form of the soft capsule.
The gelatin solution for forming soft capsules may be prepared from conventional raw materials in the art, such as purified water, glycerin, gelatin, and the like. In addition, food pigment such as brilliant blue and amaranth can be added into the glue solution.
According to the fourth aspect of the invention, the application of the health-care composition or the health-care product with the effects of soothing the nerves and helping sleep in the fields of improving sleep and soothing the nerves is also provided.
In view of the effects of the health-care composition or the health-care product with the effects of soothing nerves and helping sleep, the health-care composition or the health-care product has good application in the fields of improving sleep and soothing nerves.
The technical solution provided by the present invention is further described below with reference to examples and comparative examples.
Example 1
The embodiment provides a health-care composition with the effects of soothing nerves and helping sleep, which is prepared from the following raw materials in parts by weight:
70 parts of peanut stem leaf extract, 6 parts of aminobutyric acid, 5 parts of theanine and 6 parts of cordyceps militaris extract.
The preparation method of the peanut stem and leaf extract comprises the following steps:
(a) adding clean water into peanut stem leaves in an extraction tank according to the weight ratio (material-liquid ratio) of 1:5, heating to 92 ℃, soaking for two times, soaking for 2 hours for the first time, filtering and collecting water extract, adding water into raw materials, soaking for 1 hour for the second time at 92 ℃, then combining the two extracting solutions, filtering, centrifuging and evaporating in multiple stages to obtain peanut stem leaf concentrated solution with the specific gravity of 1.1-1.2, and performing spray drying at 200 ℃ to obtain a crude product of peanut stem leaf extract;
(b) and (3) crushing the crude peanut stem and leaf extract in a 10 ten thousand grade clean area, and sieving with a 80-mesh sieve to remove impurities and purify to obtain the peanut stem and leaf extract.
Example 2
The embodiment provides a health-care composition with the effects of soothing nerves and helping sleep, which is prepared from the following raw materials in parts by weight:
70 parts of peanut stem leaf extract, 11 parts of aminobutyric acid, 10 parts of theanine and 9 parts of cordyceps militaris extract.
The preparation method of the peanut stem and leaf extract is the same as that of example 1.
Example 3
The embodiment provides a health-care composition with the effects of soothing nerves and helping sleep, which is prepared from the following raw materials in parts by weight:
65 parts of peanut stem leaf extract, 12 parts of aminobutyric acid, 5 parts of theanine and 10 parts of cordyceps militaris extract.
The preparation method of the peanut stem and leaf extract is the same as that of example 1.
Example 4
The embodiment provides a health-care composition with the effects of soothing nerves and helping sleep, which is prepared from the following raw materials in parts by weight:
85 parts of peanut stem leaf extract, 6 parts of aminobutyric acid, 8 parts of theanine and 5 parts of cordyceps militaris extract.
The preparation method of the peanut stem and leaf extract is the same as that of example 1.
Examples 1 to 4 are processes for preparing a health composition with tranquilizing and sleep-aiding effects, which includes the following steps:
mixing the raw materials to obtain the health-care composition with the effects of soothing nerves and helping sleep.
Comparative example 1
The comparative example provides a health-care composition which is prepared from the following raw materials in parts by weight:
50 parts of peanut stem leaf extract, 20 parts of aminobutyric acid, 18 parts of theanine and 12 parts of cordyceps militaris extract.
Comparative example 2
The comparative example provides a health-care composition which is prepared from the following raw materials in parts by weight:
90 parts of peanut stem leaf extract, 3 parts of aminobutyric acid, 3 parts of theanine and 4 parts of cordyceps militaris extract.
Comparative example 3
This comparative example provides a health composition, except that aminobutyric acid was not added to the raw material of example 1, and the kinds and amounts of the raw materials were the same as those of example 1.
Comparative example 4
This comparative example provides a health composition, except that theanine was not added to the raw materials of example 1, and the kinds and amounts of the other raw materials were the same as those of example 1.

Comparative examples 13 to 24 each provide a health product (soft capsule) comprising the health composition and the adjuvant provided in comparative examples 1 to 12, respectively, and the amount of the health composition, the kind and amount of the adjuvant, and the preparation method of the health product used in the preparation are the same as those in examples 5 to 8, and are not described herein again.
In order to compare the technical effects of the respective examples and comparative examples, the following experimental examples were specifically set.
Examples of the experiments
1. Test article
Test article 1 (comparative example 12), content: 600g/kg of peanut stem and leaf extract, 200g/kg of spina date seed extract and 30g/kg of mulberry extract; specification: 200 mg/granule;
sample 2 (example 1), content: 700g/kg of peanut stem and leaf extract, 60g/kg of aminobutyric acid, 50g/kg of theanine and 60g/kg of cordyceps militaris extract; specification: 200 mg/granule;
test article 3 (comparative example 1), content: 500g/kg of peanut stem and leaf extract, 200g/kg of aminobutyric acid, 180g/kg of theanine and 120g/kg of cordyceps militaris extract; specification: 200 mg/granule.
2. Reagent
Sodium pentobarbital, lot No. 202018010925100, specification: 25 g/bottle, molecular weight: 248.25, manufacturer: merck, Germany.
Barbiturate sodium, lot No. MY4025, specification: 25 g/bottle, molecular weight: 206.17, manufacturer: jiuding chemical (Shanghai) science and technology, Inc.
3. Test animal and breeding environment
40 SPF-grade KM mice, male, were used for the test weighing 18-22g, provided by Jinan Pengyue test animal breeders Co., Ltd, production license number: SCXK (lu) 20190003, test animal quality certification number: no. 370726211101161563. Breeding mice in ten thousand-level barrier environment, trying for 20-25 ℃, ensuring that the daily temperature difference is less than or equal to 3 ℃, ventilating times are 10-20 times/h, the humidity is 40-70%, the pressure gradient is 20-50Pa, and the light and shade alternation time of day and night is 12h/12 h; the test animal uses the license number: SCXK (lu) 20180031.
4. Dosage design and basis
The human dose of the test samples 1, 2 and 3 was 400 mg/day, calculated as 70kg of adult human body weight, 5.71mg/kg of human body dose, and 57.1mg/kg of human body dose, which is 10 times of the recommended dose, was administered to mice. The intragastric administration volume of the mice of the three test sample groups is 20mL/kg, the administration is carried out once a day, the negative control group is provided with pure water with the same volume, and each index is detected after the three test samples are continuously administered for 30/33 d.
5. Pharmaceutical formulation
Test article 1, 2, 3 solution: 0.043g of each of the powders 1, 2 and 3 was weighed out, dissolved in pure water and diluted to 15mL, and the concentration of the solution was 2.87 g/mL.
6. Main instrument equipment
Model LE203E/02 electronic balance, mettler-toledo (shanghai) ltd.
Model AUW-120D electronic balance, shimadzu, japan.
Electronic balance model Uw2200H, shimadzu, japan.
7. Test method
(1) Direct sleep test
And observing whether the animals in the tested group are fed with the test products with different formulas, and whether the animals in the control group are in sleep after the animals in the control group are fed with the solvent with the same volume. The disappearance of the righting reflex is used as an index for sleeping. When the mouse is placed in the dorsal position, it can turn over right immediately. If the patient can not be righted for more than 30-60 seconds, the righting reflex is considered to disappear, and the patient enters sleep. The recovery of the righting reflex is the awakening of the animal, the time from disappearance of the righting reflex to recovery is the sleeping time of the animal, and the number of sleeping animals and the sleeping time of the negative control group and the test sample group are recorded.
(2) Pentobarbital sodium subthreshold dose hypnosis test
A pre-test is carried out before a formal test, and the sub-threshold hypnotic doses of the sodium pentobarbital are respectively determined to be 32mg/kg (administration for 7 days), 34mg/kg (administration for 14 days) and 35mg/kg (administration for 30 days), namely the maximum sub-threshold dose of the sodium pentobarbital of which 80-90% of mice turn over normal reflex does not disappear. 30min after animals are given the test article or pure water last time, the maximum sub-threshold hypnotic dose of the sodium pentobarbital is injected into the abdominal cavity of each group of animals, and the number of the animals falling asleep within 30min is recorded (the animals with the turn-over reflex disappeared for more than 1 min). The test was carried out in a quiet environment at 24-25 ℃.
(3) Test for prolonging sleep time of sodium pentobarbital
The pre-test is carried out before the formal test, the dose of the sodium pentobarbital which enables the animals to fall asleep for 100 percent but does not lead to overlong sleep time is respectively 53mg/kg (7 days and 14 days of administration) and 54mg/kg (30 days of administration), and the formal test is carried out by using the doses. After the animals are fully conscious in the sub-threshold dose hypnosis test of the pentobarbital, the pentobarbital sodium is injected into the abdominal cavity of each group of animals, the disappearance of the righting reflex is taken as an index, and the sleep time of the pentobarbital sodium prolonged by the test sample is observed.
(4) Barbiturate sodium sleep latency test
Before the formal test, a preliminary test is carried out, and the barbiturate sodium dose which enables the animals to fall asleep for 100% but does not lead to excessive sleep time is determined to be 270mg/kg (administration lasts for 33 days), and the formal test is carried out by using the dose. 30min after animals are given test samples with different formulas or pure water at the last time, injecting barbital sodium solution into abdominal cavities of animals of each group, and observing the influence of the test samples on the sleep latency of the barbital sodium by taking the disappearance of the righting reflex as an index.
(5) Statistics of test data
The metrology data was analyzed using one-way analysis of variance (ANOVA) or T-test. If the measured data has uniform variance, adopting single-factor analysis of variance LSD test. If the variance of the metrology data is not uniform, a one-way analysis of variance Dunnett's T3 test is used. The number of sleeping animals is counted by X2And (4) testing, and when the total case number of the four-table is less than 40, or the total case number is equal to or greater than 40 but the theoretical number is equal to or less than 1, applying an exact probability method.
(6) Determination of results
Direct sleep test: and comparing the difference between the number of the animals falling asleep and the sleeping time of the control group and the test group, and if the number of the animals falling asleep or the sleeping time is obviously increased, the test result is positive.
Pentobarbital sodium subthreshold dose hypnosis test: and comparing the difference between the numbers of the animals falling asleep in the control group and the test group, wherein the incidence rate of the animals falling asleep is obviously increased, and the test result is positive.
Test for prolonging sleep time of sodium pentobarbital: and comparing the difference between the test group and the control group in the sleep time extension, wherein the test result is positive if the sleep time extension is significant.
Barbiturate sodium sleep latency test: comparing the difference between the sleep latency of the control group and the sleep latency of the test group, if the sleep latency is obviously shortened, the test result is positive.
The test sample can be judged to have the function of improving the sleep function.
8. Test results
(1) Influence of three formula test samples of peanut stem leaves on direct sleep of mice
Animals in the test group were given 3 formulations of the test article and no direct sleep was observed in the negative control group after the same volume of solvent.
(2) Influence of three formula test samples of peanut stem leaves on number of mice pentobarbital sodium sleeping animals
When the test articles 1, 2 and 3 are administrated for 7 days, the number of animals falling asleep is 3, 4 and 2 respectively; at 14 days of administration, the number of sleeping animals was 2; the number of animals falling asleep at 30 days of administration was 3, 6 and 3, respectively. Through statistical analysis, the number of sleeping animals in each test sample group is increased, but the difference is not statistically significant compared with a negative control group (P > 0.05). Specific results are shown in table 1.

(3) Influence of three peanut stem leaf samples on prolonging of pentobarbital sodium sleep time of mice
Test article 2 tends to prolong the sleep time of the mice by 7 days when administered as compared with the negative control group (P ═ 0.07); the test products 1, 2 and 3 are administrated for 14 days, and the effect of prolonging the sleep time of the mouse is not found; the test article 1 and the test article 2 can be administrated for 30 days, and the sleep time of the mouse can be obviously prolonged (P is less than 0.05). The specific results are shown in Table 2.

(4) Influence of three test samples of peanut stems and leaves on sleep latency of barbital sodium in mice
Through statistical analysis, the test article 2 can shorten the sleep latency of mice after being administrated for 33 days, and the difference has statistical significance (P is less than 0.05).

9. Conclusion of the experiment
After the test article 1, 2 and 3 preparation 30/33d with the dose of 57.1mg/kg is orally administered to the mice, compared with a negative control group, the test article 2 can obviously prolong the sleep time of the pentobarbital sodium mice after being administered for 30 days, the difference has significance (P <0.05), the sleep latency period of the pentobarbital sodium mice can be obviously shortened (P <0.05) after being administered for 33 days, and the direct sleep effect is avoided. From this, it was found that sample 2 had an effect of improving the sleep function of mice.
Finally, it should be noted that: the above embodiments are only used to illustrate the technical solution of the present invention, and not to limit the same; while the invention has been described in detail and with reference to the foregoing embodiments, it will be understood by those skilled in the art that: the technical solutions described in the foregoing embodiments may still be modified, or some or all of the technical features may be equivalently replaced; and the modifications or the substitutions do not make the essence of the corresponding technical solutions depart from the scope of the technical solutions of the embodiments of the present invention.

Claims (10)

1. A health-care composition with the effects of calming nerves and helping sleep is characterized by comprising the following raw materials in parts by weight:
65-85 parts of peanut stem leaf extract, 6-12 parts of aminobutyric acid, 5-10 parts of theanine and 5-10 parts of cordyceps militaris extract.
2. The health-care composition as claimed in claim 1, which is characterized by comprising the following raw materials in parts by weight:
68-80 parts of peanut stem and leaf extract, 8-11 parts of aminobutyric acid, 6.5-10 parts of theanine and 5.5-9 parts of cordyceps militaris extract.
3. A health composition as claimed in claim 1 or 2, wherein the preparation method of the peanut stem and leaf extract comprises the steps of:
extracting stems and leaves of peanut with water to obtain an extracting solution, concentrating and drying to obtain a crude product of the extract of the stems and leaves of the peanut;
and (3) crushing and purifying the crude peanut stem and leaf extract to obtain the peanut stem and leaf extract.
4. A health composition as claimed in claim 3, wherein the extract of peanut stem and leaf is prepared by extracting peanut stem and leaf with water for 1-3 times;
preferably, the temperature of each extraction is respectively and independently 90-95 ℃, and the time of each extraction is respectively and independently 1-2 h;
preferably, the feed-liquid ratio of each extraction is 1: (5-10).
5. A health composition as claimed in claim 3, wherein the peanut stem and leaf extract is prepared by concentrating the extractive solution to specific gravity of 1.1-1.2;
preferably, the drying is spray drying;
preferably, the temperature of the spray drying is 190-210 ℃;
preferably, the pulverized particle size is 80-100 mesh.
6. A method for preparing a health composition with tranquilizing and sleep-aiding effects according to any one of claims 1 to 5, comprising the steps of:
mixing the raw materials to obtain the health-care composition with the effects of soothing nerves and helping sleep.
7. A health product, which is characterized by comprising the health composition with the effects of soothing the nerves and aiding sleep of any one of claims 1 to 5 and auxiliary materials.
8. The health product of claim 7, wherein the adjuvant comprises any one or a combination of at least two of a matrix, a suspending agent, a sweetener, or a disintegrant.
9. The health product according to claim 7 or 8, wherein the dosage form of the health product comprises any one of capsules, tablets, pills, granules or oral liquid.
10. Use of the health composition with tranquilizing and sleep-aiding effects of any one of claims 1 to 5 or the health product of any one of claims 7 to 9 in the fields of improving sleep and tranquilizing.
